Copyright © 2023 The Author(s). Background: Sleep disturbance is common following hospital admission both for COVID-19 and other causes. The clinical associations of this for recovery after hospital admission are poorly understood despite sleep disturbance contributing to morbidity in other scenarios. We aimed to investigate the prevalence and nature of sleep disturbance after discharge following hospital admission for COVID-19 and to assess whether this was associated with dyspnoea. Methods: CircCOVID was a prospective multicentre cohort substudy designed to investigate the effects of circadian disruption and sleep disturbance on recovery after COVID-19 in a cohort of participants aged 18 years or older, admitted to hospital for COVID-19 in the UK, and discharged between March, 2020, and October, 2021. Participants were recruited from the Post-hospitalisation COVID-19 study (PHOSP-COVID). Follow-up data were collected at two timepoints: an early time point 2–7 months after hospital discharge and a later time point 10–14 months after hospital discharge. Sleep quality was assessed subjectively using the Pittsburgh Sleep Quality Index questionnaire and a numerical rating scale. Sleep quality was also assessed with an accelerometer worn on the wrist (actigraphy) for 14 days. Participants were also clinically phenotyped, including assessment of symptoms (ie, anxiety [Generalised Anxiety Disorder 7-item scale questionnaire], muscle function [SARC-F questionnaire], dyspnoea [Dyspnoea-12 questionnaire] and measurement of lung function), at the early timepoint after discharge. Actigraphy results were also compared to a matched UK Biobank cohort (non-hospitalised individuals and recently hospitalised individuals). Multivariable linear regression was used to define associations of sleep disturbance with the primary outcome of breathlessness and the other clinical symptoms. PHOSP-COVID is registered on the ISRCTN Registry (ISRCTN10980107). Findings: 2320 of 2468 participants in the PHOSP-COVID study attended an early timepoint research visit a median of 5 months (IQR 4–6) following discharge from 83 hospitals in the UK. Data for sleep quality were assessed by subjective measures (the Pittsburgh Sleep Quality Index questionnaire and the numerical rating scale) for 638 participants at the early time point. Sleep quality was also assessed using device-based measures (actigraphy) a median of 7 months (IQR 5–8 months) after discharge from hospital for 729 participants. After discharge from hospital, the majority (396 [62%] of 638) of participants who had been admitted to hospital for COVID-19 reported poor sleep quality in response to the Pittsburgh Sleep Quality Index questionnaire. A comparable proportion (338 [53%] of 638) of participants felt their sleep quality had deteriorated following discharge after COVID-19 admission, as assessed by the numerical rating scale. Device-based measurements were compared to an age-matched, sex-matched, BMI-matched, and time from discharge-matched UK Biobank cohort who had recently been admitted to hospital. Compared to the recently hospitalised matched UK Biobank cohort, participants in our study slept on average 65 min (95% CI 59 to 71) longer, had a lower sleep regularity index (–19%; 95% CI –20 to –16), and a lower sleep efficiency (3·83 percentage points; 95% CI 3·40 to 4·26). Similar results were obtained when comparisons were made with the non-hospitalised UK Biobank cohort. Overall sleep quality (unadjusted effect estimate 3·94; 95% CI 2·78 to 5·10), deterioration in sleep quality following hospital admission (3·00; 1·82 to 4·28), and sleep regularity (4·38; 2·10 to 6·65) were associated with higher dyspnoea scores. Poor sleep quality, deterioration in sleep quality, and sleep regularity were also associated with impaired lung function, as assessed by forced vital capacity. Depending on the sleep metric, anxiety mediated 18–39% of the effect of sleep disturbance on dyspnoea, while muscle weakness mediated 27–41% of this effect. Interpretation: Sleep disturbance following hospital admission for COVID-19 is associated with dyspnoea, anxiety, and muscle weakness. Due to the association with multiple symptoms, targeting sleep disturbance might be beneficial in treating the post-COVID-19 condition. This is an open access article under the CC BY-NC-ND license., Modeling infiltration in water-repellent soils is difficult, as the underlying processes remain poorly quantified. However, recent work has adapted the Beerkan Soil Transfer Parameter (BEST) algorithm to include an exponential correction term for characterizing these types of soils. The original BEST-WR (WR = Water Repellent) method used a two-term approximate expansion of the Haverkamp quasi-exact implicit model. However, the BEST-WR method can have