Previous observational studies have found an association between sleep behaviors and grip strength, but the causal relationship remains unclear.Purpose: With the Mendelian randomization (MR) approach, the study aimed to determine the causal association between sleep traits (sleep duration, insomnia, daytime napping, sleep-wake disorders, chronotype) and low grip strength.Methods: The study used genetic variants from the genome-wide association study (GWAS) archived in UK Biobank and FinnGen. We assessed the potential causal relationship between sleep behaviors and grip strength using inverse variance weighting (IVW), weighted median (WM), and MR-Egger. Additionally, we performed sensitivity analyses using Cochran’s Q test, MR Egger Intercept test, funnel plots, and leave-one-out method.Results: We found that sleep duration is causally negatively associated with low grip strength (OR = 0.618, 95% CI = 0.424– 0.900, P = 0.012). Sleep-wake disorders have a positive association with low grip strength (OR = 1.018, 95% CI = 1.002– 1.034, P = 0.029). Reversely, high low grip strength risk was causally associated with increased daytime napping (OR = 1.018, 95% CI = 1.004– 1.032, P = 0.011).Conclusion: The study revealed causal associations between sleep duration, sleep-wake disorders, and low grip strength. Understanding their relationship helps in early clinical intervention to improve the life quality of the elderly.Keywords: sleep, low grip strength, Mendelian randomization

The chiral light field has attracted great attention owing to its interaction with chiral matter. The generation of chiral light fields with rich structures has become crucial as it can expand application scenarios. Herein, we introduce a chiral optical vortex lattice. As a whole, the optical vortex lattice has a chiral intensity distribution, with each spiral arm having sub-vortices (chiral phase). By using an expansion factor to adjust the involute of a circular lattice, this helical optical vortex lattice can be continuously varied from a circular lattice. The chirality of intensity and phase can be controlled independently. Furthermore, the optical tweezers using the lattice demonstrate the capability of sub-vortices to manipulate particle movement, with the chiral intensity determining the trajectory of particle motion. As the lattice possesses both intensity and phase chirality, it may also find potential applications in tasks such as chiral structure microfabrication. [ABSTRACT FROM AUTHOR]

In order to create a cost-effective Zr alloy with superior overall characteristics, Zr-xFe (x = 0, 2, 4, 6, 8, 10 at.%) binary alloys were prepared by a vacuum arc furnace system. The microstructure of these alloys was examined utilizing various techniques such as optical microscope, X-ray diffractometer, scanning electron microscopy, and transmission electron microscopy. The results showed that the addition of Fe effectively refined α-Zr slats, and the Zr3Fe compound phase was dispersed throughout the alloy. However, the composition line of the alloying elements in the phase diagram intersected with the Zr2Fe and β-Zr eutectic lines when the Fe content exceeded 6 at.%. Zr–Fe alloys were transformed from monocrystalline-eutectoid to eutectic-eutectoid reactions. The Zr3Fe compound phase accumulated inside the grain and at the grain boundary as a result of the eutectic reaction, which stabilized the microstructure of the Zr–Fe alloys. This seriously affected the mechanical and corrosion properties of the alloys. The mechanical and corrosion properties of the alloys were assessed by universal testing machine, dynamic microhardness tester, Vickers hardness tester, and electrochemical workstation. The mechanical and corrosion properties of Zr–2Fe were excellent due to the coupling effect of Zr3Fe and grain refinement. The surface of the Zr–Fe alloy was analyzed by X-ray photoelectron spectroscopy, revealing the presence of passivation film components ZrO2 and Fe2O3.

Information flow analysis can effectively model the security behavior and security properties of hardware design. However, the existing gate level information flow analysis methods cannot deal with large-scale designs due to computing power and verification effectiveness, and the register transfer level (RTL) information flow analysis methods require formal languages to rewrite hardware designs. This paper proposes a RTL hardware design information flow modeling and security verification method. Based on the RTL functional model, this method develops an information flow tracking logical model to model security behavior and security properties of RTL hardware designs from the perspective of information flow. This method can be integrated into EDA flows and uses EDA testing and verification tools to capture security property violations and detect security vulnerabilities based on non-interference security policy. The results on experiments with Trust-Hub hardware Trojan benchmarks show that the proposed method can effectively detect hardware Trojans.

By selecting five key operational parameters—inlet temperature, operating temperature, anode humidity, cathode humidity, and operating pressure—and using PEMFC power density, membrane temperature difference, and membrane water content as performance indicators, simulations were conducted using 3D CFD modeling and orthogonal experiments to identify the most significant factors influencing PEMFC performance and determine the optimal operational parameter combination. The results show that working pressure has the most significant impact on PEMFC performance. Pressure variations cause significant changes in diffusion and heat conduction within the PEMFC. When the working pressure increases from 1.0 MPa to 3.0 MPa, the power density increases by approximately 11.2 %, but the temperature difference within the membrane increases by about 30.5 %. The difference between operating temperature and inlet temperature directly leads to changes in the temperature difference within the membrane. Moreover, changes in operating temperature greatly affect the water content within the proton exchange membrane, which in turn impacts the PEMFC's power density. When the operating temperature increases from 60 °C to 100 °C, the water content within the membrane decreases by approximately 50.5 %, and the power density decreases by 60.4 %. By optimizing the combination of operating parameters using a comprehensive scoring method, the temperature difference within the membrane can be controlled at 27.78 °C while ensuring the power density is increased to 0.92 W/cm2.

Abstract Background Mitochondrial transplantation (MTx) has emerged as a novel therapeutic strategy, particularly effective in diseases characterized by mitochondrial dysfunction. This review synthesizes current knowledge on MTx, focusing on its role in modulating immune responses and explores its potential in treating post-cardiac arrest syndrome (PCAS). Methods We conducted a comprehensive narrative review of animal and human studies that have investigated the effects of MTx in the context of immunomodulation. This included a review of the immune responses following critical condition such as ischemia reperfusion injury, the impact of MTx on these responses, and the therapeutic potential of MTx in various conditions. Results Recent studies indicate that MTx can modulate complex immune responses and reduce ischemia–reperfusion injury post-CA, suggesting MTx as a novel, potentially more effective approach. The review highlights the role of MTx in immune modulation, its potential synergistic effects with existing treatments such as therapeutic hypothermia, and the need for further research to optimize its application in PCAS. The safety and efficacy of autologous versus allogeneic MTx, particularly in the context of immune reactions, are critical areas for future investigation. Conclusion MTx represents a promising frontier in the treatment of PCAS, offering a novel approach to modulate immune responses and restore cellular energetics. Future research should focus on long-term effects, combination therapies, and personalized medicine approaches to fully harness the potential of MTx in improving patient outcomes in PCAS.

