Relaxin3 (rln3) has been associated with various emotional and cognitive processes, including stress, anxiety, learning, memory, motivational behavior, and circadian rhythm. Notably, previous report revealed that Rln3a played an indispensable role in testicular development and male fertility in Nile tilapia (Oreochromis niloticus). However, the underlying molecular mechanisms remain largely unknown. We found that Rln3a is expressed exclusively in the diencephalon* (Di*) of the brain. Deficiency of Rln3a resulted in a significant increase in serum dopamine level and an upregulation of gene expression of gnrh1 and kisspeptin2. To further elucidate the role of Rln3a in fish fertility, we collected two different regions of Di* and hypothalamus (Hyp) tissues for subsequent RNA-seq analysis of both wild-type (rln3a +/+) and rln3a -/- male tilapia. Upon the transcriptomic data, 1136 and 755 differentially expressed genes (DEGs) were identified in the Di* and Hyp tissues, respectively. In Di*, the up-regulated genes were enriched in circadian rhythm, chemical carcinogenesis, while the down-regulated genes were enriched in type II diabetes mellitus, dopaminergic synapse, and other pathways. In Hyp, the up-regulated genes were enriched in circadian rhythm, pyrimidine metabolism, while the down-regulated genes were enriched in type I diabetes mellitus, autoimmune thyroid disease, and other pathways. Subsequently, the results of both qRT-PCR and FISH assays highlighted a pronounced up-regulation of core circadian rhythm genes, cry1b and per3 , whereas genes such as clocka , clockb , and arntl exhibited down-regulation. Furthermore, the genes associated with dopamine biosynthesis were significantly increased in the Hyp. In summary, the mutation of rln3a in male tilapia resulted in notable changes in circadian rhythm and disease-linked signaling pathways in the Di* and Hyp. Chromosomal ploidy manipulation is one of the means to create excellent germplasm. Triploid fish could provide an ideal sterile model for searching of a underlying mechanism of abnormality in meiosis. The complete understanding of the coding and noncoding RNAs regulating sterility caused by meiosis abnormality is still not well understood. By high-throughput sequencing, we compared the expression profiles of gonadal mRNA, long non-coding RNA (lncRNA), and microRNA (miRNA) at three different developmental stages between the diploid (XX) and triploid (XXX) female rainbow trout. These stages were gonads before differentiation (65 days post fertilisation, dpf), at the beginning of morphological differences (180 dpf) and showing clear difference between diploids and triploids (600 dpf), respectively. A majority of differentially expressed (DE) RNAs were identified, and 22 DE mRNAs related to oocyte meiosis and homologous recombination were characterized. The predicted miRNA-mRNA/lncRNA networks of 3 developmental stages were constructed based on the target pairs of DE lncRNA-miRNA and DE mRNA-miRNA. According to the networks, meiosis-related gene of ccne1 was targeted by dre-miR-15a-5p_R + 1, and 6 targeted DE lncRNAs were identified. Also, qRT-PCR was performed to validate the credibility of the network. Overall, this study explored the potential interplay between coding and noncoding RNAs during the gonadal development of polyploid fish. The development of semen cryopreservation strategies is necessary to improve the semen storage technologies of species of great commercial interest for aquaculture. Recent studies demonstrate that lipids play an important role in the fertility and cryotolerance of fish gametes. This study investigated the effect of exogenous lipids in the freezing medium on the post-thaw functional parameters of Salmo salar spermatozoa. Semen samples (n = 12) were incubated in standard extender supplemented with different concentrations of oleic acid (OA, C18:1n9), linoleic acid (LA, C18:2n6), arachidonic acid (ARA, C20:4n6) and cholesterol-loaded cyclodextrin (CLC). Post-thaw motility, membrane integrity, mitochondrial membrane potential (ΔΨm), superoxide anion (O 2 •-) and fertility rates were analyzed. The results revealed that the semen incubated with 0.003 mmol/L OA increased the motility (~7%) and ΔΨm (~2%) (P < 0.05), but membrane integrity and fertility were not increased. The addition of 0.003 mmol/L LA increased the motility (~4%) and all LA extenders increased the ΔΨm (P < 0.05); however, LA increased the O 2 •- levels and decreased the membrane integrity and fertility (P < 0.05). Semen incubated with ARA improved sperm motility (~5%), membrane integrity (~10.5%) and fertility rates (~11%) (P < 0.05). The maximum improvement in post-thaw sperm functionality was observed by adding 0.003 mmol/L ARA. In contrast, sperm quality parameters and fertility were