Economic impact assessments were conducted to estimate the potential cost savings associated with implementing a peer-support program.Results: The economic analysis reveals significant annual costs associated with SVP-induced absenteeism: approximately 1.56 billion euros for physicians and 1.87 billion euros for nurses. Implementing comprehensive peer-support programs could reduce these costs to approximately 0.85 billion (physicians) and 1.02 billion euros (nurses), respectively, demonstrating substantial potential economic benefits.Conclusion: Investing in a structured peer-support program could yield annual savings exceeding 1.55 billion euros while enhancing workforce resilience and improving patient care. Abstract This investigation aimed to develop a radiographic 3D cephalometric index to grade severity of dentofacial deformity in patients with juvenile idiopathic arthritis (JIA), and to perform a validation against expert evaluations. Data were collected from a population-based Nordic JIA cohort of 240 patients that received a cone-beam computed tomography (CBCT) scan approximately 17 years after onset of JIA. The cohort was randomized into two groups: A baseline group for establishing the index (n = 210) and a test group (n = 30). The standardized index generation process led to the inclusion of six radiographic outcome measures, each representing different facets of dentofacial deformities. Specific severity scores were established for three domains; dentofacial asymmetry (0–8), vertical deviation (0–8), and sagittal deviation (0–8). An overall dentofacial deformity index score was computed by aggregating the scores from the six variables, with the total score ranging from 0 to 24. The validity of the index was assessed by comparing the index score with expert evaluations. Based on the generated index, 10 patients exhibited severe deformities. Reliable agreement between data-generated index scores and expert assessments was found. A 3D radiographic dentofacial deformity index was generated and tested. This index can support an objective assessment of the severity of dentofacial deformities related to JIA in a clinical setting and future research.

Abstract The visceral organ-brain axis, mediated by vagal sensory neurons, is essential for maintaining various physiological functions. Here, we investigate the impact of liver-projecting vagal sensory neurons on energy balance, hepatic steatosis, and anxiety-like behavior in mice under obesogenic conditions. A small subset of vagal sensory neurons innervate the liver and project centrally to the nucleus of the tractus solitarius, area postrema, and dorsal motor nucleus of the vagus, and peripherally to the periportal areas in the liver. The loss of these neurons prevents diet-induced obesity, and these outcomes are associated with increased energy expenditure. Although males and females exhibit improved glucose homeostasis following disruption of liver-projecting vagal sensory neurons, only male mice display increased insulin sensitivity. Furthermore, the loss of liver-projecting vagal sensory neurons limits the progression of hepatic steatosis. Intriguingly, mice lacking liver-innervating vagal sensory neurons also exhibit less anxiety-like behavior compared to control mice. Modulation of the liver-brain axis may aid in designing effective treatments for both psychiatric and metabolic disorders associated with obesity and MAFLD.

Dinutuximab promotes complement-mediated reaction against disialoganglioside GD2, which is expressed in peripheral nerves and over-expressed in neuroblastoma. Dinutuximab is associated with ≥grade 3 neuropathic pain. Targeting GD2 stimulates the NMDA receptor, which makes ketamine useful in treatment of associated pain. The objective of this retrospective study is to describe the use of ketamine for pain uncontrolled by opioids, and ketamine’s impact on total opioid usage for patients receiving dinutuximab. In addition, the secondary objective is to describe the toxicities of pain management with opioids versus opioid plus ketamine.Methods: A retrospective chart review of 40 hRNBL patients receiving dinutuximab at Nationwide Children’s Hospital, from 2010 to 2022, was conducted. Demographics, pain scores, medication records, and total daily IV morphine milligram equivalents (IVMME) with and without a ketamine adjunct were collected. Linear mixed effect regression was used to model IVMME use for pain management across dinutuximab cycles and explore the effect of ketamine.Results: The study cohort included 187 dinutuximab hospitalizations from 40 patients. Age at diagnosis ranged from 1.2 to 11.4 years. 66/187 hospitalizations included ketamine. The average daily IVMME during post-consolidation dinutuximab infusions was greater in admissions with ketamine (median 11.67 mg/day vs 6.09 mg/day; p = 0.0005). Ketamine was not significantly associated (p = 0.77) with daily IVMME when examining opioid use longitudinally over dinutuximab cycles and controlling for patient age. Fever/chills was more frequent in admissions that utilized ketamine (79% vs 63%; p = 0.0297). No other significant statistical differences in adverse effects were observed in patients’ receiving opioids versus opioids plus ketamine.Conclusion: Findings suggest ketamine is safe in a non-ICU setting for treatment of complex pain during anti-GD2 immunotherapy. This paper presents a case study of the impact of continuous deformations and rock mass tremors on a single-family building, typical of Upper Silesia. In the areas subjected to the influence of ground deformation, a point method for assessing the resistance of buildings to these influences has been developed by the Central Mining Institute. As part of this method, the geometric parameters, structure, materials and technical condition of the facility, as well as the ground on which the building is located, are taken into account. A case study considers a building being simultaneously subjected to continuous deformations and mining tremors. The object’s resistance is assessed according to the point method and its technical condition is assessed after revealing the influence of ground deformation. This is used to assess the validity of the point method for the building. On the basis of the analyses of the state of deformation and the impact of tremors, conclusions are drawn regarding the need to make changes to the scale used to assess the category of resistance of buildings to the effects of continuous deformation.