considerable inaccuracy, particularly as the time of infiltration and the soil water repellency increase. Here, we extended the BEST-WR model by adapting a three-term approximation of the Haverkamp quasi-exact implicit model to water-repellent soils. We then tested the new method using analytical data. For highly water-repellent soils, the proposed method had better performance when estimating soil sorptivity (S) and soil saturated conductivity (Ks), with respective errors of less than 1.5 % and 8 %, compared to relative errors of more than 10 % and 30 % with the two-term BEST-WR method. We also tested both approaches with experimental data. The two methods provided similar estimates for hydraulic parameters, with linear correlations between methods of R2 = 0.84 for S and R2 = 0.88 for Ks. Initial infiltration was not well modeled by either the two-term or three-term model for 33 tests, thus revealing limitations in the applied exponential model that we used to account for soil repellency. AimTo identify and quantify potential determinants of antimicrobial prescribing behaviour, using the Theoretical Domains Framework (TDF). MethodsA cross-sectional survey involving doctors (∼4000) and pharmacists (∼400) working within Hamad Medical Corporation (HMC), Qatar. An online questionnaire, developed with reference to the TDF, included: personal and practice demographics, and Likert statements on potential determinants of antimicrobial prescribing practice. Analysis included principal component analysis (PCA), descriptive and inferential statistics. ResultsIn total, 535 responses were received, 339 (63.4%) from doctors. Respondents were predominantly male, 346 (64.7%). Just over half (N = 285, 53.3%) had ≤5 years' experience. PCA showed a three-component (C) solution: ‘Guidelines compliance’ (C1), ‘Influences on practice’ (C2) and ‘Self-efficacy’ (C3). The scales derived for each component had high internal consistency (Cronbach's alphas >0.7), indicating statistical appropriateness for developing scales. Respondents generally scored highly for ‘Guidelines compliance’ and ‘Self-efficacy’. The lowest levels of positive scores were in relation to the items within the ‘Influences on practice’ component, with particular focus on TDF domains, environmental context and resources, and social influences. Inferential analysis comparing component scores across demographic characteristics showed that doctors, the more qualified and those with greater experience, were more likely to be positive in responses. ConclusionsThis study has identified that environmental context and resources, and social influences, with an emphasis on pharmacists and early career clinicians, may be useful targets for behaviour change interventions to improve clinicians' antimicrobial prescribing, thereby reducing antimicrobial resistance rates. Such interventions should focus on appropriate linked behaviour-change techniques. arine wild-capture fisheries depend on the capacity of the ocean to provide a flow of harvestable resources to sustain the industry. Paradoxically, conventional fishing often undermines these resources by degrading the environment and overexploiting fish stocks. Many UK fisheries have declined for over a century due to a biased focus on their social-economic value and lack of recognition that they are social-ecological systems and need to be managed as such. With the UK’s recent transition to an independent coastal state, the Fisheries Act (2020) and associated Joint Fisheries Statement provide an opportunity to correct this. Focusing on the ecological foun- dations, a more sustainable future for UK fisheries may be achieved by: (1) implementing a conservative quota setting system based on Maximum Sustainable Yield (MSY), defined as that which would occur when the biomass of a population of the target species is at 50% of that estimated at carrying capacity, to set catch limits rather than targets. The biomass of fish stocks should be allowed to regenerate to a minimum of 120% of that which will achieve MSY to provide a buffer against the uncertainty in ecological response to climate change. (2) Fishing capacity should be reduced while redistributing a greater share of the quota to sectors of the fleet that are demonstrably more sustainable; recognising that short term compensation may be required by some to mitigate the impacts of displaced activity until the benefits of stock recovery are realised. (3) Greater restrictions should be applied to ensure the most damaging fishing techniques (e.g. bottom trawling and dredging) are prohibited as appropriate in the network of marine protected areas. Protection should be enforced to promote the regeneration of degraded habitats and restoration of fish populations to help achieve the objectives as set out in the Act.

