Coherent phonon modes supported by plasmonic nanoparticles offer prospective applications in chemical and biological sensing. Whereas the characterization of these phonon modes often requires single-particle measurements, synthetic routes to narrow size distributions of nanoparticles permit ensemble investigations. Recently, the synthesis of highly monodisperse gold tetrahedral nanoparticles with tunable edge lengths and corner sharpnesses has been developed. Herein, we characterize a size series of these nanoparticles in colloidal dispersion via transient absorption spectroscopy to examine their mechanical and plasmonic responses upon photoexcitation. Oscillations of transient absorption signals are observed in the plasmon resonance and correspond to the lowest-order radial breathing modes of the nanoparticles, the frequencies of which are affected by the edge length and truncation of the corners. Homogeneous quality factor values ranging from 24 to 34 are observed for the oscillations that convey potential utility in mass-sensing and plasmon-exciton-coupling photonics schemes. Finite-difference time domain and finite element analysis calculations establish specific optically relevant phonon modes.

The practically unlimited high-dimensional composition space of high-entropy materials (HEMs) has emerged as an exciting platform for functional material design and discovery. However, the identification of stable and synthesizable HEMs and robust design rules remains a daunting challenge. Here, we propose a mixed enthalpy-entropy descriptor (MEED) that enables highly efficient, robust, high-throughput prediction of synthesizable HEMs across vast chemical spaces from first-principles. The MEED is based on two parameters: the relative formation enthalpy with respect to the most stable competing compound and the spread of the point-defect formation energy spectrum. The former measures the relative synthesizability of an HEM to its most stable competing phase, going beyond the conventional thermodynamic understanding. The latter gauges the relative entropy forming ability of an HEM, entailing no sampling over numerous alloy configurations. By applying the MEED to two structurally distinct representative material systems (i.e., 3D rocksalt carbides and 2D layered sulfides), we not only successfully identify all experimentally reported HEMs within these systems but also reveal a cutoff criterion for assessing their relative synthesizability within each system. By the MEED, tens of new high-entropy carbides and 2D high-entropy sulfides are also predicted, which have the potential for a wide variety of applications such as coating in aerospace devices, energy conversion and storage, and flexible electronics.

Closely related protein families evolved from common ancestral genes present a significant hurdle in developing member- and isoform-specific chemical probes, owing to their similarity in fold and function. In this piece of work, we explore an allele-specific chemical rescue strategy to activate a "dead" variant of a wildtype protein using synthetic cofactors and demonstrate its successful application to the members of the alpha-ketoglutarate (αKG)-dependent histone demethylase 4 (KDM4) family. We show that a mutation at a specific residue in the catalytic site renders the variant inactive toward the natural cosubstrate. In contrast, αKG derivatives bearing appropriate stereoelectronic features endowed the mutant with native-like demethylase activity while remaining refractory to a set of wild type dioxygenases. The orthogonal enzyme-cofactor pairs demonstrated site- and degree-specific lysine demethylation on a full-length chromosomal histone in the cellular milieu. Our work offers a strategy to modulate a specific histone demethylase by identifying and engineering a conserved phenylalanine residue, which acts as a gatekeeper in the KDM4 subfamily, to sensitize the enzyme toward a novel set of αKG derivatives. The orthogonal pairs developed herein will serve as probes to study the role of degree-specific lysine demethylation in mammalian gene expression. Furthermore, this approach to overcome active site degeneracy is expected to have general application among all human αKG-dependent dioxygenases.

