The field of bacteria-based cancer therapy, which focuses on the key role played by the prevalence of bacteria, specifically in tumors, in controlling potential targets for cancer therapy, has grown enormously over the past few decades. In this review, we discuss, for the first time, the global cancer situation and the timeline for using bacteria in cancer therapy. We also explore how interdisciplinary collaboration has contributed to the evolution of bacteria-based cancer therapies. Additionally, we address the challenges that need to be overcome for bacteria-based cancer therapy to be accepted in clinical trials and the latest advancements in the field. The effective management of infectious diseases and the growing concern of antibiotic resistance necessitates accurate and targeted therapies, highlighting the importance of antibiotic susceptibility testing. This study aimed to develop a real-time impedimetric biosensor for identifying and monitoring bacterial growth and antibiotic susceptibility. The biosensor employed a gold 8-channel disk-shaped microelectrode array with specific antibodies as bio-recognition elements. This setup was allowed for the analysis of bacterial samples, including Staphylococcus aureus, Bacillus cereus, and Micrococcus luteus. These microorganisms were successfully cultured and detected within 1 h of incubation even with a minimal bacterial concentration of 10 CFU/ml. Overall, the developed biosensor array exhibits promising capabilities for monitoring S. aureus, B. cereus and M. luteus, showcasing an excellent linear response ranging from 10 to 10 4  CFU/ml with a detection limit of 0.95, 1.22 and 1.04 CFU/mL respectively. Moreover, real-time monitoring of antibiotic susceptibility was facilitated by changes in capacitance, which dropped when bacteria were exposed to antibiotic doses higher than their minimum inhibitory concentration (MIC), indicating suppressed bacterial growth. The capacitance measurements enabled determination of half-maximal cytotoxic concentrations (CC50) values for each bacteria-antibiotic pair. Global warming and climate changes have a detrimental impact on poultry production, causing substantial economic losses. This study investigated the effects of incorporating dietary betaine (BT) and organic minerals (OMs) on broilers' performance as well as their potential to mitigate the negative impacts of heat stress (HS). Six hundred 1-day-old Ross 308 chicks were randomly allocated to 12 experimental treatments with 5 replicates of 10 birds each (5 male + 5 female). The birds were provided with diets containing BT (0 and 2,000 ppm) and OMs (0, 250, and 500 ppm), either individually or in combination, under both thermoneutral and HS-inducing temperatures. The HS conditions involved exposing the birds to cyclic periods of elevated temperature (35°C ± 2°C) for 6 h daily, from 10:00 am to 4:00 pm, starting from d 10 and continuing until d 35. The exposure to HS deteriorated birds' growth performance; however, dietary BT and OMs inclusion improved the growth performance parameters bringing them close to normal levels. Carcass traits were not affected by dietary supplementation of BT, OMs, HS, or their interaction. Interestingly, while HS led to increased (P < 0.05) levels of total cholesterol, LDL-cholesterol, and hepatic malondialdehyde (MDA), these adverse effects were mitigated (P < 0.05) by the addition of BT and OMs. Moreover, dietary BT supplementation led to elevated serum total protein and globulin concentrations. Cyclic HS did not alter Mn, Zn, and Cu contents in the pectoral muscle. However, the incorporation of OMs at both levels increased concentrations of these minerals. Notably, the combination of 500 ppm OMs and 2,000 ppm BT improved Mn, Zn, Cu, and Fe digestibility, which has been compromised under HS conditions. Cyclic HS upregulated gene expression of interleukin-1β, heat shock protein 70, and Toll-like receptor-4 while downregulated the expression of claudin-1, uncoupling protein, growth hormone receptor, superoxide dismutase 1, glutathione peroxidase 1 and insulin-like growth factor 1. The aforementioned gene expressions were reversed by the combination of higher dietary levels of BT and OMs. In conclusion, the dietary supplementation of 500 ppm OMs along with 2,000 ppm BT yielded significant improvements in growth performance and mineral digestibility among broiler chickens, regardless of thermal conditions. Plasma phosphorylated-tau