1. Spontaneous cervical and mediastinal hematoma due to rupture of inferior thyroid artery

Abstract

A 62-year-old man was referred to our hospital presenting with a sore throat, dyspnea, and cervical swelling. Initial precontrast CT scans revealed a cervical and mediastinal hematoma, along with a hemothorax. Further dynamic contrast-enhanced CT scans indicated contrast media extravasation dorsal to the right thyroid gland lobe, suggesting a rupture of the right inferior thyroid artery or a parathyroid adenoma. Following endotracheal intubation, angiography confirmed extravasation from the right inferior thyroid artery. Transarterial embolization (TAE) was successfully performed using a gelatin sponge. The cervical and mediastinal hematoma were surgically excised, and the right inferior parathyroid gland was simultaneously resected. Pathological examination revealed no neoplastic components.

2. The price of attention: An analysis of the intersection of media coverage and public sentiments about eggs and egg prices

Abstract

Retail prices for eggs surged during the period from early 2022 to mid-2023 in the U.S. We investigate the impact of egg price fluctuations on online and social media discourse, analyzing the relationship between egg prices and public sentiment. Utilizing online and social media listening data from September 2019 to August 2023, we explore how the total number of statements (i.e., mentions) and sentiment respond to changes in egg prices in the U.S. We find a significant association between increases in egg prices and both the volume of mentions and the sentiment of online discussions. Notably, mentions increased and sentiment became more negative as egg prices rose, highlighting a clear public response to price changes. However, the relationship between egg prices and online and social media attention is complex, which becomes apparent when studying the timing of increased concern with the timing of online news media coverage. Importantly, using regression discontinuity in time, we show that online and social media conversations about eggs and egg prices tend to increase after the rapid rise in online news coverage. Similarly, online and social media conversations about eggs and egg prices tend to decrease after the rapid rise in online news coverage.

Abstract

Background: The shoulder motion during pitching is influenced by the trunk and pelvis motions, but their relationship during the deceleration phase of throwing on flat ground has not yet been clarified. This study aimed to investigate the relationship between shoulder, trunk, and pelvis kinematics at the maximum internal rotation (MIR) of the shoulder during the deceleration phase of throwing on flat ground. Methods: The study participants included 17 male baseball players over 20 years old and at the recreational playing level. The recreational level was illustrated by players who did not practice at high intensity and had played only 1-2 competitions per week. Reflective markers were applied to the subject, and throwing motion was assessed using a three-dimensional motion capture system. Data were captured at 1000 Hz. We assessed the angle of the shoulder, spine, and pelvis at the MIR on flat ground. Internal shoulder rotation velocity and spinal and pelvic angular velocities were also assessed. The relationship between the shoulder, spine, and pelvis kinematics at the MIR was examined using simple linear regression analysis. Results: The internal shoulder rotation angle at the MIR was negatively associated with only the spinal flexion and rotation angle at the MIR (P = .006 and P = .010, respectively). No other significant associations between shoulder, spine, and pelvis kinematics were detected at the MIR. Conclusion: For throwing on flat ground, the internal shoulder rotation motion may be suppressed by producing trunk flexion and rotation motion at the MIR.

4. Falciform ligament hernia: combination of key CT findings

Abstract

A male patient in his early 90s with no history of abdominal surgery was referred to us for abdominal pain and vomiting. An abdominal computed tomography (CT) demonstrated dilated small bowel with a double beak sign and poorly enhanced wall, which indicated a closed-loop obstruction that leads to strangulation. A closed-loop bowel was located in front of the anterior and medial segments of the liver and to the right of the round ligament of the liver on axial images. Sagittal images revealed that the round ligament has deviated downward and 2 adjacent narrowed intestines were located at its cranial side. These CT findings suggested the hernia orifice was in the falciform ligament. Emergency surgery for highly suspected bowel ischemia revealed the falciform ligament hernia. A combination of the CT findings played a key role, including the double beak sign, the location of the closed-loop small bowel, and the downward deviation of the round ligament, although preoperative CT diagnosis of falciform ligament hernia is a diagnostic challenge.

