Adverse childhood experiences (ACEs) negatively affect the function and structure of emotion brain circuits, increasing the risk of various psychiatric disorders. It is unclear if ACEs show disorder specificity with respect to their effects on brain structure. We aimed to investigate whether the structural brain effects of ACEs differ between patients with major depression (MDD) and borderline personality disorder (BPD). These disorders share many symptoms but likely have different etiologies. To achieve our goal, we obtained structural 3T-MRI images from 20 healthy controls (HC), 19 MDD patients, and 18 BPD patients, and measured cortical thickness and subcortical gray matter volumes. We utilized the Adverse Childhood Experiences (ACE) questionnaire to quantify self-reported exposure to childhood trauma. Our findings suggest that individuals with MDD exhibit a smaller cortical thickness when compared to those with BPD. However, ACEs showed a significantly affected relationship with cortical thickness in BPD but not in MDD. ACEs were found to be associated with thinning in cortical regions involved in emotional behavior in BPD, whereas HC showed an opposite association. Our results suggest a potential mechanism of ACE effects on psychopathology involving changes in brain structure. Background: Repetitive negative thinking (RNT) is a frequent symptom of major depressive disorder (MDD) that is associated with poor outcomes and treatment resistance. While most studies on RNT have focused on structural and functional characteristics of gray matter, this study aimed to examine the association between white matter (WM) tracts and interindividual variability in RNT., Methods: A probabilistic tractography approach was used to characterize differences in the size and anatomical trajectory of WM fibers traversing psychosurgery targets historically useful in the treatment of MDD (anterior capsulotomy, anterior cingulotomy, and subcaudate tractotomy) in patients with MDD and low (n = 53) or high (n = 52) RNT, and healthy control subjects (n = 54). MDD samples were propensity matched on depression and anxiety severity and demographics., Results: WM tracts traversing left hemisphere targets and reaching the ventral anterior body of the corpus callosum (thus extending to contralateral regions) were larger in the high-RNT MDD group compared with low-RNT (effect size D = 0.27, p = .042) and healthy control (D = 0.23, p = .02) groups. MDD was associated with greater size of tracts that converge onto the right medial orbitofrontal cortex regardless of RNT intensity. Other RNT-nonspecific findings in MDD involved tracts reaching the left primary motor and right primary somatosensory cortices., Conclusions: This study provides the first evidence to our knowledge that WM connectivity patterns, which could become targets of intervention, differ between high- and low-RNT participants with MDD. Adverse childhood experiences (ACEs) have lifelong effects on emotional behavior and are frequent in Borderline Personality Disorder (BPD) and Major Depressive Disorder (MDD). The Central Autonomic Network (CAN), which modulates heart rate variability (HRV), comprises brain regions that mediate emotion regulation processes. However, it remains unclear the effect of ACEs on CAN dynamics and its relationship with HRV in these disorders. We studied the effects of ACEs on the brain and HRV simultaneously, during regulation of psychological stress in 19 BPD, 20 MDD and 20 healthy controls (HC). Participants underwent a cognitive reappraisal task during fMRI with simultaneous ECG acquisition. ACEs exposure was associated with increased activity of CAN and salience network components in patients with MDD compared to BPD during cognitive reappraisal. A brain-autonomic coupling was found in BPD relative to HC during emotion regulation, whereby greater activity of left anterior cingulate and medial superior frontal gyrus areas was coupled with increased HRV. Results suggest that ACEs exposure is associated with a distinct activation of the CAN and salience network regions governing responses to psychological stress in MDD compared to BPD. A traditional hallmark of cognitive impairment associated with late-onset Alzheimer´s disease (LOAD) is episodic memory impairment. However, early alterations have been identified in brain regions associated with executive function in asymptomatic, middle-age offspring of patients with LOAD (O-LOAD) compared to those with no family history. We hypothesized that executive function among O-LOAD would correlate with structural and amyloid brain imaging differently from those without a family history of LOAD (control subjects, CS). Executive function, cortical thickness, and in-vivo Aβ deposits were quantified in 30 O-LOAD and 25 CS. Associations were observed among O-LOAD only. Cortical thickness in the left lateral orbitofrontal cortex was positively associated with Design Fluency. The Stroop Color and Word Test, correlated positively with right rostral mid-frontal cortex thickness. Trails Making Test-B was inversely related to left medial orbitofrontal thickness. Tower of London total time was positively associated with β-amyloid deposition in the right precuneus. These results support previous evidence that early executive dysfunction might reflect subtle, early changes in persons at risk of LOAD and suggests that executive function alterations deserve further exploration in the LOAD literature., (Copyright © 2021 Elsevier B.V. Background: Major depressive disorder (MDD) is the leading cause of years lived with disability worldwide, and up to 40% of individuals with MDD do not respond to current treatments. Studies suggest that peripheral inflammation