In most teleost fishes, sperm cells are quiescent in the seminal plasma and are activated by either a drop (fresh water fish) or an increase in osmolality (marine fish) when released in the water. It is most interesting to examine how the mechanisms of sperm motility activation can adapt to a broad range of salinities, as applies to some euryhaline species, and particularly to the tilapia Sarotherodon melanotheron heudelotii, which can reproduce at salinities from 0 up to 120 in the wild. Here, the gonado-somatic index, semen characteristics, and the osmotic and ionic requirements of sperm motility activation were compared in S. m. heudelotii reared in fresh water (FW), sea water (SW), or hypersaline water (HW; salinities of 0, 35, and 70, respectively). No salinity-dependent differences were found in gonado-somatic index or semen characteristics, except for an increase of seminal plasma osmolality with increasing salinity (from 318 to 349 mOsm kg(-1) in FW and HW fish, respectively). The osmolality range allowing the highest percentages of sperm activation broadened and shifted toward higher values with increasing fish ambient salinity (150-300, 300-800, and 500-1200 mOsm kg(-1), for FW, SW, and HW fish, respectively). Nevertheless, at the three fish rearing salinities, sperm could be activated in media that were hypotonic, isotonic, or hypertonic relative to the seminal plasma, at least when some calcium was present above a threshold concentration. The [Ca(2+)] required for the activation of S. m. heudelotii sperm is (1) higher in fish reared at a higher salinity (2) higher in hypertonic than that in hypotonic activation media, whatever the fish rearing salinity, and (3) higher in the presence of Na(+) or K(+), the negative effects of which increased with an increase in fish rearing salinity. The [Ca(2+)]/[Na(+)] ​ ratios allowing for maximal sperm motility in SW or HW fish are close to those observed in natural environments, either in sea or hypersaline waters. In comparison to most teleosts with external fertilization, the total duration of sperm motility in S. m. heudelotii was exceptionally long (>2 hours regardless the fish rearing salinities). The decrease in sperm activity with increasing time since activation did not result from limiting energy reserves, as the addition of calcium in the activation medium caused most spermatozoa to become motile again. The comparison of sperm characteristics of S. m. heudelotii acclimated from FW to SW or HW with those of fish maintained all lifelong at their native salinity showed that adaptive responses were completed within 2 months or less., (Copyright © 2016 Elsevier Inc. Objective: The study objective was to assess the awareness of standard precautions (SP) among healthcare professionals, 1 year after the latest national guidelines were issued., Methods: A multicenter cross-sectional survey was conducted in 34 volunteer institutions in 2010. Data was collected using an anonymous and self-administered questionnaire. The data was analyzed with a program developed from Epi-Info software., Results: Four thousand four hundred and thirty-nine questionnaires were analyzed. Most respondents were nurses (44.1%) or nurses' aides (26.7%) followed by physicians (3.5%). 25% of respondents had participated in specific PS training in the previous 5 years. The percentage of correct answers for each question ranged from 37.1 to 91%. There was 72.6% of correct answers on hand hygiene but only 7.3% of correct answers on use of appropriate barriers and disposal of needles. 39.3% of respondents gave correct answers to eight or more of the 10 SP questions. The level of knowledge of nurses was higher compared to other professionals. The lowest level of knowledge was observed in long-term care and psychiatric institutions., Conclusions: The knowledge of healthcare professionals on use of appropriate protective barriers and disposal of needles is still too limited. Protein kinases are central components of signal transduction cascades often dysregulated in cancer, and they represent some of the most promising drug targets. However, the target selectivity is a major concern because most described kinase inhibitors target the highly conserved ATP-binding pocket. Recently, new classes of inhibitors that do not compete with ATP and exhibit different mechanisms of action have been described. Overexpression of protein kinase CK2 is an unfavorable prognostic marker in several cancers. Consequently, CK2 has emerged as a relevant therapeutic target. Several classes of ATP-competitive inhibitors have been identified, showing variable effectiveness. The molecular architecture of this multisubunit enzyme could offer alternative strategies to inhibit CK2 functions, and this review illustrates these emerging possibilities.

