Introduction: Serious Staphylococcus aureus (SA) infection is one of the most life-threatening diseases. Interferon-induced protein 35 (IFP35) is a pleiotropic factor that participates in multiple biological functions, however, its biological role in SA infection is not fully understood. Ferroptosis is a new type of regulated cell death driven by the accretion of free iron and toxic lipid peroxides and plays critical roles in tissue damage. Whether ferroptosis is involved in SA-induced immunopathology and its regulatory mechanisms remain unknown. Objectives: We aimed to determine the role and underlying mechanisms of IFP35 in SA-induced lung infections. Methods: SA infection models were established using wild-type (WT) and IFP35 knockout (Ifp35−/−) mice or macrophages. Histological analysis was performed to assess lung injury. Quantitative real-time PCR, western blotting, flow cytometry, and confocal microscopy were performed to detect ferroptosis. Co-IP and immunofluorescence were used to elucidate the molecular regulatory mechanisms. Results: We found that IFP35 levels increased in the macrophages and lung tissue of SA-infected mice. IFP35 deficiency protected against SA-induced lung damage in mice. Moreover, ferroptosis occurred and contributed to lung injury after SA infection, which was ameliorated by IFP35 deficiency. Mechanically, IFP35 facilitated the ubiquitination and degradation of nuclear factor E2-related factor 2 (Nrf2), aggravating SA-induced ferroptosis and lung injury. Conclusions: Our data demonstrate that IFP35 promotes ferroptosis by facilitating the ubiquitination and degradation of Nrf2 to exacerbate SA infection. Targeting IFP35 may be a promising approach for treating infectious diseases caused by SA.

Objective: To investigate the role and the molecular mechanisms of apoptotic signaling in ferroptosis to regulate tumor radiosensitivity. Methods: Reactive oxygen species (ROS) and lipid peroxide levels were detected in Mouse embryonic fibroblasts(MEFs) with Bcl-xL or Mcl-1 deficiency induced by erastin. Colony formation, ROS, lipid peroxidation and the transcription/translation levels of PTGS2 were measured in Bcl-xL knockdown tumor cells induced by 5 ​Gy γ-rays or co-treated with ferrostatin-1 (Ferr-1). The protein levels of LPCAT3, ACSL4 and PEBP1 in Bcl-xL knockout MEF cells were evaluated in Bcl-xL knockout MEF cells post-radiation. Moreover, the interaction of heat shock protein 90 (HSP90) with Bcl-xL, GPX4, or LAMP2A was detected by protein mass spectrometry and immunoprecipitation assays. Results: Manipulating Bcl-xL levels facilitated radiation-induced ferroptosis by augmenting the enzymatic oxidation of polyunsaturated fatty acids (PUFAs) and enhancing chaperone-mediated autophagy (CMA) of glutathione peroxidase 4 (GPX4) (MEF cell line: t=4.540, P＜0.01; A549 ​cell line: t=56.16, P＜0.0001; t=4.885, P＜0.01; HCT116 ​cell line: t=14.75, P＜0.01; t=7.363, P＜0.05). Downregulating Bcl-xL expression promoted the activity of acyl-CoA synthetase long-chain family member 4 (ACSL4), thus increasing the enzymatic oxidation of PUFAs (t=4.258, P＜0.01). Moreover, depletion of Bcl-xL expedited the CMA process targeting GPX4 by facilitating the association of GPX4 with heat shock protein 90 (HSP90) and LAMP2A following radiation exposure. Subsequent degradation of GPX4 led to the accumulation of lipid peroxides, ultimately triggering ferroptosis. Conclusions: Our study provides initial insights into the regulatory role of Bcl-xL in ferroptosis and underscores the potential of targeting Bcl-xL as a promising therapeutic strategy for cancer by modulating both apoptotic and ferroptotic pathways.

Introduction: Epidemiological evidences reveal that populations with psychological stress have an increased likelihood of respiratory viral infection involving influenza A virus (IAV) and SARS-CoV-2. Objectives: This study aims to explore the potential correlation between psychological stress and increased susceptibility to respiratory viral infections and how this may contribute to a more severe disease progression. Methods: A chronic restraint stress (CRS) mouse model was used to infect IAV and estimate lung inflammation. Alveolar macrophages (AMs) were observed in the numbers, function and metabolic-epigenetic properties. To confirm the central importance of the gut microbiome in stress-exacerbated viral pneumonia, mice were conducted through microbiome depletion and gut microbiome transplantation. Results: Stress exposure induced a decline in Lactobacillaceae abundance and hence γ-aminobutyric acid (GABA) level in mice. Microbial-derived GABA was released in the peripheral and sensed by AMs via GABAAR, leading to enhanced mitochondrial metabolism and α-ketoglutarate (αKG) generation. The metabolic intermediator in turn served as the cofactor for the epigenetic regulator Tet2 to catalyze DNA hydroxymethylation and promoted the PPARγ-centered gene program underpinning survival, self-renewing, and immunoregulation of AMs. Thus, we uncover an unappreciated GABA/Tet2/PPARγ regulatory circuitry initiated by the gut microbiome to instruct distant immune cells through a metabolic-epigenetic program. Accordingly, reconstitution with GABA-producing probiotics, adoptive transferring of GABA-conditioned AMs, or resumption of pulmonary αKG level remarkably improved AMs homeostasis and alleviated severe pneumonia in stressed mice. Conclusion: Together, our study identifies microbiome-derived tonic signaling tuned by psychological stress to imprint resident immune cells and defensive response in the lungs. Further studies are warranted to translate these findings, basically from murine models, into the individuals with psychiatric stress during respiratory viral infection.

Hepatocellular carcinoma (HCC) is one of the most lethal malignancies worldwide. Sorafenib (Sf) is currently the first-line treatment for HCC. However, due to the side effects and unsatisfied efficiency of Sf, it is urgent to combine different therapeutic agents to inhibit HCC progression and increase the therapeutic efficacy. Here, our study constructed a Sf and KIAA1199-siRNA co-loaded liposome Sf-Lp-KIAA, which was prepared by electrostatic interaction of KIAA1199-siRNA and Sf loaded liposome (Sf-Lp). The particle size, zeta potential, the in vitro cumulative release was investigated. The physical and chemical properties were characterized, and the inhibition of HepG2 growth and metastasis in vitro was investigated. The cellular uptake of the co-loaded liposome was significantly higher than that of free siRNA, and the drug/siRNA could be co-delivered to the target cells. Sf-Lp-KIAA could significantly inhibit the growth, migration, invasion and down-regulate KIAA1199 expression of HepG2 cells in vitro than that of single Sf treated group. In addition, the co-delivery liposome accumulated in the HepG2 subcutaneous tumor model and suppress tumor growth after systemic administration without induce obvious toxicity. The present study implied that the co-delivery of Sf and KIAA1199-siRNA through the co-loaded liposomes exerted synergistic antitumor effects on HCC, which would lay a foundation for HCC therapy in the future.