decreased by the CLC addition (P < 0.05). Behavioural isolation between closely related species with exaggerated male mating traits is traditionally thought to be mediated by female preference for conspecific male ornaments. The role of male mate choice in maintaining boundaries between highly sexually dimorphic species is therefore comparatively neglected. However, mounting theoretical and empirical evidence supports the existence of male mate choice both within, and increasingly, between species with exaggerated male ornaments. We therefore tested the role of male mate choice in the maintenance of species boundaries for two sexually dimorphic species of darters (Percidae: Etheostoma ). Using dichotomous choice assays, we measured male preferences of sympatric species Etheosotma barrenense and Etheosotma zonale for size-matched conspecific and heterospecific females, thus reducing the possibility that males would select for general indicators of fecundity. Our results show that males of both species strongly prefer conspecific females. A comparison with published data showed that the strength of preference for conspecific mates is just as strong for males as it is for females in E. barrenense . We also estimated the relative contribution of male mate choice, female mate choice and male–male competition to behavioural isolation. We found that male mate choice contributes as much as female mate choice to total behavioural isolation and thus likely plays an important role in behavioural isolation in these sexually dimorphic species. Our results suggest that the contribution of male choice to behavioural isolation should be studied in a larger variety of animal species to appreciate the relative roles of the sexes in the maintenance of species boundaries. Because of various Congressional mandates to protect the environment from endocrine disrupting chemicals (EDCs), the United States Environmental Protection Agency (USEPA) initiated the Endocrine Disruptor Screening Program. In the context of this framework, the Office of Research and Development within the USEPA developed the Medaka Extended One Generation Reproduction Test (MEOGRT) to characterize the endocrine action of a suspected EDC. One important endpoint of the MEOGRT is fecundity of medaka breeding pairs. Power analyses were conducted to determine the number of replicates needed in proposed test designs and to determine the effects that varying reproductive parameters ( e.g . mean fecundity, variance, and days with no egg production) would have on the statistical power of the test. The MEOGRT Reproduction Power Analysis Tool (MRPAT) is a software tool developed to expedite these power analyses by both calculating estimates of the needed reproductive parameters ( e.g . population mean and variance) and performing the power analysis under user specified scenarios. Example scenarios are detailed that highlight the importance of the reproductive parameters on statistical power. When control fecundity is increased from 21 to 38 eggs per pair per day and the variance decreased from 49 to 20, the gain in power is equivalent to increasing replication by 2.5 times. On the other hand, if 10% of the breeding pairs, including controls, do not spawn, the power to detect a 40% decrease in fecundity drops to 0.54 from nearly 0.98 when all pairs have some level of egg production. Perhaps most importantly, MRPAT was used to inform the decision making process that lead to the final recommendation of the MEOGRT to have 24 control breeding pairs and 12 breeding pairs in each exposure group. This study was designed to test the effect of post-thaw storage time on sperm motility parameters of Caspian brown trout ( n = 7). Furthermore, we investigated the effect of sperm-to-egg ratios of 100,000:1, 300,000:1 and 600,000:1 on fertility of cryopreserved Caspian brown semen. Quality was assessed by measuring sperm motility parameters and fertilization rates at the eyed and hatching stages. The percentage of post-thawed sperm motility, curvilinear velocity (VCL) and amplitude of lateral head displacement (ALH) were not affected by 60 min of storage, whereas a decrease in straight line velocity (VSL), average path velocity (VAP) and linearity (LIN) were found in cryopreserved semen. Thus, the cryopreserved sperm of Caspian brown trout could be stored up to 60 min without loss of the percentage of sperm motility. The fertilization rate was not affected by 60 min of post-thaw storage and was over 70% for sperm-to-egg ratios of both 300,000 and 600,000:1. To our knowledge, this study is the first to report the high post-thaw fertilization ability of Caspian brown trout semen at a sperm-to-egg ratio as low as 300,000:1. Exposure to estrogenic endocrine disruptors (EDCs) during development affects fertility, reproductive