A new convenient and continuous fluorometric assay method for acetylcholinesterase (ACHE) and its inhibitor screening is successfully established with the ensemble of 1 [a TPE (tetraphenylethylene) compound with two sulfonate (-S[O.sub.3]) units] and myristoylcholine (an amphiphilic compound as a good substrate of AChE). This new assay method is designed by making use of the aggregation-induced emission (AIE) feature of TPE compounds. Both dynamic light scattering (DLS) and fluorescence confocal microscopic measurements indicated the formation of aggregation complex for the ensemble of 1 and myristoylcholine and further disassembly of the aggregation complex after introducing ACHE. The analysis for AChE can be carried out continuously, and AChE with concentration as low as 0.5 U/mL can be assayed. The results also clearly demonstrate the usefulness of this convenient assay method for kinetic study of AChE-catalyzed myristoylcholine hydrolysis and screening inhibitors of ACHE. Given its simplicity and easy operation, this method may extend to high-throughput screening of AChE inhibitors and relevant Alzheimer's disease (AD) drug discovery.

By making use of the aggregation-induced emission feature of silole, compound 1 with an ammonium group is designed and synthesized with a view to developing a new optical probe for fluorescence turn-on detection of DNA and label-free fluorescence nuclease assay. The fluorescence of 1 increases largely upon mixing with DNA, in particular for long DNA, indicating that I can be used for fluorescence turn-on detection of DNA. More interestingly, 1 can be employed to follow the DNA cleavage process by nuclease. Therefore, a label-free fluorescence nuclease assay method is successfully established with 1. Furthermore, this label-free fluorescence assay can also be used for inhibitor screening of nucleases. Given its simplicity, easy operation, sensitivity and cost-effectiveness, this method can be extended to other nuclease assays and high-throughput screening of nuclease inhibitors.

A new TTF-anthracene dyad fusion with the crown ether unit is described. It is interesting to find that the fluorescence of this new dyad could be modulated with (Na.sup.+) and C60, and its fluorescence intensity could be largely enhanced only in the presence of both (Na.sup.+) and C60.

A search for low energy structures of water clusters was performed with a combination of three computational tools. For the sizes of main interest, n=48,123, and 293, input configurations included spheroid structures cut from crystalline ice, and amorphous structures.

A variational multifaceted dividing surface generalization of the variable reaction coordinate (VCR) approach is described. For the addition of H atoms to propargyl radical a high level ab initio potential is employed and comparisons are made with trajectory simulations and with prior implementations of VRC-TST (transition state theory).

The quantitative photophysical description of matrix-assisted laser desorption ionization (MALDI) is explored by using a wide range of optical pumping rates. The longer laser pulse can sequentially desorb and ionize analyte molecules due to the extended interaction of the laser pulse and the MALDI plume.

Intramolecular photoinduced charge-separation and charge-recombination processes of tetrathiafulvalene-(spacer)-napthalenediimide-(spacer)-tetrathiafulvalenetriads (TTF-(sp)NIm-(sp)-TTF) were investigated. The fluorescence lifetimes of NIm moiety considerably decreased compared with that of the NIm molecule, suggesting the photoinduced intramolecular charge-separation via the singlet-excited state of the NIm moiety.

ConspectusIn recent decades, there has been rapid development in the field of polymer semiconductors, particularly those based on conjugated donor-acceptor (D-A) polymers exhibiting high charge mobilities. Furthermore, the application of polymer semiconductors has been successfully extended to a wide range of functional devices, including sensors, photodetectors, radio frequency identification (RFID) tags, electronic paper, skin electronics, and artificial synapses. Over the past few years, there has been a growing focus on stimuli-responsive polymer semiconductors, which have the potential to impart additional functionalities to conventional field-effect transistors, garnering increased attention within the research community. In this context, phototunable polymer semiconductors have received significant attention due to their ability to utilize light as an external stimulus, enabling remote control of device performance with high spatiotemporal resolution. Meanwhile, integration of field-effect transistors with polymer semiconductors can enable the realization of complex functions. To achieve this, precise and controllable patterning of polymer semiconductors becomes essential. In this Account, we discuss our research findings in the context of phototunable and photopatternable polymer semiconductors. These developments encompass the following key aspects: (i) polymer semiconductors, such as poly(diketopyrrolopyrrole-quaterthiophene) ( PDPP4T ), exhibit phototunability when blended with the photochromic compound hexaarylbiimidazole ( HABI ). The photo/thermal-responsive field-effect transistors (FETs) can be fabricated using blending thin films. Remarkably, these photo/thermal-responsive transistors can function as photonically programmable and thermally erasable nonvolatile memory devices. (ii) By incorporating photoswitchable groups like azo and spiropyran into the side chains of conjugated D-A polymers, we can create phototunable polymer semiconductors. The reversible isomerization of azo and spiropyran groups significantly influences the charge transport properties of these polymer semiconductors. Consequently, the performance of the resulting FETs can be reversibly tuned through UV/visible or near-infrared light (NIR) irradiation. Notably, the incorporation of two distinct azo groups into the side chains leads to polymer semiconductors with tristable semiconducting states, offering the ability to logically control device performance using light irradiation at three different wavelengths. (iii) Photopatterning of p -type, n -type, and ambipolar semiconductors featuring alkyl side chains can be achieved using a diazirine-based, four-armed photo-cross-linker ( 4CNN ) with a loading concentration of no more than 3% (w/w). Furthermore, the semiconducting performances of FETs with patterned thin films were found to be satisfactorily uniform. Importantly, the cross-linked thin films are robust and show good resistance to organic solvents, which is useful for fabricating all-solution processable multilayer electronic devices. (iv) The introduction of azide groups into the side chains of conjugated polymers results in a single-component semiconducting photoresist. The presence of azide groups renders the side chains with photo-cross-linking ability, enabling the successful formation of uniform patterns, even as small as 5 μm, under UV light irradiation. Benefiting from the single component feature, field-effect transistors with individual patterned thin films display satisfactorily uniform performances. Moreover, this semiconducting photoresist has proven effective for efficiently photopatterning other polymer semiconductors, demonstrating its versatility.

