Cyanide poisoning remains a significant concern for both civilian and military personnel worldwide. Rhodanese, a cyanide detoxify enzyme produced by the wild (KOJCM1665) and two selected mutant strains (KOJCM1665c and KOJCM1665d) of Klebsiella oxytoca JCM 1665 was purified and characterized using a combination of standard techniques. Extracellular rhodanese yield of the wild and selected mutants were 6.2 ± 0.6, 26.7 ± 0.7 and 18.3 ± 0.3 U/mL respectively. The overall levels of recovery of rhodanese activity and fold after purification were 49 %, 66 %, and 58 % and 2.6, 2.6, and 2.3 % for the wild, KOJCM 1665c, and KOJCM 1665d, respectively. The native molecular weights of the three enzyme preparations were 35.1, 34.9, and 34.8 kDa, while the subunit molecular weight was 35 kDa for all the enzymes. The optimum activity of the enzymes was observed at 50 °C and pH 6.0. The real km of the three enzyme preparations for KCN as a substrate were 1.03 × 10−3, 0.95 × 10−3, and 0.80 × 10−3 M, respectively, while the km for the second substrate (Na2S2O3) were 0.59 × 10−3, 0.49 × 10−3, and 0.69 × 10−3 M, respectively. The substrate specificity study showed that the enzyme preferred sodium thiosulfate as the substrate. Metal ions such as Na+ and K+ had significantly greater inhibitory effects on enzyme activity. This study demonstrated the potential of enhancing K. oxytoca JCM 1665 to overexpress extracellular rhodanese, and the physicochemical properties of the enzyme hold significant promise as a target for improving the efficiency of cyanide bioremediation processes.

Toxic effluents from cassava processing mills through leaching and percolation are transported into groundwater, which may be detrimental to the health of humans when consumed. Our knowledge on how these toxic effluents are transported into groundwater is limited and has not received the needed attention. The objective of the study is to stimulate groundwater flow and migration pattern of cassava effluents, stimulate the impact of recharge and discharge on the fate of cassava plume migration and, lastly to suggest a remediation techniques. Groundwater modelling system (GMS) software was used for the stimulation of migration of cyanide from the surface into groundwater. The results revealed that transport of Cyanide is considerably influenced by the subsurface geology of the unsaturated zone, groundwater recharge, pumping and dispersion rates, respectively. In a well at the source, a cyanide mass of 3.5 × 105 mg was observed under increased recharge conditions for 7300 days and 7.6 × 104 mg when reduced. Increased pumping rate for 7300 days, cyanide mass of 8.8 × 104 mg was detected and it was 2.6 × 105 mg when reduced. Increased dispersion results in 2.3 × 105 mg and 3.1 × 105 mg when reduced. Of these parameters simulated, recharge has the greatest impact on the transport of cyanide. The effectiveness of using MODFLOW and MT3DMS to understand the way cyanide is transported into groundwater is fully demonstrated. The pump-and treat remediation types is recommended for groundwater resource management and planning.

Cyanide (CN) is a serious concern in industrial and goldmine wastewater. Strict regulatory standards have been established by various agencies due to the detrimental effects that CN has on human health. Therefore, before discharge to water bodies or land, it is essential to create a sustainable model for the safe removal of CN. Carbon-based materials are well known for their adsorption and oxidation features, which can be conducive to CN removal. This paper reviews the relevant literature on the application of modified and unmodified carbon-based materials to CN removal in water; these materials include activated carbon (AC), graphene, graphene oxide (GO), and carbon nanotubes (CNTs). Moreover, CN removal mechanisms and photocatalytic removal of CN are comprehensively discussed, with a particular emphasis on modifying carbon-based materials. It has been observed that adding various elements to carbon-based materials improves their surface area, functional groups, CN adsorption capacity, and pore volume. Impacts of operational parameters, isotherm models, kinetics, and types of carbon-based materials are also outlined. This study provides insight into the real-scale applicability of carbon-based materials for CN removal from waters. Moreover, this review indicates that essential work on CN removal using carbon-based materials is still needed. Future research should focus on developing modified carbon-based materials to encourage multidisciplinary research. The most crucial gap in the literature is that the studies have been performed on a lab scale. Therefore, further pilot and real-scale applications should be conducted. Overall, the cost assessment, environmental effects, and human health risks of carbon-based materials should be studied in future research to achieve a realistic perspective on applicability on an industrial scale.