and non-reproductive behavior in mammals and fish. These effects can also be transferred to coming generations. In fish, the effects of developmental EDC exposure on non-reproductive behavior are less well studied. Here, we analyze the effects of 17α-ethinylestradiol (EE 2 ) on anxiety, shoaling behavior and fertility in zebrafish after developmental treatment and remediation in clean water until adulthood. Zebrafish embryos were exposed from day 1 to day 80 post fertilization to actual concentrations of 1.2 and 1.6 ng/L EE 2 . After remediation for 82 days non-reproductive behavior and fertilization success were analyzed in both sexes. Males and females from the 1.2 ng/L group, as well as control males and females, were bred, and behavior of the untreated F1 offspring was tested as adults. Developmental treatment with 1.2 and 1.6 ng/L EE 2 significantly increased anxiety in the novel tank test and increased shoaling intensity in both sexes. Fertilization success was significantly reduced by EE 2 in both sexes when mated with untreated fish of opposite sex. Progeny of fish treated with 1.2 ng/L EE 2 showed increased anxiety in the novel tank test and increased light avoidance in the scototaxis test compared to control offspring. In conclusion, developmental exposure of zebrafish to low doses of EE 2 resulted in persistent changes in behavior and fertility. Background Polybrominated diphenyl ethers (PBDEs) are ubiquitous in various environmental matrices and organisms and pose a threat to reproductive systems of organisms. However, few studies have explored the effects of PBDEs on sexual and reproductive behaviors in animals. Here we evaluated the effect of BDE-47 exposure on sexual and reproductive behaviors in zebrafish ( Danio rerio ). Methods We used a charge-coupled device camera to evaluate 3 standard male zebrafish sexual behaviors—chasing, female association and induced female spawning—and assessed effects on reproductive success in female zebrafish that mated with exposed males. Results After 21-day BDE-47 exposure, the frequency and total time of males associating with females was dose-dependently decreased. With the highest BDE-47 exposure, 1000 µg/L, the frequency of inducing spawning was decreased. Sexual behaviors and spawning outcome were closely associated in both control and exposure groups. Logistic regression analysis revealed BDE-47 exposure and total time of female association as the main factors contributing to induced female spawning behaviors, which affected final egg production. Multiple stepwise regression analysis suggested that female association and induced spawning by males were associated with egg production. Meanwhile, fecundity was lower for BDE-47-treated groups than controls, with only a significant difference with the highest dose. BDE-47 exposure at 100 and 1000 µg/L in males decreased fertilization rate, but BDE47 had no effect on hatching rate. In present study, it was examined whether addition of different antioxidants to the cryopreservation extenders had an effect on semen post-thaw fertility and motility in rainbow trout ( Oncorhynchus mykiss ) and also it was investigated the sperm characteristics post-thaw sperm characteristics and fertility. The collected semen was pooled to minimize individual variation. Each pooled ejaculate was split into 12 equal aliquots and diluted with base extenders supplemented with the antioxidants, and a base extender with no additives (control). The pooled semen samples diluted at the ratio of 1:10 by the extenders were subjected to cryopreservation. Antioxidants were separately added to the extenders (one per experimental group): catalase (250 U/l), superoxide dismutase (250 U/l), peroxidase (250 U/l), oxidized glutathione (1.5 mmol/l), reduced glutathione (1.5 mmol/l), l -methionine (1.5 mmol/l), uric acid (0.25 mmol/l), l -ascorbic acid (0.5 mmol/l), α-tocopherol (2.0 mmol/l), β-carotene (0.5 mmol/l) and carnitine (0.5 mmol/l). After dilution the semen was aspirated into 0.25 ml straws, the straws were placed on the tray, frozen for 10 min, and plunged into liquid nitrogen. Our results indicated that the post-thaw motility rate increased in extenders supplemented with uric acid, l -methionine, SOD, l -carnitine, α-tocopherol and l -reduced glutathione ( p < 0.05). The motility duration of frozen thawed semen increased in extenders supplemented with uric acid, l -methionine, SOD, α-tocopherol and l -reduced glutathione ( p < 0.05). Fertilization rate and hatching rate of frozen-thawed semen was not affected by the tested antioxidants. Consequently, the tested antioxidants affected the motility parameters and cryopreservation extenders could be supplement with antioxidants. Abstract: Individuals are commonly exposed to fluctuating levels of stressors, while