Detailed herein is the development of a photochemical intermolecular formal [3+2] cycloaddition between cyclopropylimines and substituted alkenes to generate aminocyclopentane derivatives. The Schiff base of the cyclopropylimine was designed to enable a masked N-centered radical approach in which the requisite open-shell character was achieved upon excitation with visible-light. The cycloaddition products were directly converted to N-functionalized aminocyclopentanes via solvolysis and N-acylation. The photochemical component of this reaction sequence was demonstrated to operate in continuous flow.

Radical aryl migrations are powerful techniques to forge new bonds in aromatic compounds. The growing popularity of photoredox catalysis has led to an influx of novel strategies to initiate and control aryl migration starting from widely available radical precursors. This review encapsulates progress in radical aryl migration enabled by photochemical methods─particularly photoredox catalysis─since 2015. Special attention is paid to descriptions of scope, mechanism, and synthetic applications of each method.

Copper (Cu) incorporation is a key process for fabricating efficient CdTe-based thin-film solar cells and has been used in CdTe-based solar cell module manufacturing. Here, we investigate the effects of different Cu precursors on the performance of CdTe-based thin-film solar cells by incorporating Cu using a metallic Cu source (evaporated Cu) and ionic Cu sources (solution-processed cuprous chloride (CuCl) and copper chloride (CuCl2)). We find that ionic Cu precursors offer much better control in Cu diffusion than the metallic Cu precursor, producing better front junction quality, lower back-barrier heights, and better bulk defect property. Finally, outperforming power conversion efficiencies of 17.2 and 17.5% are obtained for devices with cadmium sulfide and zinc magnesium oxide as the front window layers, respectively, which are among the highest reported CdTe solar cells efficiencies. Our results suggest that an ionic Cu precursor is preferred as the dopant to fabricate efficient CdTe thin-film solar cells and modules.

Phthalimide N-oxyl (PINO) is a potent hydrogen atom transfer (HAT) catalyst that can be generated electrochemically from N-hydroxyphthalimide (NHPI). However, catalyst decomposition has limited its application. This paper details mechanistic studies of the generation and decomposition of PINO under electrochemical conditions. Voltammetric data, observations from bulk electrolysis, and computational studies suggest two primary aspects. First, base-promoted formation of PINO from NHPI occurs via multiple-site concerted proton-electron transfer (MS-CPET). Second, PINO decomposition occurs by at least two second-order paths, one of which is greatly enhanced by base. Optimal catalytic efficiency in PINO-catalyzed oxidations occurs in the presence of bases whose corresponding conjugate acids have pKa's in the range of ∼11-15, which strikes a balance between promoting PINO formation and minimizing its decay.

A two-week summer camp was designed, implemented, and then updated for high school students focusing on organic chemistry, solar energy, and green chemistry principles. Students learn about laboratory safety, perform organic reactions, go on field trips, and take part in interactive lessons that focus on both fundamental chemistry topics as well as green chemistry. Each lab experiment implements at least one green chemistry principle by reducing waste or reagent use while employing additional safety measures. The course has been edited and modified on the basis of student feedback to include additional lessons, field trips, a student presentation, and other activities. Responses to postcourse surveys provided feedback on each component and also indicated that student interest in STEM increased as students gained physical laboratory and research skills.

A short course for high school students was converted from a two-week, in-person laboratory course to a remote and virtual four-week course due to the COVID-19 pandemic. Students met virtually each day to participate in activities that included the following: presentations, demonstrations, interviews, group discussions, lessons, and virtual tours and field trips. The virtual version of the course was able to maintain the same learning objectives and activities when compared to the in-person session and integrated additional expanded lessons, kitchen chemistry activities, and an emphasis on careers in science in order to increase student engagement. Student feedback indicated that interest in a career in STEM was increased overall, and the incorporation of some of the newly introduced activities can be implemented into future versions of the course.

