Coagulase-negative staphylococci (CNS) are the most common pathogens associated with intramammary infections (IMI) in dairy cows. We hypothesized that postmilking teat disinfection would reduce microbial colonization of the teat canal and thus reduce the prevalence of IMI caused by certain CNS species. The efficacy of iodine postmilking teat dip was tested against CNS colonization of the teat canal, and incidence of IMI was measured. Using an udder-half model, 43 Holstein cows at the Washington State University Dairy were enrolled in the trial; postmilking teat dip was applied to one udder-half, treatment (TX), and the remaining half was an undipped control (CX). Teat canal swabbing and mammary quarter milk samples were taken in duplicate once a week for 16 wk for microbial culture. Isolates from agar cultures were presumptively identified as CNS and then speciated using PCR-RFLP and agarose gel electrophoresis. Colonization of the teat canal and IMI by CNS were assessed. Thirty CNS IMI were diagnosed and the number of new IMI in CX quarters (21) was significantly greater than that in TX mammary quarters (9). The majority of CNS IMI were caused by Staphylococcus chromogenes (30%) and Staphylococcus xylosus (40%), and the latter were appreciably reduced by teat dip. Except for S. xylosus, an association was observed between teat canal colonization and IMI by all CNS species in this study, in which the majority of IMI were preceded by teat canal colonization. The total number of CNS IMI was greater for CX group cows compared with TX group cows. Low sensitivity of a single bulk tank milk culture is a major limitation for detection of mycoplasma organisms. We hypothesized that sedimentation of Mycoplasma spp. in a milk sample by centrifugation followed by resuspension in a small volume of fluid before agar plating would increase the ability to detect Mycoplasma spp. compared with direct conventional culture. The experiment was conducted to determine recovery of Mycoplasma spp. from milk as affected by 1) treatment (centrifugation vs. conventional method); 2) 2 species (Mycoplasma bovis and Mycoplasma californicum and 4 strains for each species); and 3) 4 different concentrations of Mycoplasma spp. (1,000, 100, 10, and 1 cfu/mL). A 5-mL portion of mycoplasma suspension from each strain was inoculated into 45 mL of fresh bulk tank milk to achieve concentrations of 1,000, 100, 10, and 1 cfu/mL. Treatment samples were vigorously mixed and centrifuged at 5,000 x g for 30 min. Control samples were vigorously mixed. All samples were plated on modified Hayflick agar. Plates were incubated at 37 degrees C and 5% CO(2) for 5 d. Mean (+/-SE) log(10) mycoplasma counts (cfu/mL) in the treatment groups (1.91 +/- 0.15) were higher than those in the control groups (1.70 +/- 0.16). Recovery of at least 1 mycoplasma colony on agar culture was 100% in both treatment and control groups at high, medium, and low concentrations. At the lowest concentration, recovery of at least 1 mycoplasma colony on agar culture in treatment and control groups was 75% (n = 12/16) and 18.75% (n = 3/16), respectively. Centrifugation of milk followed by suspension in a smaller volume of saline before conventional culture increased the ability to detect mycoplasma microorganisms in the milk sample compared with controls. Recovery by centrifugation appeared best at the lowest concentration where detection of a positive sample was 4 times more likely than when conventional methods were used.

Fifty-nine commercial dairy farms were sampled 7 times over 15 to 21 mo to determine the role of animal movement, including off-farm rearing of heifers, in the interherd transmission of multidrug-resistant (MDR) Salmonella spp. Farm management data were collected by on-site inspections and questionnaires on herd management practices before and after the study. Forty-four percent (26/59) of herds did not acquire any new MDR Salmonella strains. The number of newly introduced MDR Salmonella strains acquired by the remaining 56% (33/59) of herds ranged from 1 to 8. Logistic regression models indicated that off-farm heifer raising, including contract heifer raising where heifers commingle with cattle from other farms [commingled heifers, odds ratio (OR) = 8.9, 95% confidence interval (CI): 2.4, 32.80], and herd size per 100-animal increment (herd size, OR = 1.04, 95% CI, 1.01, 1.05) were significantly associated with the introduction of new MDR Salmonella strains. The negative binomial regression similarly revealed that commingled heifers [relative risk (RR) = 2.3, 95% CI: 1.1, 4.7], herd size per 100 animals (RR = 1.02, 95% CI, 1.01, 1.03), and a history of clinical salmonellosis diagnosed before the study (RR = 2.5, 95% CI, 1.3, 5.0) were significantly associated with the number of new MDR Salmonella strains that were introduced. Factors not associated with the introduction of new MDR Salmonella strains were housing of heifers and cows in the same close-up pen, a common hospital-maternity pen, and the number of purchased cattle. This study highlights the role of animal movement in the interherd transmission of MDR Salmonella spp.

