1. Evolutionary associations of drug resistance in Salmonella enteritidis in Jiamusi City

Abstract

ObjectiveThe study investigated the evolutionary associations of drug resistance in main serotype Salmonella enteritidis （S. enteritidis）in Jiamusi City， to provide a scientific basis for clinical treatment.MethodsS. enteritidis was isolated from food and patients with diarrhea. Serological typing was performed. Resistance against 16 antibiotics was investigated using the microbroth dilution method. Genes associated with antimicrobial resistance were analyzed by PCR， and the evolutionary relationships between strains were analyzed by pulsed-field gel electrophoresis （PFGE）.ResultsOf the 108 Salmonella strains identified， 55 （50.93%） were found to be S. enteritidis. Forty-nine （89.09%） of these strains showed antimicrobial resistance， with 33 strains （67.35%， 33/49） found to be resistant to multiple antibiotics. The highest resistance was against nalidixic acid and ampicillin， with 81.82% and 60.00% resistance， respectively. Homologies between 37% and 100% were observed among the 55 S. enteritidis strains.ConclusionHigh levels of multi-antimicrobial resistance were found in S. enteritidis in Jiamusi. A close relationship was found between， pathogen strain and drug resistance. S. enteritidis in the diarrhea of infected patients can cross-infect food. As S. enteritidis shows significant drug resistance in this region， it is of great importance to develop targeted diagnostic and treatment programs， as well as prevention and control measures， to ensure the safety and maintenance of public health.

Abstract

Recently, we notice that a pressure-based lattice Boltzmann (LB) method was established to recover the volume-averaged Navier-Stokes equations (VANSE), which serve as the cornerstone of various fluid-solid multiphase models. It decouples the pressure from density and exhibits excellent numerical performance, however, the widely adopted density-based LB scheme still suffers from significant spurious velocities and inconsistency with VANSE. To remedy this issue, a multiple-relaxation-time LB method is devised in this work, which incorporates a provisional equation of state in an adjusted density equilibrium distribution to decouple the void fraction from density. The Galilean invariance of the recovered VANSE is guaranteed by introducing a penalty source term in moment space, effectively eliminating unwanted numerical errors. Through the Chapman-Enskog analysis and detailed numerical validations, this novel method is proved to be capable of recovering VANSE with second-order accuracy consistently, and well-suited for handling void fraction fields with large gradients and spatiotemporal distributions., Comment: 23 pages

Abstract

Pathological diagnosis remains the definitive standard for identifying tumors. The rise of multimodal large models has simplified the process of integrating image analysis with textual descriptions. Despite this advancement, the substantial costs associated with training and deploying these complex multimodal models, together with a scarcity of high-quality training datasets, create a significant divide between cutting-edge technology and its application in the clinical setting. We had meticulously compiled a dataset of approximately 45,000 cases, covering over 6 different tasks, including the classification of organ tissues, generating pathology report descriptions, and addressing pathology-related questions and answers. We have fine-tuned multimodal large models, specifically LLaVA, Qwen-VL, InternLM, with this dataset to enhance instruction-based performance. We conducted a qualitative assessment of the capabilities of the base model and the fine-tuned model in performing image captioning and classification tasks on the specific dataset. The evaluation results demonstrate that the fine-tuned model exhibits proficiency in addressing typical pathological questions. We hope that by making both our models and datasets publicly available, they can be valuable to the medical and research communities., Comment: 10 pages, 2 figures

Abstract

Statin therapy is a cornerstone in the treatment of systemic vascular diseases. However, statins have failed to translate as therapeutics for pulmonary vascular disease. Early pulmonary vascular disease in the setting of congenital heart disease (CHD) is characterized by endothelial dysfunction, which precedes the more advanced stages of vascular remodeling. These features make CHD an ideal cohort in which to re-evaluate the potential pulmonary vascular benefits of statins, with a focus on endothelial biology. However, it is critical that the full gamut of the pleiotropic effects of statins in the endothelium are uncovered. The purpose of this investigation was to evaluate the therapeutic potential of simvastatin for children with CHD and pulmonary over-circulation, and examine mechanisms of simvastatin action on the endothelium. Our data demonstrate that daily simvastatin treatment preserves endothelial function in our shunt lamb model of pulmonary over-circulation. Further, using pulmonary arterial endothelial cells (PAECs) isolated from Shunt and control lambs, we identified a new mechanism of statin action mediated by increased expression of the endogenous Akt1 inhibitor, C-terminal modifying protein (CTMP). Increases in CTMP were able to decrease the Akt1-mediated mitochondrial redistribution of endothelial nitric oxide synthase (eNOS) which correlated with increased enzymatic coupling, identified by increases in NO generation and decreases in NOS-derived superoxide. Together our data identify a new mechanism by which simvastatin enhances NO signaling in the pulmonary endothelium and identify CTMP as a potential therapeutic target to prevent the endothelial dysfunction that occurs in children born with CHD resulting in pulmonary over-circulation.