threonine 181 (p-tau181) is a promising biomarker for predicting Alzheimer's disease (AD) and mild cognitive impairment (MCI), which is the symptomatic pre-dementia stage of AD. To date, there are limitations in the current diagnosis and classification of the two stages of MCI and AD in clinical practice remain a dilemma. In this study, we aimed to discriminate and diagnose patients with MCI, AD, and healthy participants based on the accurate, label-free, and ultrasensitive detection of p-tau181 levels in human clinical plasma samples using our developed electrochemical impedance-based biosensor, which allows to detect p-tau181 at a very low concentration of 0.92 fg mL -1 . Human plasma samples were collected from 20 patients with AD, 20 patients with MCI, and 20 individuals with healthy control. The change in charge-transfer resistance of the developed impedance-based biosensor caused by capturing p-tau181 in plasma samples was recorded to evaluate the determination of plasma p-tau181 levels in human clinical samples for discrimination and diagnosis of AD, MCI, and healthy control individuals, respectively. Receiver operating characteristic (ROC) curve, a standard analysis to judge the clinically diagnostic capability of our biosensor platform based on the estimated levels of plasma p-tau181, resulted a sensitivity of 95%, a specificity of 85%, the area under the ROC curve (AUC) value of 0.94 of the accuracy for discriminating AD patients from healthy controls; a sensitivity of 70%, a specificity of 70%, the AUC of 0.75 to discriminate MCI patients from healthy controls. Statistical analysis (one-way analysis of variance (ANOVA)) was used to compare the estimated plasma p-tau181 levels in clinical samples, indicated significantly higher for AD patients with healthy controls (***p ≤ 0.001), AD with MCI patients (***p ≤ 0.001), and MCI patients with healthy controls (*p ≤ 0.05), respectively. In addition, we compared our sensor to the global cognitive function scales and discovered that it performed noticeably improvement in diagnosing the stages of AD. These results demonstrated the good application of our developed electrochemical impedance-based biosensor in the identification of clinical disease stages. The study examined how adding phytase to nonphytate phosphorus (NPP) diets affected performance, egg quality, reproductive hormones, and plasma biochemical indices in 73- to 80-wk-old laying hens. Six treatments with 5 replicates of 18 Hy-Line brown laying hens each were randomly assigned. Three isonitrogenous, isocaloric diets containing consistent calcium levels (3.8%) were formulated to contain 0.20, 0.25, and 0.30% NPP, treated with or without phytase supplementation (1,000 FYT per kg feed, Ronozyme HiPhos-L, Aspergillus oryzae 6-phytase). The results showed that the addition of phytase to the diet containing 0.20, 0.25, and 0.30% NPP increased egg production by 1.50, 1.64, and 0.97%, respectively, and improved eggshell thickness. Also, use of phytase in the diet contain 0.25, and 0.30% NPP increased the plasma concentration of albumin (ALB), high-density lipoprotein (HDL), phosphorus (P), and plasma follicle-stimulating hormone (FSH), plasma calcium (Ca), estradiol-17β (E2β), and luteinizing hormone (LH). In contrast, the egg weight, feed intake, egg mass, feed conversion ratio, albumen height, Haugh unit, yolk, and shell color were unaffected. It can be advisable to use phytase supplementation in an elderly laying hen's diet contain 0.25, and 0.30% NPP to improve shell quality and positively impact reproductive hormones leading to the persistence of egg production., (Copyright © 2023 The Authors. Objective: Corydalis bungeana (CB) is a well-used medicinal herb in Mongolian folk medicine and has been traditionally applied as an antiobesity agent. However, the evidence-based pharmacological effects of CB and its specific metabolic alterations in the obese model are not entirely understood. This study aimed to utilize untargeted metabolomic techniques to identify biomarkers and gain mechanistic insight into the serum metabolite alterations associated with weight loss and lipid metabolism in obese rats., Methods: A high-fat high-sugar (HFHS) diet was used to induce obese models in rats. CB extract was orally gavaged at 0.18, 0.9 and 1.8 g/kg doses for six weeks, and feed intake, body weight, fat pad weight, and blood indexes were measured. Blood serum metabolites were evaluated by gas