An accurate single-molecule kinetic isotope effect (sm-KIE) was applied to circumvent the inherent limitation of conventional ensemble KIE by using graphene-molecule-graphene single-molecule junctions. In situ monitoring of the single-molecule reaction trajectories in real time with high temporal resolution has the capability to characterize the deeper information brought by KIE. The C-O bond cleavage and the C-C bond formation of the transition state (TS) were observed in the Claisen rearrangement through the secondary kinetic isotope effect, demonstrating the high detection sensitivity and accuracy of this method. More interestingly, this sm-KIE can be used to determine TS structures under different electric fields, revealing the multidimensional regulation of the TS. The detection and manipulation of the TS provide a broad perspective to understand and optimize chemical reactions and biomimetic progress.

The diallene-containing compound dACl-1 was unexpectedly obtained by the unconventional transformation of two carbonyl groups in 4,4',5,5'-tetrachloro-10 H ,10' H -[9,9'-bianthracenylidene]-10,10'-dione into diallenes. In addition, the two 1-triisopropylsilyl (TIPS) groups in dACl-1 were easily removed to yield dACl-2 . The reaction mechanism was investigated and is discussed. Moreover, both compounds are stable under ambient conditions, and, in particular, dACl-1 is thermally stable at 315 °C.

Peptides with modular structure provide a tailorable platform for constructing responsive supramolecular assemblies, which are attractive as functional biomaterials and smart sensors. In this work, the feasibility of regulating small peptides assembly with molecular design and metal ion recognition was demonstrated. Tripeptides were designed and found to have diverse response and self-assembly behavior to Hg 2+ . The incorporation of an aggregation-induced emission fluorophore TPE enabled the visualization of Hg 2+ recognition and the assembly phenomenon. A structural analogue (Pep2) to γ-glutathione was identified with high specificity and nanomolar response to Hg 2+ both in buffer solution and living cells. Driven by the coordination force and noncovalent intramolecular stacking, assembling of twisted nanofibers from Pep2-TPE and Hg 2+ were observed. Benefiting from its biocompatibility, fast and switchable fluorescence response, Pep2-TPE was applied for imaging and monitoring Hg 2+ distribution in living cells and zebrafish. With good permeability to plasma membrane and tissues, Pep2-TPE indicated the preferential distribution of Hg 2+ in cell nucleoli and brain of zebrafish, which is related with the deleterious effect of inorganic mercury in living biosystems.

Aggregation-induced emission (AIE) nanoparticles have been shown promise for fluorescence bioimaging and photodynamic therapy due to the good combination of nanoparticles and organic dyes or photosensitizers. Among several kinds of AIE nanoparticles, those that are capsulated with nanographene oxides (NGO) are easy to make, size-tunable, and have proven to be very stable in deionized water. However, the stability in saline solution still needs improvement for further applications in chemical or biomedical fields, and the efficacy of photodynamic therapy using NGO-capsulate AIE photosensitizers has not been evaluated yet. Herein, we modified NGO with polyethylene glycol (PEG) to improve the stability of NGO-capsulated AIE nanoparticles in phosphate buffer saline. Furthermore, by combining this modification method with a dual-functional molecule which has both typical AIE property and photosensitizing ability, we performed both two-photon fluorescence bioimaging and photodynamic therapy in vitro and in vivo. Our work shows that AIE nanoparticles capsulated with PEGylated nanographene oxide can be a powerful tool for future bioimaging and photodynamic therapy applications.