As a relatively young field of medicine, oncology is an important element in society, as cancer is one of the major diseases of civilisation in recent decades. As recently as the 20th century, the types of oncology centres were mainly limited to university clinics and bed wards, and the most common method of cancer treatment was invasive surgery. The development of cancer treatment methods that occurred at the turn of the 20th century influenced the architectural and functional design of cancer centres. Innovative treatment methods required new spaces, and thus cancer centres evolved into new types. An analysis was carried out in terms of what types cancer centres in Europe currently take. For further research, 12 facilities considered as reference were selected to represent different types of cancer facilities established in the 21sh century. The aim of the research was to classify contemporary types of cancer centres in the context of the development of treatment methods and to characterise them generally in an architectural context. As a result of the research, five main forms in which cancer centres occur were identified. The general characteristics of these facilities and the differences between them depending on the profile of services provided were presented. The zones present in the centres in question that define their functional layout and shape were also specified.

ABSTRACT: Substantial advances in livestock traits have been achieved primarily through selection strategies targeting variation in the nuclear genome, with little attention given to mitogenome variation. We analyzed the influence of the mitogenome on milk production traits of Holstein cattle in Croatia based on strategically generated next-generation sequencing data for 109 cows pedigree-linked to 7,115 milk production records (milk, fat, and protein yield) from 3,006 cows (first 5 lactations). Because little is known about the biology of the relationship between mitogenome variation and production traits, our quantitative genetic modeling was complex. Thus, the proportion of total variance explained by mitogenome inheritance was estimated using 5 different models: (1) a cytoplasmic model with maternal lineages (CYTO), (2) a haplotypic model with mitogenome sequences (HAPLO), (3) an amino acid model with unique amino acid sequences (AMINO), (4) an evolutionary model based on a phylogenetic analysis using Bayesian Evolutionary Analysis Sampling Trees phylogenetic analysis (EVOL), and (5) a mitogenome SNP model (SNPmt). The polygenic autosomal and X chromosome additive genetic effects based on pedigree were modeled, together with the effects of herd-year-season interaction, permanent environment, location, and age at first calving. The estimated proportions of phenotypic variance explained by mitogenome in 4 different models (CYTO, HAPLO, AMINO, and SNPmt) were found to be substantial given the size of mitogenome, ranging from 5% to 7% for all 3 milk traits. At the same time, a negligible proportion of the phenotypic variance was explained by mitogenome with the EVOL model. Similarly, in all models, no proportion of phenotypic variance was explained by the X chromosome. Although our results should be confirmed in other dairy cattle populations, including a large number of sequenced mitogenomes and nuclear genomes, the potential of utilizing mitogenome information in animal breeding is promising, especially as the acquisition of complete genome sequences becomes cost-effective.

In this study, we compared ABO blood groups with five major groups of mental disorders to attain a higher level of specificity. The analyses were conducted using data from the CoLaus|PsyCoLaus study (N=5111). They revealed that the AB blood group exhibited a positive association with both neurodevelopmental disorders (RR 2.29, CI 1.38– 3.82) and substance use disorders (RR 2.25, CI 1.38– 3.65) after adjusting for sex and childhood adversities. These associations could be replicated with respect to the familial aggregation of neurodevelopmental and substance use disorders. Large databases are needed to achieve more detailed results related to specific disorders.Plain Language Summary: Prior research has shown an interesting link between the AB blood group and mental disorders. In our study, we used data from a large Swiss population-based epidemiological cohort study to learn more about this connection. We examined the relationship between ABO blood groups and five major groups of mental disorders: neurodevelopmental, early onset anxiety, later onset anxiety, mood, and substance use disorders. Our analyses revealed associations between the AB blood group and neurodevelopmental and substance use disorders. The prevalence of childhood obesity is increasing worldwide, along with the associated common comorbidities of type 2 diabetes and cardiovascular disease in later life. Motivated by evidence for a strong genetic component, our prior genome-wide association study (GWAS) efforts for childhood obesity revealed 19 independent signals for the trait; however, the mechanism of action of these loci remains to be elucidated. To molecularly characterize these childhood obesity loci, we sought to determine the underlying causal variants and the corresponding effector genes within diverse cellular contexts. Integrating childhood obesity GWAS summary statistics with our existing 3D genomic datasets for 57 human cell types, consisting of high-resolution promoter-focused Capture-C/Hi-C, ATAC-seq, and RNA-seq, we applied stratified LD score regression and calculated the proportion of genome-wide SNP heritability attributable to cell type-specific features, revealing pancreatic alpha cell enrichment as the most statistically significant. Subsequent chromatin contact-based fine-mapping was carried out for genome-wide significant childhood obesity loci and their linkage disequilibrium proxies to implicate effector genes, yielded the most abundant number of candidate variants and target genes at the BDNF, ADCY3, TMEM18, and FTO loci in skeletal muscle myotubes and the pancreatic beta-cell line, EndoC-BH1. One novel implicated effector gene, ALKAL2 – an inflammation-responsive gene in nerve nociceptors – was observed at the key TMEM18 locus across multiple immune cell types. Interestingly, this observation was also supported through colocalization analysis using expression quantitative trait loci (eQTL) derived from the Genotype-Tissue Expression (GTEx) dataset, supporting an inflammatory and neurologic component to the pathogenesis of childhood obesity. Our comprehensive appraisal of 3D genomic datasets generated in a myriad of different cell types provides genomic insights into pediatric obesity pathogenesis.