A longitudinal observational study of 59 dairy herds was conducted in Washington State to estimate the rate of introduction of new multidrug-resistant (MDR) Salmonella enterica strains onto commercial dairy herds. Samples were collected on these herds over 7 visits separated by intervals of 2 to 4 mo over a period of 15 to 21 mo. Samples were cultured for Salmonella spp. and serogroup, serovar, and antimicrobial susceptibility patterns were identified for MDR Salmonella isolates. Fingerprinting generated by pulsed-field gel electrophoresis (PFGE) using XbaI restriction enzyme digestion generated genotyping profiles for all MDR isolates identified in the study. The rate of new MDR Salmonella strain introduction was 0.9 per herd-year (95% confidence interval: 0.6-1.4). The rates for the most commonly introduced MDR Salmonella serovars were 0.4/herd-year for Typhimurium, 1.2/herd-year for Newport, and 0.1/herd-year for Dublin. Thirty-three of 59 herds (56%) had at least one new MDR Salmonella introduction during the study period. The number of new MDR Salmonella strains acquired by dairy herds ranged from zero to 8. Thirteen of the 59 herds had a history of clinical salmonellosis. Among these 13 herds, 6 herds acquired new MDR Salmonella strains, although these strains were different than historical clinical strains. These data indicate that acquisition of new MDR Salmonella strains by dairy herds was a common event in participating herds, although the number of strains introduced varied greatly among herds.

Several recent studies have investigated the effect of shortened dry periods on milk production in the subsequent lactation. What is lacking from these studies is an understanding of the effect that a shortened dry period has on udder health. Four herds, 156 cows, were studied to determine if a shortened dry period (30 d) had a negative effect on mammary gland health during the subsequent lactation as opposed to cows assigned to a long, 45 or 60 d, dry period. Cows in 2 herds were assigned to either 30- or 60-d dry periods (group I), whereas cows in the other 2 herds were assigned to either 30- or 45-d dry periods (group II). Intramammary instillation of commercial preparations of cephapirin benzathine, 300 mg (dry cow formulation), was given to cows assigned a 45- or 60-d dry period length protocol, and 200 mg (lactating cow formulation) was administered to cows assigned a 30-d dry period. Differences in response variables to dry period length were compared within group. Cure rates for 60- vs. 30-d dry period cows were 72% (28/39) vs. 81% (30/37) and 74% (25/34) and 73% (27/37) for 45- vs. 30-d dry periods. Differences were not statistically significant for either comparison group. The majority of intramammary infections were caused by the minor pathogens, coagulase-negative staphylococci (n = 102) or Corynebacterium bovis (n = 11). Only 11 cows had intramammary infections by major pathogens. The herd average percentage of new intramammary infections ranged from 6 to 9% and did not differ among herds between treatment groups. Linear somatic cell counts were not significantly affected by dry period length during the first 6 to 7 mo of the subsequent lactation. Milk production did differ between groups. Mature equivalent milk production was greater in group I cows given a 60-d dry period (11,942 +/- 2,059 kg) as opposed to those given a 30-d dry period (10,749 +/- 2,321 kg). Cows given a 45-d dry period did not produce more milk than cows with a 30-d dry period in group II. Although shortening the dry period to 30 d did not have untoward effects on mammary gland health as measured by intramammary infections or milk somatic cell counts, production may be adversely affected when dry periods are shortened to 30 d.