Background: Recently, phytomedicine have been applied in the management of various diseases related to toxicity. In addition, attention has been on cyanide toxicity due to its harmful effects on man and animals. Thus, this research was carried out to determine the effect of Acalypha wilkesiana methanol leaf extract on cyanide induced renal damage in rats. Methods: 45 female Wister rats randomly divided into 7 groups were used for this study. Group 1:received neither cyanide nor methanol fraction of Acalypha wilkesiana (MFAW), 2 and 3 received different doses of MFAW respectively, 4 received cyanide only, 5 Rats received cyanide and sodium thiosulphate. 6 and 7 received cyanide and different doses of MFAW respectively. Renal markers (urea, creatinine, sodium, bicarbonate, potassium and chloride electrolytes) and antioxidant status (superoxide dismutase, malonaldehyde, catalase, gluthathione peroxidase) were assessed in the kidneys using standard biochemical procedures. Plants face multiple challenges from environmental pollutants and higher emissions of atmospheric CO 2 . Therefore, a hydroponic-based experiment was used to explore the combined effects of elevated [CO 2 ] (700 ppm) and exogenous cyanide (CN - ) (3.0 mg CN/L) on rice seedlings under nitrogen deficiency, utilizing metabonomic and transcriptomic analysis. Elevated [CO 2 ] significantly improved the growth of CN - -treated rice seedlings compared to those with ambient [CO 2 ] (350 ppm), and it also significantly affected CN - assimilation. Transcriptome analysis revealed distinct impacts on differentially expressed genes (DEGs) across treatments and tissues. KEGG analysis showed variability in DEGs enriched in amino acid (AA) and energy metabolism pathways due to elevated [CO 2 ] and CN - . Metabonomic indicated that higher input of [CO 2 ] and exogenous CN - more severely impacted energy metabolism elements than the individual species of AAs. Positive synergistic effects of elevated [CO 2 ] and CN - were observed for glutamine and asparagine in shoots, and methionine in roots, wherein negative effects were noted for phenylalanine in shoots, and phenylalanine, valine, and alanine in roots. Meanwhile, positive effects on fumarate in shoots and α-ketoglutarate and succinate in roots were also found. Overall, elevated [CO 2 ] enhanced growth in CN - -treated rice seedlings under nitrogen deficiency by altering AA and energy metabolism. The present study focuses on the analysis of the diffusion process of various cyanide compounds through general porins and outer membranes of gram-negative bacteria. We demonstrate the impact of the compound-to-porin radius ratio, the charge of cyanide ion, the Donnan potential, the intrinsic porin potential, the number and length of general porins, the fraction of open channels, and the size of bacteria on the effective diffusion coefficients and permeability coefficients of cyanide compounds. Moreover, we report, for the first time, the procedure for comparison of the rate of cyanide diffusion across the outer membrane with the rate of cyanide biodegradation that allows establishing the conditions for which the biodegradation is a diffusion-limited process or the diffusion is a significantly faster process than biodegradation. We apply this procedure to several experimental studies and predict the range of extracellular cyanide concentrations for which diffusion is a significantly faster process than biodegradation. Gold cyanidation facilities in the Arequipa Region of Peru are challenged by the availability and quality of water for processing in an arid environment. The facilities reuse decant water which recycles residual cyanide but also undesirable constituents. To understand the impact of intensive water recycling on cyanide and metals concentrations, we collected barren water, decant water, and tailings samples from six gold cyanidation facilities with ore capacities of 10-430 tons per day. Processing facilities in Arequipa recycle all effluents, with decant waters making up 58 ± 11 % of process waters. Decant water contained non-target metals: copper (394 ± 161 mg/L), iron (59 ± 34 mg/L), and zinc (74 ± 42 mg/L). In addition, decant water mean free and complexed cyanide concentrations were 534 ± 129 mg/L and 805 ± 297 mg/L, respectively. Complexed cyanide concentrations remained more constant than free cyanide concentrations with 786 ± 299 mg/L for barren water and 805 ± 297 mg/L for decant water. Cyanide mass balances showed between 21 % and 42 % of unaccounted free cyanide from the start of gold cyanidation and discharge to the tailings storage facility (TSF). Free cyanide estimated losses due to volatilization were 0.8 kg and 2.5 kg of hydrogen cyanide per ton of ore processed at barren water pH of 10.1 and 9.7. Together these results indicate two acute hazards: 1) volatilization of free cyanide during processing and 2) loading and retention of cyanides and metals into TSFs. This study elucidates the extent of uncontrolled vapor phase cyanide release during gold processing operation and contaminant concentrations in the tailings storage facilities. Background: As cyanide (CN - ) is a significant hazard to the environment and human health, it is essential to monitor cyanide levels in water and food samples. Moreover, real-time visualization of CN - could provide an additional understanding of its critical physiological and toxicological roles in living cells. The fluorescence approach based on small organic probes is an effective way for the detection of CN - . In this approach, a triphenylamine-xhantane conjugate was applied to detect in many samples such as sewage water, soil, sprouted potato, apricot seed, and living cells., Results: We report a new ratiometric near-infrared fluorescent probe based on a triphenylamine-xhantane derivative for CN - sensing in many samples. The probe displays high selectivity for only CN - ions among a series of analytes. The addition of cyanide to the dicyanovinyl moiety of the probe disrupts π-conjugation followed by the interruption of internal charge transfer. Consequently, the emission peak of the probe shifts hypsochromically from 655 to 495 nm. There is a linear correlation between the emission intensity (I 495 ) and cyanide level, with a detection limit of 0.036 μM. The probe has many advantages over many probes, such as NIR fluorescence, ratiometric response, low cytotoxicity (85.0 % cell viability up to 50.0 μM of the probe), good membrane permeability, fast response time (4.0 min), high selectivity, good photostability, and anti-interference capability., Significance: Although various probes have been reported in the literature, the use of triphenylamine-xhantane unit as CN - probe has yet to be explored. The probe can detect trace levels of cyanide in many samples such as sewage water, soil, sprouted potatoes, and apricot seeds. Given that mining is considered to be an essential activity for Mexico's industrial development, cyanide has been increasingly used to recover precious metals such as gold and silver. Along with that arises the need to develop new technologies to treat the wastes (mining tailings). In addition to their high cyanide content, metal and other contaminants that are found in tailings also present a problem. As a result, conventional (physicochemical) strategies have been developed to reduce contamination from tailings, nonetheless, these have high operating costs and generate unwanted by-products. For this reason, studies have begun to focus on non-conventional strategies to treat free cyanide and cyanide complexes such as fungi, bacterial consortia, and pure bacteria. These are important because of the mechanisms involved in degrading or modifying contaminants at neutral to high pH levels, which convert contaminants into non-hazardous products. The ability of microorganisms to grow at an alkaline pH prevents HCN volatilization. These studies have been performed at the laboratory level using two types of microbial binding: suspended biomass and immobilized biomass. They have used both natural (granite rock, citrus peels, cellulose, gravel) and synthetic (stainless steel, geotextiles, alginate, plastics) packing material, as well as reactors with different types of flow, namely, batch and continuous.

