Humans continuously learn new information. Here, we examined the temporal brain dynamics of explicit verbal associative learning between unfamiliar items. In the first experiment, 25 adults learned object-pseudoword associations during a 5-day training program allowing us to track the N400 dynamics across learning blocks within and across days. Successful learning was accompanied by an initial frontal N400 that decreased in amplitude across blocks during the first day and shifted to parietal sites during the last training day. In Experiment 2, we replicated our findings with 38 new participants randomly assigned to a consistent learning or an inconsistent learning group. The N400 amplitude modulations that we found, both within and between learning sessions, are taken to reflect the emergence of novel lexical traces even when learning concerns items for which no semantic information is provided. The shift in N400 topography suggests that different N400 neural generators may contribute to specific word learning steps through a balance between domain-general and language-specific mechanisms. Abstract: Picture naming covers the main stages of word production from concept retrieval to articulation. Cortical correlates of picture naming have been characterized with both haemodynamic and neurophysiological methods but the association of specific activation patterns with the hypothesized processing stages remains elusive. Here we used categorization tasks to selectively highlight different components of picture confrontation, from visual analysis (VIS) to semantic (SEM) and phonological access (PHON), and compared these time courses of activation with that obtained during picture naming (NAM). Brain activity was recorded with whole-head magnetoencephalography (MEG). Following the initially similar activation patterns in occipital and parietal areas, task effects (stronger activation in NAM/PHON than in SEM/VIS) emerged after 300 ms, in the sustained activation of the left posterior temporal and bilateral inferior frontal cortex, apparently reflecting enhancement of phonological and phonetic/articulatory processing. Abstract: The present case continues the series of anomia treatment studies with contextual priming (CP), being the second in-depth treatment study conducted for an individual suffering from semantically based anomia. Our aim was to acquire further evidence of the facilitation and interference effects of the CP treatment on semantic anomia. Based on the results of the study of , our hypothesis before the treatment was that our participant would show short-term interference and at most modest and short-term benefit from treatment. To acquire such evidence would not only be important for the choice of anomia treatment methods in individual patients, but would also prompt further development of the CP method. The CP technique used for our participant included cycles of repeating and naming items in three contextual conditions (semantic, phonological, and unrelated). As predicted, the overall improvement of naming was modest and short-term. Interestingly, the contextual condition that corresponded with the nature of our patient’s underlying naming deficit (semantic) elicited immediate interference in the form of contextual naming errors, as well as short-term improvement of naming. Based on this and a recent study by , it appears that despite short-term positive effects, in its current form the CP treatment is not sufficient for those aphasics who have a semantic deficit underlying their anomia. We tracked the evolvement of naming-related cortical dynamics with magnetoencephalography when five normal adults successfully learned names and/or meanings of unfamiliar objects. In all subjects, the learning of new names was associated with pronounced cortical effects. The learning effect was of long latency and emerged as a change of activation in the same cortical network that was active during naming of familiar items. In four out of five subjects, the cortical learning effect occurred in the inferior parietal lobe. In three of these subjects, the cortical effect was left-sided. These results suggest that the inferior parietal lobe plays an important role in the acquisition of novel words, presumably as a part of working memory systems. [Copyright &y& Elsevier]

Neuropsychological studies have demonstrated that the production of nouns and verbs can be dissociated in aphasia. These reports have been taken as evidence for separate representations of nouns and verbs in the human brain. We used whole-head magnetoencephalography to record cortical dynamics of action and object naming in 10 healthy adults and in 1 anomic patient with superior naming of verbs compared with nouns due to a left posterior parietal lesion. A single set of 100 line drawings was used for both action and object naming. In normal subjects, the activation sequences in action and object naming were essentially identical, advancing from the occipital to posterior temporoparietal and further to the left frontal cortex, without consistent involvement of the classical left inferior frontal (Broca) and temporal (Wernicke) language areas. In the anomic patient, pronounced differences between action and object naming emerged in the left hemisphere. The activation sequence was disrupted at the level of the damaged parietal cortex and did not reach the left frontal cortex even in the relatively easier action naming. The more severely impaired object naming was associated with exceptionally strong and early activation of the left inferior frontal cortex (Broca) and subsequent pronounced activation