Protein kinase CK2 is a multifunctional kinase of medical importance that is dysregulated in many cancers. In this study, polyoxometalates were identified as original CK2 inhibitors. [P2Mo18O62](6-) has the most potent activity. It inhibits the kinase in the nanomolar range by targeting key structural elements located outside the ATP- and peptide substrate-binding sites. Several polyoxometalate derivatives exhibit strong inhibitory efficiency, with IC50 values < or = 10 nM. Furthermore, these inorganic compounds show a striking specificity for CK2 when tested in a panel of 29 kinases. Therefore, polyoxometalates are effective CK2 inhibitors in terms of both efficiency and selectivity and represent nonclassical kinase inhibitors that interact with CK2 in a unique way. This binding mode may provide an exploitable mechanism for developing potent drugs with desirable properties, such as enhanced selectivity relative to ATP-mimetic inhibitors.

Of the four subunits constituting DNA polymerase delta, subunit C or p66 has been shown to mainly mediate polymerase interaction with PCNA, an auxiliary factor that greatly enhances DNA polymerase delta processivity on primed DNA templates. Here, we provide evidence that a highly conserved region located between amino acids 384 and 399 in the C-terminus of p66 is phosphorylated, most probably by Protein kinase CK2, and that another region, most probably located within the PCNA interacting domain in its extreme C-terminus, regulates its interaction with PCNA. Phosphorylation of p66 is associated with its co-localization with large subunit of DNA polymerase delta, p125, and PCNA, to the insoluble chromatin fraction at the beginning of S-phase. Taken together, the results provide evidence that concurrent phosphorylation events in p66 may positively and negatively regulate its activity and interactions with other components of the replisome during the cell cycle.

Protein kinase CK2 appears to interact with different signaling pathways and therefore represents the prototype of a multifunctional protein kinase. The enzyme is highly expressed in most cancers and this higher expression has been tentatively correlated with the involvement of CK2 in the promotion of specific phases of the cell cycle. This review summarizes our current knowledge on how CK2 may also contribute to tumorigenesis through its direct interaction with the cell-survival circuitry.

Using a synthetic peptide corresponding to the sixteen amino-acid N-terminus of the beta subunit, the structure-activity relationship study of casein kinase 2 (CK2) was performed with regard to its previously reported property to polymerize and oligomerize in vitro. Velocity sedimentation experiments show that the peptide beta 1-16 prevents the thick filament formation and stabilizes the ring-like structure of the kinase. Furthermore, the peptide beta 1-16 stimulates the kinase activity by 3-fold toward exogenous substrates as well as the intrinsic autophosphorylation of the kinase. Such observations are in agreement with the proposed model of an activated state of CK2 when the ring-like structure is adopted. Comparison of the effects of spermine and peptide beta 1-16 on CK2 structure and activity suggests that these two activating molecules may function in a different way. Our study suggests that the N-terminal region of the beta subunit of CK2 could regulate the kinase activity by controlling the quaternary structure of the enzyme.

EGF is involved in the regulation of cell proliferation in normal as well as in neoplastic tissues. The A431 cells that over-express EGFR, display in vitro ambivalent growth properties in response to EGF, since stimulation induced by low concentrations (10(-12) M-10(-10) M) is reversed with increasing concentrations (10(-9) M-10(-8) M). To assess differential mechanisms of signal transduction that determine growth stimulatory and inhibitory activity, we have studied the MAP kinase activation induced by mitotic and antimitotic concentrations of EGF. We demonstrate that the presence of a growth stimulatory concentration of EGF (10(-12) M) leads to a moderate but persistent activation of p42 MAP kinase during all the time of the EGF treatment. Conversely, an early peak of kinase activation that rapidly falls down below the basal level, is observed when a growth inhibitory concentration of EGF (10(-8) M) is used. Moreover, the addition of Na-orthovanadate in 10(-8) M EGF-treated cells leads to the rescue of the MAP kinase activity, suggesting that the loss of kinase activity induced by growth inhibitory EGF concentrations involves the dephosphorylation of the MAP kinase. In conclusion, our data demonstrate that the dual effect (stimulator/inhibitor) of EGF on the proliferation of A431 cells is associated to differential mechanisms of p42 MAP kinase regulation.

