Abstract Background Most individuals having Alzheimer’s disease generally experience increased frequency of awakenings as well as prolonged wakefulness periods at night. Brain wave research indicate reductions in both REM (dreaming stage) along with NREM (non-dreaming stage). The inherited pattern of Alzheimer’s disease is attributed to presence of genetic polymorphisms. The association of these polymorphisms and sleep pattern in Alzheimer’s disease patients was not assessed before. Purpose This research was aimed at assessing the association between sleep quality as well as genetic polymorphisms of ACE, ACE2 and Apo E among Alzheimer’s disease patients in Upper Egypt. Patients and methods Our cross-sectional study with analytic component involved 50 cases having Alzheimer’s disease evaluated utilizing Pittsburg sleep quality index for sleep quality. They were accordingly divided into good as well as poor sleep quality groups. Genetic analysis of ACE, ACE2 (rs2285666) SNPs and Apo E (rs121918398) SNPs was done using (RFLP-PCR) I. Results No statistically significant variation was documented among both groups regarding age as well as age at diagnosis. Female patients were more susceptible for poor sleep quality. In addition, patients came from rural areas were significantly higher among poor sleep quality group. Statistical significant variations were documented among both groups as regard presenting symptoms and HAM-A and HAM-D. No statistically significant variations were documented among both groups as regard ACE genotypes; however, ACE 2 AA genotype and A allele were higher with statistically significant difference among poor sleep quality group. Apo E3/E4 genotype and E4 allele were greater with statistically significant variations among poor sleep quality group. Conclusions Female patient and rural Alzheimer’s disease patients at high risk for poor sleep quality. In addition, Apo E4 carriers and ACE2 AA carriers are at increasing susceptibility for poor sleep quality.

Abstract Background Self- medication leading to wastage of limited resources in developing countries, prolonged suffering, increase resistance to drugs and may result in significant medical complications such as adverse drug responses and dependence. Self-medication is extensively used by health professionals. Undergraduate medical students as being the future physicians representing a main pillar in health care system thus have special significance. This study aimed to estimate the prevalence of self-medication and identify its pattern among medical students in Tanta university, Egypt. Methods A four-month cross-sectional study was carried out among Egyptian undergraduate medical students at Tanta University, from first to final year. (November 2023 – February 2024). The students were chosen using a two-stage cluster sampling method, and data on sociodemographic and self-medication patterns were collected using a predesigned semi-structured self-administered questionnaire. Results The prevalence of self-medication was 71%. The key determinants of self-medication was students’ medical knowledge from self- experience and studies (55.9%). Headache was a common complaint for self-medication (80.4%). Majority of them (88.3%) use pharmaceutical products where analgesics lies on top (92.4%). The degree of popularity of the medicine was the primary reason for drug selection (52.5%) and recommendation of the pharmacist was the main determinate of selecting type of drug (43.6%). However, 30.2% experienced side effects, of which 50% went to private physician and 33.5% stopped taking their medications. Half of the students took antibiotics for self-medication. Conclusions Self-medication is a common practice among medical students. where headache was the common symptom and the most commonly utilized medications for self-medication were analgesics. There is a need to augment the value of diagnosis, awareness and seriousness of this practice.

