The DNA nanomachines as excellent synthetic biological tools have been widely used for the sensitive detection of intracellular microRNA (miRNA) and DNAzyme-involved gene silencing. However, intelligent DNA nanomachines which have the ability to sense intracellular specific biomolecules and respond to external information in complex environments still remain challenging. Herein, we develop a miRNA-responsive DNAzyme cascaded catalytic (MDCC) nanomachine to perform multilayer cascade reactions, enabling the amplified intracellular miRNA imaging and miRNA-guided efficient gene silencing. The intelligent MDCC nanomachine is designed based on multiple DNAzyme subunit-encoded catalyzed hairpin assembly (CHA) reactants sustained by the pH-responsive Zeolitic imidazolate framework-8 (ZIF-8) nanoparticles. After cellular uptake, the MDCC nanomachine degrades in acidic endosome and releases three hairpin DNA reactants and Zn 2+ , and the latter can act as an effective cofactor for DNAzyme. In the presence of miRNA-21, a catalytic hairpin assembly (CHA) reaction is triggered, which produces a large number of Y-shaped fluorescent DNA constructs containing three DNAzyme modules for gene silencing. The construction of Y-shaped DNA modified with multisite fluorescence and the circular reaction realizes ultrasensitive miRNA-21 imaging of cancer cells. Moreover, miRNA-guided gene silencing inhibits the cancer cell proliferation through the DNAzyme-specific recognition and cleavage of target EGR-1 (Early Growth Response-1) mRNA, which is one key tumor-involved mRNA. Abnormal upregulation of telomerase RNA (TR) is a hallmark event at various stages of tumor progression, providing a universal marker for early diagnosis of cancer. Here, we have developed a triple signal amplification strategy for in situ visualization of TR in living cells, which sequentially incorporated the target-initiated strand displacement circuit, multidirectional rolling circle amplification (RCA), and Mg 2+ DNAzyme-mediated amplification. All oligonucleotide probes and cofactors were transfected into cells in one go, and then escaped from lysosomes successfully. Owing to the specific base pairing, the amplification cascades could only be triggered by TR and performed as programmed, resulting in a satisfactory signal-to-background ratio. Especially, the netlike DNA structure generated by RCA encapsulated high concentrations of DNAzyme and substrates (FQS) in a local region, thereby improving the reaction efficiency and kinetics of the third amplification cycle. Under optimal conditions, the proposed method exhibited ultrasensitive detection of TR mimic with a detection limit at pM level. Developing a simple, economical, and accurate diagnostic method has positive practical significance for the early prevention and intervention of Alzheimer's disease (AD). Herein, combining a closed bipolar electrode (BPE) chip with multicolor electrochemiluminescence (ECL) imaging technology, we constructed a low-voltage driven portable visualized ECL device for the early screening of AD. By introducing parallel resistance, the total resistance of the circuit was greatly reduced. A classical mixture of Ir(ppy) 3 and Ru(bpy) 3 2+ was used as multicolor emitters of the anode with TPrA as the co-reactant. Capture of amyloid-β (Aβ) through antigen-antibody recognition, and signal amplification by electroactive covalent organic frameworks (COF) probe at the cathode of BPE caused the significantly increased faradaic current. The electrical balance of the BPE system resulted in the change of the emission color from green to red at the anode. The ECL-BPE sensor shows good reproducibility and high sensitivity with detection limit of 1 pM by naked eye. The driving voltage is 3.0 V, which means the chip could be driven by two fifth batteries. G-quadruplex secondary structures are naturally found in genome sequences and play essential roles in regulating a wide variety of important biological processes. Although stabilizing effects of monovalent cations (e.g., K + and Na + ) has been recognized during the past decades, a general and reliable analytical method for accurate characterization of specific interactions of K + /Na + with G-quartets is still not well established. In the present study, we demonstrate a practical strategy that combined the use of a nanoscale ion emitter, a low-flow drying gas and a volatile salt (trimethylammonium acetate) to almost totally suppress the nonspecific cationic adduction to G-quadruplexes during the ionization process. Our combined strategy takes full advantage of the ultrasmall initial charged droplets when employing a nanoscale ion emitter, the maximum uneven fission of charged droplets under the gentle desolvation conditions, and the effective shielding of the negatively charged phosphate groups by trimethylammonium ions, to eventually producing ions of G-quadruplexes free of non-specific K + /Na + adduction. For the first time, the accurate binding states as well as the quantitative binding constants between K + /Na + and G-quadruplexes can be directly obtained even in the presence of tens of millimolar non-volatile salts, which has long been a notorious challenge in mass spectrometry., (Copyright © 2022 Elsevier B.V. Herein, a facile and sensitive dual mode sensing strategy for amantadine (AMD) level evaluation by coupling the plasmonic Au nanorod (NR) and supramolecular SH-cyclodextrin (CD) through strong Au-S bond is proposed. Methylene blue (MB) molecules can be inserted into the cavity of CD molecules through the hydrophobic interaction, which would cause the plasmon resonance energy transfer (PRET) process as well as electrochemical signal response due to the spectrum overlap between Au NR and MB molecules and the electrochemical conversion activity of MB molecules. Subsequently, AMD would induce the replacement of MB molecules because of the stronger interaction with CD, resulting the recovery of scattering intensity of Au NR and decrease of the electrooxidation current of MB. On one hand, the increase of Au NR scattering intensity is linearly proportional to AMD with the concentration from 0.4 to 3.0 μM, and reaches a limit of detection (LOD) of 0.28 μM. On the other hand, electrochemical measurement method enlarged the detection range of AMD. The variation of electrochemical oxidation peak current of MB is linearly proportional to the logarithm value of AMD concentration from 2.5 to 375.0 μM, with LOD of 1.9 μM. Subsequently, the proposed dual mode response sensing strategy was successfully employed for the detection of AMD in human serum samples with great selectivity and sensitivity, with a recovery percentage ranged from 92.6 to 112.0%. CRISPR-Cas12a system exhibits tremendous potential in accurate recognition and quantitation of nucleic acids and non-nucleic-acid targets thanks to the discovery of its cleavage capability toward