Background: This study explored the feasibility of radiofrequency (RF)-based radiomics analysis techniques for the preoperative prediction of programmed cell death protein 1 (PD-1) in patients with hepatocellular carcinoma (HCC).Methods: The RF-based radiomics analysis method used ultrasound multifeature maps calculated from the RF signals of HCC patients, including direct energy attenuation (DEA) feature map, skewness of spectrum difference (SSD) feature map, and noncentrality parameter S of the Rician distribution (NRD) feature map. From each of the above ultrasound maps, 345 high-throughput radiomics features were extracted. Then, the useful radiomics features were selected by the sparse representation method and input into support vector machine (SVM) classifier for PD-1 prediction.Results and Conclusion: Among all the RF-based prediction models and the ultrasound grayscale comparative model, the RF-based model using all of the three ultrasound feature maps had the highest prediction accuracy (ACC) and area under the curve (AUC), which were 92.5% and 94.23%, respectively. The method proposed in this paper is effective for the meaningful feature extraction of RF signals and can effectively predict PD-1 in patients with HCC. [ABSTRACT FROM AUTHOR]

Background: Tumor location served as an important prognostic factor in glioma patients was considered to postulate molecular features according to cell origin theory. However, anatomic distribution of unique molecular subtypes was not widely investigated. The relationship between molecular phenotype and histological subgroup were also vague based on tumor location. Our group focuses on the study of glioma anatomic location of distinctive molecular subgroups and histology subtypes, and explores the possibility of their consistency based on clinical background.Methods: We retrospectively reviewed 143 cases with both molecular information (IDH1/TERT/1p19q) and MRI images diagnosed as cerebral diffuse gliomas. The anatomic distribution was analyzed between distinctive molecular subgroups and its relationship with histological subtypes. The influence of tumor location, molecular stratification and histology diagnosis on survival outcome was investigated as well.Results: Anatomic locations of cerebral diffuse glioma indicate varied clinical outcome. Based on that, it can be stratified into five principal molecular subgroups according to IDH1/TERT/1p19q status. Triple-positive (IDH1 and TERT mutation with 1p19q codeletion) glioma tended to be oligodendroglioma present with much better clinical outcome compared to TERT mutation only group who is glioblastoma inclined (median overall survival 39 months VS 18 months). Five molecular subgroups were demonstrated with distinctive locational distribution. This kind of anatomic feature is consistent with its corresponding histological subtypes.Discussion: Each molecular subgroup in glioma has unique anatomic location which indicates distinctive clinical outcome. Molecular diagnosis can be served as perfect complementary tool for the precise diagnosis. Background: The Eigenspace-based beamformers, by orthogonal projection of signal subspace, can remove a large part of the noise, and provide better imaging contrast upon the minimum variance beamformer. However, wrong estimate of signal and noise component may bring dark-spot artifacts and distort the signal intensity. The signal component and noise and interference components are considered uncorrelated in conventional eigenspace-based beamforming methods. In ultrasound imaging, however, signal and noise are highly correlated. Therefore, the oblique projection instead of orthogonal projection should be taken into account in the denoising procedure of eigenspace-based beamforming algorithm., Methods: In this paper, we propose a novel eigenspace-based beamformer based on the oblique subspace projection that allows for consideration of the signal and noise correlation. Signal-to-interference-pulse-noise ratio and an eigen-decomposing scheme are investigated to propose a new signal and noise subspaces identification. To calculate the beamformer weights, the minimum variance weight vector is projected onto the signal subspace along the noise subspace via an oblique projection matrix., Results: We have assessed the performance of proposed beamformer by using both simulated software and real data from Verasonics system. The results have exhibited the improved imaging qualities of the proposed beamformer in terms of imaging resolution, speckle preservation, imaging contrast, and dynamic range., Conclusions: Results have shown that, in ultrasound imaging, oblique projection is more sensible and effective than orthogonal subspace projection. Better signal and speckle preservation could be obtained by oblique projection compare to orthogonal projection. Also shadowing artifacts around the hyperechoic targets have been eliminated. Implementation the new subspace identification has enhanced the imaging resolution of the minimum variance beamformer due to the increasing the signal power in direction of arrival. Also it has offered better sidelobe suppression and a higher dynamic range.