most laboratory experiments focus on constant exposures. We develop and test a mathematical model for predicting the effects of low dissolved oxygen (hypoxia) on growth, reproduction, and survival using laboratory experiments on fish and shrimp. The exposure-effects model simulates the hourly reductions in growth and survival, and the reduction in reproduction (fecundity) at times of spawning, of an individual as it is exposed to constant or hourly fluctuating dissolved oxygen (DO) concentrations. The model was applied to seven experiments involving fish and shrimp that included constant and fluctuating DO exposures, with constant exposures used for parameter estimation and the model then used to simulate the growth, reproduction, and survival in the fluctuating treatments. Cumulative effects on growth, reproduction, and survival were predicted well by the model, but the model did not replay the observed episodic low survival days. Further investigation should involve the role of acclimation, possible inclusion of repair effects in reproduction and survival, and the sensitivity of model predictions to the shape of the immediate effects function. Additional testing of the model with other taxa, different patterns of fluctuating exposures, and different stressors is needed to determine the model's generality and robustness. Abstract: 2,4-Dichlorophenoxyacetic acid (2,4-D) was evaluated in both the Amphibian Metamorphosis Assay (AMA) and the Fish Short Term Reproduction Assay (FSTRA). In the AMA, tadpoles were exposed to mean measured 2,4-D concentrations of 0 (water control), 0.273, 3.24, 38.0 and 113mg acid equivalents (ae)/L for either seven or 21 days. In the FSTRA, fathead minnows were exposed to mean measured 2,4-D concentrations of 0 (water control), 0.245, 3.14, 34.0, and 96.5mg ae/L for 21 days. The respective concentrations of 2,4-D were not overtly toxic to either Xenopus laevis tadpoles or fathead minnows (Pimephales promelas). In the AMA, there were no signs of either advanced or delayed development, asynchronous development, or significant histopathological effects of the thyroid gland among 2,4-D exposed tadpoles evaluated on either day seven or day 21 of the exposure. Therefore, following the AMA decision logic, 2,4-D is considered “likely thyroid inactive” in the AMA with a No Observable Effect Concentration (NOEC) of 113mg ae 2,4-D/L. In the FSTRA, there were no significant differences between control and 2,4-D exposed fish in regard to fertility, wet weight, length, gonado-somatic indices, tubercle scores, or blood plasma concentrations of vitellogenin. Furthermore, there were no treatment-related histopathologic changes in the testes or ovaries in any 2,4-D exposed group. The only significant effect was a decrease in fecundity among fish exposed to 96.5mg ae 2,4-D/L. The cause of the reduced fecundity at the highest concentration of 2,4-D tested in the assay was most likely due to a generalized stress response in the fish, and not due to a specific endocrine mode of action of 2,4-D. Understanding pre- and postcopulatory mechanisms of sexual selection (male–male competition and female choice) can provide insight into the evolution of male ornamentation. In this study, we use an integrative approach to study sexual selection by incorporating measures of both mechanisms using the redside dace, Clinostomus elongatus, a freshwater, externally fertilizing fish found in streams of eastern North America. We measured the success of individual male redside dace in both pre- and postspawning sexual selection and related the success of males to a range of traits, including spectral properties and the size of their red spawning coloration, body size metrics and sperm quality. We found significant sexual dimorphism in the relative amount and spectral properties (saturation and brightness) of the red spawning coloration, with males being more ornamented than females. Using dichotomous mate choice trials, we found that females did not favour males with relatively larger red spawning coloration area and more saturated or brighter red spawning coloration. Males with relatively more red spawning area would be favoured in male–male postspawning sperm competition because they possess sperm with higher velocity, and sperm velocity was determined to be the key predictor of sperm competition success as assessed by paternity analyses related to in vitro sperm competition trials. These results suggest that prespawning selection via female choice is not driving the evolution of red spawning coloration and that postspawning selection via male–male competition is primarily determined by sperm velocity, which was correlated with the relative amount of red spawning coloration. Abstract: The Lusitanian toadfish, Halobatrachus didactylus (Bloch and