The cold stress during overwintering is considered the bottleneck of red tilapia industry. In this study, the water temperature (WT) was reduced by 2 °C per day from 20 °C to 8 °C in the cold (C) group. Then transcriptome of brain(B), gill(G), liver(L) and skin(S) tissues and proteome of G, L and S tissues were performed in C and Normal (N) (WT: 20 °C) group. 24 transcriptomes were completed, and 168.8 Gb data were obtained, with more than 5.89 Gb clean data of each sample. A total of 30499 annotation results were obtained with 3199, 4697, 4393, and 3382 differentially expressed mRNAs in NB_vs_CB, NG_vs_CG, NL_vs_CL, NS_vs_CS. 18 DIA proteomes were performed, and 6341 proteins were obtained with 178, 500 and 166 differentially expressed proteins in NG_vs_CG, NL_vs_CL, NS_vs_CS. Our datasets can be reused for key genes and proteins identification, omics joint analysis and regulatory mechanism analysis of low temperature or cold stress in fish, which will help understanding the regulatory mechanism and facilitate the molecular selective breeding of cold-resistant varieties of fish. This study was designed to screen 6 lysosomal storage diseases (LSDs) in neonates using tandem mass spectrometry (MS/MS), and establish cutoff values for these LSDs with 3000 dried blood spots (DBS) samples. Cutoff values for α-L-iduronidase (IDUA), α-galactosidase (GLA), acid beta glucosidase (ABG), β-galactocerebrosidase (GALC), acid sphingomyelinase (ASM), and acid alpha glucosidase (GAA) were as follows: GLA, > 2.06 μmol/L·h; ABG, > 1.78 μmol/L·h; ASM, > 0.99 μmol/L·h; IDUA, > 1.33 μmol/L·h; GALC, > 0.84 μmol/L·h; and GAA, > 2.06 μmol/L·h. There were 30 positives in initial MS/MS screening test, and 15 samples were still positive with repeat testing. Their parents/guardians were recontacted and DBS samples were collected again for test. Only 1 child showed abnormal GAA enzyme activity after recontacting process, and was diagnosed with Pompe disease after genetic screening. The traditional methods for predicting the distribution of albacore (Thunnus alalunga) fishing grounds have low performance and accuracy. Uneven sampling can result in unreasonable evaluation indicators. To address these issues, three methods, equi-frequency, K-means clustering algorithm, and 1-R split, were applied to discretize the catch per unit effort (CPUE) of albacore in the South Pacific from 2016 to 2021 and partition the fishing grounds into abundance levels. Eight machine learning models were used to predict the fishing grounds. In addition to the traditional evaluation index based on confusion matrix, top-k index was also used to evaluate the accuracy of fishery abundance predictions. The results showed that (1) When sampling is unbalanced, the reported accuracy does not fully represent the actual performance of the model in predicting the abundance of albacore in the fishing ground. F1 value can be used as the index of the model effect and stability. (2) In binary classification, the quartile stacking algorithm has the best stacking performance, with F1 0.89. (3) The top-1 prediction accuracy of three-category fishery forecasting is the highest at 0.74, and the top-1 prediction accuracy of five-category fishery forecasting is the highest at 0.54. (4) The top-k accuracy of classification of fisheries with multiple abundance using K-means is significantly better than that of equal frequency discretization (p < 0.001). The top-k evaluation index was used to predict the fishing grounds of albacore across multiple abundance levels for the first time in this study, which is significant for pioneering a new method for this application and which provides a demonstration of the development of artificial intelligence techniques for fisheries in the future. To address the challenges of high experimental costs, complexity, and time consumption associated with pre-mixed combustible gas deflagration experiments under semi-open space obstacle conditions, a rapid temporal prediction method for flame propagation velocity based on Ranger-GRU neural networks is proposed. The deflagration experiment data are employed as the training dataset for the neural network, with the coefficient of determination (R2) and mean squared error (MSE) used as evaluation metrics to assess the predictive performance of the network. First, 108 sets of pre-mixed methane gas deflagration experiments were conducted, varying obstacle parameters to investigate methane deflagration mechanisms under different conditions. The experimental results demonstrate that obstacle-to-ignition source distance, obstacle shape, obstacle length, obstacle quantity, and thick and fine wire mesh obstacles all significantly influence flame propagation velocity. Subsequently, the GRU neural network was trained, and different