Abstract

Background and Aims: Japan has experienced 8 waves of the coronavirus disease 2019 (COVID-19) outbreak over the past 3 years, resulting in an increasing number of deaths and incidence of severe infections. This study aimed to analyze the data from the Japanese inflammatory bowel disease (IBD) patients with COVID-19 registry (J-COSMOS) up to the eighth wave to investigate the clinical course of IBD patients with COVID-19 and factors contributing to disease severity. Methods: In this multicenter, observational, cohort study, we analyzed a cohort of 1308 IBD patients diagnosed with COVID-19, enrolled across 77 participating facilities in the J-COSMOS registry from June 2020 to December 2022. Data on age, sex, IBD (classification, treatment, and activity), and COVID-19 (symptoms, severity, and treatment) were analyzed. Results: The majority of patients (76%) were in clinical remission. According to the World Health Organization classification of COVID-19 severity, 98.4% of IBD patients had nonsevere disease, while 1.6% of patients had severe or critical disease. COVID-19 did not affect disease activity in most IBD patients. Stepwise logistic regression analysis revealed that high body mass index, and cerebrovascular disease were risk factors for severe COVID-19. Corticosteroids could affect COVID-19 severity, whereas anti-tumor necrosis factor α antibodies and thiopurines were associated with a reduced risk of severe COVID-19. No deaths were observed among IBD patients with COVID-19 registered in this cohort. Conclusion: The impact of COVID-19 on IBD disease activity and factors associated with COVID-19 severity were consistent with findings of previous reports. No deaths in Japanese patients with IBD were observed.

Abstract

Here, we propose a two-step pervaporation system with a high-silica CHA (chabazite) membrane, which has sufficient resistance to water and acid, to demonstrate the extraction and condensation of the formic acid formed by electroreduction of CO2. The kinetic diameters of water and formic acid are similar and smaller than the pore size of CHA, while the hydrated electrolyte ions (e.g., K+ and Cl−) are larger than the pore size of CHA. Consequently, the electrolyte ions are separated from the mixture of water and formic acid in the first desalination process, and then water molecules are easily removed from the mixture in the second dehydration process. From 300 ml of an approximately 3 wt% formic acid aqueous solution containing 0.5 M KCl, 10 ml of 18.2 wt% formic acid was obtained.

Abstract

The concept of a “nutrient flap,” in which ischemic tissue is nourished by a transferred well-perfused flap, has been advocated for use since the early days of free flap procedures. Several studies have reported cases of no-option chronic limb-threatening ischemia salvaged by nutrient free flap transfer. However, it has been difficult to prove the actual dynamic flow and nutritional vascular formation. Thus, the existence of a nutrient flap has remained unproved. In the present report, we have described the case of free flap transfer for a patient with no-option chronic limb-threatening ischemia in whom we detected evidence of a nutrient flap using indocyanine green fluorescence imaging.

Abstract

Stent-graft placement is an important treatment for traumatic renal artery dissection, but it may occasionally be technically difficult to advance a catheter through the lesion of the dissection due to severe stenosis of the true rumen. A triple-coaxial (triaxial) system, which consists of a small microcatheter, a large microcatheter, and a 4-Fr. catheter, has recently become available, and it contributes to super-selective catheterization. We thought this system may be useful for passing catheters through the dissection. We herein report a 30-year-old male patient with traumatic renal artery dissection, who was successfully treated by stent-graft placement using the triaxial system.

Objectives: The novel coronavirus (SARS-CoV-2) originating from Wuhan spread rapidly throughout China. While its origin remains uncertain, accumulating evidence links a wet market with the early spread of SARS-CoV-2 in Wuhan. Similarly, the influence of the marketplace on the early transmission dynamics is yet to be investigated. Methods: Using the daily series of COVID-19 incidence, stratified according to contact history with the market, we have conducted quantitative modeling analyses to estimate the reproduction numbers (R) for market-to-human and human-to-human transmission, the reporting probability, and the early effects of public health interventions. Results: We estimated R at 0.24 (95% CrI: 0.01–1.38) for market-to-human transmission and 2.37 (95% CrI: 2.08–2.71) for human-to-human transmission during the early spread in China (2019–2020). Moreover, we estimated that the reporting rate for cases stemming from market-to-human transmission was 2–34 fold higher than that for cases stemming from human-to-human transmission, suggesting that contact history with the wet market played a key role in identifying COVID-19 cases. Conclusions: Our R estimate tied to market-to-human transmission had substantial uncertainty, but it was significantly lower compared with the reproduction number driving human-to-human transmission. Our results also suggest that asymptomatic and subclinical infections constitute a substantial component of the COVID-19 morbidity burden.

