Abstract

Prediction of retention times (RTs) is increasingly considered in untargeted metabolomics to complement MS/MS matching for annotation of unidentified peaks. We tested the performance of PredRet (http://predret.org/) to predict RTs for plant food bioactive metabolites in a data sharing initiative containing entry sets of 29–103 compounds (totalling 467 compounds, >30 families) across 24 chromatographic systems (CSs). Between 27 and 667 predictions were obtained with a median prediction error of 0.03–0.76 min and interval width of 0.33–8.78 min. An external validation test of eight CSs showed high prediction accuracy. RT prediction was dependent on shape and type of LC gradient, and number of commonly measured compounds. Our study highlights PredRet’s accuracy and ability to transpose RT data acquired from one CS to another CS. We recommend extensive RT data sharing in PredRet by the community interested in plant food bioactive metabolites to achieve a powerful community-driven open-access tool for metabolomics annotation., The authors acknowledge networking support by the European Cooperation in Science and Technology (COST) Action FA 1403 POSITIVe (Interindividual Variation in Response to Consumption of Plant Food Bioactives and Determinants Involved). Abstract

Este artículo contiene 16 páginas, 6 figuras, 2 tablas., Soil degradation by water is a serious environmental problem worldwide, with specific climatic factors being the major causes. We investigated the relationships between synoptic atmospheric patterns (i.e. weather types, WTs) and runoff, erosion and sediment yield throughout the Mediterranean basin by analyzing a large database of natural rainfall events at 68 research sites in 9 countries. Principal Component Analysis (PCA) was used to identify spatial relationships of the different WTs including three hydro-sedimentary variables: rainfall, runoff, and sediment yield (SY, used to refer to both soil erosion measured at plot scale and sediment yield registered at catchment scale). The results indicated 4 spatial classes of rainfall and runoff: (a) northern sites dependent on North (N) and North West (NW) flows; (b) eastern sites dependent on E and NE flows; (c) southern sites dependent on S and SE flows; and, finally, (d) western sites dependent on W and SW flows. Conversely, three spatial classes are identified for SY characterized by: (a) N and NE flows in northern sites (b) E flows in eastern sites, and (c) Wand SW flows in western sites. Most of the rainfall, runoff and SY occurred during a small number of daily events, and just a few WTs accounted for large percentages of the total. Our results confirm that characterization by WT improves understanding of the general conditions under which runoff and SY occur, and provides useful information for understanding the spatial variability of runoff, and SY throughout the Mediterranean basin. Their suitability regarding both yield and environmental (e.g. nitrate leaching and ammonia volatilization) sustainability, and regional barriers and opportunities for their implementation were also considered. Based on its results best strategies to abate GHG emissions in Mediterranean agro-systems were proposed. Adjusting N fertilization to crop needs in both irrigated and rain-fed systems could reduce N2O emissions up to 50% compared with a non-adjusted practice. Substitution of N synthetic fertilizers by solid manure can be also implemented in those systems, and may abate N2O emissions by about 20% under Mediterranean conditions, with additional indirect benefits associated to energy savings and positive effects in crop yields. The use of urease and nitrification inhibitors enhances N use efficiency of the cropping systems and may mitigate N2O emissions up to 80% and 50%, respectively. The type of irrigation may also have a great mitigation potential in the Mediterranean region. Drip-irrigated systems have on average 80% lower N2O emissions than sprinkler systems and drip-irrigation combined with optimized fertilization showed a reduction in direct N2O emissions up to 50%. Methane fluxes have a relatively small contribution to the total GHG budget of Mediterranean crops, which can mostly be controlled by careful management of the water table and organic inputs in paddies. Reduced soil tillage, improved management of crop residues and agro-industry by-products, and cover cropping in orchards, are the most suitable interventions to enhance organic C stocks in Mediterranean agricultural soils. The adoption of the proposed agricultural practices will require farmers training. The global analysis of life cycle emissions associated to irrigation type (drip, sprinkle and furrow) and N fertilization rate (100 and 300 kg N ha−1 yr−1) revealed that these factors may outweigh the reduction in GHG emissions beyond the plot scale. The analysis of the impact of some structural changes on top-down mitigation of GHG emissions revealed that 3–15% of N2O emissions could be suppressed by avoiding food waste at the end-consumer level. A 40% reduction in meat and dairy consumption could reduce GHG emissions by 20–30%. Abstract

