The cellulase production by Penicillium decumbens L-06 in solid state fermentation (SSF) was investigated using bagasse as the substrate in this paper. The optimum conditions for cellulase production achieved by single factor testing were: the ratio of bagasse to wheat bran 1∶1 (w/w), the ratio of water to material 3∶1 (v/w), culture temperature 30 °C, initial pH 5.0, ammonium sulphate as nitrogen source with the concentration of 1%, 6 day’s fermentation period. BoxuBehnken factorial design (BBD) and response surface methodology (RSM) were further used to optimize conditions for cellulase (Filter paper activity) production. The maximal cellulase (Filter paper activity) production (3.89 FPu g−1) was obtained under the optimized conditions (ratio of water to material 2.38∶1, initial pH 5.28, cultivation time 150.5 h). Background: The cyclic adenosine-monophosphate (cAMP) pathway is generally recognized as one of the essential pathways for the adipose conversion of rodent preadipocytes in vitro. However, divergent effects of cAMP on adipocyte differentiation have also been reported. Since there is very little data on non-rodent preadipose cells, the aim of the present work was to analyze the effects of one classic activator (dbcAMP) of the cAMP pathway on the proliferation and differentiation of sheep preadipocytes grown., Method: We retrospectively analyzed the regulation of dbcAMP on the proliferation and differentiation of sheep preadipocytes through observation on cell dynamic morphology, drawing on the growth curve, Oil Red O staining and induction of cell differentiation., Results: 1) During first 5 days of treatment, at lower levels of dbcAMP (1 nmol/L to 1 × 10 4  nmol/L), sheep cells were not increased, but at higher levels (1 × 10 5  nmol/l to 1 × 10 6  nmol/l), they were significantly increased (P < 0.05); 2) dbcAMP had the trendency to promote cell differentition, but it was not significant (P > 0.05); 3) treated for 4 days, dbcAMP at the levels of 1 nmol/L, 1 × 10 4  nmol/L and 1 × 10 6  nmol/L increased C20:0 abundance (P < 0.05), but other fatty acids had no significant changes; 4) treated for 4 days, expression of SCD mRNA had no significant change (P > 0.05), but expression of HSL mRNA increased at the level of 1 × 10 6  nmol/L dbcAMP (P < 0.05)., Conclusion: This study demonstrated that the mechanisms by which of the cAMP pathway affects on preadipocytes between sheep and rodent animals was different.

Background: Trichorhinophalangeal syndrome 1 (Trps1) gene is a member of GATA transcription factor family and has an important function in tumorigenesis and progression. However, there are rare studies on its roles in carcinogenesis and prognostic significance in human osteosarcoma., Methods: The expression of Trps1 was detected by immunohistochemistry, and MVD was evaluated to determine the amounts of microvessels by counting CD31-positive endothelial cells., Results: Of the 74 cases that underwent study, Trps1-positive cases were 24. And it was associated with MVD significantly (P = 0.008). The data also exhibited more cases of remote metastasis (P = 0.013) and higher Enneking stage (P = 0.017) in Trps1-positive group compared to Trps1-negative group. Univariate analysis revealed that distant metastasis, MVD and Trps1 expression were associated with a lower 3-year overall survival rate and disease-free survival rate (P = 0.003, and P = 0.012 respectively). Furthermore, Trps1 and distant metastasis retained their significant prognostic effects on patients survival rate by multivariate analysis (P < 0.05)., Conclusions: Trps1 plays a crucial role in osteosarcoma angiogenesis, metastasis and clinical surgical stage. Trps1 can be a novel promising prognostic marker and therapeutic target, and antiangiogenic therapy which targets Trps1 molecule in patients with osteosarcoma may lead to improved prognosis and longer-term survival.

Background: The previous study showed that the cardiac arrhythmias induced by myocardial ischemia and reperfusion were attenuated by the pretreatment of acupuncture; however, the related mechanism is not understood. The present study was therefore designed to determine whether intracellular Ca(2+) ([Ca(2+)]i) and connexin 43 (Cx43) are involved in the mediation of the anti-arrhythmic effect of electro-acupuncture (EA) pretreatment in the rats subjected to simulative global ischemia and reperfusion (SGIR)., Methods: SGIR was made in the isolated heart by a low flow perfusion followed by a flow restoration. Four groups of animals are involved in the present study, including normal control group, SGIR group, EA group and EA plus 18 beta-glycyrrhetinic acid (EAG) group. For EA pretreatment, bilateral Neiguan acupoints (PC6) of the rats were stimulated for 30 min once a day in 3 consecutive days. Cx43 antagonist was given to the rats in EAG group 30 minutes before the EA pretreatment. The resting [Ca(2+)]i concentration, calcium oscillation, the contents of total Cx43 and non-phosphrylated Cx43 and arrhythmia score were compared among different groups., Results: In EA group, the arrhythmic score, the resting [Ca(2+)]i concentration and the number of [Ca(2+)]i oscillations were all significantly less than those in SGIR group (all P < 0.05), and interestingly, after EA pretreatment, the contents of nonphosphated Cx43 in the EA group were significantly lower than that in SGIR group respectively (P < 0.05). However, when the rats were treated with Cx43 antagonist prior to the EA pretreatment, the protection effects induced by EA pretreatment were reversed., Conclusions: The results showed that EA pretreatment could produce anti-arrhythmic effect in the rats subjected to SGIR. The anti-arrhythmic effect of EA pretreatment may be due at least partially to the inhibition of SGIR-induced calcium overload and [Ca(2+)]i oscillations, reduction of non-phosphorylated Cx43 and the enhancement of the corresponding phosphorylated Cx43 in the cardiac cells.