Abstract

Neutrophils are central players in the innate immune system. They generate neutrophil extracellular traps (NETs), which protect against invading pathogens but are also associated with the development of autoimmune and/or inflammatory diseases and thrombosis. Here, we report that lactoferrin, one of the components of NETs, translocated from the cytoplasm to the plasma membrane and markedly suppressed NETs release. Furthermore, exogenous lactoferrin shrunk the chromatin fibers found in released NETs, without affecting the generation of oxygen radicals, but this failed after chemical removal of the positive charge of lactoferrin, suggesting that charge-charge interactions between lactoferrin and NETs were required for this function. In a model of immune complex-induced NET formation in vivo, intravenous lactoferrin injection markedly reduced the extent of NET formation. These observations suggest that lactoferrin serves as an intrinsic inhibitor of NETs release into the circulation. Thus, lactoferrin may represent a therapeutic lead for controlling NETs release in autoimmune and/or inflammatory diseases.

Background: Preventing anterior cruciate ligament (ACL) injuries is very important for athletes, and dynamic knee valgus is considered a risk factor for non-contact ACL injury. However, little is known about whether the functions of the hip abductor and rear-foot increase dynamic knee valgus. A two-dimensional (2D) video-based screening test focused on hip abductor and rear-foot functions among factors involved in dynamic knee valgus. The present study determined associations between hip and rear-foot dynamic alignment and dynamic knee valgus. Methods: This cross-sectional study recruited 130 female basketball players (258 legs) from nine high-school teams. The players performed single-leg squats and single-leg drop landings to provide knee-in (KID) and hip-out (HOD) distances on 2D video images. Hip and rear-foot dynamic alignment was evaluated using a dynamic Trendelenburg test (DTT) and a dynamic heel-floor test (HFT). Results: The Chi-square test revealed no significant difference in the prevalence of DTT-positivity between single-leg squats (28.7%) and single-leg drop landings (23.3%). 14. A Mouse Model of Sural Nerve Injury–Induced Neuropathy: Gabapentin Inhibits Pain-Related Behaviors and the Hyperactivity of Wide-Dynamic Range Neurons in the Dorsal Horn

Abstract

This study was conducted to make a new mouse model of neuropathic pain due to injury to a branch of the sciatic nerve. One of three branches (sural, tibial, and common peroneal nerves) of the sciatic nerve was tightly ligated, and mechanical and cool stimuli were applied to the medial part (tibial and common peroneal nerve territories) of the plantar skin. The three types of nerve injuries produced behavioral mechanical hypersensitivities, and the extent of the hypersensitivities after sural and tibial nerve ligation was larger than that of common peroneal nerve ligation. Sural nerve ligation did not affect motor function of the affected hind paw, but tibial and common peroneal nerve ligation produced motor dysfunction. These results suggest that the ligation of the sural nerve is the most suitable for behavioral study. Sural nerve ligation induced behavioral hypersensitivities to mechanical and cool stimuli, which were almost completely inhibited by gabapentin (30 mg/kg). Sural nerve ligation increased spontaneous activity and responses of the wide-dynamic range neurons in the lumbar dorsal horn, which were also almost completely inhibited by gabapentin (30 mg/kg). Sural nerve ligation provides a new mouse model of neuropathic pain, which is easy to prepare and sensitive to gabapentin. Mast cells express high-affinity IgE receptors (FcεRI) on their surface and can be activated to secrete a variety of biologically active mediators by cross-linking of receptor-bound IgE. Recent studies in animal models indicate that mouse mast cells may play a protective role in host defense against bacteria through the production of tumor necrosis factor-α, mainly as a result of Toll-like receptor (TLR) 4- or CD48-mediated activation. Moreover, several recent observations in animal models have indicated that mast cells may also play a pivotal role in coordinating the early phases of autoimmune diseases, particularly those involving auto-antibodies. We recently identified functional TLR4 and FcγRI on human mast cells, in which their expression had been upregulated by interferon-γ. We compared each of the receptor-mediated gene expression profiles with the FcεRI-mediated gene expression profile using high-density oligonucleotide probe arrays and discovered that human mast cells may modulate the immune system in a receptor-specific manner.

