Background: Heterogeneous nuclear ribonucleoprotein A2B1 (HNRNPA2B1) is closely related to tumour occurrence and development, oncogene expression, apoptosis inhibition and invasion and metastasis capacities. However, its function in the epithelial-mesenchymal transition (EMT) of pancreatic cancer is not fully understood., Methods: By comparing various wild-type pancreatic cancer cell lines, we determined which have a higher expression level of HNRNPA2B1 accompanied by the higher expression of N-cadherin and vimentin and lower expression of E-cadherin. Therefore, to elucidate the role of HNRNPA2B1 in EMT, we generated models of HNRNPA2B1 knockdown and overexpression in different types of pancreatic cancer cell lines (MIA Paca-2, PANC-1 and Patu-8988) and examined changes in expression of EMT-related factors, including CDH1, CDH2, vimentin and snail., Results: The results show that HNRNPA2B1 promotes EMT development by down-regulating E-cadherin and up-regulating N-cadherin and vimentin, and also stimulates the invasion capacity and inhibits viability in human pancreatic cancer cell lines, the similar results in vivo experiments. Moreover, we found that HNRNPA2B1 likely regulates EMT progression in pancreatic carcinoma via the ERK/snail signalling pathway., Conclusions: The results of this work suggest that HNRNPA2B1 inhibition has potential antitumour effects, which warrants in-depth investigation.

Background: Chronic hepatitis B virus (HBV) infection is a known major etiological factor for hepatocellular carcinoma (HCC) development. Alpha-fetoprotein (AFP) is widely used to detect primary HCC, whereas its sensitivity and specificity are not satisfying. Recently, circulating microRNAs (miRNAs) have been reported to be promising biomarkers for diagnosing and monitoring cancers. This study was conducted to detect the application of serum miR-150 in the diagnosis and prognosis of HBV-related HCC., Methods: The expression of miR-150 was evaluated using a real-time quantitative RT-PCR in 350 serum samples (120 samples from controls, 110 from chronic hepatitis B (CHB) patients and 120 samples from HCC patients., Results: Serum miR-150 levels were significantly reduced in HCC patients, compared with healthy controls (P < 0.0001) and CHB patients (P < 0.0001). Serum miR-150 levels were increased after surgical operation (P < 0.0001) and decreased after tumor recurrence (P < 0.0001). Receiver operating characteristic curve (ROC) analyses suggested that serum miR-150 had significant diagnostic value for HBV-related HCC. It yielded an area under the curve (AUC) of ROC of 0.931 with 82.5% sensitivity and 83.7% specificity in discriminating HCC from healthy controls, and an AUC of ROC of 0.881 with 79.1% sensitivity and 76.5% specificity in discriminating HCC from CHB patients. Moreover, Kaplan-Meier curve analysis revealed that HCC patients with lower serum miR-150 had a significantly shortened overall survival (P < 0.0001). Univariate and Multivariable Cox regression analysis indicated that serum miR-150 level was an independent risk factor for overall survival (P < 0.0001 and P = 0.015, respectively)., Conclusions: Serum miR-150 can serve as a non-invasive biomarker for the diagnosis and prognosis of HCC patients.

Background: The expression of μ-opioid receptor has important role in cognitive dysfunction in Schizophrenia (SZ). The results of studies about the association of polymorphisms of μ-opioid receptor gene (OPRM1) with SZ were inconsistent., Methods: We conducted a case-control study to investigate the genetic association between OPRM1 polymorphisms and SZ among the Han chinese population. 264 SZ patients and 264 age-matched control subjects were recruited. Four SNPs of OPRM1 were successfully genotyped by using PCR-RFLP., Results: Of four polymorphisms, rs1799971 and rs2075572 were shown to associate with SZ. Compared with the A allele of rs1799971 and C allele of rs2075572, the G allele of rs1799971 and rs2075572 was associated with an almost 0.46-fold risk (OR=0.46, 95% CI: 0.357-0.59, P<0.01) and 0.7-fold risk (OR=0.707, 95% CI: 0.534-0.937, P=0.015) of the occurrence of SZ,. When subjects were divided by gender, rs1799971 remained significant difference only in males (OR=0.309, 95% CI: 0.218-0.439 for G allele, P<0.01), and rs2075572 only in females (OR=0.399, 95% CI: 0.246-0.648 for G allele, P<0.01). In secondary analysis with subsets of patients, the G allele of rs1799971 (compared to the A allele) was associated with a decreased risk of all patients and male patients with apathy symptoms (OR=0.086, 95% CI: 0.048-0.151, P=0.01; OR=0.083, 95% CI: 0.045-0.153, P<0.01), and the G allele of rs2075572 (compared to the C allele) was associated with a decreased risk of all patients and female patients with positive family history (OR=0.468, 95% CI: 0.309-0.71, P<0.01; OR=0.34, 95% CI: 0.195-0.593, P<0.01). In addition, haplotype analysis revealed that two SNP haplotypes (A-C-C-G and G-C-C-A) were associated with decreased risks of SZ (P<0.01). The other two (G-C-C-G and G-G-C-G) with increased risks of SZ (P<0.01)., Conclusions: The present study demonstrated for the first time that the OPRM1 polymorphism may be a risk factor for schizophrenia in the Han Chinese. Further studies are needed to give a global view of this polymorphism in pathogenesis of schizophrenia in a large-scale sample, family-based association design or well-defined subgroups of schizophrenia.
