The Chinese Association of Neurorestoratology put forward these standards for training operators, standardized use and management of materials and equipment, standardized isolation and culture for neural progenitor/precursor cells, and the standardized management in preservation, transport, and related safe operation procedures of the neural progenitors. These cultures and quality control standards also include the Good Manufacturing Practice environment, routine maintenance as well as the monitoring and reporting of the clinical-grade neural progenitor cells. The aim of these standards is to improve the therapeutic efficacy and minimize the possible side effects from lake of quality control. Currently, neurorestorative strategies, including stem cell therapy products or mature/functionally differentiated cell-derived cell therapy products, can restore patients with chronic complete SCI to some degree of neurological functions. The stem cells for neurorestoration include neural stem cells, mesenchymal stem cells, embryonic stem cells, induced pluripotent stem cells, etc. A better understanding of the merits, demerits and precise function of different stem cells in the treatment of SCI may aid in the development of neurorestorative strategies. However, the efficacy, safety and ethical concerns of stem cell-based therapy continue to be challenged. Nonetheless, stem cell-based therapies hold promise of widespread applications, particularly in areas of SCI, and have the potential to be novel therapeutics, which contributes to the repair of SCI. This review mainly focused on recent advances regarding the stem cell-based therapies in the treatment of SCI and discussed future perspectives in this field. Background & aims: With the rising prevalence of non-alcoholic fatty liver disease (NAFLD) as a significant etiology for hepatocellular carcinoma (HCC), lean NAFLD-HCC has emerged as a specific distinct subtype. This study sought to investigate long-term outcomes following curative-intent hepatectomy for early-stage NAFLD-HCC among lean patients compared with overweight and obese individuals. Methods: A multicenter retrospective analysis was used to assess early-stage NAFLD-HCC patients undergoing curative-intent hepatectomy between 2009 and 2022. Fused silica is widely used in high-power laser systems, astronomy and military fields due to its excellent optical and physical properties. However, Subsurface defects(SSDs), such as microcracks, scratches, plastic deformation, and pits are often formed during mechanical processing, which can seriously reduce the optical performance and durability of fused silica. This paper first discusses the formation mechanism of SSDs in fused silica due to mechanical stress, thermal effects, etc. during mechanical processing such as cutting, grinding, and polishing. Then, the commonly used destructive and non-destructive detection methods are reviewed, and the principles and latest progress of each technique are discussed. The potential of deep learning algorithms for SSD detection is also examined. Finally, future directions for research and development are proposed to provide a valuable reference for researchers in the field.

The presented work concerns with some novel solutions of the (2+1)-dimensional Boussinesq equation (BE), which acts as an important model for shallow water wave. Some resonant soliton solutions such as the X-shape soliton (XSS) and Y-shape soliton (YSS) solutions are developed via imposing the resonant conditions on the N-soliton solutions (N-SSs) that developed by the Hirota bilinear approach(HBA). Based on the XSS and YSS solutions, the novel hybrid interactions including the interaction between the 1-soliton and the YSS, the interaction between two YSS solutions are extracted. In addition, the complex N-SSs are also explored and discussed. Finally, the travelling wave solutions including the bright solitary and kinky solitary wave solutions are studied by employing the Bernoulli sub-equation function method (BSEFM). The graphs of the attained solutions are drawn to show the physical properties. The findings of this study are expected to help us apprehend the dynamics of the (2+1)-dimensional BE better.

As the global population continues to grow, so does the demand for food. However, cold stress due to global climate change greatly threatens the safe production of rice. Piriformospora indica plays a pivotal role as a functional microbiota in improving cold tolerance in rice. However, the addition of P. indica alone has a limited effect. In this study, an attempt was made to select plant growth-promoting bacteria (Agrobacterium rhizogenes and Bacillus subtilis) as helper bacteria to help P. indica function better and further enhance rice cold tolerance. Under cold stress, the co-addition of the three beneficial microorganisms significantly increased the biomass, photosynthetic performance indicators, osmoregulatory substance contents, and antioxidant enzyme activities of rice. P. indica and helper bacteria significantly reduced the cold stress-induced increases in malondialdehyde content and electrolyte leakage rate in rice leaves and triggered a substantial upregulation of cold-tolerant genes in rice. This suggests that P. indica and helper bacteria can synergistically improve the cold resistance of rice. The findings of the research offer a conceptual foundation and practical assistance for enhancing rice's resilience to cold temperatures.

Background and objective: Magnetic resonance imaging (MRI) plays a vital role in diagnosing spinal diseases, including different types of spinal tumors. However, conventional segmentation techniques are often labor-intensive and susceptible to variability. This study aims to propose a full-automatic segmentation method for spine MRI images, utilizing a convolutional-deconvolution neural network and patch-based deep learning. The objective is to improve segmentation efficiency, meeting clinical needs for accurate diagnoses and treatment planning. Methods: The methodology involved the utilization of a convolutional neural network to automatically extract deep learning features from spine data. This allowed for the effective representation of anatomical structures. The network was trained to learn discriminative features necessary for accurate segmentation of the spine MRI data. Furthermore, a patch extraction (PE) based deep neural network was developed using a convolutional neural network to restore the feature maps to their original image size. To improve training efficiency, a combination of pre-training and an enhanced stochastic gradient descent method was utilized. Results: The experimental results highlight the effectiveness of the proposed method for spine image segmentation using Gadolinium-enhanced T1 MRI. This approach not only delivers high accuracy but also offers real-time performance. The innovative model attained impressive metrics, achieving 90.6% precision, 91.1% recall, 93.2% accuracy, 91.3% F1-score, 83.8% Intersection over Union (IoU), and 91.1% Dice Coefficient (DC). These results indicate that the proposed method can accurately segment spine tumors CT images, addressing the limitations of traditional segmentation algorithms. Conclusion: In conclusion, this study introduces a fully automated segmentation method for spine MRI images utilizing a convolutional neural network, enhanced by the application of the PE-module. By utilizing a patch extraction based neural network (PENN) deep learning techniques, the proposed method effectively addresses the deficiencies of traditional algorithms and achieves accurate and real-time spine MRI image segmentation.