A 40-year-old female patient with chronic persistent cough was diagnosed with cough variant asthma (CVA). To investigate the effect of suplatast tosilate (suplatast, 300 mg/day, three times a day), cough scores, medication scores, pulmonary function, capsaicin cough threshold, bronchial hyperresponsiveness (BHR) to methacholine and eosinophilic cationic protein (ECP) concentration in hypertonic saline-induced sputum were evaluated before and after a 6-week treatment with suplatast. Treatment with suplatast resulted in a marked decrease in persistent cough and in the percentage of eosinophils and ECP concentration in induced sputum. It also resulted in a marked improvement in capsaicin cough threshold and a slight improvement in BHR. These results suggest that suplatast tosilate may be useful for treating patients with CVA.

Thus, lactoferrin may represent a therapeutic lead for controlling NETs release in autoimmune and/or inflammatory diseases., Highlights • We developed a real-time cell imaging system which can visualize neutrophil activation, DNA and cell membrane structures. • Lactoferrin serves as an intrinsic inhibitor of neutrophil extracellular traps (NETs) release into the circulation. • Lactoferrin may be a novel therapeutic target in pathological conditions related to NETs. Neutrophils generate neutrophil extracellular traps (NETs), which protect against invading pathogens but are also associated with the development of autoimmune and/or inflammatory diseases and thrombosis. Controlling NETs release is one of the promising therapeutic strategies for the treatment of these diseases. Lactoferrin is a multifunctional protein which is found mainly in human maternal milk, tears, and neutrophils granules and so on. We found that lactoferrin serves an endogenous inhibitor of NETs release into the circulation in inflammatory conditions. Thus, lactoferrin may represent a therapeutic lead for controlling NETs release in autoimmune and/or inflammatory diseases.

Abstract

Background: Several studies have shown that diabetes confers a higher relative risk of vascular mortality among women than among men, but whether this increased relative risk in women exists across age groups and within defined levels of other risk factors is uncertain. We aimed to determine whether differences in established risk factors, such as blood pressure, BMI, smoking, and cholesterol, explain the higher relative risks of vascular mortality among women than among men. Methods: In our meta-analysis, we obtained individual participant-level data from studies included in the Prospective Studies Collaboration and the Asia Pacific Cohort Studies Collaboration that had obtained baseline information on age, sex, diabetes, total cholesterol, blood pressure, tobacco use, height, and weight. Data on causes of death were obtained from medical death certificates. We used Cox regression models to assess the relevance of diabetes (any type) to occlusive vascular mortality (ischaemic heart disease, ischaemic stroke, or other atherosclerotic deaths) by age, sex, and other major vascular risk factors, and to assess whether the associations of blood pressure, total cholesterol, and body-mass index (BMI) to occlusive vascular mortality are modified by diabetes. Findings: Individual participant-level data were analysed from 980793 adults. During 9 center dot 8 million person-years of follow-up, among participants aged between 35 and 89 years, 19686 (25 center dot 6%) of 76965 deaths were attributed to occlusive vascular disease. After controlling for major vascular risk factors, diabetes roughly doubled occlusive vascular mortality risk among men (death rate ratio [RR] 2 center dot 10, 95% CI 1 center dot 97-2 center dot 24) and tripled risk among women (3 center dot 00, 2 center dot 71-3 center dot 33; x(2) test for heterogeneity p

