Background: Chagas disease, or American trypanosomiasis, is an important neglected tropical illness caused by the flagellate protozoan Trypanosoma cruzi, which is primarily transmitted to humans by hematophagous insects of the subfamily Triatominae. Although knowledge on triatomine movement capabilities at the micro-geographical scale is of fundamental importance concerning the development of effective vector control strategies, it remains a poorly understood subject. Furthermore, survival rates and size estimates of natural populations are important topics to consider when evaluating transmission intensity. Results: The movement of adult Triatoma sordida within the peridomestic area of a rural Brazilian household was evaluated via mark-release-recapture assays. A total of 210 insects had their pronota marked with fluorescent dyes and were released at different distances from the chicken coop (two, five, ten and 20 m), and from the horse corral (27, 32, 35, 46 and 56 m). Recaptures occurred in three consecutive 15-day intervals. Specimens were successfully recaptured at all distances up to 32 m. Bayesian models were used to estimate recapture probability, survival rates (males vs females) and population size. Although recapture probability was inversely proportional to distance for both sexes, females were more affected by increased distance. On the other hand, no significant difference was detected in the survival rates between males and females in a 15-day period. Fisher-Ford and Bayesian models gave more accurate population size estimates than Lincoln method. Conclusions: Triatoma sordida adults were able to cover a distance of 32 m in 45 days. Recapture data modelling reveals that male dispersal was more effective suggesting that T. sordida males are more likely to contribute as potential colonizers of the peridomestic environment. Increasing the distance between the peridomestic structures and the sylvatic environment as much as possible appears to be a simple and feasible recommendation to reduce the contact rate between humans and infected bugs and ultimately Chagas disease transmission. Background: With field releases starting in Brazil, particular interest must be given to understanding how the endosymbiotic bacterium Wolbachia pipientis affects Aedes aegypti mosquitoes with a Brazilian genetic background. Currently, there is limited information on how the bacterium affects phenotypic traits such as larval development rate, metabolic reserves and morphometric parameters in Ae. aegypti. Here, we analyze for the first time, the effect of Wolbachia on these key phenotypes and consider how this might impact the potential of the bacterium as a disease control agent in Brazil. Methods: We examined the influence of the wMel strain of Wolbachia in laboratory Ae. aegypti with a Brazilian genetic background, reared under different larval densities. Pupae formation was counted daily to assess differences in development rates. Levels of metabolic reserves and morphometric parameters were assessed in adults resulting from each larval condition. Results: wMel infection led to more rapid larval development at higher densities for both males and females, with no effect under less crowded conditions in females. Infection also led to reduced body size at both high and low density, but not at intermediate density, although the scale of this difference was maintained regardless of larval density, in comparison to uninfected individuals. Wing shape also varied significantly between infected and uninfected mosquitoes due to larval density. Glycogen levels in uninfected mosquitoes decreased under higher larval density, but were consistently high with Wolbachia infection, regardless of larval density. Conclusions: We demonstrate that the wMel Wolbachia strain can positively influence some important host fitness traits, and that this interaction is directly linked to the conditions in which the host is reared. Combined with previously published data, these results suggest that this Wolbachia strain could be successfully used as part of the Eliminate Dengue Program in Brazil. [ABSTRACT FROM AUTHOR]

Background: Surveillance is a critical component of any dengue prevention and control programme. Herein, we investigate the efficiency of the commercial kit Platelia Dengue NS1 Ag-ELISA to detect dengue virus (DENV) antigens in Aedes aegypti mosquitoes infected under laboratory conditions. Methods: Under insectary conditions, four to five day-old mosquitoes were orally challenged with DENV-2 titer of 3.6 x 105 PFU equivalent/ml, incubated for 14 days and then killed. At ten time-points following mosquito death (0, 6, 12, 24, 72, 96, 120, 144 and 168 h), i.e., during a one-week period, dried mosquitoes were comparatively tested for the detection of the NS1 antigen with other methods of detection, such as qRT-PCR and virus isolation in C6/36 cells. Results: We first observed that the NS1 antigen was more effective in detecting DENV-2 in Ae. aegypti between 12 and 72 h after mosquito death when compared with qRT-PCR. A second round involved comparing the sensitivity of detection of the NS1 antigen and virus isolation in C6/36 cells. The NS1 antigen was also more effective than virus isolation, detecting DENV-2 at all time-points, i.e., up to 168 h after mosquito death. Meanwhile, virus isolation was successful up to 96 h after Ae. aegypti death, but the number of positive samples per time period presented a tendency to decline progressively over time. From the 43 samples positive by the virus isolation technique, 38 (88.4%) were also positive by the NS1 test. Conclusion: Taken together, these results are the first to indicate that the NS1 antigen might be an interesting complementary tool to improve dengue surveillance through DENV detection in dried Ae. aegypti females. Background The survival of adult female Aedes mosquitoes is a critical component of their ability to transmit pathogens such as dengue viruses. One of the principal determinants of Aedes survival is temperature, which has been associated with seasonal changes in Aedes populations and limits their geographical distribution. The effects of temperature and other sources of mortality have been studied in the field, often via mark-release-recapture experiments, and under controlled conditions in the laboratory. Survival results differ and reconciling predictions between the two settings has been hindered by variable measurements from different experimental protocols, lack of precision in measuring survival of free-ranging mosquitoes, and uncertainty about the role of age-dependent mortality in the field. Methods Here we apply generalised additive models to data from 351 published adult Ae. aegypti and Ae. albopictus survival experiments in the laboratory to create survival models for each species across their range of viable temperatures. These models are then adjusted to estimate survival at different temperatures in the field using data from 59 Ae. aegypti and Ae. albopictus field survivorship experiments. The uncertainty at each stage of the modelling process is propagated through to provide confidence intervals around our predictions. Results Our results indicate that adult Ae. albopictus has higher survival than Ae. aegypti in the laboratory and field, however, Ae. aegypti can tolerate a wider range of temperatures. A full breakdown of survival by age and temperature is given for both species. The differences between laboratory and field models also give insight into the relative contributions to mortality from temperature, other environmental factors, and senescence and over what ranges these factors can be important. Conclusions Our results support the importance of producing site-specific mosquito survival estimates. By including fluctuating temperature regimes, our models provide insight into seasonal patterns of Ae. aegypti and Ae. albopictus population dynamics that may be relevant to seasonal changes in dengue virus transmission. Our models can be integrated with Aedes and dengue modelling efforts to guide and evaluate vector control, better map the distribution of disease and produce early warning systems for dengue epidemics.
