The electro-polishing behavior of C- steel in ortho-phosphoric acid using the galvanostatic polarization and weight loss methods was ascertained. This was inspected via anode potential–limiting current relationship measurement and comparison in solution of regularly mounting concentrations (from 50 to 1800 ppm of methanolic Marjoram (Origanum majorana), Coriander Seeds (Coriandrum sativum), Chamomile (Matricariarecutita), and Guava leaves (Psidium guajava L)) and influence of temperature on the dissolution kinetic were investigated. Surface morphologies, roughness and reflection of investigated specimens were inspected with a scanning electron microscope (SEM), Profilometry, atomic force microscope and Vis-IR Spectroscopy. Addition of methanolic plant extract to electro-polishing solution results in a lower limiting current. This confirms the mass transport of dissolved species from the anode surface to the bulk of solution as the rate-determining step in the presence of plant extract in all concentrations investigated. Retardation percentage gained from mass loss measurement are comparable with those obtained from measurements of galvanostatic polarization. Results based on microscopy, surface profile measurement and spectroscopy indicate that, addition of methanolic plant extract, a discrete enhancement in the surface texture was observed. Addition of 500 ppm of marjoram (Origanum majorana), coriander (Coriandrum sativa), chamomile (Matricariarecutita) and guava leaves (Psidium guajava L), the degree of surface brightness and reflectance are increased to 64.9, 56.59, 27 and 24.5 respectively relative to electro-polishing electrolyte free solution 16. The roughness (Ra) decreased from 2.7 µm to 0.52 µm without addition of any material. Ra values are 0.28, 0.23, 0.21 and 0.17 μm in the presence of guava leaves (Psidium guajava L), chamomile (Matricariarecutita), coriander seeds (Coriandrum sativum) and marjoram (Origanum majorana). Surface morphology study confirmed that addition of plant extracts to C-steel dissolution bath enhance surface appearance and its texture quality to great extent.

Members of the families Circoviridae and Anelloviridae are unenveloped and have single-stranded circular DNA genomes. Both have capsids with T =1 icosahedral symmetry although they morphologically distinct. Circovirus genomes are ~1700–3800 nt. They encode a single capsid (C) protein and a replication-associated protein that is believed to facilitate rolling circle replication of the genome. Anelloviruses are variable in genome size (2000–4000 nt) and sequence. They code for 3–4 proteins. The capsid is assembled from 60 copies of the capsid protein, VP1. VP2 is a phosphatase and VP3 (also called apoptin) induces apoptosis only in cancer cells. Viruses of both families replicate in the nucleus of mitotically active cells, as they are highly dependent on host cell proteins to replicate their DNA genomes. While a few circoviruses are associated with disease, many viruses in both families have been identified through metagenomic-sequencing studies and in general appear to be highly ubiquitous, nonpathogenic agents.

The name arenavirus comes from the Latin arenosus (sandy). Ribosomes are packaged within arenavirus virions and these are responsible for the “sandy” appearance of the particles. Arenavirus virions are enveloped with helical nucleocapsids and are pleomorphic, ranging in size from 100 to 130 nm in diameter. Structural proteins include a nucleocapsid protein (N), a matrix-like protein (Z), and two glycoproteins derived from a single precursor. Arenaviruses are segmented RNA viruses that replicate using an overall strategy common to negative-strand RNA viruses. However, both genome segments (large (L) and small (S)) are ambisense in their coding strategy. Rodents are natural hosts of many arenaviruses though some can infect humans through contact with infected rodents or inhalation of infectious urine or feces.

Sleep is affected by multiple environmental factors, but specific industrial and biologic toxins produce changes in the sleep–wake functions of humans. Mercury poisoning, trypanosomiasis, and endotoxins are examples that can alter sleep. 1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine produces a parkinsonian syndrome with significant sleep effects. Alcohol also affects sleep, especially altering hypoxic and hypercapnic drive, and increasing upper airway resistance. Lastly, cannabis tends to induce sleep, but a cannabis withdrawal syndrome has been described in which sleep worsens.

Rhus javanica (Anacardiaceae) containing abundant glucopyranosidal constituents, is traditionally used to treat gastric and duodenal ulcer, dysentery, and diarrhea. Rumex hastatus (Polygonaceae) widely distributed in Pakistan, has traditional importance in treating wound healing, jaundice, rheumatism, and skin diseases. Callistemon viminalis (Myrtaceae), a rich source of essential oils, saponins, triterpenoids, phloroglucinols, and flavonoids is used in industries, perfumes, nutrition, and cosmetics. Taking the importance of the subject plants, this study is designed to synthesize silver nanoparticles via aqueous extracts of R. javanica (RJAgNPs), R. hastatus (RHAgNPs), and C. viminalis (CVAgNPs). Synthesis, surface, and sizes of silver nanoparticles (AgNPs) were confirmed using spectroscopic techniques including ultraviolet–visible (UV–Vis), Fourier transform-infrared (FT-IR), and scanning electron microscopy (SEM). AgNPs were produced in ratios 1:15, 1:16, and 1:9 and inferred via appearance of a sharp surface plasmon resonance (SPR) absorption peak (400–435 nm), which represented well-defined, stable, and spherical AgNPs. From SEM analysis, the sizes of RJAgNPs, RHAgNPs, and CVAgNPs were found to be 67 nm, 61 nm, and 55 nm, respectively. The synthesized AgNPs exhibited potential free radical scavenging, antibacterial, and catalytic properties in degradation of dyes including Congo red, methylene blue, methyl orange, rhodamine B, ortho and para-nitrophenols, and several food colours. Hence, the subject AgNPs in the current study might display promising role in drug development and remediation of environmental/industrial effluents.

Tecoma stans (L.) Juss. Ex Kunth is widely used in folk medicine. In ethnomedicine, it is applied as a cardioprotective, hepatoprotective, antiarthritic, antinociceptive, anti-inflammatory, and antimicrobial. The aqueous extract is considered antidiabetic, and is used as a traditional remedy in Mexico. More than 120 chemical constituents have been identified in its leaves, barks, and roots. However, less is known about the phytochemical properties of T. stans flower extracts. The herbal plant Nervilia concolor (Blume) Schltr. is native to Vietnam, and is used in traditional Chinese medicine to treat diseases such as bronchitis, stomatitis, acute pneumonia, and laryngitis. Only two previous reports have addressed the chemical content of this plant. Bouea macrophylla Griff., commonly known as marian plum or plum mango, is a tropical plant that is used to treat a range of illnesses. Phytochemical analysis of B. macrophylla suggests the presence of volatile components and flavonoids. However, existing data have been obtained from screening without isolation. As part of our ongoing search for alpha-glucosidase inhibitors from Vietnamese medicinal plants, we conducted bioactive-guided isolation of the whole plant N. concolor, the flowers of T. stans, and the leaves of B. macrophylla. We isolated and structurally elucidated five known compounds from T. stans: ursolic acid (TS1), 3-oxours-12-en-28-oic acid (TS2), chrysoeriol (TS3), ferulic acid (TS4), and tecomine (TS5). Three known compounds were isolated from Nervilia concolor: astragalin (NC1), isoquercitrin (NC2), and caffeic acid (NC3). From B. macrophylla, betullinic acid (BM1), methyl gallate (BM2), and 3-O-galloyl gallic acid methyl ester (BM3) were isolated. All compounds showed promising alpha-glucosidase inhibition, with IC50 values ranging from 1.4 to 143.3 µM. The kinetics of enzyme inhibition showed BM3 to be a competitive-type inhibitor. An in silico molecular docking model confirmed that compounds NC1, NC2, and BM3 were potential inhibitors of the α-glucosidase enzyme. Molecular dynamics simulations were carried out with compound BM3 demonstrating the best docking model during simulation up to 100 ns to explore the stability of the complex ligand–protein.