single-stranded DNA (ssDNA). In this study, we developed an efficient electrochemiluminescence (ECL) sensing platform based on CRISPR-Cas12a for the analysis of adenosine triphosphate (ATP). In the presence of the target, the successful release of the DNA activator is specially recognized by Cas12a-crRNA duplex and activates the cleavage of ferrocene (Fc) labeled-ssDNA (Fc-ssDNA) modified on the cathode of bipolar electrode (BPE), resulting in a decrease of ECL intensity of [Ru(bpy) 3 ] 2+ /TPrA in the anodic cell of BPE. By means of the unique combination of Cas12a with ECL technique based on BPE, it can convert the recognition of target ATP into a detectable ECL signal. The detection limit of ATP was determined to be 0.48 nM under the optimal conditions. Nanoparticle plasmon scattering can provide real-time imaging information on the formation process of noble metal-based nanomaterials. Due to the synergistic effect of the interface between metal and oxide supporting pores, metal nanoparticles (NPs), especially Au NPs, generally exhibit higher catalytic activity on oxide carriers than single-component NPs. Here, we use the dark field scattering microscope to in situ monitor the growth of Au on Cu 2 O surface by oxidation-reduction reactions and the nanostructures could be precisely controlled via the scattering signal. The prepared Cu 2 O/Au nanocomposite has a higher electrocatalytic activity toward Glucose. When being used as a potential biosensor for nonenzyme glucose detection, excellent performance, such as high sensitivity with a detection limit of 4 μM, high selectivity and outstanding stability, was obtained. Simultaneous recording of action potentials (APs) and neurotransmitter release is highly desirable in living neurons since it provides a complete framework of the physiological and pathological statuses of nerve cells. In this work, we proposed an approach coupling ultra-thin microelectrode array (MEA) with total internal reflection fluorescence microscopy (TIRFM), which served as a powerful platform to visualize both APs and vesicular exocytosis in a neuronal circuit model formed by neuron-like PC12 cells. Taking advantages of fluorescent false neurotransmitter (FFN), the transient neurotransmitter transport down an axon could be visualized with high spatial and temporal resolution. The real-time recording of APs burst and neurotransmitter release induced by hypoxia with MEA/TIRFM platform reveals the relevance of electrical and chemical activities in the neuronal model. A novel versatile locked nucleic acid modified molecular beacon probe (LNA-MB) was developed for imaging intracellular precursor miRNAs (pre-miRNAs) and disturbing Dicer-mediated cleavage process. The target recognition reaction between the smart probe and pre-miRNA can not only induce the conformational changes of probe and block the Dicer cleavage site, but also inhibit the cleavage process, and then achieve down-regulation of miRNA expression. Simultaneously, the target recognition reaction broke the fluorescence resonance energy transfer (FRET) between fluorophore donor FAM and acceptor TAMRA, which were labelled on the LNA-MB probe, further induced the relevant change of fluorescence signal, and then achieved imaging analysis of pre-miRNA and inhibition events in situ. Both in vitro and in single living cell studies showed that the versatile probes exhibited a remarkable performance in targeting with pre-miRNA-21, and nearly 65% downregulation of mature miRNA-21 was achieved with 100 nM probes. Background: Disulfiram (DSF), a drug widely used to control alcoholism, which has anticancer activity by inducing apoptosis in a copper (Cu)-dependent manner. Numerous evidences from mouse experiments indicated that some anti-cancer agents of chemotherapeutic drugs favor the induction of immunogenic cancer cell death (ICD) leading to tumor-specific immune responses. However, whether DSF could induce the colorectal tumor cells death and the mechanism involved in ICD regulatory remains elusive. The main objective of this study was to elucidate the effect of DSF/Cu on the apoptosis of colorectal cancer (CRC) cells and the expression of the two major ICD markers in CRC cells: calreticulin (CRT) and heat shock proteins (HSP) 70., Methods: Firstly, the toxicity of DSF/Cu in HCT116, SW620 and HCT8 cells was assayed by MTT. Flow cytometry was utilized to detect the apoptosis effects. The effects of DSF/Cu on the expression of ICD-related molecules in tumor tissues were further verified in the CRC xenograft mouse model., Results: The results showed that DSF/Cu increase apoptosis of these three cells in a dose dependent manner and significantly inhibited the proliferation at the concentration range from 0.05 to 1.6 μM. Furthermore, the expression of CRT and HSP70 on the cell surface also increased. The rate of transplanted tumors grew slowly, and the expression of CRT and HSP70 in colorectal cancer tissues was increased after treated with DSF/Cu., Conclusion: In conclusion, our results show that DSF/Cu exerts anti-colorectal cancer and its underlying mechanisms are associated with the enhancement of molecules expression of cell ICD. It is a challenge to ascertain the quality of polysaccharides due to their complex chemical structure; therefore, multi-fingerprint profiling was used to investigate the quality of Astragalus polysaccharides (APS) harvested from Inner Mongolia (NM) and Gansu (GS) with the help of chemometric analysis. Additionally, FT-IR and 1 H NMR were applied to characterize the chemical structure of the harvested APS. The spectral fingerprinting results indicated that APS had reduced similarity when they were from different origins. Further, PCA showed that NM and GS could be distinguished and that the main differences from the loading plots were in the absorption intensity of carbonyls and H1 signals of Galp and β-glucose. Moreover, UPLC/Q-TOF-MS fingerprints were established based on the monosaccharide composition of the APS. The concentration of monosaccharides and results of cluster analysis indicated that GlcA might be an indicator that can be used to distinguish NM and GS. In this paper, a novel prototype liquid-air dual gradient chip is introduced, which has paved the way for effective synergic effect bio-evaluation of air pollutant. The chip is composed of an array of the agarose liquid-air interfaces, top air gradient layer and bottom liquid gradient layer. The novel agarose liquid-air interface allows for non-biased exposure of cells to all the substances in the air and diffusive interactions with the liquid phase; while the dual liquid-air gradient provides powerful screening abilities, which well reduced errors, saved time and cost from repeated experiment. Coupling the two functions, the chip subsequently facilitates synergic effect evaluation of both liquid and air factors on cells. Here cigarette smoke was taken as the model