Abstract

In adult female crickets (Gryllus bimaculatus), rhythmic movements of ovipositor valves are produced by contractions of a set of ovipositor muscles that mediate egg-laying behavior. Recordings from implanted wire electrodes in the ovipositor muscles of freely-moving crickets revealed sequential changes in the temporal pattern of motor activity that corresponded to shifts between behavioral steps: penetration of the ovipositor into a substrate, deposition of eggs, and withdrawal of the ovipositor from the substrate. We aimed in this study to illustrate the neuronal organization producing these motor patterns and the pattern-switching mechanism during the behavioral sequence. Firstly, we obtained intracellular recordings in tethered preparations, and identified 12 types of interneurons that were involved in the rhythmic activity of the ovipositor muscles. These interneurons fell into two classes: 'initiator interneurons' in which excitation preceded the rhythmic contractions of ovipositor muscles, and 'oscillator interneurons' in which the rhythmic oscillation and spike bursting occurred in sync with the oviposition motor rhythm. One of the oscillator interneurons exhibited different depolarization patterns in the penetration and deposition motor rhythms. It is likely that some of the oscillator interneurons are involved in producing different oviposition motor patterns. Secondly, we analyzed oviposition motor patterns when the mecahnosensory hairs located on the inside surface of the dorsal ovipositor valves were removed. In deafferented preparations, the sequential change from deposition to withdrawal did not occur. Therefore, the switching from deposition pattern to withdrawal pattern is signaled by the hair sensilla that detect the passage of an egg just before it is expelled.

Abstract

The Cherenkov Telescope Array (CTA) is a new observatory for very high-energy (VHE) gamma rays. CTA has ambitions science goals, for which it is necessary to achieve full-sky coverage, to improve the sensitivity by about an order of magnitude, to span about four decades of energy, from a few tens of GeV to above 100 TeV with enhanced angular and energy resolutions over existing VHE gamma-ray observatories. An international collaboration has formed with more than 1000 members from 27 countries in Europe, Asia, Africa and North and South America. In 2010 the CTA Consortium completed a Design Study and started a three-year Preparatory Phase which leads to production readiness of CTA in 2014. Abstract

Mast cells express high-affinity IgE receptors (FcεRI) on their surface and can be activated to secrete a variety of biologically active mediators by cross-linking of receptor-bound IgE. Recent studies in animal models indicate that mouse mast cells may play a protective role in host defense against bacteria through the production of tumor necrosis factor-α, mainly as a result of Toll-like receptor (TLR) 4- or CD48-mediated activation. Moreover, several recent observations in animal models have indicated that mast cells may also play a pivotal role in coordinating the early phases of autoimmune diseases, particularly those involving auto-antibodies. We recently identified functional TLR4 and FcγRI on human mast cells, in which their expression had been upregulated by interferon-γ. We compared each of the receptor-mediated gene expression profiles with the FcεRI-mediated gene expression profile using high-density oligonucleotide probe arrays and discovered that human mast cells may modulate the immune system in a receptor-specific manner.

Abstract

The growth process of oxidation films on pure Mg, and AZ31B and AZ91D alloys in pure water, Na2SO4 and NaCl solutions has been analyzed by in-situ ellipsometry. The films with a refractive index of 1.42 – 1.47 grew rapidly in two consecytive stages; stage I where the film thickness increases approximately linearly with time, and stage II where the growth rate decreases with time. In every solution, the film growth rate of 6N-Mg was higher than that of AZ91D. The oxidation films were mainly composed of Mg(OH)2. The films formed on 6N-Mg and AZ91D have protective ability, while they suffer local breakdown at high potentials.

Abstract

This paper presents the development of a master arm system with gripping operation devices in a tele-operation platform system that was developed in the national project, “Humanoid and Human Friendly Robotics System.” The tele-operation platform system is connected to a humanoid robot with communication links. An operator can remotely operate the robot by using the platform system with reality of existence at the remote site. The master arm system in the tele-operation platform system can provide the operator with reacting force sensation of the robot arms. The gripping operation devices, which are installed on each master arm, can also provide the operator with gripping sensation. The master arm system will be used to investigate various applications as for humanoid and human friendly robot in the second term of the project. It is expected that the system can serve realistic tele-presence when tele-operating a humanoid and human friendly robot in the near future.

