Oppositely charged polymerized ionic liquids (PILs) were used to form complex coacervates in two different organic solvents, 2,2,2-trifluoroethanol (TFE) and hexafluoro-2-propanol (HFIP), and the corresponding phase diagrams were constructed using UV-vis, NMR, and turbidity experiments. While previous studies on complex coacervates have focused almost exclusively on aqueous environments, the use of PILs in the current work enabled studies in solvents with substantially lower dielectric constants (27.0 for TFE, 16.7 for HFIP). The critical salt concentration required to induce complete miscibility was roughly 2-fold larger in HFIP compared with TFE, and two different PIL complexes, solidlike precipitates and liquidlike coacervates, were found in both systems. Polymer science has been driven by ever-increasing molecular complexity, as polymer synthesis expands an already-vast palette of chemical and architectural parameter space. Copolymers represent a key example, where simple homopolymers have given rise to random, alternating, gradient, and block copolymers. Polymer physics has provided the insight needed to explore this monomer sequence parameter space. The future of polymer science, however, must contend with further increases in monomer precision, as this class of macromolecules moves ever closer to the sequence-monodisperse polymers that are the workhorses of biology. The advent of sequence-defined polymers gives rise to opportunities for material design, with increasing levels of chemical information being incorporated into long-chain molecules; however, this also raises questions that polymer physics must address. What properties uniquely emerge from sequence-definition? Is this circumstance-dependent? How do we define and think about sequence dispersity? How do we think about a hierarchy of sequence effects? Are more sophisticated characterization methods, as well as theoretical and computational tools, needed to understand this class of macromolecules? The answers to these questions touch on many difficult scientific challenges, setting the stage for a rich future for sequence-defined polymers in polymer physics.

In this study, we provide the first demonstration that aqueous complex coacervates can be electrospun into chemically robust polyelectrolyte complex (PEC) fiber mats. PECs form due to electrostatic complexation between oppositely charged polymers. Here, we exploit the ability of salt to plasticize PECs, thus enabling the electrospinning of solid fibers. Electrospinning solutions were composed of a pair of strong polyelectrolytes, poly(4-styrenesulfonic acid, sodium salt) and poly(diallyldimethylammonium chloride) using potassium bromide as the plasticizing salt. We systematically investigated the effect of salt concentration and electrospinning apparatus parameters on fiber formation. Electrospun PEC fiber mats were stable over a wide range of pH values, ionic strength conditions, and many organic solvents. This study demonstrates that the electrospinning of aqueous complex coacervates can generate chemically robust, free-standing PEC fiber mats while circumventing the reliance on organic solvents, the challenge of working with entangled polyelectrolytes in solution, and the need to chemically cross-link the as-spun fibers. These PEC fiber mats hold potential in applications where environmentally benign fiber mats are imperative, such as tissue engineering scaffolds and water purification technologies.

A complex coacervate is a fluid phase that results from the electrostatic interactions between two oppositely charged macromolecules. The nature of the coacervate core structure of hydrogels and micelles formed from complexation between pairs of diblock or triblock copolymers containing oppositely charged end-blocks as a function of polymer and salt concentration was investigated. Both ABA triblock copolymers of poly[(allyl glycidyl ether)-b-(ethylene oxide)-b-(allyl glycidyl ether)] and analogous poly[(allyl glycidyl ether)-b-(ethylene oxide)] diblock copolymers, which were synthesized to be nearly one-half of the symmetrical triblock copolymers, were studied. The poly(allyl glycidyl ether) blocks were functionalized with either guanidinium or sulfonate groups via postpolymerization modification. Mixing of oppositely charged block copolymers resulted in the formation of nanometer-scale coacervate domains. Small angle neutron scattering (SANS) experiments were used to investigate the size and spacing of the coacervate domains. The SANS patterns were fit using a previously vetted, detailed model consisting of polydisperse core-shell micelles with a randomly distributed sphere or body-centered cubic (BCC) structure factor. For increasing polymer concentration, the size of the coacervate domains remained constant while the spatial extent of the poly(ethylene oxide) (PEO) corona decreased. However, increasing salt concentration resulted in a decrease in both the coacervate domain size and the corona size due to a combination of the electrostatic interactions being screened and the shrinkage of the neutral PEO blocks. Additionally, for the triblock copolymers that formed BCC ordered domains, the water content in the coacervate domains was calculated to increase from approximately 16.8% to 27.5% as the polymer concentration decreased from 20 to 15 wt %.

Proteins have gained increasing success as therapeutic agents; however, challenges exist in effective and efficient delivery. In this work, we present a simple and versatile method for encapsulating proteins via complex coacervation with oppositely charged polypeptides, poly(l-lysine) (PLys) and poly(d/l-glutamic acid) (PGlu). A model protein system, bovine serum albumin (BSA), was incorporated efficiently into coacervate droplets via electrostatic interaction up to a maximum loading of one BSA per PLys/PGlu pair and could be released under conditions of decreasing pH. Additionally, encapsulation within complex coacervates did not alter the secondary structure of the protein. Lastly the complex coacervate system was shown to be biocompatible and interact well with cells in vitro. A simple, modular system for encapsulation such as the one presented here may be useful in a range of drug delivery applications.

We consider polyelectrolyte solutions which, under suitable conditions, phase separate into a liquid-like coacervate phase and a coexisting supernatant phase that exhibit an extremely low interfacial tension. Such interfacial tension provides the basis for most coacervate-based applications, but little is known about it, including its dependence on molecular weight, charge density, and salt concentration. By combining a Debye-Hückel treatment for electrostatic interactions with the Cahn-Hilliard theory, we derive explicit expressions for this interfacial tension. In the absence of added salts, we find that the interfacial tension scales as N -3/2 (η/η c -1) 3/2 near the critical point of the demixing transition, and that it scales as η 1/2 far away from it, where N is the chain length and η measures the electrostatic interaction strength as a function of temperature, dielectric constant, and charge density of the polyelectrolytes. For the case with added salts, we find that the interfacial tension scales with the salt concentration ψ as N -1/4 (1-ψ/ψ c ) 3/2 near the critical salt concentration ψ c . Our predictions are shown to be in quantitative agreement with experiments and provide a means to design new materials based on polyelectrolyte complexation.

We report on a microfluidic method for the formation of aqueous/lipid mesophases to enable screening of suitable crystallization conditions of membrane proteins from a membrane-like phase in sub-20 nanoliter volumes. This integrated microfluidic chip and the employed mixing strategy address the specific challenges associated with the mixing of fluids of highly different viscosities (here a factor of 30) as well as the non-Newtonian character of the resulting mesophases. The chip requires less than 20 nL of material per condition screened whereas typically on the order of 10 μL or more is needed for a batch preparation in the present screening methods. We validated our approach with the successful crystallization of the membrane protein bacteriorhodopsin.