In this study, we considered the compressible effect on the mutual interaction of two cavitation bubbles by correcting the sound field emitted by one bubble in the radial equations of the other bubble to first order in the Mach number of the flow, and the effect is represented by the incident wave acting on bubbles. The results illustrates that the incident wave can enhance the resonance response at the redistributed resonance frequency, which leads to an increase in radial acceleration and the secondary Bjerknes force, and rapid approach of bubbles. Furthermore, the influence of incident wave on the interaction of bubbles driven at lower frequencies is more significant, due to resonance enhancement caused by the proximity of natural frequencies and frequency multiplications of the external sound field. Our findings reveal that the compressible effect is not only critical to interaction in radial oscillations, but also in translational motion.

Subsurface drip irrigation in arid areas has the potential to replace traditional mulched drip irrigation to achieve green and sustainable cotton production. However, the suitable irrigation amount and frequency are still unclear, which seriously limits the ability of this model to improve water productivity and water-saving potential. Therefore, a field experiment was carried out from 2021 to 2023; a split plot experimental design was adopted with two irrigation amounts (W1, 3177 m3 ha−1; W2, 3840 m3 ha−1) and three irrigation frequencies (F1, 9; F2, 8; F3, 7). The effects of different irrigation strategies on the soil microenvironment, moisture content, biomass, and water use efficiency (WUE) of cotton organs were evaluated. The W2 treatment improved the soil moisture content, increased the soil temperature gradient, and reduced the soil conductivity, thereby increasing the moisture content and biomass of various organs. Moreover, compared with the F1 treatment, the F2 and F3 treatments were more likely to increase the soil moisture content, soil temperature gradient, WUEStem, WUELeaf and WUEBoll. In addition, the water consumption of the F2 and F3 treatments decreased by 3.9 % and 0.9 %, respectively, compared with that of the F1 treatment. These findings indicate that W2F2 can reduce water consumption while increasing boll biomass and WUEBoll. Further analysis revealed that under W2F2, WUEBoll was positively correlated with soil temperature gradient and soil conductivity and negatively correlated with leaf moisture content (LMC) and water consumption. In summary, with an irrigation amount of 3840 m3 ha−1, delaying the initial irrigation event and increasing the irrigation quota (8 irrigation events) improve the water environment in cotton fields, reducing soil temperature fluctuations and surface salt accumulation and synergistically increasing the boll biomass of cotton organs and WUEBoll. This irrigation strategy represents an effective cotton cultivation method to maximize cotton yield and improve resource utilization efficiency.

Objectives: Efforts to shorten rifampicin-resistant tuberculosis (RR-TB) treatment have led to concerns about hepatotoxicity in shorter regimens. We evaluated hepatotoxicity in two novel regimens against the standard shorter regimen recommended by the World Health Organization (WHO). Methods: Participants from the TB-TRUST and TB-TRUST plus trials were assigned to the WHO shorter regimen, a levofloxacin (Lfx)-based regimen, or a bedaquiline (Bdq)-based regimen. Liver function was tested bi-weekly in the first month, then monthly until treatment ended. Eligibility required receiving at least one drug dose and undergoing at least two liver function tests. Results: Of 429 patients, hepatotoxicity was most prevalent in the WHO shorter group (26.7% of 169), compared to 4.7% in the Lfx group (172 patients), and 5.7% in the Bdq group (88 patients). The median peak alanine aminotransferase levels were 1.67 × upper limit of normal (ULN) for WHO, 0.82 × ULN for Lfx, and 0.88 × ULN for Bdq groups. The incidence of drug-induced liver injury was significantly higher in the WHO group (18.3%) than in the Lfx (3.5%) and Bdq (4.6%) groups. The time to significant alanine aminotransferase elevation was about 2.8 months, with no differences between groups. Conclusions: Two novel regimens demonstrated lower hepatotoxicity compared to the WHO's shorter regimen. Entire course management monitoring is recommended in RR-TB treatment.

Objective: Circulating tumor DNA (ctDNA) is increasingly being used as a potential prognostic biomarker in cancer patients. We aimed to assess the prognostic value of ctDNA in different subtypes of breast cancer patients throughout the whole treatment cycle. Materials and methods: PubMed, Web of Science, Embase, Cochrane Library, Scopus, and clinical trials.gov databases were searched from January 2016 to May 2022. The following search terms were used: ctDNA OR circulating tumor DNA AND breast cancer OR breast carcinoma. Only studies written in English were included. The following pre-specified criteria should be met for inclusion: (i) original articles, conference abstracts, etc.; (ii) patients with breast cancer; (iii) ctDNA measurement; and (iv) clinical outcome data such as recurrence-free survival (RFS) and overall survival (OS). The random-effects model was preferred considering the potential heterogeneity across studies. The main outcomes are ctDNA detection rate and postoperative long-term outcomes (RFS and OS). Results: A total of 24 studies were screened. At every measurement time, the ctDNA detection rate of the HR+ subgroup was similar to that of the HR- subgroup (P = 0.075; P = 0.458; P = 0.744; and P = 0.578), and the ctDNA detection rate of the HER2+ subgroup was similar to that of the HER2- subgroup (P = 0.805; P = 0.271; P = 0.807; and P = 0.703). In the HR+ subgroup, RFS and OS of ctDNA positive patients were similar to those of ctDNA negative patients (P = 0.589 and P = 0.110), while RFS and OS of the ctDNA positive group was significantly shorter than those of the ctDNA negative patients in the HR- subgroup (HR = 4.03, P < 0.001; HR = 3.21, P < 0.001). According to HER grouping, the results were the same as above. In the triple negative breast cancer (TNBC) subgroup, the RFS and OS of ctDNA-positive patients was significantly shorter than of the ctDNA negative patients before and after surgery. Conclusions: ctDNA was more predictive of recurrence-free survival and overall survival in the HR- subgroup than in the HR+ subgroup, and the same result was showed in the HER2- subgroup vs. HER2+ subgroup. The prognosis of the TNBC subtype is closely related to ctDNA before and after surgery.