ABSTRACT: Control of meat quality traits is an important goal of any farm animal production, including poultry. A better understanding of the biochemical properties of muscle fiber properties that drive muscle development and ultimately meat quality constitutes one of the major challenging topics in animal production and meat science. In this paper, the existing classification methods of skeletal muscle fibers in poultry were reviewed and the relationship between contractile and metabolic characteristics of muscle fibers and poultry meat quality was described. Finally, a comprehensive review of multiple potential factors affecting muscle fiber distribution and conversion is presented, including breed, sex, hormones, growth performance, diet, muscle position, exercise, and ambient temperature. We emphasize that knowledge of muscle fiber typing is essential to better understand how to control muscle characteristics throughout the life cycle of animals to better manage the final quality of poultry meat.

This research presents a novel method for objectively evaluating college badminton players' physical function levels. It examines current evaluation methods before proposing a novel model that combines Particle Swarm Optimization (PSO) with Backpropagation (BP) neural networks and data mining. The model establishes an evaluation index system that considers physical form, function, quality, and neural mechanisms. The study uses PSO-BP neural networks to adjust indicator weights for more accurate ratings. This recurrent improvement reduces errors while increasing prediction ability, resulting in accurate assessments of athletes' physical talents and neurological insights. The model's efficiency is proved by low mistakes and high accuracy results, which are critical for training optimization and injury avoidance. The combination of PSO optimization and BP neural networks offers robustness across various athlete profiles and training scenarios. This method improves physical function evaluation in badminton and has wider implications for sports science and performance analytics. This study uses bio-inspired computing and machine learning to emphasize the relevance of data-driven techniques in enhancing athlete assessments for better training outcomes and general well-being.

Background: The predictive value of growth differentiation factor-15 (GDF-15) in coronary microvascular dysfunction (CMD) following primary percutaneous coronary intervention (PPCI) in ST-segment elevation myocardial infarction (STEMI) patients is unclear. Methods: This study continuously recruited STEMI patients treated with PPCI at the Chest Pain Center of Qilu Hospital of Shandong University from April 2023 to December 2023. Blood samples were taken before PPCI and the level of circulating GDF-15 was measured by enzyme-linked immunosorbent assay (ELISA), and the patients were divided into CMD and Control group according to angiographic microvascular resistance (AMR) (cut-off value 2.50 mmHg*s/cm). The differences in GDF-15 expression levels between the two groups were compared, and the predictive value of GDF-15 for CMD was systematically evaluated. Results: A total of 134 patients, with an average age of 59.78 ± 12.69 years and 75.37 % being male, were included in this study. Although the store-operated Ca2+ entry (SOCE) plays a critical role in maintaining Ca2+ homeostasis in vascular endothelial cells (VECs), its role in regulating endothelium-dependent hyperpolarization (EDH)-mediated vasorelaxation is largely unknown. Inflammatory bowel disease (IBD) and irritable bowel syndrome (IBS) are the most common gastrointestinal disorders with no effective cures. The present study applied N,N,N′,N′-tetrakis (2-pyridylmethyl)ethylenediamine (TPEN) as a Ca2+ chelator in the endoplasmic reticulum (ER) to study the SOCE/EDH-mediated vasorelaxation of micro-arteries and their involvements in the pathogenesis of IBD and IBS. Human submucosal arterioles and the second-order branch of 6–8 weeks male C57BL/6 mouse mesenteric arterioles were used, and TPEN-induced vasorelaxation was recorded by Danish DMT520A microvascular measuring system. The mice were fed water with 2.5 % dextran sulfate sodium for 7 days to induce mouse model of ulcerative colitis, and water avoidance stress was used to induce mouse model of IBS. The statistical significance of differences in the means of experimental groups was determined using a t-test for two groups or one-way ANOVA for more than two groups. TPEN concentration-dependently induced vasorelaxation of human colonic submucosal arterioles and the second-order branch of murine mesenteric arteries in endothelium-dependent manner. TPEN-induced vasorelaxation was much greater in the arteries pre-constricted by noradrenaline than those by high K+. While TPEN-induced vasorelaxation was unaffected by inhibitors of NO and PGI2, it was significantly inhibited by the selective inhibitors of IKCa and SKCa channels but was potentiated by their activator. Moreover, TPEN-induced vasorelaxation was attenuated by selective inhibitors of NCX, NKA, SOCE, STIM translocation and Orai transportation. Finally, TPEN-induced vasorelaxation via SOCE/EDH was impaired in colitic mice but remained intact in IBS mice. Interestingly, TPEN could rescue vagus neurotransmitter ACh-induced vasorelaxation that was impaired in IBS mice. Therefore, since TPEN-induced SOCE/EDH-mediated vasorelaxation of mesenteric arteries is well-preserved to be able to rescue ACh-induced vasorelaxation impaired in IBS, TPEN has therapeutic potentials for IBS.

Summary: Positive autoregulation (PAR), one type of network motifs, provides a high phenotypic heterogeneity for cells to better adapt to their microenvironments. Typical mechanosensitive proteins can also form PAR, e.g., integrin mediated PAR, but the role of such mechanical PAR in physiological development and pathological process remains elusive. In this study, we found that transforming growth factor β1 (TGF-β1) and integrin levels decrease with tissue softening after the development of paradentium in vivo in rat model of periodontitis (an inflammatory disease with bone defect). Interestingly, TGF-β1 could induce the formation of mechanical PAR involving the integrin-FAK-YAP axis in mesenchymal stem cells (MSCs) by both in vitro experiments and in silico computational model. The computational model predicted a mechanical PAR involving the bimodal distribution of focus adhesions, which enables cells to accurately perceive extracellular mechanical cues. Thus, our analysis of TGF-β1 mediated mechanosensing mechanism on MSCs may help to better understand the molecular process underlying bone regeneration.

Microplastics (MPs), recognized as an emerging global environmental concern, have been extensively detected worldwide, with specific attention directed towards the Yangtze River Estuary (YRE) and East China Sea (ECS) regions. Despite their critical research significance, there remains a knowledge gap concerning the distribution of MPs in the benthic layer within this area, particularly regarding interactions governing their occurrence. Here we illuminate the distribution of MPs within the benthic layer and unravel the intricate interplay between bottom water and sediment in the YRE and ECS. We find that MPs are notably more abundant in bottom water, ranging from 8 to 175 times higher than in surface water. These MPs predominantly consist of polyester fibers, exhibit a size range between 0.5 and 5.0 mm, and display distinct coloration. Co-occurrence network analysis and Principal Coordinate Analysis confirm a robust correlation between MPs in bottom water and sediment, signifying the pivotal role of bottom water in mediating the distribution and transportation of MPs within the benthic layer. Furthermore, a positive correlation between MPs in sediment and bottom water turbidity underscores the impact of surface sediment resuspension and upwelling on MPs distribution. This study clarifies the intricate interactions within the benthic layer and highlights the crucial role of bottom water as a mediator in the vertical distribution of MPs, advancing our understanding of the “source-to-sink” transport processes governing MPs within water-sediment systems.