In this study, three concentrations (250, 500, and 1000 ppm) of abamectin 2% suspension concentration (SC) were used as cucumber seeds treatment. The seeds were treated with abamectin to reduce nematodes reproduction and their ability to penetrate the roots, then seed germination and plant growth were observed. All the concentrations didn’t negatively affect seeds germination wherever the germination percent reached 80% at the concentration (1000 ppm) after 20 days of sowing. The effect of abamectin on root-knot nematode was studied by recording numbers of nematodes in 100 g/soil, numbers of the galls, egg mass on the root, and the nematode reproduction factor. All concentrations significantly affected the nematode reproduction parameters compared to control. Abamectin at (500 ppm) was the most effective concentration on reducing nematodes parameters, i.e., 26.57, 38.83, 47.40 %, and 3.15 for the above-mentioned parameters, respectively at the end of experimint. No significant difference between 500 ppm and 1000 ppm. We recommended using the abamectin in (500 ppm) concentration as a seed application to control Meloidogyne incognita in cucumber plants under greenhouse conditions to reduce its environmental toxic effect.

Crustacean waste is one of the most severe issues, posing significant environmental and health risks. This study aims to improve managing marine waste by isolating chitosan from Procambarus clarkii by devising a new methodology, incorporating technical steps, e.g., washing, decolorization and deacetylation under a reflexive condenser and dialysis purification. A comparison was made between the prepared P. clarkii chitosan and four types of shrimp chitosans: commercial, high, low, and nano. The obtained chitosan has a low molecular weight and viscosity compared to the commercial shrimp chitosan used in various applications. P. clarkii chitosan was prepared in high quality from a cheap source, as its color and quality were better than those of the commercial shrimp chitosan. The new methodology has successfully extracted chitosan from P. clarkii in a good quality and high purity, achieving 89% deacetylation, high solubility, high purity, and medium molecular weight. Analysis of the different chitosan samples with Fourier transform infrared spectroscopy (FTIR), atomic force microscopy, Raman spectrum referred indicated high similarity between the chitosan different types, regardless of its source. The 3D image of P. clarkii showed the distance between the highest and most profound points of extracted chitosan is 728.94 nm, revealing homogeneous, smooth surfaces, apparently free of pores and cracks. FTIR and Raman spectrum of P. clarkii indicated various functional groups, e.g., alcohol, amines, amides, and phenols. These active groups are responsible for about 60% of the antioxidant activity of that product. Evaluating the quality traits indicated the excellence of the chitosan prepared from P. clarkii, especially in color, viscosity, and antioxidant activity, nominating it for different food applications.

Microalgae represent promising sources of bioactive compounds for pharmaceutical and industrial applications. The emergence of antibiotic resistant bacteria leads to the need to explore new cost-effective, safe, and potent bioactive compounds from the microalgae. This study aimed to investigate the potential of local microalgae for their antimicrobial properties and bioactive compounds. Three local microalgae namely Chlorella sorokiniana (UKM2), Chlorella sp. UKM8, and Scenedesmus sp. UKM9 biomass methanol extracts (ME) were prepared and tested against Gram-positive and Gram-negative bacteria. Chlorella sp. UKM8-ME showed the highest antibacterial activity. UKM8-ME minimum inhibitory concentrations were in the range of 0.312 to 6.25 mg/mL. Cytotoxicity evaluation using MTT assay showed that the microalgae methanolic extracts did not exhibit cytotoxicity against Vero-cells. The UKM8-ME was mainly containing 28 compounds from the Gas Chromatography-Mass Spectrometry (GC–MS) analysis. Major compounds of UKM8-ME included phenol (18.5%), hexadecanoic acid (18.25%), phytol (14.43%), 9,12-octadecadienoic acid (13.69%), and bicyclo[3.1.1]heptane (7.23%), which have been previously described to possess antimicrobial activity. Hence, Chlorella sp. (UKM8) methanol extracts showed promising antibacterial activity. More comprehensive studies are required to purify these antimicrobial compounds and develop our understanding on their mechanism in UKM8-ME to unleash their specific potential.

Xanthones C-glycosides are plants secondary metabolites with diverse biological activities. Among the C-glycoside xanthones, the mangiferin (MF) is of widespread occurrence in plants while isomangiferin (IsoMF) is not very common. For the present study mangiferin (MF) and isomangiferin (IsoMF) were isolated from Dryopteris ramosa. The antibacterial potential of MF and IsoMF was evaluated by using agar well diffusion method while cytotoxic properties of MF and IsoMF were assessed by brine shrimp lethality test (BSLT). The antibacterial potential of MF and IsoMF increases in dose dependent manner. The minimum inhibitory concentration (MIC) indicated strong antibacterial potential of MF against Salmonella setubal (125 µg/mL) and Bacillus subtilis (125 µg/mL) while MF showed weak antibacterial potential against Escherichia coli (500 µg/mL). On the other hand the IsoMF showed better antibacterial potential against all the tested strain including Escherichia coli (MIC = 250 µg/mL). The MF and IsoMF showed poor cytotoxicity towards Brine shrimp nauplii as indicated by their LD50 (969.77 ± 0.67 and 768.92 ± 0.81 µg/mL respectively). The present study has highlighted the antibacterial potential of MF and IsoMF. Further evaluation of these two isomeric compounds may prove to be the future remedies for various bacterial infections and other human ailments.

Background The emergence of resistance to commonly used antibiotics by human infections occurred mostly due to their overuse, that prompted individuals to pursue novel and innovative treatments. The phytochemical characteristics, antibacterial activity, and cytotoxicity of MCF7 cells were evaluated in two Pleurotus spp. mycelial extracts in this work. Methods Pleurotus djamor and Pleurotus florida mycelial extracts from pure cultures were tested for antibacterial activity by a well-diffusion assay and antimicrobial activity against mold fungi was evaluated for biomass inhibition. Mycelial extracts were obtained from dichloromethane extracts and their biophysical characteristics are analyzed by UV–vis spectrum and FTIR analysis. By spraying detection reagents onto TLC plates, the chemicals in dichloromethane extraction of chosen mushroom fungus mycelia were identified. Using the MTT test, the cytotoxic effect of dichloromethane extracts of selected mushroom fungi was evaluated on MCF7 Cell lines. Results Mycelial extracts of P. djamor and P. florida exhibited significant antimicrobial effect on the bacterial and fungal pathogens tested. Dichloromethane mycelial extracts were obtained using soxhlet extraction which response positive for various phytochemical analysis. Detection of metabolites in thin layer chromatography using spray reagents documented one of few first accounts on flavonoids, anthroquinone and terpenoid compounds in P. djamor and P. florida. P. djamor and P. florida had dose-dependent antiproliferative activity against MCF7 cells, with an inhibitory impact of 55.72% and 64.47% percent at 125 μg/mL, respectively. Conclusion The study has reported the identification with the potent biological activity of some of the key bioactive components present in DCM extracts from the mycelia of P. djamor and P. florida.

The acute toxicity of household detergent (Ariel) on blood parameters and histology of Oreochromis niloticus was investigated using static bioassay for 96 h. Linear alkylbenzene sulfonate (LAS) is an anionic surfactant widely used in detergents and cleaners, both in industrial and household applications. LAS contaminating aquatic ecosystems as a potential toxic pollutant, was investigated in the present study for acute toxicity. The fish samples were divided into six groups, including 20 fish in each group. Normal feed was given to control group without detergents treatment. Hematological parameters (RBC count, Hb, Ht and platelets) were significantly declined, while WBC count showed a highly significant increase. Compared with the control group, significant elevation of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) was recorded in fish treated with different concentrations of detergent. Catalase (CAT), Superoxide dismutase (SOD) activities and Reduced Glutathione (GSH) concentration showed a highly significant reduction. Total proteins showed significant decrease, while total lipids, cholesterol and triglycerides significantly increased. The mean lethal concentration (LC50) for 96 h of Ariel was at concentration 10 mg/L. Relative percentage of detergent residues in fish muscles was increased with higher detergent concentrations. In conclusion, exposure to detergents resulted in great alterations in the histological structure of liver and gills.