Abstract The waiting time to deceased-donor kidney transplantation (DDKT) is long in Asian countries. We investigated the impact of sensitization and ABO blood type (ABO) on DDKT opportunity using two Korean cohorts: a hospital cohort from two centers and a national database. When recording the vegetation distribution with a camera, shadows can form due to factors like camera angle and direct sunlight. These shadows result in the loss of pixel information and texture details, significantly reducing the accuracy of fractional vegetation coverage (FVC) extraction. To address this issue, this study proposes an efficient double-exposure algorithm. The method reconstructs the pixel information in shadow areas by fusing normal-exposure and overexposed images. This approach overcomes the limitations of the camera’s dynamic range in capturing pixel information in shadowed regions. The study evaluates images with five levels of overexposure combined with five vegetation extraction indices. The aim is to determine the best-performing double-exposure combination under shadow conditions and the most suitable vegetation index. Experimental results reveal that the R² value between the best vegetation index and the FVC calculated from the fused double-exposure images and the ground truth FVC increases from 0.750 to 0.969. The root mean square error (RMSE) reduces from 0.146 to 0.046, and the intersection over union (IOU) increases from 0.856 to 0.943. These results demonstrate the excellent vegetation extraction capability of the double-exposure algorithm under shadow conditions, offering a straightforward and effective solution to low accuracy of FVC in shadowed areas.

BackgroundCardiac arrest (CA) is a significant public health concern. There is the high imminent mortality and survival in those who are resuscitated is substantively compromised by the post-CA syndrome (PCAS), characterized by multiorgan ischemia–reperfusion injury (IRI). The inflammatory response in PCAS is complex and involves various immune cell types, including lymphocytes and myeloid cells that have been shown to exacerbate organ IRI, such as myocardial infarction. Purinergic signaling, as regulated by CD39 and CD73, has emerged as centrally important in the context of organ-specific IRI. Hence, comprehensive understanding of such purinergic responses may be likewise imperative for improving outcomes in PCAS.MethodsWe have investigated alterations of immune cell populations after CA by utilizing rodent models of PCAS. Blood and spleen were collected after CA and resuscitation and underwent flow cytometry analysis to evaluate shifts in CD3+CD4+ helper T cells, CD3+CD8a+ cytotoxic T cells, and CD4/CD8a ratios. We then examined the expression of CD39 and CD73 across diverse cell types, including myeloid cells, T lymphocytes, and B lymphocytes.ResultsIn both rat and mouse models, there were significant increases in the frequency of CD3+CD4+ T lymphocytes in PCAS (rat, P < 0.01; mouse, P < 0.001), with consequently elevated CD4/CD8a ratios in whole blood (both, P < 0.001). Moreover, CD39 and CD73 expression on blood leukocytes were markedly increased (rat, P < 0.05; mouse, P < 0.01 at 24h). Further analysis in the experimental mouse model revealed that CD11b+ myeloid cells, with significant increase in their population (P < 0.01), had high level of CD39 (88.80 ± 2.05 %) and increased expression of CD73 (P < 0.05). CD19+ B lymphocytes showed slight increases of CD39 (P < 0.05 at 2h) and CD73 (P < 0.05 at 2h), while, CD3+ T lymphocytes had decreased levels of them. These findings suggested a distinct patterns of expression of CD39 and CD73 in these specific immune cell populations after CA.ConclusionsThese data have provided comprehensive insights into the immune response after CA, highlighting high-level expressions of CD39 and CD73 in myeloid cells.

The development of light composite rotor blades with acceptable load carrying capacity is an essential issue to be dealt with in the design of relatively large copter-type drones. In this paper, a method is established to determine the quasi-static blade load carrying capacity which is vital to drone reliability. The proposed method, which provides a systematic procedure to determine blade load carrying capacity, consists of three parts, namely, a procedure to determine the distributed quasi-static blade aerodynamic load via the Blade Element Momentum (BEM) approach, a finite element-based failure analysis method to identify the actual blade failure mode, and an optimization method to determine the actual blade load carrying capacity. The experimental failure characteristics (failure mode, failure thrust, failure location) of two types of composite sandwich rotor blades with different skin lamination arrangements have been used to verify the accuracy of the theoretical results obtained using the proposed load carrying capacity determination method. The skin lamination arrangement for attaining the optimal blade-specific load carrying capacity and the blade incipient rotational speed for safe drone operation has been determined using the proposed method.

Abstract Hand gesture plays an important role in many circumstances, which is one of the most common interactive methods in daily life, especially for disabled people. Human–machine interaction is another popular research topic to realize direct and efficient control, making machines intelligent and maneuverable. Here, a special human–machine interaction system is proposed and namedas computer‐vision (CV) based gesture‐metasurface interaction (GMI) system, which can be used for both direct beam manipulations and real‐time wireless communications. The GMI system first needs to select its working mode according to the gesture command to determine whether to perform beam manipulations or wireless communications, and then validate the permission for further operation by recognizing unlocking gesture to ensure security. Both beam manipulation and wireless communication functions are validated experimentally, which show that the GMI system can not only realize real‐time switching and remote control of different beams through gesture command, but also communicate with a remote computer in real time by translating the gesture language to text message. The proposed non‐contact GMI system has the advantages of good interactivity, high flexibility, and multiple functions, which can find potential applications in community security, gesture‐command smart home, barrier‐free communications, and so on.

Background: Mitochondrial dysfunction, which is triggered by systemic ischemia–reperfusion (IR) injury and affects various organs, is a key factor in the development of post-cardiac arrest syndrome (PCAS). Current research on PCAS primarily addresses generalized mitochondrial responses, resulting in a knowledge gap regarding organ-specific mitochondrial dynamics. This review focuses on the organ-specific mitochondrial responses to IR injury, particularly examining the brain, heart, and kidneys, to highlight potential therapeutic strategies targeting mitochondrial dysfunction to enhance outcomes post-IR injury. Methods and Results: We conducted a narrative review examining recent advancements in mitochondrial research related to IR injury. Mitochondrial responses to IR injury exhibit considerable variation across different organ systems, influenced by unique mitochondrial structures, bioenergetics, and antioxidative capacities. Each organ demonstrates distinct mitochondrial behaviors that have evolved to fulfill specific metabolic and functional needs. For example, cerebral mitochondria display dynamic responses that can be both protective and detrimental to neuronal activity and function during ischemic events. Cardiac mitochondria show vulnerability to IR-induced oxidative stress, while renal mitochondria exhibit a unique pattern of fission and fusion, closely linked to their susceptibility to acute kidney injury. This organ-specific heterogeneity in mitochondrial responses requires the development of tailored interventions. Progress in mitochondrial medicine, especially in the realms of genomics and metabolomics, is paving the way for innovative strategies to combat mitochondrial dysfunction. Emerging techniques such as mitochondrial transplantation hold the potential to revolutionize the management of IR injury in resuscitation science. Conclusions: The investigation into organ-specific mitochondrial responses to IR injury is pivotal in the realm of resuscitation research, particularly within the context of PCAS. This nuanced understanding holds the promise of revolutionizing PCAS management, addressing the unique mitochondrial dysfunctions observed in critical organs affected by IR injury.

