Quantifying the protein levels of drug transporters in plasma membrane fraction helps elucidate the function of these transporters. In this study, we conducted a proteomic evaluation of enriched drug-related transporter proteins in plasma membrane fraction prepared from mouse liver and kidney tissues using the membrane protein extraction kit and a bead homogenizer. Crude and plasma membrane fractions were prepared using either the Dounce or bead homogenizer, and protein levels were determined using quantitative proteomics. In liver tissues, the plasma membrane fractions were more enriched in transporter proteins than the crude membrane fractions; the average enrichment ratios of plasma-to-crude membrane fractions were 3.31 and 6.93 using the Dounce and bead homogenizers, respectively. The concentrations of transporter proteins in plasma membrane fractions determined using the bead homogenizer were higher than those determined using the Dounce homogenizer. Meanwhile, in kidney tissues, the plasma membrane fractions were enriched in transporters localized in the brush-border membrane to the same degree for both the homogenizers; however, the membrane fractions obtained using either homogenizer were not enriched in Na+/K+-ATPase and transporters localized in the basolateral membrane. These results indicate that fractionation, using the bead homogenizer, yielded transporter-enriched plasma membrane fractions from mouse liver and kidney tissues; however, no enrichment of basolateral transporters was observed in plasma membrane fractions prepared from kidney tissues.

The Ritter reaction, Brønsted- or Lewis acid-mediated amidation of alkene or alcohol with nitrile via a carbocation, represents a classical method for the synthesis of tertiary amides. Although analogous reaction through a vinyl cation or a species alike may offer a route to enamide, an important synthetic building block as well as a common functionality in bioactive compounds, such transformations remain largely elusive. Herein, we report on a Ritter-type trans- difunctionalization of alkynes with trivalent iodine electrophile and nitrile, which affords β-iodanyl enamides in moderate to good yields. Mediated by benziodoxole triflate (BXT), the reaction proves applicable to a variety of internal alkynes as well as to various alkyl- and arylnitriles. The benziodoxole group in the product serves as a versatile handle for further transformations, thus allowing for the preparation of various tri- and tetrasubstituted enamides that are not readily accessible by other means.

A saturated N-heterocyclic carbene–phenolate bidentate ligand, 3,3a,4,5-tetrahydroimidazo[1,5-a]quinolin-9-olate-1-ylidene (SIzQO), was synthesized and characterized. The SIzQO ligand was then treated with PdClMe(pyridine)2 and [Pd(μ-Cl)Me(2,6-lutidine)]2, which afforded C,C-cis-(SIzQO)PdMe(pyridine) and the thermodynamically unstable trans isomer C,C-trans-(SIzQO)PdMe(2,6-lutidine), respectively. The latter isomerizes at 40 °C into the corresponding cis isomer via a dissociative mechanism. These palladium/SIzQO complexes catalyze the polymerization of ethylene at 100–120 °C, although the catalytic activity is lower than that of a previously reported palladium/imidazo[1,5-a]quinolin-9-olate-1-ylidene (IzQO) system.

Nickel complexes bearing bidentate alkylphophine–phenolate ligands were synthesized and applied to the polymerization of propylene and the copolymerization of propylene and polar monomers. Therein, the use of bulky alkyl substituents on the phosphorus atom was crucial for the formation of highly regioregular polypropylenes. Theoretical calculations suggested that the 1,2-insertion of propylene is favored over the 2,1-insertion in these nickel-catalyzed (co)polymerization reactions. The substituent at the ortho position relative to the oxido group greatly influences the polymerization activity, the molecular weight, and stereoregularity of the polypropylenes. This method delivers moderately isotactic ([mm] ≤ 68%) crystalline polypropylenes. The present system represents the first example for a nickel-catalyzed regiocontrolled copolymerization of propylene and polar monomers such as but-3-en-1-ol, but-3-en-1-yl acetate, and tert-butyl allylcarbamate.

Coordination–insertion copolymerization of ethylene with 1,1-disubstituted ethylenes bearing a polar functional group, such as me-thyl methacrylate (MMA), is a long-standing challenge in catalytic polymerization. The major obstacle for this process is the huge difference in reactivity of ethylene versus 1,1-disubstituted ethylenes towards both coordination and insertion. Herein we report the copolymerization of ethylene and 1,1-disubstituted ethylenes by using an imidazo[1,5-a]quinolin-9-olate-1-ylidene (IzQO)-supported palladium catalyst. Various types of 1,1-disubstituted ethylenes were successfully incorporated into the polyethylene chain. In-depth characterization of the obtained copolymers and mechanistic inferences drawn from stoichiometric reactions of alkylpalladium com-plexes with methyl methacrylate and ethylene indicate that the copolymerization proceeds by the same coordination–insertion mech-anism that has been postulated for ethylene.

