The Maipo River estuary is a low-inflow bar-built estuary that includes a protected wetland, which harbors a rich ecosystem. The estuary and wetland have been threatened by a persistent drought for more than a decade, which has resulted in greater salinity intrusion and increased residence times. Previous studies have described salinity and pollutants in estuaries; however, almost all have focused on deeper and/or wider estuaries with dimensions much larger than those of the small-scale Maipo River estuary. In this study, we used the numerical model FVCOM to simulate the dynamics of the Maipo River estuary under drought scenarios and explored the interactions between river discharge and tides in terms of saline intrusion and particle dispersal. The model was validated against observations collected during a field campaign near the river mouth. The simulations successfully reproduced the water surface elevation but underestimated salinity values, such that the vertical salinity structure observed in the field was not captured by the model in this shallow and morphologically complex estuary. Consequently, our model results provide qualitative insight related to salinity and baroclinic dynamics. Results of maximum saline intrusion showed an exponential decay with increasing river discharge, and the analysis of salinity intrusion time series revealed that droughts may cause permanent non-zero salinity levels in the estuary, potentially affecting ecological cycles. The incorporation of passive tracers showed that decreasing river discharge increases the residence time of particles by allowing the tracers to re-enter the estuary. The Lord Howe Volcanic Complex (LHVC) is part of a seamount chain associated with the detached continental crust of Zealandia. We present petrographic and geochemical data for the LHVC, including the first reported data for Balls Pyramid and the first trace element data for Lord Howe Island, clarifying the tectonic affiliation and general mantle source characteristics. Classification and discrimination diagrams based on concentrations and ratios of immobile elements, such as rare earth and high field strength elements, Th, Ti, Y, and Mg, show the LHVC comprises transitional to alkali basalts and at least two, and probably three, shield building complexes, consistent with an intraplate tectonic setting. Primitive-mantle normalised trace element patterns, variation in Dy/Yb for a restricted range of Dy/Dy*, Zr/Yb > 90, and variations in Zr/Hf and P2O5/TiO2 through the eruptive sequences indicate the mantle source may be a laterally heterogeneous enriched garnet peridotite that has undergone varying degrees of partial melting and carbonatitic metasomatism, both within the LHVC and along the Lord Howe Seamount Chain. The LHVC is thus an ideal natural laboratory for probing the probability of small intraplate volcanic fields originating via mantle plumes or from previously subducted slabs stagnating at the mantle transition zone. Governments have developed Indigenous procurement policies (IPPs) in Australia and other colonised countries with Indigenous populations to collaboratively close socioeconomic inequities experienced by Indigenous communities. These policies are widely used in the construction industry because of the volume of government spending on this sector and its significant social impact. However, recent research argues that Indigenous peoples' notions of social value are not reflected in IPP designs, implementation, or evaluation frameworks. To investigate this claim, this research uses a holistic Aboriginal evaluation framework called 'Ngaa-bi-nya' to investigate how IPPs promote social value creation. Using community-based participatory research principles, semi-structured interviews were held with owners and senior managers of Indigenous construction companies in Australia. Findings highlight how social value is created by IPPs when Indigenous contractors use strategic actions to improve Indigenous socioeconomic outcomes, social and cultural development, create stronger connections in the construction industry and promote self-determination. The findings provide new conceptual and operational insights into how IPPs can create social value through partnerships with Indigenous contractors. The insights can be used by buyers and builders to strengthen social value in their supply chains through new ways of collaborative working to support social value creation. Background: Suicide is a manner of death resulting from complex environmental and genetic risks that affect millions of people globally. Both structural and functional studies identified the hippocampus as one of the vulnerable brain regions contributing to suicide risk., Methods: We have identified the hippocampal tissue transcriptomes, gene ontology, cell type proportions, and dendritic spine morphology in controls (n = 28) and suicide decedents (n = 22). In addition, the transcriptomic signature in iPSC-derived neuronal precursor cells (NPCs) and neurons were also investigated in controls (n = 2) and suicide decedents (n = 2)., Results: The hippocampal tissue transcriptomic data revealed that NPAS4 gene expression was downregulated while ALDH1A2, NAAA, and MLXIPL gene expressions were upregulated in hippocampal tissue of suicide decedents. The gene ontology identified 29 significant pathways including NPAS4-associated gene ontology terms "excitatory post-synaptic potential", "regulation of postsynaptic membrane potential" and "long-term memory" indicating alteration of glutamatergic synapses in the hippocampus of suicide decedents. The cell type deconvolution identified decreased excitatory neuron proportion and an increased inhibitory neuron proportion providing evidence of excitation/inhibition imbalance in the hippocampus of suicide decedents. In addition, suicide decedents had increased dendric spine density in the hippocampus, due to an increase of thin (relatively unstable) dendritic spines, compared to controls. The CA3 region is central to hippocampal function during learning and memory and has a unique connectivity. CA3 pyramidal neurons are the targets of huge, excitatory mossy fiber synapses from DG axons and have a high degree of excitatory recurrent connectivity. Thus, inhibition likely plays an outsized importance in constraining excitation and shaping CA3 ensembles during learning and memory. Here, we investigate the function of a never-before studied set of dendrite-targeting, GABAergic neurons defined by expression of the synaptic adhesion molecule, Kirrel3. We discovered that activating Kirrel3-expressing GABAergic neurons specifically impairs memory discrimination and inhibits CA3 pyramidal neurons in novel contexts. Kirrel3 is required for DG-to-GABA synapse formation and variants in Kirrel3 are strong risk factors for neurodevelopmental disorders. Thus, our work suggests that Kirrel3-GABA neurons are a critical source of feed-forward inhibition from DG to CA3 during the encoding and retrieval of contextual memories, a function which may be specifically disrupted in some brain disorders., Competing Interests: DECLARATION OF INTERESTS The authors declare no competing interests.

Borrelia burgdorferi , the spirochetal agent of Lyme disease, utilizes a variety of strategies to evade and suppress the host immune response, which enables it to chronically persist in the host. The resulting immune response is characterized by unusually strong IgM production and a lack of long-term protective immunity. Previous studies in mice have shown that infection with B. burgdorferi also broadly suppresses host antibody responses against unrelated antigens. Here, we show that mice infected with B. burgdorferi and concomitantly immunized with recombinant severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein had an abrogated antibody response to the immunization. To further define how long this humoral immune suppression lasts, mice were immunized at 2, 4, and 6 weeks post-infection. Suppression of host antibody production against the SARS-CoV-2 spike protein peaked at 2 weeks post-infection but continued for all timepoints measured. Antibody responses against the SARS-CoV-2 spike protein were also assessed following antibiotic treatment to determine whether this immune suppression persists or resolves following clearance of B. burgdorferi . Host antibody production against the SARS-CoV-2 spike protein returned to baseline following antibiotic treatment; however, anti-SARS-CoV-2 IgM remained high, comparable to levels found in B. burgdorferi -infected but untreated mice. Thus, our data demonstrate restored IgG responses following antibiotic treatment but persistently elevated IgM levels, indicating lingering effects of B. burgdorferi infection on the immune system following treatment., Competing Interests: The authors declare no conflict of interest.
