This study introduces the development of a photothermal-based microfluidic paper analytical device (PT-µPAD) integrated with near-field communication (NFC) technology and smartphone readout for enzyme-free glucose quantification in human samples. With the properties of gold nanoparticles (AuNPs) both as a nanozyme and as a photothermal substrate, there is no need for costly reagents like enzymes or a readout instrumentation for the selective and sensitive detection of glucose. In PT-µPADs, AuNPs are etched by hydrogen peroxide (H2O2) generated from glucose catalysis. Photothermal detection from the plasmonic heating of these AuNPs when illuminated by a 533nm LED light source is achieved by inserting the PT-µPAD sensor into a portable NFC platform suitable for smartphone readout. Temperature variation is directly proportional to the glucose concentration. After optimization, we acquired a linear range between 5.0 and 20.0 µmol L−1 (R2 = 0.9967) and a limit of detection (LOD) of 25.0 nmol L−1 for glucose. Additionally, while our sensor does not utilize any enzyme, it is remarkably selective to glucose with no effects from interferences. Recovery studies in various human control samples indicated a range of 99.73–102.66% with the highest RSD of 3.53%, making it highly accurate and precise. Moreover, our method is more sensitive than other methods relying on conventional µPADs for glucose sensing. By integrating the potential benefits of microfluidics, nanomaterials as nanozymes, and NFC technology for wireless readout, our sensor demonstrates great promise as an accessible, affordable, and shelf-stable device for glucose quantification. This article presents the development of a distance-based thread analytical device (dTAD) integrated with an ion-imprinted polymer (IIP) for quantitative monitoring of zinc ions (Zn2+) in human urine samples. The IIP was easily chemically modified onto the thread channel using dithizone (DTZ) as a ligand to bind to Zn2+ with methacrylic acid (MAA) as a functional monomer and ethylene glycol dimethacrylate (EGDMA) as well as 2,2-azobisisobutyronitrile (AIBN) as cross-linking agents to enhance the selectivity for Zn2+ detection. The imprinted polymer was characterized using Attenuated Total Reflectance-Fourier Transform Infrared (ATR-FTIR) spectroscopy and Scanning Electron Microscopy-Energy Dispersive X-ray Spectroscopy (SEM-EDS). Under optimization, the linear detection range was from 1.0 to 20.0 mg L-1 (R2 = 0.9992) with a limit of detection (LOD) of 1.0 mg L-1. Other potentially interfering metal ions and molecules did not interfere with this approach, leading to high selectivity. Furthermore, our technique exhibits a remarkable recovery ranging from 100.48% to 103.16%, with the highest relative standard deviation (% RSD) of 5.44% for monitoring Zn2+ in human control urine samples, indicating high accuracy and precision. Similarly, there is no significant statistical difference between the results obtained using our method and standards on zinc supplement sample labels. The proposed method offers several advantages in detecting trace Zn2+ for point-of-care (POC) medical diagnostics and environmental sample analysis, such as ease of use, instrument-free readout, and cost efficiency. Overall, our developed dTADbased IIP method holds potential for simple, affordable, and rapid detection of Zn2+ levels and can be applied to other metal ions' analysis. [ABSTRACT FROM AUTHOR]

Selectivity and enhanced response remain an issue in gaseous acetone sensing. This report demonstrates MoS2 on MXene (Ti3C2Tx) contact based on a low-cost flexible sensor for selective detection of acetone using principle component analysis and binary logistic regression. The piezotronic effect enhances the response of the sensor. The fabricated acetone sensor exhibited an excellent response of ∼5.84% at lower concentrations of acetone ranging from 5 to 30 ppm at room temperature (25 °C). The sensor's response values with (5%) and without applied external strain were calculated to be approximately 5.84 and 7.21%, respectively. This suggested an increment in the sensing performance under the applied external strain with an improved response time (448 s). A detailed study is presented in terms of comparative performance of MoS2 and MXene/MoS2 under strained and unstrained conditions. Principle component analysis and binary logistic regression techniques were used to successfully selectively detect acetone among other eight VOC (volatile organic compound) (toluene, benzene, isopropyl alcohol, formaldehyde, hydrogen sulfide, dimethyl sulfide, ethanol, and methanol) gases. A real-time breath analyzer is presented wherein an artificial breath consisting of different humidities and acetone was created, and acetone was sensed using the fabricated sensor. Finally, a proof of concept for the flexible bracelet is presented for the on-site detection of acetone levels in human breath to monitor diabetes at the point-of-care setting. The small intestinal (SI) microbiota plays an important role in human health and disease. Current methods based on fecal analysis do not provide an accurate representation of the SI microbiome. This article introduces an ingestible sensor for the quantification of Escherichia coli (E. coli) in the SI by developing the fluorescent thread‐based ingestible pill analytical device (TPAD) using fluorescent gold nanoclusters (AuNCs) coupled with molecularly imprinted polymers (MIPs). Quantitative detection lies in the change in fluorescence signal on thread as proportional to target bacteria concentrations using a smartphone. Utilizing a pH‐sensitive enteric coating, this device facilitates localized sampling of the SI microbiome. Hydrophilic beads are employed to autonomously seal the device after sampling, offering a self‐locking mechanism. The sensor demonstrates a linear range for E. coli detection between 2.0 × 103 and 14.0 × 103 CFU mL−1 with an R2 of 0.9975 and a detection limit (LOD) of 400.0 × 102 CFU mL−1. The sensor also shows remarkable precision and acceptable accuracy for monitoring E. coli levels in in vivo animal studies compared to commercially available test kits. The developed TPAD, therefore, offers a novel, low‐cost, and non‐invasive method for rapidly diagnosing gastrointestinal (GI)‐related medical conditions and studying host–microbiome interactions. Technologies capable of noninvasively sampling different locations in the gut upstream of the colon will enable new insights into the role of organ‐specific microbiomes on human health. Herein, an ingestible pill for the sampling of gut lumen based on one of the earliest hydraulic machines known as an Archimedes screw is reported. The design contains twisted wires as Archimedes screw driven by a motor, wirelessly activated using a magnet. The sampling performance of the screw‐pump pill is characterized using realistic in vitro models and validated in vitro using E. coli expressing different fluorescent proteins. The use of the Archimedes screw enables the pill to sample the dense GI environment. The pill is also tested ex vivo in the pig intestine and in vivo in pigs. Herein, the results show that the bacterial populations recovered from the pill's chamber closely resemble the targeted bacterial population of the microenvironment to which the pill is exposed. Such ingestible devices have the potential to revolutionize the understanding of the spatial diversity of the gut microbiome and its response to medical conditions and treatments.