Although back-surface passivation plays an important role in high-efficiency photovoltaics, it has not yet been definitively demonstrated for CdTe. Here, we present a solution-based process, which achieves passivation and improved electrical performance when very small amounts of oxidized Al3+ species are deposited at the back surface of CdTe devices. The open circuit voltage (Voc) is increased and the fill factor (FF) and photoconversion efficiency (PCE) are optimized when the total amount added corresponds to ∼1 monolayer, suggesting that the passivation is surface specific. Addition of further Al3+ species, present in a sparse alumina-like layer, causes the FF and PCE to drop as the interface layer becomes blocking to current flow. The optimized deposit increases the average baseline PCE for both Cu-free devices and devices where Cu is present as a dopant. The greatest improvement is found when the Al3+ species are deposited prior to the CdCl2 activation step and Cu is employed. In this case, the best-cell efficiency was improved from 12.6 to 14.4%. Time-resolved photoluminescence measurements at the back surface and quantum efficiency measurements performed at the maximum power point indicate that the performance enhancement is due to a reduction in the interface recombination current at the back surface.

Herein, an organocatalytic method for photochemical C-O bond cleavage of lignin systems is reported. The use of photochemistry enabled fragmentation of the β-O-4 linkage, the primary linkage in lignin, provides the fragmentation products in good to high yields. The approach was merged with reported oxidation conditions in a one-pot, two-step platform without any intermediary purification, suggesting its high fidelity. The future utility of the organocatalytic method was illustrated by applying the visible light-mediated protocol to continuous flow processing.

Cocaine addiction remains a major public concern throughout the world especially in developed countries. In the last three decades, significant achievements have led to a greater understanding of the signaling pathways involved in the development of cocaine addiction; however, there are no FDA-approved treatments available to reverse or block this brain disease due to either the unsatisfactory therapeutic efficacy or severe side effects. Previous studies have demonstrated that chronic exposure to cocaine elevates levels of cyclic AMP (cAMP) as a neuroadaptative response in reward-related brain regions. Phosphodiesterase 4 (PDE4) inhibitors, which elevate cAMP levels, have been shown to block cocaine-mediated behavioral changes related to psychoactive and reinforcing properties. Unfortunately, previously studied PDE4 inhibitors induce severe side-effects, which limit their clinical usage. In this study, we identified a novel PDE4B inhibitor, KVA-D-88, with an improved selectivity profile compared to previous compounds (e.g., rolipram). Pharmacokinetic studies have shown that this compound is brain penetrant and preferably acts on PDE4B compared to PDE4D in vitro, alluding to less unwanted side effects with KVA-D-88 in vivo. Interestingly, pretreatment with KVA-D-88 significantly inhibited cocaine-induced hyperlocomotor activity. In cocaine self-administering mice with differential schedules, KVA-D-88 strikingly decreased the number of active nose-pokes and cocaine infusions and reduced the break point. Taken together, our findings demonstrate that this novel PDE4 inhibitor, KVA-D-88, could inhibit cocaine-mediated rewarding effects implying its potential clinical usage for cocaine addiction.

[Image: see text] Herein we report the synthesis, SAR, and biological evaluation of a series of 1H-pyrrolo[2,3-b]pyridine-2-carboxamide derivatives as selective and potent PDE4B inhibitors. Compound 11h is a PDE4B preferring inhibitor and exhibited acceptable in vitro ADME and significantly inhibited TNF-α release from macrophages exposed to pro-inflammatory stimuli (i.e., lipopolysaccharide and the synthetic bacterial lipopeptide Pam3Cys). In addition, 11h was selective against a panel of CNS receptors and represents an excellent lead for further optimization and preclinical testing in the setting of CNS diseases.