The current works report the bio-efficacy of Pimenta dioica leaf derived silver nanoparticles (Pd@AgNPs) and leaf extract obtained trough different solvents against the larvae of malaria, filarial and dengue vectors. Synthesis of silver nanoparticles (AgNPs) was done by adding 10 ml of P. dioica leaf extract into 90 ml of 1 mM silver nitrate solution, a slow colour change was observed depicting the formation of AgNPs. Further, Pd@AgNPs was confirmed through Ultraviolet–visible spectroscopy which exhibited characteristic absorption peak at 422 nm wavelength. X-ray diffraction and selected area electron diffraction analysis confirmed monodispersed and crystalline nature of Pd@AgNPs with 32 nm an average size. Scanning electron microscopy and transmission electron microscopy showed the most of Pd@AgNPs were spherical and triangular in shape and energy-dispersive X-ray spectroscopy revealed silver elemental nature of nanoparticles. Zeta potential of Pd@AgNPs is highly negative which confirmed its stable nature. Pd@AgNPs showed prominent absorption peaks at 1015, 1047, 1243, 1634, 2347, 2373, 2697 and 3840 cm−1 which are corresponding to following compounds polysaccharides, carboxylic acids, water, alcohols, esters, ethers, amines, amides and phenol, respectively as reported by Fourier-transform infrared spectroscopy analysis. Gas chromatography–mass spectrometry and Liquid chromatography–mass spectrometry analysis revealed 39 and 70 compounds, respectively, which might be contributed for bio-reduction, capping, stabilization and larvicidal behavior of AgNPs. A comparable lethality (LC50 and LC90) was observed in case of Pd@AgNPs over leaf extract alone. The potential larvicidal activity of Pd@AgNPs was observed against the larvae of Aedes aegypti,(LC50, 2.605; LC90, 5.084 ppm) Anopheles stephensi (LC50, 3.269; LC90, 7.790 ppm) and Culex quinquefasciatus (LC50, 5.373; LC90, 14.738 ppm without affecting non-targeted organism, Mesocyclops thermocyclopoides after 72 hr of exposure. This study entails green chemistry behind synthesis of AgNPs which offers effective technique for mosquito control and other therapeutic applications.

Background: The use of single cell protein (SCP) has become a method for alleviating the shortage of protein feed that microorganisms propagate in a suitable culture medium. In this study, SCP was produced by yeast to use the nutrition contained in the biogas slurry of chicken manure. Results: The results showed that Galactomyces candidum was the most efficient at producing SCP among the seven yeasts studied. The maximum cell dry weight (CDW) 6.79 g/L and protein content 39.39%, were obtained under the fermentation conditions of initial NH4+-N concentration of 2000 mg/L and a C/N ratio of 6:1 with acetate as the pH regulator. The total CDW increased to 9.24 g/L after secondary fermentation. Metal elements had a little effect on the growth of G. candidum. The addition of sulfur not only promoted the synthesis of sulfur-containing amino acid cysteine but also increased protein content by promoting the synthesis of glutamic acid and glutamine. Conclusions: Future experiments should focus more on achieving high-density cultivation and more efficient utilization of ammonia nitrogen in the biogas slurry.How to cite: Zhou Pan, Zhang Lu, Ding Hongxia, et al. Alzheimer’s disease (AD) is characterized by progressive cognitive decline, and the number of affected individuals has increased worldwide. However, there are no effective treatments for AD. Therefore, it is important to prevent the onset of dementia. Oxidative stress and endoplasmic reticulum (ER) stress are increased in the brains of AD patients, and are postulated to induce neuronal cell death and cognitive dysfunction. In this study, Centella asiatica, a traditional Indian medicinal herb, were fractionated and compared for their protective effects against glutamate and tunicamycin damage. Araliadiol was identified as a component from the fraction with the highest activity. Further, murine hippocampal cells (HT22) were damaged by glutamate, an oxidative stress inducer. Centella asiatica and araliadiol suppressed cell death and reactive oxygen species production. HT22 cells were also injured by tunicamycin, an ER stress inducer. Centella asiatica and araliadiol prevented cell death by mainly inhibiting PERK phosphorylation; additionally, Centella asiatica also suppressed the expression levels of GRP94 and BiP. In Y-maze test, oral administration of araliadiol (10 mg/kg/day) for 7 days ameliorated the arm alternation ratio in mice with scopolamine-induced cognitive impairment. These results suggest that Centella asiatica and its active component, araliadiol, have neuroprotective effects, which may prevent cognitive dysfunction.

Background/purpose: The use of fluoride is known to reduce the risk of dental caries. There is limited information on the relationship between Streptococcus mutans (S. mutans) and fluoride exposure. This study investigated the association between the count of S. mutans on supragingival biofilm and fluoride exposure of scholar children. Materials and methods: In this cross-sectional study, 56 children from 9 to 11 years of age were selected. Fluoride concentration in drinking water, urine and saliva of each participant were assessed. The count of S. mutans was estimated by calculating the DNA copy number through a quantitative real time polymerase chain reaction (qPCR) assay. Also, sociodemographic data, oral and general health information and variables related to caries risk were evaluated. A stepwise multiple linear regression was performed in all caries related predictor variables with the count of S. mutans as the dependent variable. Infections caused by virus are one of the foremost causes of morbidity and mortality in the world. Although a number of antiviral drugs are currently used for treatment of various kinds of viral infection diseases, there is still no available therapeutic agent for the most of virus in clinical practice. Coumarin is a chemical compound which is found naturally in a variety of plants, as well as it can be synthetically produced possessing diverse biological effects. More recently, reports have highlighted the potential role of coumarin derivatives as antiviral agents. This review outlines the advances in coumarin-based compounds against various viruses including human immunodeficiency virus, hepatitis virus, herpes simplex virus, Chikungunya virus and Enterovirus 71, the structure activity relationship and the possible mechanism of action of the most potent coumarin derivatives are also discussed.

The rise of antibiotic resistance has increased the need for alternative ways of preventing and treating enteropathogenic bacterial infection. Various probiotic bacteria have been used in animal and human. However, Saccharomyces boulardii is the only yeast currently used in humans as probiotic. There is scarce research conducted on yeast species commonly found in kefir despite its claimed potential preventative and curative effects. This work focused on adhesion properties, and antibacterial metabolites produced by Kluyveromyces lactis and Saccharomyces unisporus isolated from traditional kefir grains compared to Saccharomyces boulardii strains. Adhesion and sedimentation assay, slide agglutination, microscopy and turbidimetry assay were used to analyze adhesion of Salmonella Arizonae and Salmonella Typhimurium onto yeast cells. Salmonella growth inhibition due to the antimicrobial metabolites produced by yeasts in killer toxin medium was analyzed by slab on the lawn, turbidimetry, tube dilution and solid agar plating assays. Alcohol and antimicrobial proteins production by yeasts in killer toxin medium were analyzed using gas chromatography and shotgun proteomics, respectively. Salmonella adhered onto viable and non-viable yeast isolates cell wall. Adhesion was visualized using scanning electron microscope. Yeasts-fermented killer toxin medium showed Salmonella growth inhibition. The highest alcohol concentration detected was 1.55%, and proteins with known antimicrobial properties including cathelicidin, xanthine dehydrogenase, mucin-1, lactadherin, lactoperoxidase, serum amyloid A protein and lactotransferrin were detected in yeasts fermented killer medium. These proteins are suggested to be responsible for the observed growth inhibition effect of yeasts-fermented killer toxin medium. Kluyveromyces lactis and Saccharomyces unisporus have anti-salmonella effect comparable to Saccharomyces boulardii strains, and therefore have potential to control Salmonella infection.

The effects of adsorbent dosage, reaction time, temperature and pH on the adsorption of Pb2+, Zn2+, Fe2+ and Cu2+ from produced water using activated carbon prepared from Citrullus lanatus peel via calcination at 681.10 °C for 2.61 h were studied. L9 orthogonal Taguchi approach was used for the parametric optimization. Activated carbon was characterized using scanning electron microscopy, Brunauer-Emmett-Teller analysis, X-ray diffraction, X-ray fluorescence and Fourier transform infrared spectroscopy and; exhibited a porous-graphite-crystalline structure, presence of different metal oxides and active functional groups with large surface area of 1249.7 m2/g. Batch adsorption experiments revealed the efficacy of prepared activated carbon to remove heavy metals from produced water. At optimum points, predicted values were 76.31, 59.3, 69.91 and 84.08% for Pb2+, Zn2+, Fe2+ and Cu2+ removed respectively. The values of R2, Adj-R2 and pred-R2 revealed reasonable agreements between experimental and predicted data which proved developed models to be effective.