Preparturient heifers (n = 561) from 9 herds in 6 US states and 1 Canadian province were enrolled in a study to test the hypothesis that prepartum intramammary therapy would cure existing intramammary infections (IMI) and lead to increased milk production, reduced linear somatic cell count (LSCC), and improved reproductive performance. Mammary secretions were collected 10 to 21 d before expected calving from each quarter. Heifers were then assigned by identification number to receive intramammary therapy consisting of infusion of one tube per mammary quarter of a lactating cow commercial antibiotic preparation containing cephapirin or to a nontreated control group. Overall, 34.1% of mammary quarters were infected with a mastitis pathogen before parturition and 63.4% of heifers had at least one mammary quarter infected. The coagulase-negative staphylococci (CNS) caused the majority (74.8%) of prepartum IMI. Coagulase-positive staphylococci, environmental streptococci, and coliforms accounted for 24.5% of prepartum infections. Treatment had a significant effect on the cure rate of infected mammary quarters. Mammary quarters that were infected prepartum and treated with antibiotics had a 59.5% efficacy of cure rate and the percentage reduction in heifers with IMI was 51.9. Control quarters had a spontaneous cure rate of 31.7%. Treatment did not significantly affect milk production or LSCC in the first 200 d of lactation; however, there was a significant treatment by herd interaction for milk production. Quarters cured of either CNS or major pathogens had a lower LSCC in the first 200 d of lactation. No significant effect on services per conception or days open between treatment and control groups was observed. This trial demonstrated that prepartum intramammary antibiotic therapy did reduce the number of heifer IMI postpartum. Milk production, LSCC, and reproductive performance during the first 200 d of the first lactation were not significantly affected by treatment. Given these results, use of prepartum intramammary antibiotic therapy in heifers as a universal strategy to increase milk production in first-lactation dairy cows may not be warranted.

This study was executed to determine the effects of storage and thawing on the viability of Mycoplasma spp. in milk from cows with intramammary infections. The trial was designed using a control sample and seven handling regimens subjected to two methods of thawing. There was a significant treatment effect on the recovery of colony-forming units in milk samples when comparing the control sample with handling regimens 1 through 7. There was a continuous decline in log (10) mean number of cfu/mL recovered. Mean concentrations were 6.29, 4.64, 3.69, 3.01, 1.86, 4.41, 4.13, and 3.18 for control and handling regimens 1 to 7, respectively. To determine the best thawing method, handling regimen 1 through 7 samples were thawed using two methods. On average, more mycoplasma were recovered from milk samples thawed at ambient temperature (4.04 cfu/mL) than milk samples thawed in a 37 degrees C water bath (3.76 cfu/mL). A final comparison was made between individual treatments. With the exception of the handling regimen 5 to 6 pair-wise comparison, all pair-wise comparisons between handling regimens were significantly different. The results of this study indicate that storage and thawing of milk samples is harmful to mycoplasma organisms. Fresh samples should be used to improve detection of Mycoplasma spp. from milk of infected cattle. If frozen samples are used, then length of storage time should be minimized, and thawing milk at ambient temperature will improve recovery of mycoplasma as opposed to thawing in a 37 degrees C water bath.

The microbial quality of livestock drinking water was evaluated in 473 cattle water troughs located at 99 different cattle operations. The mean log10-transformed coliform and Escherichia coli concentrations per milliliter of trough water were 1.76 +/- 1.25 (SD) and 0.98 +/- 1.06 (SD), respectively. The degree of E. coli contamination was positively associated with the proximity of the water trough to the feedbunk, protection of the trough from direct sunlight, lower concentrations of protozoa in the water, and warmer weather. Salmonella sp. were isolated from 2/235 (0.8%) troughs and shigatoxigenic-E. coli O157 was recovered from 6/473 (1.3%) troughs. Four experimental microcosms simulating cattle water troughs were used to further evaluate the effects of protozoal populations on the survival of E. coli O157 in cattle water troughs. Escherichia coli O157 of bovine fecal origin proliferated in all microcosms. Reduction of protozoal populations by treatment with cycloheximide was associated with increased persistence of E. coli O157 concentrations in the microcosms. Water troughs are a major source of exposure of cattle to enteric bacteria, including a number of foodborne pathogens, and this degree of bacterial contamination appeared to be associated with potentially controllable factors.

