Effective governance of public forests depends, in part, on public support for changes in forest management, particularly those responding to changes in socio-ecological conditions driven by climate change. Trust in managing authorities and knowledge about forest management have proven influential in shaping public support for policy across different forest managemen contexts. However, little is known about the relationship between public trust and knowledge as it relates to policy support for emerging management strategies for climate adaptation in forests. We use the example of genomics-based assisted migration (within and outside of natural range) in British Columbia's (BC) forests to examine the relative roles of and interactions between trust in different forestry actors and knowledge of forestry in shaping public support for this new and potentially controversial management alternative. Our results, based on an online survey (n = 1953 BC residents), reveal low public trust in governments and the forest industry combined with low levels of public knowledge about forest management. We find that individuals who are more trusting of decision-makers and other important forestry actors hold higher levels of support for assisted migration. Higher levels of forestry knowledge are linked with support for assisted migration within native range, whereas no knowledge effect is observed for assisted migration outside of native range. We discuss the implications of these observations and provide recommendations to more fully engage with the challenges of low levels of trust and knowledge in this context., (Copyright © 2019 Elsevier Ltd. In recent years, the provision of economic incentives through carbon financing and carbon offsetting has been central to efforts at forest carbon mitigation. However, notwithstanding their potentially important roles in climate policy, forest carbon offsets face numerous barriers which have limited widespread implementation worldwide. This paper uses the case study of the Canadian province of British Columbia to explore the barriers associated with achieving widespread implementation of forest carbon offsets in the next several decades. Drawing on interviews with experts from government, non-governmental organizations, the private sector and First Nations, six main barriers are identified and discussed: (1) deficiencies of carbon markets, (2) limited economic benefits, (3) uncertain climate effectiveness, (4) negative public opinion, (5) limited and uncertain property rights, and (6) governance issues. While respondents from different sectors agreed on various points, divergence was also observed, notably on the trade-off between generating environmentally sound offsets and promoting cost-effective ways to achieve mitigation. We discuss these differences in the context of the goals and objectives of different actors, and offer insights for understanding the uptake (or not) of carbon offset policies., (Copyright © 2017 Elsevier Ltd. The analysis of the differential expression of genes has been the key goal of many molecular biology methods for decades and will remain with us for decades to come. It constitutes a fundamental resource at our disposal for determining the relationship between products of transcription, biology and disease. The completed genome sequencing of many common species allowed microarrays and RNA sequencing (RNAseq) to become major tools in Systems Biology. However, we estimate that at least half of all experiments ignore transcripts that change less than some subjectively chosen threshold, typically around 2-3 fold. Here we show that a majority of the informative RNAs and differentially expressed transcripts can exhibit fold changes less than 2. We use highly quantitative single-molecule sequencing of total cellular RNA derived from a time course of inflammatory response, a process critical to a large number of diseases. Furthermore, we show that enrichment of biologically-relevant functions occurs even at very low fold changes in RNA levels. In addition, we show that most of the common statistical methods can reliably detect transcripts with low fold change when as few as 3 biological replicates are sequenced using single-molecule based RNAseq. In conclusion, given the prevalence of expression profiling in current research, the loss of data in half of all expression studies results in a significant, yet needless drain on the discovery process., (Copyright © 2013 Elsevier Inc. Transient gene expression in mammalian cells is a valuable alternative to stable cell lines for the rapid production of large amounts of recombinant proteins. While the establishment of stable cell lines takes 2-6 months, milligram amounts of protein can be obtained within a week following transfection. The polycation polyethylenimine (PEI) is one of the most utilized reagents for small- to large-scale transfections as it is simple to use and, when combined with optimized expression vectors and cell lines, provides high transfection efficiency and titers. As with most transfection reagents, PEI-mediated transfection involves the formation of nanoparticles (polyplexes) which are obtained by its mixing with plasmid DNA. A short incubation period that allows polyplexes to reach their optimal size is performed prior to their addition to the culture. As the quality of polyplexes directly impacts transfection efficiency and productivity, their formation complicates scalability and automation of the process, especially when performed in large-scale bioreactors or small-scale high-throughput formats. To avoid variations in transfection efficiency and productivity that arise from polyplexes formation step, we have optimized the conditions for their creation directly in the culture by the consecutive addition of DNA and PEI. This simplified approach is directly transferable from suspension cultures grown in 6-well plates to shaker flasks and 5-L WAVE bioreactors. As it minimizes the number of steps and does not require an incubation period for polyplex formation, it is also suitable for automation using static cultures in 96-well plates. This "direct" transfection method thus provides a robust platform for both high-throughput expression and large-scale production of recombinant proteins., (Crown Copyright © 2011.