Phosphorus (P) fertilizer is a significant cost in crop production. Understanding the mechanisms behind P fertilizer–soil–crop interactions can enhance phosphorus use efficiency (PUE) and increasing yield. We conducted a rice cultivation bucket experiment in red soil (pH = 5.9) and lime concretion black soil (pH = 7.8) and applied single superphosphate (SSP), calcium magnesium phosphate (CMP), diammonium phosphate (DAP), triple superphosphate (TSP), ammonium polyphosphate (APP), and a control group (CK, no P fertilizer). We analyzed rice P uptake and utilization patterns, evaluated the impact of varying P fertilizer formulations on rice root morphology, yield, and PUE, and investigated changes in soil P pools. In red soil, the APP treatment produced the greatest total root length, rice yield, PUE and increased soil Olsen-P, NaHCO3-Pi at anthesis. In lime concretion black soil, the TSP and APP treatments had the highest rice yields and increased Olsen-P, H2O-P, NaHCO3-Pi at anthesis. Moreover, the TSP treatment had the greatest total root length and root surface area at anthesis and the APP treatment had the largest PUE. Random forest regression analysis revealed that residual-P and Olsen-P significantly impacted rice yield in red soil and lime concretion black soil, respectively. We recommend using APP in red soil and TSP and APP in lime concretion black soil for rice cultivation to optimize soil P pool characteristics and root morphology for nutrient uptake, ultimately leading to the highest yields and PUE.

Tunnel boring machines (TBMs) have been widely utilised in tunnel construction due to their high efficiency and reliability. Accurately predicting TBM performance can improve project time management, cost control, and risk management. This study aims to use deep learning to develop real-time models for predicting the penetration rate (PR). The models are built using data from the Changsha metro project, and their performances are evaluated using unseen data from the Zhengzhou Metro project. In one-step forecast, the predicted penetration rate follows the trend of the measured penetration rate in both training and testing. The autoregressive integrated moving average (ARIMA) model is compared with the recurrent neural network (RNN) model. The results show that univariate models, which only consider historical penetration rate itself, perform better than multivariate models that take into account multiple geological and operational parameters (GEO and OP). Next, an RNN variant combining time series of penetration rate with the last-step geological and operational parameters is developed, and it performs better than other models. A sensitivity analysis shows that the penetration rate is the most important parameter, while other parameters have a smaller impact on time series forecasting. It is also found that smoothed data are easier to predict with high accuracy. Nevertheless, over-simplified data can lose real characteristics in time series. In conclusion, the RNN variant can accurately predict the next-step penetration rate, and data smoothing is crucial in time series forecasting. This study provides practical guidance for TBM performance forecasting in practical engineering.

This study aimed to investigate the effect of oxidation temperature on the physical properties of β-Ga2O3 grown by thermal oxidation of GaN in the O2 ambient at high oxidation temperatures from 900 to 1400 °C. Notably, the analysis of the oxidation temperatures ranging from 1100 to 1400 °C remains unexplored in the other studies. The effects of the oxidation temperature on the physical properties were investigated via X-ray diffraction, X-ray photoelectron spectroscopy, and field-emission scanning electron microscopy (FESEM). At all oxidation temperatures, XPS results revealed oxygen deficiency. The top FESEM images indicated disordered nanocolumn structures on the surface of the oxide layer, which increased in abundance with oxidation temperatures exceeding 1100 °C. Energy-filtered high-resolution transmission electron microscopy images and the selected area electron diffraction patterns revealed that the top nanocolumns and the oxidized layer comprised polycrystalline β-Ga2O3. Additionally, the cross-sectional lamellae prepared through focused ion beam milling revealed an increase in the thickness of the β-Ga2O3 layer with oxidation temperature. To account for the variations in the oxide layer thickness, the oxidation temperatures were classified into two ranges of 900–1100 °C and 1100–1400 °C for calculating the activation energy. Moreover, this study assumed that the activation energy required for the oxidation process remained constant within each temperature interval. According to Arrhenius's model, the activation energy from 900 to 1100 °C was calculated to be 286.15 kJ/mol (2.97 eV). Furthermore, the activation energy from 1100 to 1400 °C was determined to be 111.33 kJ/mol (1.15 eV).

Summary: Background: Previous conventional epidemiological studies found a J-shape relationship between alcohol consumption and dementia, but this result was subject to confounding biases and reverse causation. Therefore, we aimed to investigate the potential linear or non-linear causal association between alcohol consumption and the incident risk of dementia in current drinkers. Methods: This study used data from the UK Biobank to investigate the relationship between alcohol consumption and dementia risk. 313,958 White British current drinkers, who were free of dementia during 2006–2010, were followed up until 2021. Alcohol consumption was self-reported and calculated according to the National Health Service guideline. The primary outcome was all-cause dementia identified through hospital and mortality records. We used multivariable Cox models with restricted cubic splines for conventional analysis and both non-linear and linear Mendelian Randomization (MR) analyses to assess causal relationships, employing a genetic score based on 95 SNPs identified from a meta-genome-wide association study of 941,280 people from Europe. Findings: 313,958 current drinkers consumed an average of 13.6 [IQR: 7.1–25.2] units/week alcohol (men averaged 20.2 [11.1–33.9] units/week and women 9.5 [5.3–16.7] units/week). During a mean follow-up of 13.2 years, 5394 (1.7%) developed dementia. Multivariable Cox model with restricted cubic spline functions identified a J-shaped relationship between alcohol consumption and dementia risk, with the lowest risk at 12.2 units/week. The non-linear MR failed to identify a significant non-linear causal relationship (p = 0.45). Both individual-level (HR: 2.22 95%CI [1.06–4.66]) and summary-level (1.89 [1.53–2.32]) linear MR analyses indicated that higher genetically predicted alcohol consumption increased dementia risk. Interpretation: This study identified a positive linear causal relationship between alcohol consumption and dementia among current drinkers. The J-shaped association found in conventional epidemiological analysis was not supported by non-linear MR analyses. Our findings suggested that there was no safe level of alcohol consumption for dementia. Objectives: To determine the efficacy and safety of respiratory syncytial virus (RSV) vaccines in infants and older adults. Methods: We performed a systematic review and meta-analysis of randomized control trials that evaluated the efficacy of maternal RSV immunization against infections in infants, as well as the efficacy of RSV vaccines in older adults. The primary outcome was the vaccine efficacy against RSV-related lower respiratory tract disease (LRTD). Grading of Recommendations Assessment, Development and Evaluation criteria was used to evaluate the level of evidence. Results: Ten trials were included in the review. For maternal vaccination, the RSV vaccine showed favourable efficacy against RSV-related LRTD (vaccine efficacy 57.3%, 95% confidence interval [CI] 31.3-73.5; low certainty) and RSV-related severe LRTD (vaccine efficacy 81.9%, 95% CI 56.8-92.4; moderate certainty) in infants within 90 days after birth. For older adults, Meta-analysis showed that RSV vaccines could also reduce the risk of RSV-related LRTD (vaccine efficacy 78.3%, 95% CI 65.6-86.3; moderate certainty) and RSV-related severe LRTD (vaccine efficacy 86.5%, 95% CI 68.3-94.3; moderate certainty). There was no significant difference in serious adverse events between RSV vaccines and placebo. Conclusion: RSV vaccines have the potential to offer protection against RSV disease in both infants and older adults, without apparent safety concerns.