Magnetic metals have attracted extensive attention in microwave absorption for comprehensive merits of high magnetic loss and suitability for mass productions. Nevertheless, the applications have been limited by shortcomings of high conductivity and inadequate impedance matching. In this study, we have successfully synthesized a biphasic magnetic Sm2O3@Co composite through a facile one-step precipitation method. The resultant features a well-designed structure with spherical cores and flaky shells, exhibiting highly-tunable absorption performance over 2–18 GHz. Particularly, under 20 wt% filling ratio with paraffin, the Sm2O3@Co composite achieved minimum reflection loss (RLmin) of −30.1 dB and effective absorbing bandwidth (EAB) of 6.22 GHz at only 1.7 mm. The dielectric and magnetic losses could be easily modulated by simply changing the induced Co ratios. Through a systematic study, the microwave absorption mechanisms of the Sm2O3@Co magnetic composite have been comprehensively elucidated. This research offers a fresh perspective on developing highly efficient electromagnetic wave absorbents based on traditional magnetic metals.

Patients with advanced hepatocellular carcinoma (HCC) are not sensitive to sorafenib (SOR), therefore, combination therapy is required. In this study, an improved thin-film dispersion and post-insertion anchoring technique was utilized to construct a dual-targeted co-delivery SOR and docetaxel (DTX) liposome drug delivery system, folate/chondroitin sulfate with SOR/DTX-modified liposomes (FA/CS@SDLP), to jointly enhance the anti-recurrence and metastasis of HCC. FA/CS@SDLP can establish the gradual release of the two drugs because of successful lysosomal escape in the condensed hyaluronidase environment. The results indicated that modification with folate (FA) and chondroitin sulfate (CS) significantly enhanced the cellular uptake of FA/CS@SDLP and the internalization of SOR/DTX in HepG2 cells through FA and CD44 receptor-mediated endocytosis. Compared to free drugs or the mono-targeted liposomal system (FA@SDLP), FA/CS@SDLP presented higher potency against HepG2 cells regarding pro-apoptosis, anti-proliferation, and anti-metastasis (migration and invasion). Moreover, a more satisfactory antitumor efficacy was observed for FA/CS@SDLP in the pulmonary metastasis of HCC in a mouse model. In summary, dual-targeted co-delivery of liposomes can synergistically treat HCC recurrence and metastasis, providing a new approach for the clinically accurate treatment of HCC.

Cynoglossus semilaevis (half-smooth tongue sole) is an important aquaculture species that has a ZW/ZZ sex-determination system (females, ZW; males, ZZ). Some genotypic females may develop phenotypic males, and are known as pseudomales (ZW). These pseudomales can develop mature testes and produce sperm. However, previous studies have shown that pseudomales cannot produce sperms bearing W chromosome (W sperm). In this study, transcriptomic, proteomic, and ubiquitinated proteomic analyses were employed simultaneously to investigate the differences in gonads between pseudomales and normal males. Functional enrichment analysis of the differentially expressed genes/proteins showed that non-coding RNA metabolic processes and RNA degradation were more active in the gonads of pseudomales, and they showed higher levels of ubiquitination compared to males. In addition, most genes/proteins related to sperm morphogenesis expressed downwards in pseudomales. These genes included genes located exclusively on the Z chromosome, such as sperm flagellar protein 2 (spef2), doublesex and mab-3 related transcription factor 1 (dmrt1), doublesex and mab-3 related transcription factor 3 (dmrt3), and cilia- and flagella-associated protein 157 (cfap157). These findings characterized the transcriptional and proteomic profiles of the gonads of pseudomales and males, and suggested that W sperm were absent in pseudomales, possibly due to the disruption of sperm morphogenesis, resulting from W chromosome lacking spermatogenesis-related genes.

Out-of-hospital cardiac arrest is a major public health problem worldwide due to its high burden and poor outcomes. Despite progress in treatment, patient outcomes remain unsatisfactory, particularly in low-resource settings. The establishment of a registry is the first step towards gaining a comprehensive understanding of prevailing local conditions and identifying potential opportunities for improving patient survival. Here, we provide a narrative review of the BASeline Investigation of Out-of-hospital Cardiac Arrest (BASIC-OHCA), the first national OHCA registry in China, to introduce its development history, current state, challenges and future directions. We aim to enhance cross-cultural understanding by providing insights from China, while also serving as a reference for the implementation of large-scale registries in low-resource settings.

Many studies have examined the influence of digital technologies, such as robots and artificial intelligence, on enterprise labor, but few have investigated the underlying mechanisms and impact paths of digital empowerment on labor employment. Therefore, this study uses data on manufacturing enterprises listed on China's Shanghai and Shenzhen A-share markets from 2011 to 2020, and applies a panel fixed effect model to test the relationship between digital empowerment and labor employment, and the mechanisms underlying this relationship. We find that digital empowerment increases labor employment. However, the effects are heterogeneous: firms with better corporate governance, more competitive industry, and less favorable regional business environments are more motivated to optimize the structure of their labor resources. Through robustness test and mediation effect model test, we find that digital empowerment can improve enterprise human capital by increasing economies scale and managerial efficiency, especially the employment of R&D and innovation personnel and management personnel; it can also affect the amount of human capital by improving total factor productivity.

Summary: It is imperative to explore biomarkers that are both precise and readily accessible in the comprehensive management of breast cancer. A multicenter cohort, including 512 breast cancer patients and 198 nonneoplastic individuals, was recruited to detect the level of tumor-derived extracellular vesicles using our method based on dual DNA tetrahedral nanostructures. The level of tumor-derived extracellular vesicles was significantly higher in newly diagnosed breast cancer patients than in nonneoplastic individuals at a cutoff value of 3.58 U/μL. For postoperative metastasis monitoring, the level of tumor-derived extracellular vesicles was significantly higher in breast cancer patients with metastasis than in those without metastasis at a cutoff value of 3.91 U/μL. Its efficacy of diagnosis and metastasis monitoring was superior to traditional tumor markers. Elevated level of tumor-derived extracellular vesicles served as a predictive biomarker for diagnosis and metastasis monitoring in breast cancer patients.