An incomplete block split-plot design with repeated measures was used to test the effects of eight combinations of premilking and postmilking teat disinfectants on teat skin and teat end condition over a 6-wk period. These combinations included iodine, sodium chlorite, lactic acid, and orthophosphoric acid. Visual assessments of teat skin and teat ends were made on the teats of 101 cows. Teat skin thickness, teat end thickness, and transepidermal water loss were assessed for the teats of 50 cows. The interaction of predip and postdip had no effect on teat condition. Postdip had a significant effect on transepidermal water loss, visual teat skin condition, and visual teat end condition. The 1% iodine postdip was associated with the poorest teat skin condition.

A split-plot design with repeated measures was used to test the relationship between visual teat skin condition score, the degree of transepidermal water loss from the skin, and the colonization by Staphylococcus aureus on experimentally chapped and inoculated teats of 20 lactating Holstein cows. Visual teat skin chapping score and the number of S. aureus colonies obtained from a teat skin swab were correlated (r = 0.53). Transepidermal water loss and S. aureus count were not correlated (r = 0.02). Results indicated that visual teat skin evaluation is superior to measurements of transepidermal water loss in the prediction of the susceptibility of teat skin to colonization by S. aureus.

Sixty-three of 209 (30.1%) samples of cattle feed that were collected from multiple commercial sources and from farms were found to contain Escherichia coli. However, none of the feed samples examined were culture-positive for E. coli O157. Replication of fecal E. coli, including E. coli O157, was demonstrated in a variety of feeds at temperatures that were similar to those found on farms in summer months. Fresh mixed rations containing corn silage were sampled from 16 dairies. Rations from 12 of these dairies were found to contain E. coli, and the rations from 5 dairies had concentrations of E. coli that were greater than 1000 cfu/g. The ability of experimental mixed rations to support the replication of E. coli was correlated with the concentration of organic acids in the corn silage that was used in the ration. Widespread contamination of cattle feeds with E. coli and the ability of E. coli to replicate in feeds suggest that feeds are a potentially important factor in the ecology of organisms that can be transmitted from feces to mouth, such as E. coli O157.

The study objective was to identify probable sources and modes of transmission of 91 Staphylococcus aureus isolates obtained from the colostrum of 76 heifers at parturition. Sources cultured were milk (including colostrum), heifer body sites (teats, muzzle, rectum, vagina, and lacteal secretions), and environmental sites (bedding, insects, housing, water, feedstuffs, humans, nonbovine animals, air, and equipment). Staphylococcus aureus isolates were characterized by 63 phenotypic traits. A similarity coefficient was calculated by herd to identify the S. aureus that most closely resembled the S. aureus obtained from heifer colostrum. Staphylococcus aureus from a heifer's colostrum was compared with all preexisting S. aureus isolates from that heifer's herd. Isolates that were > or = 90% similar were considered to be identical. Because 30 (of the 91) S. aureus isolates from heifer colostrum were collected prior to environmental sampling, only 61 S. aureus isolates from heifer colostrum were available for comparison among all three sources. Possible sources of S. aureus from heifer colostrum at parturition were milk (70%, 43 of 61 isolates), heifer body sites (39%, 24 of 61), environmental sites (28%, 17 of 61), or no identified source (16%, 10 of 61). Three heifers with intramammary infection (IMI) from S. aureus at parturition had the same S. aureus on their teats prior to parturition. Milk was the only source identified for 41% (25 of 61) of isolates from heifer colostrum. Isolates from heifer body sites were the only source identified for 5% (3 of 61) of heifer colostrum isolates. Staphylococcus aureus from the environment was never the sole possible source for S. aureus from heifer colostrum. Data suggest that the major sources of S. aureus IMI in heifers at parturition are milk and heifer body sites. Contact among heifers may be an important mode of transmission of S. aureus leading to IMI in heifers at parturition.

Telephone interviews were conducted to evaluate the association of teat chapping with combinations of premilking and postmilking teat disinfectants (predip and postdip), other management practices, and climatic conditions. Holstein dairies (n = 100) in the DHIA SCC program were randomly selected from three northwest regions. During nonwinter seasons, herd managers using an iodophor dip without skin conditioner both premilking and postmilking were 9.7 times more likely to report chapping than were those using iodophor with conditioner. The herd managers least likely to report chapping problems were those using conditioner in both the predip and postdip. Use of conditioner in postdip only was associated with intermediate frequency of reported chapping. No conditioner in either predip or postdip was associated with the greatest frequency of reported chapping. Herd managers using a different germicide in the predip and postdip reported more chapping than herd managers using the same germicide. The frequency of chapping was highest during winter in all regions, and the percentage of herd managers reporting chapping within a region was associated with the temperature extremes of the region. For seasons other than winter, characteristics of predip and postdip combinations and parlor exit or barn design were the management factors that best described an association with reported chapping.