air pollutant, and its strong synergic effects with inflammatory level of A549 lung cancer cells on their fate were successfully quantified for the first time. A paper-based SERS test strip combining strengths of paper chip and Raman active substrate was demonstrated to overcome challenges in spectroscopic sensing of complicated samples and realize quantitative detection of disease markers in whole blood. The precisely controlled Au NPs were not only capable of generating condensed hot spots on the fibers, but also enhanced the size exclusion effect of paper, resulting in the novel performance on both SERS detection and sample pretreatment. A biosensor for Mucin-1 is developed by equipping the Au NPs with aptamer. A simple yet powerful polymerase chain reaction (PCR)-free strategy for visualized assay of human telomerase activity was reported in this work. Gold nanoparticles (AuNPs) based colorimetric strategy was applied with well-designed enzyme-aided cyclic amplification. Briefly, the detection relies on the elongated primers of telomerase substrate (TS) induced by telomerase, which open the hairpin DNA and hybridize with linker DNA, the trigger of AuNPs aggregation. Nicking endonuclease was added in the sensing system, which cleaved linker DNA after hybridization and released complimentary strand for cyclic hybridization with linker DNA, resulted in high sensitivity for the detection of telomerase. Down to 25 HeLa cells with high expression of telomerase could be recognized. The direct separation and analysis of chiral drugs in the complex matrix systems are meaningful and challenging. As the most common broad-spectrum antibiotic, levofloxacin has a strong antibacterial ability, but its enantiomer, dextrofloxacin can cause serious harm to human health. In this work, we reported a rapid on-line extraction/ionization device coupled with Electrospray Mass Spectrometry (ESI-MS) for chiral analysis of ofloxacin enantiomers in complex matrix of milk. Since ofloxacin is difficult to dissolve in water and most organic solvents, the procedure of separating ofloxacin in complex system is often complicated. Using the homemade apparatus, the sample pretreatment process was greatly simplified. Milk sample was directly injected and chiral ofloxacin in the sample was extracted at PTFE membrane for further ionization. It took less than 10s to finish all the procedures including sampling, extraction, reagents mixing, ionization and mass analysis. Utilizing reaction thermodynamics method, trimeric cluster ion [Ni ΙΙ (ref) 2 Ofloxacin-H] + was formed and collisionally dissociated to get chiral resolution of levofloxacin and dextrofloxacin due to the different relative stabilities of the two diastereomeric clusters produced through the dissociation of Ni ΙΙ bound trimeric clusters. A highly sensitive colorimetric/fluorescent dual-mode sensor based on hybridization chain reaction (HCR) combining multifunctional Au NPs is presented for the detection of Hg 2+ in aqueous solution. In Hg 2+ absent solution, the surface of Au NPs was covered by hairpin auxiliary DNAs and a single strand DNA (ssDNA), which prevented Au NPs from salt-induced aggregation. At the same time, the fluorescence intensity of the dye-labeled hairpin probes was significantly quenched by Au NPs. In the presence of Hg 2+ , T-Hg 2+ -T coordination chemistry between helper DNA and the hairpin probes was induced which triggered the formation of extended double-stranded DNA (dsDNA) polymers via HCR. The formed dsDNA polymers were stiffer which couldn't attach to Au NPs, resulting in a red-to-blue color change along with salt-induced aggregation of Au NPs for colorimetric sensing. Meanwhile, the fluorescence of dye-labeled DNA turns on. Due to the HCR amplification effect, a highly sensitive detection of Hg 2+ was achieved with detection limit of 0.1nM. Colorimetry is suitable for the analysis in salt solution with concentration lower than 100mM. It serves as an intuitive method that Hg 2+ down to 1.0nM could be identified by naked eyes. For high-concentration salt solutions such as industrial effluent, fluorescent sensing acts as a better choice. Both the color and fluorescence changes of the proposed sensor exhibited high selectivity against other metal ions. A highly sensitive electrogenerated chemiluminescence (ECL) biosensor was developed on the basis of a closed bipolar electrode (BPE) apparatus for the analysis of prostate specific antigen (PSA). Bipolar modifications bring up two different stages of enhancement on the same electrode. Anodic enhancement was conducted by modifying gold nanoparticles (Au NPs) to catalyze the anodic ECL reaction between luminol and hydrogen peroxide. Cathodic introduction of thionine tagged PSA antibody led to a further pertinently enhancement synchronized with the PSA amount variation, because the existence of thionine greatly increased the rate of electron gains on cathode, leading to the corresponding acceleration of anodic ECL reaction. The more thionine modified target molecules were introduced, the faster luminol was oxidized, the higher faraday current approached, and sensitive quantification was realized in correlation with the responsive ECL intensity differences. The quantification resulted in a good determination range between 0.1pg/mL and 0.1µg/mL. This strategy mainly took advantage of the special structure of closed BPE to realize a simultaneous amplification on both ends of BPE. In this work, we report a novel microfluidic gas collecting platform aiming at simultaneous sample extraction and multiplex mass spectrometry (MS) analysis. An alveolar-mimicking elastic polydimethylsiloxane (PDMS) structures was designed to move dynamically driven by external pressure. The movement was well tuned both by its amplitude and rhythm following the natural process of human respiration. By integrating the alveolar units into arrays and assembling them to gas channels, a cyclic contraction/expansion system for gas inhale and exhale was successfully constructed. Upon equipping this system with a droplet array on the alveolar array surface, we were able to get information of inhaled smoke in a new strategy. Here, with cigarette smoke as an example, analysis of accumulation for target molecules during passive smoking is taken. Relationships between the breathing times, distances away from smokers and inhaled content of nicotine are clarified. Further, by applying different types of extraction solvent droplets on different locations of the droplet array, simultaneous extraction of nicotine, formaldehyde and caproic acid in sidestream smoke (SS) are realized. Since the extract droplets are spatially separated, they can be directly analyzed by MS which is fast and can rid us of all complex sample separation and purification steps. In this work, poly-thymine stabilized copper nanoclusters have been used as a fluorescence probe for melamine