This paper systematically analyzes the current status of the mental health of young children, summarizing both the positive and negative aspects. It explores the underlying causes of these phenomena and proposes corresponding suggestions for improvement. Through a detailed analysis of the literature, the paper highlights how increasing attention and the popularization of mental health education have allowed more children to access mental health knowledge and counselling early on. Despite this progress, challenges persist due to modern lifestyle changes, rapid technological advancements, and urbanization, which negatively impact children’s mental health. Many children suffer from a lack of emotional support, high psychological pressure, and exposure to harmful information within their family and social environments. To address these issues, the paper emphasizes the need for improvements in family environments, societal support, and policies, as well as enhancements to the mental health service system. The recommendations include fostering better parent-child communication, using electronic devices judiciously, and creating supportive home environments. On a broader scale, it calls for stronger policy support, improved mental health services, and widespread mental health education.

Triterpenoidal saponins have been reported to be able to restrain SARS-CoV-2 infection. To isolate antiviral compounds against SARS-CoV-2 from the leaves of Aster koraiensis, we conducted multiple steps of column chromatography. We isolated six triperpenoidal saponins from A. koraiensis leaves, including three unreported saponins. Their chemical structures were determined using HR-MS and NMR data analyses. Subsequently, we tested the isolates to assess their ability to impede the entry of the SARS-CoV-2 pseudovirus (pSARS-CoV-2) into ACE2+ H1299 cells and found that five of the six isolates displayed antiviral activity with an IC50 value below 10 μM. Notably, one unreported saponin, astersaponin J (1), blocks pSARS-CoV-2 in ACE2+ and ACE2/TMPRSS2+ cells with similar IC50 values (2.92 and 2.96 μM, respectively), without any significant toxic effect. Furthermore, our cell-to-cell fusion and SARS-CoV-2 Spike-ACE2 binding assays revealed that astersaponin J inhibits membrane fusion, thereby blocking both entry pathways of SARS-CoV-2 while leaving the interaction between the SARS-CoV-2 Spike and ACE2 unaffected. Overall, this study expands the list of antiviral saponins by introducing previously undescribed triterpenoidal saponins isolated from the leaves of A. koraiensis, thereby corroborating the potency of triterpenoid saponins in impeding SARS-CoV-2 infection.

AbstractBackgroundMethodsResultsConclusionThe essential tremor (ET) course to 54 months post-unilateral VIM/PSA magnetic resonance-guided focused ultrasound (MRgFUS) in the treated arm (TA) and non-treated arm (NTA) of 12 patients is reported.Tremor severity was rated using Bain Findley spirography (BFS) scores in the TA and NTA. We divided follow-up into ‘Early’ (0–6 months) and ‘Late’ (6–54 months) phases, to minimise the effect of peri-lesion oedema resolution on the latter.The mean baseline BFS score was 6.2 in TA and 5.7 in the NTA. After unilateral VIM/PSA MRgFUS, mean BFS improved in TA at all subsequent time points (p < 0.001), with no significant differences between BFS scores at consecutive assessments or between 1 and 54 months, while the NTA BFS scores worsened between 12 and 24 months (p < 0.003). Three patients showed worsening of their TA BFS scores and an increasing NTA–TA BFS difference, indicating slower tremor worsening in TA compared to NTA, whilst one patient showed a greater rate of worsening in the TA compared to NTA BFS.After 54 months, the beneficial effect of MRgFUS is usually maintained with any worsening of BFS scores in TA slower than in NTA. Layered vanadium oxides with preintercalated cations have been recognized to be among the most promising cathode materials for aqueous zinc-ion batteries (AZIBs). However, their underlying structure-property relationships are still poorly understood owing to the lack of systematic comparison. Herein, a series of layered V 2 O 5 nanobelts including single-layer α-V 2 O 5 and bilayer hydrated δ-V 2 O 5 with preintercalation (M x V 2 O 5 · n H 2 O, M = Mg 2+ , Ca 2+ , or Ba 2+ ) with similar morphology are fabricated through a controllable synthesis protocol as models. The side-by-side comparisons of the energy storage performances and related kinetics in these samples decouple the reconstruction effects of nanostructuring and preintercalation. Results demonstrate that nanostructuring tends to improve the capacity and rate retention of α-V 2 O 5 but leaves the rapid capacity decay unsettled, whereas preintercalation can convert α-V 2 O 5 into δ-V 2 O 5 with simultaneously enhanced discharge capacity, rate retention, and cycling stability. Regulation of the cations' species/content and their associated structural water appears to accelerate the electron/ion transport in V 2 O 5 and stabilize the framework of V 2 O 5 , thereby boosting the energy storage properties. The optimized sample with a stoichiometric formula of Mg 0.255 V 2 O 5 ·0.809H 2 O exhibits outstanding comprehensive energy storage performance that is comparable to the most advanced vanadium oxide hydrate cathodes reported in the literature. The quantification and understanding of the reconstruction effects of nanostructuring and preintercalation in this work offer insights into the structural engineering of advanced cathodes for AZIBs and beyond.