Esophageal adenocarcinoma (EAC) is an aggressive gastrointestinal (GI) malignancy with increasing incidence. Despite the recent progress in targeted therapies and surgical approaches, the survival rates of esophageal adenocarcinoma patients remain poor. The Dickkopf (DKK) proteins are secretory proteins known mainly as antagonists of the Wnt/β-catenin signaling pathway, which is considered an oncogene. However, it has been shown that in several GI cancers, including esophageal cancer, DKK1 may act as an oncogene itself through Wnt-independent signaling pathways. LRP5\6 and Kremen1/2 (Krm1/2) are transmembrane receptors to which the DKK proteins are mainly known to bind. CKAP4 (cytoskeleton-associated protein 4) is a novel receptor of DKK1, and the DKK1-CKAP4 pathway seems to be crucial in the role of DKK1 as an oncogene. The aim of this review is to feature the essential role of DKK1 and its receptors in carcinogenesis with a focus on EAC in an era of urgent need for specific biomarkers along with improved targeted therapies.

Mushrooms are a raw material rich in many nutritional compounds, and that is why a number of them are widely known as functional food. They contain fatty acids, carbohydrates, lycopene, sterols, lovastatin, trace elements, and other valuable compounds that show a wide range of properties, such as hepatoprotective, anticancer, antiviral, etc. For more efficient utilisation of mushrooms’ biologically active substances, widespread supercritical carbon dioxide extraction (Sc-CO2) was used as an efficient way to isolate the high-value phytoconstituents from this type of raw material. Using Sc-CO2, the extracts of five types of edible mushrooms—Lycoperdon saccatum, Pleurotus ostreatus, Craterellus cornucopioides, Russula Cyanoxantha and Cantharellus cibarius—were obtained. During the Sc-CO2 process, the extraction time was reduced to 4 h compared to the prolonged process time applied in the typical traditional techniques (6–24 h). The extraction pressure (30 MPa) and temperature (40 °C) were constant. Fatty acids and the compounds of steroid structures were determined in the obtained extracts using GC–MS and GC–FID methods of analysis. The dominant compounds identified in the lipid extracts were fatty acids (linoleic, oleic, palmitic and stearic) and sterols (ergosterol, 7,22-ergostadienone and 7,22-ergostadienol). For complete insight into the process and to obtain the value of the extracts, chemometric analysis is provided. Principal component analysis (PCA) and hierarchical cluster analysis (HCA), as well as k-means clustering, showed that Craterellus cornucopioides was distinguished based on the extraction yield results.

Phosphonic acid cation exchange materials (PCX) are synthesized by atom transfer radical polymerization (ATRP) followed by alkyne-azide click-chemistry. ATRP is used to synthesize polymeric chains of diethyl 4-vinylbenzylphosphonate with different chain lengths, which are covalently bonded to the surface of monodisperse polystyrene-divinylbenzene (PS/DVB) particles by click-chemistry. The functionalized particles are characterized by FIB-SEM, IR and Schoeniger combustion followed by chromatographic experiments. The effect of chain length on chromatographic behavior is investigated by comparing four PCX columns with different chain lengths. Polar ionic pesticides and non-polar triazines are used as model analytes for chromatographic characterization. The chain length showed an influence on the retention of doubly charged analytes, which can be explained by the increased local charge density with longer chains. The resolution of the constitutional isomers prometryn and terbutryn increased with the chain length, leading to an isocratic separation of seven triazines in 35 min. In this study, a graphene oxide combined with poly(2-diethylaminoethyl methacrylate) (GO@PDEAEMA) nanocomposite was synthesized for the separation and enrichment of Cd and Pb from food and water samples using the dispersive micro-solid phase extraction (d-μ-SPE) technique. The GO@PDEAEMA nanocomposite was synthesized using surface-initiated atom transfer radical polymerization (SI-ATRP) and characterized using various analytical techniques, such as FTIR, FE-SEM, TGA, BET, and XRD. The optimal experimental conditions were pH 8, 0.5 M HNO₃ as eluent, 5 mg of sorbent, and adsorption/desorption times of 0.5 and 1 min, respectively, with a recovery range of 89-101 %. The suggested method showed low limits of detection (LOD) and quantification (LOQ) of 0.11 μg L -1 and 0.37 μg L -1 for Cd and 0.28 μg L -1 and 0.93 μg L -1 for Pb, respectively. The optimal procedure was successfully applied to real water and food samples. This study demonstrates the development and application of an ultra-high-performance liquid chromatography coupled with high-resolution mass spectrometry (UHPLC-HRMS) method for the rapid and sensitive identification of intact bovine milk proteins following simulated gastrointestinal infant digestion. The new method enables the differentiation between partially hydrolysed/modified and fully intact proteins. In the raw milk, intact α-lactalbumin was visible on SDS - PAGE until the end of the gastrointestinal digestion, while it was not detected with UHPLC-HRMS. Analysis of both raw and heated milk samples revealed that the method is applicable to various milk types. Interestingly, heated milk showed additional signals in the mass spectrum, indicating non-enzymatic post-translational modifications. The relative abundance of these proteoforms could be followed along digestion. The construction of ultrathin membranes from linearly aligned π-electron systems is advantageous for targeted energy, charge, or mass transfer. The Langmuir-Blodgett (LB) technique enables the creation of such membranes, especially with amphiphilic π-electron systems. However, these systems often aggregate, forming rigid Langmuir monolayers with defects or holes. In this study we introduce plasticizers to effectively address this issue. To create anisotropic membranes, we used an oligo(phenylene ethynylene) derivative (OPE-NH 2 ) as an linear amphiphile and bisphenol A di-tert-butyl ester (BPAE) as a plasticizer. We analyzed surface pressure (mean molecular area) (Π(mma)) isotherms and characterized Langmuir monolayers with Brewster Angle Microscopy (BAM), to determine the optimal miscibility of OPE-NH 2 with BPAE. Detailed analysis of hole areas filled was performed through