Herein, we first prepared a novel anti-TROP2 antibody-drug conjugate (ADC) hIMB1636-MMAE using hIMB1636 antibody chemically coupled to monomethyl auristatin E (MMAE) via a Valine-Citrulline linker and then reported its characteristics and antitumor activity. With a DAR of 3.92, it binds specifically to both recombinant antigen ( K D ∼ 0.687 nM) and cancer cells and could be internalized by target cells and selectively kill them with IC 50 values at nanomolar/subnanomolar levels by inducing apoptosis and G2/M phase arrest. hIMB1636-MMAE also inhibited cell migration, induced ADCC effects, and had bystander effects. It displayed significant tumor-targeting ability and excellent tumor-suppressive effects in vivo, resulting in 5/8 tumor elimination at 12 mg/kg in the T3M4 xenograft model or complete tumor disappearance at 10 mg/kg in BxPc-3 xenografts in nude mice. Its half-life in mice was about 87 h. These data suggested that hIMB1636-MMAE was a promising candidate for the treatment of pancreatic cancer with TROP2 overexpression.

The current global issue of antibiotic resistance is serious, and there is an urgent requirement of developing novel antibiotics. Octapeptins have recently regained interest because of their activities against resistant Gram-negative bacteria. We synthesized four natural octapeptins and 33 derivatives with diverse polarity, amphiphilicity, and acid-base properties by solid-phase synthesis and investigated their in vitro antibacterial activity and renal cytotoxicity. We also assessed the structure-activity relationship and structure-toxicity relationship of the cyclic lipopeptide compounds. Some compounds showed increased activity against Gram-negative and/or Gram-positive bacteria, with improved renal cytotoxicity. C-02 showed remarkable in vitro antibacterial activity and low renal cytotoxicity. We found that C-02 showed high antibacterial activity against Escherichia coli in vivo and manifested its effects preliminarily by increasing outer membrane permeability. Therefore, C-02 might be a new antibiotic lead compound with not only high efficacy but also low renal cytotoxicity.

Sodium (Na) metal is considered a promising anode material for high-energy Na batteries due to its high theoretical capacity and abundant resources. However, uncontrollable dendrite growth during the repeated Na plating/stripping process leads to the issues of low Coulombic efficiency and short circuits, impeding the practical applications of Na metal anodes. Herein, we propose a silver-modified carbon nanofiber (CNF@Ag) host with asymmetric sodiophilic features to effectively improve the deposition behavior of Na metal. Both density functional theory (DFT) calculations and experiment results demonstrate that Na metal can preferentially nucleate on the sodiophilic surface with Ag nanoparticles and uniformly deposit on the whole CNF@Ag host with a "bottom-up growth" mode, thus preventing unsafe dendrite growth at the anode/separator interface. The optimized CNF@Ag framework exhibits an excellent average Coulombic efficiency of 99.9% for 500 cycles during Na plating/stripping at 1 mA cm -2 for 1 mAh cm -2 . Moreover, the CNF@Ag-Na symmetric cell displays stable cycling for 500 h with a low voltage hysteresis at 2 mA cm -2 . The CNF@Ag-Na//Na 3 V 2 (PO 4 ) 3 full cell also presents a high reversible specific capacity of 102.7 mAh g -1 for over 200 cycles at 1 C. Therefore, asymmetric sodiophilic engineering presents a facile and efficient approach for developing high-performance Na batteries with high safety and stable cycling performance.

The development of methods toward the construction of quaternary carbon centers has been a hot topic in recent years. In this work, an Au(I)-catalyzed intramolecular cyclization/semipinacol rearrangement of allene-containing allylic silyl ether was developed to provide a direct strategy for the construction of multisubstituted cyclohexene-type compounds with a quaternary carbon center in moderate to good yields. In particular, this method provides an alternative synthetic strategy for the construction of a multisubstituted spirocyclo[4.5]decane skeleton and may be applied to the synthesis of related bioactive molecules and their derivatives, thus facilitating the corresponding functional studies.

In this paper, 26 natural polymyxin components and a new derivative S 2 were synthesized, and their differences in efficacy and toxicity have been investigated. Almost all of the synthesized components showed strong activity against both susceptible and resistant strains of E. coli, K. pneumoniae, P. aeruginosa, and A. baumannii. The toxicities were obviously different between the components. Only some of the components were tested for toxicity in vivo. Compounds E 2 , E 2 -Val, A 2 , M 2 , D 2 , and S 2 showed obviously lower renal cytotoxicity and acute toxicity than polymyxins B and E. The in vivo nephrotoxicity of E 2 , M 2 , and S 2 was similar to that of polymyxin E. Compound S 2 , with four positive charges, was especially interesting as it possessed both increased efficacy and decreased toxicity. The SAR and toxicity studies indicated that further structural modification could concentrate on polymyxin S. The results also indicated that S 2 could be a new drug candidate.

We present a method for synthesizing MoS(2)/Graphene hybrid heterostructures with a growth template of graphene-covered Cu foil. Compared to other recent reports, (1, 2) a much lower growth temperature of 400 °C is required for this procedure. The chemical vapor deposition of MoS(2) on the graphene surface gives rise to single crystalline hexagonal flakes with a typical lateral size ranging from several hundred nanometers to several micrometers. The precursor (ammonium thiomolybdate) together with solvent was transported to graphene surface by a carrier gas at room temperature, which was then followed by post annealing. At an elevated temperature, the precursor self-assembles to form MoS(2) flakes epitaxially on the graphene surface via thermal decomposition. With higher amount of precursor delivered onto the graphene surface, a continuous MoS(2) film on graphene can be obtained. This simple chemical vapor deposition method provides a unique approach for the synthesis of graphene heterostructures and surface functionalization of graphene. The synthesized two-dimensional MoS(2)/Graphene hybrids possess great potential toward the development of new optical and electronic devices as well as a wide variety of newly synthesizable compounds for catalysts.

Phloem-mobile insecticides are preferred to achieve economically useful activity. However, only a few phloem-mobile synthetic insecticides are available. One approach to converting nonmobile insecticides into phloem-mobile types is introducing sugar to the parent compound. To test whether the addition of a glucose group to a non-phloem-mobile insecticide enables conversion into phloem-mobile, N-[3-cyano-1-[2,6-dichloro-4-(trifluoromethyl)phenyl]-4-[(trifluoromethyl)sulfinyl]-1H-pyrazol-5-yl]-1-(β-D-glucopyranosyl)-1H-1,2,3-triazole-4-methanamine (GTF) was prepared through click chemistry. A phloem-mobility test in Ricinus communis L. seedlings confirmed that GTF was mobile in the sieve tubes. Although GTF exhibited lower insecticidal activity against the third-instar larvae of Pzlutella xylostella than fipronil did, it can be reconverted into fipronil in adult plants of castor bean, thereby offsetting the decrease of insecticidal activity. Therefore, the presence of a glucose core confers phloem mobility to fipronil.

In this contribution we demonstrate a method of synthesizing a hexagonal boron nitride (h-BN) thin film by ambient pressure chemical vapor deposition on polycrystalline Ni films. Depending on the growth conditions, the thickness of the obtained h-BN film is between ∼5 and 50 nm. The h-BN grows continuously on the entire Ni surface and the region with uniform thickness can be up to 20 μm in lateral size which is only limited by the size of the Ni single crystal grains. The hexagonal structure was confirmed by both electron and X-ray diffraction. X-ray photoelectron spectroscopy shows the B/N atomic ratio to be 1:1.12. A large optical band gap (5.92 eV) was obtained from the photoabsorption spectra which suggest the potential usage of this h-BN film in optoelectronic devices.