Among all kinds of chemical warfare agents, only cyanide and nerve agents can cause massive mortality at low concentrations. In this work, a dual-channel fluorescent probe CWAs-Thia capable of detecting cyanide and nerve agents is presented. The two reactive recognition units, pyridine and the thiazole-2-carbonyl group, of the probe for cyanide and nerve agents, respectively, produced red and blue fluorescent responses, respectively, which were attributed to excited-state intramolecular proton transfer and intramolecular charge transfer. CWAs-Thia is the first probe that can selectively recognize cyanide and nerve agent. Most frequently used biological method was improved anaerobic/anoxic/aerobic (A/A/O) process for treating actual pesticide wastewater presently. However, the effluent results were far away from what the national standard expects due to pesticide wastewater characteristics of high COD concentration, high total nitrogen (TN) concentration and high toxicity. In this study, a novel two-phase anaerobic/aerobic/integrated deoxygenated and anoxic reactor associated with membrane process (P1) were regarded as excellent candidates for the treatment of pesticide wastewater, compared the removal efficiency of pollutants with improved A/A/O process (P2) under different hydraulic retention times (HRTs). Effluent ethylene thiourea (ETU) concentration was reduced effectively by 67.1 % in P1. The average cyanide (CN - ) effluent concentration in P1 and P2 was 0.40 mg/L and 6.67 mg/L, respectively. During the entire HRT change period, P1 significantly increased the biological oxygen demand (BOD 5 ) removal efficiency by 12.98 %. The average effluent COD concentration of P1 was 36.41 mg/L, while that of P2 was 984.42 mg/L. In addition, TN removal efficiency of P1 exhibited distinct superiority. When HRT was 72 h, the average TN effluent concentration of P1 and P2 was 12.67 mg/L and 67.66 mg/L. The sludge performance indicators have been determined and results showed that the sludge viscosity of the membrane reactor in P1 had merely 32.6 % higher than that of the secondary sedimentation tank in P2, and the average vertical displacement of sludge settleability in P1 was 23.0 % slight lower than that of P2. The overall sludge performance proved that higher sludge concentration was allowed in P1. Moreover, experiment improved nitrite reductase (NIR) and nitrate reductase (NAR) in P1 were less susceptible to be suppressed by ETU and CN - increase compared with P2. Ribonuclease A is a major hydrolyzing enzyme involved in the hydrolysis of RNA. The crystals of bovine pancreatic RNase A (bpRNase A) were grown at pH 5.5. The effect of sodium cyanide on bpRNase A was assessed by adding it directly to the crystal containing well. Treating the crystals of bpRNase A with sodium cyanide resulted in the displacement of the sulfate ion from the active site of bpRNase A, while the additional sulfate ion, bound to Ala-4, remained unaffected. The addition of sodium cyanide to bpRNase A crystals did not show change in the secondary structure elements of the enzyme. The objective of this study was to compare the use of hydroxocobalamin (B12a) and a succinate prodrug to evaluate for improvement in mitochondrial function in an in vitro model of cyanide poisoning. Peripheral blood mononuclear cells (PBMC) and human aortic smooth muscle cells (HASMC) incubated with 50 mM of sodium cyanide (CN) for five minutes serving as the CN group compared to controls. We investigated the following: (1) Mitochondrial respiration; (2) Superoxide and mitochondrial membrane potential with microscopy; (3) Citrate synthase protein expression. All experiments were performed with a cell concentration of 2−3 × 106 cells/ml for both PBMC and HASMC. There were four conditions: (1) Control (no exposure); (2) Cyanide (exposure only); (3) B12a (cyanide exposure followed by B12a treatment); (4) NV118 (cyanide followed by NV118 treatment). In this study the key findings include: (1) Improvement in key mitochondrial respiratory states with the succinate prodrug (NV118) but not B12a; (2) Attenuation of superoxide production with treatment of NV118 but not with B12a treatment; (3) The changes in respiration were not secondary to increased mitochondrial content as measured by citrate synthase; (4) The use of easily accessible human blood cells showed similar mitochondrial response to both cyanide and treatment to HASMC. The use of a succinate prodrug to circumvent partial CIV inhibition by cyanide with clear reversal of cellular respiration and superoxide production that was not attributed to changes in mitochondrial content not seen by the use of B12a.

We have determined the effects of azure B (AzB), the main demethylated metabolite of methylene blue (MB), on a model of lethal cyanide intoxication. Our rationale was the following: AzB 1- possesses redox properties very similar to those of MB, which is a potent cyanide antidote, 2- may present a higher intracellular diffusibility than MB, 3- is already present in commercially available solutions of MB, and 4- appears very quickly in the blood after MB administration. AzB could therefore be a member of the phenothiazium chromophore family of interest to treat cyanide intoxication. We found, in spontaneously breathing urethane sedated rats, that AzB mimicked the effects of MB by increasing metabolism, ventilation and cardiac contractility up to 30−40 mg/kg. AzB had a lethal toxicity when the dose of 60 mg/kg was reached. Doses of AzB were therefore chosen in keeping with these data and the doses of MB previously used against cyanide intoxication (4–20 mg/kg) in the rat – doses corresponding to those used in humans to treat methemoglobinemia. KCN, infused at the rate of 0.375 mg/kg/min iv for 13 min, was fatal within 15 min in 100 % of our un-anesthetized rats. AzB at the dose of 4 mg/kg (n = 5) or 10 mg/kg (n = 5) administered 3 min into cyanide infusion allowed 100 % of the animals to survive with no clinical sequelae. The onset of coma was also significantly delayed and no apnea or gasping occurred. At the dose of 20 mg/kg, AzB was much less effective.At 4 mg/kg, the antidotal effects of AzB were significantly better than those produced by MB at the same dose and were not different from the effects produced by 20 mg/kg MB. We conclude that AzB is a potent cyanide antidote at relatively low doses.

A novel organic probe (R) possessing multiple binding sites has rationally been designed, synthesized and characterized using NMR and mass spectral techniques. The probe R displays an excellent selective and sensitive response to CN– and SO32– ions over other anions. In aq. DMSO solution, R exhibits dual emission (463 and 590 nm) upon interaction with CN– ion, which is ascribed to the deprotonation of the NH group of the probe. Upon addition of SO32– ion, the enhancement of fluorescence for R at 478 nm is attributed to the formation of 1:1 adduct between R and SO32– ion by the nucleophilic addition of SO32– to C-atom of the ring CCH moiety. The fluorescence responses of R toward CN– and SO32– ions are pH independent in the 5 – 9 range. Limits of detection down to 28 and 3.7 nM were determined for CN– and SO32– quantitation, respectively. Theoretical calculations (DFT and TDDFT) are applied to interpret the absorption and dissimilar emission behaviour of the probe and its adduct with CN– and SO32– ions. The CN– induced dual fluorescence in R may be due to relaxation of the excited state to two different ground states.