The coexistence of microorganisms in complex soil environments greatly affects the environmental behavior and ecological effects of microplastics (MPs). However, relevant studies are sparse, and internal mechanisms remain unclear. Herein, arbuscular mycorrhizal fungi (AMF), a common symbiotic microorganism in the soil–plant system, was proved to significantly affect MPs absorption and migration with a “size effect”. Specifically, the existence of AMF accelerated small-sized MPs (0.5 μm) uptake but slowed large-sized MPs (2 μm) uptake in lettuce. The content of 0.5 μm MPs absorbed by plants with AMF was 1.26 times that of the non-AMF group, while the content of 2 μm MPs was only 77.62 % that of non-AMF group. Additionally, the different effects of microorganisms on the intake content of MPs with different particle sizes in plants also led to different toxic effects of MPs on lettuce, that is, AMF exacerbated small-size MPs toxicity in lettuce (e.g., reduced plant biomass, photosynthesis, etc), and it weakened large-sized MPs toxicity (e.g., increased plant height, antioxidant enzyme activity, etc). The above phenomenon mainly because of the change in AMF on the plant root structure, which can be visually observed through the intraradical and extraradical hyphae. The symbiotic structure (hyphae) formed by AMF and host plants root could enhance the absorption pathway for small-sized MPs in lettuce, although not for large-sized MPs. Additionally, the effects of AMF varied with the soil environment of differently sized MPs, which promoted the migration of small-particle MPs to plants but aggravated large-particle MPs fixation at the soil interface. These findings could deepen the understanding of MPs pollution in terrestrial systems and provide theoretical basis and technical support to accurately assess soil MPs pollution.

Background: Glyphosate is a widely used herbicide. Clinical presentations of glyphosate intoxication show variation, but hepatic portal venous gas(HPVG) caused by glyphosate poisoning is rarely reported. Herein, we report a rare case of ominous HPVG after ingesting glyphosate. HPVG, which used to be an ominous abdominal radiologic sign, is associated with numerous underlying abdominal pathologies, ranging from benign conditions that require no invasive treatment to potentially lethal diseases that necessitate prompt surgical intervention. Case summary: A young woman who ingested 100 mL glyphosate 6-h prior was admitted to the emergency intensive care unit. Before admission to our hospital, the patient was administered gastric lavage treatment with 10000 mL of normal saline in the local hospital. After 14 h, her laboratory examinations showed systemic inflammatory response syndrome and multiple organ dysfunction syndrome, while the condition deteriorated. Computed tomography of the abdomen showed multilinear air densities in the portal vein, hepatic branches, and mesenteric vessels, intestinal obstruction, and intestinal necrosis. Septic shock and a severe abdominal infection were diagnosed. The patient was treated conservatively as they could not tolerate surgery and, after 20 h died of septic shock. Conclusion: We reviewed 289 cases of ''hepatic portal venous gas'' in PUBMED and analyzed the etiology and treatment of HPVG accompanied by the underlying pathology. We concluded that HPVG is a radiological sign associated with various diseases, and the prognosis mainly depends on the underlying cause and clinical condition. As glyphosate may erode the digestive tract, attention should be paid to the volume, pressure, and speed of gastric lavage in treating glyphosate poisoning to avoid fatal complications such as HPVG. Abdominal symptoms need to be closely observed, and changes in the early onset of the condition in clinical practice need to be responded to promptly.

Intestinal dysbiosis and disrupted bile acid (BA) homeostasis are associated with obesity, but the precise mechanisms remain insufficiently explored. Hepatic protein phosphatase 1 regulatory subunit 3G (PPP1R3G) plays a pivotal role in regulating glycolipid metabolism; nevertheless, its obesity-combatting potency remains unclear. In this study, a substantial reduction was observed in serum PPP1R3G levels in high-body mass index (BMI) and high-fat diet (HFD)-exposed mice, establishing a positive correlation between PPP1R3G and non-12α-hydroxylated (non-12-OH) BA content. Additionally, hepatocyte-specific overexpression of Ppp1r3g (PPP1R3G HOE) mitigated HFD-induced obesity as evidenced by reduced weight, fat mass, and an improved serum lipid profile; hepatic steatosis alleviation was confirmed by normalized liver enzymes and histology. PPP1R3G HOE considerably impacted systemic BA homeostasis, which notably increased the non-12-OH BAs ratio, particularly lithocholic acid (LCA). 16S ribosomal DNA (16S rDNA) sequencing assay indicated that PPP1R3G HOE reversed HFD-induced gut dysbiosis by reducing the Firmicutes/Bacteroidetes ratio and Lactobacillus population, and elevating the relative abundance of Blautia, which exhibited a positive correlation with serum LCA levels. A fecal microbiome transplantation test confirmed that the anti-obesity effect of hepatic PPP1R3G was gut microbiota-dependent. Mechanistically, PPP1R3G HOE markedly suppressed hepatic cholesterol 7α-hydroxylase (CYP7A1) and sterol-12α-hydroxylase (CYP8B1), and concurrently upregulated oxysterol 7-α hydroxylase and Takeda G protein-coupled BA receptor 5 (TGR5) expression under HFD conditions. Furthermore, LCA administration significantly mitigated the HFD-induced obesity phenotype and elevated non-12-OH BA levels. These findings emphasize the significance of hepatic PPP1R3G in ameliorating diet-induced adiposity and hepatic steatosis through the gut microbiota-BA axis, which may serve as potential therapeutic targets for obesity-related disorders.

