Research has been conducted on the paramagnetic lanthanide(III) complexes. The solution structure of these complexes has been determined via the use of NMR spectroscopy and the comparison of the complexes' solution and solid state structures has been performed and is presented.

Complexes between the Py(2)N(6)Ac(4) (H(4)L) ligand containing four carboxylate pendant arms and trivalent lanthanide ions have been synthesized, and structural studies have been made both in the solid state and aqueous solution. The crystal structures of the La, Ce, Sm, Tb, Dy, Ho, Er, Tm, and Lu complexes, with chemical formulas [LaH(2)L](NO(3)).3H(2)O (1), [Ce(4)L(2)](NO(3))(4).30H(2)O (2), [SmHL].EtOH.3H(2)O (5), [TbHL].EtOH.3H(2)O (8), [DyHL].2EtOH.2H(2)O (9), [HoHL].3H(2)O (10), [ErHL].EtOH.3H(2)O (11) [TmHL].EtOH.3H(2)O (12), and [LuHL].3H(2)O (14), have been determined by single-crystal X-ray crystallography. In the solid state, the complexes of the lighter lanthanide ions La(3+)-Dy(3+) show a 10-coordinated geometry close to a distorted bicapped antiprism, where the carboxylate pendants are situated alternatively above and below the best plane that contains the nitrogen donor atoms. The complexes of the heavier ions, Ho(3+)-Lu(3+), have a 9-coordinated geometry close to distorted tricapped trigonal prism, with one of the pendant carboxylate groups uncoordinated. The ligand is in a "twist-fold" conformation, where the twisting of the pyridine units is accompanied by an overall folding of the major ring of the macrocycle so that the pyridine nitrogen atoms and the metal are far from linear. The aqueous solution structures of the complexes were thoroughly characterized, the diamagnetic ones (La(3+) and Lu(3+)) by their COSY NMR spectra, and the paramagnetic complexes using a linear least-squares fitting of the (1)H LIS (lanthanide-induced shift) and LIR (lanthanide-induced relaxation) data with rhombic magnetic susceptibility tensors. The solution structures obtained for the La(3+)-Dy(3+) complexes (10-coordinate) and for the Tm(3+)-Lu(3+) complexes (9-coordinate) are in very good agreement with the corresponding crystal structures. However, the 10-coordinate structure is still exclusive in solution for the Ho(3+) complex and predominant for the Er(3+) complex.

Two linear polyamide conjugates of Gd(DTPA)2- were synthesized and characterized by high-resolution nuclear magnetic resonance (NMR) spectroscopy and size exclusion chromatography (SEC). DTPA was copolymerized with two different diamines, 1,6-hexanediamine and trans-1,4-cyclohexanediamine, yielding the polymers DTPA-HMD and DTPA-CHD, with low polydispersity. Their molecular flexibility in solution was studied using 13C spin-lattice relaxation time measurements, indicating that the cyclohexanediamine linking moiety of the DTPA-HMD polymer is more rigid than that of DTPA-CHD. The influence of the flexibility of the linking functionalities on the relaxivity of the Gd3+-DTPA-polymer conjugates was studied by water nuclear magnetic relaxation dispersion (NMRD). The relaxivity of the Gd(DTPA-CHD) polymer was only slightly higher than that of the Gd(DTPA-HMD) polymer, and only two times higher than the usual values for small Gd-DTPA-like chelates. The low relaxivities obtained for both polymers, much lower than expected from the polymer apparent molecular weights, result from their substantial residual flexibility, and also from a too long, nonoptimal, value of the inner-sphere water exchange rate. These polymeric compounds are also cleared very quickly from the blood of rats, indicating that they are of limited value as blood pool contrast agents for MRA.