31. Neural mechanism for generating and switching motor patterns of rhythmic movements of ovipositor valves in the cricket

Abstract

In adult female crickets (Gryllus bimaculatus), rhythmic movements of ovipositor valves are produced by contractions of a set of ovipositor muscles that mediate egg-laying behavior. Recordings from implanted wire electrodes in the ovipositor muscles of freely-moving crickets revealed sequential changes in the temporal pattern of motor activity that corresponded to shifts between behavioral steps: penetration of the ovipositor into a substrate, deposition of eggs, and withdrawal of the ovipositor from the substrate. We aimed in this study to illustrate the neuronal organization producing these motor patterns and the pattern-switching mechanism during the behavioral sequence. Firstly, we obtained intracellular recordings in tethered preparations, and identified 12 types of interneurons that were involved in the rhythmic activity of the ovipositor muscles. These interneurons fell into two classes: 'initiator interneurons' in which excitation preceded the rhythmic contractions of ovipositor muscles, and 'oscillator interneurons' in which the rhythmic oscillation and spike bursting occurred in sync with the oviposition motor rhythm. One of the oscillator interneurons exhibited different depolarization patterns in the penetration and deposition motor rhythms. It is likely that some of the oscillator interneurons are involved in producing different oviposition motor patterns. Secondly, we analyzed oviposition motor patterns when the mecahnosensory hairs located on the inside surface of the dorsal ovipositor valves were removed. In deafferented preparations, the sequential change from deposition to withdrawal did not occur. Therefore, the switching from deposition pattern to withdrawal pattern is signaled by the hair sensilla that detect the passage of an egg just before it is expelled.

32. Crowbar Experiments by Ionizing Shock Waves and New Methods to Produce Torus Like Plasmas

Abstract

Various data on basic processes concerning crowbar methods by ionizing shock waves were gathered to apply this technique to the real and larger machines. A pair of crowbar electrodes were flush mounted at the end of a pressure driven shock tube which was terminated with a dielectric plate for shock reflection. Rather restricted shape of crowbar electrodes and use of ambient air in laboratory as a test gas did not decrease so much the insulating voltage of the crowbar gaps. To breakdown the crowbar gaps by reflected shock waves with the incident mach number of 7 ~ 16 in air at the initial pressure of 5 ~ 250 μHg, electrodes voltage must be applied by 700 ~ 400 V and the deduced conductivity of shocked plasmas was 10−11 ~ 10−9 Ω/cm. Two kinds of test circuits were tried to be crowbared. CRT traces of the discharge current with the crowbar gap closed by reflected shock waves showed successful and monotonous envelopes of the oscillating ones without the crowbarring. Various important improvements and another application of this type of experiments are proposed and shown briefly. Among them is a new method to produce torus-like plasmas, which is a different type of application of this crowbarring-idea, itself.

Abstract

Background: The long-axis view and in-plane needle approach (LAX-IP) for ultrasound-guided central vein catheterization is considered ideal because of the quality of real-time imaging. We describe a novel technique, using a step-by-step procedure, to overcome the pitfalls associated with the LAX-IP. This study was undertaken to demonstrate the clinical utility of this approach., Methods: All operators underwent training before participation in this study. One hundred patients were enrolled in this study and underwent central venous catheterization using this method. Using a portable ultrasound and vein catheterization kit, patients were appropriately positioned and a straight portion of the vein identified (Step 1). A needle guide was used (Step 2) and the vein imaged in real time in two directions (Step 3), to identify the true long axis and prevent damage to surrounding tissues., Results: The overall success rate for catheterization was 100% with a median of one puncture for each patient. All catheterizations were performed within three punctures. Problems with the first puncture included difficult insertion of the guide-wire due to coiling, difficult anterior wall puncture, less experience with the procedure, and other reasons. There were no complications associated with the procedure., Conclusions: This three-step method is not dependent on an operator's ability to proceed based on spatial awareness, but rather depends on logic. This method can prevent difficulties associated with a two-dimensional ultrasound view, and may be a safer technique compared with others. Further clinical trials are needed to establish the safety of this technique.

