Individuals with major depressive disorder (MDD) exhibit cognitive impairment, while childhood trauma (CT) is associated with an elevated risk of both MDD and cognitive dysfunction. The effect of CT on cognitive function in MDD patients and healthy controls (HCs) is unclear. MDD patients and HCs were enrolled between December 2013 and December 2016. The Childhood Trauma Questionnaire (CTQ) was used to assess CT. Depressive symptoms and cognitive function were assessed at baseline and after 8-week acute-phase treatment with selective serotonin reuptake inhibitors (SSRIs) in MDD patients. A total of 909 people were included in the analysis. The interaction between MDD and CT had a main effect on Digit Symbol-Coding Test (DSCT), Stroop Color Test (SCT), and Stroop Color-Word Test (SCWT) scores. The effect of CT on cognitive function disappeared after adjusting for MDD diagnosis and years of education. Neglect could predict poor performance on SCT and SCWT in the HC group. After acute-phase treatment with SSRIs, CT did not significantly predict changes in cognitive function or depressive symptoms. The CTQ assessment might cause recall bias, and the cross-sectional design could not establish the causal link between CT and cognitive function. The effect of CT on cognitive function was modulated by MDD diagnosis and years of education. CT did not predict changes in depressive symptoms or cognitive function after acute-phase treatment with SSRIs. The direct influence of CT on cognitive function in MDD patients may be over-estimated. Background: To date, few data to transcranial Doppler sonography (TCD) are available in patients with mild vascular cognitive impairment (VCI) at risk for vascular or mixed dementia. In a previous study in patients with mild VCI and cerebral small vessels disease, a hemodynamic pattern of cerebral hypoperfusion and enhanced vascular resistance were observed; however, longitudinal data are currently lacking. Here, we perform a clinical, psychopathological, and neurosonological follow-up of patients with VCI in order to monitor any progression and to identify TCD measures to detect it. Methods: From the original cohort of 161 patients, 127 with VCI (mean age 73.6 ± 7.1; 67 males) were re-evaluated after 5.0 ± 1.8 years. Namely, the Montreal Cognitive Assessment (MoCA), the 17-items Hamilton Depression Rating Scale (HDRS), and the Stroop Color-Word Interference Test (StroopT) were administered to screen for global cognitive status, to quantify depressive symptoms, and to explore executive functions, respectively. Mean blood flow velocity (MBFV), peak systolic blood flow velocity (PSV), end-diastolic blood flow velocity (EDV), pulsatility index (PI), and resistivity index (RI) were recorded from the middle cerebral artery, bilaterally. Results: At follow up, patients exhibited a significant worsening of both MoCA (21.7 ± 2.1 vs. 20.7 ± 2.0) and StroopT scores (57.4 ± 19.4 vs. 59.7 ± 18.6), whereas HDRS showed an improvement, although the mean raw score remained above the cut-off value for depression (10.3 ± 6.6 vs. 9.8 ± 6.3). MBFV, PSV, and EDV showed a significant increase in PSV and PI and a reduction in EDV. When focused to younger patients (<65 years), we confirmed the significant worsening of both MoCA and StroopT but not HDRS, as well as the significant changes in PI and RI. Finally, considering the differences (D) between baseline and follow-up, the following significant correlations emerged, although with a small-to-medium effect size for all of them: positive correlation between MBFV-D and MoCA-D and between RI-D and STROOP-D, and a negative significant correlation between RI-D and MoCA-D. Conclusions: Notwithstanding some limitations, such as the lack of a control group and neuroimaging data at follow-up, TCD may contribute to the early detection, monitoring, and management of VCI patients at risk for dementia. Together with compatible clinical and cognitive features, the exploration of early TCD markers that possibly indicate a higher risk of progression might represent an intriguing research direction and a significant clinical perspective. Background: Klotho and neurotrophic factors, including brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and glial cell line-derived neurotrophic factor (GDNF), have been shown to play a role in cognitive functions. However, these molecules have not been investigated in bipolar disorder simultaneously to assess the interactions among them and their relationships with cognitive functions. This study investigated the relationships among cognitive function, klotho, and neurotrophic factors in patients with bipolar disorder in the remission period. Methods: Male, bipolar disorder (BD) patients (patient group, n = 48) in the remission period and healthy volunteers (control group, n = 48) were included in the study. The Stroop test and Wechsler Memory Scale–Visual Production Subtest were applied, and the serum levels of Klotho, BDNF, GDNF, and NGF were measured with an ELISA reader. Results: The klotho protein levels (0.12 ± 0.15 and 0.17 ± 0.16) and NGF levels (34.36 ± 41.99 