image binarization. We identified an optimal BPAE concentration of 4 mol-% in the OPE-NH 2 Langmuir monolayer. Our BAM image evaluation via binarization determined the difference between the mean molecular areas of close-packed Langmuir domains and those quantified via the Π(mma) isotherm. Thevinols and their 3-O-demethylated relatives, orvinols, are derivatives of the Diels-Alder adduct of natural alkaloid thebaine with methyl vinyl ketone. Taken together, thevinols and orvinols constitute an important family of opioid receptor (OR) ligands playing an important role in both the OR mediated antinociception and OR antagonism. Herein, we disclose for the first time the antagonist activity of the N-allyl substituted orvinol derivative fluorinated within the pharmacophore associated with C(20) and its surrounding. This compound was prepared via a novel synthetic sequence from 18,19-dihydrothevinone bearing an allyl substituent at N(17) and three fluorine atoms at C(21). Preliminary trials reported earlier demonstrated that the compound exhibited no analgesic activity. However, in vivo experiments conducted in an acute pain model (tail-flick test in mice) demonstrated that this fluorinated compound, when administered at doses of 5-10 mg/kg (sc) 30 min before morphine, exhibited antagonistic activity at the level of naloxone (1 mg/kg, sc) for a longer duration (at least 120 min) compared to naloxone (60 min). Together with the analgesic activity that has been reported for the C(21)-trifluorinated relatives bearing methyl or cyclopropylmethyl substituent at N(17), this result highlights C(21)-fluorinated thevinols and orvinols as the family of opioid receptor ligands (structurally related to buprenorphine, diprenorphine, etc.) covering the full range of activity profiles from agonists to antagonists, which is promising for tuning of their pharmacological properties via a substitution of hydrogen atoms within the pharmacophore associated with C(20) and its surrounding for fluorine., Competing Interests: Declaration of competing interest The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (Copyright © 2024 Elsevier Masson SAS. The development of a new Neodymium/Dysprosium metal-organic framework, referred to as Nd/Dy-BTC MOF, based on benzene-1,2,4-tricarboxylic acid, has been achieved through an in situ growth process on 2D transition metal carbides (MXene) surfaces. This synthesis was conducted via a solvothermal method utilizing a solvent mixture of water, ethanol, and dimethylformamide. The primary objective of this research is to investigate the framework's efficacy as a photocatalyst for the degradation of anionic dye, as well as its potential for sensing certain explosives. The Nd/Dy-MOF@MXene has been characterized by various analysis techniques. The prepared Nd/Dy-MOF@MXene was utilized as catalyst in selective degradation of methyl orange dye. The study examined the influence of pH and catalyst concentration, revealing that the catalyst achieves peak efficiency of 93.55% when exposed to sunlight in an acidic medium. The reusability of the catalyst shows the highest efficient photocatalyst after four cycles of reusability. The detection of explosives, specifically 2,4,6-trinitrophenol, through photoluminescence techniques has been conducted, utilizing the Stern-Volmer equation to evaluate the quenching efficiency. The findings indicated remarkable efficiency and selectivity of Nd/Dy-MOF@MXene for 2,4,6-trinitrophenol, achieving a quenching efficiency of 91%. Furthermore, the reusability tests demonstrated that Nd/Dy-MOF@MXene exhibits outstanding recyclability, maintaining performance over five cycles. The antibacterial activity of Nd/Dy-MOF@MXene was investigated against Gram-positive and Gram-negative strains due to indication of medicine properties of Nd/Dy-MOF@MXene. Exploring high-efficiency photocatalysts for solar hydrogen (H 2 ) generation through water splitting is of great significance for addressing both energy shortage and environmental contamination. In this work, a facile self-assembly strategy was developed to couple NiPS 3 nanosheets (NPS NSs) with ZnIn 2 S 4 (ZIS) microspheres to synthesize NPS NSs/ZIS (NPS/ZIS) composites, featuring a characteristic of S-scheme charge transfer mechanism. The NPS/ZIS composites possess broad-spectrum light absorption property, improved photothermal effect and efficient charge transfer, showcasing exceptional solar-to-chemical energy conversion capability for visible-light-driven photocatalytic hydrogen evolution (PHE). The photothermal effect derived from NPS NSs loading can facilitate charge carrier transfer across the interfaces and surface reaction kinetics. By carefully adjusting the mass ratio of NPS NSs, the optimized 4-NPS/ZIS exhibits excellent stability and significantly improved PHE activity (1827.6 μmol⋅g -1 ⋅h -1 ) in water, which is 18.4 times higher than that of bare ZIS (99.4 μmol⋅g -1 ⋅h -1 ). Furthermore, the 4-NPS/ZIS also shows the high PHE efficiency of 312.2 μmol⋅g -1 ⋅h -1 in seawater. Diverse characterization results reveal that the remarkably enhanced PHE performance primarily arises from the synergistic effect of S-scheme heterostructure, heightened light harvesting capacity, and enhanced photothermal effect. On the basis of density functional theory (DFT) simulations and experimental verifications, a possible PHE mechanism via the S-scheme heterojunction with photothermal assistance in NPS/ZIS is proposed. The use of nanoparticulate systems for the transport of active ingredients into hair follicles has been researched for almost two decades, resulting in countless publications with a wide variety of particle types, release mechanisms and active ingredients. The production of a stable dispersion is often time-consuming and costly. In this publication, we demonstrate for the first time that simply adding diverse submicron particles to a drug solution significantly increases follicular penetration depth by over 160% to 190%, allowing the targeting of subinfundibular structures. Our results indicate that the increase in follicular penetration is independent of the type or sphericity of the particles (nanocrystals (NC) or lipid submicron particles (LN)). Furthermore, this principle can be used with both small molecules and large molecule therapeutics, as demonstrated with the model drugs fluorescein sodium, 6-carboxyfluorescein, green fluorescent protein and FITC-BSA. This highlights the high