A common experimental technique for viewing in vivo angiogenesis utilises tumours implanted into a test animal cornea. The cornea is avascular but the tumour promotes vascularisation from the limbus and the new blood vessels can be readily observed through the transparent cornea. Many of the early mathematical models for tumour angiogenesis used this scenario as their experimental template and as such assumed that there is a large gap, of the order of 2mm, between the tumour and neighbouring vasculature at the onset of angiogenesis. In this work we consider whether the assumption that there is a significant gap between the tumour and neighbouring vasculature is unique to intra-cornea tumour implants, or whether this characterises avascular tumour growth more generally. To do this we utilise a simple scaling argument, derive a multi-compartment model for tumour growth, and consider in vivo images. This analysis demonstrates that the corneal implant experiments and the corresponding mathematical models cannot generally be applied to a clinical setting., (Copyright © 2011 Elsevier Ltd. A hybrid cellular automaton model is described and used to simulate early tumor growth and examine the roles of host tissue vascular density and tumor metabolism in the ability of a small number of monoclonal transformed cells to develop into an invasive tumor. The model incorporates normal cells, tumor cells, necrotic or empty space, and a random network of native microvessels as components of a cellular automaton state vector. Diffusion of glucose and H(+)ions (the latter largely resulting from the tumor's excessive reliance on anaerobic metabolism) to and from the microvessels, and their utilization or production by cells, is modeled through the solution of differential equations. In this way, the cells and microvessels affect the extracellular concentrations of glucose and H(+)which, in turn, affect the evolution of the automaton. Simulations of the model demonstrate that: (i) high tumor H(+)ion production is favorable for tumor growth and invasion; however for every H(+)ion production rate, there exists a range of optimal microvessel densities (leading to a local pH favorable to tumor but not to normal cells) for which growth and invasion is most effective, (ii) at vascular densities below this range, both tumor and normal cells die due to excessively low pH, (iii) for vascular densities above the optimal range the microvessel network is highly efficient at removing acid and therefore the tumor cells lose their advantage over normal cells gained by high local H(+)concentration. While significant spatial gradients of glucose formed, no regions of detrimentally poor glucose perfusion (for either cell type) were observed, regardless of microvessel density. Depending on metabolic phenotype, a variety of tumor morphologies similar to those clinically observed were realized in the simulations. Lastly, a sharp transition (analogous to that of the adenoma-carcinoma sequence) between states of initial tumor confinement and efficient invasiveness was observed when H(+)production reached a critical value., (Copyright 2001 Academic Press.)

Faecal strongyle egg counts were monitored in peri-parturient ewes and dry ewes grazing in a rubber plantation in North Sumatra, Indonesia. A significant rise in faecal egg counts was observed during the post-parturient period. Haemonchus contortus was the dominant species in the faecal cultures of the lactating ewes. The intensity of the peri-parturient rise was affected by the genotype of the sheep, which were the local Sumatra and its F1 crosses with St. Croix, Barbados Blackbelly and Java Fat-tail. The peri-parturient rise in egg counts was highest in Sumatra and lowest in the Barbados Blackbelly x Sumatra cross. A significant effect of litter size on the peri-parturient rise was also demonstrated. These findings are discussed in relation to the epidemiology and control of gastrointestinal nematodes in the humid tropics.

Fifty-five rams aged about 18-24 months weighing 30-42 kg were used in this study. Ten rams of each of four genotypes, S (Sumatra), H1 (50% Sumatra-50% Virgin Island), B1 (50% Sumatra-50% Barbados Blackbelly), E1 (50% Sumatra-50% Java Fat-tail), were infected orally with a newly isolated strain of Haemonchus contortus. Each animal received 2000 infective larvae 3 times week-1 for 3 weeks, with a total of 18,000 larvae. Fifteen rams belonging to Sumatra and its crosses with Virgin Island were used as uninfected controls. Peak egg counts were observed on Day 35 for genotype B1 and on Day 42 for genotype H1 and S. In genotype E1 a slow but consistent increase in EPG continued until Day 49 when the experiments terminated. Overall faecal egg counts at all sampling dates were not statistically different between genotypes (P > 0.05). There was a large variation in the EPG of individual rams within a genotype. The overall average EPG (geometric means) of individual rams within a genotype ranged from 3 to 1028 for B1, 4 to 261 for E1, 7 to 3119 for H1 and 9 to 506 for S. The analysis of packed cell volume (PCV) for four genotypes and controls from all sampling times showed significant differences (P < 0.05). The overall mean PCV was highest in S (31.1) and lowest in H1 (28.4). The ranking of four genotypes for PCV was S > E1 > B1 > H1. The decrease of PCV during the course of infection was highly significant for all genotypes of infected rams (B1, E1, H1, S) (P < 0.01). PCV of the control group did not exhibit much change during the course of the experimental period. Weight gain of infected rams was lower than those of uninfected controls (P < 0.5) but there was no significant difference between the four genotypes of infected rams. Individual variation within genotype in susceptibility to infections was generally more important than between genotype differences. Two major conclusions of the present study are: (1) The imported breeds with higher body weight, namely the Barbados Blackbelly and Virgin Island, may be used in cross breeding to increase the body size of local Sumatra sheep. (2) Based on the faecal egg counts it is possible to identify the animals for use in selective breeding programmes for higher resistance to H. contortus.

Zenker's diverticula are esophageal outpouchings in the region of the cricopharyngeus which typically grow in a posterolateral and inferior direction and can cause esophageal compression if the diverticulum is large enough. We describe a patient in whom a Zenker's diverticulum expanded posteriorly and superiorly so that it was positioned in the posterior pharyngeal space, causing both esophageal and tracheal narrowing.

Population ecology mathematical models examine tumors not as an isolated collection of transformed individuals but as part of a dynamic society of interacting malignant and normal cells. This approach investigates the mechanisms by which a small clone of neoplastic cells is able to replace the much larger and previously stable population of normal cells, despite the numerical advantage of the latter and the inhibitory effects of the host response. The models define a sequence of different stable equilibria with critical mathematical parameters which control the outcome of different stages of the neoplasm-host competition--parameters which can be correlated with cellular physiologic properties. When neoplasm is viewed as a network of interacting tumor and normal populations, a unifying hypothesis can be developed that allows the diverse but inconsistent properties of transformed cells to be understood according to their specific contributions to tumorigenesis within this network. It predicts general sequences of genetic changes necessary for tumor survival and invasion and demonstrates that apparently disparate properties found in different tumor models can be functionally equivalent. The paper proposes novel modes of therapy requiring classification and treatment of tumors according to the strategies they employ, rather than the traditional criteria of cell type and organ of origin.