chromatography/quadrupole time-of-flight tandem mass spectrometry (GC-TOF/MS)., Results: The results showed that compared with the obese group, the administration of CB extract caused significant decreases in body weight ( P  < 0.05), feed intake, Lee's index, and perirenal, mesenteric, epididymal fat weight. CB extract also reduced blood triglyceride and total cholesterol levels ( P  < 0.05) of obese rats. Metabolomic findings showed that nine differential metabolites, including pyruvic acid, D -glucuronic acid, malic acid, dimethylglycine, oxoglutaric acid, pantothenic acid, sorbitol acid, fumaric acid and glucose 6-phosphate were identified under CB treatment and altered metabolic pathways such as TCA cycle, pantothenate and CoA biosynthesis, and glycolysis/gluconeogenesis., Conclusion: This study demonstrated weight loss and lipid lowering effects of CB on HFHS diet-induced obese rats and identified nine metabolites as potential biomarkers for evaluating the favorable therapeutic mechanism of CB via regulation of lipid and glucose metabolism., Competing Interests: The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (© 2022 Tianjin Press of Chinese Herbal Medicines. In this paper, we developed a new ultrasensitive capacitance sensor for detection of amyloid beta 1-40 (aβ40) protein (one of Alzheimer's disease core biomarkers) in human serum based on the high supramolecular recognition of the β-cyclodextrin/reduced graphene oxide (β-CD/RGO) nanohybrid toward the anti-aβ40 antibody molecule. The sensor was established by immobilizing specific anti-aβ40 antibody onto the β-CD/RGO nanohybrid functionalized on indium tin oxide micro-disk electrode (anti-aβ40/β-CD/RGO/ITO). Detection of aβ40 in the human serum (HS) using the sensor anti-aβ40/β-CD/RGO/ITO is carried out by capacitance measurement without a redox probe to prevent protein denaturation, serving as a convenient strategy for point-of-care diagnosis. In comparison with other studies, the sensor shows a very low limit of detection of 0.69 fg mL -1 in HS, demonstrating its ability for the ultrasensitive detection of aβ40. Using this sensor, the dissociation constant K D of the binding interaction between anti-aβ40 and aβ40 in HS is found to be 2.9 × 10 -7  nM, indicating the high binding affinity of antibody-antigen and the suitability of the anti-aβ40/β-CD/RGO/ITO sensor for aβ40 protein detection. Redox cycling based on an interdigitated electrode (IDE) was used as a highly sensitive immunoassay for carcinogenic embryonic antigen (CEA) through the quantification of 3,3',5,5'-tetramethylbenzidine (TMB). For the redox cycling process, one pair of interdigitated finger electrodes was used as the first working electrode (generator) for cyclic voltammetry of TMB, and another pair of interdigitated finger electrodes was used as the second working electrode (collector) for sequential application of potentials for reduction and oxidation of TMB. The reduction (and oxidation) products of TMB at the collector were supplied to the generator, and following sequential oxidization (and reduction) at the generator, again supplied to the collector. Such redox recycling processes between the generator and collector allowed signal amplification. In this work, the influences of the following factors on the redox cycling of TMB were analyzed: (1) the redox potential at the collector, (2) the gap between the interdigitated finger electrodes, and (3) the scan rate of the generator. The redox potential and electrode gap influences were simulated with COMSOL software and compared with empirical results. At the optimum redox potentials and electrode gap, redox cycling was estimated to be five-fold more sensitive for the quantification of TMB than conventional cyclic voltammetry using one pair of interdigitated finger electrodes as the working electrode. Finally, redox cycling was applied to a commercial immunoassay for CEA, and the sensitivity of redox cycling was three-fold higher than that of conventional cyclic voltammetry using a single set of interdigitated finger electrodes as the working electrode., (Copyright © 2017 Elsevier B.V. For label-free and direct detection of C-reactive protein (CRP), an impedimetric sensor based on an indium tin oxide (ITO) electrode array functionalized with reduced graphene oxide-nanoparticle (rGO-NP) hybrid was fabricated and evaluated. Analytical measurements were performed to examine the properties of rGO-NP-modified ITO microelectrodes