sensing for the first time. Melamine can bind to thymine through hydrogen bond, which could dramatically enhance the fluorescence intensity of poly-thymine stabilized copper nanoclusters. The enhancement factors (I-I0)/I0 increase linearly with the lgCmelamine over the melamine concentration range of 0.1 µM to 6 µM. The detection limit of melamine is 95 nM, which is 200 times lower than the US Food and Drug Administration estimate melamine safety limit 20 µM. Melamine in milk was detected with good recovery, which suggested that this novel fluorescence probe has great potential in practical application., (Copyright © 2015 Elsevier B.V. Here we have developed a sensitive cancer cell amplified detection method which combined Au NPs enhanced electrochemiluminescence (ECL) of CdS nanocrystals (NCs) film, with isothermal circular amplification reaction of polymerase. In DNA circular amplification detection system, hairpin DNA beacon/Au NPs composite modified CdS NCs film was used as an ECL emitter. Messenger DNA is hybridized with the aptamer modified on magnetic beads (MBs) to form MB-Au bioconjugates. In the presence of HL-60 cell, the aptamer would conjugate with the glycoprotein at cell surface and messenger DNA sequence would be released. The released messenger DNA sequence was then introduced into the cycle amplification system to trigger circular polymerizations. A novel strategy for preparing aptamer-based organic-silica hybrid monolithic column was developed via "thiol-ene" click chemistry. Due to the large specific surface area of the hybrid matrix and the simplicity, rapidness and high efficiency of "thiol-ene" click reaction, the average coverage density of aptamer on the organic-silica hybrid monolith reached 420 pmol μL(-1). Human α-thrombin can be captured on the prepared affinity monolithic column with high specificity and eluted by NaClO4 solution. N-p-tosyl-Gly-Pro-Arg p-nitroanilide acetate was used as the sensitive chromogenic substrate of thrombin. The thrombin enriched by this affinity column was detected with a detection of limit of 0.01 μM by spectrophotometry. Furthermore, the extraction recovery of thrombin at 0.15 μM in human serum was 91.8% with a relative standard deviation of 4.0%. These results indicated that "thiol-ene" click chemistry provided a promising technique to immobilize aptamer on organic-inorganic hybrid monolith and the easily-assembled affinity monolithic material could be used to realize highly selective recognition of trace proteins., (Copyright © 2015 Elsevier B.V. In this work, a sensitive and selective detection method based on fluorescence resonance energy transfer (FRET) was developed for analyzing thiol compounds by using a novel fluorescent probe. The new fluorescent probe contains a disulfide bond which selectively reacts with nucleophilic thiolate through the thiol-disulfide exchange reaction. An obvious fluorescence recovery can be observed upon addition of the thiol compound in the fluorescent probe solution due to the thiol-disulfide exchange reaction and the destruction of FRET. This novel probe was successfully used to determine dithiothreitol (DTT), glutathione (GSH) and cysteine (Cys). The limits of detection (LOD) were 2.0μM for DTT, 0.6μM for GSH, and 0.8μM for Cys. Carbon decorated Fe3O4 nanoparticles (Fe3O4/C) are promising magnetic solid-phase extraction (MSPE) sorbents in environmental and biological analysis. Fe3O4/C based MSPE method shows advantages of easy operation, rapidness, high sensitivity, and environmental friendliness. In this paper, the MSPE mechanism of Fe3O4/C nanoparticles has been comprehensively investigated, for the first time, through the following three efforts: (1) the comparison of extraction efficiency for polycyclic aromatic hydrocarbons (PAHs) between the Fe3O4/C sorbents and activated carbon; (2) the chromatographic retention behaviors of hydrophobic and hydrophilic compounds on Fe3O4/C nanoparticles as stationary phase; (3) related MSPE experiments for several typical compounds such as pyrene, naphthalene, benzene, phenol, resorcinol, anisole and thioanisole. It can be concluded that there are hybrid hydrophobic interaction and hydrogen bonding interaction or dipole-dipole attraction between Fe3O4/C sorbents and analytes. In this work, a rapid, sensitive and low-cost colorimetric method for detection of quaternary ammonium surfactants using citrate-capped silver nanoparticles (Ag NPs) was developed. The quaternary ammonium surfactants induce the aggregation of Ag NPs through the hydrophobic effect, which is a novel aggregation mechanism of Ag NPs. The addition of cationic surfactant results in color change of Ag NPs solution from yellow to red and finally to colorless, which is due to the broadening of the surface plasmon band. The color change was monitored using a UV-vis spectrophotometer. The LOD of different cationic surfactants was in the range of 0.5-5 µM. Aims: Indoleamine 2,3-dioxygenase (IDO) inhibits T-cell proliferation by catalyzing the conversion of l-tryptophan to l-kynurenine. IDO-induced immune tolerance weakens the clinical outcomes of immunotherapies. Sodium butyrate (NaB), one of the histone deacetylase inhibitors (HDACIs), has potential anti-tumor effects. Our previous studies revealed that NaB could inhibit IFN-γ induced IDO expression in nasopharyngeal carcinoma cells, CNE2. In the present study, we aim to investigate to the mechanism of NaB interfering with the interferon-gamma (IFN-γ)-mediated IDO expression signaling transduction., Main Methods: IDO expression and STAT1 phosphorylation in CNE2 cells were analyzed by western blotting and STAT1 acetylation was evaluated by immunoprecipitation. STAT1 nuclear translocation and NF-κB activity were detected by transient transfection and reporter gene assay., Key Findings: We found that NaB inhibited IFN-γ-induced IDO expression in CNE2 cells via decreasing phosphorylation and nuclear translocation of STAT1, but not via down-regulation of IFN-γ-receptor (IFNGR). Immunoprecipitation assays revealed that NaB increased STAT1 acetylation. Furthermore, NaB elevated the activity of NF-κB in CNE2 cells, and blocking the NF-κB activity had no effect on the IFN-γ-induced IDO expression., Significance: These results suggest that NaB inhibited IFN-γ-induced IDO expression via STAT1 increased acetylation, decreased phosphorylation, and reduced nuclear translocation. A capillary poly(trimethyl-2-methacroyloxyethylammonium chloride-co-ethylene glycol dimethacrylate) monolith was in situ synthesized by thermally initiated free radical co-polymerization using trimethyl-2-methacroyloxyethylammonium chloride (MATE) and ethylene glycol dimethacrylate (EGDMA) as functional monomer and cross-linker, respectively. N,N-dimethylformamide and polyethylene glycol 6000 were used as solvent and porogen, respectively. The morphology and porous structure of the resulting monoliths were