Aeromonas sobria is an opportunistic pathogen that can infect humans, animals and aquatic species, which is widely distributed in different aquatic environments and products. In recent years, with the rapid expansion of intensive aquaculture, the disease caused by A. sobria has occurred. This study aims to understand the pathogenic characteristics of A. sobria and provide scientific basis for the prevention and control of the epidemic. The dominant strain As012 was isolated from the diseased rainbow trout during the outbreak. Through physiological and biochemical experiments, sequencing and phylogenetic tree analysis of 16S rRNA and gyrB genes, the strain As012 was identified as A. sobria. The clinical signs of the diseased rainbow trout in the experimental infection were consistent with those in the farm, and the LD50 was 1.0 × 106.6 CFU/mL. The histopathological lesions in the gills, heart, liver, spleen and intestines were mainly extensive hemorrhage. In addition, eight virulence genes were screened from strain As012, including Act, Aer, AexT4, Alt, ahyB, ascV, Nuc and Hly. The strain As012 can grow in the environment with pH 1–11, temperature 8–43°C and NaCl concentration 0–8%. The drug sensitivity results showed that it was resistant to 12 antibiotics including penicillin G, vancomycin, and clindamycin, and highly sensitive to 16 antibiotics including cefazolin, ciprofloxacin, and furadantin. The results showed that A. sobria , the dominant strain isolated from diseased rainbow trout, was the main pathogen causing the epidemic in the farm. The strain As012 has a very wide range of growth and strong pathogenicity, causing widespread hemorrhaging in various tissues of rainbow trout. It is multi-resistant, but highly sensitive to cephalosporins, quinolones, nitrofurans and sulfonamides. Drought is one of the most severe environmental factors limiting plant growth and crop yield, necessitating the identification of genes that enhance drought resistance for crop improvement. Through screening an ethyl methyl sulfonate‐mutagenized rice mutant library, we isolated the PEG tolerance mutant 97‐1 (ptm97‐1), which displays enhanced resistance to osmotic and drought stress, and increased yield under drought conditions. A point mutation in OsMATE6 was identified as being associated with the drought‐resistant phenotype of ptm97‐1. The role of OsMATE6 in conferring drought resistance was confirmed by additional OsMATE6 knockout mutants. OsMATE6 is expressed in guard cells, shoots and roots and the OsMATE6‐GFP fusion protein predominantly localizes to the plasma membrane. Our ABA efflux assays suggest that OsMATE6 functions as an ABA efflux transporter; mutant protoplasts exhibited a slower ABA release rate compared to the wild type. We hypothesize that OsMATE6 regulates ABA levels in guard cells, influencing stomatal closure and enhancing drought resistance. Notably, OsMATE6 knockout mutants demonstrated greater yields under field drought conditions compared to wild‐type plants, highlighting OsMATE6 as a promising candidate for improving crop drought resistance. Located in the guard cell plasma membrane, OsMATE6 acts as an efflux transporter for abscisic acid (ABA), a key hormone regulating stomatal closure. This paper is devoted to studying the existence and nonexistence of monotone traveling wave solutions for a discrete diffusive vector-borne epidemic model without monotonicity. After a deep observation, we realize that the wave profile system of this three-dimensional non-monotone epidemic model has a special structure that can be transformed into a two-dimensional special cooperative system. Based on this, we pay attention to the existence and nonexistence of monotone traveling wave solutions for this special cooperative system. It is observed that whether the monotone traveling wave solutions exist or not depend on the so-called basic reproduction ratio $ R_0 $ of the corresponding kinetic system and the critical wave speed $ c^* $. More specifically, the model admits a positive monotone traveling wave solution if $ R_0 >1 $ and $ c\geq c^* $. The proofs are based on the upper and lower solutions, Schauder's fixed point theorem, and a limiting argument. When $ R_0>1 $ and $ 00 $, the non-existence of traveling wave solutions for this model is proved by using the contradictory approach and two-sided Laplace transforms. [ABSTRACT FROM AUTHOR]

Exploring the molecular mechanisms underlying insulin analogs is important for protein engineering to design innovative drug proteins. Insulin aspart (IAsp) and insulin degludec (IDeg) are representative examples of insulin analogs with distinct release profiles synthesized by targeted mutagenesis in protein engineering. Despite their importance in diabetes treatment, there remains a gap in our understanding of the molecular basis for their differential release mechanisms. In this study, ordinary molecular dynamics simulation and steered molecular dynamics are utilized to investigate the structural stability, solubility analysis, and monomer interactions of these insulins, with the aim to explain the mesoscale differences between the two insulin release mechanisms. Simulation findings have further been validated through experimental verification, shedding light on the intricate mechanisms underlying insulin release and providing valuable insights into pharmaceutical implications and potential advancements in the design of insulin therapy. A novel approach was applied to synthesize B6O using CaB6 and B2O3 powders as reactants. CaB6 and B2O3 were reacted to generate B6O and CaB2O4 at 1573 K, and the by‐product CaB2O4 was removed by acid leaching. It was found that the samples prepared in this work were oxygen deficient (B6Ox, x ≈.76–.77). The optimal molar ratio of CaB6 to B2O3 was determined to be 3:5, and excess B2O3 could not lead to a change in the value of oxygen occupancy. Scanning electron microscope images showed that regular holes similar to the morphologies of CaB6 particles were exhibited in the final samples, which could be explained by the diffusion of CaB6 during the reaction process. Objective SNOMED CT provides a standardized terminology for clinical concepts, allowing cohort queries over heterogeneous clinical data including Electronic Health Records (EHRs). While it is intuitive that missing and inaccurate subtype (or is-a) relations in SNOMED CT reduce the recall and precision of cohort queries, the extent of these impacts has not been formally assessed. This study fills this gap by developing quantitative metrics to measure these impacts and performing statistical analysis on their significance. Material and Methods We used the Optum de-identified COVID-19 Electronic Health Record dataset. We defined micro-averaged and macro-averaged recall and precision metrics to assess the impact of missing and inaccurate is-a relations on cohort queries. Both practical and simulated analyses were performed. Practical analyses involved 407 missing and 48 inaccurate is-a relations confirmed by domain experts, with statistical testing using Wilcoxon signed-rank tests. Simulated analyses used two random sets of 400 is-a relations to simulate missing and inaccurate is-a relations. Results Wilcoxon signed-rank tests from both practical and simulated analyses (P -values <.001) showed that missing is-a relations significantly reduced the micro- and macro-averaged recall, and inaccurate is-a relations significantly reduced the micro- and macro-averaged precision. Discussion The introduced impact metrics can assist SNOMED CT maintainers in prioritizing critical hierarchical defects for quality enhancement. These metrics are generally applicable for assessing the quality impact of a terminology's subtype hierarchy on its cohort query applications. Conclusion Our results indicate a significant impact of missing and inaccurate is-a relations in SNOMED CT on the recall and precision of cohort queries. Our work highlights the importance of high-quality terminology hierarchy for cohort queries over EHR data and provides valuable insights for prioritizing quality improvements of SNOMED CT's hierarchy.
