It is unsatisfactory that regarding the problem of entangled macromolecules driven out of equilibrium, experimentally based understanding is usually inferred from the ensemble average of polydisperse samples. Here, confronting with single-molecule imaging this common but poorly understood situation, over a wide range of shear rate we use single-molecule fluorescence imaging to track alignment and stretching of entangled aqueous filamentous actin filaments in a homebuilt rheo-microscope. With increasing shear rate, tube "softening" is followed by "hardening." Physically, this means that dynamical localization first weakens from molecular alignment, then strengthens from filament stretching, even for semiflexible biopolymers shorter than their persistence length.

The spatially resolved diffusive dynamic cross correlations of a pair of colloids in dense quasi-two-dimensional monolayers of identical particles are studied experimentally and theoretically at early times where motion is Fickian. In very dense systems where strong oscillatory equilibrium packing correlations are present, we find an exponential decay of the dynamic cross correlations on small and intermediate length scales. At large separations where structure becomes random, an apparent power law decay with an exponent of approximately -2.2 is observed. For a moderately dense suspension where local structural correlations are essentially absent, this same apparent power law decay is observed over all probed interparticle separations. A microscopic nonhydrodynamic theory is constructed for the dynamic cross correlations which is based on interparticle frictional effects and effective structural forces. Hydrodynamics enters only via setting the very short-time single-particle self-diffusion constant. No-adjustable-parameter quantitative predictions of the theory for the dynamic cross correlations are in good agreement with experiment over all length scales. The origin of the long-range apparent power law is the influence of the constraint of fixed interparticle separation on the amplitude of the mean square force exerted on the two tagged particles by the surrounding fluid. The theory is extended to study high-packing-fraction 3D hard sphere fluids. The same pattern of an oscillatory exponential form of the dynamic cross correlation function is predicted in the structural regime, but the long-range tail decays faster than in monolayers with an exponent of -3.

High-speed movies are taken when bubbles grow at gold surfaces heated spotwise with a near-infrared laser beam heating water below the boiling point (60-70 °C) with heating powers spanning the range from very low to so high that water fails to rewet the surface after bubbles detach. Roughly half the bubbles are conventional: They grow symmetrically through evaporation until buoyancy lifts them away. Others have unorthodox shapes and appear to contribute disproportionately to heat transfer efficiency: mushroom cloud shapes, violently explosive bubbles, and cavitation events, probably stimulated by a combination of superheating, convection, turbulence, and surface dewetting during the initial bubble growth. Moreover, bubbles often follow one another in complex sequences, often beginning with an unorthodox bubble that stirs the water, followed by several conventional bubbles. This large dataset is analyzed and discussed with emphasis on how explosive phenomena such as cavitation induce discrepancies from classical expectations about boiling.

We show, using a large new data set, that the temporally resolved speed of active cargo transport in living cells follows a scaling law over several decades of time and length. The statistical regularities display a time-averaged shape that we interpret to reflect stress buildup, followed by rapid release. The scaling power law agrees quantitatively with those reported in inanimate systems (jammed colloids and granular media, and magnetic Barkhausen noise), suggesting a common origin in pushing through a crowded environment in a weak force regime. The implied regulation of the speed of active cellular transport due to environmental obstruction results in bursts of speed and acceleration. These findings extend the classical notion of molecular crowding.

Using single-molecule fluorescence imaging, we track Brownian motion perpendicular to the contour of tightly entangled F-actin filaments and extract the confining potential. The chain localization presents a small-displacement Hookean regime followed by a large amplitude regime where the effective restoring force is independent of displacement. The implied heterogeneity characterized by a distribution of tube width is modeled.

We investigate, from single-particle tracking of jumps, the cluster configurations that allow hopping over a geometric activation barrier in surface diffusion. Spherical colloidal particles, their dimers, and their isomeric planar trimers are compared on hexagonal surface lattices commensurate with the elemental size of one particle. The experiments reveal that translational and rotational mobility depend on the shape of these clusters, not only on their mass, because the jump process favors a restricted family of cluster configurations. The resulting strong decoupling between rotation and translation demonstrates the limitations of a naïve Arrhenius picture, even for a simple gravitational potential.

Computer simulations of a polymer chain of length N strongly adsorbed at the solid-liquid interface in the presence of explicit solvent are used to delineate the factors affecting the N dependence of the polymer lateral diffusion coefficient, D(||). We find that surface roughness has a large influence, and D(||) scales as D(||) approximately N(-x), with x approximately 3/4 and x approximately 1 for ideal smooth and corrugated surfaces, respectively. The first result is consistent with the hydrodynamics of a "particle" of radius of gyration R(G) approximately N(nu) (nu=0.75) translating parallel to a planar interface, while the second implies that the friction of the adsorbed chains dominates. These results are discussed in the context of recent measurements.