and 48.54 ± 41.06) in the patient group were significantly lower than those in the control group (Z = -3.071, p = 0.002 and Z = -2.217, p = 0.027, respectively). In the patient group, there was a positive correlation between the klotho and NGF levels (p = 0.003, rs = 0.413), and a negative correlation was detected between the NGF and GDNF levels (p = 0.013, rs = -0.355). Klotho and NGF were predictors of Weshler 40-min test results (adjusted R2 = 0.467), and Klotho and BDNF were predictors of Stroop test colour word reading time (adjusted R2 = 0.391) with other variables. Conclusion: In BD patients, klotho, BDNF, GDNF and NGF are associated with cognitive functions and exhibit different characteristics from those of the control group. A recent meta-analysis has found that patients who have achieved remission of major depressive disorder (MDD) show cognitive dysfunction. Moreover, anticholinergic activity levels are associated with cognitive dysfunction, although the extent of these effects is unclear. Therefore, we measured serum anticholinergic activity (SAA) in blood samples of patients with remitted MDD and examined its relationship with cognitive function. We recruited 49 patients with remitted MDD following treatment and 165 healthy subjects. Subjects completed the Stroop test and the logical memory (LM) and visual reproduction (VR) subtests from the Wechsler Memory Scale-Revised. We compared cognitive function scores among those with SAA below the limit of quantification (SAA [−]), those with SAA above the limit of quantification (SAA [+]), and healthy controls. The SAA (+) group scored significantly lower (p < 0.001) than the healthy control group on all tests, and the VR score of the SAA (−) group was significantly lower than that of the healthy control group (p = 0.024). LM scores in the SAA (+) group were significantly lower than that of the SAA (−) group (p = 0.033). Multiple regression analysis revealed a significant effect of SAA on the LM score (p = 0.015). Our study was a cross-sectional analysis of a small number of patients. Our results support previous findings that the anticholinergic effect of antidepressants adversely affects cognitive function. Background: Tele-neuropsychology has already been employed in neurocognitive disorders, however, in Italy, the evidence of its psychometric quality and satisfaction is still limited. Objective: This study aimed to: (1) evaluate the reliability of a standardized battery of neuropsychological screening and domain-specific tests delivered at home via videoconference to a sample of Italian people with cognitive disorders, compared with traditional face-to-face administration; (2) assess the feasibility and satisfaction about remote administration. Methods: This crossover study enrolled patients with subjective cognitive disorder, mild neurocognitive disorder, or dementia. All participants performed a brief neuropsychological screening assessment in face-to-face or remote mode. People with milder cognitive disorders (Mini-Mental State Examination ≥ 20) also performed an extended neuropsychological battery. After 15 days, each participant repeated the same assessment in the opposite mode. Finally, participants completed a satisfaction questionnaire. Results: 114 participants were initially enrolled in the study and 100 individuals performed both face-to-face and remote assessments (14 dropouts). All neuropsychological tests showed agreement between the two modes of administration, except for the Digit Span Forward (p = 0.009) and oral-Symbol Digit Modalities Test (p < 0.001). According to Lin's concordance correlation coefficient (LCCCs), tests also showed good or excellent reliability (LCCCs between 0.609 and 0.964); only the Digit Span Backward and the Stroop test showed moderate reliability (LCCCs =0.514–0.441, respectively). Among 100 participants, 65 patients answered the satisfaction questionnaire, declaring high satisfaction for the remote evaluation. Conclusions: Tele-neuropsychology seems to work with in-home assessments among Italian patients with cognitive disorders and seems to be acceptable to them. Objective: The literature on clinical psychophysiology highlights the possibility of using Heart Rate Variability (HRV) as an index of psychophysical balance and resilience to stress. This study investigates the differences in stress reactivity and subsequent recovery between a group of migraineurs and healthy controls. Methods: Socio-demographic (i.e., sex, age, profession, marital status, and level of education) and psychophysiological (HR and HRV) measures of a group of thirty subjects with migraine (26 migraineurs without aura (86.7%), 2 migraineurs with aura (6.7%), and 2 migraineurs with and without aura (6.7%)) and from thirty healthy control subjects were collected. In particular, HRV was analyzed through frequency-domain parameters, including Low-Frequency (LF; 0.04–0.15 Hz) and High-Frequency (HF; 0.15–0.4 Hz) bands as well as LF/HF ratio during a Psychophysiological Stress Profile (PSP) structured in seven phases: (1) Baseline, (2) Objective stressor 1 (Stroop