Here, we enhanced the popular yeast display method by multiple rounds of DNA and protein engineering. We introduced surface exposure-tailored reporters, eUnaG2 and DnbALFA, creating a new platform of C and N terminal fusion vectors. The optimization of eUnaG2 resulted in five times brighter fluorescence and 10 °C increased thermostability than UnaG. The optimized DnbALFA has 10-fold the level of expression of the starting protein. Following this, different plasmids were developed to create a complex platform allowing a broad range of protein expression organizations and labeling strategies. Our platform showed up to five times better separation between nonexpressing and expressing cells compared with traditional pCTcon2 and c-myc labeling, allowing for fewer rounds of selection and achieving higher binding affinities. Testing 16 different proteins, the enhanced system showed consistently stronger expression signals over c-myc labeling. In addition to gains in simplicity, speed, and cost-effectiveness, new applications were introduced to monitor protein surface exposure and protein retention in the secretion pathway that enabled successful protein engineering of hard-to-express proteins. As an example, we show how we optimized the WD40 domain of the ATG16L1 protein for yeast surface and soluble bacterial expression, starting from a nonexpressing protein. As a second example, we show how using the here-presented enhanced yeast display method we rapidly selected high-affinity binders toward two protein targets, demonstrating the simplicity of generating new protein-protein interactions. While the methodological changes are incremental, it results in a qualitative enhancement in the applicability of yeast display for many applications.

Herein we introduce a transition-metal-free protocol that involves a commercially available, inexpensive pyrazole molecule to conduct C-C cross-coupling reactions at room temperature via a radical pathway. Using this method, an aryldiazonium salt has been coupled to a wide range of arenes and heteroarenes including benzene, mesitylene, thiophene, furan, benzoxazole to result the corresponding biaryl products. The full reaction mechanism is elucidated along with the crystallographic probation of an active initiator species. A potassium-stabilized deprotonated pyrazole steers single-electron transfer to the substrate and behaves as an initiator for the reaction.

The emergence of new viral infections and drug-resistant bacteria urgently necessitates expedient therapeutic development. Repurposing and redesign of existing drugs against different targets are one potential way in which to accelerate this process. Suramin was initially developed as a successful antiparasitic drug but has also shown promising antiviral and antibacterial activities. However, due to its high conformational flexibility and negative charge, suramin is considered quite promiscuous toward positively charged sites within nucleic acid binding proteins. Although some suramin analogs have been developed against specific targets, only limited structure-activity relationship studies were performed, and virtual screening has yet to be used to identify more specific inhibitor(s) based on its scaffold. Using available structures, we investigated suramin's target diversity, confirming that suramin preferentially binds to protein pockets that are both positively charged and enriched in aromatic or leucine residues. Further, suramin's high conformational flexibility allows adaptation to structurally diverse binding surfaces. From this platform, we developed a framework for structure- and docking-guided elaboration of suramin analog scaffolds using virtual screening of suramin and heparin analogs against a panel of diverse therapeutically relevant viral and bacterial protein targets. Use of this new framework to design potentially specific suramin analogs is exemplified using the SARS-CoV-2 RNA-dependent RNA polymerase and nucleocapsid protein, identifying leads that might inhibit a wide range of coronaviruses. The approach presented here establishes a computational framework for designing suramin analogs against different bacterial and viral targets and repurposing existing drugs for more specific inhibitory activity.

We present a comprehensive theory of the magnetic phases in twisted bilayer chromium trihalides through a combination of first-principles calculations and atomistic simulations. We show that the stacking-dependent interlayer exchange leads to an effective moiré field that is mostly ferromagnetic with antiferromagnetic patches. A wide range of noncollinear magnetic phases can be stabilized as a function of the twist angle and Dzyaloshinskii-Moriya interaction as a result of the competing interlayer antiferromagnetic coupling and the energy cost for forming domain walls. In particular, we demonstrate that for small twist angles various skyrmion crystal phases can be stabilized in both CrI 3 and CrBr 3 . Our results provide an interpretation for the recent observation of noncollinear magnetic phases in twisted bilayer CrI 3 and demonstrate the possibility of engineering further nontrivial magnetic ground states in twisted bilayer chromium trihalides.

Conditional remodeling of enzyme catalysis is a formidable challenge in protein engineering. Herein, we have undertaken a unique active site engineering tactic to command catalytic outcomes. With ten-eleven translocation (TET) enzyme as a paradigm, we show that variants with an expanded active site significantly enhance multistep C-H oxidation in 5-methylcytosine (5mC), whereas a crowded cavity leads to a single-step catalytic apparatus. We further identify an evolutionarily conserved residue in the TET family with a remarkable catalysis-directing ability. The activating variant demonstrated its prowess to oxidize 5mC in chromosomal DNA for potentiating expression of genes including tumor suppressors.