In this study, we evaluated the potency ofR. crispusextracts on the model organism,Caenorhabditis elegansand the bioactive compounds of the extracts were also identified. The solvent extracts ofR. crispuswere tested againstC. elegansfor up to 72h. The effect of the extracts onC. eleganswas examined using light microscopy (LM) and scanning electron microscopy (SEM). LM and SEM analysis showed damage on the body wall, reduced body and slight modifications of the nematode organs. Thelethalitytest reveals a significant reduction in the viability of the nematode with the water extract of leaf (LF-WAE), among others, having the strongest potency against the nematode, with 83% lethality. Anlysis done with Fourier-transform infrared spectroscopy (FTIR) spectra reveals various characteristic vibration bands and fingerprint bands at 3400–600cm−1, identifying phenols, organic acids, aromatics, amines, among others in the plant. The compounds were identified with liquid chromatography-mass spectrometry (LC-MS), under the categories of flavonoids, steroidalalkaloidsand proanthocyanidin. In conclusion, this study confirmed thatR. crispushas anthelmintic potential, using standardisedC. elegansmodels as a tool and suggests that there could be novel compounds yet to be explored in the studied plant that could be of great benefit to livestock and humans.

To increase the agricultural production, nano fertilizers have allowed the development of new technologies. In this study, zinc oxide nanoparticles (ZnO-NPs), iron oxide nanoparticles (FeO-NPs) and magnesium oxide nanoparticles (MgO-NPs) have been prepared and applied on the medicinal plants (Caesalpinia bonducella) to increase the yield, nutrient and chlorophyll contents. This study intended to compare the impacts of synthesised NPs with the control plants and their conventional fertilizers such as ZnSO4·7H2O (Zn-bulk), FeSO4·7H2O (Fe-bulk) and MgSO4·7H2O (Mg-bulk). All fertilizers were applied in a completely randomized design including six treatments and three replications of each treatment by using a combination of seed priming method and soil mixing process. Moreover, the NPs were also compared for the best gaining of results. The concentrations selected for applications were 10, 20, 30, 40 ppm for iron and magnesium fertilizers and 25, 50, 75, 100 ppm for zinc-based fertilizers. The results indicated the increase in morphological parameters of the plants of 28–50% by the application of conventional fertilizers while 50–93% by NPs application as compared to control plants. Furthermore, the increase in chlorophyll contents was 5–28% with conventional fertilizers while 30–80% in case of NPs application than control plants. The obtained results indicate that the best increase in growth, nutrient and chlorophyll contents of plants were achieved at the dose concentrations of 40 ppm for Mg and Fe fertilizers and 100 ppm for Zn fertilizers. The comparative study of NPs showed that the highest growth and yield was obtained by the use of ZnO-NPs in comparison to others. Finally, it is worth highlighting that NPs have a huge potential to increase the productivity of crops if applied in optimal concentrations.

Background: Antimicrobial substances from medicinal plants have been widely applied in many industries, without the concern of side effects. Nonwoven fabric products such as face masks, paper wipes, and wound dressings contain antibacterial substances for reduction of bacterial accumulation. Therefore, the use of bioactive compounds from medicinal plants improves the properties of fabrics. This study used a crude extract from Caesalpinia sappan to perform phytochemical screening and investigate its antibacterial activity, stability, hemolytic activity, and preliminary application in nonwoven fabric products. Results: The alcoholic crude extracts of C. sappan heartwood consisted of alkaloids, anthraquinones, coumarins, flavonoids, saponins, tannins, terpenoids, and cardiac glycosides. The C. sappan extract (CSE) had the highest inhibitory activity against Staphylococcus aureus with inhibition zone of 13.67 ± 1.56 mm in the agar well diffusion assay. The MIC value of CSE was at 1.95, 1.95, and 0.98 mg/ml, while the MBC value was 62.5, 3.91, and 31.25 mg/ml for S. aureus, Bacillus cereus, and Vibrio parahaemolyticus, respectively. The phenolic content was stable at 28 ± 2°C and at pH 4.0 for 12 h (∼95%) and 10 d (>90%), and it had low hemolytic activity on human erythrocytes at 5.04–18.95%. The major chemical component was flavonoids, and brazilein was 1.88% area sum. For the application to antibacterial fabrics, the highest inhibitory activity against S. aureus was found in 2MIC of CSE-coated fabric with 99% for 3 h. Conclusions: The CSE had high inhibitory toward 3 bacterial strains and stability in a wide range of pH and temperature. In order to investigate temporal variations in phytoplankton connected with hydrography, high-resolution (twice a week) samplings were carried out between January and December 2018 at a single, shallow water station in the Dardanelles. Diatoms dominated in early and mid-winter, late spring and early summer, and between August and October. Whereas dinoflagellates dominated in the period from mid-February to the end of the third week of May, mid-summer, and late autumn. Other groups were generally more abundant in the early summer and mid-autumn than any other period. Late spring and early summer, mid-autumn and middle winter were the important bloom periods. The average contribution of diatom abundance (5.00 × 105±7.80 × 105 cells L−1) to average total phytoplankton abundance (9.63 × 105±7.88 × 105 cells L−1) was above 50% (average: 51.88%) during the year except spring (34.32%). However, the average contribution of dinoflagellates (43.32±20.69%) and others (4.81±6.99%) to the total phytoplankton abundance were lower than the abundance of the diatoms (51.88±21.61%). The study revealed a decrease both in the total number of algal blooms which also included HABs and their densities in the region compared to the previous findings. These observations seem to evidence environmental status improvement of the investigated area, manifested in an average chlorophyll a concentration (0.71±0.52 µg L−1) and quantitative phytoplankton structure (103 to 105 cells L−1).

Due to drug addiction and the emergence of antibiotic resistance in pathogens, the disease load and medication intake have risen worldwide. The alternative treatment for drug-resistant infections is Nano formulation-based antimicrobial agents. The plant extract of Conocarpus Lancifolius fruits was used to synthesize silver nanoparticles in the current study, and it was further employed as an antimicrobial and anticancer agent. Nanoparticles have been characterized by UV–visible spectrometer revealed the notable peak of λmax = 410–442 nm, which confirms the reduction of silver ion to elemental silver nanoparticles, and the biological moieties in the synthesis were further confirmed by FTIR analysis. The stability and crystalline nature of materials were approved by XRD analysis and expected the size of the nanomaterials of 21 to 173 nm analyzed by a nanophox particle-size analyzer. In vitro, synthesized materials act as an antibacterial agent against Streptococcus pneumonia and Staphylococcus aureus. The inhibition zones of 18 and 24 mm have been estimated to be antibacterial activity against both bacteria. The potency of up to 100% of AgNPs for bacterial strains was incubated overnight at 60 μg/ml. Based on our results, biogenic AgNPs reveal significant activity against fungal pathogen Rhizopusus stolonifera and Aspergillus flavus that cause leading infectious diseases. Additionally, nanomaterials were biocompatible and demonstrated the potential anticancer activities against MDA MB-231 cells after 24-hour exposure.

Fusarium crown rot (FCR), typically caused by Fusarium pseudograminearum, is a severe soil-borne disease that, in recent years, has become an emerging threat to Chinese wheat crops. For the first time in this study, we investigated and compared the proteomic characteristics of two Chinese wheat varieties (04 Zhong 36 and Xinmai 26) at 24, 48, and 72 h post-inoculation using label-free quantitative proteomic analysis. A total of 9 234 proteins were successfully quantified, of which 783 were differentially expressed after inoculation. These proteins were mainly involved in metabolic, single-organism, and cellular processes. Thirty-three proteins associated with defense, cell wall formation, photosynthesis, etc., showed consistently different expression between the two genotypes at multiple time points. In particular, chitinase, which degrades chitin in the fungal cell wall and limits fungal growth, was exclusively and consistently upregulated in 04 Zhong 36 across the three time points. Other proteins such as flavonoid O-methyltransferase, glycosyltransferase, and peroxidase were only upregulated in 04 Zhong 36, and proteins, including the berberine bridge enzyme and rubisco large subunit-binding protein, were specifically downregulated in Xinmai 26. The expression of transcripts encoding eight selected proteins through qRT-PCR analysis supported the proteomic profiles. Overall, the results of this study allow us to understand FCR resistance in wheat at the protein level. Some proteins and their corresponding genes may be useful resources for the genetic improvement of FCR resistance in wheat.