Background: Some COVID-19 patients deteriorate to severe cases with relatively higher case-fatality rates, which increases the medical burden. This necessitates identification of patients at risk of severe disease. Early assessment plays a crucial role in identifying patients at risk of severe disease. This study is to assess the effectiveness of SUPER score as a predictor of severe COVID-19 cases. Methods: We consecutively enrolled COVID-19 patients admitted to a comprehensive medical center in Wuhan, China, and recorded clinical characteristics and laboratory indexes. The SUPER score was calculated using parameters including oxygen saturation, urine volume, pulse, emotional state, and respiratory rate. In addition, the area under the receiver operating characteristic curve (AUC), specificity, and sensitivity of the SUPER score for the diagnosis of severe COVID-19 were calculated and compared with the National Early Warning Score 2 (NEWS2). Results: The SUPER score at admission, with a threshold of 4, exhibited good predictive performance for early identification of severe COVID-19 cases, yielding an AUC of 0.985 (95% confidence interval [CI] 0.897–1.000), sensitivity of 1.00 (95% CI 0.715–1.000), and specificity of 0.92 (95% CI 0.775–0.982), similar to NEWS2 (AUC 0.984; 95% CI 0.895–1.000, sensitivity 0.91; 95% CI 0.587–0.998, specificity 0.97; 95% CI 0.858–0.999). The asperity wear of rock joints significantly affects their shear behaviour. This study discusses the wear damage of the asperities on the joint surface, highlighting the roughness degradation characteristics during the shear process. The direct shear experiment of artificial specimens containing rock joints was conducted under different normal stresses based on three-dimensional scanning technology. These experimental results showed the contribution of joint wear to roughness degeneration, such as the height, zone, and volume of asperity degeneration. The wear coefficient of the rock joint was obtained based on the volume wear of asperities in the laboratory experiment. The functional relationship between the friction coefficient and wear coefficient is subsequently determined. To quantitatively analyse the wear damage of a joint surface, a calculation method for determining the wear depth of the rock joint after shearing was proposed based on wear theory. The relationship between the ultimate dilation and wear depth was analysed. A coefficient m, which can describe the damage degree of the joint surface, and a prediction method of joint surface roughness after shearing are established. Good agreement between analytical predictions and measured values demonstrates the capability of the developed model. Lastly, the sensitivity factors on the wear depth are explored.

The emission trading system (ETS) is one of the important policy instruments that help to achieve the “double carbon” goals, however, how the ETS affects the carbon emissions for an urban agglomeration remains uncertain. This study aims to explore the ETS’s impacts to carbon emission intensity (CEI) in Urban Agglomeration in the Middle Reaches of the Yangtze River (UAMRYR) of China during 2006–2018. By using the Difference-in-Differences (DID) model, the positive effects of ETS to CEI reduction were confirmed, and the robustness of findings were proved via the parallel trend test, placebo test, and propensity score matching DID (PSM-DID) method. This study also recognized two mediation impacting paths, which were economic intensity improvement and green technology innovation, and provided the evidence that the ETS could further reduce the CEI through improving the economic intensity as well as the green technology innovation. Furthermore, the study investigated the heterogeneous impacts from ETS by dividing the cities within the agglomeration into different orientations. The ETS performed better in the edge cities, and the same phenomenon occurred when considering the mediation effect from economic intensity. Policy recommendations about ETS were also provided.

The building energy performance gap (EPG) seriously restricts the improvement of building energy efficiency. Currently, although many studies on EPG, it is not yet fully understood and addressed. To fill this gap, this paper conducted an extensive review of EPG research. Firstly, the magnitude of EPG was summarized from many case studies, and the results showed that it varied greatly among building types, with gap ratios ranging from 0.5 to 4 for educational/research buildings, concentrated between 0.5 and 2.5 for residential buildings, and between 0 and 1 for office building. Then, fifteen direct causes and seven in-depth drivers of EPG were analyzed from simulation and lifecycle perspectives, and the linkages between them were established. Furthermore, solutions for EPG were summarized, including some state-of-the-art technical and “soft” measures, and their correspondence with the underlying causes. Finally, eight future research recommendations were proposed based on the limitations of existing strategies.

When working alongside proactive colleagues, do you elevate yourself through benign envy or resort to malicious envy? To address this intriguing question, we constructed a model based on social comparison theory to measure the double-edged sword effects of proactive personality on employee outcomes. We hypothesized that proactive employees would induce two distinct tendencies in their peers—workplace ostracism and employee creativity—due to peer envy. The study analyzed 389 valid responses from full-time employees in Chinese organizations using structural equation modeling. Results indicate that proactive personality positively influences benign envy among peers, which in turn positively affects employee creativity. Moreover, benign envy mediates the relationship between proactive personality and employee creativity. On the other hand, proactive personality positively influences malicious envy among peers, which in turn positively affects workplace ostracism. Additionally, malicious envy mediates the relationship between proactive personality and workplace ostracism. This study intertwines personality, emotions, and workplace outcomes, thereby advancing the existing literature on social comparison theory. Additionally, it furnishes valuable insights for organizational human resource management, particularly in the realms of employee recruitment and workplace relationship management.

Sargentodoxa cuneata is a traditional Chinese herb commonly used for the treatment of ulcerative colitis (UC) in clinical practice. However, the effects of total flavonoids of Sargentodoxa cuneata (TFSc) remain unclear. In this study, we explored the effects and mechanisms of action of TFSc in mice with dextran sulfate sodium-induced UC. The TFSc components were identified using liquid chromatography-tandem mass spectrometry. Mice with UC were orally administered TFSc to evaluate its effects on colon and liver damage. Changes in gut microbiota were analyzed using 16S rRNA gene sequencing. Moreover, the levels of short-chain fatty acids (SCFAs) and few organic acids in the intestinal contents were measured. Furthermore, the correlation amongst gut flora, SCFAs, and organic acids was studied. A total of 304 compounds were identified. TFSc alleviated the symptoms of UC in mice, inhibited weight loss, reduced the disease activity index, and improved colon shortening along with cardiac and hepatic indices. Furthermore, TFSc mitigated pathological changes in the colon and liver, and reduced the TNF-α levels in colon tissues of mice with UC. TFSc also modulated intestinal flora disorders, increased the levels of four SCFAs (isocaproic acid, 3-hydroxyisovaleric acid, isobutyric acid, and ethylmethylacetic acid), and decreased the levels of two organic acids (malonic acid and succinic acid). The differential regulation of SCFAs and organic acids by TFSc was strongly associated with specific gut flora. TFSc showed potential for improving colon and liver injury in mice with UC by regulating the intestinal flora and associated metabolites (SCFAs and organic acids). These findings provide new evidence for developing TFSc as a drug for treating UC and UC-related liver injuries.

One patient with rifampin-resistant tuberculosis underwent emergency left pneumonectomy and thoracic gauze packing for hemoptysis due to recurrent hemoptysis after transcatheter arterial embolization. Vital signs were maintained by mechanical ventilation and medication. Tracheotomy and anti-tuberculosis treatment were performed. After half a year of follow-up, the patient's condition was stable.