Several toll-like receptors (TLRs) reside inside endosomes of specific immune cells-among them, aberrant activation of TLR7 and TLR9 is implicated in myriad contexts of autoimmune diseases, making them promising therapeutic targets. However, small-molecule TLR7 and TLR9 antagonists are not yet available for clinical use. We illustrate here the importance of C2, C6, and N9 substitutions in the purine scaffold for antagonism to TLR7 and TLR9 through structure-activity relationship studies using cellular reporter assays and functional studies on primary human immune cells. Further in vitro and in vivo pharmacokinetic studies identified an orally bioavailable lead compound 29 , with IC 50 values of 0.08 and 2.66 μM against TLR9 and TLR7, respectively. Isothermal titration calorimetry excluded direct TLR ligand-antagonist interactions. In vivo antagonism efficacy against mouse TLR9 and therapeutic efficacy in a preclinical murine model of psoriasis highlighted the potential of compound 29 as a therapeutic candidate in relevant autoimmune contexts.

Membrane fusion, a key step in the early stages of virus propagation, allows the release of the viral genome in the host cell cytoplasm. The process is initiated by fusion peptides that are small, hydrophobic components of viral membrane-embedded glycoproteins and are typically conserved within virus families. Here, we attempted to identify the correct fusion peptide region in the Spike protein of SARS-CoV-2 by all-atom molecular dynamics simulations of dual membrane systems with varied oligomeric units of putative candidate peptides. Of all of the systems tested, only a trimeric unit of a 40-amino-acid region (residues 816-855 of SARS-CoV-2 Spike) was effective in triggering the initial stages of membrane fusion, within 200 ns of simulation time. Association of this trimeric unit with dual membranes resulted in the migration of lipids from the upper leaflet of the lower bilayer toward the lower leaflet of the upper bilayer to create a structural unit reminiscent of a fusion bridge. We submit that residues 816-855 of Spike represent the bona fide fusion peptide of SARS-CoV-2 and that computational methods represent an effective way to identify fusion peptides in viral glycoproteins.

A theoretical study on the coupled electron-nuclear dynamics of HD + molecular ions under ultrashort, intense laser pulses is performed by employing a well-established quasi-classical model. The influence of the laser carrier-envelope phase on various channel (H + D + , D + H + , and H + + D + ) probabilities is investigated at different laser field intensities. The carrier-envelope phase is found to govern the dissociation (H + D + and D + H + ) and Coulomb explosion (H + + D + ) channel probabilities. The kinetic energy release distributions of the fragments are also found to be sensitive to the carrier-envelope phase of the laser pulse. Our results are in agreement with the previously reported quantum dynamics studies and experiments.

Photochemistry induced by phase-coherent laser light is an intriguing topic. The possibility of weak-field (one-photon) phase-only control of photoisomerization is controversial. Experimental studies on the weak-field coherent control of cis-trans isomerization have led to conflicting results. Here we address this issue by quantum dynamical calculations, focusing on a mechanism where different "phase-shaped" wave packets are quickly stabilized ("dumped") in the trans configuration because of prompt energy dissipation. We systematically investigate different relaxation rates with the system-bath dynamics described within the time-dependent Hartree approximation leading to a friction-type force. We find evidence for phase control of trans-isomer yields (about 5%) in this model with pure energy dissipation given sufficiently strong dampening.