Background: Stroke recurrence readmission poses an additional burden on both patients and healthcare systems. Risk stratification aims to accurately divide patients into groups to provide targeted interventions at reducing readmission. To accurately predict short and intermediate-term risks of readmission and provide information for further temporal risk stratification, we developed and validated an interpretable machine learning risk scoring tool., Methods: In this retrospective study, all stroke admission episodes from January 1st 2015 to December 31st 2019 were obtained from the Shanghai Health and Health Development Research Centre database, which covers medical records of all patients hospitalized in 436 medical institutes in Shanghai. The outcome was time to stroke recurrence readmission within 90 days post discharge. The Score for Stroke Recurrence Readmission Prediction (SSRRP) tool was derived via an interpretable machine learning-based system for time-to-event outcomes. SSRRP as six-variable survival score includes sequelae, length of stay, type of stroke, random plasma glucose, medical expense payment, and number of hospitalizations., Results: A total of 339,212 S admission episodes were finally included in the whole cohort. Among them, 217,393 episodes were included in the training dataset, 54,347 episodes in the internal validation dataset, and 67,472 in the temporal validation dataset. Readmission within 90 days was documented in 33922(9.97 %) episodes, with a median time to emergency readmission of 19 days (Interquartile range: 8-43). In the temporal validation dataset, the SSRRP achieved an integrated area under the curve of 0.730(95 % CI, 0.724-0.737). In addition, SSRRP demonstrated good calibration and clinical benefit rate., Conclusions: In this retrospective cohort study, the SSRRP, a parsimonious and point-based scoring tool, was developed to predict the risk of recurrent readmission for stroke. Background: Fibroinflammatory cholangiopathies, such as primary sclerosing cholangitis (PSC) and primary biliary cholangitis (PBC), are characterized by inflammation and biliary fibrosis, driving disease-related complications. In biliary fibrosis, cholangiocytes activated by transforming growth factor-β (TGFβ) release signals that recruit immune cells to drive inflammation and activate hepatic myofibroblasts to deposit the extracellular matrix (ECM). TGFβ regulates stearoyl-CoA desaturase (SCD), an enzyme that catalyzes the synthesis of monounsaturated fatty acids, in stimulating fibroinflammatory lipid signaling. However, the role of SCD or its inhibitor, Aramchol, has not been investigated in biliary fibrosis or TGFβ-mediated cholangiocyte activation., Method: 10-16-week-old multi-drug resistance 2 knockout (Mdr2 -/- ) and 3,5-diethoxycarboncyl-1,4-dihydrocollidine (DDC) diet-fed mice were orally gavaged daily with Aramchol at 12.5 mg/kg/day for 4 and 3 weeks, respectively. Liver and serum were harvested for the assessment of fibrosis and inflammation. Transformed human cholangiocyte cells (H69) and mouse large biliary epithelial cells (MLEs) were used to test the effects of the SCD inhibitor, Aramchol, at varying doses on TGFβ-mediated expression of fibroinflammatory signals and were confirmed in PSC-derived cholangiocytes (PSC-Cs) using ELISA, qPCR, and Western blot analyses., Results: Aramchol treatment of Mdr2 -/- mice with established biliary fibrosis (treatment) and DDC diet-induced (prevention) models of cholestatic injury and fibrosis demonstrated significant reductions in both measures of ECM synthesis (mRNA expression of ECM components in the liver), collagen content of the liver (picrosirius red staining and hydroxyproline content) and myofibroblast activation (αSMA staining). Il6 and Tnfa were also reduced with Aramchol in the liver. RNA-seq analysis of H69 cells showed that Aramchol co-treatment led to significant inhibition of TGFβ-induced hepatic fibrosis pathways while upregulating peroxisome proliferator-activated receptor (PPAR) signaling. SCD expression was significantly increased in TGFβ-treated H69 cells (2-fold, p<0.05). Aramchol in a dose-dependent manner significantly attenuated the increased expression of the fibrotic marker, plasminogen activator inhibitor-1 (PAI-1/SERPINE1), and hepatic stellate cell-activating genes ( VEGFA and PDGFB ) in TGFβ-activated H69 and MLEs. Aramchol also markedly reduced the expression of the inflammatory cytokine, interleukin 6 (IL6). SCD siRNA knockdown produced similar results in H69 cells. Furthermore, in PSC-Cs, the expressions of SCD, VEGFA and IL6 were significantly reduced with Aramchol. The expression of the anti-fibroinflammatory factors PPARα and -γ were modestly increased in cholangiocyte cell lines with increased expression of PPAR-responsive genes and increased nuclear binding of DNA PPAR response elements with Aramchol co-treatment compared to TGFβ only., Conclusion: Aramchol, an SCD inhibitor, both attenuates and prevents biliary fibrosis in mouse models of cholestatic injury and fibrosis. This effect is partially due to Aramchol inhibiting TGFβ-induced fibroinflammatory mediators in cholangiocytes by upregulating PPARα and -γ expression and activity. These findings, along with Aramchol's excellent safety profile in clinical trials, provide the rationale for assessing Aramchol in further clinical studies in patients with biliary fibrosis, particularly PSC, where a treatment is desperately needed.

German chamomile (Matricaria chamomilla L.) is a traditional medicinal aromatic plant, and the sesquiterpenoids in its flowers have important medicinal value. The (E)-β-farnesene (EβF) is one of the active sesquiterpenoid components and is also a major component of aphid alarm pheromones. In this study, two EβF synthase (βFS) genes (McβFS1 and McβFS2), were cloned from German chamomile. Subcellular localization analysis showed that both McβFS1 and McβFS2 were localized in the cytoplasm and nucleus. Tissue-specific expression analysis revealed that McβFS1 and McβFS2 were expressed in all flower stages, with the highest levels observed during the tubular flower extension stage. Prokaryotic expression and enzyme activity results showed that McβFS1 and McβFS2 possess catalytic activity. Overexpression of McβFS1 and McβFS2 in the hairy roots of German chamomile led to the accumulation of EβF, demonstrating enzyme activity in vivo. The promoters of McβFS1 and McβFS2 were cloned and analyzed. After treating German chamomile with methyl jasmonate (MeJA) and methyl salicylate (MeSA), the transcription levels of McβFS1 and McβFS2 were found to be regulated by both hormones. In addition, feeding experiments showed that aphid infestation upregulated the expression levels of McβFS1 and McβFS2. Background: The metastatic cascade, a multifaceted and highly aggressive process, is the primary cause of mortality. The survival of quiescent cancer cells in circulatory system during metastasis is crucial, yet our comprehension is constrained by the absence of universally accepted quiescent cancer models., Method: We developed a quiescent cancer cell model using high-density cultivation. Based on the scRNA-seq analysis, IP-MS, metabolomics, mouse lung metastasis models, cholesterol assay, PLA and other molecular experiments, we explored the molecular mechanism. Immunofluorescence, atomic force microscope, FluidFM, and shear stress stimulation were used to analyze the cytoskeleton and membrane properties contributing to mechanical force resistance., Result: We established a quiescent cancer cell model induced by high-density cultivation. Single-cell RNA sequencing (scRNA-seq) analysis reveals that CDC25A plays a crucial role in the transition to quiescence, with its expression significantly elevated in the quiescent state. Depletion of CDC25A leads to an increased proliferative capacity, and reduced metastasis under high-density conditions. Mechanistically, upregulated CDC25A in quiescent cells enhances cholesterol metabolism via endosome pathways, leading to cell cycle arrest. This increase in cholesterol reinforces the cytoskeleton, alters membrane properties, and improves resistance to mechanical forces in circulatory system., Conclusion: CDC25A significantly increased the cholesterol metabolism through endosome pathway in quiescent cancer cells, leading to the significant changes in cytoskeleton and membrane properties so as to enhance the resistance of mechanical force in circulatory system, facilitating lung metastasis. Walking-assistive devices require adaptive control methods to ensure smooth transitions between various modes of locomotion. For this purpose, detecting human locomotion modes (e.g., level walking or stair ascent) in advance is crucial for improving the intelligence and transparency of such robotic systems. This study proposes Deep-STF, a unified end-to-end deep learning model designed for integrated feature extraction in spatial, temporal, and frequency dimensions from surface electromyography (sEMG) signals. Our model enables accurate and robust continuous prediction of nine locomotion modes and 15 transitions at varying prediction time intervals, ranging from 100 to 500 ms. Experimental results showcased Deep-STP's cutting-edge prediction performance across diverse locomotion modes and transitions, relying solely on sEMG data. When forecasting 100 ms ahead, Deep-STF achieved an improved average prediction accuracy of 96.60%, outperforming seven benchmark models. Even with an extended 500ms prediction horizon, the accuracy only marginally decreased to 93.22%. The averaged stable prediction times for detecting next upcoming transitions spanned from 31.47 to 371.58 ms across the 100-500 ms time advances. Although the prediction accuracy of the trained Deep-STF initially dropped to 71.12% when tested on four new terrains, it achieved a satisfactory accuracy of 92.51% after fine-tuning with just 5 trials and further improved to 96.27% with 15 calibration trials. These results demonstrate the remarkable prediction ability and adaptability of Deep-STF, showing great potential for integration with walking-assistive devices and leading to smoother, more intuitive user interactions.