Water used for domestic and irrigation purposes are sometime contaminated by organic and inorganic pollutants from cassava processing centres. Accordingly, groundwater from ten randomly wells were analyzed for pH, turbidity, total suspended solids (TSS), total dissolved solids (TDS), electrical conductivity (EC), sodium adsorption ratio (SAR), CN−, Na+, K+, Ca++, Mg++ and NO3− ion concentrations and the microbial content to determine the suitability of these waters for drinking and/or irrigation purposes. The reduction of CO 2 with low overpotential and high selectivity is a crucial challenge in catalysis. Fortunately, natural systems have evolved enzymes that achieve this catalytic reaction very efficiently at a complex nickel-iron-sulfur cluster within carbon monoxide dehydrogenase (CODH). Extensive biochemical, crystallographic, and spectroscopic work has been done to understand the structures and mechanism involved in the catalytic cycle, which are summarized here from the perspective of mechanistic organometallic chemistry. We highlight the ambiguities in the data and suggest experiments that could lead to clearer understanding of the mechanism and structures of intermediates at the active-site cluster. These include parallel crystallography and spectroscopy, as well as the preparation of synthetic analogues that help to interpret structural and spectroscopic signatures.

Polarity is a significant intracellular environmental parameter associated with cancer, while cyanide (CN - ) is known to be highly toxic to humans. In this work, we designed a dual-functional fluorescent probe (TPABT) for simultaneous detection of polarity and CN - . As a polarity sensor, the probe exhibits NIR emission at 766 nm in 1,4-dioxane (non-polar solvent), whose emission intensity is 71-fold stronger than that in water (polar solvent). Meanwhile, the fluorescence intensity and quantum yield are linearly related to solvent polarity, confirming the polarity response ability of TPABT. For cell polarity detection, low cytotoxicity and polarity sensitivity of probe enable the applications for differentiating cancer cells (HeLa, 4TI) from normal cells (HUV, 3 T3) and monitoring the polarity changes of 4TI cells. As a CN - sensor, TPABT displays a turn-on fluorescence at 640 nm upon the addition of CN - , with advantages of anti-interference, response in aqueous media and low detection limit (22 nM). Additionally, we further explored the practical applications of TPABT for CN - determination in three types of real water samples (drinking water, tap water and lake water) and living cells. Notably, TPABT responses to polarity and CN - in two independent fluorescence channels of 766 and 640 nm, respectively, ensuring the dual functions for polarity and CN - sensing. This research deepens the analysis of the mineral water footprint, especially that of gold, in regions that are understudied and where mining has been an intensified extractive activity since the colonial era, as is the case in the northern part of department of Cauca in Colombia. Thus, the purpose was to estimate the water footprint indicators in gold mining in Suárez (Cauca, Colombia), to quantify the impacts generated by the non-returned water in the production process and the levels of pollutants in the wastewater, aimed to strength public policies, control strategies and mitigation that generates reductions in the impacts from mining activities on the environment. The blue water footprint was estimated in 79.91 m3 per kg of gold extracted and the gray water footprint was found to be in the range of 272,125.39 to 404,825.11 m3 per kg of gold extracted. The water footprint values obtained were compared with other mines with similar operations. These results generate a baseline for decision making, providing elements for environmental strategic planning, regulations and showing the great environmental pressure that gold activity exerts on water resources and the territories.

Several causes of infertility have been identified, and several papers have documented some compounds that cause infertility. One of the compounds reported to be toxic to the reproductive system is cyanide. In the management of infertility, various mechanisms ranging from synthetic drugs, natural products and supplements have been employed. Quercetin is an antioxidant supplement that has been used in the treatment of a variety of ailments. This work is aimed at investigating the role of quercetin in attenuating spermato-toxicity and testicular-histopathology induced by cyanide.Seventy-two (72) male wistar rat (weight 190 g ± 10 g) were divided into nine groups (n = 8) except for groups 4 and 5 with (n = 16). Group 1 (control) received physiological saline while Groups 2 and 3 received 0.5 and 1 mg/kg body weight (bwt) cyanide respectively for 56 days, groups 4 and 5 received 0.5 and 1 mg/kg bwt cyanide respectively for 30 days. At day 30, eight animals were sacrificed from Groups 4 and 5 and the remaining eight (8) rats were subdivided into groups (6 and 7) and were given 20 and 40 mg/kg bwt of quercetin respectively for twenty-six days. Co-administration of cyanide and quercetin at a dose of 0.5 mg/kg cyanide +20 mg/kg quercetin and 1 mg/kg cyanide +40 mg/kg quercetin were given to group 8 and 9 respectively for 56 days.Significant decreases in sperm parameters (count, motile and normal sperm) and increases in malondiadehyde concentration were observed in the cyanide treated groups. Testicular histoarchitecture showed few to no spermatozoa in the lumen of rats treated with cyanide. All these effects were attenuated by quercetin.In conclusion, quercetin regulates testicular histopathology induced by cyanide in Wistar rats. Data from this work suggests potential preventive or therapeutic applications of quercetin for individuals subjected to cyanide environmental pollution.