Purpose: To assess the value of the driving pressure variation rate (ΔP%) in predicting the outcome of weaning from invasive mechanical ventilation in patients with acute respiratory distress syndrome. Methods: In this case-control study, a total of 35 patients with moderate-severe acute respiratory distress syndrome were admitted to the intensive care unit between January 2022 and December 2022 and received invasive mechanical ventilation for at least 48 h were enrolled. Patients were divided into successful weaning group and failed weaning group depending on whether they could be removed from ventilator support within 14 days. Outcome measures including driving pressure, PaO2:FiO2, and positive end-expiratory pressure, etc. were assessed every 24 h from day 0 to day 14 until successful weaning was achieved. The measurement data of non-normal distribution were presented as median (Q1, Q3), and the differences between groups were compared by Wilcoxon rank sum test. And categorical data use the Chi-square test or Fisher's exact test to compare. The predictive value of ΔP% in predicting the outcome of weaning from the ventilator was analyzed using receiver operating characteristic curves. Results: Of the total 35 patients included in the study, 17 were successful vs. 18 failed in weaning from a ventilator after 14 days of mechanical ventilation. Objective: Circulating tumor DNA (ctDNA) is increasingly being used as a potential prognosis biomarker in patients of breast cancer. This review aims to assess the clinical value of ctDNA in outcome prediction in breast cancer patients throughout the whole treatment cycle. Methods: PubMed, Web of Science, Embase, Cochrane Library, Scopus, and clinical trials.gov were searched from January 2016 to May 2022. Conference abstracts published in last three years were also included. The following search terms were used: ctDNA OR circulating tumor DNA AND breast cancer OR breast carcinoma. Only studies written in English languages were included. The following pre-specified criteria should be met for inclusion: (1) observational studies (prospective or retrospective), randomized control trials, case-control studies and case series studies; (2) patients with breast cancer; (3) ctDNA measurement; (4) clinical outcome data such as objective response rate (ORR), pathological complete response (pCR), relapse-free survival (RFS), overall survival (OS), and so on. The random-effect model was preferred considering the potential heterogeneity across studies. The primary outcomes included postoperative short-term outcomes (ORR and pCR) and postoperative long-term outcomes (RFS, OS, and relapse). Secondary outcomes focused on ctDNA detection rate. Results: A total of 30 studies, comprising of 19 cohort studies, 2 case-control studies and 9 case series studies were included. The baseline ctDNA was significantly negatively associated with ORR outcome (Relative Risk [RR] = 0.65, 95% confidence interval [CI]: 0.50–0.83), with lower ORR in the ctDNA-positive group than ctDNA-negative group. ctDNA during neoadjuvant therapy (NAT) treatment was significantly associated with pCR outcomes (Odds Ratio [OR] = 0.15, 95% CI: 0.04–0.54). The strong association between ctDNA and RFS or relapse outcome was significant across the whole treatment period, especially after the surgery (RFS: Hazard Ratio [HR] = 6.74, 95% CI: 3.73–12.17; relapse outcome: RR = 7.11, 95% CI: 3.05–16.53), although there was heterogeneity in these results. Pre-operative and post-operative ctDNA measurements were significantly associated with OS outcomes (pre-operative: HR = 2.03, 95% CI: 1.12–3.70; post-operative: HR = 6.03, 95% CI: 1.31–27.78). Conclusions: In this review, ctDNA measurements at different timepoints are correlated with evaluation indexes at different periods after treatment. The ctDNA can be used as an early potential postoperative prognosis biomarker in breast cancer, and also as a reference index to evaluate the therapeutic effect at different stages.

Summary: People often display stronger aversion to losses than appetite for equivalent gains, a widespread phenomenon known as loss aversion. The prevailing theory attributes loss aversion to a valuation bias that amplifies losses relative to gains. An alternative account attributes loss aversion to a response bias that avoids choices that might result in loss. By modeling the temporal dynamics of scalp electrical activity during decisions to accept or reject gambles within a sequential sampling framework, we decomposed valuation bias and response bias from a single event-related neural signal, the P3. Specifically, we found valuation bias manifested as larger sensitivity of P3 to losses than gains, which was localizable to reward-related brain regions. By contrast, response bias manifested as larger P3 preceding gamble acceptance than rejection and was localizable to motor cortex. Our study reveals the dissociable neural biomarkers of response bias and valuation bias underpinning loss-averse decisions.

Cell surface molecules transiently upregulated on activated T cells can play a counter-regulatory role by inhibiting T cell function. Deletion or blockade of such immune checkpoint receptors has been investigated to improve the function of engineered immune effector cells. CD38 is upregulated on activated T cells, and although there have been studies showing that CD38 can play an inhibitory role in T cells, how it does so has not fully been elucidated. In comparison with molecules such as PD1, CTLA4, LAG3, and TIM3, we found that CD38 displays more sustained and intense expression following acute activation. After deleting CD38 from human chimeric antigen receptor (CAR) T cells, we showed relative resistance to exhaustion in vitro and improved anti-tumor function in vivo. CD38 is a multifunctional ectoenzyme with hydrolase and cyclase activities. Reintroduction of CD38 mutants into T cells lacking CD38 provided further evidence supporting the understanding that CD38 plays a crucial role in producing the immunosuppressive metabolite adenosine and utilizing nicotinamide adenine dinucleotide (NAD) in human T cells. Taken together, these results highlight a role for CD38 as an immunometabolic checkpoint in T cells and lead us to propose CD38 deletion as an additional avenue for boosting CAR T cell function.

