Abstract

The addition of lead to diphenyl diselenide in ethylenediamine (en) or pyridine (py) allowed for the observation of the solvento complexes, (en)Pb(SePh)2 or (py)2Pb(SePh)2, respectively. Performing this reaction in dimethyl sulfoxide and subsequent crystallization was found to afford Pb(SePh)2. Inductively coupled plasma optical emission spectroscopy revealed a 1:2 lead to selenium ratio for all three complexes. Nuclear magnetic resonance spectroscopy confirms that Pb(SePh)2 is readily solubilized by ethylenediamine, and electrospray ionization mass spectrometry supports the presence of Pb(SePh)2 moieties in solution. Single-crystal X-ray diffraction analysis of the pyridine adduct, (py)2Pb(SePh)2, revealed a seesaw molecular geometry featuring equatorial phenylselenolate ligands. Crystals of Pb(SePh)2 grown from dimethyl sulfoxide revealed one-dimensional polymeric chains of Pb(SePh)2. We believe that the lead(II) phenylselenolate complexes form via an oxidative addition reaction.

Abstract

A strategy is described that results in the trapping of the substrate hydrazine (H2N-NH2) as an adduct bound to the active site metal cluster of nitrogenase, and this bound adduct is characterized by EPR (electron paramagnetic resonance) and ENDOR (electron-nuclear double resonance) spectroscopies. The EPR signal showed Curie law temperature dependence similar to the resting state EPR signal.

Abstract

A study is conducted to show that the iron-molybdenum (FeMO)-cofactor of the nitrogenase alpha-70(super lle) molybdenum-iron (MoFe) protein variant accumulates a novel S = 1/2 state that can be trapped during the reduction of protons to H(sub 2). The data does not favor any model involving protonation of sulfides, however it suggests that the intermediate contains two chemically equivalent bound hydrides.

Abstract

PCR cassette mutagenesis helps design zinc-finger yeast ADR1 mutants with different binding specificity to 16 UAS1 variants of thymine bases T5 and T6. Substitution of His for Leu146 decreases the binding of wild type UASI having T6, but decreases the binding to sequences having purine A6 or G6. Substitution of Lys for Leu146 increases the binding of both G6 and T6. Analysis of mutants with Glu for Arg 149 confirms the interaction of Arg 149 with G5 or T5 in UAS1 DNA.

Nitrogenase is the metalloenzyme that catalyzes the nucleotide-dependant reduction of N2, and reduction of variety of other triply bonded substrates, including the alkyne, acetylene. Substitution of the alpha-70(super val) residue in the nitrogenase MoFe protein by alanine expands the range of substrates to include short-chained alkynesnot reduced by the unaltered protein.

Abstract

The intermediate generated at a high concentration during reduction of the natural nitrogenase substrate ([N.sub.2]) by wild-type MoFe protein has shown that it contains [N.sub.2] bound to the active-site FeMo cofactor. The single type of [super 15]N-coupled nucleus from the field dependence, along with the absence of an associated exchangeable [super 1]H ENDOR signal, is shown to be consistent with the [N.sub.2] molecule bound end-on to the FeMo cofactor.

The structure of NifB-co, an Fe-S cluster produced by the enzyme NifB, bound to NifX protein from Azotobacter vinelandii is studied. The results revealed that the interstitial light atom is inserted at the NifB-co stage in iron molybdenum cofactor (FeMo-co) biosynthesis.

A molybdenum K-edge X-ray absorption spectroscopy (XAS) study is conducted to analyze the ligand environment of the Mo bound on the as purified NifEN. It is strongly proposed that the Mo is a part of a cluster whose first two coordination spheres represent a [Mo[Fe.sub.3][S.sub.4]] cubane fragment as part (or whole ) of some cluster on NifEN.

Abstract

An innovative emulsification technology that produces modern emulsions at a reasonable cost has been demonstrated. This process is useful for producing conventional emulsions, and so-called "surfactant free" emulsions, which are stabilized by particles and/or amphipathic polymers. Such emulsions present significant formulation and processing challenges because conventional emulsification techniques rely upon shear force to break up the droplets formed by the discontinuous phase and to transport the emulsifier to the interface. Microchannel emulsification technology is unlike traditional methods that use high-shear forces to form small droplets. This technology, which consists of intervening microchannels with apertured substrates, adds the discontinuous to the continuous phase, one droplet at a time, and provides better control than conventional methods. The droplet size distribution is controlled by adjusting critical process parameters (mixing energy, mixing time), and precise heating and/or cooling.

Abstract

It has been hypothesized that the ribosome gains additional fidelity during protein translation by probing structural differences in tRNA species. We measure the translocation kinetics of different tRNA species through ~3 nm diameter synthetic nanopores. Each tRNA species varies in the time scale with which it is deformed from equilibrium, as in the translocation step of protein translation. Using machine-learning algorithms, we can differentiate among five tRNA species, analyze the ratios of tRNA binary mixtures, and distinguish tRNA isoacceptors. [ABSTRACT FROM AUTHOR]