Organic semiconductors have received increasing attentions in recent years because of their promising applications in various optoelectronic devices. The key performance metric for organic semiconductors is charge carrier mobility, which is governed by the electronic structures of conjugated backbones and intermolecular/interchain π-π interactions and packing in both microscopic and macroscopic levels. For this reason, more efforts have been paid to the design and synthesis of conjugated frameworks for organic semiconductors with high charge mobilities. However, recent studies manifest that appropriate modifications of side chains that are linked to conjugated frameworks can improve the intermolecular/interchain packing order and boost charge mobilities. In this Account, we discuss our research results in context of modification of side chains in organic semiconductors for charge mobility enhancement. These include the following: (i) The lengths of alkyl chains in sulfur-rich thiepin-fused heteroacences can dramatically influence the intermolecular arrangements and orbital overlaps, ushering in different hole mobilities. Inversely, the lamellar stacking modes of alkyl chains in naphthalene diimide (NDI) derivatives with tetrathiafulvalene (TTF) units are affected by the structures of conjugated cores. (ii) The steric hindrances owing to the bulky branching chains can be weakened by partial replacement of the branching alkyl chains with linear ones for diketopyrrolopyrrole (DPP)-based D (donor)-A (acceptor) conjugated polymers. Such modification of side chains makes the polymer backbones more planar and thus interchain packing order and charge mobilities are improved. The incorporation of hydrophilic tri(ethylene glycol) (TEG) chains into the polymers also leads to improved interchain packing order. In particular, the polymer in which TEG side chains are distributed uniformly exhibits relatively high charge mobility without thermal annealing. (iii) The incorporation of urea groups in the side chains induces the polymer chains to pack more orderly and form large domains because of the additional H-bonding among urea groups. Accordingly, thin film mobilities of the conjugated D-A polymers with side chains entailing urea groups are largely boosted in comparison with those of polymers of the same backbones with either branching alkyl chains or branching/linear alkyl chains. (iv) The torsions of branching alkyl chains in conjugated D-A polymers can be inhibited to some extent upon incorporation of tiny amount of NMe 4 I in the thin film. As a result, the polymer thin films with NMe 4 I exhibit improved crystallinity, and charge mobilities can be boosted by more than 20 times. (v) Side chains with functional groups in the conjugated polymers can endow the thin film field-effect transistors (FETs) with sensing functionality. FETs with the conjugated polymer with -COOH groups in the side chains show sensitive, selective, and fast responses toward ammonia and amines, while FETs with the ultrathin films of the polymer containing tetra(ethylene glycol) (TEEG) in the side chains can sense alcohol vapors (in particular ethanol vapor) sensitively and selectively with fast response.

In this paper, we report Zincke's salt-substituted tetraphenylethylenes 1a and 1b with Cl - and PF 6 - as counteranions, respectively. The crystal structure of 1b was determined. Both 1a and 1b are almost nonemissive even in the aggregated states. This is attributed to the photoinduced electron transfer from 2,2-bis(4-methoxyphenyl)-1-phenylvinyl-phenyl unit to 1-(2,4-dinitrophenyl) pyridinium unit within 1a and 1b. The results demonstrate that the emissions of 1a and 1b in aqueous solution can be switched on upon either reaction with GSH or light irradiation. On the basis of the reaction between 1a and GSH, 1a can be utilized for the fluorescence turn-on detection of GSH selectively, and GSH with concentration as low as 36.9 nM can be detected. The transformation of 1b into 2 under light irradiation results in the fluorescence imaging of Hela and U2OS cells and phototoxicity toward Hela and U2OS cells after the protonation of pyridine unit in 2 because of the acidic environment of tumor cells. Aggregates of 1b can be up-taken by Hela and U2OS cells and fluorescence imaging has been successfully recorded with CLSM. Moreover, the protonated form of 2 can function as photosensitizer and 1b shows phototoxicity toward tumor cells such as Hela and U2OS cells.

An endo-polygalacturonase gene (pga-zj5a) was cloned by reverse transcription from cDNAs synthesized from Aspergillus niger ZJ5 total RNA. The open reading frame of pga-zj5a was 1089 base pairs encoding 362 amino acids. Pga-zj5a lacking a signal peptide sequence was successfully amplified using A. niger ZJ5 cDNA as the template and was ligated into the pPIC9 vector. The resulting plasmid was transformed into competent cells of Pichia pastoris GS115 for heterologous expression. The polygalacturonase showed a maximum activity level of 10436 U/mL in the culture supernatant from a 3 L fermenter. Assays of enzymatic properties showed that the optimal pH and temperature of the recombinant PGA-ZJ5A were 4.5 and 40 °C, respectively. PGA-ZJ5A was effective in pear juice clarification, increased the volume of pear juice by 41.8%, and improved its light transmittance 3-fold.