Wildlife conservation and management in human-dominated landscapes are major concerns for wildlife ecologists and managers. The dynamics of human disturbance, combined with seasonal limitations in the availability of nutritious foods, may restrict wildlife population growth and recovery. However, understanding how large mammal species adjust their population distribution in forest habitats with seasonal changes in food and disturbances requires a deeper and more extensive analysis. In this study, we found that three ungulate species, roe deer (Capreolus pygargus), sika deer (Cervus nippon), and wild boar (Sus scrofa), employ robust, conservative, and flexible distribution strategies, respectively, to adapt to the effects of seasonal changes and human disturbances. Moreover, croplands, villages, and grazing have some negative effects on the distribution of roe deer and sika deer, while wild boar can be highly abundant near human land use. Additionally, roe deer, sika deer, and wild boar are also affected by the abundance of shrub species they consume. During the cold season, the populations of the Amur tiger (Panthera tigris altaica) and Amur leopard (P. pardus orientalis) were primarily located near roads and dense forests, respectively. In the warm season, the distribution of both big cats was influenced by prey abundance, and Amur tigers also avoided grazing livestock. Nevertheless, the negative effects of human land use on Amur tigers and wild boars increased during the warm season, which was attributed to more frequent human activities during that time. Consequently, it is crucial to implement season-specific habitat management, particularly by regulating human disturbances during the warm season, in order to promote the recovery and expansion of populations of big cats and ungulates.

The inverse kinematics problem plays a crucial role in robotic manipulator planning, autonomous control, and object grasping. This problem can be solved in simple environments based on existing studies. However, it is still challenging to quickly find a feasible inverse kinematic solution when obstacle avoidance is required. In this paper, we present a nonconvex composite programming method to solve the inverse kinematics problem with overhead obstacle-avoidance requirements. Our method enables efficient obstacle avoidance by directly calculating the minimum distance between the manipulator and the overhead environment. We construct end-effector error functions based on the Product of Exponentials model and explicitly provide their gradient formula. We derive the minimum distance based on the geometry parametric equation and directly utilize it to construct the obstacle avoidance function. We propose an enhanced version of adaptive moment estimation based on short-time gradient information to improve optimization performance. Finally, we conduct simulations and experiments in overhead line environments. Comparative results with other optimization methods demonstrate that our proposed method achieves a high success rate with a low solution time.

Aim: To compare the survival outcomes between male and female patients with intrahepatic cholangiocarcinoma who underwent liver resection. Methods: Data from 976 consecutive intrahepatic cholangiocarcinoma patients undergoing liver resection between January 2005 and May 2013 at the Eastern Hepatobiliary Surgery Hospital were prospectively collected and retrospectively reviewed. Patient clinicopathological characteristics, overall survival, and cumulative recurrence rates were compared between male and female patients using propensity score matching. Results: Propensity score matching generated 313 matched pairs of patients. Among the entire cohort, the 1-, 3-, and 5-year overall survival and recurrence rates of the male and female patients were 60.2 %, 37.3 %, and 27.7 % vs. 65.8 %, 40.4 %, and 31.0 % (P = 0.380) and 50.6 %, 67.4 %, and 74.2 % vs. 44.4 %, 63.5 %, and 69.6 % (P = 0.123), respectively. In the matched cohort, the 1-, 3-, and 5-year overall survival and recurrence rates of the male and female patients were 60.6 %, 35.9 % and 22.4 % vs. 66.4 %, 40.6 % and 31.1 % (P = 0.041) and 51.5 %, 69.3 % and 83.9 % vs. 44.3 %, 63.6 %, and 69.9 % (P = 0.041), respectively. After adjustment for other confounding variables by multivariate Cox regression analysis, male sex was independently associated with worse overall survival (hazard ratio = 1.322, 95 % confidence interval: 1.079–1.621, P = 0.007) and tumor recurrence (hazard ratio = 1.337, 95 % confidence interval: 1.088–1.645, P = 0.006). A subgroup analysis of patients younger than 55 years old after propensity score matching showed that male patients had significantly worse overall survival and higher recurrence rates than female patients after surgery, while no significant difference in long-term overall survival and recurrence was observed between male and female patients older than 55 years old after propensity score matching. Conclusion: Male sex was an independent risk factor for overall survival and tumor recurrence in patients after liver resection for intrahepatic cholangiocarcinoma.

Background: Major depressive disorder (MDD) is a widespread health issue in many countries, which has an extremely negative impact on the health of children and adolescents in particular. In the context of depression and metabolic disorders, dyslipidemia and metabolism-related problems become more prominent comorbidities. However, they continue to be the main barrier to the successful recovery of the clinical progress. In this study we investigated the rate of dyslipidemia, additional risk factors among Chinese children and adolescents with MDD, and association of the suicidal behavior with lipid levels. Methods: The study took 756 people from the Third People's Hospital of Fuyang between January 2020 and December 2021, aged between 8 and 18, with major depressive disorders diagnosed according to the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM-5). We determined the FBG (fasting blood glucose) and lipid parameters in all subjects and also investigated the history of suicidal ideation, the cases of attempted suicide, and the scores of depressive symptoms. Sociodemographic and clinical data were gathered and analyzed using the SPSS-23.0 version. Results: The prevalence of hypercholesterolemia, hypertriglyceridemia, high LDL-C, and low HDL-C were 5.42 % (41/756), 10.58 % (80/756), 3.84 % (29/756) and 5.42 % (41/756) respectively. For hypercholesterolemia and hypertriglyceridemia, they were positive associated with suicidal ideation and suicide attempts, and the positive correlation is shown between low HDL-C levels and suicide attempts. Nevertheless, non-ideation and inversely suicidal attempts were not discovered among high-LDL-C subjects. Purpose: Atrophic nonunion is one of the most difficult complications of fracture. The cellular factors that contribute to atrophic nonunion are poorly understood, and mesenchymal stem cells (MSCs) are recognized as the key contributor to bone formation. This study aimed to characterize the MSCs isolated from the fibrotic tissue of atrophic nonunion (AN-MSCs) from the perspective of proliferation, differentiation potential, senescence, and paracrine function. Methods: Human atrophic fibrotic tissue was obtained from four donors aged 29–37 for isolating AN-MSCs, and donor-matched bone marrow acquired from the iliac crest for isolating MSCs (IC-MSCs) as control. The AN-MSCs or IC-MSCs in passage 3 were applied for the following evaluations. The surface markers expressed on the two cells were evaluated using flow cytometry. The proliferation of the two cells for up to 11 days was comparatively investigated. After osteogenic, chondrogenic, or adipogenic induction, multi-lineage differentiation of AN-MSCs or IC-MSCs was comparatively evaluated using lineage-specific stains and lineage-specific gene expression. Enzyme-linked immunosorbent assay (ELISA) assessment was applied to evaluate the paracrine function of AN-MSCs or IC-MSCs. Cellular senescence of AN-MSCs or IC-MSCs was evaluated using senescence-associated β-galactosidase (SA-β-gal) staining. Results: AN-MSCs or IC-MSCs from the four donors showed morphologic and immunophenotypic characteristics of MSCs, with the expression of MSCs markers and negative expression of hematopoietic markers. In general, AN-MSCs showed similar proliferation and adipogenic capacity with IC-MSCs. In contrast, IC-MSCs showed significantly higher osteogenic and chondrogenic capacity compared to AN-MSCs. Moreover, the culture medium of IC-MSCs contains significantly higher levels of VEGF, TGF-β1, PDGF-BB, and IGF-1 than the culture medium of AN-MSCs. Lastly, the AN-MSCs are more prone to cellular senescence than the IC-MSCs. Conclusions: In-vitro, AN-MSCs were similar to IC-MSCs in proliferation and adipogenic capacity, but inferior to IC-MSCs in osteogenic and chondrogenic capacity, paracrine function, and anti-senescence.