assessed by scanning electron microscope. In order to prepare practically useful poly(MATE-co-EGDMA) monoliths with low flow resistance and good mechanical strength, some parameters such as PEG-6000 to DMF ratio, total monomer to porogen ratio, and crosslinker to monomer ratio were optimized systematically. Moreover, the extraction mechanism was evaluated using two series of compounds, alkylbenzenes and weak acids, as model compounds on poly(MATE-co-EGDMA) monoliths as liquid chromatographic stationary phase. Finally, the monoliths were applied as the solid phase microextraction medium, and a simple off-line method for simultaneous determination of three brominated flame retardants, 2,4,6-tribromophenol (TBP), tetrabromobisphenol A (TBBPA) and 4,4'-dibrominated diphenyl ether (DBDPE), in environmental waters was developed by coupling the polymer monolith microextraction to HPLC with UV detection. The regression equations for these three brominated flame retardants showed good linearity from their limit of quantification to 5000ng/mL. The limits of detection were 0.20, 0.15 and 0.10ng/mL for TBP, TBBPA and DBDPE, respectively. A simple and facile needle-adapter was designed for constructing manual on-line polymer monolith microextraction-high performance liquid chromatography (PMME-HPLC). A capillary poly(4-vinylpyridine-co-ethylene glycol dimethacrylate) [poly(VP-co-EGDMA)] monolith was prepared by in situ polymerization, using 4-vinylpyridine (VP) and ethylene glycol dimethacrylate (EGDMA) as functional monomer and cross-linker, respectively. The synthesized monolith was used as the extraction medium for concentrating four EPA priority pollutants, 2-chlorophenol, 2,4-dichlorophenol, 2,4,6-trichlorophenol and pentachlorophenol, from water samples. The effect of the dosage of porogen polyethylene glycol 6000 (PEG) on back pressure and extraction performance of the capillary monolith was also investigated. Moreover, the influences of several parameters (such as extraction time, desorption time, content of MeOH in sample solution and sample pH) were examined to obtain the optimal PMME conditions. As a result, the established on-line PMME-HPLC protocol, with good extraction efficiency (80.6-91.7%), satisfactory recovery (94.7-106% and 76.8-86.3% for water and serum samples, respectively) and low detection limit (0.3-1.4 μg/L), exhibited potential applicability for the analysis of chlorophenols in environmental and biological samples., (Copyright © 2012 Elsevier B.V. The retention factor corresponding to pure water in reversed-phase high performance liquid chromatography (RP-HPLC), k(w), was commonly obtained by extrapolation of retention factor (k) in a mixture of organic modifier and water as mobile phase in tedious experiments. In this paper, a relationship between logk(w) and logk for directly determining k(w) has been proposed for the first time. With a satisfactory validation, the approach was confirmed to enable easy and accurate evaluation of k(w) for compounds in question with similar structure to model compounds. Eight PCB congeners with different degree of chlorination were selected as a training set for modeling the logk(w)-logk correlation on both silica-based C(8) and C(18) stationary phases to evaluate logk(w) of sample compounds including seven PCB, six PBB and eight PBDE congeners. These eight model PCBs were subsequently combined with seven structure-similar benzene derivatives possessing reliable experimental K(ow) values as a whole training set for logK(ow)-logk(w) regressions on the two stationary phases. Consequently, the evaluated logk(w) values of sample compounds were used to determine their logK(ow) by the derived logK(ow)-logk(w) models. The logK(ow) values obtained by these evaluated logk(w) were well comparable with those obtained by experimental-extrapolated logk(w), demonstrating that the proposed method for logk(w) evaluation in this present study could be an effective means in lipophilicity study of environmental contaminants with numerous congeners. As a result, logK(ow) data of many PCBs, PBBs and PBDEs could be offered. Aiming to find an alternative electrochemiluminescence (ECL) quenching route of high efficiency, biocatalytic precipitation (BCP) was firstly coupled with ECL for investigating its insulating effect on ECL. Experimental data revealed that an insulating layer could be formed via BCP onto the electrode surface, inhibiting the reaction between the coreactant and luminophore and thereby impairing the ECL emission dramatically. Since the extent of insulating effect associated strongly with analyte concentration, a new ECL biosensor was successfully realized. In a model horseradish peroxidase (HRP)-based system, the fabricated biosensor possessed high sensitivity and wide linear range from 1.0 × 10(-10)M to 1.0 × 10(-6)M for H(2)O(2) determination with a detection limit of 4 × 10(-11)M (S/N=3). In this paper, gold nanoparticle-thionine-reduced graphene oxide (GNP-THi-GR) nanocomposites were prepared to design a label-free immunosensor for the sensitive detection of carcinoembryonic antigen (CEA). The nanocomposites with good biocompatibility, excellent redox electrochemical activity and large surface area were coated onto the glassy carbon electrode (GCE) surface and then CEA antibody (anti-CEA) was immobilized on the electrode to construct the immunosensor. The morphologies and electrochemistry of the formed nanocomposites were investigated by using scanning electron microscopy (SEM), ultraviolet-visible (UV-vis) spectrometry, electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). CV and differential pulse voltammetry (DPV) studies demonstrated that the formation of antibody-antigen complexes decreased the peak current of THi in the GNP-THi-GR nanocomposites. The decreased currents were proportional to the CEA concentration in the range of 10-500 pg/mL with a detection limit of 4 pg/mL. As a label-free alternative of conventional flow cytometry, chip-based impedance measurement for single cell analysis has attracted increasing attentions in recent years. In this paper, we designed a T-shape microchannel and fabricated a pair of gold electrodes located horizontally on each side of the microchannel using a transfer printing method. Instant electric signals of flowing-through single cells were then detected by connecting the electrodes to a Keithley resistance and capacitance measurement system. Experimental results based on the simultaneous measurement of resistance and capacitance demonstrated that HL-60 and SMMC-7721 cells could be differentiated effectively. Moreover, SMMC-7721 cells at normal, apoptotic and necrotic status can also be discriminated in the flow. We discussed the possible mechanism for the discrimination of cell size and cell status by electrical analysis, and it is believed that the improvement of detection with our design results from more uniform