and to determine the influence upon sensory performance of using nanostructures modified for antibody immobilization and for recognition of CRP binding events. Impedimetric measurements in the presence of the redox couple [Fe(CN)6](3-/4-) showed significant changes in charge transfer resistance upon binding of CRP. The impedance measurements were highly target specific, linear with logarithmic CRP concentrations in PBS and human serum across a 1 ng mL(-1) and 1000 ng mL(-1) range and associated with a detection limits of 0.06 and 0.08 ng mL(-1) respectively., (Copyright © 2015 Elsevier B.V. A capacitive biosensor based on an interdigitated electrode (IDE) with nanoislands was developed for label-free detection of antigen-antibody interactions. To enable sensitive capacitive detection of protein adsorption, the nanoislands were fabricated between finger electrodes of the IDE. The effect of the nanoislands on the sensitive capacitive measurement was estimated using horseradish peroxidase (HRP) as a model protein. Additionally, a parylene-A film was coated on the IDE with nanoislands to improve the efficiency of protein immobilization. By using HRP and hepatitis B virus surface antigen (HBsAg) as model analytes, the effect of the parylene-A film on the capacitive detection of protein adsorption was demonstrated., (Copyright © 2014 Elsevier B.V. Label-free and real-time monitoring of stem cells based on electrical impedance measurement is increasingly utilized for the quality control of the isolated stem cells to be used in stem cell-based tissue therapy or regenerative medicine. In spite of that the proliferative capacity and multipotency of stem cells are dependent on the type and age of the source tissue, however, the effect of the cell senescence on the impedance measurement of stem cells has not yet been studied. We investigated whether the senescence of adipose tissue-derived stem cells (ADSCs) can be detected by electrical impedance spectroscopy. For this, ADSCs at passage 9 and 31 were prepared and those genetic characteristics and growth kinetics were evaluated by quantitative polymerase chain reaction and cell counting. While the identified ADSCs were grown on the indium tin oxide electrodes, the impedance spectra were measured and interpreted by fitting analysis with an equivalent circuit model. ADSCs at passage 9 adhered on the electrode were small and spindle-shaped whereas the cells at passage 31 were flattened and larger than younger cells. At the beginning of culture time when the cell adhesion occurred, the resistance at 4.6kHz of passage 31 cells was higher than passage 9 due to the larger size of older cells. Afterwards, the value of passage 9 cells increased higher than passage 31, since younger cells proliferated more than old cells. Objective: The aim of this in vitro study was to investigate the feasibility of a new highly flexible microelectrode on human tissue and its potential of differentiating atherosclerotic lesions by electric impedance spectroscopy (EIS)., Methods: Electric impedance measurements (EIM) were performed on 148 spots of 7 aortic and 6 femoral human arteries at 1kHz, 10kHz and 100kHz., Results: According to the AHA classification 33 (25%) grade I lesions (PI), 34 (26%) grade II (PII), 21 (16%) grade III (PIII), 21 (16%) grade IV (PIV), 13 (10%) grade Va (PVa) and 10 (8%) grade Vb (PVb) could be identified by histology. At 1kHz, 10kHz and 100kHz the mean electric impedance (MEI) of PI, PII, PIII and PIV was statistically not different. At 100kHz the MEI of PVa showed significantly higher values compared to the MEI of PI (455+/-66Omega vs. 375+/-47Omega, p=0.05), PII (455+/-66Omega vs. 358+/-63Omega, p=0.007), PIII (455+/-66Omega vs. 342+/-52Omega, p=0.003), PIV (455+/-66Omega vs. 356+/-41Omegap=0.013) and the MEI of PVb was significantly increased compared to the MEI of PI (698+/-239Omega vs. 375+/-47Omega, p<0.001), PII (698+/-239Omega vs. 358+/-63Omegap<0.001), PIII (698+/-239Omega vs. 342+/-52Omegap<0.001), PIV (698+/-239Omega vs. 356+/-41Omegap<0.001), PVa (698+/-239Omega vs. 455+/-66Omega, p<0.001). Performing ROC analyses for the detection of grouped PVa/PVb lesions, the largest AUC was found at 100kHz with a cut-off value of 441Omega presenting a sensitivity of 74% and a specificity of 94%., Conclusions: EIM could be performed on human aortic and femoral tissue. The results show that EIS has the potential to distinguish between different plaque types.