The purposes of this study were to identify sources of Staphylococcus aureus on dairies and to determine whether S. aureus colonization of heifer body sites increases the risk of S. aureus IMI at parturition. In herds with high (> 10%) or low (< 3%) prevalence of S. aureus IMI, S. aureus was isolated from heifer teat skin, heifer external orifices, housing, feedstuffs, humans, nonbovine animals, air, and equipment. Additionally, in herds with high prevalence, S. aureus was isolated from bedding, insects, and water. The predominant sources of S. aureus for both groups were other IMI and heifer body sites. Heifers with prepartum lacteal secretions with S. aureus were at greater risk of S. aureus IMI at parturition than were prepartum heifers with lacteal secretions that were negative for S. aureus. Heifers with teat skin colonized by S. aureus were 3.34 times more likely to have S. aureus IMI at parturition than were noncolonized heifers. Overall, 35% of 700 heifers were colonized with S. aureus on a body site at least once. Although colonizations of most body sites appeared to be transient, a few heifers were colonized on the same site for 1 yr. Persistently colonized heifers may represent the primary reservoirs of S. aureus for other heifers.

Hydrometer measurement in globulin and IgG1 concentration measured by the radial immunodiffusion technique were compared for 915 samples of first milking colostrum from Holstein cows. Least squares analysis of the relationship between hydrometer measurement and IgG1 concentration was improved by log transformation of IgG1 concentration and resulted in a significant linear relationship between hydrometer measurement and log10 IgG1 concentration; r2 = .469. At 50 mg of globulin/ml of colostrum, the recommended hydrometer cutoff point for colostrum selection, the sensitivity of the hydrometer as a test of IgG1 concentration in Holstein colostrum was 26%, and the negative predictive value was 67%. The negative predictive value and sensitivity of the hydrometer as a test of IgG1 in Holstein colostrum was improved, and the cost of misclassification of colostrum was minimized, when the cutoff point for colostrum selection was increased above the recommended 50 mg/ml.

Objectives were to determine the prevalence of coagulase-positive staphylococcal IMI in primiparous cows at first parturition, to contrast the differences in coagulase-positive staphylococcal IMI in primiparous cows at parturition in herds with high and low prevalences of coagulase-positive staphylococcal IMI in the lactating herd, and to determine the percentage of primiparous cows having persistent coagulase-positive staphylococcal IMI. Milk samples were collected aseptically from cows at the start and end of the study, at dry-off, and at parturition. Herds (n = 18) were split evenly into two categories: high (> 10%) or low (< 5%) prevalence of coagulase-positive staphylococcal IMI. At the start, the mean prevalence of coagulase-positive staphylococcal IMI in high prevalence herds was 30%, ranging from 13 to 65%, and in low prevalence herds was 2%, ranging from 0 to 5%. Overall the prevalence of coagulase-positive staphylococcal IMI in primiparous cows at parturition was 8.1% (67 of 828), ranging from 0 to 27%. Although primiparous cows from high prevalence herds had a higher prevalence of coagulase-positive staphylococcal IMI (9.2%; 40 of 436) at parturition than did primiparous cows from low herds (6.9%; 27 of 392), the difference was not significant. Of primiparous cows with coagulase-positive staphylococcal IMI at parturition, 43% had coagulase-positive staphylococcal IMI at least 2 mo after parturition. Primiparous cows with coagulase-positive staphylococcal IMI at parturition may represent significant reservoirs of infection to uninfected herdmates.

Bacteriological culture results were compared between 336 pairs of quarter milk samples collected premilking and postmilking. Using a positive result on either premilking or postmilking samples as the definitive diagnosis, premilking sampling sensitivity was 91% for Staphylococcus aureus, 91% for coagulase-negative staphylococci, and 97% for Streptococcus other than agalactiae. Postmilking sampling sensitivities were 81, 45, and 58%, respectively, for the same pathogens. Requiring both premilking and postmilking samples for the definitive diagnosis, specificities were 92, 86, and 95% for premilking sampling alone and 96, 98, and 99% for postmilking sampling alone. Such differences in specificity would result in major differences in predictive value of a positive culture for herds with a low prevalence. Multiple isolates were significantly more common from premilking samples.