of the left middle temporal cortex, silent in action naming. Verb and noun retrieval thus utilized a spatiotemporally similar neuronal network in healthy individuals. A clear dissociation in cortical correlates of verb and noun retrieval only became evident in our anomic patient, in whom damage to the language network has resulted in disproportionately worse performance in object than action naming. • We examined lexical acquisition in aphasia using a brief novel word learning task. • Most people with aphasia (PWA) demonstrated learning on recall and recognition tests. • Phonological cues on recall testing provide a sensitive measure of learning ability. • Language, cognitive and lesion factors modulate verbal learning ability in PWA. • Brief effective word learning tasks may have clinical value for PWA when validated. Novel word learning ability has been associated with language treatment outcomes in people with aphasia (PWA), and its assessment could inform prognosis and rehabilitation. We used a brief experimental task to examine novel word learning in PWA, determine the value of phonological cueing in assessing learning outcomes, and identify factors that modulate learning ability. Twelve PWA and nineteen healthy controls completed the task, and recall and recognition tests of learning ability. Most PWA showed comparable learning outcomes to those of the healthy controls. Learning assessed via expressive recall was more clearly evidenced with phonological cues. Better single word processing abilities and phonological short-term memory and higher integrity of the left inferior frontal gyrus were related to better learning performance. Brief learning tasks like this one are clinically feasible and hold promise as screening tools of verbal learning in PWA once validated and evaluated for their capacity to predict treatment outcomes. This article presents the results of a study, which presents a new PET method that can be applied to the study of the cortical dynamics of a key neurotransmitter in the brain, dopamine. Regions of interest analysis showed an average decrease of 11.3% of the blood pressure in the ventrolateral frontal cortex and a 14% decrease in the left medial temporal lobe during the verbal working memory task compared to the vigilance task. Even though the ultimate mechanisms at the receptor level remain to be examined as also dopamine-mediated receptor internalization might be involved, the present results provide the first evidence on regional cortical dopaminergic activity in the healthy human brain during cognitive task performance.

Abstract: Despite considerable research interest, it is still an open issue as to how morphologically complex words such as “car + s” are represented and processed in the brain. We studied the neural correlates of the processing of inflected nouns in the morphologically rich Finnish language. Previous behavioral studies in Finnish have yielded a robust inflectional processing cost, i.e., inflected words are harder to recognize than otherwise matched morphologically simple words. Theoretically this effect could stem either from decomposition of inflected words into a stem and a suffix at input level and/or from subsequent recombination at the semantic–syntactic level to arrive at an interpretation of the word. To shed light on this issue, we used magnetoencephalography to reveal the time course and localization of neural effects of morphological structure and frequency of written words. Ten subjects silently read high- and low-frequency Finnish words in inflected and monomorphemic form. Morphological complexity was accompanied by stronger and longer-lasting activation of the left superior temporal cortex from 200 ms onwards. Earlier effects of morphology were not found, supporting the view that the well-established behavioral processing cost for inflected words stems from the semantic–syntactic level rather than from early decomposition. Since the effect of morphology was detected throughout the range of word frequencies employed, the majority of inflected Finnish words appears to be represented in decomposed form and only very high-frequency inflected words may acquire full-form representations. Abstract: By employing visual lexical decision and functional MRI, we studied the neural correlates of morphological decomposition in a highly inflected language (Finnish) where most inflected noun forms elicit a consistent processing cost during word recognition. This behavioral effect could reflect suffix stripping at the visual word form level and/or subsequent meaning integration at the semantic-syntactic level. The first alternative predicts increased activation for inflected vs. monomorphemic words in the left occipitotemporal cortex while the second alternative predicts left inferior frontal gyrus and/or left posterior temporal activation increases. The results show significant activation effects in the latter areas. This provides support for the second alternative, i.e., that the morphological processing cost stems from the semantic-syntactic level. [Copyright &y& Elsevier]