Casein kinase II is a ubiquitous serine-threonine kinase made of two different (alpha and beta) subunits, with an alpha 2 beta 2 stoichiometry, alpha bearing the catalytic site of the enzyme. Two different genes encode two different (alpha and alpha') catalytic subunits. The present work shows that at least two forms of CKII (CKIIa and CKIIb) can be isolated from bovine tissues and adrenocortical cells in culture. The use of specific antibodies developed against the alpha and alpha' subunits disclosed that while CKIIa contains both alpha and alpha', the alpha subunit was highly prominent in CKIIb. This suggests that CKIIb may be an alpha 2 beta 2 oligomer whereas CKIIa may be an alpha alpha' beta 2 moiety or an alpha alpha' beta 2 and alpha' 2 beta 2 mixture. Transition from quiescence to proliferation was concomitant with an average two fold increase in total cell CKII activity involving both CKIIa and CKIIb isoforms. The two isoforms were found in cytosolic and particulate adrenocortical subcellular preparations and they similarly increased their association to intracellular organelles in response to growth stimulatory conditions.

Treatment of thyroid differentiated carcinoma is controversial. In case of papillary microcarcinoma a low malignancy is usually considered and a limited surgical excision is currently used. We describe 49 cases with papillary tumor < 1 cm. Their high frequency (51% of all differentiated carcinoma discovered in the same period), current mode of their diagnosis (incidental histological findings: N = 48), and constant tumoral situation in extra nodular parenchyma are emphasised. In spite of median tumor size < 2 mm, 8% had extra thyroidal tumoral extent at diagnosis (node metastasis: N = 3; bone: N = 1). In one case with multifocal lesions in both lobes, an unilateral thyroidectomy would have missed contralateral periglandular metastatic nodes. Diagnosis of the case with asymptomatic bone metastasis was clearly attributable to radioiodine therapeutic use. These results suggest an heterogeneous prognosis of papillary microcarcinoma, with some cases requiring total bilateral thyroidectomy and radioiodine remnants ablation (e.g. Tumors invading peripheral thyroid tissue that seem at higher risk of extra glandular extension).

A decreased bone mineral density is a well known complication of thyrotoxicosis. If bone is also adversely affected by a subclinical hyperthyroidism is still debated. Such a situation is deliberately induced for treatment of thyroid differentiated carcinoma. We compared bone mineral measurements in case of thyroid carcinoma exposed to prolonged suppressive hormonal treatment (288 patient year, LT3 in 87%) and in age and weight matched controls. We did not find any difference in both sexes between the two populations in term of fracture rate and vertebral mineral density measured by dual photon absorptiometry, nor any influence of gonadal status in women.

Protein phosphorylation by the ubiquitous casein kinase II (CKII) is known to be sensitive to naturally occurring polyamines. Using isolated recombinant alpha and beta subunits of the kinase, as well as the alpha 2 beta 2 oligomeric enzyme, it is shown that (i) CKII binds [3H]-spermine with Kds in the micromolar range. (ii) The beta subunit appears mostly responsible for this binding activity. (iii) The isolated alpha (catalytic) subunit is not activated by polyamines. (iv) The polyamine-dependent activation of the oligomeric CKII requires the beta subunit, which appears as a regulatory component in the native kinase. These observations suggest that there may be a functional interaction between polyamines and CKII in living cells, especially in the response to cell growth factors and trophic hormones.

The ubiquitous casein kinase II (CKII) has been shown to accumulate in the cell nuclear compartment, following exposure to extracellular growth stimuli (O. Filhol et al., Biochemistry, 1990, 29, 9928-9936). The aim of the present study was to examine whether intracellular polyamines, whose levels are increased under similar conditions, could be related to this process. It is shown that (i) CKII accumulates in nuclei of adrenocortical cells exposed to their trophic hormone ACTH; (ii) this CKII nuclear translocation is concomitant with an increase in nuclear polyamine content resulting from ACTH-induced polyamine synthesis; (iii) selective inhibition of polyamine biosynthesis by DFMO results in the inhibition of both ACTH-induced cellular polyamine increase and CKII nuclear accumulation. These observations suggest that polyamines may be examined as intracellular messengers in the regulation of CKII activity and subcellular distribution in the cell response to growth factors and trophic hormones.