In this paper, the research on bending and low resistance drift of In2Se3/Sb composite multilayer films based on flexible substrate were investigated. The flexible film can still complete the phase transition after being at different bending radius for 24 h and bending 2 × 104 times. With the increase of bending times, the crystalline resistance of the film increased gradually, the band gap widened. Compared with the flat film, the stretched and pressed films had a lower resistance drift. External forces such as bending, stretching and pressing will reduce the surface roughness of the flexible film. The stability of the multilayer structure for flexible thin film was checked by transmission electron microscopy. The devices based on In2Se3/Sb composite multilayer thin film were prepared on a flexible substrate polyether ether ketone. The device can achieve Set and Reset operation in the states of flat, bending and after bending 100 times. It had low power consumption. This study could provide a potential option for flexible information storage.

Objectives: Long, ineffective, and toxic regimens hinder the treatment of patients with multidrug-resistant tuberculosis (MDR-TB) and pre-extensive drug-resistant tuberculosis (pre-XDR-TB). Methods: We conducted a multicenter cohort study to prospectively evaluate the safety and efficacy of three 9-month, all-oral, 5-drug regimens. Regimen A (bedaquiline [Bdq]+linezolid [Lzd]+moxifloxacin [Mfx]+cycloserine [Cs]+pyrazinamide [Pza]) and Regimen B (Lzd+Mfx+Cs+clofazimine [Cfz]+Pza) were used to treat MDR-TB patients (Groups A and B, respectively, assigned according to the patient's treatment preference), while Regimen C (Bdq+Lzd+Cs+Cfz+Pza) was used to treat pre-XDR-TB patients (Group C). The primary endpoint was the occurrence of an unfavorable outcome within 12 months of treatment completion, regardless of regimen. Results: A total of 104 patients (34 in Group A, 46 in Group B, and 24 in Group C), with a median age of 35.5 (29.0-54.0) years, were included in the analysis population. At 12 months after treatment completion, five patients were deemed non-assessable. Of the remaining 99 participants, seven (7.1%) had an unfavorable outcome (including two deaths from any cause, four with treatment failure, and one loss to follow-up) and 92 (92.9%) had a favorable outcome. Culture conversion was achieved in 82.5% (80/97) of participants at month 2 and in 97.9% (94/97) of participants at month 6. Adverse events (AEs) resulting in drug adjustment occurred in 69.2% (72/104) of participants, mainly due to Lzd and Pza use. A QT interval prolongation of ≥ 500 ms occurred in 5.8% (6/104) of participants. Conclusion: The primary outcome of the three tailored, 9-month, all-oral, 5-drug regimens was satisfactory in the vast majority of MDR-TB and pre-XDR-TB patients, with manageable and reversible AEs.

Background/purpose: The success of transcrestal sinus floor elevation (TSFE) is primarily reliant upon the experience of the surgeon owing to the limited operative visibility. To evaluate the accuracy associated with the use of a dynamic navigation system when conducting posterior maxilla implant surgery with TSFE. Materials and methods: Twenty-eight implants were placed in 28 patients requiring implantation in the posterior maxilla via a TSFE approach. The drills were used to access the planned position (within 1 mm of the bottom of the maxillary sinus floor) under dynamic navigation system. TSFE was then accomplished using osteotomes and a piezoelectric device. Lastly, the implant was inserted under the dynamic navigation. Three effective deviations between planned and actual implant placement were then measured including angular deviation (AD, degrees), entry point horizontal deviation (EPHD, mm), and apical point horizontal deviation (APHD, mm). Results: The AD, EPHD, and APHD between the planned and actual implant placement were 3.656 ± 1.665°, 1.073 ± 0.686 mm, and 1.086 ± 0.667 mm, respectively. Premolar site AD values were less than those for molar sites (P = 0.004). No significant differences in these outcomes were observed in different surgeons. Obvious sinus perforation was not detected by immediate postoperative cone beam computed tomography imaging. Conclusion: The accuracy associated with using a dynamic navigation system when conducting posterior maxilla implant surgery via a TSFE approach using piezoelectric devices was comparable. This technique thus achieved appropriate interventional precision and safety while decreasing the morbidity associated with the TSFE approach.

