1. Methylation alterations of imprinted genes in different placental diseases

Abstract

Abstract Background Imprinted genes play important functions in placentation and pregnancy; however, research on their roles in different placental diseases is limited. It is believed that epigenetic alterations, such as DNA methylation, of placental imprinting genes may contribute to the different pathological features of severe placental diseases, such as pre-eclampsia (PE) and placenta accreta spectrum disorders (PAS). Results In this study, we conducted a comparative analysis of the methylation and expression of placental imprinted genes between PE and PAS using bisulfite sequencing polymerase chain reaction (PCR) and quantitative PCR, respectively. Additionally, we assessed oxidative damage of placental DNA by determining 8-hydroxy-2′-deoxyguanosine levels and fetal growth by determining insulin-like growth factor 2 (IGF2) and cortisol levels in the umbilical cord blood using enzyme-linked immunosorbent assay. Our results indicated that methylation and expression of potassium voltage-gated channel subfamily Q member 1, GNAS complex locus, mesoderm specific transcript, and IGF2 were significantly altered in both PE and PAS placentas. Additionally, our results revealed that the maternal imprinted genes were significantly over-expressed in PE and significantly under-expressed in PAS compared with a normal pregnancy. Moreover, DNA oxidative damage was elevated and positively correlated with IGF2 DNA methylation in both PE and PAS placentas, and cortisol and IGF2 levels were significantly decreased in PE and PAS. Conclusions This study suggested that DNA methylation and expression of imprinted genes are aberrant in both PE and PAS placentas and that PE and PAS have different methylation profiles, which may be linked to their unique pathogenesis.

Abstract

Autism spectrum disorder (ASD) is a complex neurodevelopmental condition characterized by diverse clinical features. EEG biomarkers such as spectral power and functional connectivity have emerged as potential tools for enhancing early diagnosis and understanding of the neural processes underlying ASD. However, existing studies yield conflicting results, necessitating a comprehensive, data-driven analysis. We conducted a retrospective cross-sectional study involving 246 children with ASD and 42 control children. EEG was collected, and diverse EEG features, including spectral power and spectral coherence were extracted. Statistical inference methods, coupled with machine learning models, were employed to identify differences in EEG features between ASD and control groups and develop classification models for diagnostic purposes. Our analysis revealed statistically significant differences in spectral coherence, particularly in gamma and beta frequency bands, indicating elevated long range functional connectivity between frontal and parietal regions in the ASD group. Machine learning models achieved modest classification performance of ROC-AUC at 0.65. While machine learning approaches offer some discriminative power classifying individuals with ASD from controls, they also indicate the need for further refinement.

Abstract

With the rapid growth of computer technology and multimedia technology, VR technology is regularly developed and applied to entire walks of life. This paper first introduces the VR double master teaching model into the practical teaching of Civics in colleges and universities (CU) and analyzes the shortcomings and limitations of the traditional teaching design scheme. Secondly, under the guidance of constructivist learning theory and contextual cognitive theory, as well as the basic theory of humanistic theory, combined with the characteristics of venue learning, and also with the implementation of gesture interaction technology and face expression detection technology with the help of depth sensors, a teaching design scheme with immersion and interactivity based on red VR venues is designed. The results show that the response time of logging into the business through the performance test of the VR Civics teaching system is 5.239s - 3s, and the thinking time is 2.271 s, which is in line with the result of logging in less than 3 s. Moreover, the Civics performance of students taught through VR has improved by 11.2 percentage points, and the teaching effect and system performance are in line with the practice expectation. It shows that the virtual reality technology proposed in this paper and Civics teaching have achieved an effective combination and realized the fundamental purpose of implementing moral education.

