30. Vitamin D and omega-3 fatty acid supplements in children with autism spectrum disorder: a study protocol for a factorial randomised, double-blind, placebo-controlled trial

Background: There is strong mechanistic evidence to suggest that vitamin D and omega-3 long chain polyunsaturated fatty acids (n-3 LCPUFAs), specifically docosahexaenoic acid (DHA), have the potential to significantly improve the symptoms of autism spectrum disorder (ASD). However, there are no trials that have measured the effect of both vitamin D and n-3 LCPUFA supplementation on autism severity symptoms. The objective of this 2 × 2 factorial trial is to investigate the effect of vitamin D, n-3 LCPUFAs or a combination of both on core symptoms of ASD. Methods/design Children with ASD living in New Zealand (n = 168 children) will be randomised to one of four treatments daily: vitamin D (2000 IU), n-3 LCPUFAs (722 mg DHA), vitamin D (2000 IU) + n-3 LCPUFAs (722 mg DHA) or placebo for 12 months. All researchers, participants and their caregivers will be blinded until the data analysis is completed, and randomisation of the active/placebo capsules and allocation will be fully concealed from all mentioned parties. The primary outcome measures are the change in social-communicative functioning, sensory processing issues and problem behaviours between baseline and 12 months. A secondary outcome measure is the effect on gastrointestinal symptoms. Baseline data will be used to assess and correct basic nutritional deficiencies prior to treatment allocation. For safety measures, serum 25-hydroxyvitamin D 25(OH)D and calcium will be monitored at baseline, 6 and 12 months, and weekly compliance and gastrointestinal symptom diaries will be completed by caregivers throughout the study period. Discussion To our knowledge there are no randomised controlled trials assessing the effects of both vitamin D and DHA supplementation on core symptoms of ASD. If it is shown that either vitamin D, DHA or both are effective, the trial would reveal a non-invasive approach to managing ASD symptoms. 31. Bayesian Models for Pooling Microarray Studies with Multiple Sources of Replications

Background: Biologists often conduct multiple but different cDNA microarray studies that all target the same biological system or pathway. Within each study, replicate slides within repeated identical experiments are often produced. Pooling information across studies can help more accurately identify true target genes. Here, we introduce a method to integrate multiple independent studies efficiently. Results: We introduce a Bayesian hierarchical model to pool cDNA microarray data across multiple independent studies to identify highly expressed genes. Each study has multiple sources of variation, i.e. replicate slides within repeated identical experiments. Our model produces the gene-specific posterior probability of differential expression, which provides a direct method for ranking genes, and provides Bayesian estimates of false discovery rates (FDR). In simulations combining two and five independent studies, with fixed FDR levels, we observed large increases in the number of discovered genes in pooled versus individual analyses. When the number of output genes is fixed (e.g., top 100), the pooled model found appreciably more truly differentially expressed genes than the individual studies. We were also able to identify more differentially expressed genes from pooling two independent studies in Bacillus subtilis than from each individual data set. Finally, we observed that in our simulation studies our Bayesian FDR estimates tracked the true FDRs very well. Conclusion: Our method provides a cohesive framework for combining multiple but not identical microarray studies with several sources of replication, with data produced from the same platform. We assume that each study contains only two conditions: an experimental and a control sample. 32. Inactivation of the Huntington's Disease Gene (Hdh) Impairs Anterior Streak Formation and Early Patterning of the Mouse Embryo

- Abstract
Background: Huntingtin, the HD gene encoded protein mutated by polyglutamine expansion in Huntington's disease, is required in extraembryonic tissues for proper gastrulation, implicating its activities in nutrition or patterning of the developing embryo. To test these possibilities, we have used whole mount in situ hybridization to examine embryonic patterning and morphogenesis in homozygous Hdhex4/5 huntingtin deficient embryos. Results: In the absence of huntingtin, expression of nutritive genes appears normal but E7.0–7.5 embryos exhibit a unique combination of patterning defects. Notable are a shortened primitive streak, absence of a proper node and diminished production of anterior streak derivatives. Reduced Wnt3a, Tbx6 and Dll1 expression signify decreased paraxial mesoderm and reduced Otx2 expression and lack of headfolds denote a failure of head development. In addition, genes initially broadly expressed are not properly restricted to the posterior, as evidenced by the ectopic expression of Nodal, Fgf8 and Gsc in the epiblast and T (Brachyury) and Evx1 in proximal mesoderm derivatives. Despite impaired posterior restriction and anterior streak deficits, overall anterior/posterior polarity is established. A single primitive streak forms and marker expression shows that the anterior epiblast and anterior visceral endoderm (AVE) are specified. Conclusion: Huntingtin is essential in the early patterning of the embryo for formation of the anterior region of the primitive streak, and for down-regulation of a subset of dynamic growth and transcription factor genes. These findings provide fundamental starting points for identifying the novel cellular and molecular activities of huntingtin in the extraembryonic tissues that govern normal anterior streak development. This knowledge may prove to be important for understanding the mechanism by which the dominant polyglutamine expansion in huntingtin determines the loss of neurons in Huntington's disease.

Background: Admission committees are responsible for creating fair, defensible, reliable, and valid processes that assess those attributes considered important for professional success. There is evidence for the continuing use of academic ability as a selection criterion for health professional schools; however, there is little evidence for the reliability and validity of measures currently in place to assess personal characteristics. The Ontario Veterinary College (OVC) initiated a review of its admissions criteria in order to implement an evidence-based method to determine which characteristics veterinary stakeholders consider important to assess for admission., Methods: Eleven characteristics were identified by the OVC Admissions Committee and a survey was sent to all licensed veterinarians in Ontario (n=4,068), OVC students (n=450), and OVC faculty, interns and residents (n=192). A paired comparison method was used to identify the relative rank order of the characteristics, and multivariate analysis of variance with post hoc analyses was used to determine between group differences in the returned survey data., Results: Surveys were returned from 1,312 participants (27.86% response rate; female 59.70%). The relative rank of the characteristics was reasonably consistent among participant groups, with ethical behaviour, sound judgment, communication, and critical and creative thinking being ranked as the top four. However, the importance of certain characteristics like communication and empathy were perceived differently by groups. For instance, females scored communication (F(1, 1289) = 20.24, p < .001, d = .26) and empathy (F(1, 1289) = 55.41, p < .001, d = 0.42) significantly higher than males, while males scored knowledge of profession (F(1, 1289) = 12.81, p < .001, d = 0.20), leadership (F(1, 1289) = 10.28, p = .001, d = 0.18), and sound judgment (F(1, 1289) = 13.56, p < .001, d = 0.21) significantly higher than females., Conclusions: The data from the paired comparison method provide convergent evidence for the characteristics participant groups identify as most important in determining who should be admitted to a veterinary program. The between group analyses provides important information regarding characteristics most important to various subgroups; this has implications for what characteristics are selected for at admission as well as on who is selecting for them.