We investigated the mechanism of competition between Li+ and Mg2+ in Li(+)-loaded human red blood cells (RBCs) by making 7Li and 31P NMR and fluorescence measurements. We used 7Li NMR relaxation times to probe Li+ binding to the human RBC membrane and ATP; an increase in Mg2+ concentration caused an increase in both 7Li T1 and T2 values in packed Li(+)-loaded RBCs, in suspensions of Li(+)-loaded RBC ghosts, in suspensions of Li(+)-containing RBC membrane, and in aqueous solutions of ATP, indicating competition between Li+ and Mg2+ for binding sites in the membrane and ATP. We found that increasing concentrations of either Li+ or Mg2+ in the presence of human RBC membrane caused an increase in the 31P NMR chemical shift anisotropy parameter, which describes the observed axially symmetric powder pattern, indicating metal ion binding to the phosphate groups in the membrane. Competition between Li+ and Mg2+ for phosphate groups in ATP and in the RBC membrane was also observed by both fluorescence measurements and 31P NMR spectroscopy at low temperature. The ratio of the stoichiometric binding constants of Mg2+ to Li+ to the RBC membrane was approximately 20; the ratio of the conditional binding constants in the presence of a free intracellular ATP concentration of 0.2 mM was approximately 4, indicating that Li+ competes for approximately 20% of the Mg(2+)-binding sites in the RBC membrane. Our results indicate that, regardless of the spectroscopic method used, Li+ competes with Mg2+ for phosphate groups in both ATP and the RBC membrane; the extent of metal ion competition for the phosphate head groups of the phospholipids in the RBC membrane is enhanced by the presence of ATP. Competition between Li+ and Mg2+ for anionic phospholipids or Mg(2+)-activated proteins present in cell membranes may constitute the basis of a general molecular mechanism for Li+ action in human tissues.

We used 7Li NMR spin-lattice (T1) and spin-spin (T2) relaxation time measurements to investigate the binding of Li+ in human red blood cell (RBC) suspensions. In RBCs containing 1.4 mM Li+, the intracellular 7Li NMR T2 relaxation value (0.30 +/- 0.03 s) was much smaller than the corresponding T1 value (6.0 +/- 0.1 s), yielding a ratio of T1 to T2 of 20. For 1.5 mM LiCl solutions whose viscosities were adjusted to 5 cP with glycerol, the values of the T1/T2 ratios were as follows: 49 for unsealed RBC membrane (2.0 mg of protein/mL); 4.4 for spectrin (1.9 mg/mL); 1.5 for 5.4 mM 2,3-bisphosphoglycerate (BPG); 2.2 for 2.7 mM carbonmonoxyhemoglobin (COHb); 1.6 for 2.0 mM ATP; and 1.2 for a 50/50% (v/v) glycerol-water mixture. Intracellular viscosity and the electric field gradients experienced by Li+ when traversing the spectrin-actin network therefore are not responsible for the large values of the T1/T2 ratios observed in Li(+)-loaded RBCs. We conclude that the RBC membrane is the major Li+ binding site in Li(+)-loaded RBCs (Kb = 215 +/- 36 M-1) and that the binding of Li+ to COHb, BPG, spectrin-actin, or ATP is weak. Partially relaxed 7Li NMR spectra of Li(+)-containing RBC membrane suspensions indicated the presence of two relaxation components, one broad and one narrow.(ABSTRACT TRUNCATED AT 250 WORDS)

Helical complexes formed between fd DNA and reductively methylated fd gene 5 protein were indistinguishable by electron microscopy from complexes formed with the nonmethylated protein. 13C NMR spectroscopy of 13C-enriched N epsilon, N epsilon-dimethyllsyl residues of the protein showed that three of these residues (Lys-24, Lys-46, and Lys-69) were selectively perturbed by binding of the oligomer d(pA)7. These were the same lysyl residues that we previously found to be most protected from methylation by binding of the protein to poly[r(U)] [Dick, L. R., Sherry, A. D., Newkirk, M. M., & Gray D. M. (1988) J. Biol. Chem. 263, 18864-18872]. Thus, these lysines are probably directly involved in the nucleic acid binding function of the protein. Negatively charged chelates of lanthanide ions were used to perturb the 13C NMR resonances of labeled lysyl and amino-terminal residues of the gene 5 protein. The terbium chelate was found to bind tightly (Ka approximately 10(5) M-1) to the protein with a stoichiometry of 1 chelate molecule per protein dimer. 13C resonances of Lys-24, Lys-46, and Lys-69 were maximally shifted by the terbium chelate and were maximally relaxed by the gadolinium chelate. Also, the terbium chelate was excluded by the oligomer d(pA)7. Computer fits of the induced chemical shifts of 13C resonances with those expected for various positions of the terbium chelate failed to yield a possible chelate binding site unless the chemical shift for Lys-24 was excluded from the fitting process.(ABSTRACT TRUNCATED AT 250 WORDS)