The extensive use of fossil fuels has sped up the global development of the world economy and is accompanied by significant problems, such as energy shortages and environmental pollution. Solar energy, an inexhaustible and clean energy resource, has emerged as a promising sustainable alternative. Light irradiation can be transformed into electrical/chemical energy, which can be used to remove pollutants or transform contaminants into high-value-added chemicals through photocatalytic reactions. Therefore, photocatalysis is a promising strategy to overcome the increasing energy and environmental problems. As is well-known, photocatalysts are key components of photocatalytic systems. Among the widely investigated photocatalysts, titanium dioxide (TiO 2 ) has attracted great attention owing to its excellent light-driven redox capability and photochemical stability. However, its poor solar light response and rapid recombination of electron-hole pairs limit its photocatalytic applications. Therefore, strategies to enhance the photocatalytic activity of TiO 2 by narrowing its bandgap and inhibiting the recombination of charges have been widely accepted. Constructing heterojunctions with other components, including cuprous oxide (Cu 2 O), has especially narrowed the bandgap, providing a promising means of solving the present challenges. This paper reviews the advances in research on heterostructured Cu 2 O/TiO 2 photocatalysts, such as their synthesis methods, mechanisms for the enhancement of photocatalytic performance, and their applications in hydrogen production, CO 2 reduction, selective synthesis, and the degradation of pollutants. The mechanism of charge separation and transfer through the Cu 2 O/TiO 2 heterojunctions and the inherent factors that lead to the enhancement of photocatalytic performance are extensively discussed. Additionally, the current challenges in and future perspectives on the use of heterostructured Cu 2 O/TiO 2 photocatalysts are also highlighted.