Banana peel-derived porous carbon from natural biowaste had been prepared with different annealing temperatures at 300, 400, 500, 600, and 700 °C. The as-prepared carbon powders were carefully characterized with XRD, SEM, TEM, EIS, and XPS measurements and their antibacterial activities were tested toward Escherichia coli (E. coli - ATCC 25922). It was found that 600 °C as-prepared porous carbon exhibited the most significant inhibitor against E. coli colonies’ growth. It was found that the natural potassium chloride in the banana peel-derived porous carbon played an essential role in preventing the bacterial growth. Furthermore, the as-prepared nanosized porous carbon also directly contacted the bacterial cell wall, which seriously affected the cellular activities, including the metabolic processes and transfer of substances in the cell interfaces. Therefore, the synergistic effect between natural potassium chloride and porous carbon gave rise to antimicrobial activity. The generated electron and hole from low thermal-induced excitation in porous carbon and released electron from potassium significantly deactivated the bacteria activity. The bactericides of banana peel-derived porous carbon were investigated at 37 °C without light illumination and the mechanism of antibacterial growth was elucidated and proposed in this work.

In recent years, many research on the quantity of lignocellulosic waste have been developed. The production, partial purification, and characterisation of ligninolytic enzymes from various fungi are described in this work. On the 21st day of incubation in Potato Dextrose (PD) broth, Hypsizygus ulmarius developed the most laccase (14.83 × 10−6 IU/ml) and manganese peroxidase (24.11 × 10−6 IU/ml), while Pleurotus florida produced the most lignin peroxidase (19.56 × −6 IU/ml). Laccase (Lac), lignin peroxidase (LiP), and manganese peroxidase (MnP), all generated by selected basidiomycetes mushroom fungi, were largely isolated using ammonium sulphate precipitation followed by dialysis. Laccase, lignin peroxidase, and manganese peroxidase purification findings indicated 1.83, 2.13, and 1.77 fold purity enhancements, respectively. Specific activity of purified laccase enzyme preparations ranged from 305.80 to 376.85 IU/mg, purified lignin peroxidase from 258.51 to 336.95 IU/mg, and purified manganese peroxidase from 253.45 to 529.34 IU/mg. H. ulmarius laccase (376.85 IU/mg) with 1.83 fold purification had the highest specific activity of all the ligninolytic enzymes studied, followed by 2.13 fold purification in lignin peroxidase (350.57 IU/mg) and manganese peroxidase (529.34 IU/mg) with 1.77-fold purification. Three notable bands with molecular weights ranging from 43 to 68 kDa and a single prominent band with a molecular weight of 97.4 kDa were identified on a Native PAGE gel from mycelial proteins of selected mushroom fungus. The SDS PAGE profiles of the mycelial proteins from the selected mushroom fungus were similar to the native PAGE. All three partially purified ligninolytic isozymes display three bands in native gel electrophoresis, with only one prominent band in enzyme activity staining. The 43 kDa, 55 kDa, and 68 kDa protein bands correspond to laccase, lignin peroxidase, and manganese peroxidase, respectively.

A controlled fermentation was conducted to develop and design a bioorganic fertilizer based on three wastes generated by the food industry: olive mill wastewater (OMW), molasses and rice hulls by inoculation with selected strains of lactic acid bacteria (LAB), of the species “Lactiplantibacillus plantarum” and “Levilactobacillus brevis” and of the yeast of the species “candida famata”. The fermentation with commercial yeasts and particularly the species of “saccharomyces” produced an unstable fertilizer with a high pH value (6.14).Whereas, the fermentation test with an inoculum composed of 20% yeast and 80% lactic acid bacteria showed a decrease in pH at 4.66 and a maximum enzymatic activity of 43.900 IU/l after 30 h. This combination allowed an appropriate morphological and microbiological quality of the bioorganic fertilizer. To determine with precision, the optimum conditions of this fermentation an optimization design was applied. As a result of experiments and statistical validation of the fitted model, the bioorganic fertilizer with the lowest pH value was achieved after fermentation of 53.2% OMW, 19.1% molasses, 21.3% rice hulls and 6.4% inoculum content. The final pH was able to reach a value of 4.07 with desirability of the order of 99.7%. A fertilizer with the lowest possible pH allowed farmers longer storage periods and required no special storage conditions.

Biological synthesis of silver nanoparticles (AgNPs) by Cheatomorpha antennia and its in vitro and in vivo antibacterial activity against Vibrio harveyi in Macrobrachium rosenbergii was demonstrated in the study. In vitro growth curve analysis, cell viability and bacterial inhibitory assays were performed to test the efficacy of synthesised AgNPs against bacteria. Sodium caseinate was used as an encapsulating agent to deliver the antibacterial drugs and the commercial process of microencapsulation comprises the antibacterial bioelements for oral administration to improve the disease resistance of AgNPs against V. harveyi due to the eco-friendly for non-toxic behaviour of nanoparticle and their treatment. Characterisation of antibacterial silver was performed by UV spectroscopy, X-ray diffraction, Fourier Transform Infrared spectroscopy and Scanning Electron Microscopy. The peak at 420 nm showed the presence of nanoparticles in the solution and the crystal nature of the particle was identified by the XRD. FTIR characterised the functional harveyi biomolecules and further SEM confirmed the size of the nanoparticles around 24 ± 2.4 nm. Experimental pathogenicity of V. harveyi showed 100% mortality at the 120th hour. Treatment of encapsulated AgNPs was administered orally for the relative percentage of survival which acquired almost 90% of survival till 30 days of exposure. In conclusion, the microencapsulation of AgNPs in the biopolymer matrices promotes the health, growth responses, immunity and disease resistance of encapsulated AgNPs with an improved relative percentage of survival.

Human malaria continues to be a public health problem and an important cause of morbidity and mortality in the world. Malaria control is achieved through both individual protection against mosquito bites and drug treatment, which is hampered by the spread of Plasmodium falciparum resistance to most antimalarials, including artemisinin derivatives. One of the key pharmacological strategies for controlling malaria is to block transmission of the parasites to their mosquito vectors. Following this rational, MEFAS, a synthetic hybrid salt derived from artesunate (AS) and mefloquine has been previously reported for its activity against asexual P. falciparum parasites in vitro, in addition to a pronounced reduction in the viability of mature gametocytes. Herein, MEFAS was tested against asexual forms of Plasmodium vivax and for its ability to block malaria transmission in Anopheles darlingi mosquitoes in a membrane feeding assay using P. vivax field isolates. MEFAS demonstrated high potency, with a IC50 of 6.5 nM against asexual forms of P. vivax. At 50 μM, MEFAS completely blocked oocyst formation in mosquitoes, regardless of the oocyst number in the control group. At lower doses, MEFAS reduced oocyst prevalence by greater than 20%. At equivalent doses, AS irregularly reduced oocyst formation and caused only slight inhibition of mosquito infections. These results highlight the potential of MEFAS as a novel transmission-blocking molecule, as well as its high blood schizonticidal activity against P. vivax and P. falciparum field isolates, representing a starting point for further development of a new drug with dual antimalarial activity., Graphical abstract Image 1