Cyanide is among the ubiquitous chemicals that humans are usually exposed to and it is well documented that cyanide induces infertility in humans and experimental rodents. However, the pathogenesis remains unknown. Likewise, quercetin is an important nutraceutical that detoxifies reactive oxygen species, but its effects on testicular damage is not clear. The present study investigated the role of nutraceutical, quercetin on cyanide-induced testicular toxicity and probable involvement of cAMP-response-element modulator (CREM) which is a transcription factor necessary for the process of spermatogenesis. Thus, this work hypothesized that quercetin will mitigate endocrine dysfunction induced by cyanide. Seventy-two adult male Wistar rats were divided into seven groups (A to G). Groups A, B, C, F and G comprised of eight (8) rats per group while groups D and E comprised of sixteen (16) rats per group. Group A was designated as control while Groups B and C were given 0.5 and 1 mg/kg of cyanide respectively for 56 days. Group D and E received 0.5 and 1 mg/kg body weight cyanide respectively for 30 days. At day 30, eight animals were sacrificed from Group D and E and the remaining eight (8) rats were subdivided into sub-groups (D1 and E1) and were given 20 and 40 mg/kg of quercetin respectively for twenty-six (26) days. Group F and G were given concurrent administration of cyanide and quercetin at a dose of 0.5 + 20 mg/kg and 1 + 40 mg/kg respectively for 56 days. Body and testicular weight were significantly reduced in cyanide treated groups while quercetin modulates the reduction. Significant down-regulation of CREM gene and reduction in serum level of follicle stimulating hormone (FSH), Luteinizing hormone (LH), testosterone, glutathione peroxidase (GPx) and zinc in cyanide-treated groups, whereas administration of quercetin concomitantly with cyanide exposure or post-treated significantly reversed the alterations.

Background: Detecting and neutralizing Pd 2+ ions are a significant challenge due to their cytotoxicity, even at low concentrations. To address this issue, various chemosensors have been designed for advanced detection systems, offering simplicity and the potential to differentiate signals from different analytes. Nonetheless, these chemosensors often suffer from limited emission response and complex synthesis procedures. As a result, the tracking and quantification of residual palladium in biological systems and environments remain challenging tasks, with only a few chemosensing probes available for commercial use., Results: In this paper, a straightforward approach for the selective detection of Pd 2+ ions is proposed, which involves the design, synthesis, and utilization of a propargylated naphthalene-derived probe (E)-N'-((2-(prop-2-yn-1-yloxy)naphthalen-1-yl)methylene)benzohydrazide (NHP). The NHP probe exhibits sensitive dual-channel colorimetry and fluorescence Pd 2+ detection over other tested metal ions. The detection process is performed through a catalytic depropargylation reaction, followed by an excited state intramolecular proton transfer (ESIPT) process, the detection limit is as low as 11.58 × 10 -7  M under mild conditions. Interestingly, the resultant chemodosimeter adduct (E)-N'-((2-hydroxynaphthalen-1-yl)methylene)benzohydrazide (NHH) was employed for the consecutive detection of CN - ions, exhibiting an impressive detection limit of 31.79 × 10 -8  M. Validation of both detection processes was achieved through 1 H nuclear magnetic resonance and density functional theory calculations. For real-time applications of the NHP and NHH probes, smartphone-assisted detection, and intracellular detection of Pd 2+ and CN - ions within HeLa cells were studied., Significance: This research presents a novel naphthalene derivative for visually detecting environmentally toxic Pd 2+ and CN - ions. A series fluorescent probes (TBM-Cx (x = 1, 4, 8)) were designed based on embedding various alkoxy chains on the electron donor of triphenylamine (TPA)-based dicyanovinyl (MT) compound with an electron-deficient benzothiadiazole (BTD) for sensitive, selective, and visualizing detection of cyanide in aqueous solution. Due to the nucleophilic addition of CN - , the intramolecular charge transfer (ICT) of these probes was inhibited by the destroyed conjugated structure, exhibiting excellent "turn-on" fluorescence response toward cyanide anion (CN - ) in tetrahydrofuran (THF). However, the alkoxy chains with different lengths embedded in TPA not only enhance the sensitivity and solubility, but also regulate the emission behavior from ICT to aggregation-induced emission (AIE) characteristics. The binding mechanism and AIE sensing performances between the probes and CN - have been investigated and compared in THF/water mixture by spectral tools and theoretical calculations. The results showed that the ICT-based TBM-C1 probe with methoxy chain showed significantly turn-on fluorescence response to CN - as low as 0.077 μM in THF/water solution at high water fraction (90 %). The extraordinary accumulation of cyanide ions within biological cells is a severe health risk. Detecting and tracking toxic cyanide ions within these cells by simple and ultrasensitive methodologies are of immense curiosity. Here, continuous tracking of ultimate levels of CN - -ions in HeLa cells was reported employing biocompatible branching molecular architectures (BMAs). These BMAs were engineered by decorating colorant-laden dendritic branch within and around the molecular building hollows of the geode-shelled nanorods of organic-inorganic Al-frameworks. Batch-contact methods were utilized to assess the potential of hollow-nest architecture for inhibition/evaluation of toxicant CN - -ions within HeLa cells. The nanorod BMAs revealed significant potential capabilities in monitoring and tracking of CN - ions (88 parts per trillion) in biological trials within seconds. These results demonstrated sufficient evidence for the compatibility of BMAs during HeLa cell exposure. Under specific conditions, the BMAs were utilized for in-vitro fluorescence tracking/sensing of CN - in HeLa cells. Cyanide is widely utilized in the extraction of precious metal extraction even though it has been deemed as the most toxic compound. Fusarium oxysporum has been shown to degrade cyanide through the activity of the Nitrilase enzyme. In this study, the coding sequence of nitrilase gene from F. oxysporum genomic DNA was optimized for cloning and expression in E. coli. The pUC57 containing synthetic optimized nitrilase gene was transferred into E. coli DH5α strain. This nitrilase gene was sub-cloned into pET26b (+) expression vector containing an in-built His-tag at the C-terminal end to facilitate its purification. The recombinant plasmid, pETAM1, was confirmed by PCR, digestion pattern, and sequencing. The recombinant protein was overproduced in E. coli BL21 (DE3). The results of the SDS-PAGE pattern and Western blot analysis confirmed the expression of the expected recombinant protein. For expression optimization of Nitrilase protein, M16 orthogonal experimental design of the Taguchi method was used. The effect of induction time, temperature and IPTG concentration were examined using four levels for each factors. Estimation of the amount of the expressed protein was calculated via densitometry on SDS-PAGE. The enzyme activity and expression in E. coli proved to be successful since there was ammonia production when potassium cyanide and acrylonitrile were used as substrates while the highest enzyme activity of 88% was expressed at 30 °C. The novel glutamic acid crosslinked chitosan membrane (CsG) was fabricated and tested for its adsorption capabilities for the removal of multiple pollutants like Cr (VI), cyanide, fluoride and diclofenac sodium from wastewater. This fabricated CsG membrane was characterized by various techniques like FT-IR, SEM, EDX and XRD, BET to assess its structural, compositional and morphological properties. The working parameters studied by batch experiments were solution pH, CsG dose, contact time, pollutant concentration and solution temperature. The CsG membrane exhibited maximum adsorption capacity of 410.7 mg/g, 310.2 mg/g, 14.3 mg/g, 132.7 mg/g for Cr (VI), cyanide, fluoride and diclofenac respectively. The validation of the operational parameters was performed by Response Surface Methodology (RSM). The experimental data fitted well with Langmuir isotherm model and followed pseudo second order kinetics for all the four targeted contaminants. The spontaneity of the process was checked by thermodynamics studies. The high partition coefficients of 7669 L/kg Cr(VI), 23,309 L/kg (CN - ), 649 L/kg (F - ) and 2613 L/kg (DFC) are the indicators of excellent attractive interaction between CsG membrane and target toxicants. The CsG membrane showed efficient regenerative adsorption properties up to 5 adsorption-desorption cycles. The continuous discharge of untreated refinery wastewater containing pollutants into the water bodies have raised enormous concern on the safety of human, aquatic species and therefore calls for concerted efforts towards developing effective and sustainable treatment method. This study focused on the development of MWCNTs/TiO2 nanoadsorbent for the removal of phenol and cyanide from refinery wastewater. MWCNTs was prepared via cracking of acetylene gas on nickel ferrites nanoparticles in a long tubular reactor. The MWCNTs and MWCNTs/TiO2 nanoadsorbent were characterized for surface properties. HR-SEM and HR-TEM results revealed successive dispersion of TiO2 on the pores of MWCNTs with average crystallite size of 37.32 nm and surface area of 360.66 m2/g. The batch adsorption process revealed maximum removal of cyanide and phenol onto MWCNTs/TiO2 adsorbent at 70 min, 0.3 g and 40 °C contact time, adsorbent dosage and temperature respectively. The experimental data were better described by Freundlich Isotherm and Pseudo second order kinetic model, an indication of multilayer adsorption. The values of the free energy change, (ΔG) were negatives which suggested the existence of strong bond formation between the adsorbates and the adsorbents. The study demonstrated that the developed MWCNTs/TiO2nanoadsorbent is an excellent material for the sequestration of phenol and cyanide from refinery wastewater.