Schneider, 1801), is a subtropical sound-producing marine teleost that inhabits estuaries and other brackish water ecosystems in its northern distribution area. The relative isolation of populations within estuaries may cause important modifications in the bio-ecology of this species. The males of this species present two morphotypes which are more accurately distinguishable during the reproductive season. In the Tagus estuary population, the two types of reproductive males were present: type I, the “characteristic” morphotype, and type II, the alternative morphotype. The opportunistic males had more developed testis while type I males had more developed accessory glands. Type I males presented more developed swimbladders and larger body size and live longer than type II males. On the contrary, type II males grew faster and matured earlier. The feeding habits of the two male morphotypes were similar during the reproductive season but differences were found between reproductive specimens and non-reproductive specimens. The diet of reproductive specimens was mainly composed of benthic prey in contrast to predominance of nectobenthic prey in the diet of non-reproductive specimens. The females of H. didactylus generally had an intermediate pattern between the two male morphotypes. The percentage of type II males tended to be higher in denser areas of the estuary. In comparison with populations from different size ecosystems, smaller ecosystems revealed important modifications on the bio-ecology of the species due to the increased competition for nesting sites and mating partners, favouring alternative male tactics. Abstract: It is well known that slow and fast muscles are used for long-term sustained movement and short bursts of activity, respectively, in adult animal behaviors. However, the contribution of the slow and fast muscles in early animal movement has not been thoroughly explored. In wild-type zebrafish embryos, tactile stimulation induces coilings consisting of 1–3 alternating contractions of the trunk and tail at 24hours postfertilization (hpf) and burst swimming at 48hpf. But, embryos defective in flightless I homolog (flii), which encodes for an actin-regulating protein, exhibit normal coilings at 24hpf that is followed by significantly slower burst swimming at 48hpf. Interestingly, actin fibers are disorganized in mutant fast muscle but not in mutant slow muscle, suggesting that slower swimming at 48hpf is attributable to defects of the fast muscle tissue. In fact, perturbation of the fast muscle contractions by eliminating Ca2+ release only in fast muscle resulted in normal coilings at 24hpf and slower burst swimming at 48hpf, just as flii mutants exhibited. In contrast, specific inactivation of slow muscle by knockdown of the slow muscle myosin genes led to complete loss of coilings at 24hpf, although normal burst swimming was retained by 48hpf. These findings indicate that coilings at 24hpf is mediated by slow muscle only, whereas burst swimming at 48hpf is executed primarily by fast muscle. It is consistent with the fact that differentiation of fast muscle follows that of slow muscle. This is the first direct demonstration that slow and fast muscles have distinct physiologically relevant contribution in early motor development at different stages. [Copyright &y& Elsevier]

Abstract: Potamopyrgus antipodarum is a promising test organism that is often used in ecotoxicology, both in laboratory and field exposures. As no data are available on the physiological variation range of its life-traits and the biomarkers it uses, we studied the variation of fecundity, steroid levels and energy reserves over the course of a year in a field population. The reproductive cycle was described and showed seasonal activity during summer and autumn. Steroid levels (17β-estradiol and testosterone) varied significantly during the year and were correlated with the reproductive cycle, which suggested a potential role for sex-steroids in P. antipodarum reproduction. Energy status also showed seasonal variations. Triglycerides (TG) seemed to be the main energy lipid, whereas cholesterol appeared to be mostly used as a structural lipid. Proteins were also involved in the reproductive cycle, but only when TG were not sufficient to support the reproductive strain, similar to cholesterol. Glycogen seemed to be used as an early reserve. Threshold values under which no reproduction occurred were defined in starved snails. We proposed a range of variation in the measured parameters, allowing for a better understanding and interpretation of their levels during laboratory or in situ exposures. Abstract: Gravid females of Caligus rogercresseyi were collected from Atlantic salmon (Salmo salar) from a farm located at Chiloe Island (42°40′S73°15′W), Chile, to obtain