Abstract

The transformylase activity of the Escherichia coli 4-amino-4-deoxy-L-arabinose (ArnA) contained in its 300N-terminal residues is demonstrated. The enzyme adopts a bilobal structure with an N-terminal Rossman fold domain containing the N-10-formyltetrahydrofolate binding site and a C-terminal subdomain resembling an OB fold.

Abstract

Gram negative bacteria including Escherichia coli, Salmonella typhimurium, and pseudomonas aeruginosa can modify the structure of lipid A in their outer membrane with 4-amino-4-deoxy-L-arabinose (Ara4N). It is shown that NAD+ dependent decarboxylating activity is contained in the 360 amino acid C-terminal domain of ArnA.

Abstract

Results indicate that the site involved in the binding of soluble N-ethylmaleimide sensitive factor (NSF) attachment protein is located between the NSF-N subdomains. Data discuss the sites of the NSF protomer mediating nucleotide hydrolysis and the ATP binding site on NSF-D2 limiting ATPase activity.