Transforming growth factor beta (TGF beta) is a multifunctional polypeptide presenting various biological activities depending on the cellular context where it is studied. In the adrenal cortex, TGF beta is a potent inhibitor of corticosteroïdogenesis. The sum of data that we have collected in the past years has allowed us to establish that TGF beta (in particular TGF beta 1) is implicated in a physiological autocrine/paracrine regulatory loop. We determined that primary cultures of bovine fasciculatareticularis cells synthesize and secrete about 5 ng/24 h x 10(6) cells of TGF beta but that most of this activity is under a latent form. In the adrenal gland, TFG beta 1 could be localized in the fasciculata-reticularis zone by immunohistochemistry. We also reported that adrenocortical cells possess type I and type III (betaglycan) receptors, whose number is positively regulated by ACTH. Moreover, we have identified 3 major targets of TGF beta in our cell model: cytochrome P-450(17) alpha (17 alpha-hydroxylase) whose gene expression is decreased, angiotensin II receptors and LDL receptors whose number is reduced after TGF beta treatment. As a conclusion, TGF beta appears to participate together with other regulatory peptides (IGF-1, TNF alpha) in the fine tuning of corticosteroid secretion levels which are under the major control of the hypothalamo-pituitary axis.

Casein kinase II, an ubiquitous, oligomeric, messenger-independent protein kinase has previously been shown to concentrate in the nuclear compartment when cells are stimulated to proliferate. The present communication reports that purified mammalian CKII interacts with genomic DNA preparations in vitro. This interaction led to an apparent activation of the kinase, most likely explained by prevention of its aggregation and subsequent denaturation. Binding of CKII was optimum with double stranded DNA preparations; duplex lambda phage DNA exhibited at least two types of binding sites and the high affinity system (Kd approximately equal to 6 x 10(-13) M) represented a binding capacity of about 1 mol CKII per mol DNA. CKII-DNA interaction was stimulated in the presence of a polyamine and inhibited by heparin. Blotting experiments disclosed that DNA binds CKII through its alpha subunit. These observations are in line with the hypothesis that casein kinase II may be examined as a component in the transduction of the mitogenic signal from the cell membrane to the nucleus, in response to growth factors.

Epidermal Growth Factor (EGF) was assayed in saliva collected from control subjects and cystic fibrosis (CF) patients, using both radioimmuno (RIA) and radioreceptor (RRA) assays. An intriguing finding was that the average ratio of the values found by RRA over those obtained by RIA was of 1.7 for normal subjects and of about 0.4 for CF patients. This observation could be understood following gel filtration analysis of EGF-like material in these salivary fluids. Whereas control saliva contained the expected 6 kDa EGF active peptide, the immunoreactive EGF material from CF patients eluted as a polydisperse macromolecular moiety. The poor biological reactivity of this material as assessed by radioreceptor assay suggests that this EGF anomaly may contribute to the physiopathology of cystic fibrosis, especially as the upper gastrointestinal tract differentiated functions may be the target of normal salivary EGF.

Ten independant cellular hybrids were obtained from Chimpanzee (Pan troglodytes) fibroblasts and the murine cell line C11D. The comparison of electrophoretic and cytogenetic studies showed that 9 markers with known localizations in Man could also be localized on the homologous chromosomes of the Chimpanzee: pyrophosphate hydratase (PPH), phosphoglucomutase-1 (PGM1), and peptidase-C (Pep-C) on the No. 1; malate dehydrogenase MDH(NAD) on the No. 2; lactico dehydrogenase-A (LDH-A) on the No. 11; lactico dehydrogenase-B (LDH-B) on the No. 12; thymidine kinase (TK) on the No. 17; superoxide dismutase-1 (SOD-1) on the No. 21; glucose-6-phosphate dehydrogenase (G6PD) on the X. The localization of mannose phosphate isomerase (MPI) on the No. 7 could be excluded. One discrepancy between Chimpanzee and Man was noted: the localization of superoxide dismutase-2 (SOD-2) on the 6 is excluded.

A twofold CuZnSOD activity was observed in an orangutan as compared to man and a chimpanzee. This suggests that a submicroscopic duplication of chromosome 21 (similar to those demonstrated in patients with the trisomy 21 phenotype but with a normal karyotype) has occurred in the homozygous state during evolution of the orangutan phylum. Such duplications could be important evolutionary mechanisms, together with visible chromosome rearrangements and classical gene mutations.