The cyanide, haematology and histopathology profiles of white albino rats fed with ‘fufu’-based diets were monitored. The cassava tubers were subjected into different processing operations: spontaneous-soaked traditional method (SWI), grated before spontaneously-fermented without starter culture (CWI) and those grated, blanched at 65 °C for 15 min before fermented with microorganisms isolated, purified and identified from spontaneously fermented ‘fufu’ categorized into Bacteria only (BAP), Bacteria and Yeast (BYP), Yeast only (YAP), Bacteria and Mould (BMP), Mould and Yeast (MYP) and Mould only (MAP) and were oven dried and milled. The commercial ready-to-eat ‘Fufu’ (CWF) was served as control for laboratory processed samples. Albino rats of the wister strain with four (4) rats per group were fed with 50 % of Commercial vital feed (CVF) and 50% each of the remaining nine (9) “Fufu” samples in ratio 1:1 before haematology and histopathology profile of the animals were investigated. It was found out that Samples BMP and MAP had abnormal high Neutrophil (58–60; 41–42 %) lower Lymphocyte (40–43; 58–61 %), lower Packed cell volume (46–48; 45–47 %) and higher cyanide in the blood (0.004–0.006 mg/L) with sample BMP highest white blood cell (23–24 × 10⁹/L) and sample MAP least white blood cell (6.5–6.6 × 10⁹/L) when compared with other samples which were within the acceptable recommended values for animal studied for haematology and histopathology profiles. The “fufu” samples had reduced cyanide levels ranging from 0.001 mg/L to 0.006 mg/L in the blood of the animals assayed due to the processing operations used. Therefore, combined use of bacteria isolated as starter cultures in the production of odourless “Fufu” have acceptable haematology, histopathology and reduced cyanide level which suggests the suitability in terms of safety for human consumption.

Sustainability of nutrient requirements for microbial proliferation on a large scale is a challenge in bioremediation processes. This article presents data on biochemical properties of a free cyanide resistant and total nitrogen assimilating fungal isolate from the rhizosphere of Zea mays (maize) growing in soil contaminated with a cyanide-based pesticide. DNA extracted from this isolate were PCR amplified using universal primers; TEF1-α and ITS. The raw sequence files are available on the NCBI database. Characterisation using biochemical data was obtained using colorimetric reagents analysed with VITEK® 2 software version 7.01. The data will be informative in selection of biocatalyst for environmental engineering application.