Suppression of cellular O-linked β-N-acetylglucosaminylation (O-GlcNAcylation) can repress proliferation and migration of various cancer cells, which opens a new avenue for cancer therapy. Based on the regulation of insulin gene transcription, we designed a cell-based fluorescent reporter capable of sensing cellular O-GlcNAcylation in HEK293T cells. The fluorescent reporter mainly consists of a reporter (green fluorescent protein (GFP)), an internal reference (red fluorescent protein), and an operator (neuronal differentiation 1), which serves as a “sweet switch” to control GFP expression in response to cellular O-GlcNAcylation changes. The fluorescent reporter can efficiently sense reduced levels of cellular O-GlcNAcylation in several cell lines. Using the fluorescent reporter, we screened 120 natural products and obtained one compound, sesamin, which could markedly inhibit protein O-GlcNAcylation in HeLa and human colorectal carcinoma-116 cells and repress their migration in vitro. Objectives The objective of this study was to evaluate the efficacy of different dosing regimens of ceftazidime/avibactam (CZA) in patients with Klebsiella pneumoniae carbapenemase-producing K. pneumoniae (KPC-Kp) pulmonary infections. Methods A total of 70 KPC-Kp strains were isolated from sputum and bronchoalveolar lavage samples of patients with pulmonary infections in three hospitals in northern China from April 2015 to October 2015. Monte Carlo simulation (MCS) was performed using population pharmacokinetic parameters of CZA combined with the minimum inhibitory concentration (MIC) distributions gained from antimicrobial susceptibility testing to predict the efficacy of different dosing regimens. Various CZA dosing regimens were modelled using MCS. Results The in vitro study showed potent activity of CZA against KPC-Kp strains with MIC50/90 values of 1/2 mg/L, with a susceptibility rate of 95.7%. The values of cumulative fraction of response (CFR) for bactericidal (50%fT>5 × MIC) target were as follows: for patients with creatinine clearance (CLCr) >51 mL/min, the CFR was 96.01% for 2.5 g CZA every 12 h (q12h) and 97.14% for 2.5 g CZA every 8 h (q8h); and for patients with moderate renal impairment (CLCr >30 to ≤50 mL/min), the CFR was 95.75% for 1.25 g CZA q12h and 97.09% for 1.25 g CZA q8h. Conclusion This study indicated that the recommended dose of CZA can provide adequate pharmacodynamic exposure for treating KPC-Kp pneumonia.

Objectives The purpose of this study was (i) to investigate whether nanocomposite poly(methyl-methacrylate)-zinc oxide nanowires (PMMA-ZnO-NWs) have C. albicans antibiofilm activity; (ii) to evaluate the interaction between components of the nanocomposites based on PMMA-ZnO-NWs by Raman spectroscopy; and (iii) to assess ultrastructural alterations. Design Sixty-eight rectangles (17 PMMA (control) and 51 PMMA–ZnO-NWs (250, 500, 1000 ppm ZnO nanowires) were fabricated. C. albicans ATCC 10231 and a C. albicans clinical strain were tested. Adherence, biofilm formation and ultrastructural alterations were assessed by transmission electron microscopy. Raman mapping images and spectra were analyzed using main component analysis. Results Nanocomposite PMMA-ZnO-NWs inhibited the formation of C. albicans biofilms 94% at 1000 ppm and 80% at 500 ppm against both C. albicans strains. PMMA-ZnO-NWs induced ultrastructural alterations, including cell wall damage and disorganization of the cytoplasmic membrane, resulting in cell lysis. Raman spectroscopy showed new vibrational modes (300–365–485–600 cm−1) for PMMA and ZnO-NW interactions. Conclusions PMMA-ZnO-NWs inhibited C. albicans dose-dependent biofilm formation and led to changes in the structures and cell membrane. Raman spectroscopy showed chemical interactions between ZnO-NWs and PMMA, as suggested by the appearance of new bands at 301 and 485 cm−1.

Background Trachyspermum ammi (commonly known as Ajwain), a medicinal plant of the Apiaceae family is scientifically acknowledged to harbor potential bioactive compounds for the treatment of gastrointestinal issues, loss of appetite, bronchial difficulties, cough, inflammation, diarrhoea, headache, hypertension, stomach discomfort, bronchitis and influenza. Candida albicans is generally a commensal fungus found in the gastrointestinal and genitourinary systems. Objective This study was focused on secondary metabolites of T. ammi and its effects towards candidiasis infection as caused by C. albicans. Methods Phytochemical components of T. ammi as a crude extract were extracted through maceration method using three polar (ethanol, methanol and water) and two non-polar (chloroform and diethyl ether) solvents and subjected to 14 phytochemical tests. Further, the crude extract of T. ammi was analyzed over gas chromatography–mass spectroscopy (GC–MS) and Fourier transform infrared spectroscopy (FTIR). Evaluation of antimicrobial property of the extract was carried out by minimum fungal concentration (MFC) and minimal inhibitory concentration (MIC). In addition, cell reduction assay was performed using flowcytometry to confirm the antifungal effect of Ajwain crude extract. Results The aqueous extract showed high presence of phytochemicals including alkaloids, carbohydrates, flavonoids, resins, steroids, tannins, inorganic acids, organic acids, phenolic compounds, amino acids, protein and coumarins. GCM S analysis revealed seven bioactive compounds, in which thymol was detected in significant amount in the chromatogram. FTIR performance showed the presence of various stretching vibration including O H, C H, C C, C O, C N and C O C. However, the MFC and MIC of Ajwain extracts using different solvent showed that the methanolic extract possesses maximum antifungal efficacy at 250 μg/ml and 15.6 μg/ml, respectively. In addition, cell reduction assay exhibited significant cell reduction in Ajwain methanolic extract compared to the other crude extracts used in the study. Conclusion Overall, the findings revealed that Ajwain methanolic crude extract has antifungal activity against C. albicans; however, that further needs to be established at molecular level.

The present study. has successfully identified the nitrate reducing bacteria present in the cooling water system and also investigated the performance of industrially applied biocide and inhibitor on the bacterial inhibition. In order to carry out the objective of this study, facilities and methods such as 16S rRNA gene sequencing, Lowry assay, SEM, EIS, ICP-MS and weight loss analysis were being utilized. In this study, two out of the five morphologically dis- similar colonies identified through 16S rRNA gene sequencing, namely the Massilia timonae and the Pseudomonas, were being utilized in the biocorrosion study on copper metal. From the surface analysis using SEM demonstrated the phenomenon of biofilm formation on the copper surface. 2-methylbenzimidazole has the addition of methyl group in the diazole ring position of benzimidazole it has create basicity environment and inhibit the metal deterioration. Meanwhile, it is also deducible from the EIS and protein analysis that com- bination of biocide with either of the inhibitors gives rise to better biocorrosion suppression (0.00178 mpy and 0.00171mpy) as compared to the sole effect of either biocide or inhibitor (0.00219 mpy, 0.00162 and 0.00143). Biocorrosion system biocide with MBM was found to exhibit 65% corrosion inhibition efficiency. Moreover, adoption of 2-Methylbenzimidazole seems to display better performance as compared to Multionic 8151, which is adopted in cooling water system.

Graphical abstract, Natural products and traditional medicine are rich sources for developing therapeutics for chronic inflammatory diseases. However, the way from natural products/traditional medicines to Western pharmaceutical practices is not always straightforward. According to the World Health Organization (WHO), chronic diseases are the greatest threat to human health. 3 of 5 people die due to chronic inflammatory disorders worldwide like chronic respiratory diseases, stroke, cardiovascular diseases, cancer, diabetes, and obesity. Various nonsteroidal anti-inflammatory drugs (NSAIDs) are used to reduce inflammation and pain, but there are many side effects of these drugs' administration. Medicinal plants have therapeutic anti-inflammatory effects with low or no side effects. Ziziphus plant species are generally safe and not toxic to humans. Many studies on the Ziziphus species have shown that their therapeutic properties are attributed to the roots, leaves and fruits. Unfortunately, Ziziphus species from different regions worldwide with anti-inflammatory properties have not been documented in a single review paper. Therefore, it is crucial to establish ethnobotanical knowledge and applications of Ziziphus species against chronic inflammatory diseases. The current article exhaustively reviews phytochemical profile, pharmacological studies, toxicological effects, and ethnobotanical uses of Genus Ziziphus in chronic anti-inflammatory diseases. The present review article also highlights the most promising experimental data on Ziziphus extracts and pure compounds active in clinical trials and animal models of chronic inflammatory diseases. This review would be a valuable resource for contemporary researchers in the field to understand the promising role of the Ziziphus genus in chronic inflammatory disorders.