Background The impact of COVID-19 on physical and mental health and employment after hospitalisation with acute disease is not well understood. The aim of this study was to determine the effects of COVID-19-related hospitalisation on health and employment, to identify factors associated with recovery, and to describe recovery phenotypes. Methods The Post-hospitalisation COVID-19 study (PHOSP-COVID) is a multicentre, long-term follow-up study of adults (aged ≥18 years) discharged from hospital in the UK with a clinical diagnosis of COVID-19, involving an assessment between 2 and 7 months after discharge, including detailed recording of symptoms, and physiological and biochemical testing. Multivariable logistic regression was done for the primary outcome of patient-perceived recovery, with age, sex, ethnicity, body-mass index, comorbidities, and severity of acute illness as covariates. A post-hoc cluster analysis of outcomes for breathlessness, fatigue, mental health, cognitive impairment, and physical performance was done using the clustering large applications k-medoids approach. The study is registered on the ISRCTN Registry (ISRCTN10980107). Findings We report findings for 1077 patients discharged from hospital between March 5 and Nov 30, 2020, who underwent assessment at a median of 5·9 months (IQR 4·9–6·5) after discharge. Participants had a mean age of 58 years (SD 13); 384 (36%) were female, 710 (69%) were of white ethnicity, 288 (27%) had received mechanical ventilation, and 540 (50%) had at least two comorbidities. At follow-up, only 239 (29%) of 830 participants felt fully recovered, 158 (20%) of 806 had a new disability (assessed by the Washington Group Short Set on Functioning), and 124 (19%) of 641 experienced a health-related change in occupation. Factors associated with not recovering were female sex, middle age (40–59 years), two or more comorbidities, and more severe acute illness. The magnitude of the persistent health burden was substantial but only weakly associated with the severity of acute illness. Four clusters were identified with different severities of mental and physical health impairment (n=767): very severe (131 patients, 17%), severe (159, 21%), moderate along with cognitive impairment (127, 17%), and mild (350, 46%). Of the outcomes used in the cluster analysis, all were closely related except for cognitive impairment. Three (3%) of 113 patients in the very severe cluster, nine (7%) of 129 in the severe cluster, 36 (36%) of 99 in the moderate cluster, and 114 (43%) of 267 in the mild cluster reported feeling fully recovered. Persistently elevated serum C-reactive protein was positively associated with cluster severity. Interpretation We identified factors related to not recovering after hospital admission with COVID-19 at 6 months after discharge (eg, female sex, middle age, two or more comorbidities, and more acute severe illness), and four different recovery phenotypes. The severity of physical and mental health impairments were closely related, whereas cognitive health impairments were independent. In clinical care, a proactive approach is needed across the acute severity spectrum, with interdisciplinary working, wide access to COVID-19 holistic clinical services, and the potential to stratify care. Background Evidence from animal models and observational epidemiology points to a role for chronic inflammation, in which interleukin 6 (IL-6) is a key player, in the pathophysiology of type 2 diabetes (T2D). However, it is unknown whether IL-6 mediated inflammation is implicated in the pathophysiology of T2D. Methods We performed a meta-analysis of 15 prospective studies to investigate associations between IL-6 levels and incident T2D including 5,421 cases and 31,562 non-cases. We also estimated the association of a loss-of-function missense variant (Asp358Ala) in the IL-6 receptor gene (IL6R), previously shown to mimic the effects of IL-6R inhibition, in a large trans-ethnic meta-analysis of six T2D case-control studies including 260,614 cases and 1,350,640 controls. Findings In a meta-analysis of 15 prospective studies, higher levels of IL-6 (per log pg/mL) were significantly associated with a higher risk of incident T2D (1·24 95% CI, 1·17, 1·32; P = 1 × 10−12). In a trans-ethnic meta-analysis of 260,614 cases and 1,350,640 controls, the IL6R Asp358Ala missense variant was associated with lower odds of T2D (OR, 0·98; 95% CI, 0·97, 0·99; P = 2 × 10−7). This association was not due to diagnostic misclassification and was consistent across ethnic groups. IL-6 levels