Background: Increased density of 5-HT2A receptors was observed in the platelets of depressive patients with suicidal ideation. Enhanced 5-HT2A receptor-mediated platelet calcium mobilization has been proposed as a biological marker for the pathophysiology of major depression in cancer patients as well as in physically healthy patients. To examine whether depressive cancer patients with suicidal ideation have enhanced 5-HT2A receptor-mediated platelet response compared with those without suicidal ideation, we compared 5-HT-induced platelet calcium mobilization in depressive cancer patients with and without suicidal ideation., Methods: 5-HT-induced platelet calcium mobilization was examined in 24 cancer patients diagnosed as having major depression according to the DSM-IV criteria. Suicidal ideation was evaluated by the Hamilton Depression Rating Scale and Zung's Self Depression Scale, as well as by the DSM-IV criteria., Results: There was no significant differences in 5-HT-induced platelet calcium response between the depressive cancer patients with (n = 8) and without suicidal ideation (n = 16). 5-HT-induced platelet calcium response was also not significantly associated with the severity of suicidal ideation or with the severity of depression assessed by Hamilton Depression Rating Scale and Zung's Self Depression Scale., Conclusions: These findings suggest that enhanced 5-HT2A receptor-mediated response was not associated with suicidal ideation in cancer patients with depression.

Abstract

Electroconvulsive shock (ECS) therapy is considered to be an effective treatment for depression, but its mechanism of action is still unknown. We investigated the effect of chronic ECS in rats treated for 14 days with dexamethasone (Dex), a glucocorticoid receptor agonist. Chronic injection of sesame oil decreased body weight change and increased serotonin (5-HT)-2A receptor number and DOI (5-HT-2A, 2C receptor agonist)-induced wet-dog shake (WDS) behaviors. Dex treatment for 14 days decreased body weight of rats, but repeated ECS did not reverse this decrease. Dex also abolished plasma corticosterone levels, and ECS failed to restore these levels. These results indicate that chronic ECS does not antagonize the effect of Dex. The treatment with Dex increased 5-HT-2A receptor binding density of rat frontal cortex and the number of DOI-induced WDS behaviors. Chronic ECS reduced the enhanced WDS behaviors by Dex but had little effect on receptor density. These results suggest that chronic ECS might suppress 5-HT-2A receptor function at the postreceptor signaling level rather than at the receptor itself, without changing HPA axis function in Dex-treated rats.

Abstract

It is well known that some psychiatric sequelae exist after CO poisoning, but few animal studies on serotonergic neuronal function after CO exposure have been carried out. We investigated the effects of successive carbon monoxide (CO) exposure (6000 ppm, 10 min, 3 repetitions) on serotonergic neuronal systems in rat brain. Serotonin (5-HT) concentrations were significantly decreased only in the frontal cortex from 1 hr to 7 days after CO exposure. 5-Hydroxyindoleacetic acid (5-HIAA) concentrations were significantly increased at 1 hr in all six brain regions measured (frontal cortex, striatum, hypothalamus, hippocampus, midbrain, and pons). 5-HT synthesis, measured by the accumulation of 5-hydroxytryptophan (5-HTP) after the administration of m-hydroxybenzylhydrazine (NSD-1015), was significantly decreased in all regions from 1 hr to 7 days after CO exposure. [3H]Ketanserin (5-HT2A) binding sites in the frontal cortex were not affected by CO exposure. DOI-induced head shakes, a 5-HT2A receptor mediated behavior, were not changed after CO exposure. These findings indicated that CO exposure caused presynaptic serotonergic neuronal dysfunctions that consisted mainly of decreased concentration of 5-HT in the frontal cortex or a decrease of 5-HT synthesis in all six regions, without compensatory hyperfunction of 5-HT2A receptors.