versatility of this new formulation principle. The presence of multiple hydroxyl groups at positions C2, C3 and C6 on the cyclodextrin (CD) ring structure allows for extensive functionalisation, enabling the development of biomaterials with significant potential for therapeutic siRNA delivery. To identify structural modifications that enhance activity, a range of cationic amphiphilic CDs, including both β- and γ-CDs, were synthesised, compared and evaluated. Each CDs incorporated a C 12 lipid chain on the primary face of the CD. On the secondary rim, at positions C2 and C3, either primary or tertiary amine groups with different pKa values were introduced via either a triazole or a thiopropyl linker. Nanoparticles (NPs) encapsulating small interfering RNA (siRNA) were formulated at mass ratios 7.5:1 and 10:1 (CD:siRNA) and their physicochemical properties evaluated. A comparative in vitro study was conducted to assess the gene silencing efficacy of these NPs using the luciferase reporter gene in A549-luc cells. Gene silencing levels for both β- and γ-CDs increased when modified with a primary amine compared to a tertiary amine group at position C2. Gene expression inhibition was further improved when the CDs were functionalized with amine functionalities at positions C2 and C3. Modification of the secondary side of γ-CDs with two sets of primary amine functionalities via a thiopropyl linker, as compared to a triazole linker, achieved up to 80% gene knockdown, regardless of dose. In this contribution, we compute the 1H nuclear magnetic resonance (NMR) relaxation rate of liquid water at ambient conditions. We are using structural and dynamical information from Coupled Cluster Molecular Dynamics (CCMD) trajectories generated at CCSD(T) electronic structure accuracy while also considering nuclear quantum effects in addition to consulting information from x-ray and neutron scattering experiments. Our analysis is based on a recently presented computational framework for determining the frequency-dependent NMR dipole-dipole relaxation rate of spin 1/2 nuclei from Molecular Dynamics (MD) simulations, which allows for an effective disentanglement of its structural and dynamical contributions and includes a correction for finite-size effects inherent to MD simulations with periodic boundary conditions. A close to perfect agreement with experimental relaxation data is achieved if structural and dynamical information from CCMD trajectories is considered, leading to a re-balancing of the rotational and translational dynamics, which can also be expressed by the product of the self-diffusion coefficient and the reorientational correlation time of the H-H vector D0 × τHH. The simulations show that this balance is significantly altered when nuclear quantum effects are taken into account. Our analysis suggests that the intermolecular and intramolecular contributions to the 1H NMR relaxation rate of liquid water are almost similar in magnitude, unlike what was predicted earlier from fully classical MD simulations., (© 2025 Author(s). In this present work, we describe the syntheses of a new series of 32 1H-indole-based-meldrum linked 1H-1,2,3-triazole derivatives (2-13, 15a-15f, 16a-16f, 17a-17f and 19a, 19b, 20a), which constitute a new class of 1H-1,2,3-triazoles. Compounds 15a-15f, 16a-16f, 17a-17f have been prepared by employing "click" reactions between substituted 1H-indole-based meldrum alkynes (11, 12 and 13) and substituted aromatic azides (14a-14f) in the presence of copper iodide (CuI) and Hünig's base. Then, the synthesis of compounds 19, 20 through decomposition of meldrum moiety. The resulting compounds have been screened for their dihydrofolate reductase (DHFR) inhibition activity. All the newly synthesized compounds were characterized by 1 H NMR, 13 C NMR, 19 F NMR (spectroscopy when applicable), and HR-ESI-MS spectroscopy techniques. The X-ray crystallography studies have unambiguously confirmed the structure of compounds 6, 11 and 13. Furthermore, their DHFR-inhibitory activity was evaluated in-vitro. The results obtained from the DHFR-inhibitory assay revealed that all the synthesized 1H-indole-based-meldrum linked 1H-1,2,3-triazole derivatives were highly potent inhibitors, with IC 50 values in the range 3.48 ± 0.16-30.37 ± 1.20 μM. Ten compounds (15c-15f, 16c-16f, 17e and 17f) among the 32 synthesized 1H-indole-based-meldrum linked 1H-1,2,3-triazole compounds were found to exhibit exceptional inhibitory while the rest of the derivatives showed moderate activities. Additionally, molecular docking analysis of the most active (16f), moderate (15c) and least active (16a) inhibitors reflect excellent binding of 16f with the binding residues of DHFR with higher docking score (-9.13 kcal/mol) than that of 15c and 16a. The docking analysis correlates well with the inhibitory potential of these synthesized molecules. Aqueous solutions of glycols, on the one hand, are widely used in many applications; on the other hand, they can serve as simple and representative models for studying intra- and intermolecular hydrogen bonds. In this work, we analyze the possibilities and limitations of Raman spectroscopy for fundamental and applied researches of such solutions on the examples of ethylene glycol (EG) and 1,3-propylene glycol (1,3-PG). It is shown that Raman spectroscopy is an effective tool for monitoring temporal changes in the structure of glycol solutions deposited on substrates. This study demonstrates that the water content in the solutions on the substrates decreases rapidly with time, and the rate of this decrease depends on the chemical structure of both glycol and substrate. It was found that the reduction in the water content leads to slight decrease in the contents of gauche-conformers in the backbones of EG and 1,3-PG molecules. We report on the formation of the Cr 2 C compound using chemical etching-free methodology to extract Al from a Cr 2 AlC MAX phase thin film. Cr 2 AlC/Cu assemblies were deposited on sapphire substrates, using magnetron sputtering, and were subsequently annealed in vacuum. The Al from the MAX phase was shown to diffuse into Cu resulting in the formation of Al 4 Cu 9 and causing the MAX phase to collapse into Cr 2 C grains. These carbide grains were characterized by transmission electron microscopy and the interatomic distances extracted were in good agreement with ab initio calculations predicting the equilibrium volume of the Cr 2 C phase.