mediated up to 5% of the association between higher body mass index and T2D. Interpretation Large-scale human prospective and genetic data provide evidence that IL-6 mediated inflammation is implicated in the etiology of T2D but suggest that the impact of this pathway on disease risk in the general population is likely to be small. The dataset presented here supports the research paper entitled "A calculator to quantify cover crop effects on soil health and productivity". Soil health (sometimes used synonymously with soil quality) is a concept that describes soil as a living system to sustain plants, animals, and human. Soil physical, chemical, and biological properties, along with their interactions, are required to quantify soil health. The use of cover crops in agricultural rotations may enhance soil health, yet there has been little progress in understanding how external factors such as climate, soil type, and agronomic practices affect soil and cash crop responses. In response, this dataset compiles measurements from 281 studies and provides an analysis of field-measured changes in 38 soil health indicators due to cover crop usage. Environmental and background indicators were also compiled to assess how climatic and management practices affect soil and cash crop responses to cover crops, with specific categories including climate type (tropical, arid, temperate, and continental), soil texture (coarse, medium, and fine), cover crop type (legume, grass, multi-species mixture, and other), and cash crop type (corn, soybean, wheat, vegetable, corn-soybean rotation, corn-soybean-wheat rotation, and other). An unbalanced analysis of variation was used to determine the hierarchy of most to least important factors that affected responsiveness of each soil health indicator. Based on the hierarchy structure, a soil health calculator was then developed to quantify the response of 13 parameters - erosion, runoff, weed suppression, soil aggregate stability, leaching, infiltration, microbial biomass carbon, soil bulk density, soil organic carbon, soil nitrogen, microbial biomass nitrogen, cash crop yield, and saturated hydraulic conductivity - to cover crops. The presented data in the calculator report the mean change in parameter values based on all combinations of climate, soil texture, cover crop type, and cash crop type. (c) 2020 The Author(s). Background: Evidence of the association between daily variation in air pollution and risk of stroke is inconsistent, potentially due to the heterogeneity in stroke etiology. Objectives: To estimate the associations between daily variation in ambient air pollution and risk of stroke and its subtypes among participants of the Women's Health Initiative, a large prospective cohort study in the United States. Methods: We used national-scale, log-normal ordinary kriging models to estimate daily concentrations of fine particulate matter (PM2.5), respirable particulate matter (PM10), nitrogen dioxide (NO2), nitrogen oxides (NOx), sulphur dioxide, and ozone at participant addresses. Stroke was adjudicated by trained neurologists and classified as ischemic or hemorrhagic. Ischemic strokes were further classified according to the Trial of ORG 10172 in Acute Stroke Treatment (TOAST) classification. We used a time-stratified case-crossover approach to estimate the odds ratio (OR) of the risk of stroke associated with an interquartile range (IQR) increase in concentrations of each air pollutant. We performed stratified analysis to examine whether associations varied across subgroups defined by age at stroke onset, US census region, smoking status, body mass index, and prior history of diabetes mellitus, hypertension, heart or circulation problems, or arterial fibrillation at enrollment. Results: Among 5417 confirmed strokes between 1993 and 2012, 4300 (79.4%) were classified as ischemic and 924 (17.1%) as hemorrhagic. No association was observed between day-to-day variation in any pollutant and risk of total stroke, ischemic stroke, or specific etiologies of ischemic stroke. We observed a positive association between risk of hemorrhagic stroke and NO2 and NOx in the 3 days prior to stroke with OR of 1.24 (95% CI: 1.01, 1.52) and 1.18 (95% CI: 1.03, 1.34) per IQR increase, respectively. The observed associations with hemorrhagic stroke were more pronounced among non-obese participants. Conclusions: In this large cohort of post-menopausal US women, daily NO2 and NOx were associated with higher risk of hemorrhagic stroke, but ambient levels of four other air pollutants were