One of the holy grails of contemporary science is to understand and manipulate chemical reactions to obtain desired products preferentially. To achieve this goal, chemists traditionally choose the correct starting materials and reaction conditions, but it often lacks selectivity and efficiency. A promising alternative is to design laser control schemes and apply them to guide and control chemical reactions. A concise synthetic strategy to 5-dihydroxyboryldexoyuridine (5boU) phosphoramidite has been developed. 5boU was introduced into short oligonucleotides in a site-specific manner, demonstrating compatibility of the boronic acid moiety with standard solid-phase DNA synthesis chemistry. Electrophilic 5boU DNAs inhibited thymine DNA glycosylase, a cancer-relevant DNA-modifying enzyme. We envisage diverse applications of 5boU in organic synthesis, medicinal chemistry, and chemical biology.

A theoretical study on the ionization dynamics of carbon atom irradiated with a few-cycle, intense laser field is performed within a quasiclassical model to get mechanistic insights into an earlier reported carrier-envelope phase dependency of ionization probabilities of an atom [ Phys. Rev. Lett. 2013, 110, 083602]. The carrier-envelope phase of the laser pulse is found to govern the overall dynamics, reflecting its importance in controlling electronic motion. To understand the origin of this effect, individual trajectories were analyzed at a particular laser intensity. We found that a variation in the carrier-envelope phase affects the angle of ejection of the electrons and subsequently the attainment of the desired final state.

Quantum dynamical behavior of H 2 + in the presence of a linearly polarized, ultrashort, intense, infrared laser pulse has been studied by numerically solving the time-dependent Schrödinger equation with nuclear motion restricted in one-dimension along the direction of laser polarization and electronic motion in three-dimensions. On the basis of the time-dependent Born-Oppenheimer approximation, we have constructed time-dependent potentials for the ground electronic state (1sσ g ) of H 2 + . Subsequent nuclear dynamics is then carried out on these field-dressed potential energy surfaces, and the dissociation dynamics is investigated. Our analyses reveal that although the electronic longitudinal degree of freedom plays the major role in governing the dissociation dynamics, contributions from the electronic transverse degree of freedom should also have to be taken into account to obtain accurate results. Also, modeling electron-nuclei Coulomb interactions in a one-dimensional calculation with an artificially chosen constant softening parameter leads to a discrepancy with the exact results. Comparing our results with other quantum and classical dynamical studies showed a good agreement with exact results.

Oxidative C-H hydroxylation of methyl groups, followed by their removal from DNA, RNA, or histones, is an epigenetic process critical to transcriptional reprogramming and cell fate determination. This reaction is catalyzed by Fe(II)-dependent dioxygenases using the essential metabolite 2-ketoglutarate (2KG) as a cofactor. Given that the human genome encodes for more than 60 2KG-dependent dioxygenases, assigning their individual functions remains a significant challenge. Here we describe a protein-ligand interface engineering approach to break the biochemical degeneracy of these enzymes. Using histone lysine demethylase 4 as a proof-of-concept, we show that the enzyme active site can be expanded to employ bulky 2KG analogues that do not sensitize wild-type demethylases. We establish the orthogonality, substrate specificity, and catalytic competency of the engineered demethylation apparatus in biochemical assays. We further demonstrate demethylation of cognate substrates in physiologically relevant settings. Our results provide a paradigm for rapid and conditional manipulation of histone demethylases to uncloak their isoform-specific functions.

Preferential breaking of chemical bonds using few-cycle, intense laser pulse to obtain desired products offer a formidable challenge in understanding ultrafast chemical reactivity. In a recent study [J. Chem. Phys. 2015, 143, 244310], it was found that carrier-envelope phase influences the bond-selective fragmentation in HOD with up to 3-fold enhancement. We present a detailed theoretical study to understand the influence of initial vibrational states governing the dissociation dynamics. We have carried out a time-dependent quantum mechanical wave packet study on the ground electronic state (X̃ (3)B1) of HOD(2+). Analytical potential energy surface for the ground electronic states of both the neutral molecule and dication has been developed at multireference configuration interaction level of theory with aug-cc-pVQZ basis set. Branching ratio is computed from the accumulated flux in H(+) + OD(+) and D(+) + OH(+) dissociation channels. Our investigation demonstrate a strong dependency on the initial conditions, and thereby preferential cleavage of bonds can be achieved. We have also compared our results with experimental and other theoretical studies.