With the frequent occurrence of worldwide extreme climate events, human-induced ecosystem degradation has seriously threatened the realization of the Sustainable Development Goals (SDGs), especially in arid ecologically fragile areas. Macro-scale ecological quality (EQ) monitoring and exploration of its driving mechanisms have become research hotspots. However, this research field still lacks a method framework with strong comparability, adaptability and transferability, which significantly restricts the applicability of research results. In this study, a method framework for exploring spatial and temporal changes in EQ and their driving mechanisms based on text summarization and information extraction is constructed. Taking Central Asia as a typical case, this study outlines its EQ driving paths, explores the influencing mechanisms of representative drivers, and verifies the effectiveness of this method framework based on the comparison of the spatial and temporal evolution of EQ at multiple scales. The results indicate that the overall EQ in Central Asia exhibited a slight inverted U-shaped trend, with its driving paths falling into five categories: urban expansion, agricultural development, resource extraction, climate change, and ecological protection. The fragmentation of areas with high EQ is the main landscape characteristic in Central Asia. Furthermore, land use intensity and agricultural water use efficiency are significant factors in Central Asia's EQ evolution. Over time, the interaction between anthropogenic and natural factors has played an essential role in EQ evolution in Central Asia, with interactions between altitude, climate aridity, agricultural water use efficiency, and land use intensity gradually intensifying. This study has an implication for the construction of method framework for EQ-related study in ecologically fragile areas at the macro scale.

This paper conducted a comprehensive investigation into the role of polymers in cement-based materials. First, the modification mechanism of polymers on cement-based materials based on physical and chemical effects are analyzed. The polymers are adsorbed on the surface of cement particles and cement hydration products through electrostatic attraction or hydrogen bonding, and filled in the pores or interfacial areas, and then transformed into a three-dimensional interpenetrating network structure, so as to densify the matrix materials. Then, the effects of polymers on the cement hydration reaction were introduced. It is mainly attributed to the adsorption behavior between polymer particles, and polymer films with cement causes them to cover the surfaces of cement particles and hydration products, which to some extent restricts the effective diffusion of various ions, such as Ca2+, AlO2−, SiO42−, SO42−, etc., as well as water in the system. And the complexation of functional groups in the polymer with Ca2+ hinders the nucleation and precipitation of calcium-containing hydration products, which in turn delays the hydration reaction process. In terms of strength, as it can be due to the agglomeration of polymer chains, increased porosity arised from generated CO2 gas and entrained air during preparation, the improvement effect of the polymers on cement-based material's compressive strength is not apparent or it shows a decreasing trend. As the role of “microfibers” and “bridge”, many filamentous or network structures composed of flexible polymer membranes dramatically improve the flexural strength and toughness, and also enhance the bonding property of the matrix material. It is expected that the polymers containing designed specific structures according to target property will be applied in cement-based materials, and the universal guidelines and regulations will be provide for expanded application of polymer modified cement-based materials (PMCBMs) in the construction industry.

Nowadays, the development of magnetic energy-conversion devices stimulates the exploration of non-rare-earth materials and their polymer composites with large strain output under magnetic field. In this paper, we will provide a basic strategy to produce [100]-oriented Mn0.988Ni0.012CoSi/epoxy composites with large and reversible magnetic-field-induced strain at room temperature. The mixture of Mn0.988Ni0.012CoSi powders and epoxy is cured under a rotating magnetic field of 1 T, and [100] preferred orientation is obtained along the rotation axis. The formation of [100] preferred orientation is related to the ferromagnetic structure of Mn0.988Ni0.012CoSi and greatly enhances the magnetic-field-induced strain. For the composite with the weight fraction of epoxy = 15 wt. %, the absolute value of magnetic-field-induced strain under a magnetic field of 1 T reaches 1506 ppm, comparable to that of Terfenol-D single crystal and Terfenol-D-based magnetostrictive polymer composite. Since the magnetic-field-induced strain is originated from second-order metamagnetic transition, the observed magnetic-field-induced strain is reversible and independent of the direction of the magnetic field. Besides, these composites show good processing ability. Our results suggest that the [100]-oriented Mn0.988Ni0.012CoSi/epoxy composites have application potential in energy-conversion devices.

Background: T cells and natural killer (NK) cells are essential components of the immune system and are regulated by coinhibitory and costimulatory molecules in which the B7 family and CD28 family play significant roles. Previous immune checkpoint studies on B7/CD28 family members, such as PD-1, have led to remarkable success in cancer immunotherapy. However, there is still a need to find new immune checkpoint molecules. Recent studies have demonstrated that HHLA2 exerts inhibitory and stimulatory functions on the immune system by binding to different receptors on different sites. However, the pathways between HHLA2 and its two receptors on T cells and NK cells remain controversial. Aim of Review: Here, we reviewed recent studies about HHLA2 ligand interactions with KIR3DL3 and TMIGD2. We focused on elucidating the pathways between KIR3DL3/TMIGD2 and HHLA2 as well as their function in tumour progression. We also addressed the relationship between HHLA2 expression and the clinical prognosis of cancer patients. Key Scientific Concepts of Review: KIR3DL3/TMIGD2-HHLA2 may represent novel pathways within the tumour microenvironment and serve as crucial immune checkpoints for developing novel therapeutic drugs against human cancer.

The aim of this study was to establish a new method for the determination of homoisoflavanones (Ⅲ, Ⅳ, V) in polygonatum odoratum(POD) by combination of thin layer chromatography (TLC) and chemical derivative resonance Raman spectroscopy (RRS). The twice chromatography method of TLC was used to improve the specificity of the component to be tested; the method of the relative Rf was used to reduce the use of the reference substance of the component to be tested; the chemical derivatization method was used to improve the signal intensity of Raman spectrum for the component to be tested in POD, so as to obtain a trace amount fingerprint structure information of the measured component. The method exhibits robust specificity, high sensitivity, and reliable stability, there by offering a novel reference approach for the identification and evaluation of homoisoflavanones (Ⅲ, Ⅳ, V) in POD.

High-entropy alloys (HEAs) have gained significant attentions in recent years, due to their unique properties derived from the combination of multiple elements in equimolar or near-equimolar ratios. The mechanical properties of HEAs are influenced by microstructural characteristics. In this study, MnCrFeCoNi HEA ribbons were produced using a technique called melt spinning, for which the wheel speed was adjusted to control the undercooling levels. The rapid solidification process under undercooling condition resulted in refined grain sizes to micrometers in the ribbons. One notable feature was the appearance of twin boundaries, which especially accounted for approximately 7.36 % of the microstructure for the ribbons produced at a wheel speed of 10 m/s. For the ribbons with thickness of micrometer scale, the mechanical properties (ultimate tensile strength up to 2.5 GPa and hardness up to 300 MPa) were analyzed by microstructure (grain boundaries and homogeneity) and exterior factors (e.g. thickness). Overall, this study provides a new approach for tailoring the microstructures and mechanical properties of HEAs via melt spinning technique. The HEA ribbons present a novel form that could potentially broaden the scope of applications for these materials.