The relationship between COVID-19 and ischemic stroke (IS) has attracted significant attention, yet the precise mechanism at the gene level remains unclear. This study aims to reveal potential biomarkers and drugs for COVID-19-related IS through bioinformatics methods. We collected two gene expression profiling datasets, GSE16561 and GSE213313, and selected GSE179879 and GSE196822 as validation sets for analysis. Through analysis, we identified 77 differentially expressed genes (DEGs) shared between COVID-19 and IS. Further gene enrichment analysis revealed that these genes are primarily involved in immune regulation. By constructing a protein-protein interaction network, we screened out nine hub genes, including FCGR3A, KLRB1, IL2RB, CD2, IL7R, CCR7, CD3D, GZMK, and ITK. In LASSO regression analysis, we evaluated the ROC curve's area under the curve (AUC) scores of key genes to assess their diagnostic accuracy. Subsequently, we performed random forest (RF), Support Vector Machine Recursive Feature Elimination (SVM-RFE), and neural network construction on hub genes to ensure accurate diagnosis of IS. Finally, by intersecting the results of three algorithms (LASSO regression, random forest, and SVM), CD3D and ITK were identified as the ultimate key genes. Based on this, we predicted potential targeted drug Blinatumomab. There is an urgent need to identify predictive biomarkers of Parkinson's disease (PD) with cognitive impairment (PDCI) in order to individualize patient management, ensure timely intervention, and improve prognosis. The aim of this study was to screen for these biomarkers by comparing the plasma proteome and metabolome of PD patients with or without cognitive impairment. Proteomics and metabolomics analyses were performed on a discover cohort. A machine learning model was used to identify candidate protein and metabolite biomarkers of PDCI, which were validated in an independent cohort. The predictive ability of these biomarkers for PDCI was evaluated by plotting receiver operating characteristic curves and calculating the area under the curve (AUC). Moreover, we assessed the predictive ability of these proteins in combination with neuroimaging. In the discover cohort (n = 100), we identified 25 protein features with best results in the machine learning model, including top-ranked PSAP and H3C15. The two-proteins were used for model construction, achieving an Area under the curve (AUC) of 0.951 in the train set and AUC of 0.981 in the test set. Similarly, the model gives a rank list of endogenous metabolite features, Glycocholic Acid and 6-Methylnicotinamide were two top features. Combining these two markers further got the AUC of 0.969 in train set and 0.867 in the test set. To validate the performance of the protein biomarkers, we performed targeted analysis of selected proteins (H3C15 and PSAP) and proteins likely associated with PDCI (NCAM2 and LAMB2) using parallel reaction monitoring in validation cohort (n = 116). The AUC of the classifier built with H3C15 and PSAP is 0.813. Moreover, when combining H3C15, PSAP, NCAM2, and LAMB2, the model achieved AUC of 0.983 in the train set, AUC of 0.981 in the test set, and AUC of 0.839 in the validation set. Furthermore, we verified that these protein markers we discovered can improve the predictive effect of neuroimaging on PDCI: the classifier built with neuroimaging features had AUC of 0.833, which improved to 0.905 when combined with H3C15. Taken together, our integrated proteomics and metabolomics analysis successfully identified potential biomarkers for PDCI. Background: Complete preoperative comprehension of the adjacent structures of the kidney and location of renal vessels is essential for robot-assisted partial nephrectomy (RAPN). The effectiveness of three-dimensional (3D) visualization techniques in improving perioperative outcomes of RAPN has been inconsistent and has not been reported in Northeastern China., Methods: In this cohort study, we reviewed patients with renal tumours who underwent RAPN between April 2019 and April 2024. Three-dimensional visualization models were reconstructed to evaluate resectability parameters, including vascular variations, collection system infiltration, and lymphatic involvement. Subsequently, a meta-analysis combining previous studies utilising 3D visualization techniques for partial nephrectomy was conducted., Results: Of the 324 patients in the cohort, 147 were preoperatively evaluated using the 3D technology. Group 3D had significantly less estimated blood loss (P < 0.001) and a shorter operative time (P = 0.016) than in group No 3D. We also found that the rates of intraoperative ultrasound use (P = 0.015), intraoperative complications (P = 0.007), intraoperative transfusions (P = 0.007), and postoperative Clavien complications (P < 0.001) in group 3D were significantly lower than in group No 3D. The above findings were consistent in the subgroup with R.E.N.A.L. ≥ 8 points partly. Furthermore, a meta-analysis identified 11 studies that included 1522 patients who underwent RAPN. Use of 3D visualization technology resulted in decreased 55 % risk of opening the collecting system (Risk Ratio [RR] = 0.45[0.22-0.92], P = 0.030) and 79 % incidence of conversion to radical nephrectomy (RR = 0.21[0.08-0.57], P = 0.002). Background: According to Japanese law, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection during the COVID-19 pandemic falls under Category 2, implying that it necessitates hospitalization, isolation, and significant government participation. The category of infection was lowered to Category 5 in May 2023, meaning that individuals were going back to live their lives as they did before the COVID-19 pandemic. This study aimed to explore changes in the prevalence of non-motor symptoms over a longer four-year period, spanning before and after the COVID-19 pandemic., Methods: We conducted a questionnaire survey between January and February in the following years: 2021, 2022, 2023, and 2024., Results: The Parkinson's fatigue scale score in 2021 (mean: 35.3) increased significantly in 2022 (49.84), 2023 (49.33), and 2024 (54.71) (p < 0.001). After adjusting for baseline score using a mixed linear model with random intercept, the Parkinson's fatigue scale was significantly increased by 15.9 points (95 % CI: 10.9 to 20.9) in 2022, 13.1 points (7.9-18.3) in 2023, and 16.9 points (11.3-22.6) in 2024 independently of all potential confounders including other non-motor symptoms., Conclusions: The four years of longitudinal observation during the recurrent COVID-19 pandemic revealed that patients with PD experienced an increase in fatigue. Tinnitus is a phantom auditory sensation that commonly co-occurs with hearing loss. Both tinnitus and hearing loss can impact the quality of life, emotional well-being, and cognitive functioning of the affected individuals. While previous studies have highlighted structural alterations in hearing loss and/or tinnitus, the fundamental neural mechanisms underpinning tinnitus severity remain poorly understood. In this study, we conducted a voxel-based morphometry to investigate gray matter (GM) volume differences among groups of participants with varying tinnitus severity and hearing status, and controls within a large sample. We observed reduced GM volume in the left anterior insula and right planum polare in participants with hearing loss, regardless of their tinnitus status, compared to normal hearing controls. We noted decreased GM volume in the bilateral anterior and posterior insula for those with tinnitus and normal hearing compared to a normal hearing control group. Further, the tinnitus with hearing loss group showed decreased GM volume in the left planum polare, left inferior temporal gyrus, bilateral anterior temporal gyri, and right superior frontal gyrus compared to the normal hearing control group, suggesting a combined effect of hearing loss and tinnitus. While tinnitus severity did not show a significant overall effect, there was a significant positive correlation between tinnitus distress and GM volume in bilateral planum polare. Symmetrical batteries hold great promise as cost-effective and safe candidates for future battery technology. However, they realistically suffer low energy density due to the challenge in integrating high specific capacity with high voltage plateau from the limited choice of bipolar electrodes. Herein, a high-voltage all-V 2 O 5 symmetrical battery with clear voltage plateau is conceptualized by decoupling the cathodic/anodic redox reactions based upon the episteme of V 2 O 5  intercalation chemistry. As the proof-of-concept, a hierarchical V 2 O 5 -carboncomposite (VO-C) bipolar electrode with boosted electron/ion transport kinetics is fabricated, which shows high performance as both cathode and anode in their precisely clamped working potential windows. Accordingly, the symmetrical full-battery exhibits a high capacity of 174 mAh g -1 along with peak voltage output of above 2.9 V at 0.5C, remarkable capacity retention of 81% from 0.5C to 10C, and good cycling stability of 70% capacity retention after 300 cycles at 5C. Notably, its energy density reaches 429 Wh kg -1 at 0.5C estimated by the cathode mass, which outperforms most of the existing Li/Na/K-based symmetrical batteries. Ray-wave structured vortex beams have attracted increasing attention due to their unique spatial geometric coupling to control complex orbital angular momentum (OAM). Still, current models were constrained by circular symmetry with limited modulation freedom. Herein, we propose a generalized class of ray-wave light fields called Mathieu geometric modes (MGMs) fulfilling the form of a stationary coherent state but based on a set of helical Mathieu modes (HMMs), in which geometrically tunable elliptical accelerating vortices are obtained by tuning their eccentricity-related parameters. MGMs also possess intriguing properties of coordinate transformation, self-healing, and multilayer tunable angular acceleration upon propagation. MGMs have higher degrees of freedom to control spatial accelerating vortices, paving the way for higher-dimensional optical tweezers and complex particle manipulation.