A study on self-assembly of anisotropically substituted penta-aryl fullerenes in water has been reported. The penta-phenol-substituted amphiphilic fullerene derivative [C60Ph5(OH)5] exhibited self-assembled vesicular nanostructures in water under the experimental conditions. The size of the vesicles was observed to depend upon the kinetics of self-assembly and could be varied from ∼300 to ∼70 nm. Our mechanistic study indicated that the self-assembly of C60Ph5(OH)5 is driven by extensive intermolecular as well as water-mediated hydrogen bonding along with fullerene-fullerene hydrophobic interaction in water. The cumulative effect of these interactions is responsible for the stability of vesicular structures even on the removal of solvent. The substitution of phenol with anisole resulted in different packing and interaction of the fullerene derivative, as indicated in the molecular dynamics studies, thus resulting in different self-assembled nanostructures. The hollow vesicles were further encapsulated with a hydrophobic conjugated polymer and water-soluble dye as guest molecules. Such confinement of π-conjugated polymers in fullerene has significance in bulk heterojunction devices for efficient exciton diffusion.

The basic requirement for understanding the nonviral gene delivery pathway is a thorough biophysical characterization of DNA polyplexes. In this work, we have studied the interactions between calf-thymus DNA (ctDNA)and a new series of linear cationic block copolymers (BCPs). The BCPs were synthesized via controlled radical polymerization using [3-(methacryloylamino)propyl] -trimethylammonium chloride (MAPTAC) and poly(ethyleneglycol) methyl ether (PEGMe) as comonomers. UV−visible spectroscopy, ethidium bromide dye exclusion, and gel electrophoresis study revealed that these cationic BCPs were capable of efficiently binding with DNA. Steady-state fluorescence, UV melting, gel electrophoresis, and circular dichroism results suggested increased binding for BCPs containing higher PEG. Hydrophobic interactions between the PEG and the DNA base pairs became significant at close proximity of the two macromolecules, thereby influencing the binding trend. DLS studies showed a decrease in the size of DNA molecules at lower charge ratio (the ratio of “+” charge of the polymer to “−” charge of DNA) due to compaction, whereas the size increased at higher charge ratio due to aggregation among the polyplexes. Additionally, we have conducted kinetic studies of the binding process using the stop-flow fluorescence method. All the results of BCP−DNA binding studies suggested a two-step reaction mechanism--a rapid electrostatic binding between the cationic blocks and DNA, followed by a conformational change of the polyplexes in the subsequent step that led to DNA condensation. The relative rate constant(k'(1)) of the first step was much higher compared to that of the second step (k'(2)), and both were found to increase with an increase in BCP concentration. The charge ratios as well as the PEG content in the BCPs had a marked effect on the kinetics of the DNA−BCP polyplex formation. Introduction of a desired PEG chain length in the synthesized cationic blocks renders them potentially useful as nonviral gene delivery agents.

Formation and disintegration of self-assembled nanostructures in response to external stimuli are important phenomena that have been widely explored for a variety of biomedical applications. In this contribution, we report the thermally triggered assembly of block copolymer molecules in aqueous solution to form vesicles (polymersomes) and their disassembly on reduction of temperature. A new thermoresponsive diblock copolymer of poly(N-isopropylacrylamide) poly((3-methacrylamidopropyl)trimethylammonium chloride) (PNIPA-b-PMAPTAC) was synthesized by reversible addition-fragmentation chain transfer technique. The solution properties and self-assembling behavior of the block copolymer molecules were studied by turbidimetry, temperature-dependent proton nuclear magnetic resonance, fluorescence spectroscopy, dynamic light scattering, and transmission electron microscopy. Fluorescence resonance energy transfer studies between coumarin-153 (C-153, donor) and rhodamine 6G (R6G, acceptor) have been performed by steady-state and picosecond-resolved fluorescence spectroscopy to probe the structural and dynamic heterogeneity of the vesicles. The occurrence of efficient energy transfer was evident from the shortening of donor lifetime in the presence of the acceptor. The capability of the vesicles to encapsulate both hydrophobic and hydrophilic molecules and release them in response to decrease in temperature makes them potentially useful as drug delivery vehicles.