activation functions (Sigmoid, Relu, PReLU) and optimizers (Lookahead, RAdam, Adam, Ranger) were incorporated into the backpropagation updating process of the network. The training results show that the Ranger-GRU neural network based on the PReLU activation function achieves the highest mean R2 value of 0.96 and the lowest mean MSE value of 7.16759. Therefore, the Ranger-GRU neural network with PReLU activation function can be a viable rapid prediction method for flame propagation velocity in pre-mixed methane gas deflagration experiments under semi-open space obstacle conditions. Organic fertilization is considered an effective approach in promoting agricultural green development, dramatically affecting soil phosphorus (P) availability. Nonetheless, limited information is available on the comprehensive impact of full substitution of organic fertilizer for chemical fertilizer on P speciation, phytoavailability, and apparent balance throughout different rice‐growth stages. To address this gap, a 5‐year field experiment was conducted, implementing five organic P gradients ranging from 0 (P0), 70 (P70), 140 (P140), 210 (P210) to 280 (P280) kg P2O5 ha−1 of organic fertilizer. To assess P phytoavailability in the root zone with submillimetre spatial resolutions, this study employed techniques such as the one‐ and two‐dimensional diffusive gradients in thin films (DGT) technique and the high‐resolution soil solution sampling technology (HR‐Peeper). The findings revealed that increasing P rates enhanced soil Olsen‐P and biological‐based P fractions across rice‐growth stages, primarily driven by variation in mineral‐associated P. Notably, the P140 treatment demonstrated the highest P uptake efficiency among the different rice‐growth stages, with a significant increase in soil DGT‐P, particularly in the 0–60 mm soil layer (p <0.05), providing tangible evidence for enhanced P uptake. Moreover, compared with higher P treatments (P210 and P280), the P140 treatment markedly increased P use efficiency by 31.7% and 99.0%, respectively (p <0.05). Further, with a high ratio of DGT‐P to Peeper‐P and a low apparent balance of P, organic fertilization at the rate of 140 kg P2O5 ha−1 effectively struck a balance between ensuring adequate P supply for yield stability and mitigating potential P loss risks. These results underscore the significance of optimal organic fertilization in enhancing agronomic benefits while reducing environmental risks. The late Mesozoic igneous rocks along the coastal South China Block (SCB) exhibit complex parental sources involving a depleted mantle, subducted sediment-derived melt, and melted crust. This period aligns with the magmatic flare-up and lull in the SCB, debating with the compression or extension in coastal region. Our study employs numerical models to investigate the dynamics of the ascent of underplating magma along the Changle–Nan'ao Belt (CNB), simulating its intrusion and cooling processes while disregarding the formational background. The rheological structure of the lithospheric mantle significantly influences magma pathways, dictating the distribution of magmatism. This work reveals that the ascent of magma in the presence of faults is considerably faster than in the absence of faults, and contemporaneous magmatic melts could produce different cooling and diagenetic processes. Additionally, the influence of pre-existing magma accelerated the emplacement of underplating magma. The magma beneath the fault ascended rapidly, reaching the lower crust within 20 million years, with a cooling rate of approximately ∼ 35 °C Myr -1. Conversely, the thickened magma took 40–50 million years to ascend to the lower crust, cooling at a rate of ∼ 10 °C Myr -1. In contrast, magma without thickening and fault would take a considerably longer time to reach the lower crust. The ascent of magma formed a mush-like head, contributing to magmatic circulation beneath the crust and decreasing crustal thickness. Multiphase magmatism increases the geothermal gradient, reducing lithospheric viscosity and promoting underplating magma ascent, leading to magmatic flare-ups and lulls. Our findings suggest that the Cretaceous magmatism at different times in the coastal SCB may be associated with the effects of lithospheric faults under similar subduction conditions. Boundary compression forces delay magma ascent, while rising magma induces a significant circulation, decreasing the crustal thickness of the coastal SCB. This study provides new insights into the complex interplay of magmatic processes during subduction, emphasizing the role of lithospheric structure in shaping the temporal and spatial evolution of coastal magmatism. Background: In prior research employing iTRAQ (Isobaric Tags for Relative and Absolute Quantitation) technology, we