This study explored potential associations of bacterial overgrowth in the small intestine, as detected based on levels of hydrogen and methane in breath after lactulose consumption, with cortical thickness and resting-state functional connectivity in different brain regions. Prospective comparison of 35 patients with Parkinson's disease (PD) involving mild cognitive impairment, 35 patients with PD with normal cognitive function and 17 healthy controls showed the largest level of hydrogen alone and the largest combined level of hydrogen and methane in patients with mild cognitive impairment. The comparison also revealed a significant negative correlation between those levels and thickness of the right insular cortex. Mild cognitive patients showed different functional connectivity between the right insula and cognition-related brain networks from normal cognitive patients. Our results suggest that bacterial overgrowth in the small intestine may contribute to cortical thinning and alterations in resting-state functional connectivity in PD involving mild cognitive impairment. Background: Huntington's disease (HD) is a neurodegenerative disorder for which effective therapies are currently lacking. Studies suggest that increasing physical activity (PA) and reducing leisure sedentary behavior (LSB) mitigate the progression of HD, but their causal relationship with the age at onset (AAO) of HD remains uncertain. To investigate this, we conducted the Two-sample Mendelian Randomization (MR)., Methods: Exposure were retrieved from the UK BioBank's (UKB) Genome-Wide Association Study (GWAS). PA included accelerometer-based average PA, vigorous PA, self-reported moderate-to-vigorous PA (MVPA), and light do-it-yourself activity. LSB included television (TV) time, computer time, and driving time. Outcome came from the GWAS of the GEM-HD Consortium. We applied several MR methods such as inverse variance weighted (IVW), MR-Egger regression, weighted median (WM) for sensitivity analysis., Results: Increases in light PA (β = 8.53 years, 95 % CI = 10.64 to 44.09, P = 0.001) and accelerometer-based vigorous PA (β = 5.18, 95 % CI = 0.92 to 9.43, P = 0.017) delayed AAO of HD, while longer TV time was associated with earlier AAO of HD (β = -2.88 years, 95 % CI = -4.99 to -0.77, P = 0.007). However, other PA and LSB phenotypes did not significantly affect AAO of HD., Conclusion: The study revealed a unidirectional causality between PA, LSB and the AAO of HD. Laryngeal squamous cell carcinoma (LSCC) is a prevalent human cancer with a complex pathogenesis that remains incompletely understood. Here, we unveil a long non-coding RNA (lncRNA) associated with LSCC tumorigenesis and progression. LOC730101 exhibits significant overexpression in human LSCC tissues, and elevated LOC730101 levels correlate with malignant clinicopathological characteristics. Moreover, we demonstrate that LOC730101 is encapsulated into exosomes in an hnRNPA2B1-dependent manner, serving as a promising plasma biomarker for discriminating LSCC patients from healthy individuals (AUC = 0.92 with 89.36% sensitivity and 86.36% specificity). Exosomes derived from LSCC cells enhance the viability, DNA synthesis rate, and invasiveness of normal nasopharynx epithelial cells, with pronounced effects observed upon LOC730101 overexpression. Additionally, exosomal LOC730101 promotes tumor growth in vivo. Mechanistically, exosomal LOC730101 internalization by normal nasopharynx epithelial cells leads to increased H3K4me3 levels on the p38 MAPK gamma (p38γ) promoter via direct interaction with hnRNPA2B1. This interaction activates p38γ transcription, ultimately driving LSCC tumorigenesis. Parthenolide is a germacrane sesquiterpene lactone separated from the traditional medicinal plant feverfew. Previous studies have shown that parthenolide possesses many pharmacological activities, involving anti-inflammatory and anticancer activities. However, the antitumor mechanism of parthenolide has not been fully elucidated. Thus, we investigate the potential antitumor mechanisms of parthenolactone. We predicted through network pharmacology that parthenolide may target HIF-1α to interfere with the occurrence and development of cancer. We found that parthenolide inhibited PD-L1 protein synthesis through mTOR/p70S6 K /4EBP1/eIF4E and RAS/RAF/MEK/MAPK signaling pathways and promoted PD-L1 protein degradation through the lysosomal pathway, thereby inhibiting PD-L1 expression. Immunoprecipitation and Western blotting results demonstrated that parthenolide inhibited PD-L1 expression by suppressing HIF-1α and RAS cooperatively. We further proved that parthenolide inhibited cell proliferation, migration, invasion, and tube formation via down-regulating PD-L1. Moreover, parthenolide increased the effect of T cells to kill tumor cells. In vivo xenograft assays further demonstrated that parthenolide suppressed the growth of tumor xenografts. Collectively, we report for the first time that parthenolide enhanced T cell tumor-killing activity and suppressed cell proliferation, migration, invasion, and tube formation by PD-L1. The current study provides new insight for the development of parthenolide as a novel anticancer drug targeting PD-L1.

Objective: Chronic obstructive pulmonary disease (COPD) is a risk factor for pneumonia following esophagectomy. This study aimed to investigate the efficacy of perioperative inhaled tiotropium in patients with COPD undergoing esophagectomy., Methods: This open-label, randomized controlled trial randomly assigned 32 patients with COPD undergoing esophagectomy to conventional management or addition of tiotropium inhalation. The intervention group received tiotropium from two weeks before esophagectomy until the final evaluation one month after esophagectomy. The primary outcome was the incidence of pneumonia within 30 postoperative days. We also assessed the changes and the percentages from baseline in pulmonary function and walking distance of the incremental shuttle walking test to just before esophagectomy and final evaluation., Results: Enrolled patients were randomly assigned to the control group (n = 18) and the intervention group (n = 14). Pneumonia was recorded in 4 (28.6%) and 5 (27.8%) patients in the intervention and control groups, respectively (risk difference: 0.8%, 95% confidence interval: - 30.6 to 32.2). The intervention group demonstrated a significant improvement in pulmonary function and walking distance preoperatively. Further, the pulmonary function test was significantly better preoperatively in the intervention group than in the control group. Postoperatively, pulmonary function deterioration was more significant in the control group than in the intervention group., Conclusions: Preoperative tiotropium inhalation significantly improved pulmonary function and exercise tolerance in patients with COPD undergoing esophagectomy. The exploration of noble metal-free nanoarrays as high-activity catalytic electrodes for glucose biosensing holds great significance. Herein, we propose a Ni nanoparticle-decorated TiO 2 nanoribbon array on a titanium plate (Ni@TiO 2 /TP) as an effective non-enzymatic glucose biosensing electrode. The as-prepared Ni@TiO 2 /TP electrode demonstrates rapid glucose response, a wide linear response range (1 μM to 1 mM), a low detection limit (0.08 μM, S/N = 3), and high sensitivity (10 060 and 3940 μA mM -1 cm -2 ), with good mechanical flexibility and stability. Moreover, it proves efficient in glucose biosensing in real human blood serum and cell culture fluid. Thus, it is highly promising for practical applications.

Systemic lupus erythematosus (SLE) is a multifaceted autoimmune disorder characterized by diverse clinical manifestations and organ damage. Despite its elusive etiology, dysregulated subsets and functions of B cells are pivotal in SLE pathogenesis. Peoniflorin-6'-O-benzene sulfonate (CP-25), an esterification modification of Paeoniflorin, exhibits potent anti-inflammatory and immunomodulatory properties in autoimmune diseases (AID). However, the involvement of CP-25 and its target, GRK2, in SLE development has not been explored. In this study, we demonstrate that both genetic deficiency and pharmacological inhibition of GRK2 attenuate autoantibodies production, reduce systemic inflammation, and mitigate histopathological alterations in the spleen and kidney in the pristane-induced mouse SLE model. Importantly, our findings highlight that both genetic deficiency and pharmacological inhibition of GRK2 suppress plasma cells generation and restore dysregulated B-cell subsets by modulating two crucial transcription factors, Blimp1 and IRF4. As a new micromanipulation tool with the advantages of small size, flexible movement and easy manipulation, light-driven microrobots have a wide range of prospects in biomedical fields such as drug targeting and cell manipulation. Recently, microrobots have been controlled in various ways, and light field has become a research hotspot by its advantages of noncontact manipulation, precise localization, fast response, and biocompatibility. It utilizes the force or deformation generated by the light field to precisely control the microrobot, and combines with the drug release technology to realize the targeted drug application. Therefore, this paper provides an overview of light-driven microrobots with drug targeting to provide new ideas for the manipulation of microrobots. Here, this paper briefly categorizes the driving mechanisms and materials of light-driven microrobots, which mainly include photothermal, photochemical, and biological. Then, typical designs of light-driven microrobots with different driving mechanisms and control strategies for multiple physical fields are summarized. Finally, the applications of microrobots in the fields of drug targeting and bioimaging are presented as well as the future prospects of light-driven microrobots in the biomedical field are demonstrated.