Poly(amidoamine) (PAMAM) dendrimers are known to be highly efficient nonviral carriers in gene delivery. Dendrimer-mediated transfection is known to be a function of the dendrimer to DNA charge ratio as well as the size of the dendrimer. In the present study, the binding kinetics of four PAMAM dendrimers (G1, G2, G3, and G4) with calf thymus DNA (CT-DNA) has been studied using stopped-flow fluorescence spectroscopy. The effect of dendrimer-to-DNA charge ratio and dendrimer generation on the binding kinetics was investigated. In most cases, the results of dendrimer-CT-DNA binding can be explained by a two-step reaction mechanism: a rapid electrostatic binding between the dendrimer and DNA, followed by a conformational change of the dendrimer-DNA complex that ultimately leads to DNA condensation. It was observed that the charge ratio on the dendrimer and the DNA phosphate groups, as well as the dendrimer generation (size), has a marked effect on the kinetics of binding between the DNA and the dendrimers. The rate constant (k'1) of the first step was much higher compared to that of the second step (k'2), and both were found to increase with an increase in dendrimer concentration. Among the four generations of dendrimers, G4 exhibited significantly faster binding kinetics compared to the three smaller generation dendrimers.

A methodology involving a "transition metal-free" intramolecular biaryl-coupling of o-halo-N-arylbenzylamines has been developed in the presence of potassium tert-butoxide and an organic molecule as catalyst. The reaction appears to proceed through KO(t)Bu-promoted intramolecular homolytic aromatic substitution (HAS). Interestingly, this biaryl coupling also works in the presence of potassium tert-butoxide as sole promoter. On extending our approach further, we found that N-acyl 2-bromo-N-arylbenzylamines undergo a one-pot N-deprotection/biaryl coupling followed by oxidation, thus offering an expeditious route to the phenanthridine and benzo[c]phenanthridine skeletons. The strategy has been applied to a concise synthesis of Amaryllidaceae alkaloids viz. oxoassoanine (1b), anhydrolycorinone (1d), 5,6-dihydrobicolorine (2d), trispheridine (2b), and benzo[c]phenanthridines alkaloids dihydronitidine (3b), dihydrochelerythidine (3d), dihydroavicine (3f), nornitidine (3h), and norchelerythrine (3j).

An efficient route to 2-oxindoles bearing an all-carbon quaternary center at the pseudobenzylic position has been developed via a DDQ-mediated Intramolecular-Dehydrogenative-Coupling (IDC). The methodology involves a one-pot C-alkylation of β-N-arylamido esters (7) concomitant with dehydrogenative-coupling in the presence of stoichiometric amount of DDQ. A tentative mechanistic route has been proposed for the oxidative coupling. The methodology provides a two-step entry to the ergoline structure of ergot alkaloids.

Fluorescence in poly(ethylene glycol) (PEGs 400-12000) solutions is reported here for the first time. PEG solutions form a vesicular organization with the hydrophilic groups attached at both ends which arrange themselves beyond a particular concentration and offer electron-dense regions at the center of the vesicle. These vesicles provide an inherent site for fluorescence generation in PEG solutions. Fluorescence emission was observed at ~380 nm with an excitation wavelength of 300 nm. PEG of molecular weight 6000 was found to show maximum emission intensity at a particular concentration. The formation of PEG vesicles (~1 nm size) was confirmed by dynamic light scattering (DLS) and confocal laser microscopy. On addition of metal ions the polymeric vesicle breaks up to monomeric PEG, and hence, the fluorescence intensity decreases with a red shift. Fluorescence lifetime measurements indicate the nature of complexation of the metals with PEG. Since PEGs are used as one of the phases in aqueous biphasic systems (ABS) of liquid-liquid extractions, the nature of the fluorescence emission spectrum of the PEG phase after extraction was studied. Metal extraction in the PEG-rich phase of an ABS leads to quenching of fluorescence in PEG.

