Abstract

Research was conducted to examine the metal-catalyzed oxidation mechanism that causes the in vitro instability of the phenylalanine-sensitive 3-deoxy-D-arabino-heptulosonate-7-phosphate synthase (DAHPS) from Escherichia coli. A disulfide linkage between the two cysteines, namely, Cys61 and Cys328, was observed from peptide mapping of the Cu2+-inactivated enzyme. A proposed inactivation mechanism of DAHPS features a metal redox cycle that needs a sequential oxidation of its two active-site cysteines.