Background: The measurement of dynamic parameters, such as the length of applanation and the amplitude of deformation, is significant for evaluating corneal properties. Most of the corneal properties (related to shape) including the anterior corneal curvature and the thickness of cornea can be easily measured using some existing techniques. However, they only provide the static or pseudo-dynamic analysis. Based on Corvis ST images, the dynamic features after corneal boundaries detection and parameter estimation will be helpful for corneal analysis., Material: The study included 40 eyes in normal group (ranging from 19 to 45 years old) and 30 eyes in keratoconus group (ranging from 16 to 40 years old). These eyes were examined by Corvis ST and for each one a sequence of 140 images was obtained. Besides, 11 subjects of each group were also tested by Pentacam., Methods: By analyzing the video from the Corvis ST imaging, the fully dynamic curvature topography is proposed to evaluate the response of the anterior corneal surface to the air puff. The new method not only quantitatively measures the intact variation of anterior corneal surface but also provides an intuitive way to observe the dynamic change of the anterior corneal surface in the whole air stream process. The proposed method consists of three main steps: cornea segmentation, curvature estimation and integrated visualization. An automatic segmentation method based on the combination of prior knowledge with phase symmetry and asymmetry theory is firstly presented to detect the corneal boundaries. The Landau-new method is then used to estimate the anterior corneal surface. The corneal dynamic topography is finally obtained by combining the dynamic parameters with the original Corvis ST video, which is an improvement of the fusion technique proposed by Li et al., Results and Conclusion: By comparing the segmentation results with manual method and built-in method of Corvis ST, the accuracy and robustness of our proposed segmentation method is demonstrated. The correctness of the estimated corneal anterior curvatures is also evaluated by comparing it with that of Pentacam which is considered to be able to provide the first-class measurement currently. The dynamic topography may be used to distinguish the dynamic behavior of normal corneas from that of keratoconus.

Background: Classification of breast ultrasound (BUS) images is an important step in the computer-aided diagnosis (CAD) system for breast cancer. In this paper, a novel phase-based texture descriptor is proposed for efficient and robust classifiers to discriminate benign and malignant tumors in BUS images., Method: The proposed descriptor, namely the phased congruency-based binary pattern (PCBP) is an oriented local texture descriptor that combines the phase congruency (PC) approach with the local binary pattern (LBP). The support vector machine (SVM) is further applied for the tumor classification. To verify the efficiency of the proposed PCBP texture descriptor, we compare the PCBP with other three state-of-art texture descriptors, and experiments are carried out on a BUS image database including 138 cases. The receiver operating characteristic (ROC) analysis is firstly performed and seven criteria are utilized to evaluate the classification performance using different texture descriptors. Then, in order to verify the robustness of the PCBP against illumination variations, we train the SVM classifier on texture features obtained from the original BUS images, and use this classifier to deal with the texture features extracted from BUS images with different illumination conditions (i.e., contrast-improved, gamma-corrected and histogram-equalized). The area under ROC curve (AUC) index is used as the figure of merit to evaluate the classification performances., Results and Conclusions: The proposed PCBP texture descriptor achieves the highest values (i.e. 0.894) and the least variations in respect of the AUC index, regardless of the gray-scale variations. It's revealed in the experimental results that classifications of BUS images with the proposed PCBP texture descriptor are efficient and robust, which may be potentially useful for breast ultrasound CADs.