We report a type of infrared switchable plasmonic quantum cascade laser, in which far field light in the midwave infrared (MWIR, 6.1 μm) is modulated by a near field interaction of light in the telecommunications wavelength (1.55 μm). To achieve this all-optical switch, we used cross-polarized bowtie antennas and a centrally located germanium nanoslab. The bowtie antenna squeezes the short wavelength light into the gap region, where the germanium is placed. The perturbation of refractive index of the germanium due to the free carrier absorption produced by short wavelength light changes the optical response of the antenna and the entire laser intensity at 6.1 μm significantly. This device shows a viable method to modulate the far field of a laser through a near field interaction.

We present spatial mapping of optical force generated near the hot spot of a metal-dielectric-metal bowtie nanoantenna at a wavelength of 1550 nm. Maxwell's stress tensor method has been used to simulate the optical force and it agrees well with the experimental data. This method could potentially produce field intensity and optical force mapping simultaneously with a high spatial resolution. Detailed mapping of the optical force is crucial for understanding and designing plasmonic-based optical trapping for emerging applications such as chip-scale biosensing and optomechanical switching.

The photophysical behavior of acridine (Acr) shows facilitated water-assisted protonation equilibrium between its deprotonted (Acr* ∼ 10 ns) and protonated forms (AcrH(+*) ∼ 28 ns) within confined region of ordered water molecules inside AOT/H(2)O/n-heptane reverse micelles (RMs). The time-resolved-area-normalized-emission spectra confirm both Acr* and AcrH(+*), while time-resolved-emission spectra depict time evolution between them. Quenching of AcrH(+*) with N,N-dimethylaniline (DMA) is a purely diffusion-controlled bimolecular quenching with linear Stern-Volmer (S-V) plot, while nonlinearity arises with triethylamine (TEA) that forms ground state complex with AcrH(+) (AcrH(+)··H(2)O··TEA) indicating both static and dynamic quenching. Transient intermediates, DMA(•+) and AcrH(•) infer photoinduced electron transfer from DMA to Acr, while those from AcrH(+)··H(2)O··TEA complex suggest water mediated excited-state proton transfer (ESPT) between AcrH(+) and TEA. The ESPT becomes faster in larger RMs due to enhanced mobility of hydronium ions in AcrH(+)··H(2)O··TEA, which reduces in smaller RMs as water becomes much more constrained owing to stronger complexation by excess confinement.

Photoinduced intramolecular electron transfer occurs in the triplet state within the complex [Htyr-Cu-phen](+) (Htyr = l-tyrosinato; phen = 1,10-phenanthroline) from tyrosine to phenanthroline. For this linked donor-acceptor system, a prominent magnetic field effect (MFE) is observed for the triplet-born radicals. The competitive binding study in the presence of ethidium bromide suggests that the complex interacts with calf thymus DNA (CT DNA) through partial intercalation. The photoexcited copper complex can oxidize DNA in a deoxygenated environment. Though the oxidation of tyrosine is thermodynamically more favorable than the oxidation of guanine, the primary electron transfer occurs from the DNA base to the phen ligand. A prominent MFE is observed for this noncovalently bound triplet-born guanine radical and phen radical anion. The process of partial intercalation of the copper complex within DNA is responsible for this rare observation.

A wide range of vinyl sulfone-modified carbohydrates have been prepared as starting materials for the synthesis of polysubstituted chiral pyrroles. All these vinyl sulfones reacted efficiently with ethylisocyanoacetate to generate a plethora of new pyrrole derivatives. Furanosyl rings opened up during pyrrole synthesis, and pyranosyl rings were opened up by reacting the pyrrole with POCl(3)/DMF. This paper also reports one of the most efficient and practical routes for the synthesis of beta-substituted pyrroles.