Background Biomedical research, recently, focus more on searching for biomasses that contain extractable biologically active components for formulating new drugs. Halophytes growing in hyper saline conditions are expected to produce stress alleviating bioactive compounds. These phytochemicals could be the better raw materials for formulating new drugs. Methods The purpose of this work was to describe physiologically active compounds from Acanthus ilicifolius and Heliotropium curassavicum that had antimicrobial, antioxidant and nutraceutical properties utilizing a variety of solvents. Analysis of bioactive compounds included the application of common phytochemical screening assays, proximate analysis, FTIR analysis and antioxidant assays. The disc diffusion technique was used to determine the antibacterial activity of the plant extracts. Results Highest extraction yield was observed with methanol. A. ilicifolius methanolic extracts included a variety of bioactive components, including alkaloids, saponins, phenolics, flavonoids, steroids, cardiac glycosides, tannins, and terpenoids. H. curassavicum extracts showed the presence of all the phytochemicals except cardiac glycosides. The overall phenolic concentration and antioxidant capacity of A. ilicifolius were substantially greater. The antimicrobial assays explored that among the tested bacterial pathogens viz., Bacillus subtilis, Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, Streptococcus pneumoniae, significant level of control was observed in E. coli, K. pneumoniae and B. subtilis, which were significantly susceptible to both the plant extracts at a concentration of 50 μg/ml. Conclusion The reports from the current investigation explored the possibility of utilizing these halophytes in nutraceutical formulations. The current study sheds light on the possibility of halophytes as natural secondary metabolites and bioactive chemicals with potential for antimicrobials.

Emerging artemisinin resistance in Plasmodium falciparum malaria has the potential to become a global public health crisis. In Southeast Asia, this phenomenon clinically manifests in the form of delayed parasite clearance following artemisinin treatment. Reduced artemisinin susceptibility is limited to the early ring stage window, which is sufficient to allow parasites to survive the short half-life of artemisinin exposure. A screen of known clinically-implemented antimalarial drugs was performed to identify a drug capable of enhancing the killing activity of artemisinins during this critical resistance window. As a result, lumefantrine was found to increase the killing activity of artemisinin against an artemisinin-resistant clinical isolate harboring the C580Y kelch13 mutation. Isobologram analysis revealed synergism during the early ring stage resistance window, when lumefantrine was combined with artemether, an artemisinin derivative clinically partnered with lumefantrine. A high concentration of nitrate and total nitrogen (N) in Chinese violet (CV) weed (Asystasia gangetica) has a potential benefit as a digestible protein source for ruminant production in humid tropical regions. We conducted a study using twelve Kacang goats in a crossover design to investigate the effect of CV weed on intake, nutrient utilization, and growth. Four dietary treatments were tested: Guinea grass (Panicum maximum) ad libitum (GG), GG ad libitum and CV (1% live weight, LW) (GG + CV), CV ad libitum (CV), and CV ad libitum plus cassava chip (1% LW) (CV + CC) (dry matter, DM basis). The apicomplexan parasite Neospora caninum is an important causative agent of congenital neosporosis, resulting in abortion, birth of weak offspring and neuromuscular disorders in cattle, sheep, and many other species. Among several compound classes that are currently being developed, two have been reported to limit the effects of congenital neosporosis: (i) bumped kinase inhibitors (BKIs) target calcium dependent protein kinase 1 (CDPK1), an enzyme that is encoded by an apicoplast-derived gene and found only in apicomplexans and plants. CDPK1 is essential for host cell invasion and egress; (ii) endochin-like quinolones (ELQs) are inhibitors of the cytochrome bc1 complex of the mitochondrial electron transport chain and thus inhibit oxidative phosphorylation. We here report on the in vitro and in vivo activities of BKI-1748, and of ELQ-316 and its respective prodrugs ELQ-334 and ELQ-422, applied either as single-compounds or ELQ-BKI-combinations. In vitro, BKI-1748 and ELQ-316, as well as BKI-1748 and ELQ-334, acted synergistically, while this was not observed for the BKI-1748/ELQ-422 combination treatment. In a N. caninum-infected pregnant BALB/c mouse model, the synergistic effects observed in vitro were not entirely reproduced, but 100% postnatal survival and 100% inhibition of vertical transmission was noted in the group treated with the BKI-1748/ELQ-334 combination. The present study makes an attempt to describe spectral characteristics of the surface features in some parts of the Khuzestan region, using reflectance spectroscopy and ASTER data related to the hydrocarbon micro-seepage in Maroun and Mansouri oil fields. The study involves surface sediment sampling with appropriate dispersion of the samples, and laboratory analysis in terms of sediment and mineralogy through spectroscopy analysis and geological map. Soil samples were collected from 45 and 20 locations points for Maroun and Mansouri oilfields, respectively. ASD-Fieldspec3 spectroradiometer and The Spectral Geologist (TSG) software were used to determine the hydrocarbon absorptions and their mineral constituents. Four classes were identified as hydrocarbon absorption ranges, i.e.: 1220–1320 and 1350–1550 nm indicating presence of light hydrocarbons; 1700–1750 and 2290–2360 nm pertaining to heavy hydrocarbons. Results suggest that Maroun oilfield samples have stronger absorptions in heavy hydrocarbons absorption but weaker for the Mansouri oilfield samples. The Mansouri samples have an acceptable absorption in light hydrocarbons absorption (1175 nm). Furthermore, these spectra were used as ground truth for creating mineral zonation maps using Spectral Angle Mapper (SAM) classification applied to ASTER data. The results also suggest that surface indicators of hydrocarbon are associated with alteration minerals such as kaolinite, siderite and goethite for the Maroun oilfield whereas Mansouri oilfield exhibits only kaolinite group of minerals. In addition, a comparison by reflectance between the contaminated and uncontaminated soil samples was also carried out.

In fruit production, the application of the plant growth regulator 1-(2-chloro-4-pyridyl)-3-phenylurea (CPPU) dulls the fruit aroma. Gas chromatography–mass spectrometry and transcriptome analyses were performed on CPPU-treated and pollinated fruits to determine how CPPU affects the production of aroma in melon fruit. The results showed that the contents of two important esters (benzyl acetate and phenethyl acetate) in the CPPU-treated fruits were significantly lower than those in the pollinated fruits. Transcriptome sequencing data revealed that most differentially expressed genes were involved in “phenylalanine metabolism” pathway, and their expression was significantly decreased in the CPPU-treated fruits. Further analysis showed that the phenylalanine content in the CPPU-treated fruits was significantly higher than that in the pollinated fruits. In summary, CPPU application interferes with phenylalanine metabolism in melon fruits and affects the production of aromatic esters.

Complex diseases caused by Meloidogyne incognita and Fusarium fungus in cucumber is the most destructive disease under polyhouses. The experiment was conducted in the polyhouse of the Department of Horticulture, CCS HAU, Hisar, Haryana, India during summer season (2015–16) to evaluate the potential of bacterial and fungal biocontrol agents against Meloidogyne incognita and Fusarium oxysporum f. sp. cucumerinum in cucumber. Bioagents - Trichoderma viride (Tv), Pseudomonas fluorescence (Pf), Purpureocillium lilacinum (Pl) were taken 10 and 20 g kg−1 seed and bioagents liquid formulation, 10- and 15-ml kg−1 seed, were mixed with the potted soil. Chemical as well as untreated check were also maintained. All the treatments significantly improved the plant growth parameter, viz., shoot length (SL), root length (RL), fresh shoot weight (FSW), fresh root weight (FRW), dry shoot weight (DSW) and dry root weight (DRW) as compared to untreated check. However, significant reduction in nematode population and maximum improvement in plant growth parameter was recorded with carbofuran followed by higher dose of bioagents liquid formulation. Among the bioagents, bioagents liquid formulation was most effective in suppressing root knot nematode galling (43 / root system) and final population in soil (131 J2s / 200 cc soil) and fungus wilt incidence (25 %) at 30th day of after germination and significantly improved the plant growth parameters - shoot length (147.3 cm), fresh shoot weight (55.6 g), dry shoot weight (22.51 g) and dry root weight (4.50 g) from other bioagents. Bioagents liquid formulation was effective in suppression of root-knot nematode and fungus complex disease than the powder formulations of bioagents. More studies should be needed in future to evaluate the efficacy of bioagents as seed treatments and soil applications under field conditions.