not associated with higher risk of total stroke, ischemic stroke, or ischemic stroke subtypes. Objective To examine changes in outcome by the Leibovich score using contemporary and historic cohorts of patients presenting with renal cell carcinoma (RCC) Patients and Methods Prospective observational multicenter cohort study, recruiting patients with suspected newly diagnosed RCC. A historical cohort of patients was examined for comparison. Metastasis-free survival (MFS) formed the primary outcome measure. Model discrimination and calibration were evaluated using Cox proportional hazard regression and the Kaplan-Meier method. Overall performance of the Leibovich model was assessed by estimating explained variation. Results Seven hundred and six patients were recruited between 2011 and 2014 and RCC confirmed in 608 (86%) patients. Application of the Leibovich score to patients with localized clear cell RCC in this contemporary cohort demonstrated good model discrimination (c-index = 0.77) but suboptimal calibration, with improved MFS for intermediate- and high-risk patients (5-year MFS 85% and 50%, respectively) compared to the original Leibovich cohort (74% and 31%) and a historic (1998-2006) UK cohort (76% and 37%). The proportion of variation in outcome explained by the model is low and has declined over time (28% historic vs 22% contemporary UK cohort). Conclusion Prognostic models are widely employed in patients with localized RCC to guide surveillance intensity and clinical trial selection. However, the majority of the variation in outcome remains unexplained by the Leibovich model and, over time, MFS rates among intermediate- and high-risk classified patients have altered. These findings are likely to have implications for all such models used in this setting.

Background Major disparities between women and men in the diagnosis, management, and outcomes of acute coronary syndrome are well recognized. Objectives The aim of this study was to evaluate the impact of implementing a high-sensitivity cardiac troponin I assay with sex-specific diagnostic thresholds for myocardial infarction in women and men with suspected acute coronary syndrome. Methods Consecutive patients with suspected acute coronary syndrome were enrolled in a stepped-wedge, cluster-randomized controlled trial across 10 hospitals. Myocardial injury was defined as high-sensitivity cardiac troponin I concentration >99th centile of 16 ng/l in women and 34 ng/l in men. The primary outcome was recurrent myocardial infarction or cardiovascular death at 1 year. Results A total of 48,282 patients (47% women) were included. Use of the high-sensitivity cardiac troponin I assay with sex-specific thresholds increased myocardial injury in women by 42% and in men by 6%. Following implementation, women with myocardial injury remained less likely than men to undergo coronary revascularization (15% vs. 34%) and to receive dual antiplatelet (26% vs. 43%), statin (16% vs. 26%), or other preventive therapies (p, Central Illustration

The WUDAPT (World Urban Database and Access Portal Tools project goal is to capture consistent information on urban form and function for cities worldwide that can support urban weather, climate, hydrology and air quality modeling. These data are provided as urban canopy parameters (UCPs) as used by weather, climate and air quality models to simulate the effects of urban surfaces on the overlying atmosphere. Information is stored with different levels of detail (LOD). With higher LOD greater spatial precision is provided. At the lowest LOD, Local Climate Zones (LCZ) with nominal UCP ranges is provided (order 100 m or more). To describe the spatial heterogeneity present in cities with great specificity at different urban scales we introduce the Digital Synthetic City (DSC) tool to generate UCPs at any desired scale meeting the fit-for-purpose goal of WUDAPT. 3D building and road elements of entire city landscapes are simulated based on readily available data. Comparisons with real-world urban data are very encouraging. It is customized (C-DSC) to incorporate each city's unique building morphologies based on unique types, variations and spatial distribution of building typologies, architecture features, construction materials and distribution of green and pervious surfaces. The C-DSC uses crowdsourcing methods and sampling within city Testbeds from around the world. UCP data can be computed from synthetic images at selected grid sizes and stored such that the coded string provides UCP values for individual grid cells.