Objective: The incidence and mortality of hepatocellular carcinoma (HCC) have been increasing around the world. Current guidelines recommend HCC screening in high-risk population. However, the strength of evidence of benefits and harms of HCC screening to support the recommendation was unclear. The objective is to systematically synthesize current evidence on the benefits and harms of HCC screening. Methods: We searched PubMed and nine other databases until August 20, 2021. We included cohort studies and RCTs that compared the benefits and harms of screening and non-screening in high-risk population of HCC. Case series studies that reported harms of HCC screening were also included. Pooled risk ratio (RR), according to HCC screening status, was calculated for each benefit outcome (e.g., HCC mortality, survival rate, proportion of early HCC), using head-to-head meta-analysis. The harmful outcomes (e.g., proportion of physiological harms provided by non-comparative studies were pooled by prevalence of meta-analysis. Analysis on publication bias and quality of life, subgroup analysis, and sensitivity analysis were also conducted. Results: We included 70 studies, including four random clinical trials (RCTs), 63 cohort studies,three case series studies. The meta-analysis of RCTs showed HCC screening was significantly associated with reduced HCC mortality (RR [risk ratio], 0.73 [95% CI, 0.56–0.96]; I2 = 75.1%), prolonged overall survival rates (1-year, RR, 1.72 [95% CI, 1.13–2.61]; I2 = 72.5%; 3-year, RR, 2.86 [95% CI, 1.78–4.58]; I2 = 10.1%; and 5-year, RR, 2.76 [95% CI, 1.37–5.54]; I2 = 28.3%), increased proportion of early HCC detection (RR, 2.68 [95% CI, 1.77–4.06]; I2 = 50.4%). Similarly, meta-analysis of cohort studies indicated HCC screening was more effective than non-screening. However, pooled proportion of physiological harms was 16.30% (95% CI: 8.92%–23.67%) and most harms were of a mild to moderate severity. Conclusion: The existing evidence suggests HCC screening is more effective than non-screening in high-risk population. However, harms of screening should not be ignored.

ARPC1B encodes the protein known as actin-related protein 2/3 complex subunit 1 B (ARPC1B), which controls actin polymerization in the human body. Although ARPC1B has been linked to several human malignancies, its function in these cancers remains unclear. TCGA, GTEx, CCLE, Xena, CellMiner, TISIDB, and molecular signature databases were used to analyze ARPC1B expression in cancers. Visualization of data was primarily achieved using R language, version 4.0. Nineteen tumors exhibited high levels of ARPC1B expression, which were associated with different tumor stages and significantly affected the prognosis of various cancers. The level of ARPC1B expression substantially connected the narrative of ARPC1B expression with several TMB cancers and showed significant changes in MSI. Additionally, tolerance to numerous anticancer medications has been linked to high ARPC1B gene expression. Using Gene Set Variation Analysis/Gene Set Enrichment Analysisanalysis and concentrating on Rectum adenocarcinoma (READ), we thoroughly examined the molecular processes of the ARPC1B gene in pan-cancer. Using WGCNA, we examined the co-expression network of READ and ARPC1B. Meanwhile, ten specimens were selected for immunohistochemical examination, which showed high expression of ARPC1B in READ. Human pan-cancer samples show higher ARPC1B expression than healthy tissues. In many malignancies, particularly READ, ARPC1B overexpression is associated with immune cell infiltration and a poor prognosis. These results imply that the molecular biomarker ARPC1B may be used to assess the prognosis and immune infiltration of patients with READ.

For bottom-up bone tissue engineering, microcarriers play crucial roles in serving as scaffolds that provide structural support and environmental cues for cell growth. Studies have demonstrated that the physicochemical properties of microcarriers, such as surface structure and chemistry, have significant effects on cell phenotype. However, current research on microcarriers has primarily focused on hydrogels or polymers, neglecting the exploration of hard materials that mimic the bone tissue environment. In this study, a new facile method to fabricate magnetic calcium phosphate (CaP)-based microcarriers were presented. By this method, magnetic calcium phosphate was obtained without changing the surface nanostructure of CaP microspheres. These magnetic CaP microspheres were utilized as cell-adhesive microcarriers, allowing for cell-guided assembly into spherical microtissue under a relatively low number of cells. However, when using CaP microspheres as microcarriers, only loose aggregates were observed. To enhance the biofunctionality of the magnetic CaP microcarriers, bisphosphonate-modified growth differentiation factor-5 (PGDF-5) were synthesized and selectively deposited onto the magnetic CaP surfaces. Coculture with preosteoblastic cells demonstrated that PGDF-5 modified microcarriers effectively improved cell osteogenic differentiation and angiogenesis, thereby facilitating the formation of highly stable cell spheroids. Collectively, all findings suggest that PGDF-5 modified magnetic CaP microcarriers hold great potential as tools for fabricating bottom-up tissue engineered bone-mimicking tissue.

Summary: Background: Liver cirrhosis (LC) is the highest risk factor for hepatocellular carcinoma (HCC) development worldwide. The efficacy of the guideline-recommended surveillance methods for patients with LC remains unpromising. Methods: A total of 4367 LCs not previously known to have HCC and 510 HCCs from 16 hospitals across 11 provinces of China were recruited in this multi-center, large-scale, cross-sectional study. Participants were divided into Stage Ⅰ cohort (510 HCCs and 2074 LCs) and Stage Ⅱ cohort (2293 LCs) according to their enrollment time and underwent Tri-phasic CT/enhanced MRI, US, AFP, and cell-free DNA (cfDNA). A screening model called PreCar Score was established based on five features of cfDNA using Stage Ⅰ cohort. Surveillance performance of PreCar Score alone or in combination with US/AFP was evaluated in Stage Ⅱ cohort. Findings: PreCar Score showed a significantly higher sensitivity for the detection of early/very early HCC (Barcelona stage A/0) in contrast to US (sensitivity of 51.32% [95% CI: 39.66%–62.84%] at 95.53% [95% CI: 94.62%–96.38%] specificity for PreCar Score; sensitivity of 23.68% [95% CI: 14.99%–35.07%] at 99.37% [95% CI: 98.91%–99.64%] specificity for US) (P

Summary: Under stress conditions, translationally stalled mRNA and associated proteins undergo liquid-liquid phase separation and condense into cytoplasmic foci called stress granules (SGs). Many viruses hijack SGs for their pathogenesis; however, whether pathogenic bacteria also exploit this pathway remains unknown. Here, we report that members of the OspC family of Shigella flexneri induce SG formation in infected cells. Mechanistically, the OspC effectors target multiple subunits of the host translation initiation factor 3 complex by ADP-riboxanation. The modification of eIF3 leads to translational arrest and thus the formation of SGs. Furthermore, OspC-mediated SGs are beneficial for S. flexneri replication within infected host cells, and bacterial strains unable to induce SGs are attenuated for virulence in a murine model of infection. Our findings reveal a mechanism by which bacterial pathogens induce SG assembly by inactivating host translational machinery and promote bacterial proliferation in host cells.