The excessive and uncontrolled release of neutrophil extracellular traps (NETs) is increasingly linked to the pathogenesis of various inflammatory diseases, cardiovascular disorders, and cancers. Real-time, non-invasive detection of NETs is crucial for understanding their role in disease progression and developing targeted therapies. Current NETs detection methods often lack the necessary specificity and resolution, particularly in vivo and ex vivo settings. To address this, we have developed novel near-infrared squaraine-peptide conjugates by rational molecular design as reporters of NETosis by targeting the protease activity of neutrophil elastase (NE). These self-quenching, cell-impermeable probes enable the precise real-time detection and imaging of NETs. The Förster resonance energy transfer (FRET)-based probe, Hetero-APA , demonstrated high specificity in detecting NETs in vitro and in vivo, generating strong fluorescence in NETs-rich environments. To overcome the limitations of FRET-based probes for ex vivo imaging, we designed SQ-215-NETP , a non-FRET-based probe that covalently binds to the NE. SQ-215-NETP achieved an unprecedented imaging resolution of 90 nm/pixel in human coronary thrombi, marking the first report of such high resolution with a low molecular weight probe. Additionally, SQ-215-NETP effectively detected NETs by flow cytometry. These results highlight the potential of these probes in NETosis detection, offering promising tools for enhanced diagnostics and therapeutic strategies in managing NET-mediated inflammatory diseases and cancers.

Mutations in the human FLNC gene encoding filamin C (FLNc) cause a broad spectrum of sporadic and familial cardiomyopathies and myopathies. We report on the genetic, clinical, morphological and biochemical findings in a German family harboring an FLNC variant that leads to severe cardiac disease comprising sudden cardiac death and arrhythmogenic cardiomyopathy. Genetic analysis identified a novel heterozygous FLNC variant in exon 16 (NM_001458.4:c.2495_2498delAGTA, het; p.K832TfsX45) in i) the index patient suffering from dilated cardiomyopathy necessitating heart transplantation, ii) a son, who died from sudden cardiac death, iii) a second son, who survived an episode of sudden cardiac arrest and iv) a third son affected by isolated skeletal muscle myopathy. FLNc protein levels were markedly reduced in cardiac tissue obtained from the index patient, implying that the p.K832TfsX45 FLNc variant most probably caused nonsense-mediated decay of the corresponding mRNA. Morphological analysis of the diseased cardiac tissue revealed extensive fibrotic remodeling, and marked degenerative changes of the contractile apparatus of cardiomyocytes and severe structural alterations of intercalated discs. Connexin-43 signal intensity at intercalated discs was diminished and FLNc labelling of myofibrils was attenuated or even absent. Proteome analyses demonstrated complex alterations of extracellular matrix and intercalated disc proteins. Our findings demonstrate that this novel, truncating FLNC mutation likely leads to haploinsufficiency, thereby causing a deleterious sequence of degenerative changes of cardiac tissue with extensive fibrotic remodeling and intercalated disc pathology as the structural basis for FLNC-related cardiomyopathy with life-threatening cardiac arrhythmias., (© The Author(s) 2025. Background: Ensuring food safety is a fundamental priority for public health. The catering sector has become prominent as a convenient and cost-effective method of food supply worldwide. Adherence to proper food hygiene practices is crucial for preventing foodborne diseases. The inspection of food items is a key component of internal controls that enables the identification of non-compliance with food hygiene standards. The present study aims to assess Attica restaurant businesses' compliance with international food hygiene standards (FAO/WHO Codex Alimentarius) and European legislation on unsafe food., Methods: From January to July 2023, 74 randomly selected restaurants of small, medium, and large capacity in Attica were examined through inspections in terms of compliance for food hygiene standards. The inspections were based on (a) the completion of forms and (b) the collection of water and food samples for laboratory microbiological analysis. Data were collected using a predefined form, referencing the manual of Codex Alimentarius regarding food hygiene standards (CAC/RCP 1-1969/CAC/RCP 39-1993). Additionally, the assessment included the identification of unsafe food placement in accordance with Regulation EC/178/2002 on food safety, as well as Greek national legislation (European Commission, 2002; Hellenic Republic, 2006, 2014)., Results: The highest non-compliance rates are in "adequacy of facilities" (14.12%), "equipment maintenance and sanitization" (12.30%), "pest control" (12.45%), "personal hygiene" (7.58%), and "efficient separation of raw materials" (9.76%). Non-compliance rates for other food hygiene parameters (cooking practices, meal apportionment, storage, transport, reheating, etc.) were considerably lower. The inspection results showed that medium-sized restaurant businesses present the highest rate (56.41%) of total non-compliance compared to large-sized businesses (29.68%) and small-sized businesses (13.91%)., Conclusions: This study demonstrates that restaurant businesses generally adhere to food hygiene and safety standards at a satisfactory level. Chronic back pain (CBP) is a disabling condition with a lifetime prevalence of 40% and a substantial socioeconomic burden. Because of the high heterogeneity of CBP, subphenotyping may help to improve prediction and support personalized treatment of CBP. To investigate CBP subphenotypes, we decomposed its genetic background into a shared one common to other chronic pain conditions (back, neck, hip, knee, stomach, and head pain) and unshared genetic background specific to CBP. We identified and replicated 18 genes with shared impact across different chronic pain conditions and two genes that were specific for CBP. Among people with