Abstract

In this paper, we show the importance of intracellular calcium (Ca2+) signaling systems in the pathophysiology of mood disorders based on our recent work. Patients with affective disorders appear to have an enhanced intracellular Ca2+ rise in response to serotonin. We have observed effects of antidepressant drugs on intracellular Ca2+ signaling in rat cultured neuronal cells and glioma cells, and found that acute application of several classes of antidepressant drugs inhibited intracellular Ca2+ signaling and Ca2+-related signaling. It is important to investigate the role of intracellular Ca2+ signaling system for an understanding of the pathophysiology of affective disorders.

Abstract

We have investigated the effects of interleukin (IL)-1 beta and lipopolysaccharide (LPS) on endothelin (ET)-induced intracellular Ca2+ rise in C6 rat glioma cells in order to study the mechanisms of their effects on Ca2+ signaling systems. Pretreatment with IL-1 beta (10(3) U/mL) and LPS (1 microgram/mL) for 24 h significantly inhibited 100 nM ET-1-induced increase in intracellular Ca2+ either in the presence or absence of external Ca2+. Their inhibitory effects were in dosedependent (IL-1 beta; 50-1000 U/mL, LPS; 10-1000 ng/mL) and time-dependent (12-24 h) manners. A tyrosine kinase antagonist genistein (50 microM) but not a protein kinase C inhibitor H7 (30 microM) prevented the inhibition of the ET response by IL-1 beta and LPS. These results suggest that activation of tyrosine kinase may be essential for the inhibition of the ET receptor-mediated Ca2+ signaling systems by IL-1 beta and LPS.

Abstract

The effects of chronic treatment with antidepressants, verapamil, or lithium on serotonin (5-HT)-induced Ca2+ increase were investigated in single C6BU-1 glioma cells with digital imaging microscopy. Clomipramine and citalopram, at a concentration of 100 nM, decreased the peak values of 5-HT-induced [Ca2+]i changes. Verapamil (100 nM), a calcium antagonist, and lithium (1 mM) also inhibited the peak amplitudes in the same way. The present findings suggest that chronic treatment with antidepressants, verapamil, or lithium, at therapeutic concentrations, have the common action of inhibiting 5-HT-mediated [Ca2+]i increase.

Abstract

The time-course of 5-HT-induced intracellular Ca2+ mobilization in platelets displays biphasic curves; a rapid peak occurs within 10 sec, followed by a prolonged plateau phase. In platelets of patients with affective disorders, many reports have suggested that there is an increase in the rapid peak in intracellular Ca2+ mobilization, but there is no report concerning the plateau phase in intracellular Ca2+ mobilization. We, then, assessed the time course of 5-HT-induced Ca2+ mobilization to compare untreated manic patients with euthymic bipolar disorders and normal subjects. Not only peak amplitude but also plateau phase were more significantly enhanced in the platelets of untreated manic patients than in those of normal controls. These results suggest that the serotonergic neural transmission by means of intracellular Ca2+ was enhanced by the prolonged plateau phase as well as by increased peak amplitude in platelets of mania. The enhanced rapid peak and plateau phase in untreated bipolar mania were restored to their control levels in treated euthymic bipolar disorders. These findings suggest that the reduction of the enhancement in Ca2+ mobilization might be related to either the effects of chronic treatment with lithium or the affective states of the patients.

Abstract

Positron emission computed tomography (PET) is regarded an excellent technique for quantitative measurements. However, its accuracy is related to the spatial resolution of the system. The relation between myocardial wall thicknesses as measured by X-ray CT or MRI and the radioactivity as measured using PET was studied in 37 patients. 1. In patients with transmural infarction, the infarcted myocardium was imaged as a region of low radioactivity. However, the myocardium usually exhibited wall thinning, so that partial volume effects must be taken into account in evaluating the radioactivity. 2. In the infarcted regions, the regions of the low radioactivity tended to be larger than those of wall thinning. 3. There were cases with the regional low radioactivity without wall thinning in myocardial infarction and in hypertrophic cardiomyopathy. Because patients with myocardial infarction frequently had regional wall thinning, it seems necessary to correct partial volume effects for the infarcted regions which differ from the normal. It was concluded that, to estimate regional myocardial blood flow or metabolism using PET, it is necessary to supplement another morphological diagnostic method to evaluate myocardial wall thickness.