Transcriptomic, proteomic, phosphoproteomic, and metabolomic analyses were combined to determine the role of pregnane X receptor (PXR) in nongenotoxic signaling and energy homeostasis in liver after rats were repeatedly orally dosed with the PXR agonist pregnenolone carbonitrile (PCN) for 7 days. Analyses of mRNAs and proteins in the supernatant, membrane, and cytosolic fractions of enlarged liver homogenates showed diverse expression profiles. Gene set enrichment analysis showed that the synchronous increase in mRNAs and proteins involved in chemical carcinogenesis and the response to drug was possibly mediated by the PXR pathway and proteasome core complex assembly was possibly mediated by the Nrf2 pathway. In addition, levels of proteins in the endoplasmic reticulum lumen and involved in the acute-phase response showed specific increase with no change in mRNA level, and those composed of the mitochondrial inner membrane showed specific decrease. The analysis of phosphorylated peptides of poly(A) RNA binding proteins showed a decrease in phosphorylation, possibly by casein kinase 2, which may be related to the regulation of protein expression. Proteins involved in insulin signaling pathways showed an increase in phosphorylation, possibly by protein kinase A, and those involved in apoptosis showed a decrease. Metabolomic analysis suggested the activation of the pentose phosphate and anaerobic glycolysis pathways and the increase of amino acid and fatty acid levels, as occurs in the Warburg effect. In conclusion, the results of combined analyses suggest that PXR's effects are due to transcriptional and post-transcriptional regulation with alteration of nongenotoxic signaling pathways and energy homeostasis.

Although the stabilization has been demonstrated by radialene-type structures - fusing aromatic rings to anti-aromatic rings - in solution, such molecules have never been studied at a single molecular level. Here, we synthesize a cyclobutadiene derivative, dibenzo[b,h]biphenylene, by an on-surface intramolecular reaction. With a combination of high-resolution atomic force microscopy and density functional theory calculations, we found that a radialene structure significantly reduces the anti-aromaticity of the cyclobutadiene core, extracting π-electrons, while the small four-membered cyclic structure keeps a high density of the total charge.

The mechanism of linear polyethylene formation catalyzed by palladium/phosphine−sulfonate and the effect of the ligand structure on the catalytic performance, such as linearity and molecular weight of the polyethylene, were reinvestigated theoretically and experimentally. We used dispersion-corrected density functional theory (DFT-D3) to study the entire mechanism of polyethylene formation from (R2PC6H4SO3)PdMe(2,6-lutidine) (R = Me, t-Bu) and elucidated the key steps that determine the molecular weight and linearity of the polyethylene. The alkylpalladium ethylene complex is the key intermediate for both linear propagation and β-hydride elimination from the growing polymer chain. On the basis of the key species, the effects of substituents on the phosphorus atom (R = t-Bu, i-Pr, Cy, Men, Ph, 2-MeOC6H4, biAr) were further investigated theoretically to explain the experimental results in a comprehensive manner. A palladium-catalyzed alternating copolymerization of [2.2.1]oxabicyclic alkenes 1 with carbon monoxide afforded isomer mixtures of polyketones 2ktn and polyketals 2ktl. Subsequent acid-induced dehydration of polymer 2 furnished novel aromatic polymers consisting of polyketones 3ktn and polyketals 3ktl units. This formal aryne/carbon monoxide copolymerization thus generated the first example of poly(aryne-alt-carbon monoxide)s, in which o-arylene and carbonyl units are incorporated in an alternating fashion.

A series of palladium/alkylphosphine-sulfonate catalysts were synthesized and examined in the homopolymerization of ethylene and the copolymerization of ethylene and polar monomers. Catalysts with alkylphosphine-sulfonate ligands containing sterically demanding alkyl substituents afforded (co)polymers whose molecular weight was increased by up to 2 orders of magnitude relative to polymers obtained from previously reported catalyst systems. The polymer molecular weight was found to be closely correlated to the Sterimol B5 parameter of the alkyl substituents in the alkylphosphine-sulfonate ligands. Thus, the use of bulky alkylphosphine-sulfonate ligands represents an effective and versatile method to prepare high-molecular-weight copolymers of ethylene and various polar monomers, which are difficult to obtain by previously reported methods.

Utilization of palladium catalysts bearing a P-chiral phosphine-sulfonate ligand enabled asymmetric copolymerization of vinyl acetate with carbon monoxide. The obtained γ-polyketones have head-to-tail and isotactic polymer structures. The origin of the regio- and stereoregularities was elucidated by stoichiometric reactions of acylpalladium complexes with vinyl acetate. The present report for the first time demonstrates successful asymmetric coordination-insertion (co)polymerization of vinyl acetate.

Coordination-insertion copolymerization of allyl monomers with ethylene was developed by using a palladium/phosphine-sulfonate catalyst. A variety of allyl monomers, including allyl acetate, allyl alcohol, protected allylamines, and allyl halides, were copolymerized with ethylene to form highly linear copolymers that possess in-chain -CH(2)CH(CH(2)FG)- units.

Coordination copolymerization of vinyl acetate (VAc) with ethylene, leading to linear copolymers that possess in-chain -CH(2)CH(OAc)- units, has been accomplished using novel palladium complexes bearing alkylphosphine-sulfonate ligands.