Purpose: The weight-adjusted waist index (WWI) is a recently developed index for measuring obesity. The aim of this study was to investigate the association between WWI levels and overactive bladder (OAB) in a nationally representative population., Methods: This cross-sectional study used data from the National Health and Nutrition Examination Survey (NHANES) database between 2007 and 2016. OAB was defined as the Overactive Bladder Syndrome Symptom Score (OABSS, score ≥ 3). The WWI index was calculated as the square root of waist circumference (WC, cm) divided by body weight (kg). We used weighted logistic regression models to assess the relationship between the WWI index and OAB in adult women. The reliability of the findings was assessed using restricted cubic spline, subgroup analysis., Results: A total of 10,563 individuals were included in the study, and the prevalence of OAB was 18.6%. Higher WWI was associated with an increased risk of overactive bladder syndrome. In model 1 with unadjusted variables (OR = 1.148; 95% CI = 1.148-1.149, p < 0.001), model 2 (OR = 1.253; 95% CI = 1.253-1.254, p < 0.001) and model 3 with fully adjusted variables (OR = 1.215; 95% CI = 1.214-1.215, p < 0.001) in which the association was significant. The results of the subgroup analyses showed that age stratification and stroke status could modify this association between WWI and OAB. Restricted cubic spline showed a nonlinear relationship between WWI and OAB (p for nonlinear < 0.05)., Conclusion: Weight-adjusted waist circumference index (WWI) values are positively associated with the risk of developing OAB in adult women in the United States, but further studies are needed to elucidate the causal relationship between WWI and OAB., (© 2024. Molybdenum disulfide (MoS 2 ) is heralded as an exemplary two-dimensional (2D) functional material, largely attributed to its distinctive layered structure. Upon forming heterojunctions with reducing species, MoS 2 displays remarkable photocatalytic properties. In this research, we fabricated a novel heterojunction photocatalyst, FeS/MoS 2 -0.05, through the integration of FeS with hollow MoS 2 . This composite aims at the efficient photocatalytic reduction of hexavalent chromium (Cr(VI)). A comprehensive array of characterization techniques unveiled that MoS 2 flakes, dispersed on FeS, provide numerous active sites for photocatalysis at the heterojunction interface. The inclusion of FeS seemingly promotes the formation of sulfur vacancies on MoS 2 . Consequently, this heterojunction catalyst exhibits photocatalytic activity surpassing pristine MoS 2 by a factor of 3.77. The augmented activity of the FeS/MoS 2 -0.05 catalyst is attributed chiefly to an internal electric field at the interface. This field enhances the facilitation of charge transfer and separation significantly. Density functional theory (DFT) calculations, coupled with experimental analyses, corroborate this observation. Additionally, DFT calculations indicate that sulfur vacancies act as pivotal sites for Cr(VI) adsorption. Significantly, the adsorption energy of Cr(VI) species shows enhanced favorability under acidic conditions. Background: Liver cirrhosis is a progressive hepatic disease whose immunological basis has attracted increasing attention. However, it remains unclear whether a concrete causal association exists between immunocyte phenotypes and liver cirrhosis., Aim: To explore the concrete causal relationships between immunocyte phenotypes and liver cirrhosis through a mendelian randomization (MR) study., Methods: Data on 731 immunocyte phenotypes were obtained from genome-wide association studies. Liver cirrhosis data were derived from the Finn Gen dataset, which included 214403 individuals of European ancestry. We used inverse variable weighting as the primary analysis method to assess the causal relationship. Sensitivity analyses were conducted to evaluate heterogeneity and horizontal pleiotropy., Results: The MR analysis demonstrated that 11 immune cell phenotypes have a positive association with liver cirrhosis [ P < 0.05, odds ratio (OR) > 1] and that 9 immunocyte phenotypes were negatively correlated with liver cirrhosis ( P < 0.05, OR < 1). Liver cirrhosis was positively linked to 9 immune cell phenotypes ( P < 0.05, OR > 1) and negatively linked to 10 immune cell phenotypes ( P < 0.05; OR < 1). None of these associations showed heterogeneity or horizontally pleiotropy ( P > 0.05)., Conclusion: This bidirectional two-sample MR study demonstrated a concrete causal association between immunocyte phenotypes and liver cirrhosis. Background: Insufficiently managed incisional (INC) pain severely affects patients' life quality and rehabilitation after a major operation. However, mechanisms underlying INC pain still remain poorly understood. Methods: A mouse model of INC pain was established by skin plus deep muscle incision. Biochemistry assay, in vivo reactive oxygen species (ROS) imaging, Ca 2+ imaging combined with retrograde labelling, neuron tracing and nocifensive behavior test, etc. were utilized for mechanism investigation. Results: We found pro-nociceptive cytokine interleukin -33 (IL-33) ranked among top up-regulated cytokines in incised tissues of INC pain model mice. IL-33 was predominantly expressed in keratinocytes around the incisional area. Neutralization of IL-33 or its receptor suppression of tumorigenicity 2 protein (ST2) or genetic deletion of St2 gene ( St2 -/- ) remarkably ameliorated mechanical allodynia and improved gait impairments of model mice. IL-33 contributes to INC pain by recruiting macrophages, which subsequently release ROS in incised tissues via ST2-dependent mechanism. Transfer of excessive macrophages enhanced oxidative injury and reproduced mechanical allodynia in St2 -/- mice upon tissue incision. Overproduced ROS subsequently activated functionally up-regulated transient receptor potential ankyrin subtype-1 (TRPA1) channel innervating the incisional site to produce mechanical allodynia. Neither deleting St2 nor attenuating ROS affected wound healing of model mice. Conclusions: Our work uncovered a previously unrecognized contribution of IL-33/ST2 signaling in mediating mechanical allodynia and gait impairment of a mouse model of INC pain. A comprehensive evaluation of the relationship between the densities of various cell types in the breast cancer tumor microenvironment and patient prognosis is currently lacking. Additionally, the absence of a large patch-level whole slide imaging (WSI) dataset of breast cancer with annotated cell types hinders the ability of artificial intelligence to evaluate cell density in breast cancer WSI. We first employed Lasso-Cox regression to build a breast cancer prognosis assessment model based on cell density in a population study. Pathology experts manually annotated a dataset containing over 70,000 patches and used transfer learning based on ResNet152 to develop an artificial intelligence model for identifying different cell types in these patches. The results showed that significant prognostic differences were observed among breast cancer patients stratified by cell density score (P = 0.0018), with the cell density score identified as an independent prognostic factor for breast cancer patients (P < 0.05). In the validation cohort, the predictive performance for overall survival (OS) was satisfactory, with area under the curve (AUC) values of 0.893 (OS) at 1-year, 0.823 (OS) at 3-year, and 0.861 (OS) at 5-year intervals. We trained a robust model based on ResNet152, achieving over 99% classification accuracy for different cell types in patches. These achievements offer new public resources and tools for personalized treatment and prognosis assessment.