In this Review, we showcase the upsurge in the development and fundamental photophysical studies of more than 100 metal-organic frameworks (MOFs) as versatile stimuli-responsive platforms. The goal is to provide a comprehensive analysis of the field of photoresponsive MOFs while delving into the underlying photophysical properties of various classes of photochromic molecules including diarylethene, azobenzene, and spiropyran as well as naphthalenediimide and viologen derivatives integrated inside a MOF matrix as part of a framework backbone, as a ligand side group, or as a guest. In particular, the geometrical constraints, photoisomerization rates, and electronic structures of photochromic molecules integrated inside a rigid MOF scaffold are discussed. Thus, this Review reflects on the challenges and opportunities of using photoswitchable MOFs in next-generation multifunctional stimuli-responsive materials while highlighting their use in optoelectronics, erasable inks, or as the next generation of sensing devices.

For the first time, we report the ability to control radionuclide species release kinetics in metal-organic frameworks (MOFs) as a function of postsynthetic capping linker installation, which is essential for understanding MOF potential as viable radionuclide wasteform materials or versatile platforms for sensing, leaching, and radionuclide sequestration. The radiation damage of prepared frameworks under γ radiation has also been studied. We envision that the presented studies are the first steps toward utilization of the reported scaffolds for more efficient nuclear waste administration.

Transient receptor potential canonical (TRPC) channels are highly homologous, nonselective cation channels that form many homo- and heterotetrameric channels. These channels are highly abundant in the brain and kidney and have been implicated in numerous diseases, such as depression, addiction, and chronic kidney disease, among others. Historically, there have been very few selective modulators of the TRPC family in order to fully understand their role in disease despite their physiological significance. However, that has changed recently and there has been a significant increase in interest in this family of channels which has led to the emergence of selective tool compounds, and even preclinical drug candidates, over the past few years. This review will cover these new advancements in the discovery of TRPC modulators and the emergence of newly reported structural information which will undoubtedly lead to even greater advancements.

Arylethylamines are abundant motifs in myriad natural products and pharmaceuticals, so efficient methods to synthesize them are valuable in drug discovery. In this work, we disclose an intramolecular alkene aminoarylation cascade that exploits the electrophilicity of a nitrogen-centered radical to form a C–N bond, then repurposes the nitrogen atom’s sulfonyl activating group as a traceless linker to form a subsequent C–C bond. This photoredox catalysis protocol enables the preparation of densely substituted arylethylamines from commercially abundant aryl sulfonamides under mild conditions. Reaction optimization, scope, mechanism, and synthetic applications are discussed.

Compounds containing bridged bicyclic carbon skeletons are desirable building blocks for medicinal chemistry. However, as a result of their inefficient, linear syntheses, commercially available compounds of this sort are plagued by high costs and/or a lack of diversity in substitution patterns. Herein we report the conversion of the readily available bicyclo[1.1.1]pentan-1-amine substructure to a wide range of poly-substituted bicyclo[3.1.1]pentan-1-amines using imine photochemistry. To our knowledge, this is the first reported method to convert the bicyclo[1.1.1]pentane skeleton to the bicyclo[3.1.1]heptane skeleton. Hydrolysis of the imine products gives complex, sp3-rich primary amine building blocks.

We report the development of a droplet microfluidic/mass spectrometry platform for performing high-throughput photochemical reaction discovery on picomole scale. We also describe the successful translation of picomole scale flow screening conditions to millimole scale flow operations, highlighting the utility of this platform for discovery chemistry applications.

Electron donor-acceptor (EDA) complexes can controllably generate radicals under mild conditions through selective photoexcitation events. However, unproductive reactivity from fast deactivation of the photoexcited complexes through back electron transfer has slowed the development of EDA complexes in synthetic methodology. Here, we disclose the study of EDA complexes derived from 2-methoxynaphthalene donor and acylated ethyl isonicotinate N-oxide acceptor that undergo a fast N–O bond fragmentation event upon photoexcitation. This reaction design not only overcomes the limitations of back electron transfer but also enables the regeneration of the donor species, representing a rare example EDA photochemistry in a catalytic regime. The synthetic utility is demonstrated through visible light-driven radical trifluoromethylation and Minisci alkylation reactions. The scalability of the EDA complex promoted reaction evidenced by the successful multigram-scale trifluoromethylation of methyl N-Boc pyrrole-2-carboxylate in a continuous flow manifold.