identified a range of proteins in breast cancer tissues exhibiting high levels of acetylation. Despite this advancement, the specific functions and implications of these acetylated proteins in the context of cancer biology have yet to be elucidated. This study aims to systematically investigate the functional roles of these acetylated proteins with the objective of identifying potential therapeutic targets within breast cancer pathophysiology. Methods: Acetylated targets were identified through bioinformatics, with their expression and acetylation subsequently confirmed. Proteomic analysis and validation studies identified potential acetyltransferases and deacetylases. We evaluated metabolic functions via assays for catalytic activity, glucose consumption, ATP levels, and lactate production. Cell proliferation and metastasis were assessed through viability, cycle analysis, clonogenic assays, PCNA uptake, wound healing, Transwell assays, and MMP/EMT marker detection. Results: Acetylated proteins in breast cancer were primarily involved in metabolism, significantly impacting glycolysis and the tricarboxylic acid cycle. Notably, PGK1 showed the highest acetylation at lysine 323 and exhibited increased expression and acetylation across breast cancer tissues, particularly in T47D and MCF-7 cells. Notably, 18 varieties acetyltransferases or deacetylases were identified in T47D cells, among which p300 and Sirtuin3 were validated for their interaction with PGK1. Acetylation at 323 K enhanced PGK1's metabolic role by boosting its activity, glucose uptake, ATP production, and lactate output. This modification also promoted cell proliferation, as evidenced by increased viability, S phase ratio, clonality, and PCNA levels. Furthermore, PGK1-323 K acetylation facilitated metastasis, improving wound healing, cell invasion, and upregulating MMP2, MMP9, N-cadherin, and Vimentin while downregulating E-cadherin. Conclusion: PGK1-323 K acetylation was significantly elevated in T47D and MCF-7 luminal A breast cancer cells and this acetylation could be regulated by p300 and Sirtuin3. Background: Fish reproduction, development and growth are directly affected by temperature, investigating the regulatory mechanisms behind high temperature stress is helpful to construct a finer molecular network. In this study, we systematically analyzed the transcriptome and miRNA information of American shad (Alosa sapidissima) liver tissues at different cultivation temperatures of 24 ℃ (Low), 27 ℃ (Mid) and 30 ℃ (High) based on a high-throughput sequencing platform. Results: The results showed that there were 1594 differentially expressed genes (DEGs) and 660 differentially expressed miRNAs (DEMs) in the LowLi vs. MidLi comparison group, 473 DEGs and 84 DEMs in the MidLi vs. HighLi group, 914 DEGs and 442 DEMs in the LowLi vs. HighLi group. These included some important genes and miRNAs such as calr, hsp90b1, hsp70, ssa-miR-125a-3p, ssa-miR-92b-5p, dre-miR-15a-3p and novel-m1018-5p. The DEGs were mainly enriched in the protein folding, processing and export pathways of the endoplasmic reticulum; the target genes of the DEMs were mainly enriched in the focal adhesion pathway. Furthermore, the association analysis revealed that the key genes were mainly enriched in the metabolic pathway. Interestingly, we found a significant increase in the number of genes and miRNAs involved in the regulation of heat stress during the temperature change from 24 °C to 27 °C. In addition, we examined the tissue expression characteristics of some key genes and miRNAs by qPCR, and found that calr, hsp90b1 and dre-miR-125b-2-3p were significantly highly expressed in the liver at 27 ℃, while novel-m0481-5p, ssa-miR-125a-3p, ssa-miR-92b-5p, dre-miR-15a-3p and novel-m1018-5p had the highest expression in the heart at 30℃. Finally, the quantitative expression trends of 10 randomly selected DEGs and 10 DEMs were consistent with the sequencing data, indicating the reliability of the results. Conclusions: In summary, this study provides some fundamental data for subsequent in-depth research into the molecular regulatory mechanisms of A. sapidissima response to heat stress, and for the selective breeding of high temperature tolerant varieties. Introduction: The seeds of Plantago asiatica L., a folk herb, are rich in polysaccharides that possess antioxidant, antidiabetic, and anti‐inflammatory properties. Polysaccharides with lower molecular weights generally exhibit higher biological activity, so a method to efficiently extract low‐molecular‐weight polysaccharides from P. asiatica L. seeds (PLPs) is needed. Objectives: The aim was to establish an efficient method for extracting polysaccharides from P. asiatica L. seeds while preserving their activity. Materials and Methods: Response