At a world scale, tomato is an important horticultural crop, but its productivity is highly reduced by drought stress. Combining the application of beneficial microbial inoculants with breeding and grafting techniques may be key to cope with reduced tomato yield under drought. This study aimed to investigate the growth responses and physiological mechanisms involved in the performance under drought stress of four tomato recombinant inbred lines (RIL) after inoculation with the arbuscular mycorrhizal (AM) fungus Rhizophagus irregularis and the plant growth promoting rhizobacteria (PGPR) Variovorax paradoxus 5C-2. Results showed a variation in the efficiency of the different tomato RILs under drought stress and a differential effect of the microbial inoculants, depending on the RIL involved. The inoculants affected plant parameters such as net photosynthetic capacity, oxidative damage to lipids, osmolyte accumulation, root hydraulic conductivity or aquaporin abundance and phosphorylation status. RIL66 was the one obtaining maximum benefit from the microbial inoculants under drought stress conditions, due likely to improved CO2-fixation capacity and root hydraulic conductivity. We propose that RIL66 could be selected as a good plant material to be used as rootstock to improve tomato growth and productivity under water limiting conditions. Abstract

Elicited pumpkin was evaluated as a potential daily consumption product able to modulate the gut microbiota. An in vitro dynamic colonic fermentation performance with microbiota from obese volunteers was used. Prebiotic effects were observed after the pumpkin treatment. Bifidobacterium abundance was maintained during the treatment period whereas Lactobacillus increased in the transversal and descending colon. Conversely, Enterobacteriaceae and Clostridium groups were more stable, although scarce decreasing trends were observed for same species. Increments of Lactobacillus acidophilus and Limosilactobacillus fermentum (old Lactobacillus fermentum) were observed in the whole colonic tract after the treatment period. However, modulatory effects were mainly observed in the transversal and descending colon. Diverse bacteria species were increased, such as Akkermansia muciniphila, Bacteroides dorei, Cloacibacillus porcorum, Clostridium lactatifermentans, Ruminococcus albus, Ruminococcus lactaris, Coprococcus catus, Alistipes shahii or Bacteroides vulgatus. The prebiotic effect of the elicited pumpkin was provided by the fiber of the pumpkin, suggesting a release of pectin molecules in the transversal and distal colonic tract through low cellulosic fiber degradation, explaining the increases in the total propionic and butyric acid in these colonic sections. Also, a possible modulatory role of carotenoids from the sample was suggested since carotenes were found in the descending colon. Abstract

Foliar fertilization is becoming very popular since it is highly efficient, minimizes contamination, and is target oriented. However, the low mobility of several nutrients and low penetration through the leaf cuticle should be addressed, to establish an efficient leaf applied fertilizer. During the last few years, nanotechnologies - such as nanocarriers - have been studied with regard to improving the penetration and delivery of compounds. This work describes the use of membrane vesicles obtained from Brassica oleracea L. as nanobiocarriers of Zn and the evaluation of their potential as a foliar fertilizer, also in Brassica. The results show a high Zn encapsulation efficiency and high delivery into protoplasts. Abstract

The effect of the pH and the type of acidulant (citric acid or glucono-δ-lactone) of the recovery medium on the thermal resistance of Bacillus stearothermophilus ATCC 12980 was studied. The spores were heated in bidistilled water as a reference substrate and in acidified mushroom extract using citric acid or glucono-δ-lactone as acidulants (pH 6.2) and subcultured in reference (pH 7) and acidified (pH 6.2) media. A period of treatment-dependent pH sensitization was observed in heat-treated spores. In all cases, D values were lower in the acidified recovery medium than those obtained in the reference medium, but the type of acidulant used in the recovery medium had no influence on the D values. No influence on z values was observed as a consequence of the different recovery media, but they changed within a range of 7 to 10°C as a function of the different heating substrates. Glucono-δ-lactone proved to be as effective as citric acid in controlling the microbiological spoilage of foods. The pH has a great influence on decreasing the D values, mainly when acidification of the substrate and acidification of the recovery medium are combined. Consequently, it would be appropriate to take this effect into account in process calculations or validation.

8. Thermal Resistance of Bacillus stearothermophilus Spores Heated in Acidified Mushroom Extract.

Abstract

The thermal resistance of Bacillus stearothermophilus spores was studied in bidistilled water as the reference medium, mushroom extract and acidified mushroom extract. Citric acid and glucono-δ-lactone were used as acidulants. Results indicated that mushroom extract affects the heat resistance of spores; D values were lower than for those in bidistilled water. The pH effect was lower with higher treatment temperatures. Acidification reduced the thermal resistance of spores, the reduction being similar for both types of acidulants, and in general it also had the effect of increasing the z values. It has been confirmed that acidification of the canned mushrooms could actually help to control the thermophilic spoilage. This acidification could also be obtained by the use of glucono-δ-lactone, which has been shown to be as effective as citric acid in reducing the heat resistance of spores.

Abstract

Spores of Clostridium botulinum 213B were heated in mushroom extract acidified to pH 6 with citric acid or glucono-deltalactone at temperatures of 121.1, 125, and 130°C using a thermoresistometer. Decimal reduction times were similar in acidified and natural pH (6.7) mushroom extract. At 121.1, 125, and 130°C, D values were in the range 2.44 - 2.55 s, 0.91-1.45 s, and 0.51-0.75 s, respectively. There was no evidence that mild acidification reduced thermal resistance at high temperatures.