distribution of the electric field. This paper presents a simple and cost-effective UV-ablation technique for fabrication of size-tunable nanofluidics devices via photochemical decomposition reaction. UV-irradiation through a PET photomask results in continuous decomposition of poly(carbonate) (PC), forming nanochannel and carboxyl groups on the surface of the etched PC. This photochemical decomposition process occurs at molecular scale, therefore, the depth of nanochannels can be controlled at nanometer level. The etching rate is estimated to be ca. 0.015 nms(-1). To demonstrate the potential application of the present UV-ablation technique, a nanochannel was fabricated and integrated with microchannels to form a micro/nanofluidics chip for protein concentration. Using this device, about 10(3)-10(5) fold protein concentration can be achieved within 10 min. The present approach offers a simple and practical solution to fabricate nanofluidics devices at low-cost, and the resulting device could provide ideal platforms for μTAS towards various applications in biology and chemistry., (Copyright © 2011 Elsevier B.V. An electrochemical approach for the sensitive detection of sequence-specific DNA has been developed. Horseradish peroxidase (HRP) assembled on the Fe(3)O(4) nanoparticles (NPs) were utilized as signal amplification sources. High-content HRP was adsorbed on the Fe(3)O(4) NPs via layer-by-layer (LbL) technique to prepare HRP-functionalized Fe(3)O(4) NPs. Signal probe and diluting probe were then immobilized on the HRP-functionalized Fe(3)O(4) NPs through the bridge of Au NPs. Thereafter, the resulting DNA-Au-HRP-Fe(3)O(4) (DAHF) bioconjugates were successfully anchored to the gold nanofilm (GNF) modified electrode surface for the construction of sandwich-type electrochemical DNA biosensor. The electrochemical behaviors of the prepared biosensor had been investigated by the cyclic voltammetry (CV), chronoamperometry (i-t), and electrochemical impedance spectroscopy (EIS). Under optimal conditions, the proposed strategy could detect the target DNA down to the level of 0.7 fmol with a dynamic range spanning 4 orders of magnitude and exhibited excellent discrimination to two-base mismatched DNA and non-complementary DNA sequences., (Copyright © 2011 Elsevier B.V. Boronate affinity solid phase microextraction (BA-SPME) is a new format appeared recently with great potential for specific extraction of cis-diol-containing compounds. Unlike conventional SPME, BA-SPME relies on covalent interactions and thereby features with specific selectivity, eliminated matrix effect and manipulable capture/release. However, only on-fiber BA-SPME and its off-line combination with high performance liquid chromatography (HPLC) have been reported so far. In this study, we report on-line coupling of in-tube BA-SPME with HPLC-electrospray ionization tandem mass spectroscopy (in-tube BA-SPME-HPLC-ESI-MS/MS) for the specific and sensitive determination of cis-diol-containing biomolecules. A boronate affinity extraction phase was prepared onto the inner surface of the capillary by copolymerization of vinylphenylboronic acid (VPBA) and ethylene glycol dimethacrylate (EDMA). The extraction conditions were optimized by choosing appropriate extraction/desorption solutions and extraction time. The extraction capacity, linear range, reproducibility and life-time were investigated. The developed method was successfully applied for the determination of dopamine in urine samples. We proposed herein a novel approach for fabricating nanoband microelectrodes for electrochemical detection on an electrophoresis microchip. The metal films were first obtained via region-selective electroless deposition of gold or copper films on PDMS substrates by selective region plasma oxidation through shadow masking. Both metal films show uniform surfaces with the thickness at the level of 100 nm. By casting another PDMS layer on the metal films, the cross section of the sandwich structures can be used as nanoband microelectrodes, which can be renewed just by cutting. These nanoband microelectrodes are successfully used as electrochemical detectors in microchip electrophoresis for the detection of amino acids, proteins and neurotransmitter molecules. Moreover, integrating an Au-Cu double-metal detector with a double-channel electrophoresis system, we can easily distinguish electroactive amino acids from that of non-electroactive amino acids., (Copyright 2010 Elsevier B.V. A novel covalent strategy was developed to modify the poly(dimethylsiloxane) (PDMS) surface. Briefly, dextran was selectively oxidized to aldehyde groups with sodium periodate and subsequently grafted onto amine-functionalized PDMS surface via Schiff base reaction. As expected, the coated PDMS surface efficiently prevented the biomolecules from adsorption. Electro-osmotic flow (EOF) was successfully suppressed compared with that on the native PDMS microchip. Moreover, the stability of EOF was greatly enhanced and the hydrophilicity of PDMS surface was also improved. To apply thus-coated microchip, the separation of peptides, protein and neurotransmitters was investigated in detail. For comparison, these analytes were also measured on the native PDMS microchips. The results demonstrated that these analytes were efficiently separated and detected on the coated PDMS microchips. Furthermore, the relative standard deviations of their migration times for run-to-run, day-to-day, and chip-to-chip reproducibilities were in the range of 0.6-2.7%. In recent years, functionalized magnetic nanoparticles (MNPs) have drawn continuously increasing attention due to their great potential for capturing biological molecules or species. However, functionalized MNPs as nanoextraction probes and the coupling with a separation platform for chemical analysis have not extensively investigated yet. In this study, boronate functionalized MNPs were synthesized and employed as extracting probes to capture and enrich cis-diol-containing biomolecules, and an off-line coupling method of the MNPs-based extraction with capillary electrophoresis (CE) was established by using pH junction, an on-line preconcentration technique in CE, as a bridge for the coupling. The prepared MNPs exhibited specific selectivity and sufficient capacity. The pH junction compressed a large injected sample volume into a much narrower sample zone and therefore significantly improved the detection sensitivity, solving the sensitivity mismatch between the MNPs-based extraction and CE. Experimental conditions for the pH junction and the desorption were optimized. Under the optimized conditions, the sensitivity was enhanced by 42-fold as compared with regular CE. N,N-dimethylformamide was found to be an effective desorption promoter, which reduced the desorption time to a few minutes. With the established method, riboflavin in a human urine sample was determined.