The Xinjiang ground-jay (Podoces biddulphi), which is endemic to China, has a limited distribution and a small population in the Taklimakan Desert. In this study, we described the breeding biology and nestling growth patterns of this understudied species. Eleven nests were monitored over three consecutive breeding seasons (2017–2019). The Xinjiang ground-jay breeding season extends from March to May with a clutch size of two or three. The incubation period was 18 d, and females spent most of their time incubating their eggs (63 %), followed by collecting leaves (25 %), resting (10 %), turning the eggs (1 %), grooming (0.6 %), and being vigilant (0.4 %). Both parents fed the nestlings with similar feeding frequencies and durations. The growth curve of the nestlings was S-shaped, and the parameters related to nestling mortality, such as tarsus length, showed faster growth than others. Over the three breeding seasons, 32 eggs were laid across 11 nests. Fifteen (47 %) eggs hatched, and 12 hatchlings across six nests fledged (37.5 %). Our study provides detailed information on the breeding biology and nestling growth of the Xinjiang ground-jay. Given its low reproductive success rate, small population size, and limited distribution, this bird species, which is endemic to China, requires further research, and its conservation should be prioritized.

As an essential tool to evaluate land use sustainability, land use functions (LUFs) have become a hot topic in the field of land use science for promoting sustainable regional development. Previous studies focused more on the interaction analysis among LUFs and the zoning management of LUFs, ignoring the importance of geospatial characteristics of LUFs for regional development patterns. In this study, a series of geospatial analysis methods were adopted to identify the abrupt changes in LUFs, and evaluate the geospatial characteristics of LUFs in the Yangtze River Economic Belt (YREB), including agglomeration, coordination, and inequality. Finally, we combined the evaluation results to distinguish the development patterns of provinces and urban agglomerations (UAs). The findings show that the abrupt changes in LUFs occurred in 2002 and 2013, and the changes of LUFs experienced three stages: sharp decline, rapid rise, and steady development from 1990 to 2018. There was a significant agglomeration of LUFs in low value areas, and gradually to high value areas over time. The coordination of LUFs exhibited moderately coordinated, which was higher in the east than in the west, though the gap has been narrowing. The major contributor to LUF inequality was from within the province rather than among them, and so did UAs. All provinces and UAs were classified into radiation-driven, multi-core driven, stable, and low-level ones. Our research provides a reference for large-scale regional development and land use management.

Summary: Fatty acid metabolism plays a critical role in both tumorigenesis and cancer radiotherapy. However, the regulatory mechanism of fatty acid metabolism has not been fully elucidated. NSD2, a histone methyltransferase that catalyzes di-methylation of histone H3 at lysine 36, has been shown to play an essential role in tumorigenesis and cancer progression. Here, we show that NSD2 promotes fatty acid oxidation (FAO) by methylating AROS (active regulator of SIRT1) at lysine 27, facilitating the physical interaction between AROS and SIRT1. The mutation of lysine 27 to arginine weakens the interaction between AROS and SIRT1 and impairs AROS-SIRT1-mediated FAO. Additionally, we examine the effect of NSD2 inhibition on radiotherapy efficacy and find an enhanced effectiveness of radiotherapy. Together, our findings identify a NSD2-dependent methylation regulation pattern of the AROS-SIRT1 axis, suggesting that NSD2 inhibition may be a potential adjunct for tumor radiotherapy.

The practical applications of hydrogel-based sensors are limited by their low mechanical strength, insufficient sensitivity and machining difficulties caused by electrically conductive fillers. It is of great practical importance for facile preparation of hydrogel sensor combined with excellent mechanical properties and conductive sensitivity. In this work, a series of ion-conductive hybrid hydrogels based on in-situ polymerized polyacrylamide with double and triple network structures have been fabricated via both chemical and physical cross-linking. The triple network structured hydrogels achieve the tensile strength of 2.105 MPa and compression strength of 57.56 MPa, which are much higher than those of hydrogels with double network due to the established energy dissipation gradient. The reversible non-covalent cross-linking in the network also leads a relatively fast recovery in tensile cycle tests and double compression tests, enabling the excellent stability of strain sensing in the durability test. The hydrogels show the tensile resistance sensitivity factor of 0.9352 at 0–120% strain and a high pressure sensitivity factor of 5.134 kPa−1 with the pressure ranged from 0 to 6 kPa. The hydrogels are also assembled as flexible sensors for human motion monitoring, and the signals of motions can be distinguished by different resistance variation values and certain curve shape, showing the potential of applications on flexible device and human-machine interface.

Summary: Background: Mediastinal neoplasms are typical thoracic diseases with increasing incidence in the general global population and can lead to poor prognosis. In clinical practice, the mediastinum's complex anatomic structures and intertype confusion among different mediastinal neoplasm pathologies severely hinder accurate diagnosis. To solve these difficulties, we organised a multicentre national collaboration on the basis of privacy-secured federated learning and developed CAIMEN, an efficient chest CT-based artificial intelligence (AI) mediastinal neoplasm diagnosis system. Methods: In this multicentre cohort study, 7825 mediastinal neoplasm cases and 796 normal controls were collected from 24 centres in China to develop CAIMEN. We further enhanced CAIMEN with several novel algorithms in a multiview, knowledge-transferred, multilevel decision-making pattern. CAIMEN was tested by internal (929 cases at 15 centres), external (1216 cases at five centres and a real-world cohort of 11 162 cases), and human–AI (60 positive cases from four centres and radiologists from 15 institutions) test sets to evaluate its detection, segmentation, and classification performance. Findings: In the external test experiments, the area under the receiver operating characteristic curve for detecting mediastinal neoplasms of CAIMEN was 0·973 (95% CI 0·969–0·977). In the real-world cohort, CAIMEN detected 13 false-negative cases confirmed by radiologists. The dice score for segmenting mediastinal neoplasms of CAIMEN was 0·765 (0·738–0·792). The mediastinal neoplasm classification top-1 and top-3 accuracy of CAIMEN were 0·523 (0·497–0·554) and 0·799 (0·778–0·822), respectively. In the human–AI test experiments, CAIMEN outperformed clinicians with top-1 and top-3 accuracy of 0·500 (0·383–0·633) and 0·800 (0·700–0·900), respectively. Meanwhile, with assistance from the computer aided diagnosis software based on CAIMEN, the 46 clinicians improved their average top-1 accuracy by 19·1% (0·345–0·411) and top-3 accuracy by 13·0% (0·545–0·616). Interpretation: For mediastinal neoplasms, CAIMEN can produce high diagnostic accuracy and assist the diagnosis of human experts, showing its potential for clinical practice. The management and treatment of disease are achieved via the use of pharmacologically active substances or drugs. Drugs do not, however, have an intrinsic ability to be effective; rather, how well they work depends on how they are administered or supplied. Treatment of a variety of biological illnesses, such as autoimmune disorders, cancer, and bacterial infections, requires effective drug delivery. Drug absorption, distribution, metabolism, duration of therapeutic impact, pharmacokinetics, excretion, and toxicity can all be impacted by drug administration. Improved chemistry and materials are required for the delivery of therapeutic concentration of novel treatments to the specified targets within the body, as well as for the necessary duration of time. This requirement is accompanied by the development of new therapeutics. Formulating a medication as a DDS is a promising strategy for directly addressing numerous typical barriers to adherence, such as frequent dosage, such as frequent dosage, side effects, and a delayed beginning of the action. In the current review, we give a compendium of drug delivery and controlled release and subsequently highlight some of the newest developments in the realm, with a particular emphasis on cutting-edge methods for targeted therapy. In each instance, we outline the obstacles to efficient drug administration as well as the chemical and material developments that are allowing the sector to overcome these obstacles and have a positive clinical impact.

