Factors affecting survival of parasites introduced to new geographical regions include changes in environmental temperature. Protopolystoma xenopodis is a monogenean introduced with the amphibian Xenopus laevis from South Africa to Wales (probably in the 1960s) where low water temperatures impose major constraints on life-cycle processes. Effects were quantified by maintenance of eggs from infections in Wales under controlled conditions at 10, 12, 15, 18, 20 and 25°C. The threshold for egg viability/ development was 15°C. Mean times to hatching were 22 days at 25°C, 32 days at 20°C, extending to 66 days at 15°C. Field temperature records provided calibration of transmission schedules. Although egg production continues year-round, all eggs produced during >8 months/ year die without hatching. Output contributing significantly to transmission is restricted to 10 weeks (May-mid-July). Host infection, beginning after a time lag of 8 weeks for egg development, is also restricted to 10 weeks (July-September). Habitat temperatures (mean 15·5°C in summer 2008) allow only a narrow margin for life-cycle progress: even small temperature increases, predicted with 'global warming', enhance infection. This system provides empirical data on the metrics of transmission permitting long-term persistence of isolated parasite populations in limiting environments.

The monogenean Protopolystoma xenopodis has been established in Wales for >40 years following introduction with Xenopus laevis from South Africa. This provides an experimental system for determining constraints affecting introduced species in novel environments. Parasite development post-infection was followed at 15, 20 and 25°C for 15 weeks and at 10°C for ⩾1 year and correlated with temperatures recorded in Wales. Development was slowed/arrested at ⩽10°C which reflects habitat conditions for >6 months/year. There was wide variation in growth at constant temperature (body size differing by >10 times) potentially attributable in part to genotype-specific host-parasite interactions. Parasite density had no effect on size but host sex did: worms in males were 1·8 times larger than in females. Minimum time to patency was 51 days at 25°C and 73 days at 20°C although some infections were still not patent at both temperatures by 105 days p.i. In Wales, fastest developing infections may mature within one summer (about 12 weeks), possibly accelerated by movements of hosts into warmer surface waters. Otherwise, development slows/stops in October-April, delaying patency to about 1 year p.i., while wide variation in developmental rates may impose delays of 2 years in some primary infections and even longer in secondary infections.

Gyrodactylus specimens infecting both anadromous Arctic charr (Salvelinus alpinus) from River Signaldalselva (northern Norway) and resident Arctic charr from Lake Pålsbufjorden (southern Norway) were identified as G. salaris using molecular markers and morphometrics. The infection in Pålsbufjorden represents the first record of a viable G. salaris population infecting a host in the wild in the absence of salmon (Salmo salar). G. salaris on charr from Signaldalselva and Pålsbufjorden bear different mitochondrial haplotypes. While parasites infecting charr in Signaldalselva carry the same mitochondrial haplotype as parasites from sympatric Atlantic salmon, G. salaris from charr in Pålsbufjorden bear a haplotype that has previously been found in parasites infecting rainbow trout (Oncorhynchus mykiss) and Atlantic salmon, and an IGS repeat arrangement that is very similar to those observed earlier in parasites infecting rainbow trout. Accordingly, the infection may result from 2 subsequent host-switches (from salmon via rainbow trout to charr). Morphometric analyses revealed significant differences between G. salaris infecting charr in the 2 localities, and between those on sympatric charr and salmon within Signaldalselva. These differences may reflect adaptations to a new host species, different environmental conditions, and/or inherited differences between the G. salaris strains. The discovery of G. salaris on populations of both anadromous and resident charr may have severe implications for Atlantic salmon stock-management as charr may represent a reservoir for infection of salmon.

The southern fiddler ray, Trygonorrhina fasciata (Rhinobatidae), is parasitized by 3 monogenean (platyhelminth) species from 3 families on 3 different sites of the host: Calicotyle australis (Monocotylidae) from the cloaca, Pseudoleptobothrium aptychotremae (Microbothriidae) from the skin and Branchotenthes octohamatus (Hexabothriidae) from the gills. Cues that promote egg hatching were investigated for each species and the behaviour of their larvae was also documented. Eggs were laid by parasites in vivo and maintained at 22 degrees C. Three different egg hatching and host finding strategies were discovered. Calicotyle australis eggs hatched spontaneously with a strong diurnal rhythm that is likely to be under circadian control. The larva is ciliated, photo-responsive and can survive for up to 24 h at 22 degrees C after hatching. Pseudoleptobothrium aptychotremae may have a 'bet-hedging' strategy. Some eggs hatched spontaneously and rhythmically. However, since the hatching success was low, it is possible that other eggs require a different cue provided by the host. The larva is also ciliated but shows no photo-response and was observed to remain active for <4 h at 22 degrees C after hatching. Branchotenthes octohamatus has a 'sit-and-wait' strategy that depends on mechanical disturbance to stimulate hatching. The larva is unciliated, shows no photo-response but may survive for more than 2 days at 22 degrees C after hatching. The implications of hatching strategy, larval behaviour and morphology in the goal to find a host are discussed for each species.