Despite great interest, two randomised controlled trials (RCTs) of cytoreductive nephrectomy in the tyrosine kinase inhibitor setting in metastatic renal cell carcinoma have either closed early (SURTIME) or are recruiting very slowly (CARMENA) after 7 yr. Challenges in RCT delivery in uro-oncologic surgery are many. Multiple steps are needed to ensure strong recruitment to trials addressing important urologic cancer questions. Feasibility/pilot studies are key stepping stones towards successful delivery of surgical RCTs.

Chiral alcohol synthesis by enzymatic carbonyl reduction has become a mainstream methodology, particularly in industry. Collections of alcohol dehydrogenases are commercially available, which provide simple starting points for process development. Genome sequence data now provide an enormous range of potentially useful enzymes, although identifying the subset with the desired substrate-, regio- and stereoselectivities still requires significant effort. Directed evolution offers an alternative; although many advances have been made, such projects still remain resource-intensive. If catalyst discovery, catalyst improvement, and process development can become more streamlined, alcohol dehydrogenases can play an even greater role in the future of chiral organic synthesis.

Intraspecific antagonistic coevolution, also called a genetic arms race, occurs when the evolutionary interests of individuals within a population diverge from one another. While arms races can occur between individuals of the same sex, siblings, or parents and their offspring, here we discuss a sample of the myriad of antagonistic interactions that result from the conflict between the evolutionary interests of males and females. Unlike other forms of coevolution, an intraspecific arms race can have no ‘winner.’ Yet, because of the tethered fate that males and females share, the adaptations that drive the arms race can both lead to population extinction as well as promote speciation.

This chapter defines the scope and limits of the book along with its focus on using enzymes for preparative synthesis. Enzymes are highly selective catalysts, and their contributions to chemo-, regio-, and stereoselectivities are described. Commonly required cofactors are briefly described along with relevant aspects of enzyme kinetics. A brief survey of common reaction types in biocatalysis covers both the reaction and associated enzyme types; a comprehensive table is provided as well. The various sources of biocatalysts (animals, plants, and microorganisms) are discussed along with their relative advantages and disadvantages. The uses of soluble versus insoluble enzyme forms are introduced and some benefits of each strategy are cited.

Global landings of scallops have grown dramatically in recent decades and these fisheries are now among the most lucrative in several countries around the world. Despite this apparent success story, concerns have arisen about the wider ecosystem effects of scallop fisheries. This is particularly the case for the most common type of fisheries that use dredges to rake scallops off the seafloor. Here the evidence for negative effects arising from this practice is reviewed and suggestions offered as to ways in which scallop fisheries might be better managed. In general, dredging causes loss of biodiversity and reduces the complexity of benthic habitats by flattening substrates and removing structurally complex species such as hydroids, bryozoans and seaweeds. This is significant because such habitats are key nursery and feeding areas for a wide range of species, including commercially important fish and shellfish. Scallop dredging also catches a variety of more mobile species such as crustaceans, echinoderms, fishes, and in certain areas, sea turtles, which is clearly of concern. Despite these general rules, the magnitude of effects varies considerably in different habitats. The most severe are in biogenic reefs such as formed by maerl and mussels, so there is a strong argument for fully protecting such areas. Reef and cobble habitats also appear relatively susceptible, but soft sediments such as sand, mud and gravel (which are the focus of most scallop fisheries) appear more resilient, particularly in areas adapted to high levels of natural disturbance. Determining the full effects of dredging remains difficult, however, because most fishing grounds have been exploited for decades, long before scientific study began. Long-term protected areas are beginning to provide insights into the recovery and composition of benthic communities in the absence of dredging. Continued study of these areas will be a key to gaining a better understanding in the future. In terms of reducing the ecosystem effects of dredging, an approach that combines effort control, gear modifications, and spatial management is suggested. Spatial management is showing great promise where it has been applied as it can offer a win–win scenario, which protects vulnerable habitats while boosting scallop stocks by providing breeding and nursery refuges; however, spatial management must be carefully planned to maximise biological benefits while accounting for socio-economic factors. Scallop fisheries must also be managed in unison with other fisheries in order to restore diversity and resilience to oceans facing an uncertain future of climate change and growing anthropogenic pressure.

