The detrimental environmental effects of surfactant-like contaminants (SLCs) with distinctive amphiphilic structures have garnered significant attention, particularly since perfluorooctanesulfonate was classified as a persistent organic pollutant. Despite the numerous absorbents developed for SLCs removal, the underlying interaction mechanisms remain speculative and lack experimental validation. To address this research gap, we elucidate the mechanistic insights into the selective removal of SLCs using mesoporous polydopamine nanospheres (MPDA) fabricated via a novel soft-template method. We employed low-field nuclear magnetic resonance to quantitatively characterize the hydrophilicity of the absorbents using water molecules as probes. The results demonstrated that MPDA with uniform mesopores exhibited a remarkable threefold enhancement in SLCs' adsorption capacity compared to conventional polydopamine particles via intraparticle diffusion. We further demonstrated the dominant effects of electrostatic and hydrophobic interactions on the selective removal of SLCs with MPDA by regulating the isoelectric pH value and performing a comparative analysis. The mechanism-inspired SLC-removal strategy achieved an average removal rate of 76.3% from highly contaminated wastewater. Our findings offer new avenues for applying MPDA as an efficient adsorbent and provide innovative and mechanistic insights for targeted SLC removal in complex wastewater matrices.

H2S plays important physiological and pathological roles in the cardiovascular system and nervous system. However, recent evidence imply that hydrogen polysulfides (H2Sn) are the actual signaling molecules in cells. Although H2Sn have been demonstrated to be responsible for mediating tumor suppressors, ion channels, and transcription factors, more of their biological effects are still need to be elaborated. On one hand, H2Sn have been suggested to be generated from endogenous H2S upon reaction with reactive oxygen species (ROS). On the other hand, H2Sn derivatives are proposed to be a kind of direct antioxidant against intracellular oxidative stress. This conflicting results should be attributed to the regulation of redox homeostasis between ROS and H2Sn. Superoxide anion (O2(•-)) is undoubtedly the primary ROS existing in mitochondria. We reason that the balance of O2(•-) and H2Sn are pivotal in physiological and pathological processes. Herein, we report two near-infrared fluorescent probes Hcy-Mito and Hcy-Biot for the detection of O2(•-) and H2Sn in cells and in vivo. Hcy-Mito is conceived to be applied in mitochondria, and Hcy-Biot is designed to target tumor tissue. Both of the probes were successfully applied for visualizing exogenous and endogenous O2(•-) and H2Sn in living cells and in tumor mice models. The results demonstrate that H2Sn can be promptly produced by mitochondrial oxidative stress. Flow cytometry assays for apoptosis suggest that H2Sn play critical roles in antioxidant systems.

A theoretical study is presented of the template-assisted formation of crystalline superstructures of magnetic-dielectric core-shell particles. The templates produce highly localized gradient fields and a corresponding magnetic force that guides the assembly with nanoscale precision in particle placement. The process is studied using two distinct and complementary computational models that predict the dynamics and energy of the particles, respectively. Both mono- and polydisperse colloids are studied, and the analysis demonstrates for the first time that although the particles self-assemble into ordered crystalline superstructures, the particle formation is not unique. There is a Brownian motion-induced degeneracy in the process wherein various distinct, energetically comparable crystalline structures can form for a given template geometry. The models predict the formation of hexagonal close packed (HCP) and face centered cubic (FCC) structures as well as mixed phase structures due to in-plane stacking disorders, which is consistent with experimental observations. The polydisperse particle structures are less uniform than the monodisperse particle structures because of the irregular packing of different-sized particles. A comparison of self-assembly using soft- and hard-magnetic templates is also presented, the former being magnetized in a uniform field. This analysis shows that soft-magnetic templates enable an order-of-magnitude more rapid assembly and much higher spatial resolution in particle placement than their hard-magnetic counterparts. The self-assembly method discussed is versatile and broadly applies to arbitrary template geometries and multilayered and multifunctional mono- and polydisperse core-shell particles that have at least one magnetic component. As such, the method holds potential for the bottom-up fabrication of functional nanostructured materials for a broad range of applications. This work provides unprecedented insight into the assembly process, especially with respect to the viability and potential fundamental limitations of realizing structure-dependent material properties for applications.

Inhibition of mitotic kinesins represents a novel approach for the discovery of a new generation of anti-mitotic cancer chemotherapeutics. We report here the discovery of the first potent and selective inhibitor of centromere-associated protein E (CENP-E) 3-chloro-N-{(1S)-2-[(N,N-dimethylglycyl)amino]-1-[(4-{8-[(1S)-1-hydroxyethyl]imidazo[1,2-a]pyridin-2-yl}phenyl)methyl]ethyl}-4-[(1-methylethyl)oxy]benzamide (GSK923295; 1), starting from a high-throughput screening hit, 3-chloro-4-isopropoxybenzoic acid 2. Compound 1 has demonstrated broad antitumor activity in vivo and is currently in human clinical trials.