CBP, we demonstrated that polygenic risk scores accounting for the shared and unshared genetic backgrounds of CBP may underpin different CBP subphenotypes. In recent years, the interest of consumers in fruit juice extracts as nutraceuticals has increased. Fruits, especially red berries, contain valuable bioactive compounds such as polyphenols. Polyphenols are often associated with anti-oxidant, anti-inflammatory, anti-diabetic, anti-cancer, cardioprotective and gastroprotective properties. However, the relationship between the various effects of fruit juice extracts and their influence on the permeability of the intestinal barrier, as well as their influence on glucose transport across the intestinal membrane, is not known. Therefore, in the present study, anthocyanins and copigments were obtained from 11 fruit juice extracts by XAD7 column chromatography and characterized their health-promoting effects, as well as their influence on the intestinal membrane. Chokeberry, pomegranate and blueberry extracts showed the highest antioxidant activity, but showed incomplete regeneration of the intestinal membrane upon treatment-induced higher permeability. This may depended on the high anthocyanin level of these extracts. Treatments with gojiberry extract, elderberry extract and the copigment fraction of apple achieved the best suitable regeneration of the intestinal barrier. The transcription of epithelial glucose transporters GLUT1 und GLUT2 as well as for the oxidative stress genes catalase (CAT) and superoxide dismutase (SOD) were most effectively reduced by chokeberry extract. To sum up, fruit juice extracts possess high antioxidant potentials and can reduce the expression of antioxidant enzymes and glucose transporters in colon cells. While the glucose uptake may be reduced, the intestinal permeability is increased, which varies due to the extract composition. Therefore, fruit juice extracts need to be fractionated and characterized in more detail to identify the health-beneficial compounds.

Surface-enhanced Raman spectroscopy (SERS) has evolved significantly over fifty years into a powerful analytical technique. This review aims to achieve five main goals. (1) Providing a comprehensive history of SERS's discovery, its experimental and theoretical foundations, its connections to advances in nanoscience and plasmonics, and highlighting collective contributions of key pioneers. (2) Classifying four pivotal phases from the view of innovative methodologies in the fifty-year progression: initial development (mid-1970s to mid-1980s), downturn (mid-1980s to mid-1990s), nano-driven transformation (mid-1990s to mid-2010s), and recent boom (mid-2010s onwards). (3) Illuminating the entire journey and framework of SERS and its family members such as tip-enhanced Raman spectroscopy (TERS) and shell-isolated nanoparticle-enhanced Raman spectroscopy (SHINERS) and highlighting the trajectory. (4) Emphasizing the importance of innovative methods to overcome developmental bottlenecks, thereby expanding the material, morphology, and molecule generalities to leverage SERS as a versatile technique for broad applications. (5) Extracting the invaluable spirit of groundbreaking discovery and perseverant innovations from the pioneers and trailblazers. Psoriasis has been considered a systemic immune-mediated disease that can affect function of the heart. However, certain herbal therapy approaches may nullify side effects of psoriasis on the heart. The aim of this study was to investigate the cardioprotective effects of Galium verum extract administration of the heart of psoriatic rats. The study included 24 Wistar albino male rats, divided into 4 groups: control (CTRL), G.verum (GV), psoriasis (PSORI), and psoriasis with G.verum (PSORI + GV). Seven-day topical application of 5% imiquimod cream was used for induction of psoriasis. After induction, animals were received 125 mg/kg G.verum extract for 4 weeks. Isolated hearts were perfused on the Langendorff apparatus and measured: dp/dt max/min, SLVP, DLVP, HR and coronary flow. The oxidative stress biomarkers: TBARS, NO2, O2- and H2O2 were measured in coronary venous effluent. Isolated hearts were fixed and stained with H/E and Picro-sirius red staining. Psoriasis decreased cardiac contractility and relaxation and increased SLVP and DLVP at all perfusion pressure. Treatment with G.verum in psoriasis rats improved contractility and relaxation of the heart and rise SLVP and DLVP. In PSORI + GV group, the decrease of oxidative stress biomarkers were observed in comparison to PSORI group. Diameter and cross-section area of cardiomyocytes were increased in PSORI and PSORI + GV groups compared to the control. Collagen content was increased in PSORI group by 283% and in PSORI + GV group by 188% compared to control. Rapeseed products, such as protein concentrates, hold promise for addressing global protein demands, but their application in food products is limited by their bitter and astringent taste. This study investigates the use of β-glucosidase (BG) and laccase (LAC) enzymatic treatment, individually and combined, to enhance the flavor of rapeseed protein concentrate (RPC). Untargeted metabolomics and sensory analysis reveal that LAC reduces the bitter compound kaempferol 3- O -(2‴- O -sinapoyl-β-D-sophoroside) (K3OSS) as well as a general reduction in other phenolic compounds, which correlates with a significant decrease in bitterness and astringency. In contrast, BG treatment elevates the levels of K3OSS and is accompanied by increased bitterness due to the conversion of precursor compounds to K3OSS. In addition, the synergistic use of both enzymes significantly reduces the concentration of K3OSS, resulting in a lower perception of bitterness. The LC-MS analysis of pure reference compounds treated with LAC and BG confirms that BG-mediated treatment facilitates the breakdown of larger kaempferol glycosides into K3OSS, while LAC treatment promotes polyphenol polymerization. Consequently, LAC treatment seems to be an effective strategy to improve the sensory quality of RPC and make it more suitable for human consumption.