Diarrhea is a prevalent gastrointestinal problem associated with fatal implications. It is a huge public health concern that requires better alternatives to current drugs. This study investigated the mechanisms involved in the antidiarrheal activity of Anacardium occidentale (Ao) stem bark extract, a plant commonly used in the management of diarrhea in Nigeria. Methanolic stem bark extract of the plant was partitioned into 3 fractions; hexane fraction (AoHF), ethyl acetate fraction (AoEF) and methanol fraction (AoMF). In vitro studies on the effect of these fractions using guinea pig ileum (GPI) muscle contraction, as well as the modulatory effect of standard agonists and antagonists on such contraction, revealed AoEF as the most active fraction. In vivo studies to assess the effect of AoEF on the dopaminergic, muscarinic and serotonergic pathways were carried out using gastric emptying (GE) and gastrointestinal transit (GT) as experimental end points. AoEF was subjected to GC-MS analysis, while the identified compounds were docked with the muscarinic acetylcholinesterase receptor (M3) using AutodockVina. Results indicated that AoEF inhibited GE and GT via inhibition of M3. In addition, GC-MS analysis revealed the presence of 24 compounds in AoEF; while docking indicated that octadecanoic acid 2-(2-hydroxylethoxy) ethyl ester exhibited the highest binding affinity for M3. This study indicated that the antidiarrheal activity of Ao is through its antimotility effect via the inhibition of the muscarinic pathway. And since none of the identified compounds exhibited higher binding affinity for M3 relative to loperamide, the antimotility activity of these phytoconstituents may be via synergism.

Background Arbekacin is a broad-spectrum aminoglycoside with activity against some Gram-positive and Gram-negative bacteria. Methods Arbekacin minimum inhibitory concentration (MIC) values were determined for 296 drug-resistant Gram-negative bacilli, and compared to previously determined plazomicin, amikacin, gentamicin, and tobramycin MIC values. Results The MIC values required to inhibit 50% and 90% of isolates (MIC50 and MIC90, respectively) were 16 and >128 μg/ml, respectively. Conclusions Arbekacin showed similar MIC50 values to amikacin and gentamicin, a lower MIC50 value than tobramycin, and a higher MIC50 value than plazomicin.

The present study aimed to determine in vitro antimicrobial activities and phytochemical analysis of Harrisonia abyssinica and Vepris simplifolia which are commonly used as traditional medicine in Tanzania. Sensitivity of bacterial and fungal species against plants extracts were determined using serial microdilution method. In this method, the lowest inhibitory concentration which prevented microbial growth considered as minimum inhibitory concentration (MIC). The study also evaluated phytochemical compounds present in the leaf, stem and root barks of H. abyssinica. It was revealed that eight extracts from H. abyssinica inhibited growth of three bacteria namely Staphylococcus aureus, Enterococcus faecium and Streptococcus uberis at MIC value less than 1 mg/mL. It was further revealed that, three extracts from V. simplifolia exhibited high antibacterial and antifungal activity. The preliminary phytochemical analysis revealed presence of various phytochemicals namely alkaloids, terpenoid, flavonoid, tannin and saponin. It was concluded that presence of large number of phytochemicals in the plant extracts may be associated with pharmacological properties of H. abyssinica and V. simplifolia and therefore this study provide alternative to synthetic antimicrobial agents.

Treatment of burn injuries with Mesenchymal stem cells (MSCs) is a great promise due to their unique properties. As two natural and functional wound dressing, Chitosan and Aloe-Vera gel assist wound regeneration by providing a proper environment. In the current study, we aimed to compare the effect of encapsulated BMSCs in Chitosan-based gel and Aloe-Vera gel on the healing of grade-II burn injuries compared to other groups in the rat. After creation of a 2 × 2 cm grade-II burn on dorsal skin of rats, treatments were performed for each group. The wound closure rate and healing properties were evaluated by histopathological analysis on 7, 14, 21 and, 28 days post-treatment. The expression rate of VEGF, Collagen-I and Collagen-III genes was also assessed on days 3, 7, 14, 21 and 28 performing qRT-PCR. The full wound healing with inconsiderable scar formation was achieved for Aloe-vera/BMSCs and Chitosan/BMSCs group on 28th day post-treatment. Pathological results also demonstrated the highest angiogenesis and granulation tissue formation for Aloe-vera/BMSCs and Chitosan/BMSCs groups respectively. The expression level of VEGF, Collagen-I, and Collagen-III genes was significantly higher in these groups on days 14 and 21, compared to other groups. Results demonstrated the synergistic effect of BMSCs when combined with Chitosan or Aloe-vera gel enhanced the healing process of wound healing more than chitosan gel treatment. Therefore, this gel can be considered as effective approaches for treatment of burn injuries.

The success of Next generation sequencing (NGS) technology over the past decades has led to a better understanding of complex microbial communities in such environment. Here, we employed NGS to monitor sulfur oxidizing bacteria (SOB) community from cassava starch industry (BPT), which can oxidize H2S into sulfate or elemental sulfur under aerobic conditions. Microbial sludge was collected from two sampling points comprising starting sludge (BPT1) and recirculating sludge (BPT2) of the biotrickling filter. gDNA was extracted from BPT1 and BPT2 samples and removed other contaminations by using the GenepHarM Gel/PCR Kit (Geneaid). DNA concentrations and DNA purity were analyzed by using gel electrophoresis and NanoDrop™ 1000 Spectrophotometer, respectively. Analysis of SOB species using NGS technique was based on the hypervariable regions V4 on the 16S rRNA gene. The comparative genomic hybridization (CGH) microarray probes for detection and identification of SOB species-strain that high expression of soxAXBYZ and fccAB genes in the cassava starch industry. For the recirculating sludge (BPT2), the bacterial communities were dominated by Sulfurimonas (13.7%) followed by Synechococcus (9.4%), Hydrogenophaga (6.5%), Methanosaeta (2.4%), and Acidithiobacillus (2.1%), the same dominant genera were also found in the starting sludge (BPT1), but with slightly different abundancy. The heatmap revealed that Acidithiobacillus caldus species (33.9%–49.9%) could play a vital role in eliminating H2S in biogas from the cassava starch industry. The CGH microarray indicated that Thiothrix, Paracoccus, Sulfurimonas, Hydrogenophaga, Magnetospirillum, Rhodoplanes, Syntrophomonas, Pseudomonas, Sulfuricurvum, and Arcobacter were dominant genus, that has a high expression of sulfur oxidation genes in biotrickling filter.

Background: Microalgae are microorganisms that produce various products, for example, pigments, mainly carotenoids. This study aimed to used the strain of Muriellopsis sp. and to evaluate their behavior when grown in freshwater and seawater, along with indoor and outdoor conditions for both cultures. Growth of the strain was evaluated by determining its biomass, lutein productivity with high-performance liquid chromatography (HPLC), and antioxidant activity by using the 2,2-diphenyl-1-picrilhydrazil (DPPH method). Results: Muriellopsis sp. strain in indoor cultures showed an increased antioxidant activity. In outdoor conditions, both cultures showed increased cells number, concentration of biomass, and lutein productivity. The percentage of lutein obtained from the strain MCH in indoor conditions was 25 times higher than that reported for calendula, reaching 0.75% of lutein in Muriellopsis sp. cultured in seawater, followed by 0.6% in Muriellopsis sp., cultures in freshwater at day 12 of both cultures. These values exceed that of microalgae Scenedesmus almeriensis , which reaches 0.53% lutein. Conclusions: The results show that the native strain of the Atacama Desert is one of the largest producers of lutein as compared to those reported to date. The study demonstrated the feasibility of producing this carotenoid with well-known properties to prevent some diseases due to its high nutritional value. Muriellopsis sp. cultivation in open-air seawater is a good precedent for developing mass production of this species in an area where freshwater is scarce and costly.