The viviparous monogenean Gyrodactylus salaris continues to devastate Norwegian Atlantic salmon populations despite the extreme measures taken to control this pathogen. Increased understanding of parasite biology is needed to develop alternative control and management strategies of wild Atlantic salmon. We have examined temperature-dependent survival of G. salaris, both on and off the host. At 18 degrees C, survival off the host was 1 day, but at 3 degrees C parasites survived for 4 days. However, in contrast to assumptions made by earlier authors, many parasites remained with their host following its death. Ultrastructural evidence indicated that G. salaris individuals can feed on a dead host, and laboratory tests demonstrated that worms on their hosts more than double their life-span compared with individuals maintained off the host. Experimental infections also demonstrated that establishment and subsequent population growth of parasites previously maintained on dead hosts for 3 days, was similar to that of parasites transferred directly between living hosts. Hence, for G. salaris, dead infected hosts may increase the chances of successful transmission and be a potential important infection source in rivers and hatcheries.

Using indirect immuno- and enzyme-cytochemical techniques, interfaced with confocal scanning laser microscopy and standard optical microscopy, neuronal pathways have been demonstrated in whole-mount preparations of the unpaired diporpae and freshly paired juvenile stages of Eudiplozoon nipponicum (Monogenea: Diplozoidae). All 3 main classes of neuronal mediators, cholinergic, aminergic and peptidergic, were identified throughout both central and peripheral elements of a well-differentiated orthogonal nervous system. Neural mapping revealed considerable overlap and similarity in staining of the nervous systems of the diporpa and adult worm. The main differences in the diporpa relate to the innervation of the temporary ventral sucker and dorsal papilla, structures which are unique to the larva and which enable fusion between worms but then disappear. Branches from the longitudinal nerve cords innervate these structures and appear to be involved in the process of somatic fusion, probably giving rise to the inter-specimen connections that later link the 2 central nervous systems in paired adult parasites. In the hindbody, there is extensive haptoral innervation associated with the developing clamps and small central hooks. Reactive neuronal components were found associated with the early stages of clamp development prior to connections being made with the extrinsic adductor muscle bundles. The muscle systems of the diporpa and juvenile stages comprise a lattice-like arrangement of circular, longitudinal and diagonal fibres that make up the body wall, together with buccal suckers, haptoral clamps and associated adductor muscles, and the transient ventral sucker. All have obvious importance to diporpae when they migrate over the gill and undertake body contact, torsion and fusion during the process of pairing. Behaviour during the pairing of diporpae is described.

Reproductive success of Gvrodactylus salaris from River Lierelva was compared experimentally on 3 stocks of salmon (12.5 degrees C +/- 0.2). Isolated fish from 2 susceptible Norwegian stocks (Rivers Lier and Alta) and 1 resistant Baltic stock of salmon (River Neva) were infected with a single gravid worm in order to record the temporal sequence of births and age at death of individual parasites. Establishment success (proportion of worms surviving to give birth) was generally low and mortality high, but significantly fewer worms survived on Neva (45% with mean survival of 3.5 days) compared to Alta and Lier fish (60 % mean survival 7.9 and 5.2 days, respectively). There was a dramatic difference in parasite fecundity between the host stocks: only 2 births occurred on Neva fish compared to third and fourth births on both Alta and Lier hosts. The timing of the first birth was more variable on Neva hosts and was significantly extended (mean 2.3 days) relative to that on Alta and Neva fish (1.8 days). However, timing of the second birth did not vary on any of the 3 salmon stocks. Age-specific mortality and fecundity data are consistent with exponential population growth of G. salaris on Alta and Lier fish but eventual extinction on Neva hosts. This is the first demonstration that gyrodactylids maintained on different host stocks exhibit variations in fecundity, development and mortality, which may in turn account for the variable virulence noted when G. salaris infects different salmonid hosts.