This chapter summarizes the future of biocatalysis, focusing particularly on the short- and long-term needs for growth in this field. The former category includes issues such as new types of enzyme-catalyzed reactions and their development into viable synthetic processes, catalytic promiscuity facilitating nonnative reactions of enzymes, computational enzyme design and hybrid biocatalysts that combine enzymatic and organometallic strategies. Long-term issues include the need for increased supplies of synthetically useful enzymes with very high consistencies vital for commercial applications, a need for more process and engineering data, and more rapid enzyme-improvement strategies that yield enzymes with better properties avoiding the need for individual campaigns for each new substrate.

Threonine aldolases catalyze the pyridoxal phosphate-dependent condensation between small amino acids (principally glycine) and aldehydes such as acetaldehyde. Carbon–carbon bond formation involves forming two adjacent chiral centers. As a rule, threonine aldolases are very stereoselective for α-carbon configuration but show modest selectivity at the β-carbon. On the other hand, these enzymes accept a wide variety of synthetically useful acceptor aldehydes, making them important additions to the synthetic toolkit. This review briefly summarizes the reaction mechanism and then lists all published synthetic reactions by threonine aldolases as of early 2014. The current state of the art in crystallographic and protein engineering studies of these enzymes is also presented.

Computational models of the natural hip joint are needed to examine and optimise tissue sparing interventions where the natural cartilage remains part of the bearing surfaces. Although the importance of interstitial fluid pressurisation in the performance of cartilage has long been recognized, few studies have investigated the time dependent interstitial fluid pressurisation in a three dimensional natural hip joint model. The primary aim of this study was to develop a finite element model of the natural hip incorporating the biphasic cartilage layers that was capable of simulating the joint response over a prolonged physiological loading period. An initial set of sensitivity studies were also undertaken to investigate the influence of hip size, clearance, cartilage properties, thickness and hemiarthroplasty on the contact mechanics of the joint. The contact stress, contact area, fluid pressure and fluid support ratio were calculated and cross-compared between models with different parameters to evaluate their influence. It was found that the model predictions for the period soon after loading were sensitive to the hip size, clearance, cartilage aggregate modulus, thickness and hemiarthroplasty, while the time dependent behaviour over 3000s was influenced by the hip clearance and cartilage aggregate modulus, permeability, thickness and hemiarthroplasty. The modelling methods developed in this study provide a basic platform for biphasic simulation of the whole hip joint onto which more sophisticated material models or other input parameters could be added in the future.

Yellowtail kingfish (Seriola lalandi; YTK) hatchery technology has been largely developed in New Zealand and Australia where a burgeoning seacage grow-out industry exists. Wild-caught broodstock can be maintained in land-based tanks and induced to spawn using hormone therapy or more commonly will spawn spontaneously within one to two breeding seasons of domestication using phototherm manipulation. The chapter looks at seriola aquaculture, detailing broodstock management and larviculture. Two peaks of mortality have been found to occur – one at 3–4 dph when larvae commence feeding, and a second caused by cannibalism which occurs when larvae are 6–10 mm total length – and the chapter discusses strategies for mitigating these problems. The major bottlenecks to YTK juvenile production have included low survival and high rates of deformity, and the chapter finishes by looking at how commercial and government research institutes are addressing these.