Both diabetes and Corona Virus Disease 2019 (COVID-19) are seriously harmful to human health, and they are closely related. It is of great significance to find drugs that can simultaneously treat diabetes and COVID-19. Based on the theory of traditional Chinese medicine for treating COVID-19, this study first sorted out the compounds of Guizhou Miao medicine with “return to the lung channel” and “clear heat and detoxify” effects in China. The active components against COVID-19 were screened by molecular docking with SARS-CoV-2 PLpro and angiotensin-converting enzyme II as targets. Furthermore, the common target dipeptidyl peptidase 4 (DPP4) of diabetes and COVID-19 was used as a screening protein, and molecular docking was used to obtain potential components for the treatment of diabetes and COVID-19. Finally, the mechanism of potential ingredients in the treatment of diabetes and COVID-19 was explored with bioinformatics. More than 80 kinds of Miao medicine were obtained, and 584 compounds were obtained. Further, 110 compounds against COVID-19 were screened, and top 6 potential ingredients for the treatment of diabetes and COVID-19 were screened, including 3-O-β-D-Xylopyranosyl-(1-6)-β-D-glucopyranosyl-(1-6)-β-D-glucopyranosyl oleanolic acid 28-O-β-D-glucopyranosyl ester, Glycyrrhizic acid, Sequoiaflavone, 2-O-Caffeoyl maslinic acid, Pholidotin, and Ambewelamide A. Bioinformatics analysis found that their mechanism of action in treating diabetes and COVID-19 may be related to regulating the expression of DPP4, angiotensin II type 1 receptor, vitamin D receptor, plasminogen, chemokine C–C-motif receptor 6, and interleukin 2. We believe that Guizhou Miao medicine is rich in potential ingredients for the treatment of diabetes and COVID-19.

Purpose: To examine the relationship between the positive rate and types of necrosis in pathological examinations of tuberculosis granulomas with necrosis, to improve the detection rate of positive cases. Methods: Specimens from 381 patients were collected in Wuhan Pulmonary Hospital from Jan 2022 to Feb 2023. The samples were examined using various methods such as AFB smear microscopy, mycobacterial culture, PCR, SAT-TB, and X-pert MTB/RIF rapid molecular detection. Result: There were 3 types of necrosis. Including 270 cases of caseous necrosis, 30 cases of coagulation necrosis, and 76 cases of an abscess. Five cases were non-necrotizing granulomas.In the pathological specimen testing for tuberculosis, five detection techniques were used and their positive rates detected in descending order were X-pert, TBDNA, SAT-TB, tuberculosis culture, AFB. Cardiac fibrosis is a common pathophysiological remodeling process that occurs in a variety of cardiovascular diseases and greatly influences heart structure and function, progressively leading to the development of heart failure. However, to date, few effective therapies for cardiac fibrosis exist. Abnormal proliferation, differentiation, and migration of cardiac fibroblasts are responsible for the excessive deposition of extracellular matrix in the myocardium. Acetylation, a widespread and reversible protein post-translational modification, plays an important role in the development of cardiac fibrosis by adding acetyl groups to lysine residues. Many acetyltransferases and deacetylases regulate the dynamic alterations of acetylation in cardiac fibrosis, regulating a range of pathogenic conditions including oxidative stress, mitochondrial dysfunction, and energy metabolism disturbance. In this review, we demonstrate the critical roles that acetylation modifications caused by different types of pathological injury play in cardiac fibrosis. Furthermore, we propose therapeutic acetylation-targeting strategies for the prevention and treatment of patients with cardiac fibrosis.

With the development of offshore wind power development to deep and far sea, offshore flexible low-frequency AC (LFAC) transmission technology has attracted more and more attention from academia and industry in recent years. The control and protection system is the brain of flexible LFAC transmission, which is directly related to the performance, safety and benefit of LFAC transmission. In order to improve its operation reliability and limit the impact caused by any control link fault, in this paper, the control system is divided into system level control strategy, converter station level control strategy and valve level control strategy, which is from high level to low level according to the stratification principle. For the protection system, a specific zoning protection scheme is proposed. Finally, a two-terminal flexible LFAC transmission system is built in PSCAD/EMTDC. The simulation results prove the validity and correctness of the proposed control and protection strategy.

The hybrid DC system combined with line commutated converter (LCC) and hybrid modular multilevel converter (HY-MMC), which is named LCC-HY-MMC, has technical advantages and economy. Shutdown is a key problem that any DC system needs to face. In order to ensure that the DC system and the sub-module capacitors of HY-MMC can be discharged quickly during shutdown stage, a complete shutdown control strategy is proposed in this paper. From the characteristics of shutdown, it can be basically regarded as three stages: DC energy feedback stage, controllable dissipation stage of sub-module capacitor energy and uncontrollable dissipation stage of sub-module capacitor energy. Finally, based on the 200 MW/ 200 kV digital simulation model built in the electromagnetic transient simulation software PSCAD/EMTDC, the effectiveness of the proposed method is verified.

Introduction: Although capsaicin has long been used as food additive and medication worldwide, its actions on gastrointestinal tract as its most delivery pathway have not been well addressed. Objectives: In the present study, we aimed to study GI actions of capsaicin on mesenteric arterioles in normal and colitis mice and to elucidate the underlying mechanisms. Methods: Vasorelaxation of human submucosal arterioles and the mesenteric arterioles from wide-type (WT) mice, TRPV1−/− and TRPV4−/− (KO) mice were measured. The expression and function of TRPV channels in endothelial cells were examined by q-PCR, immunostaining, Ca2+ imaging and membrane potential measurements. Results: Capsaicin dose-dependently induced vasorelaxation of human submucosal arterioles and mouse mesenteric arterioles in vitro and in vivo through endothelium-dependent hyperpolarization (EDH), nitric oxide (NO), and prostacyclin (PGI2). Using TRPV1 and TRPV4 KO mice, we found that capsaicin-induced vasorelaxation was predominately through TRPV4/EDH, but marginally through TRPV1/NO/PGI2. Capsaicin induced hyperpolarization through activation of endothelial TRPV4 channels and intermediate-conductance of Ca2+-activated K+ channels to finally stimulate vasorelaxation. Importantly, capsaicin exerted anti-colitis action by rescuing the impaired ACh-induced vasorelaxation in WT colitis mice but not in TRPV4 KO colitis mice. Conclusions: Capsaicin increases intestinal mucosal blood perfusion to potentially prevent/treat colitis through a novel TRPV4/EDH-dependent vasorelaxation of submucosal arterioles in health and colitis. This study further supports our previous notion that TRPV4/EDH in mesenteric circulation plays a critical role in the pathogenesis of colitis.