Synchrotron x-ray reflectivity measurements of the interface between water and methyl-terminated octadecylsilane monolayers with stable contact angle >100 degrees conclusively show a depletion layer, whether or not the water is degassed. The thickness is of order one water molecule: 2-4 Angstrom with electron density <40% that of bulk water. Considerations of coherent and incoherent averaging of lateral inhomogeneities show that the data cannot be explained by "nanobubbles." When the contact angle is lower, unstable in time, or when monolayers fail to be sufficiently smooth over the footprint of the x-ray beam, there is no recognizable depletion.

The translational diffusion of a fluorescent dye embedded at a dilute concentration in a confined fluid was compared at rest and during shear. The fluid, octamethylcyclotetrasiloxane (OMCTS), was confined between step-free muscovite mica to thickness 3-4 layers. Fluorescence correlation spectroscopy showed that the time scales of intensity-intensity autocorrelation functions were essentially the same during shear and at rest, except they were faster during shear by a factor of 2 to 5. This dynamical probe of how liquids order in molecularly thin films fails to support the hypothesis that shear produced a melting transition.

Using the method of Frantz and Salmeron to cleave mica [Tribol. Lett. 5, 151 (1998)]] we investigate alkane fluids in a surface forces apparatus and confirm several predictions of molecular dynamics (MD) simulation. An oscillatory force-distance profile is observed for the methyl-branched alkane, squalane. Boundary slip is inferred from the frictional sliding of molecularly thin fluids and also from the hydrodynamic flow of thicker films. These findings resolve the paradox that prior experiments disagreed with these aspects of MD predictions, and demonstrate that exceptionally low energy dissipation is possible when fluids move past solid surfaces that are sufficiently smooth.

We report the results of in situ AFM measurements examining the phase transition of bilayers formed from the zwitterionic phospholipid, DMPC, 1,2-dimyristoyl-sn-glycero-3-phosphocholine, supported on mica. The images show that the fluid to gel phase transition process features substantial tearing of the bilayer due to the density change between the two phases. The gel to fluid transition is strongly affected by the resultant stress introduced into the gel phase, which changes the degree of cooperativity, the shape of developing fluid phase regions, and the course of the transition.

We study, using fluorescence correlation spectroscopy, translational diffusion in molecularly thin liquids confined within a surface forces apparatus. The diffusion coefficient decreases exponentially from the edges towards the center of the Hertzian contact and further suggests the presence of a small number of distinct diffusion processes. This holds alike a crystallizable fluid (OMCTS) and a glass-former (1,2-propane diol), both of which displayed static friction. We conclude that friction, the average of an ensemble of molecules, masked massively heterogeneous molecular mobility.

A controversial point in fluid dynamics is to distinguish the relative importance of surface roughness and fluid-surface intermolecular interactions in determining the boundary condition. Here hydrodynamic forces were compared for flow of Newtonian fluids past surfaces of variable roughness but similar, poorly wetted, surface chemistry. The critical shear stress and shear rate to observe deviations from predictions using the no-slip boundary condition increased nearly exponentially with increasing roughness and diverged at approximately 6 nm rms roughness. We conclude that local intermolecular interactions dominated when the surface was very smooth, but roughness dominated otherwise. This quantifies the limits of both ideas.

The effective shear viscosity and frequency-dependent dynamic oscillatory shear spectra of water containing monovalent or divalent ions (ionic strength 25 mM), confined between mica crystals at 1-2 water molecules thickness, oscillated with twist angle with the period expected for the pseudohexagonal surface lattice. The effective viscosity varied by orders of magnitude as the twist angle was changed. Confinement appeared to imprint lateral spatial correlation on the ultrathin liquid, the more so the better the confining lattices were aligned, but the oft-proposed "ice structure" was not observed dynamically.

Newtonian fluids were placed between molecularly smooth surfaces whose spacing was vibrated at spacings where the fluid responded as a continuum. Hydrodynamic forces agreed with predictions from the no-slip boundary condition only provided that flow rate (peak velocity normalized by spacing) was low, but implied partial slip when it exceeded a critical level, different in different systems, correlated with contact angle (surface wettability). With increasing flow rate and partially wetted surfaces, hydrodynamic forces became up to 2-4 orders of magnitude less than expected by assuming the no-slip boundary condition that is commonly stated in textbooks.
