: ObjectivesTo investigate in vitro natural killer (NK) cell activity and expression of signal-transducing ζ chains in patients with cervical cancer.: Patients and methodsExperiments were performed with frozen lymphocytes from patients at all disease stages and from healthy controls. Thawed NK were activated by overnight incubation in interferon-γ (IFN-γ); activity against two target cell lines was assessed by 4-h 51Cr release assay. Targets chosen were K562, an erythroleukemic cell line, and a cervical carcinoma cell line designated 808. T and NK cell ζ chain expression was measured by flow cytometry.: ResultsPatients’ NK were found to be as cytotoxic as those of normal controls against cell lines K562 and 808. Patient T and NK cells did not show significant down-regulation of the ζ chain.: ConclusionsWe have found no evidence to suggest that loss of ζ chains is a mechanism for immunocompromise in patients with cervical carcinoma. IFN-recoverable patient NK activity is not reduced compared to matched controls. This may be clinically relevant since NK are active against cells exhibiting class I human leukocyte antigen (HLA) down-regulation and many cervical cancers show loss of HLA. Background Endometrial cancer (EC) is the most common gynaecological cancer in developed nations and its incidence is rising. As a direct consequence, more women are dying from EC despite advances in care and improved survivorship. There is a lack of research activity and funding, as well as public awareness about EC. We sought to engage patients, carers and healthcare professionals to identify the most important unanswered research questions in EC. Methodology The priority setting methodology was developed by the James Lind Alliance and involved four key stages: gathering research questions; checking these against existing evidence; interim prioritisation; and a final consensus meeting during which the top ten unanswered research questions were agreed using modified nominal group methodology. Results Our first online survey yielded 786 individual submissions from 413 respondents, of whom 211 were EC survivors or carers, and from which 202 unique unanswered research questions were generated. 253 individuals, including 108 EC survivors and carers, completed an online interim prioritisation survey. The resulting top 30 questions were ranked in a final consensus meeting. Our top ten spanned the breadth of patient experience of this disease and included developing personalised risk scoring, refining criteria for specialist referral, understanding the underlying biology of different types of EC, developing novel personalised treatment and prevention strategies, prognostic and predictive biomarkers, increasing public awareness and interventions for psychological issues. Conclusion Having established the top ten unanswered research questions in EC, we hope this galvanises researchers, healthcare professionals and the public to collaborate, coordinate and invest in research to improve the lives of women affected by EC. Abstract: Objective: We evaluated baseline data from the PApilloma TRIal against Cancer In young Adults (PATRICIA; NCT00122681) on the association between behavioral risk factors and HPV infection and cervical abnormalities. Methods: Women completed behavioral questionnaires at baseline. Prevalence of HPV infection and cervical abnormalities (detected by cytological or histological procedures) and association with behavioral risk factors were analyzed by univariate and stepwise multivariable logistic regressions. Results: 16782 women completed questionnaires. Among 16748 women with data for HPV infection, 4059 (24.2%) were infected with any HPV type. Among 16757 women with data for cytological abnormalities, 1626 (9.7%) had a cytological abnormality, of whom 1170 (72.0%) were infected with at least one oncogenic HPV type including HPV-16 (22.7%) and HPV-18 (9.3%). Multivariable analysis (adjusted for age and region, N =14404) showed a significant association between infection with any HPV type and not living with a partner, smoking, age <15years at first sexual intercourse, higher number of sexual partners during the past 12months, longer duration of hormonal contraception and history of sexually transmitted infection (STI). For cervical abnormalities, only history of STI (excluding Chlamydia trachomatis) remained significant in the multivariable analysis after adjusting for HPV infection. Conclusions: Women reporting 3+ sexual partners in the past 12months had the highest risk of HPV infection at baseline. HPV infection was the main risk factor for cervical abnormalities, and history of STIs excluding Chlamydia trachomatis increased risk to a lesser extent. Although behavioral factors can influence risk, all sexually active women are susceptible to HPV infection. [Copyright &y& Elsevier]

Abstract: Background : The aim of this study is to assess the impact of two leaflets upon intentions to stop smoking in the next month in women receiving abnormal cervical smear test results. Methods : 330 women smokers who received an abnormal smear test result were randomly allocated to one of three groups: Group 1: sent a leaflet about the threat of cervical cancer from smoking and the benefits to cervical health in stopping, containing a detailed explanation of how smoking harms the cervix; Group 2: sent the same leaflet but without the detailed explanation; Group 3: no leaflet. Questionnaires were completed 1 month later. Results : Women in Group 1 had higher intentions to stop smoking compared with Group 3. This effect was mediated by perceived coherence of the link between smoking and cervical cancer and moderated by self-efficacy (confidence): only women with high levels of self-efficacy had higher intentions to stop smoking. Conclusion : Explaining the link between smoking and cervical cancer increases intentions to stop smoking in women receiving abnormal cervical smear test results. Further studies are needed to determine how self-efficacy can be increased and whether, in addition to the detailed leaflet, this increases rates of smoking cessation. [Copyright &y& Elsevier]