Laser flash photolysis and an external magnetic field have been used to study the interaction of two quinone molecules, namely, 9,10-anthraquinone (AQ) and 2-methyl 1,4-naphthoquinone, commonly known as menadione (MQ), with one of the DNA bases, guanine (G) and its nucleoside guanosine hydrate (dG). In organic homogeneous medium, it has been observed that G undergoes a predominant hydrogen (H) abstraction reaction with both the quinones while dG supports photoinduced electron transfer (PET) along with H abstraction. On the other hand, in SDS medium, G supports PET with AQ but not with MQ. However, behavior of dG remains unperturbed toward AQ and MQ with the change in medium. All of these observations have been explained on the basis of stabilization of radical ion pair and difference in size of the quinones, which can affect the distance of approach among the interacting molecules.

The magnetic field effect (MFE) on the photoinduced electron transfer (PET) reaction between the [Cu(phen)2]2+ complex and DNA has been studied in homogeneous buffer medium and in reverse micelles. The copper complex on photoexcitation can oxidize DNA in a deoxygenated environment. A prominent MFE is found even in a homogeneous aqueous medium for the triplet born radicals. The process of partial intercalation of [Cu(phen)2]2+ complex within DNA is responsible for such a rare observation. In reverse micelles, the MFE is not very much prominent because of the large separation distance between the component radicals of the geminate radical ion pairs generated through PET.

A derivative of phenazine, dibenzo[a,c]phenazine (DBPZ), can be used as a very good hydrogen-bonding probe unlike its parent phenazine molecule. Steady-state absorption and fluorescence studies reveal that DBPZ is completely insensitive to polarity of the medium. However, DBPZ can form a hydrogen bond very efficiently in its first excited singlet state. The extent of this excited-state hydrogen-bond formation depends both on size and on hydrogen-bond donor ability of the solvents. Time-resolved fluorescence studies and theoretical calculations also suggest that this hydrogen-bond formation is much more favorable in the excited state as compared to the ground state. In the excited state, the electron density is pushed toward the nitrogen atoms from the benzene rings, thereby increasing the dipole moment of the DBPZ molecule. Although the dipole moment of DBPZ increases upon photoexcitation, like other polarity probes, the molecule remains fully insensitive to the polarity of the interacting solvent. This unusual behavior of DBPZ as compared to simple phenazine and other polarity probes is due to the structure of the molecule. Hydrogen atoms at the 1 and 8 positions of DBPZ are sterically interacting with a lone pair of electrons on the proximate nitrogen atoms and make both of the nitrogen atoms inaccessible to solvent molecules. For this reason, DBPZ cannot sense the polarity of the medium. However, DBPZ can only sense solvents, those that have hydrogen with some electropositive nature, that is, the hydrogen-bond donating solvents. Hydrogen being the smallest among all elements can only interact with the lone pair of electrons of nitrogen atoms. Thus, DBPZ can act as a sensor for the hydrogen-bond donating solvents irrespective of their dielectrics.

Unlike the simple phenazine (PZ) molecule, one of its derivatives, dibenzo[a,c]phenazine (DBPZ) forms a charge-transfer complex in the triplet state (3ECT) with different amines, e.g., N,N-dimethylaniline (DMA), 4,4'-bis(dimethylamino)diphenylmethane (DMDPM), and triethylamine (TEA). Formation of the 3ECT and radical ion pairs (RIPs) due to electron transfer is identified by laser flash photolysis. The RIPs are much more abundant in the cases of DMA and DMDPM rather than in TEA. Interestingly, a prominent magnetic field effect (MFE) is observed in both the cases of 3ECT and RIPs in homogeneous acetonitrile-water (MeCN/H2O) mixtures. This rare observation of the 3ECT and MFE in non-viscous medium could be explained by considering the extended planar structure of DBPZ and inter-radical hydrogen bonding, mediated by the intervening water molecules. The magnetic field behavior is consistent with the hyperfine mechanism; however, the low B1/2 value for DBPZ-TEA system is ascribed to fast electron exchange due to the close proximity of the corresponding radical ions.
