Simple Summary: This study presents an integrated conservation framework for the globally threatened fishing cat, synthesizing both ex situ management and in situ strategies. The strategy aligns with the One Plan Approach, which views captive populations as part of a broader metapopulation network, and the Opportunities to Thrive framework, which emphasizes animal welfare in captivity. This study analyzed the genetic diversity of a recently initiated fishing cat captive breeding population by the West Bengal Zoo Authority, India, and conducted a population viability analysis. The results suggest that the current genetic diversity (56%) and population carrying capacity (30 individuals) can be sustained for over 100 years through a strategy of supplementation and harvesting. Introducing two adult males and females annually will allow for sustainable harvesting to supplement wild populations. Additionally, this study identified 21 potential reintroduction zones in the Sundarbans and Terai regions of West Bengal, using environmental criteria that favor fishing cat occurrence. This work offers a comprehensive blueprint for applying the One Plan Approach to ensure the long-term survival of the fishing cat. Here, we present an integrated framework for the conservation of a globally threatened wetland wild cat species, the fishing cat, by synthesizing optimal ex situ management practices and in situ conservation strategies. Further, we examined the genetic constitution of the founder population in a fishing cat captive breeding program that was recently initiated by the West Bengal Zoo Authority, India and conducted a population viability analysis to suggest how best to maintain the genetic diversity of the population. We found that the present genetic diversity of 56% and maximum carrying capacity of the captive population (30 individuals) can be maintained for more than 100 years with a combination of supplementation and harvesting. Keeping stochastic events in mind, the introduction of two adult males and females to the existing population each year will seamlessly allow the harvesting of two adult males and two adult females every alternate year to supplement wild populations. Further, we adopted the proposed integrated framework to delineate recommendations for the supplementation of wild populations in West Bengal. We used environmental criteria known to influence fishing cat occurrence to identify 21 potential reintroduction zones in the Sundarbans landscape and Terai region in northern West Bengal with habitable areas for the fishing cat that are larger than the maximum known species' home range. Simple Summary: In Thailand, the flat-headed cat, fishing cat, and Asiatic golden cat are among nine wild Felidae species requiring urgent conservation efforts. This study assessed routine sperm characteristics and detected protein biomarkers related to fertilization in these species. IZUMO1, involved in gamete interaction, showed the highest levels in the flat-headed cat. CRISP2 and CRISP3, crucial for sperm–egg fusion, had the highest levels in the fishing cat. Cleaved caspase-3, linked to DNA fragmentation, was found in all species and associated with decreased semen quality. These findings suggest that analyzing these biomarkers, alongside routine sperm characteristics, could enhance breeding management for wild Felidae, especially the flat-headed and fishing cats. The highest levels of IZUMO1 were found in both the sperm pellet and the seminal plasma of the flat-headed cat, as determined by immunoblotting. CRISP2, a sperm–egg fusion assisting protein, and CRISP3 were found in both the sperm pellet and the seminal plasma, and the highest levels were observed in the fishing cat. Positive correlations between certain semen parameters and IZUMO1, CRISP2, and CRISP3 expression were also demonstrated. Cleaved caspase-3 was found in all sperm samples in all three species and was associated with an increase in DNA fragmentation and a decrease in certain semen characteristics such as motility, viability, and intact acrosomes. Our results suggest that the analysis of IZUMO1, the CRISP family, and cleaved caspase-3, along with the routine sperm characteristics, may allow for better success in breeding management in wild Felidae, particularly in the flat-headed cat and the fishing cat. Introduction: Human-wildlife interactions are on the rise due to anthropogenic pressures. Understanding these interactions is crucial for preventing conflict and promoting coexistence, ultimately benefitting humans and wildlife. Objective: In this study, we analysed the interactions between humans and jaguars Panthera onca in Tortuguero National Park, Costa Rica. Methods: We examined data concerning jaguar sightings within Tortuguero National Park, Costa Rica compiled from 2000 to 2022. Results: A total of 381 jaguar sightings were recorded, of which 75% occurred in the coastal habitat; 81% occurred during the leatherback sea turtle Dermochelys coriacea (March-May) and the green sea turtle Chelonia mydas (June-October) nesting seasons. From photographic records, 26 individual jaguars were identified (14 females, 12 males). Furthermore, 9% of the sightings were reported within the boundaries of Tortuguero village, where at least 48 domestic dogs Canis lupus familiaris were attacked by jaguars. Conclusions: Despite the jaguar's cryptic nature, the data show an increase in the frequency and duration of jaguar sightings over the years. This trend is likely due to an increase in the local jaguar population and possibly an increase in jaguars' tolerance towards humans, possibly triggered by people's inappropriate behaviour (e.g. approaching jaguars while feeding, mating or with their cubs) during the sightings. Our study also highlights the potential for conflict due to jaguar predation of domestic dogs, which is increasing within the study area. Therefore, there is a need for management action to promote coexistence between humans and the local jaguar population in Tortuguero National Park, Costa Rica.