Introduction: Estrogen plays an important role in the osteogenic differentiation of mesenchymal stem cells, while stem cells from apical papilla (SCAP) can contribute to the formation of dentin/bone-like tissues. To date, the effects of estrogen on the differentiation of SCAP remain unclear., Methods: SCAP was isolated and treated with 10⁻⁷ M 17beta-estradiol (E2). The odonto/osteogenic potency and the involvement of mitogen-activated protein kinase (MAPK) signaling pathway were subsequently investigated by using methyl-thiazolyl-tetrazolium (MTT) assay, and other methods., Results: MTT and flow cytometry results demonstrated that E2 treatment had no effect on the proliferation of SCAP in vitro, while alkaline phosphatase (ALP) assay and alizarin red staining showed that E2 can significantly promote ALP activity and mineralization ability in SCAP. Real-time reverse transcription polymerase chain reaction (RT-PCR) and western blot assay revealed that the odonto/osteogenic markers (ALP, DMP1/DMP1, DSPP/DSP, RUNX2/RUNX2, OSX/OSX and OCN/OCN) were significantly upregulated in E2-treated SCAP. In addition, the expression of phosphor-p38 and phosphor-JNK in these stem cells was enhanced by E2 treatment, as was the expression of the nuclear downstream transcription factors including phosphor-Sp1, phosphor-Elk-1, phosphor-c-Jun and phosphor-c-Fos, indicating the activation of MAPK signaling pathway during the odonto/osteogenic differentiation of E2-treated SCAP. Conversely, the differentiation of E2-treated SCAP was inhibited in the presence of MAPK specific inhibitors., Conclusions: The ondonto/osteogenic differentiation of SCAP is enhanced by 10⁻⁷ M 17beta-estradiol via the activation of MAPK signaling pathway.

Multiple myeloma is a common hematological malignancy that leads to the occurrence of bone lesions. The combination of traditional cytotoxic and novel agents is usually taken to treat multiple myeloma-related bone diseases. However, the curative effect is not very satisfactory. Stem cell-based therapy has been recently introduced and investigated, which represents a new frontier in the treatment of multiple myeloma. In a recent interesting study, Dr Atsuta and colleagues provide a new insight into the effects of mesenchymal stem cells on multiple myeloma via Fas/Fas ligand pathway, which rekindles the fire of hope for those patients suffering from multiple myeloma.

Background: Dental pulp stem cells (DPSCs) can be driven into odontoblast, osteoblast, and chondrocyte lineages in different inductive media. However, the differentiation potential of naive DPSCs after serial passaging in the routine culture system has not been fully elucidated., Results: DPSCs were isolated from human/rat dental pulps by the magnetic activated cell sorting based on STRO-1 expression, cultured and passaged in the conventional culture media. The biological features of STRO-1+ DPSCs at the 1st and 9th passages were investigated. During the long-term passage, the proliferation ability of human STRO-1+ DPSCs was downregulated as indicated by the growth kinetics. When compared with STRO-1+ DPSCs at the 1st passage (DPSC-P1), the expression of mature osteoblast-specific genes/proteins (alkaline phosphatase, bone sialoprotein, osterix, and osteopontin), odontoblast-specific gene/protein (dentin sialophosphoprotein and dentin sialoprotein), and chondrocyte-specific gene/protein (type II collagen) was significantly upregulated in human STRO-1+ DPSCs at the 9th passage (DPSC-P9). Furthermore, human DPSC-P9 cells in the mineralization-inducing media presented higher levels of alkaline phosphatase at day 3 and day 7 respectively, and produced more mineralized matrix than DPSC-P9 cells at day 14. In vivo transplantation results showed that rat DPSC-P1 cell pellets developed into dentin, bone and cartilage structures respectively, while DPSC-P9 cells can only generate bone tissues., Conclusions: These findings suggest that STRO-1+ DPSCs consist of several interrelated subpopulations which can spontaneously differentiate into odontoblasts, osteoblasts, and chondrocytes. The differentiation capacity of these DPSCs changes during cell passaging, and DPSCs at the 9th passage restrict their differentiation potential to the osteoblast lineage in vivo.