5. ARTS is essential for di-2-ethylhexyl phthalate (DEHP)-induced apoptosis of mouse Leydig cells

Abstract

As an extensively employed plasticizer in industrial applications, di-2-ethylhexyl phthalate (DEHP) can induce apoptosis of mouse Leydig cells, yet the precise mechanism remains elusive. In the current study, we identified that DEHP could specially induced apoptosis in the Leydig cells of the testis tissue, accompanied with the upregulation of apoptosis-related protein in the TGF-β signaling pathway (ARTS) in the cells. Overexpression of ARTS significantly induced apoptosis of TM3 cells, while knockdown of ARTS inhibited apoptosis. Furthermore, DEHP-induced apoptosis of TM3 cells could be alleviated by knockdown of ARTS, which indicated that ARTS was involved in DEHP-induced apoptosis of mouse Leydig cells. Bioinformation assay predicts that there are four potential p53-responsive elements (p53-REs) located at − 6060, − 5726, − 5631 and − 5554 before the transcription start site of ARTS gene, implying that gene transcription of ARTS could be regulated by p53. Interestingly, DEHP was shown to specifically upregulate the expression of p53 in the Leydig cells of the testis tissue and TM3 cells. Consistently, p53 was proved to bind to the RE4 site of the ARTS gene promoter and transcriptionally activated the promoter-driven expression of the luciferase reporter gene. Overexpression of p53 could induce apoptosis of TM3 cells; while knockdown of p53 could not only rescue DEHP-induced apoptosis of the cells, but also inhibit DEHP-caused upregulation of ARTS. Meanwhile, we showed that oxidative stress could induce apoptosis of TM3 cells, accompanied with the increased protein levels of p53 and ARTS; while inhibition of oxidative stress dramatically alleviated DEHP-induced apoptosis and the up-regulation of p53 and ARTS. Taken together, these results indicated that DEHP-induced oxidative stress activates the p53-ARTS cascade to promote apoptosis of mouse Leydig cells.

6. $B\to K\bar K(\pi\eta)h$ decays in the presence of isovector scalar resonances $a_0(980,1450)$

Abstract

Different from the previous treatment in a two-body framework, we introduce the dimeson distribution amplitudes (DAs) to describe the strong dynamics between the S-wave resonances $a_0(980, 1450)$ and the $K\bar K (\pi\eta)$ pair, where the Gegenbauer coefficient required is determined from the experimental data on the time-like form factors involved. The branching ratios and direct CP asymmetries of the decays $B \to a^{(\prime)}_0 h \to K\bar K(\pi\eta) h$, with $a_0=a_0(980)$, $a^{\prime}_0=a_0(1450)$ and $h$ referring to a pion or a kaon, are then calculated in the perturbative QCD (PQCD) approach. We find that the branching ratios of the corresponding quasi-two-body decays $B\to a^{(\prime)}_0 K$ obtained with the narrow width approximation are closer to those predicted in the QCD factorization (QCDF) approach compared to the previous PQCD calculations, no matter a three-body or a two-body framework is assumed. Furthermore, all our predictions for these $B\to a^{(\prime)}_0 K$ decays are below the current experimental upper limits except for those of decays $B^0\to a^{(\prime)-}_0K^+$, which are (slightly) larger than the upper limits. Under the narrow width approximation, the branching ratios of the decays $B^+\to a^{(\prime)+}_0\pi^0$, $B^0\to a^{(\prime)+}_0\pi^-$ and $B^0\to a^{(\prime)0}_0\pi^0$ are comparable to or agree well with the previous PQCD and the QCDF calculations. While for the decays $B^+\to a^{(\prime)0}_0\pi^+$ and $B^0\to a^{(\prime)-}_0\pi^+$, their branching ratios are predicted to be unexpectedly large, for example, the obtained branching ratio of decay $B^+\to a^0_0\pi^+$ is even higher than the current experimental upper limit., Comment: 22 pages, 2 figures

7. Semileptonic and nonleptonic decays of $B_{u,d,s,c}^{*}$ in the covariant light-front approach

Abstract

The semileptonic and nonleptonic decays of the b-flavor vector mesons $B^{*}_{u,d,s}$ and $B_{c}^{*}$ are investigated within the covariant light-front quark model (CLFQM). By calculating the form factors of the transitions $B_{u, d, s, c}^{*}\to P$ under the CLFQM, with $P$ denoting a pseudoscalar meson, i.e., $\pi, K, \eta_c(1S,2S), D_{(s)}, B_{(s)}$, we predict and discuss several physical observables, including the branching ratios, polarization fractions $f_{L}, f_{\|}$, and forward-backward asymmetries $A_{FB}$. The total widths of the single-photon radiative decay channels for these b-flavor vector mesons are estimated using their partial widths. In these considered decays, one can find that the semileptonic decays $B_{s}^{*0}\to D_{s}^{-}\ell^{\prime+}{\nu}_{\ell^\prime}$ and $B_{c}^{*+}\to B_{s}^{0}\ell^{\prime+}{\nu}_{\ell^\prime}, \eta_{c}\ell^{\prime+}{\nu}_{\ell^\prime}$, with $\ell^\prime$ being $e$ or $\tau$, and the nonleptonic channels $B_{c}^{*+}\to B^0_{s} \pi^{+}, B^0_{s} \rho^{+}$ have the largest branching ratios, which can reach up to the $10^{-7}$ order, and are most likely to be accessible at the future high-luminosity LHCb and Belle-II experiments., Comment: 30 pages, 4 figures,accepted for publication in Chin. 8. TALENT: A Tabular Analytics and Learning Toolbox