Immunoglobulin G1 concentration was measured in 919 first milking colostrums from Holstein cows during a 4-yr period on a commercial dairy farm. Sources of variation analyzed for effect on colostral IgG1 concentration were season of calving, lactation number, dry period length, intercalving interval, complete lactation milk and fat production, weight of first milking colostrum, and time from calving to first milking. Weight of first milking colostrum was the variable most highly correlated (negatively) with colostral IgG1 concentration (r = -.29). Weight of first milking colostrum and lactation number of the cow were the most significant discriminators between colostrum of low and high IgG1 concentration. The implications of these results for colostrum feeding management are discussed.

Effects of differences in herd mastitis control management in maintaining low herd average SCC, was studied. Washington State DHI herds with Holstein cattle and enrolled in the SCC program (n = 309) were ranked by percentage of cows in their herd with SCC less less than or equal to 283,000 cells/ml. "Low herds" (n = 28) were among the 56 herds with the highest percentage of cows with SCC less than or equal to 283,000 cells/ml and "high herds" (n = 31) were among the 75 with the lowest percentage of cows with SCC less than or equal to 283,000 cells/ml. Herds were visited annually for 2 yr by a technician who collected samples and recorded data. Geometric mean bulk tank SCC during the year between herd visits was 175,000 and 460,000 cells/ml for low and high herds. Milking time hygiene practices, teat dipping, and dry cow therapy were practiced with equal frequency on low and high herds. Differences in function and maintenance of milking equipment did not discriminate between herd groups. Differences in mastitis control management of low versus high SCC herds were that managers of excellent control herds more frequently had highest producers milked first and clinical cows milked last; had automatic milking unit detachers; kept moisture content of cow bedding lower; and had workers disinfect teat ends prior to intramammary antibiotic treatment. Managers of low herds were more likely to use computers and attend dairy informational meetings. Results suggest subtle differences in mastitis control strategies differentiate the low and high SCC herd groups.

Blood serum immunoglobulin concentrations of calves during their 2nd wk of life had a .97 correlation with 1st wk concentrations. Correlations between 1st wk concentrations and those in the 3rd, 4th, 5th, and 6th wk of life were .91, .90, .81, and .69. Ten herds were divided into those with mortality rates above and below the median. The major difference in distributions of immunoglobulin concentration between the two groups was percent of calves with concentrations less than 2.5 mg/ml during the 1st wk of life. Calves were sampled monthly for a full year in 19 herds, and all but one herd had one or more calves with immunoglobulin concentrations under 5.0 mg/ml. Overall, calves having immunoglobulin concentrations under 5.0 mg/ml were exposed to excess mortality risk; however, most of the excess risk was concentrated in three herds with high mortality rates (16 to 20%). Evaluating the efficiency of passive immune transfer through the use of management data was unreliable in comparison with assaying serum samples for immunoglobulin concentration.

Selenium injections and oral vitamin E supplementation prepartum were related to: postpartum uterine involution (decrease in uterine size per unit time) and days to minimum uterine size in a 2 X 2 factorial design. Complete data were analyzed from 64 cows. Groups were selenium plus vitamin E, vitamin E, selenium, and control. Factors significantly affecting uterine size between 14 and 50 d postpartum were cow weight, days postpartum-linear, days postpartum-quadratic, day X metritis, and day X metritis X selenium treatment. Days to minimum uterine size were significantly less in cows with metritis and selenium treated when compared with cows with metritis and not selenium treated (32.9 vs. 35.8).

Incidence of clinical mastitis and duration of clinical symptoms for complete lactations were evaluated for 80 cows randomly assigned to one of four groups: vitamin E supplemented- and selenium injected, selenium injected, vitamin E supplemented, and controls. Vitamin E supplementation and selenium injection were during the dry period. Log-linear analysis of incidence data revealed a significant 37% reduction of clinical mastitis by vitamin E. Incidence was not affected by selenium alone, nor was there any evidence for interaction of vitamin E with selenium on incidence. However, duration of clinical symptoms (calendar months clinical/quarter lactating) was reduced by 46% for the selenium group, 44% for the vitamin E group, and 62% for the vitamin E-selenium group as compared to controls. We conclude that dairy cow diets deficient of vitamin E may elevate incidence of clinical mastitis. Selenium deficiency may result in greater duration of clinical symptoms, and selenium may interact with vitamin E. Coliform bacteria and species of streptococcus other than Streptococcus agalactiae were isolated from 70% of the clinical cases.

