Abstract

Substitution of smooth muscle myosin's phosphorylation site Ser-19 and Thr-18 with aspartic acid or alanine results in a negative charge that is required at a specific spatial position for myosin activation. An analysis of the effects of Thr-18 phosphorylation on myosin function uses a regulatory light chain mutant that contains Ser-substituted with either Asp or Als. Asp incorporation increases phosphorylation and myosin configuration whereas Ala-substituted light chain has no effect on ATPase.