Abstract

Tabular data is one of the most common data sources in machine learning. Although a wide range of classical methods demonstrate practical utilities in this field, deep learning methods on tabular data are becoming promising alternatives due to their flexibility and ability to capture complex interactions within the data. Considering that deep tabular methods have diverse design philosophies, including the ways they handle features, design learning objectives, and construct model architectures, we introduce a versatile deep-learning toolbox called TALENT (Tabular Analytics and LEarNing Toolbox) to utilize, analyze, and compare tabular methods. TALENT encompasses an extensive collection of more than 20 deep tabular prediction methods, associated with various encoding and normalization modules, and provides a unified interface that is easily integrable with new methods as they emerge. In this paper, we present the design and functionality of the toolbox, illustrate its practical application through several case studies, and investigate the performance of various methods fairly based on our toolbox. 9. A Closer Look at Deep Learning on Tabular Data

Abstract

Tabular data is prevalent across various domains in machine learning. Although Deep Neural Network (DNN)-based methods have shown promising performance comparable to tree-based ones, in-depth evaluation of these methods is challenging due to varying performance ranks across diverse datasets. In this paper, we propose a comprehensive benchmark comprising 300 tabular datasets, covering a wide range of task types, size distributions, and domains. We perform an extensive comparison between state-of-the-art deep tabular methods and tree-based methods, revealing the average rank of all methods and highlighting the key factors that influence the success of deep tabular methods. Next, we analyze deep tabular methods based on their training dynamics, including changes in validation metrics and other statistics. For each dataset-method pair, we learn a mapping from both the meta-features of datasets and the first part of the validation curve to the final validation set performance and even the evolution of validation curves. This mapping extracts essential meta-features that influence prediction accuracy, helping the analysis of tabular methods from novel aspects. Based on the performance of all methods on this large benchmark, we identify two subsets of 45 datasets each. The first subset contains datasets that favor either tree-based methods or DNN-based methods, serving as effective analysis tools to evaluate strategies (e.g., attribute encoding strategies) for improving deep tabular models. The second subset contains datasets where the ranks of methods are consistent with the overall benchmark, acting as a probe for tabular analysis. These ``tiny tabular benchmarks'' will facilitate further studies on tabular data.

10. Fukaya categories of hyperplane arrangements

Abstract

To a simple polarized hyperplane arrangement (not necessarily cyclic) $\mathbb{V}$, one can associate a stopped Liouville manifold (equivalently, a Liouville sector) $\left(M(\mathbb{V}),\xi\right)$, where $M(\mathbb{V})$ is the complement of finitely many hyperplanes in $\mathbb{C}^d$, obtained as the complexifications of the real hyperplanes in $\mathbb{V}$. The Liouville structure on $M(\mathbb{V})$ comes from a very affine embedding, and the stop $\xi$ is determined by the polarization. In this article, we study the symplectic topology of $\left(M(\mathbb{V}),\xi\right)$. In particular, we prove that their partially wrapped Fukaya categories are generated by Lagrangian submanifolds associated to the bounded and feasible chambers of $\mathbb{V}$. A computation of the Fukaya $A_\infty$-algebra of these Lagrangians then enables us to identity these wrapped Fukaya categories with the $\mathbb{G}_m^d$-equivariant hypertoric convolution algebras $\widetilde{B}(\mathbb{V})$ associated to $\mathbb{V}$. This confirms a conjecture of Lauda-Licata-Manion (arXiv:2009.03981) and provides evidence for the general conjecture of Lekili-Segal (arXiv:2304.10969) on the equivariant Fukaya categories of symplectic manifolds with Hamiltonian torus actions., Comment: Fix graphical issues, add some references and expand some of the expositions in section 3.