A new format of solid phase microextraction (SPME), boronate affinity SPME, was proposed for the first time for covalent extraction of cis-diol containing biomolecules. This new SPME format is based on the reversible complex formation between boronic acids and 1,2- and 1,3-cis-diols. The complex formation and dissociation can be facilely controlled by changing pH. An extracting phase of poly-3-aminophenylboronate (polyAPBA) electrochemically deposited on a metal wire was employed to demonstrate the concept of this new methodology. Catechol and riboflavin were used as the test analytes, and the SPME extraction was combined off-line with high-performance liquid chromatographic (HPLC) separation followed by UV absorbance or fluorescence detection. Fundamental aspects, such as selectivity, extraction/desorption equilibrium, linearity, effect of competing compounds, reproducibility and life-time, were first investigated. Then the developed method was applied to beer samples since the content of riboflavin plays an important role in the flavor stability of beverages. Excellent performance of the SPME fibers was observed for both standard and real samples. Particularly, the expected excellent features of the polyAPBA extracting phase were experimentally verified, which include specific selectivity, eliminated matrix effect and manipulable capture/release. The new methodology of SPME can be a promising tool since a lot of 1,2- and 1,3-cis-diol-containing compounds are of great biological importance.

We report a simple and novel method of stirring-only-driven accumulation and electrochemical determination of arsenite (As(III)) with both of the oxidation and reduction peaks associated with As(0)/As(III) using a gold nanofilm electrode in neutral solution. Under stirring, a large amount of As(III) was deposited on the modified electrode and the electrochemical response was greatly amplified. The accumulated As(III) on the electrode showed well-defined redox couple in 0.1M blank phosphate buffer solution (pH 7.0), which could be used for the measurement of As(III). Under optimal conditions, As(III) could be detected in the range from 0.20 to 375 ppb with a detection limit of 0.04 ppb. In particular, with the use of the reduction peak of As(III) the modified electrode exhibits excellent performance for As(III) determination even in the presence of abundant Cu(II). The regeneration of the electrodes is facile with good reproducibility. The electrochemical system was applied to analyze As(III) in lake water, As(III) spiked tap water and drinking water.

Single-walled carbon nanotubes (SWCNTs) selectively wrapped by a water-soluble, environmentally friendly, biocompatible polymer chitosan (CHI) were employed for the construction of a bioelectrochemical platform for the direct electron transfer (DET) of glucose oxidase (GOD) and biosensing purposes. Scanning electron microscopy and Raman spectroscopy were used to investigate the properties of the SWCNT-CHI film. The results show that the preferentially wrapped small-diameter SWCNTs are dispersed within the CHI film and exist on the surface of the electrode as small bundles. The DET between GOD and the electrode surface was observed with a formal potential of about ca. -460 mV vs. SCE in phosphate buffer solution. The heterogeneous electron transfer rate constant and the surface coverage of GOD are estimated to be 3.0 s(-1) and 1.3 x 10(-10)mol/cm(2), respectively. The experimental results demonstrate that the immobilized GOD retains its catalytic activity towards the oxidation of glucose. Such a GOD/SWCNT-CHI film-based biosensor not only exhibits a rapid response time, a wide linear rang and a low detection limits at a detection potential of -400 mV but also shows the effective anti-interference capability. Significantly improved analytical capabilities of the GOD/SWCNT-CHI/GC electrode could be ascribed to the unique properties of the individual SWCNTs and to the biocompatibility of CHI.

Human erythropoietin (hEPO) is a glycoprotein hormone produced primarily by the kidney, which stimulates red blood cell production. Recombinant human erythropoietin (rhEPO), generally produced in Chinese hamster ovary (CHO) cells, can be used as not only a therapeutic protein but also a doping agent in sports. Profiling of EPO glycoforms is a critical means for quality control in pharmaceutical industrial and anti-doping analysis of misuse in sports. However, the existing methods for the analysis of EPO are associated with either time consuming or poor resolution. In this work, a rapid and high-resolution glycoform profiling method was presented based on capillary isoelectric focusing (cIEF) with whole column imaging detection (WCID). Experimental conditions that influence the separation were investigated. Under optimized conditions, rhEPO from three different sources were resolved into distinct populations within 5 min with excellent reproducibility. As compared with existing methods, the presented method exhibited the advantages of speed and high resolution. If combined with an effective sample enrichment step and a much more sensitive WCID version, the method can be a potential alternative for the detection of rhEPO misuse in sports.