Background: APRV has been used for ARDS in the past. Little is known about the risk of ventilator- induced lung- injury (VILI) in APRV vs. BIPAP in the management of in COVID19-associated ARDS (CARDS). This study aimed to compare transpulmonary pressures (TPP) in APRV vs. BIPAP in CARDS in regard to lung protective ventilator settings., Methods: This retrospective, monocentric cohort study (ethical approval: 21-1553) assessed all adult ICU- patients with CARDS who were ventilated with BIPAP vs. APRV and monitored with TPP from 03/2020 to 10/2021. Ventilator-settings / -pressures, TPP, hemodynamic and arterial blood gas parameters were compared in both modes., Results: 20 non- spontaneously breathing patients could be included in the study: Median TPPendexpiratory was lower / negative in APRV (-1.20mbar; IQR - 4.88 / +4.53) vs. positive in BIPAP (+ 3.4mbar; IQR + 1.95 / +8.57; p < .01). Median TPPendinspiratory did not differ. In APRV, mean tidal- volume per body- weight (7.05 ± 1.28 vs. 5.03 ± 0.77 ml; p < .01) and mean airway- pressure (27.08 ± 1.67 vs. 22.68 ± 2.62mbar; p < .01) were higher. There was no difference in PEEP, peak-, plateau- or driving- pressure, compliance, oxygenation and CO 2 - removal between both modes., Conclusion: Despite higher tidal- volumes / airway-pressures in APRV vs. BIPAP, TPPendinspiratory was not increased. However, in APRV median TPPendexpiratory was negative indicating an elevated risk of occult atelectasis in APRV- mode in CARDS. Advanced Oxidation Processes (AOPs) have proven to be an effective solution for chemical wastewater treatment, particularly for degradation of organic pollutants, especially dyes. Ozonation is recognized as one of the most prevalent AOPs. Nevertheless, some cases show a lowered efficiency of O 3 utilization which is attributed to its inadequate distribution in the treated water causing low residence time, low mass transfer coefficient as well as shorter half-life. This study demonstrates the application of ozone nanobubbles to enhance the degradation of organics under high pollution load conditions. We propose an integrated method that utilizes bulk nanobubbles to enhance the reactivity of ozone for the degradation of organics. We examined the degradation of organic pollutants under parameters such as varying pH levels, ozone concentrations and presence of salts and surfactants. The degradation of the organic pollutant by ozone nanobubbles (0.177 L mg -1 min -1 ) demonstrated a threefold increase in reaction rate constants compared to microbubbles (0.025 L mg -1 min -1 ). The plausible reason for these findings is (i) higher mass transfer coefficient (ii) higher ozone solubility (iii) NBs may act as a surface for chemical reaction (iv) NBs may increase the half-life of ozone. The presence of reactive oxygen species was verified using scavenging tests. This part of the studies revealed contribution of reactive oxygen species (ROSs) such as hydroxyl radical ( • OH), superoxide radical anion (O 2 •- ) and singlet oxygen ( 1 O 2 ) in the degradation process. The conditions that provide total degradation of 100% in 300 s for both organic pollutants (Green rit and methylene blue dye) are: 5 LPM (Litre per minute) ozone flow rate, acidic pH, monovalent salts (NaCl, 2 mM) and lower concentration of surfactants (CTAB and SDS, 0.3 CMC). A degradation mechanism has been outlined based on intermediates identified by LC-MS. Performance: Spontaneous dissections of the cerebral arteries are among the leading causes of stroke in young adults. They result from hemorrhage into the outer layers of the arterial wall, which can lead to stenosis or even complete vessel occlusion. Clinical presentations vary, ranging from localized pain to cerebral ischemic complications. Early recognition of dissection warning signs and diagnosis through imaging is crucial due to the high risk of recurrent transient ischemic attack (TIA) or stroke within the first two weeks following a dissection, necessitating immediate therapeutic or prophylactic intervention., Achievements: Magnetic resonance imaging (MRI) is the imaging modality of choice, as it can reliably visualize all characteristic features of dissection. Typically, thin-slice, fat-saturated 3D black-blood sequences are sufficient to detect the pathognomonic intramural hematoma., Practical Recommendations: Acute treatment and secondary prevention depend on the extent of neurological deficits, the location and size of the dissection, any sequelae, concomitant pathologies, and risk factors. Background: Happiness is a concept in positive psychology. Studies have shown links between happiness, courses of diseases and health. In dermatology the role of happiness has not yet been sufficiently investigated., Research Question: What impact do skin diseases have on individual happiness and what contribution can happiness make to holistic dermatological care?, Material and Methods: Processing of the current literature for a selective overview of primary scientific studies on happiness based on heuristic happiness and subjective well-being measured by positive and negative affects as well as satisfaction with life in dermatology., Results: Patients with skin diseases show significant impairments in happiness, especially in the positive affect and satisfaction with life compared to the healthy population. The impairments appear to be expressed differently for different dermatoses; for example, patients with skin cancer show significantly higher life satisfaction than other dermatological patients. Influencing factors vary in their strength and direction depending on the operationalization of happiness and the skin disease., Discussion: Dermatological diseases appear to be associated with impairments in happiness, particularly in the positive affect. Integrating happiness into dermatological care could help to have a better understanding of patients' individual needs in the context of holistic, person-centered healthcare.