surface methodology was applied to determine the optimal polysaccharide extraction conditions. Subsequently, the extracted polysaccharides were characterized to determine their monosaccharide composition, physicochemical properties, and molecular weight. Their antioxidant activity was evaluated by measuring their ability to scavenge DPPH and ABTS free radicals. Results: An extraction yield of 9.17% was achieved under an ethanol concentration of 18.0% (w/w), a K2HPO4 concentration of 27.8% (w/w), a solvent‐to‐material ratio of 30:1 (mL/g), an ultrasound power of 203 W, and an extraction time of 39 min. Structural analyses indicated that this method might cause physicochemical changes in the conformation of PLPs and induce the degradation of PLP side chains but not the backbone. The antioxidant assay results showed that the DPPH and ABTS radical scavenging rates of PLPs were 48.3% and 49.2%, respectively, while in the control group the radical scavenging rates were 35.5% and 37.1%, respectively. Conclusion: The established method for extracting polysaccharides from P. asiatica L. seeds is efficient and reliable. The polysaccharides could be used as an important resource with antioxidant activity. In this study, an ultrasound‐assisted aqueous two‐phase extraction (UAATPE) method was established to extract polysaccharides from Plantago asiatica L. seeds (PLPs). The conditions for the UAATPE process were optimized with response surface methodology. Structual analysis showed that the method could reduce the molecular weight of polysaccharides and showed stronger DPPH and ABTS free radical scavenging ability. Collichthys lucidus is a small fish found in offshore waters that is economically important for China. It is imperative to understand its distribution characteristics and driving factors. Based on survey data of trawl fishery resources offshore of Zhejiang province, China, in spring (April) and autumn (November) from 2018 to 2022, the spatial and temporal distributions of C. lucidus in this area were analyzed. The random forest (RF) model was used to determine the important marine factors affecting the distribution of C. lucidus. The relationship between the distributions of the important variables was analyzed. The results showed that C. lucidus was mainly distributed in coastal waters. The tail density of the species exhibited obvious seasonal variation and was significantly greater in autumn than in spring. The most important factor affecting the distribution of this species in spring and autumn was water depth. The bottom temperature, bottom salinity and dissolved oxygen concentration were also important influencing factors. The importance of these factors differed among the different seasons, while the chlorophyll a concentration and pH had no significant effect on the species distribution. This study revealed the distribution pattern of C. lucidus in offshore waters of Zhejiang Province and the influence of important marine factors on its distribution. The crystallization behavior of neodymium-based rare earth polybutadiene rubber (Nd-BR) is studied in the presence of small-molecule treated distillate aromatic extract (TDAE) and high-molecular-weight polybutadiene–isoprene copolymer rubber (BIR). Pronounced inhibitory effects on the crystallization of Nd-BR are exhibited by both materials, as evidenced by reductions in the crystallization temperature (Tc), melting point (Tm), and corresponding enthalpy change. It is found that, at equal concentrations, a greater influence on the crystallization rate is exerted by TDAE oils, whereas nucleation inhibition is more potently affected by BIR. Incomplete crystallization during cooling is exhibited by Nd-BR when the TDAE oil concentration reaches 40 parts per hundreds of rubber (PHR) (31 wt.%), or BIR achieves a 60 wt.% concentration; subsequently, a noticeable cold crystallization phenomenon is observed upon heating. Insights into the isothermal crystallization kinetics are offered by the data, which reveal that the Avrami index n value for Nd-BR predominantly ranges between 2.5 and 3.0. A decrease in the n value is induced by a small amount of TDAE oil, while a noticeable decline in the n value is observed only when the BIR concentration is 60 wt.%. A correlation between the crystallization activation energy, the concentration of TDAE oil and BIR, and the crystallization temperature is established; a negative activation energy is recorded, and a decrease in the crystallization rate is noted when both concentrations are low and the crystallization temperature exceeds −50 °C. In contrast, positive activation energy and an increase in the crystallization rate are observed when the BIR concentration reaches 60%, and the crystallization temperature resides between −50 °C and −70 °C.