In this work, chloride, chlorate and perchlorate are fast separated on PDMS microchip and detected via in-channel indirect amperometric detection mode. With PDMS/PDMS microchip treated by oxygen plasma, anions chloride (Cl-), chlorate (ClO3-), and perchlorate (ClO4-) are separated within 35s. Some parameters including buffer salt concentration, buffer pH, separation voltage and detection potential are investigated in detail. The separation conditions using 15 mM (pH 6.12) of 2-(N-morpholino)ethanesulfonic acid (MES)+L-histidine (L-His) as running buffer, -2000 V as separation voltage and 0.7 V as detection potential are optimized. Under this condition, the detection limits of Cl-, ClO3-, and ClO4- are 1.9, 3.6, and 2.8 microM, respectively.

Direct electrochemistry of hemoglobin (Hb) was observed at glassy carbon electrode (GCE) modified with undoped nanocrystalline diamond (UND) and Hb multilayer films via layer-by-layer assembly. UV-VIS absorbance spectroscopy, electrochemical impedance spectroscopy and cyclic voltammograms were employed to characterize the film. The results showed that the UND had the effect of enhancing the electron transfer between Hb and the electrode surface. Hb in the multilayer films maintained its bioactivity and structure. It also exhibited a good catalytic activity towards the reduction of H(2)O(2). The reciprocal of catalytic current showed a linear dependence on the reciprocal of H(2)O(2) concentration ranging from 0.5 microM to 0.25 mM with a detection limit of 0.4 microM. The apparent Michaelis-Menten constant was estimated to be 0.019 mM.

This paper proposed a method with capillary zone electrophoresis (CZE) for analysis of eight heterocyclic amines in a commercial meat matrix. The influence of composition, pH and concentration of buffer, and applied voltage were investigated. A 5 mmol/L formic acid-ammonium formate solution at pH 2.20 was chosen as the running electrolyte. Also several solid-phase extraction actions were performed for the clean-up of the samples. With 3s hydrodynamic injection, detection limits ranging from 0.554 to 1.783 microg/g was obtained. To improve sensitivity, field-amplified sample injection (FASI) was used with the conditions of 3 s hydrodynamic injection of a water plug and 25 s electrokinetic injection of the sample. And methanol-water (1:1 in volume) was applied as the sample solvent. Under above conditions, detection limits ranged from 1.329 to 19.39 ng/g, which were at least 50 times lower than those with hydrodynamic injection.

The separation of three kinds of aminophenol isomers were achieved within 1min in polyelectrolytes multilayers modified PDMS microchips by layer-by-layer assembly with electrochemical detection (EC). Two polyelectrolytes, poly(dially dimethyl ammonium chloride) (PDDA) and poly(sodium-4-styrene-sulfonate) (PSS) were used to form polyelectrolyte multilayers (PEMs). The surface characteristic of the modified microchip was studied by XPS. The electroosmotic flow (EOF) on PEMs modified PDMS microchips was more stable than that of the native PDMS microchips and the adsorption of samples was greatly reduced on PEMs modified PDMS microchips during the electrophoretic process. The column efficiencies on PEMs modified microchip were increased by 100 times and the signals enhanced by 2 times compared with those of native microchips. The separation conditions such as running buffer pH, running buffer concentration and separation voltage were also optimized.

A novel film of chitosan-gold nanoparticles is fabricated by a direct and facile electrochemical deposition method and its application in glucose biosensor is investigated. HAuCl(4) solution is mixed with chitosan and electrochemically reduced to gold nanoparticles, which can be stabilized by chitosan and electrodeposited onto glassy carbon electrode surfaces along with the electrodeposition of chitosan. Then a model enzyme, glucose oxidase (GOD) is immobilized onto the resulting film to construct a glucose biosensor through self-assembly. The resulting modified electrode surfaces are characterized with both AFM and cyclic voltammetry. Effects of chitosan and HAuCl(4) concentration in the mixture together with the deposition time and the applied voltage on the amperometric response of the biosensor are also investigated. The linear range of the glucose biosensor is from 5.0 x 10(-5) approximately 1.30 x 10(-3) M with a Michaelis-Menten constant of 3.5 mM and a detection limit of about 13 microM.

In this paper, a simple and green modification method is developed for biomolecules analysis on poly(dimethylsiloxane) (PDMS) microchip with successful depression of nonspecific biomolecules adsorption. O-[(N-succinimdyl)succiny]-o'-methyl-poly(ethylene glycol) was explored to form hydrophilic surface via in-situ grafting onto pre-coated chitosan (Chit) from aqueous solution in the PDMS microchannel. The polysaccharide chains backbone of Chit was strongly attracted onto the surface of PDMS via hydrophobic interaction combined with hydrogen bonding in an alkaline medium. The methyl-poly(ethylene glycol) (mPEG) could produce hydrophilic domains on the mPEG/aqueous interface, which generated brush-like coating in this way and revealed perfect resistance to nonspecific adsorption of biomolecules. This strategy could greatly improve separation efficiency and reproducibility of biomolecules. Amino acids and proteins could be efficiently separated and successfully detected on the coated microchip coupled with end-channel amperometric detection at a copper electrode. In addition, it offered an effective means for preparing biocompatible and hydrophilic surface on microfluidic devices, which may have potential use in the biological analysis.

A mixture of five amino acids including arginine, histidine, phenylalanine, serine and glutamic acid was successfully separated in microchip capillary electrophoresis and detected with laser-induced fluorescence (LIF) detector. These amino acids were labeled with 5-(4, 6-dichloro-s-triazin-2-ylamino) fluorescein (DTAF). The analyses were performed on two kinds of modified poly(dimethylsiloxane) (PDMS) microchips. One kind of chip was simply treated with oxygen plasma (OP-chip), and the other was further modified by coating double layers of non-ionic polymer poly(vinyl alcohol) (PVA) after plasma oxidization (PVA-chip). The derivatization condition of amino acids by DTAF was optimized. The properties of the two modified PDMS microchips were studied and separation conditions, such as the buffer pH, buffer concentration and separation voltage, were also optimized. The column efficiencies of the two microchips were in the range of 193,000-1,370,000 plates/m. The DTAF-labeled amino acids were sufficiently separated within 50s and 90s in 2.5cm channels on OP-chip and PVA-chip, respectively.