plays an important role in the striatal mesolimbic dopamine pathway and corticostriatal reward circuitry in MDD. Although MDD patients show blunted striatal responses to reward, the link between degree of inflammation and attenuation of reward processing is unclear. We investigated whether MDD patients with elevated peripheral inflammation exhibit attenuated reward responses to enhance our understanding of MDD pathophysiology and develop more effective treatments for current non-responders., Methods: MDD subjects varying on serum C-reactive protein (CRP) concentrations (MDD-High CRP, >3 mg/L, n = 44; MDD-Low CRP, <3 mg/L, n = 44) and healthy comparisons (HC, n = 44) completed a monetary incentive delay (MID) task and provided blood samples to measure inflammation-related markers. MDD-High and MDD-Low were propensity score-matched on age, sex, body mass index (BMI), smoking status, exercise and MID task head motion. Percent change in blood oxygen level-dependent (BOLD) signal during anticipation of wins and losses was extracted from bilateral nucleus accumbens, dorsal caudate and dorsolateral putamen regions of interest (ROIs). A linear mixed-effects model was used to test group (MDD-High, MDD-Low and HC), condition (large-win, small-win and no win), and their interaction for these ROIs as well as whole-brain voxelwise data. Analyses also tested group differences in inflammatory mediators. Correlations were used to explore the relationship between inflammatory mediators and brain regions showing differences between MDD-High and MDD-Low., Results: MDD-High exhibited: (a) lower BOLD signal change in dorsal caudate, thalamus, left insula and left precuneus during anticipation of small wins than MDD-Low; and (b) higher serum soluble intercellular adhesion molecule 1 (sICAM-1) and interleukin 6 (IL-6) concentrations than MDD-Low and HC. MDD as a whole, regardless of CRP-based inflammation, exhibited: (a) lower precuneus BOLD signal change to large wins than HC; and (b) higher Interleukin 1 receptor antagonist (IL-1ra), macrophage-derived chemokine (MDC) and macrophage inflammatory protein-1 alpha (MIP-1α) concentrations than HC. Higher serum sICAM-1 concentrations were associated with lower caudate BOLD signal change to small wins only within the MDD-High group., Conclusion: Within MDD patients, high inflammation (CRP, sICAM-1) was linked to reduced striatal activation recruited to discriminate intermediate reward magnitudes. The present study sought to determine whether autonomic activity is associated with dominance in verbal over spatial reasoning tasks. A group of 19 healthy adults who performed a verbal and spatial aptitude test was evaluated. Autonomic function was assessed by means of heart rate variability analysis, before and during the tasks. Heart rate variability (HRV) is a complex signal that results from the contribution of different sources of oscillation related to the autonomic nervous system activity. Although linear analysis of HRV has been applied to sleep studies, the nonlinear dynamics of HRV underlying frequency components during sleep is less known. We conducted a study to evaluate nonlinear HRV within independent frequency components in wake status, slow-wave sleep (SWS, stages III or IV of non-rapid eye movement sleep), and rapid-eye-movement sleep (REM). The sample included 10 healthy adults. Polysomnography was performed to detect sleep stages. HRV was studied globally during each phase and then very low frequency (VLF), low frequency (LF) and high frequency (HF) components were separated by means of the wavelet transform algorithm. HRV nonlinear dynamics was estimated with sample entropy (SampEn). A higher SampEn was found when analyzing global variability (Wake: 1.53+/-0.28, SWS: 1.76+/-0.32, REM: 1.45+/-0.19, p=0.005) and VLF variability (Wake: 0.13+/-0.03, SWS: 0.19+/-0.03, REM: 0.14+/-0.03, p<0.001) at SWS. REM was similar to wake status regarding nonlinear HRV. Reduction in overall heart rate variability (HRV) associated with aging is determined by a decreased amplitude of heart rate oscillations at all frequency levels, including high frequency (HF) oscillations attributed to respiratory sinus arrhythmia, low frequency (LF) oscillations attributed to Meyer waves and very low frequency (VLF) oscillations of an uncertain origin, presumably linked among others to thermoregulation. However, no studies were conducted to determine whether heart rate oscillations at independent frequency levels show themselves reduced HRV. Wavelet transform was applied to filter specific frequency components of HRV in a sample of younger (21-34 years old) and older (68-85 years old) healthy subjects. HRV indexes were measured within HF, LF and VLF components. The standard deviation of all RR intervals (SDNN) and the square root of the mean squared differences of successive RR intervals (RMSSD) were used as conventional linear time-domain measures. Sample entropy (SampEn) was used as a measure of nonlinear variability. Aged subjects showed lower SDNN at VLF (p < 0.001), LF (p = 0.007) and HF (p < 0.001). Lower RMSSD was observed in older people at VLF (p < 0.001), LF (p = 0.005) and HF (p < 0.001). SampEn was reduced by aging only at VLF level (p < 0.001). In aged people, linear variability was diminished within all frequency components, while nonlinear variability was lower only at VLF level. Preserved central, nonreflex autonomic modulation over the baroreflex control and the central cardiopulmonary coupling might explain this observation. Potential applications of this method include the study of heart rate regulation during sleep in which a complex interaction between the sympathetic and the parasympathetic activity takes place.