Test), (3) Rest 1, (4) Objective stressor 2 (Mental Arithmetic Task), (5) Rest 2, (6) Subjective stressor (recount a significant life event), and (7) Rest 3. The LF, HF, and LF/HF ratio values were transformed into a logarithmic scale (i.e., log-LF, log-HF, and log LF/HF ratio). Additionally, LF and HF were converted into normalized units (0-100) (i.e., LF% and HF%) which, in turn, were used to obtain reactivity and recovery to stress through delta values (Δ) calculation. Results: Subjects with migraine reported greater ΔLF% levels of reactivity and recovery to subjective stressor, demonstrating a prevalence of sympathetic activity while recounting a personal life event. At the same time, a lowering of the same values was found in the subjects of the group control. Discussion: Our results underline the importance of conducting a psychophysiological assessment in patients with headaches because reduced stress management skills could influence the clinical manifestations of the disease, considering stress as one of the most common triggers for migraine patients. Conceptual metaphors crucially shape how people understand and represent abstract concepts like love. Numerous Chinese linguistic metaphors associate love (beginning/continuing a romantic relationship) with red (e.g. 红豆相思) and lovelorn (breakup/end of a romantic relationship) with gray (e.g. 万念俱灰). However, empirical evidence supporting these color metaphors for love concepts is lacking. This study investigated the red-gray color metaphor for love concepts in Chinese. Experiment 1 used a Stroop test where participants categorized love/lovelorn words in red/gray fonts. Results showed significantly faster latencies for love words in red versus gray fonts, but no difference for lovelorn words. Experiment 2 used an implicit association test (IAT), where the compatible block paired red squares with love words and gray squares with lovelorn words, and vice versa in the incompatible block. Results showed that participants implicitly associated love words with red colors and lovelorn words with gray colors. Together, these experiments provide novel empirical evidence that native Chinese speakers activate metaphorical red-gray color mappings for love concepts, contributing insights into how abstract love concepts are grounded in embodied experiences. Although cognitive training has been proposed as a possible therapeutic modality for mild cognitive impairment (MCI), most serious games focus on specific tasks. This study aimed to investigate the feasibility and efficacy of narrative video game-based cognitive intervention for MCI. A four-week (± 1-week) mobile game intervention was given to 17 MCI participants (mean age (SD) = 72.8(4.75)). At baseline and post-intervention, the participants received neuropsychological tests and a depression scale. Frontal function was assessed using the Corsi block-tapping test, Color Word Stroop Test, Controlled Oral Word Association Test, Digit Symbol Coding, and Trail Making Test-Elderly's Version; depression was assessed using the Geriatric Depression Scale. User's compliance and gaming experience were also evaluated. MCI patients showed significant improvements in frontal function, particularly in Digit Symbol Coding (mean ± SD, 0.47 ± 0.49, p = 0.007) and phonemic fluency (mean ± SD, 0.39 ± 0.55, p = 0.024). Each frontal subtest's mean z-score was increased (mean ± SD, 0.44 ± 0.38, p = 0.008). Block span and depression scale remained unchanged. High adherence rates (122.35%) and favorable feedback on the gaming experience indicated that the game intervention's usability boosted patients' motivation and engagement. Background/Objectives: For the rapid, objective characterization of the physiological stress response, there is currently no generally recognized standard. The stress measurement methods used in practice (e.g., for psychological measures of stress) are often subjective, or in the case of biological markers (e.g., cortisol, amylase), they usually require a blood test. For this reason, the use of heart rate variability (HRV) to characterize stress has recently come to the fore. HRV is the variability in the length of heartbeat intervals, which indicates the ability of the heart to respond to various physiological and environmental stimuli. However, the conventional HRV metrics are not corrected for heart rate dependence; hence, they fail to fully account for the complex physiology of stress and relaxation. In order to remedy this problem, here we introduce a novel HRV parameter, the normalized variability derived from an RMSSD "Master Curve", and we compare it with the conventional metrics. Methods: In Study 1, the relaxation state was induced either by heart rate variability biofeedback training (N = 21) or by habitual relaxation (N = 21), while in Study 2 (N = 9), the Socially Evaluated Cold Pressor Test and the Socially Evaluated Stroop Test were used to induce stress in the subject. For a statistical evaluation of the data, the Kolmogorov–Smirnov test was used to compare the distributions of mean HR, log(RMSSD), log(SDNN), and normalized