Whereas TGF-beta exhibited no detectable effect on DNA synthesis, it was found to exert a striking inhibitory effect on the steroidogenic activities of bovine adrenocortical cells in culture. Basal, as well as ACTH- and angiotensin II- activated adrenocortical cortisol productions were inhibited in a time and dose-dependent manner following TGF-beta treatment. Half-maximum inhibition of ACTH- and AII-activated steroidogenesis was observed with TGF-beta concentrations of 0.40 and 0.12 ng/ml, respectively. This effect was half maximal after 6 hours of cell exposure to optimally effective TGF-beta concentrations (1 ng/ml) and reached a plateau after 12-15 hours, resulting in an average 60% inhibition in the steroidogenic response to ACTH and 90% in the case of AII. Supply of different exogenous steroid substrates to support steroidogenesis in adrenocortical cells pointed to a marked loss in steroid-17 alpha hydroxylase activity as a major alteration following TGF-beta treatment. TGF-beta thus appears as a potent modulator of differentiated adrenocortical cell functions in vitro; in this regard it may play a significant role in the development and the regulation of adrenal cortex in vivo.

Analysis of cellular hybrids confirms the assignment of alpha-L-fucosidase (alpha-FUC) to 1p in man. Discordant results are in favour of the following gene order: 1pter (ENO-1, alpha-FUC,AK2) PGM1 centromere Pep-C but give no information on the relative positions of ENO-1,alpha-FUC, and AK2. The assignments of alpha-FUC to chromosome 1 in the chimpanzee and to chromosome 4 in the African green monkey are demonstrated (chromosome nomenclature by Finaz et al, 1976). These results confirm the homology of chromosome 4 of the African green monkey and 1p of man and the chimpanzee.

Eight new gene assignments were demonstrated in the baboon (Papio papio, PPA) by cosegregation analysis of twelve hybrid clones obtained by fusion between PPA fibroblasts and a mouse cell line deficient in thymidine kinase. The following markers and syntenic groups were assigned: SOD1 to PPA3, GLO-ME1 to PPA-4, PGM2 to PPA5, CKBB-SORD to PPA7, LDHB to PPA11 and LDHA to PPA14. These localizations are in agreement wit hthe following homoeologies with the human karyotype: PPA3-HSA21, PPA4-HSA6, PPA5-HSA4, PPA7-HSA14 and 15, PPA11-HSA12, PPA14-HSA11.

The ACTH and cortisol responses after CRF administration were studied in 21 patients with adrenal failure and compared with those obtained in 15 control subjects. In patients with Addison's disease, cortisol and ACTH responses were respectively abolished or excessive. In patients with secondary adrenal failure, a unique bolus injection of CRF allows the differentiation between pituitary or hypothalamic secretory defect. With regard to previous reports, the usefulness of CRF test in primary and secondary adrenal failure is stressed.

Exposure of various cell types (rat-1 fibroblasts, bovine adrenocortical cells, human lymphoid cells) to nanomolar concentrations of TPA, resulted in a rapid, apparent loss of cellular protein kinase C content, when the enzyme was assayed by its phospholipid and Ca2+-dependent histone (H1)-kinase activity, following solubilization and DEAE-cellulose chromatography isolation. By contrast, no loss of protein kinase C was detected when the enzyme was probed by its high affinity PDBu binding capacity nor when the kinase activity was assayed with protein substrates other than histones, such as vinculin and a cytochrome P-450. It is concluded that, in addition to the previously reported enzyme subcellular redistribution, following TPA treatment, the phorbol ester induces striking alterations of the cellular protein kinase C catalytic activities. The molecular mechanisms of these changes and their implication in the tumor promotion process remain to be clarified.

These results are consistent with the following homoeologies of PPA and HSA chromosomes : PPA3-HSA7 ; PPA7-HSA14 and 15 ; PPA10-HSA20 and PPA12-HSA2q

Type beta Transforming Growth Factor (TGF beta)-like activity was identified in conditioned medium obtained from immature porcine Sertoli cell-enriched cultures using the following criteria: (i) stimulation of anchorage independent growth of mesenchymal cell lines, (ii) competition with pure human TGF beta in a radioreceptor assay. The secretion of the receptor reactive TGF beta-like material in Sertoli cell conditioned medium is decreased to very low or undetectable levels by Follicle Stimulating Hormone, one of the major hormones involved in the physiological testicular activities. The effects of this factor are probably exerted in the context of the local control of the male gonad functions.