Cells of most eukaryotic species contain mitochondria, which play a role in physiological processes such as cellular senescence, metabolism, and autophagy. Viscosity is considered a key marker for many illnesses and is involved in several crucial physiological processes. Cyanide (CN - ) can target cytochrome-c oxidase, disrupting the mitochondrial electron transport chain and causing cell death through asphyxiation. In this study, a fluorescent probe named HL-1, which targets mitochondria and measures viscosity and CN - levels, was designed and synthesized. HL-1 is viscosity-sensitive, with a linear correlation coefficient of up to 0.992. In addition, HL-1 was found to change color substantially during a nucleophilic addition reaction with CN - , which has a low detection limit of 47 nM. HL-1 not only detects viscosity and exogenous CN - in SKOV-3 cells and zebrafish but also monitors viscosity changes during mitochondrial autophagy in real time. Furthermore, HL-1 has been used successfully to monitor changes in mitochondrial membrane potential during apoptosis. Endogenous CN - in plant samples was quantified. As cyanide is a huge hazard to the environment and human health, the study of the method of detecting low concentrations of cyanide is of great significance. In general, materials with strong positive electrostatic properties can use electrostatic attraction to enrich anions in the water near the materials, then realize rapid detection of low concentration anions by fluorescent probes. In this paper, fluorescent probes PI-S, PI-I and PI-N with cyanide-specific recognition and different charges were synthesized to study the relationship between the charge effect of probes and the sensing sensitivity. Through the zeta potential test and the calculation of the surface electrostatic potential, the positive electricity of PI-S, PI-I and PI-N gradually increased, the ΔG < 0 of the adsorption process gradually decreased, CN - could be aggregated to the vicinity of probes. As a result, the detection limit of the probe was gradually reduced from 1.07 × 10 -6 to 5.03 × 10 -8 M, the sensitivity was significantly enhanced. Therefore, this is expected to be a new strategy to improve the sensitivity of anion probes by increasing the positive electricity of molecules. The disposal of mining wastes that contain cyanide are dumped in tailings from gold extraction is a public concern in mining countries such as South Africa. Many studies have shown the potential of microorganisms to degrade cyanide. However, no in-situ exploration, in tailings contaminated with cyanide, of the capability of indigenous microorganisms to act as a natural barrier for cyanide attenuation has been performed. Here we aim to combine geochemical and metagenomics techniques to reveal the genomic machinery of indigenous bacteria to degrade cyanide in gold tailings. Indigenous bacteria (i.e., Alicyclobacillus, Sulfobacillus, Acinetobacter, Achromobacter, and among others) pose the genomic machinery to trigger hydrolytic cyanide degradation using enzymes such as nitrilase, nitrile hydratase (Nhase) and thiocyanate. It seems that the lack of nitrogen sources induces the use of cyanide, which would contribute to its natural attenuation in the gold tailings. A new naked-eye chromogenic and fluorogenic probe KS5 has been developed for the detection of CN - ions in neat DMSO and H 2 O:DMSO (1:1 v/v) media. The probe KS5 exhibited selectivity towards CN - and F - ions in organic and high selectivity towards CN - ions in aquo-organic media resulting in a colour change from brown to colourless and a turn-on fluorescence response. The probe could able to detect CN - ions via a deprotonation process, which was conceived by consecutive addition of hydroxide and hydrogen ions and confirmed using 1 H NMR studies. The limit of detection (LOD) of KS5 towards CN - ions were in the range of 0.07-0.62 µM in both these solvent systems. Suppression of intra-molecular charge transfer (ICT) transition and photoinduced electron transfer (PET) process of KS5 by the added CN - ions are responsible for the chromogenic and fluorogenic changes observed, respectively. Density Functional Theory (DFT) and Time-Dependent Density Functional Theory (TD-DFT) calculations strongly supported the proposed mechanism along with the optical properties of the probe before and after the addition of CN - ions. In the present study, comparative investigation for the bioremoval of cyanide from synthetic aqueous solution by Prunus amygdalus (Almond) shell (PAS) and Tectona grandis (Sagwan) leaves (TGL) powder have been performed as a function of solution pH, bioadsorbent dose, contact time and initial cyanide concentration. The cyanide uptake by PAS and TGL powder has been quantitatively estimated using sorption isotherms. The equilibrium data are best fitted to Freundlich and Langmuir isotherm model for PAS and TGL powder respectively. The monolayer capacity of PAS and TGL powder for cyanide adsorption as calculated by Langmuir isotherm are 32.05 mg/g and 18.45 mg/g respectively. The kinetic data are found to follow closely the pseudo-second order kinetic model for both the bioadsorbents. The present study shows that such low cost materials could be used as efficient bioadsorbents for the removal of cyanide from aqueous solutions.

In the recent years, great attention had been focused on cyanide toxicity because of its widespread use in industries and considered to be a ubiquitous pollutant in the environment. Therefore, the current study aimed to evaluate the toxic effect of cyanide on rat sperms at molecular level by using FT-IR technique. For this purpose, rats were randomly divided into four groups and treated with 0.0, 0.64, 1.2 and 3.2 mg kg−1 body weight (BW) for the period of 90 days. The group treated with lower dose (0.64 mg kg−1 BW) showed an insignificant change in all the peaks, except the peaks assigned to olefinic CH, CH2 asymmetric and CH2 symmetric stretching vibration in the lipids. While, the groups treated with higher doses (1.2 and 3.2 mg kg−1 BW) showed the significant decrease in the area under the peaks corresponds to different bio-molecules. In addition, spectral second derivative analysis showed the significant alteration in α-helix, turns, β-sheet, aggregated β-sheet and random coil structures in the proteins. In conclusion, the selected higher dosage of cyanide had caused significant decrease in the biochemical composition of rat sperms along with structural changes in the proteins. The FT-IR technique is an excellent tool used for the analysis of oxidative damage in the sperms.