variability before, during, and after relaxation and stress. Results: The results of this study indicate that while log(RMSSD) and log(SDNN) did not change significantly, the normalized variability did undergo a significant change both in relaxation states and in stress states induced by the Socially Evaluated Cold Pressor Test. Conclusions: Overall, we suggest this novel type of normalized variability ought to be used as a sensitive stress indicator, and in general, for the characterization of the complex processes of the vegetative nervous system. Older adults were found to struggle with tasks that require cognitive control. One task that measures the ability to exert cognitive control is the color-word Stroop task. Almost all studies that tested cognitive control in older adults using the Stroop task have focused on one type of control – Information control. In the present work, we ask whether older adults also show a deficit in another type of cognitive control – Task control. To that end, we tested older and younger adults by isolating and measuring two types of conflict – information conflict and task conflict. Information conflict was measured by the difference between color identification of incongruent color words and color identification of neutral words, while task conflict was measured by the difference between color identification of neutral words and color identification of neutral symbols and by the reverse facilitation effect. We tested how the behavioral markers of these two types of conflicts are affected under low task control conditions, which is essential for measuring task conflict behaviorally. Older adults demonstrated a deficit in information control by showing a larger information conflict marker, but not in task control markers, as no differences in task conflict were found between younger and older adults. These findings supported previous studies that work against theories that link the larger Stroop interference in older adults to a generic slowdown or a generic inhibitory failure. Directional judgments of an arrow became slower when the direction and location were incongruent in a spatial Stroop task (i.e., a standard congruency effect). In contrast, gaze judgments were slower when they were congruent (i.e., a reversed congruency effect). This study examined the reaction time (RT) distribution of interference effects in a spatial Stroop task to clarify the temporal characteristics of the standard congruency effect, which is known to be reversed for social targets, such as gaze direction. Participants responded to laterally presented targets (i.e., arrows, gaze, fish-only, and fish with mosaic) while ignoring their location. The standard congruency effect of arrows decreased as the overall RT increased, reflecting the temporal decay of automatically activated task-irrelevant codes (i.e., location). Critically, the reversed congruency effect of gaze increased as the overall RT increased. This result supports the dual-stage hypothesis and reflects the late-arriving selective inhibition of task-irrelevant codes. Similar results were replicated in Experiment 2, in which we manipulated the complexity of the backgrounds of nonsocial targets and in the reanalysis of existing data. These findings imply that the interplay between task-irrelevant activation and subsequent inhibition is modulated by specific stimulus characteristics, influencing spatial response selection. One tool for promoting mental health is human stress detection through multitasks of electroencephalography (EEG) recordings. This study proposed a short-term stress detection approach using VGGish as a feature extraction and convolution neural network (CNN) as a classifier based on EEG signals from the SAM 40 dataset. This database was recently available and was collected from 40 patients using 32 channels to identify performance on four tasks including Stroop color-word test (SCWT), answering arithmetic problems, finding mirror-identical images, and relaxing. Each task took 25 s to complete and was then repeated three times to record three trials. This means that the total EEG data contain 480 signals for four tasks recorded using 120 trials per task. The primary objective of this research was to track the amount of short-term stress that patients experienced while they engaged in the four mental tasks. Moreover, the VGGish-CNN model is applied to the SAM 40 dataset using five stages including signal preprocessing, segmentation, filtration, spectrogram, and classification process. We compared the VGGish-CNN model and the VGGish model for stress-based EEG classification to determine the best classification accuracy. The proposed approach for stress detection is the preliminary study that achieved an accuracy of 99.25% using the VGGish-CNN model on the SAM 40 dataset. Next, k-fold cross validation is performed to verify the efficiency of the VGGish-CNN model. This study can advance the application of brain–computer interface (BCI) and its use to identify patterns in EEG data that invoke stress-related inferences to aid in the diagnosis of mental disorders.