• fMRI and multivariate pattern analysis elucidate attention to lifelike AV speech. • Attention in cocktail-party settings strongly modulates cortical activity patterns • Semantics affect attention-related activity patterns in primary auditory cortex. • Dynamics of attention to speech are context-dependent. Recent studies utilizing electrophysiological speech envelope reconstruction have sparked renewed interest in the cocktail party effect by showing that auditory neurons entrain to selectively attended speech. Yet, the neural networks of attention to speech in naturalistic audiovisual settings with multiple sound sources remain poorly understood. We collected functional brain imaging data while participants viewed audiovisual video clips of lifelike dialogues with concurrent distracting speech in the background. Dialogues were presented in a full-factorial design, comprising task (listen to the dialogues vs. ignore them), audiovisual quality and semantic predictability. We used univariate analyses in combination with multivariate pattern analysis (MVPA) to study modulations of brain activity related to attentive processing of audiovisual speech. We found attentive speech processing to cause distinct spatiotemporal modulation profiles in distributed cortical areas including sensory and frontal-control networks. Semantic coherence modulated attention-related activation patterns in the earliest stages of auditory cortical processing, suggesting that the auditory cortex is involved in high-level speech processing. Our results corroborate views that emphasize the dynamic nature of attention, with task-specificity and context as cornerstones of the underlying neuro-cognitive mechanisms. [ABSTRACT FROM AUTHOR]

Individuals with attention-deficit/hyperactivity disorder (ADHD) have difficulties navigating dynamic everyday situations that contain multiple sensory inputs that need to either be attended to or ignored. As conventional experimental tasks lack this type of everyday complexity, we administered a film-based multi-talker condition with auditory distractors in the background. ADHD-related aberrant brain responses to this naturalistic stimulus were identified using intersubject correlations (ISCs) in functional magnetic resonance imaging (fMRI) data collected from 51 adults with ADHD and 29 healthy controls. A novel permutation-based approach introducing studentized statistics and subject-wise voxel-level null-distributions revealed that several areas in cerebral attention networks and sensory cortices were desynchronized in participants with ADHD (n = 20) relative to healthy controls (n = 20). Specifically, desynchronization of the posterior parietal cortex occurred when irrelevant speech or music was presented in the background, but not when irrelevant white noise was presented, or when there were no distractors. We also show regionally distinct ISC signatures for inattention and impulsivity. Finally, post-scan recall of the film contents was associated with stronger ISCs in the default-mode network for the ADHD and in the dorsal attention network for healthy controls. This study investigated effects of multisession transcranial direct-current stimulation on learning and maintenance of novel memory content and scrutinised effects of baseline cognitive status and the role of multi-session tDCS on overnight memory consolidation. In a prospective, randomized, double-blind, parallel-group, sham-tDCS controlled design, 101 healthy young and older adults completed a five-day verbal associative learning paradigm while receiving multisession tDCS to the task-relevant left prefrontal cortex. In older adults, active multisession tDCS enhanced recall performance after each daily training session. Effects were maintained the next morning and during follow-up assessments (one week; three months). In young adults, multisession tDCS significantly increased long-term recall. Unlike previous findings in the motor domain, beneficial effects of multisession tDCS on cognitive learning and memory were notexclusively due to enhanced memory consolidation. Positive stimulation effects were primarily found in participants with lower baseline learning ability, suggesting that multisession tDCS may counteract memory impairment in health and disease. • We characterized a cascade of brain responses modulated by working memory training. • These effects include P2-N2-P3 complex and slow waves that were differentially affected. • Reduced load-effect for P2-N2-P3 complex in the training group may reflect automatization. • Enhanced slow waves in a difficult condition was taken to indicate improved maintenance. The brain mechanisms of working memory (WM) training in humans remain unclear. Here we examined how WM updating training modulates a cascade of event-related potentials (ERPs) elicited at different processing stages. We hypothesized that WM updating training results to decreases in the early responses reflecting stimulus selection and response preparation, and increases the late slow responses reflecting maintenance of to-be-remembered materials. Healthy adults were randomized to a WM updating group that trained an adaptive dual n-back task (n = 20), and an active control group that played a computer game (n = 20). Both groups performed three 25-min training sessions per week for five weeks. Pretest-posttest comparisons showed that the training group significantly improved their performance as compared to the active controls, but this was limited to the trained task. In line with our hypothesis, P2-N2-P3 complex showed changes from pre- to posttest. In the training group this was observed as decreased load-effect while in the control group there was an opposite pattern at some latencies. Slow waves elicited during the maintenance were decreased in the easy task and increased in the difficult task. Taken together, our findings suggest that the early and late ERPs are differentially affected by training. When task demands are high, training may lead to an improved ability to actively maintain several stimuli in memory, and when they are low, training results in more efficient processing and automatization.