Experiment 1 was conducted to determine NAGase activity in skim, fat, and cell pellet fractions of foremilk and stripping milk from infection-free quarters. Changes in milk NAGase activity during a 12 h in vitro incubation were also determined. Eight cows, two quarters per cow, were used. One quarter of each cow received an intramammary infusion of oyster glycogen. N-Acetyl-beta-D-glucosaminidase activity was highest in stripping milk and in milk from infused quarters. The percentages of NAGase activity in skim, fat, and cell pellet fractions were 62.6, 22.4, and 12.6. The NAGase activity of milk incubated in vitro did not significantly change over time. Experiment 2 was conducted to determine if neutrophils lost NAGase activity during extravasation into milk. Leukocytosis was induced in infection-free quarters of five cows. The NAGase activities of peripheral neutrophils and milk neutrophils were not significantly different. Results from both studies suggest that the major source of milk NAGase is the mammary epithelial cell and that milk somatic cells contribute less than 15% of the total milk NAGase activity.

Effectiveness of segregating cows with Staphylococcus aureus intramammary infections was studied over 1 yr. Nine herds were split into control (n = 5) or segregated (n = 4) groups. Cows with S. aureus intramammary infections were milked last in segregated herds. Monthly milk samples were collected aseptically for microbiologic analysis. Mean incidences of S. aureus intramammary infections were 3.7 and 4.3 cases/100 cow-mo in segregated and control herds. The mean prevalence of S. aureus intramammary infections decreased in both segregated and control herds during the study. Mean percentages of cows with S. aureus intramammary infections at the beginning and end of the study were 33.7 and 21.5 in segregated herds and 25.3 and 15.0 in control herds. Cows in all herds with S. aureus intramammary infections were preferentially culled. There were no significant differences in incidence and prevalence of S. aureus intramammary infections between groups, suggesting that S. aureus intramammary infections can be controlled without segregation.

Selenium injections and oral vitamin E supplementation prepartum were related to incidence of retained placenta, metritis, and cystic ovaries in a 2 X 2 factorial experiment. Groups were: 1) selenium and vitamin E, 2) vitamin E, 3) selenium, and 4) control. Incidence of retained placenta was 17.5% in cows of groups 2, 3, and 4, whereas it was reduced to 0% in cows receiving both selenium and vitamin E. Incidence of metritis was 60% for cows injected with selenium and 84% for those not receiving selenium. Cystic ovaries were diagnosed in 19% of cows injected with selenium, and incidence was 47% for cows not treated with selenium. Supplementation of vitamin E was required in addition to selenium for prevention of retained placenta of cows fed stored ensiled forage, and prepartum selenium injections were effective for reducing the incidence of metritis and cystic ovaries during the early postpartum period.

The effect of intramammary antibiotic therapy at calving on mastitis infection prevalence, linear score milk somatic cell count, and milk NAGase activity, 30 d postpartum, and on milk production, 90 to 120 d postpartum, was tested. Cows (n = 175) were split into treatment and control groups at drying off. All cows received commercial dry cow therapy. At calving, treated cows received commercial lactating cow therapy in all quarters after the first two milkings; control cows were not treated. Composite milk samples were aseptically collected from all cows at drying off, calving, and 30 d postpartum. Udder health traits: linear score milk SCC, NAGase activity, and bacterial content in milk, were determined on all samples. The first three DHI milk weights were recorded for all cows. Treatment and control cows had similar prevalences of intramammary infections during the dry and 30-d postpartum periods. Least squares means of linear score milk SCC and NAGase activities were similar at drying off and calving. Cell count scores were similar between groups; NAGase activities were higher in control cows at 30 d postpartum. Control cows tended to produce more milk postpartum. Results demonstrated no advantage of intramammary therapy at calving in improving milk production or udder health.
