[Objective] In order to provide theoretical basis for pollen viability identification, artificial pollination and cross breeding of large- fruited yellow passion fruit, the effects of different culture medium in vitro and storage conditions on pollen germination were analyzed. [ Method] Pollens were collected from newly blooming flowers of large-fruited yellow passion fruit The suitable concentrations of sucrose, boric acid, calcium nitrate and suitable pH for pollen in vitro culture were screened by Li, (5 4) onhogonal design. The effects of sampling time of flowers, storage humidity and temperature on pollen viability were studied by using the most suitable pollen culture medium. The suitable storage condition for pollen of large-fruited yellow passion fruit was determined. [Result] The results of onhogonal test showed that the order of influencing factors on pollen germination rate of large-fruited yellow passion fruit was calcium nitrate > sucrose > boric acid > pH. The optimum medium for pollen germination in vitro was 100. 00 g/L sucrose + 0. 02 g/L boric acid + 0. 40 g/L calcium nitrate, and the medium pH was 5. 0. The pollen germination rate in the optimum medium was 85. 55 % . The pollen germination rate of large-fruited yellow passion fruit was more than 80. 00% when the sampling time was from 11 :30 to 17 :30. The pollen viability was the highest in the third hour ( 16: 30) after flowering, which was significantly higher than the germination rate of pol- lens collected at other times except 13 : 30 and 15 : 30 (P <0. 05, the same as below). The results of storage test showed that, the pollen germination rate of natural air drying on sulfur paper treatment for 3 - 48 hours was significantly higher than that of sealed centrifugation tube storage at room temperature and storage in desiccator on sulfur paper for the same time, and the pollen germination rate of 48 h storage was still 34. 28%. The pollen germination rate was 61. 49% after natural air drying for 5 hours and sealed in centrifugal tube at 4 °C for 24 hours, which was significantly higher than that (55. 22%) after natural air drying for 5 hours and sealed in centrifuge tube at - 20 °C for 24 hours. [Conclusion] The optimum medium for pollen germination in vitro of large-fruited yellow passion fruit is 100. 00 g/L sucrose+ 0. 02 g/L boric acid+ 0. 40 g/L calcium nitrate, and pH is 5. 0. 【Objective】stem disinfection, bud induction and proliferation were explored during tissue culture to provide technical support for mass seedling production.【Method】 Taken stem of Hylocereus undatus cultivar Guihonglong No. 1 as the explants, the effects of different factors on stem disinfection, bud inducement and proliferation were studied.【Result】 Stem size was the main factor affecting the pollution rate of stem disinfection, followed by the effect of sterilization time and stem mature degrees. The sterilization time had the greatest influence on the bud induction, followed by the 6-BA concentration. Both the stem mature degrees and stem size had little effect on bud induction. 6-BA 4.0 mg/L had the best induction effect. The higher concentration of 6-BA showed the higher multiplication ratio of buds, but when 6-BA concentration reached 4.0 mg/L, the buds appeared vitrification. The effect of IAA was better than that of NAA, and the effect of bud proliferation was better when IAA was 0.1 mg/L. PP333 was better than AC in inhibiting vitrification and promoted subculture multiplication of Hylocereus undatus. 【Conclusion】 The optimal treatment for stem disinfection and bud induction was suggested as: sliced the health stems of about 1 months into pieces with single bud, and disinfected them with 0.1 % HgCl2 solution, and inoculated the explants in medium of MS + 6-BA 4.0 mg/L. [Objective] This paper aimed to the tolerance and behavioral changes of Triplophysa (Itedinichthys) yarkandensis (Day) under the salinity and alkalinity.[Method] Triplophysa (Itedinichthys) yarkandensis (Day) was used as objects, defining a low-temperature environment (13 ± 0.5) Ti, their preferences under the gradient concentrations of salinity (S)/alkalinity (Aik ) were determined, and the effects of salinity (S)/alkalinity (Aik) toxicity on the Triplophysa (iledinichthys) yarkandensis (Day) in Tarim river were studied.[Result] When Aik was around pH 6.5 -7.5 with temperature(13 ± 1 ) Ti, the TC%) of S to T.(H.)yarkandensis (Day) were 20.0870 %c, 16.2490 %c, 13.3300 %c, 12.5680 %c and 12.3930 %c at 12, 24, 48, 72 and 96 hours, respectively, and the safe S was 4.1676 %c.The LC50 of Alkto T.(H.) yarkandensis (Day) were 12.1590, 9.9776, 9.5076, 8.6485 and 4.5220 g · I.1 at 12, 24, 48, 72 and 96 hours, respectively, and the safe Aik was 2.7179 g · L-l.Among them, 4 %c was the main preference area, and 3 -4 g · L-l was the range of Aik preference.In conclusion, although the low temperature of S/Alk toxicity tolerance slowed, behavior preferences were also different of T.(II.) yarkandensis (Day), and the S/Alk stress of Tarim water salinization seriously affected the growth and development of T.(II.) yarkandensis (Day).[Conclusion]The present study would provide a scientific basis for the enhancement and protection of T.(II.) yarkandensis (Day). The effects of salinity, alkali and weight on oxygen consumption and ammonia excretion rate of Triplophysa (Hedinichthys) yarkandensis (Day) were studied by means of the experimental ecology method in laboratory. Results showed that salinity, alkali and weight had significant effects on the oxygen consumption and ammonia excretion rate( P<0.05), when salinity was 5 g · L-1and the oxygen consumption rate of T. H. yarkandensis (Day) increased as the salinity decreased. The ammonia excretion rate of T. H. yarkandensis (Day) decreased as the salinity increased, the ammonia excretion rate was 0. 084 mg · （g · h）-1 when the salinity was 20 g · L-1. The oxygen consumption rate from the first of T. H. yarkandensis (Day), then as the alkali decreased, the alkali maximum was 11.922 mmol · L-1, and when alkali was 31.922 mmol · L-1the ammonia excretion rate from the first of T. H. yarkandensis (Day) increased. The oxygen consumption rate of T.H. yarkandensis (Day) increased as weight increased, and ammonia excretion rate of T. H. yarkandensis (Day) increased as weight decreased. The O/N ratio of T. H. yarkandensis (Day) increased with the salinity and alkali decreased, and the O/N ratio T. H. yarkandensis(Day) increased as weight increased, when the salinity was 5 g · L-1and the alkali was 11. 922 mmol · L-1. Therefore, so that salinity, alkali and weight were involved in osmoregulation of T. H. yarkandensis (Day) adapting for different salinity, to change the oxygen consumption and ammonia excretion rate of T. H. yarkandensis (Day). It is good at the oxygen consumption and ammonia excretion rate of T. H.yarkandensis (Day), when the salinity is 5 g · L-1and the alkali is 11.922 mmol · L-1, may be related to the Na+/K+-ATP.
