Background: Socio demographic factors besides dietary factors play important role in determining the health status of an individual. Health and nutritional Intervention programs stand a greater chance of success if planned, keeping the socio demographic characteristics of a certain population in focus. The present study was conducted to identify those socio demographic determinants which have a significant association with poor nutritional status in adolescent girls belonging to economically deprived households of Lahore. Methods: A cross-sectional analytical study of 140 adolescent girls living in the peri urban communities of Lahore was conducted. Socioeconomic and demographic data of the participants was recorded through a pretested questionnaire. Body mass index (BMI), a commonly used anthropometric measurement was taken as an indicator of nutritional status. Below normal (<18.5 m2) BMI was considered a benchmark of malnutrition in adolescent girls. Frequencies and percentages for socio demographic variables were calculated and Fisher Exact test was used to find out the association of nutritional status with socio-demographic predictors. Stepwise backward logistic regression analysis was then run to identify the most significant determinants associated with poor nutritional status in the adolescents. P < 0.05 was considered statistically significant. Results: Chronic energy deficiency was highly prevalent among adolescent girls, 58% had BMI < 18.5 m2. Binary regression analysis result showed that the participants who lived in rented houses (AOR = 3.513; 95% CI = 1.366 to 9. 031) who fell ill frequently (AOR = 2.996; 95% CI = 1.198 to 7.491) whose fathers were illiterate (AOR = 2.941; 95% CI = 1.187 to 7.287) were at greater odds of having poor nutritional status. Interestingly, the participants who lived in joint families (AOR = 0.411; 95% CI = 0.145 to 1.168) and were more frequently food insecure (AOR = 0.431; 95% CI = 0.164 to 1.133) had lesser odds of having poor nutritional status than those who lived in nuclear families and were food secure. Conclusion: Frequency of illness, house ownership and fathers' education are the determinants positively associated with poor nutritional status of adolescent girls. Food insecurity and joint family structure were negatively associated with poor nutritional status. The study will help in planning interventions for improving nutritional status of adolescent girls by targeting the significant socio demographic determinants of poor nutritional status among this group. Background: The purple pitcher plant, Sarracenia purpurea L., is a widely distributed species in North America with a history of use as both a marketed pain therapy and a traditional medicine in many aboriginal communities. Among the Cree of Eeyou Istchee in northern Québec, the plant is employed to treat symptoms of diabetes and the leaf extract demonstrates multiple anti-diabetic activities including cytoprotection in an in vitro model of diabetic neuropathy. The current study aimed to further investigate this activity by identifying the plant parts and secondary metabolites that contribute to these cytoprotective effects. Methods: Ethanolic extracts of S. purpurea leaves and roots were separately administered to PC12 cells exposed to glucose toxicity with subsequent assessment by two cell viability assays. Assay-guided fractionation of the active extract and fractions was then conducted to identify active principles. Using high pressure liquid chromatography together with mass spectrometry, the presence of identified actives in both leaf and root extracts were determined. Results: The leaf extract, but not that of the root, prevented glucose-mediated cell loss in a concentration-dependent manner. Several fractions elicited protective effects, indicative of multiple active metabolites, and, following subfractionation of the polar fraction, hyperoside (quercetin-3-O-galactoside) and morroniside were isolated as active constituents. Phytochemical analysis confirmed the presence of hyperoside in the leaf but not root extract and, although morroniside was detected in both organs, its concentration was seven times higher in the leaf. Conclusion: Our results not only support further study into the therapeutic potential and safety of S. purpurea as an alternative and complementary treatment for diabetic complications associated with glucose toxicity but also identify active principles that can be used for purposes of standardization and quality control. Background: Artemisinin is the current drug of choice for treatment of malaria and a number of other diseases. It is obtained from the annual herb, Artemisia annua and some microbial sources by genetic engineering. There is a great concern that the artemisinin production at current rate will not meet the increasing demand by the pharmaceutical industry, so looking for additional sources is imperative. Methods: In current study, artemisinin concentration was analysed and compared in the flowers, leaves, roots and stems of Artemisia annua and 14 other Artemisia species including two varieties each for Artemisia roxburghiana and Artemisia dracunculus using high performance liquid chromatography (HPLC). Results: The highest artemisinin concentration was detected in the leaves (0.44 ± 0.03%) and flowers (0.42 ± 0.03%) of A. annua, followed by the flowers (0.34 ± .02%) of A. bushriences and leaves (0.27 ± 0%) of A. dracunculus var dracunculus. The average concentration of artemisinin varied in the order of flowers > leaves > stems > roots. Conclusion: This study identifies twelve novel plant sources of artemisinin, which may be helpful for pharmaceutical production of artemisinin. Background: An important limiting factor in the development of centrally acting pharmaceuticals is the blood-brain barrier (BBB). Transport of therapeutic peptides through this highly protective physiological barrier remains a challenge for peptide drug delivery into the central nervous system (CNS). Because the most common strategy to treat moderate to severe pain consists of the activation of opioid receptors in the brain, the development of active opioid peptide analogues as potential analgesics requires compounds with a high resistance to enzymatic degradation and an ability to cross the BBB., Results: Herein we report that tetrapeptide analogues of the type H-Dmt1-Xxx2-Yyy3-Gly4-NH2 are transported into the brain after intravenous and subcutaneous administration and are able to activate the μ- and δ opioid receptors more efficiently and over longer periods of time than morphine. Using the hot water tail flick test as the animal model for antinociception, a comparison in potency is presented between a side chain conformationally constrained analogue containing the benzazepine ring (BVD03, Yyy3: Aba), and a "ring opened" analogue (BVD02, Yyy3: Phe). The results show that in addition to the increased lipophilicity through amide bond N-methylation, the conformational constraint introduced at the level of the Phe3 side chain causes a prolonged antinociception. Further replacement of NMe-D-Ala2 by D-Arg2 in the tetrapeptide sequence led to an improved potency as demonstrated by a higher and maintained antinociception for AN81 (Xxx2: D-Arg) vs. BVD03 (Xxx2: NMe-D-Ala). A daily injection of the studied opioid ligands over a time period of 5 days did however result in a substantial decrease in antinociception on the fifth day of the experiment. The compact opioid agonist-NK1 antagonist hybrid SBCHM01 could not circumvent opioid induced tolerance., Conclusions: We demonstrated that the introduction of a conformational constraint has an important impact on opioid receptor activation and subsequent antinociception in vivo. Further amino acid substitution allowed to identify AN81 as an opioid ligand able to access the CNS and induce antinociception at very low doses (0.1 mg/kg) over a time period up to 7 hours. However, tolerance became apparent after repetitive i.v. administration of the investigated tetrapeptides. This side effect was also observed with the dual opioid agonist-NK1 receptor antagonist SBCHM01.