A novel fluorescent probe, with advanced features including "turn-on" fluorescence response, high sensitivity, good compatibility, and mitochondria-targeting function, has been synthesized based on structural design for detecting and visualizing cyanide in foods and biological systems. An electron-donating triphenylamine group (TPA) was employed as the fluorescent and an electron-accepting 4-methyl-N-methyl-pyridinium iodide (Py) moiety was used as a mitochondria-targeted localization unit, which formed intramolecular charge transfer (ICT) system. The "turn-on" fluorescence response of the probe (TPA-BTD-Py, TBP) toward cyanide is attributed two reasons, one is the insertion of an electron-deficient benzothiadiazole (BTD) group into the conjugated system between TPA and Py, and the other is the inhibition of ICT induced by the nucleophilic addition of CN - . Two active sites for reacting with CN - were involved in TBP molecule and high response sensitivity were observed in tetrahydrofuran solvent containing 3 % H 2 O. The response time could be reduced to 150 s, the linear range was 0.25-50 μM, and the limit of detection was 0.046 μM for CN - analysis. The TBP probe was successfully applied to the detection of cyanide in food samples prepared in aqueous solution, including the sprouting potato, bitter almond, cassava, and apple seeds. Furthermore, TBP exhibited low cytotoxicity, clear mitochondria-localizing capability in HeLa cells and excellent fluorescence imaging of exogenous and endogenous CN - in living PC12 cells. Moreover, exogenous CN - with intraperitoneal injection in nude mice could be well monitored visually by the "turn-on" fluorescence. Background: Cyanide anion can be found in foodstuffs, tobacco smoke and a variety of types of waters, mainly originating from anthropogenic activities. Due to its highly toxic nature, several agencies have established limits for cyanide levels in water. Additionally, monitoring cyanide levels in biological samples, such as blood and urine, is crucial for obtaining clinical information about the health condition of patients. Therefore, there is a pressing need for the development of simple, cost-effective, and reliable analytical methods capable of quantifying cyanide at low concentrations., Results: This study presents a novel analytical method for the selective and sensitive determination of cyanide based on analyte volatilization, pre-concentration via single-drop microextraction (SDME) using a selective reagent, and colorimetric quantification using a paper-based analytical device. For this, 10 mL of a liquid sample was acidified with phosphoric acid and the generated HCN was collected using a single drop of 3 μL of a palladium dimethylglyoximate solution (Pd (DMG) 2 2- ) positioned in the flask headspace using a syringe. The reaction of Pd (DMG) 2 2- leads to the formation of Pd(CN) 4 2- and the demasking of the organic ligand. After 15 min of extraction time, the reagent drop was added to a paper-based analytical device that has been previously impregnated with 3 μL of nickel chloride, resulting in the formation of a red precipitate of nickel (II) dimethylglyoximate. Digital images of the paper-based device were captured and the red channel (R) was used for quantification purposes. Under optimized conditions, the method demonstrates a suitable linear relation (r 2  > 0.99) ranging from 26 to 286 μg L -1 and a limit of detection of 5 μg L -1 ., Significance: As a proof of concept, cyanide levels were quantified in water and urine samples using this method. Artisanal and small-scale gold mining (ASGM) is the leading global source of anthropogenic mercury (Hg) release to the environment. Top-down mercury reduction efforts have had limited results, but a bottom-up embrace of cyanide (CN) processing could eventually displace mercury amalgamation for gold recovery. However, ASGM transitions to cyanidation nearly always include an overlap phase, with mercury amalgamation then cyanidation being used sequentially. This paper uses a transdisciplinary approach that combines natural and social sciences to develop a holistic picture of why mercury and cyanide converge in gold processing and potential impacts that may be worse than either practice in isolation. We show that socio-economic factors drive the comingling of mercury and cyanide practices in ASGM as much or more so than technical factors. The resultant Hg-CN complexes have been implicated in increasing the mobility of mercury, compared to elemental mercury used in Hg-only processing. To support future inquiry, we identify key knowledge gaps including the role of Hg-CN complexes in mercury oxidation, transport, and fate, and possible links to mercury methylation. The global extent and increase of mercury and cyanide processing in ASGM underscores the importance of further research. Adsorption of cyanide out of aqueous media by moderate density fiberboard saw dust (MDFSD) was investigated using a batch system under controlled conditions. The kinetic data were followed more closely to the pseudo-first-order model. The adsorption rate and the related parameters, such as the MDFSD relative quantity, cyanide initial concentration, pH, temperature, and ionic strength were studied. The saturation equilibrium was also studied as a function of the related parameters (MDFSD quantity, initial concentration, pH and temperature). The adsorption isotherm was established and compared to other isotherms of activated carbon, TDRP and polyurethane foam. Linearized Langmuir, Freundlich and Frumkin models were applied to estimate the adsorption constants. It was estimated through this study that the adsorption rate is positively affected by increasing MDFSD’s relative quantity, adsorbent concentration, acidity and temperature. The adsorption equilibrium was shifted toward higher adsorption capacity by the substrate concentration, temperature and acidity. The adsorption data fit to Freundlich, Langmuir and Frumkin models, but attained a better correlation with the order of models as Langmuir > Frumkin > Freundlich. Activation thermodynamic parameters of adsorption, the adsorption thermodynamic parameters, and Freundlich, Langmuir and Frumkin constants were obtained.

The current study was undertaken to address the role of mitochondrial reactive oxygen species (ROS), and hypoxia inducible factor-1 alpha (HIF-1α) signaling pathway in the protection against high glucose levels in brain endothelial and NT2 neuron-like cells. Rat brain endothelial cells (RBE4) treated with non-toxic concentrations of cyanide (≤1 μM; 1 h) exhibited an increase in ROS levels, particularly hydrogen peroxide (H2O2). Cyanide also induced a modest mitochondrial depolarization, an increase in oxygen consumption and a structural (smaller mitochondria) and spatial (perinuclear region) reorganization of mitochondrial network. The stabilization and nuclear activation of HIF-1α in the presence of cyanide were also observed, which resulted in an increase in vascular endothelial growth factor (VEGF), endothelial nitric oxide synthase (eNOS) and erythropoietin (EPO) protein levels reflecting an adaptive response. Importantly, preconditioning induced by cyanide protected brain endothelial cells against high glucose-mediated damage by the prevention of apoptotic cell death. In mitochondrial DNA-depleted NT2 (NT2 ρ0) cells, cyanide (0.1 μM) was unable to stimulate ROS production and, consequently, protect against glucotoxicity. Conversely, in NT2 cells, the parental cells with functional mitochondria, cyanide significantly increased ROS levels protecting against high glucose-induced neuronal cell loss and activation of caspase-3. The free radical scavenger N-acetyl-L-cysteine and the specific HIF-1α inhibitor 2-methoxyestradiol completely abolished the protective effects of cyanide preconditioning. Altogether our results demonstrate that mitochondrial preconditioning induced by cyanide triggers a protective response mediated by mitochondrial ROS and HIF-1α activation and signaling, which render brain endothelial and neuronal cells resistant against glucotoxicity.