Potent tumor promoter TPA (1-100 nM) has previously been shown to induce a striking alteration of protein kinase C catalytic properties in target cells (C. Cochet et al., 1986, Biochem. Biophys. Res. Comm. 134, 1031-1037). This alteration contributes to the apparent loss of cellular protein kinase C, secondary to TPA treatment, when the enzyme is probed by its phospholipid-dependent histone kinase activity. This effect was observed as well when rat-1 cells were treated by other tumor promoters such as mezerein, teleocidin, aplysiatoxin and palytoxin, whereas inactive phorbol ester structures were ineffective. On the other hand, 1,2-dioctanoyl glycerol did not induce that effect. This protein kinase C alteration was shown to occur at the cellular membrane level. It is suggested that membrane translocation and activation of protein kinase C induced by potent tumor promoter structures are not functionally equivalent to that secondary to physiological stimuli. Although the mechanisms underlying this phenomenon remains to be understood at the molecular level, it may be of significance in the process of tumor promotion.

Incorporation of ouabain (G-strophantin) in the culture medium 24 h after that of Sandai virus, at a final concentration of 2.10(-6)M/1, increases considerably the yield of man/mouse or primate/mouse cellular hybridization.

We have investigated the effects of epidermal growth factor (EGF) on calcium ion channels in A431 epidermoid carcinoma cells. We have found that: -1- EGF stimulates Ca2+ channels. -2- EGF stimulated Ca2+ channels are voltage independent, exhibit a low conductance (8 pS) and a bursting multichannels activity (BMC). -3- Activation of the tyrosine-kinase function of the EGF receptor is required to generate Ca2+ current. -4- Inositol (1,4,5) triphosphate (Ins (1,4,5) P3) and EGF have similar effect on the channel activation. These results suggest that: stimulation of tyrosine-kinase activity of the EGF receptor, production of Ins (1,4,5)P3 and calcium entry via voltage independent channels are important connected steps in mediating the mitogenic effect of this growth factor.

The various protein kinase activities characterized in adrenal cortex from different mammalian species do not differ in their molecular and catalytic properties from similar types of enzymes described in other tissues. The cyclic AMP-dependent system has been the most thoroughly investigated in adrenocortical tissue and is generally accepted as an obligatory intermediate in the intracellular response to ACTH action. In addition, adrenocortical cells contain two types of messenger independent protein kinases, one of which being selectively activated by polyamines in vitro and possibly related to the control of adrenocortical growth processes. The definition of intracellular phosphorylatable targets related to specific adrenocortical functions might have been obscured by the pleiotypic effects displayed by ACTH, the major effector of these cell activities. Cholesterol esterase appears one of these targets and 11 beta-hydroxylase cytochrome P-450 has been found activated in vitro by a cAMP dependent process. Future research should provide a better understanding of the role of protein phosphorylation-dephosphorylation processes as a regulatory device in adrenocortical cell functions. This includes possible complex modulation by multisite phosphorylations of a given target protein, involving several different protein kinase systems under the dependence of separate intracellular effectors.

To increase the efficiency of the human gene mapping, a panel of 25 man-rodent hybrids was selected out of 200 independent man-rodent hybrids produced in our laboratory since 1969. The hybrid panel was selected in such a fashion as to allow the asignment of a human marker M by respecting the two following complementary criteria: correlation between M and a chromosome and exclusion of the other chromosomes. The panel characteristics allowing the use of these two criteria were presented and discussed. A first series of enzyme markers were analysed to test the validity of the hybrid panel for the human gene mapping. The different possibilities and limits of the hybrid panel were also discussed, especially for the assignment of markers with DNA probes.

By cellular hybridization with C11D cells, the following gene assignments to specific chromosomes were demonstrated for the chimpanzee: beta-glucuronidase on chromosome 7, enolase-2 on chromosome 12, and the phosphoglycerate kinase on the X.

The karyotype of Cercopithecus aethiops was analysed by RHG, GTG, and C banding, and a band nomenclature is proposed.