information about the reproductive output of this parasite in vitro. The egg strings removed from the females were incubated under controlled conditions to obtain virgin adult females. One female which had mated only once produced eleven generations of eggs strings in a period of 74days. The first egg strings of the females obtained in vitro were produced at 389 degree days (°D) after egg incubation, while the next generations of eggs strings were produced with a periodicity between 4 and 6days dependent on the water temperature. The average length of the egg string was 3.1mm and the mean number of eggs per string was 31. The values recorded in captivity for the egg string length and the number of eggs per string, were lower than the values recorded in gravid females from the field. Abstract: Elongate ilisha (Ilisha enlongata) is a commercially important species that contributes to clupeoid fisheries in Asian countries. In the present study, the reproductive biology of I. elongata in Ariake Sound, Japan is determined for the first time. Six maturity stages were described using ovarian and testicular histology throughout the annual cycle. The spawning season in Ariake Sound lasts from May to July, with peak spawning activity in May and June. Age at first maturity was estimated to be 2years, with a few exceptions of 1year in well-developed males. Ovaries that contained both tertiary yolk oocytes and postovulatory follicles occurred from late May to late July, indicating that I. elongata is a multiple spawner. The size–frequency distribution of oocytes provided evidence for its multiple spawning and accuracy of the fecundity estimates. The batch fecundity of this species was estimated at between 22,200 and 270,900 eggs per individual, increasing with age between two and six years. The present findings on the reproductive strategy of I. elongata in Ariake Sound are generally consistent with those in temperate or subtropical populations, but quite different from those of tropical population where first maturation occurs around 200days and life spans are shorter, with a maximum age less than 3years. Abstract: Breeding pairs of rare minnows were exposed to 5, 10, 20μg/L 4-nonylphenol (NP) or 4ng/L 17α-ethinylestradiol (EE2) for 21 days. Among reproductive parameters investigated, fecundity, fertility, and laying interval were significantly affected by 4ng/L EE2, but were not affected at any exposure level of NP. These effects coincided with male feminization, ova-testis, increased plasma VTG in both males and females as well as the accumulation of eosinophilic material in kidneys and livers. Histopathologic evaluation revealed severe kidney lesions in both EE2 and NP-exposed fish. The bioassay succeeded in demonstrating that EE2 was several orders of magnitude more potent than the industrial chemical NP. The rare minnow was more sensitive to EE2 than medaka, but comparable to zebrafish in terms of sensitivity. The results of the present investigation indicated that a short-term reproductive bioassay is feasible using the Chinese rare minnow as the test species. [Copyright &y& Elsevier]

The research was carried out to determine whether there are individual-level differences in the bicolor damselfish (Stegastes partitus) within the altered fish communities located on the north coast of Havana, Cuba. There was strong evidence of changes in some morphological and physiological characteristics associated with the impact of land-based pollution in the coastal zone. A combination of impaired recruitment due to habitat degradation with increased food supply due to eutrophication seems to be the best explanation for fishes being heavier and longer at polluted sites. The change in the proportion of color patterns and a very high number of atretic oocytes in the ovaries of fish caught near the mouth of the Almendares River strongly support the idea that not only is the pollution of river waters affecting the marine life in the coastal zone, but also that this pollution has greater effects than the pollution coming from the discharge of Havana Harbor. We investigated the effect of storage times of frozen–thawed Persian sturgeon ( Acipenser persicus ) semen on the duration of sperm motility, percentage of motile sperm, and fertilization and hatching rates of fresh sperm and sperm stored for 0, 30, and 60 min at 4 °C post-thawing. Frozen thawed semen analyzed immediately after thawing had similar quality characteristics as fresh semen. For cryopreserved semen stored for 30 min after thawing the characteristics did not differ to fresh semen and cryopreserved semen. For cryopreserved semen stored for 60 min a significant decline in the parameters was observed. Fertilization and hatching rates were not affected by storage times of maximally 30 min of storage.