Summary: DHX9 is a member of the DEAH (Asp-Glu-Ala-His) helicase family and regulates DNA replication and RNA processing. DHX9 dysfunction promotes tumorigenesis in several solid cancers. However, the role of DHX9 in MDS is still unknown. Here, we analyzed the expression of DHX9 and its clinical significance in 120 MDS patients and 42 non-MDS controls. Lentivirus-mediated DHX9-knockdown experiments were performed to investigate its biological function. We also performed cell functional assays, gene microarray, and pharmacological intervention to investigate the mechanistic involvement of DHX9. We found that overexpression of DHX9 is frequent in MDS and associated with poor survival and high risk of acute myeloid leukemia (AML) transformation. DHX9 is essential for the maintenance of malignant proliferation of leukemia cells, and DHX9 suppression increases cell apoptosis and causes hypersensitivity to chemotherapeutic agents. Besides, knockdown of DHX9 inactivates the PI3K-AKT and ATR-Chk1 signaling, promotes R-loop accumulation, and R-loop-mediated DNA damage.

Diabetes mellitus (DM) is a metabolic disease caused by multiple factors such as genetics, environment, and lifestyle. Bisphenol A (BPA), as one of the most common endocrine-disrupting chemicals (EDCs), has been strongly implicated in the development of type 2 diabetes mellitus (T2DM). BPA exposure is associated with target organ damage in DM and may exacerbate the progression of some chronic complications of DM. This paper reviews relevant epidemiological, in vivo, and in vitro studies to better understand BPA's potential risk associations and pathological mechanisms in several chronic diabetic complications.

Background: Effective antiplatelet therapy is critical for patients with ST-segment elevation myocardial infarction (STEMI) and receiving primary percutaneous coronary interventions (PPCI). Intracoronary (IC) and intravenous (IV) administration of tirofiban are commonly used during the procedure of PPCI. Methods: A comprehensive literature search of RCTs that comparing IC with IV tirofiban in STEMI patients undergoing PPCI was conducted, which were published as of May 7, 2022, in PubMed, Embase, Cochrane Library, Web of Science, Scopus and ClinicalTrials.gov. The primary efficacy endpoint was 30-day major adverse cardiovascular events (MACE) and the primary safety endpoint was in-hospital bleeding events. Results: This meta-analysis included 9 trials involving 1177 patients. Introduction: : Zuojin pills(ZP) have been shown to be effective in the treatment of gastric ulcer (GU); however, the efficacy of Zuojin gastric floating bio-adhesive pellets(ZFP) in GU treatment is yet to be evaluated. Methods: : The pharmacokinetics(PK) and protective effects of ZFP against acetic acid-induced GU was examined using rat models. Mass spectrometry(MS) analysis was performed using a TSQ LC-MS system. A total of 50male Sprague-Dawley(SD) rats were randomly assigned to five groups: normal, acetic acid-induced, positive control (rabeprazole, 2 mg/kg/day), ZP (500 mg/kg/day), and ZFP (500 mg/kg/day) groups, to elucidate the anti-ulcerogenic effect of ZFP. Results: : ZFP prolonged the residence time of the drug (MRT, 10.88 ± 0.93 h; T1/2, 42.83 ± 2.56 h) in rats and promote the effective absorption of the drug with obvious sustained-release properties. Compared with the control group, ZFP treatment for 7 d significantly increased the weight of the rats, significantly reduced ulcer index, increased gastric tissue Superoxide dismutase(SOD) activity, reduced gastric tissue Malondialdehyde(MDA) content, and increased serum IgA, IgM, IgG, and Interleukin-2(IL-2) levels. Additionally, histopathological analysis showed that ZFP treatment reduced inflammatory cell infiltration to mainly the mucosal layer, anda small amount of fibrous scar proliferative repair was observed in the mucosal defect area and the underlying inflammatory granulation tissue. Conclusion: : These results indicated that ZFP prolonged the residence time of the drug in rats with sustained-release properties and had an obvious protective effect against acetic acid-induced GU in rats.

Velocity-selective inversion (VSI) based velocity-selective arterial spin labeling (VSASL) has been developed to measure cerebral blood flow (CBF) with low susceptibility to the prolonged arterial transit time and high sensitivity to brain perfusion signal. The purpose of this magnetic resonance imaging study is to evaluate the test-retest reliability of a VSI-prepared 3D VSASL protocol with whole-brain coverage to detect baseline CBF variations among cognitively normal participants in different brain regions. Coefficients of variation (CoV) of both absolute and relative CBF across scans or sessions, subjects, and gray matter regions were calculated, and corresponding intraclass correlation coefficients (ICC) were computed. The higher between-subject CoV of absolute CBF (13.4 ± 2.0%) over within-subject CoV (within-session: 3.8 ± 1.1%; between-session: 4.9 ± 0.9%) yielded moderate to excellent ICC (within-session: 0.88±0.08; between-session: 0.77±0.14) to detect normal variations of individual CBF. The higher between-region CoV of relative CBF (11.4 ± 3.0%) over within-region CoV (within-session: 2.3 ± 0.9%; between-session: 3.3 ± 1.0%) yielded excellent ICC (within-session: 0.92±0.06; between-session: 0.85±0.12) to detect normal variations of regional CBF. Age, blood pressure, end-tidal CO2, and hematocrit partially explained the variability of CBF across subjects. Together these results show excellent test-retest reliability of VSASL to detect both between-subject and between-region variations supporting its clinical utility.

Objectives: The study aimed to explore the efficacy and safety of linezolid-based chemotherapeutic regimens for patients with postoperative multidrug-resistant spinal tuberculosis. Methods: The randomized controlled study included 50 Mycobacterium tuberculosis culture or pathological-confirmed multidrug resistant tuberculosis patients who received spinal surgery from January 2018 to February 2020. Twenty-five patients were assigned to the control group and the study group, respectively. Random number method was used for patient allocation and they were treated with levofloxacin, pyrazinamide, thioisonicotinamide enteric-coated tablet, amikacin sulfate injection, and sodium p-amino salicylate injection, accompanied by linezolid or not.