Abstract Background Hydroxyapatite (HAP) resembles the components of biological hard tissue. Recent research has been interested in the biomedical application of HAP nanoparticles (HAP-NPs) in cancer treatment, HAP-NPs have high cytotoxic activity against cancerous cells, in addition, they are nontoxic to healthy normal cells, biocompatible, biodegradable, and have a high absorption rate within the tissue. Therefore, this study evaluated HAP-NPs' antitumoral activity in Ehrlich solid carcinoma (ESC)-bearing mice, in addition, we examined the anticancer efficacy of combined treatment of a common chemotherapeutic drug such as Cisplatin (CDDP) and HAP-NPs in ESC-bearing mice. Methods Forty female mice were inoculated with 200 µl of diluted ascites fluid containing approximately two million viable cancer cells in the mice's left thigh, after 14 days of inoculation, the mice were distributed into four groups: 10 mice in each. ESC group was administrated distilled water, the HAP-NPs group was treated orally with 100 mg/kg of hydroxyapatite nanoparticles, the CDDP group was administrated intraperitoneally with 5 mg/kg of Cisplatin, the HAP-NPs + CDDP group was treated with both doses of hydroxyapatite and cisplatin, the animal treatment was conducted for 20 days. Antitumor activity was assessed for two durations after 10 and 20 days. DNA damage assessment was performed using comet assay in ESC, in addition, we measured the expression of the following genes (P53, Bcl2, and Bax,) using quantitative real-time PCR, and the apoptotic-related proteins (P53 and Ki-67) using immunohistochemical analysis. A histopathological examination of ESC was performed. Results The obtained data illustrated a promising anticancer activity of HAP-NPs, and the combined treatment of HAP-NPs and CDDP illustrated a higher anticancer efficacy. Abstract Background Childhood is a crucial period that shapes a person’s growth and development. For orphans, a lack of familial support affects their upbringing, making orphanages crucial for care. Children living in orphanage centers are vulnerable to several conditions, including dermatological disorders, due to factors such as malnutrition, overcrowding, and poor hygiene. The current study was carried to determine the prevalence and patterns of skin disorders among orphan children in Egypt, and to identify associated risk factors among orphans in Egypt. Methods A cross-sectional study was conducted among 415 children aged 1–18 years living in orphanage centers in Fayoum and Giza governorates, Egypt. Results Among the children, 77.1% had at least one skin disorder, with noninfectious conditions being more prevalent than infectious conditions (67.4 vs. 24.3%). The most common noninfectious conditions were dermatitis/eczema, pigmentary disorders, skin appendage disorders, and psycho-cutaneous issues. Fungal infections were the most common infectious condition. There was a significant relationship between hygiene and the type of skin disorder. Conclusion The prevalence of skin diseases in orphanages in Egypt is relatively high and is dominated by noninfectious skin diseases, and the prevalence of psycho-cutaneous disorders is high among these children. Comprehensive care strategies focusing on improving hygiene and emotional support and preventing abuse are essential for reducing the incidence of skin disorders and enhancing the overall health of orphaned children.

Abstract This study was designed to assess the effect of brentuximab vedotin on several breast cancer cell lines in terms of promoting apoptosis and managing cancer progression. Additionally, the study investigated the potential of repurposing this drug for new therapeutic reasons, beyond its original indications. The study evaluates the cytotoxic effects of Brentuximab vedotin across five cell lines: normal human skin fibroblasts (HSF), three breast cancer cell lines (MCF-7, MDA-MB-231, and T-47D), and histiocytic lymphoma (U-937). Brentuximab treatment was administered at four time points (0, 24, 48, and 72 h), with cell viability assessed at each interval. HSF cells, serving as controls, exhibited minimal viability loss (above 70%), indicating limited toxicity in normal fibroblasts. In contrast, MCF-7 and MDA-MB-231 cells demonstrated time-dependent reductions in viability, with a pronounced decline by 72 h, suggesting Brentuximab’s efficacy in both ER-positive and triple-negative breast cancer. T-47D cells also showed decreased viability, though at a slower rate. U-937 cells exhibited the most substantial reduction, highlighting Brentuximab’s potent activity against hematologic malignancies. Wound healing assays further revealed that Brentuximab significantly impaired the migration and healing capacity of cancer cells compared to untreated controls. Additionally, cell cycle analysis indicated G2/M phase arrest in cancer cells, particularly in MCF-7 and MDA-MB-231, while HSF cells remained largely unaffected. Apoptosis detection confirmed Brentuximab-induced cell death, with significant increases in late apoptosis in cancer lines, especially by 72 h. Gene expression analysis revealed upregulation of pro-apoptotic genes (BAX, Caspase 3, and Caspase 9) in cancer cells, alongside a decrease in anti-apoptotic BCL-2 expression. These findings suggest Brentuximab’s selective cytotoxicity against cancer cells and its potential as an effective therapeutic agent, particularly in breast cancer and histiocytic lymphoma.

Abstract With freshwater resources becoming scarce worldwide, mariculture is a promising avenue to sustain aquaculture development, especially by incorporating brackish and saline groundwater (GW) use into fish farming. A 75-day rearing trial was conducted to evaluate fish growth, immune response, overall health, and water quality of Chelon ramada cultured in brackish GW and fed on a basal diet (BD) augmented with rosemary oil (RO) or RO + zymogen forte™ (ZF) as an anti-flatulent. Abstract Background This study investigated the effect of B. Subtilis bacteria on the properties of cement mortar. This was done by using soil samples from Sharkia, Egypt, to isolate 48 bacterial strains, after which they were cultured using the Johnson method and various media. Bacteria were then added to the cement mortar in amounts of 5% and 10% by weight to evaluate their effect on the mechanical and chemical properties of the modified mortar. Results The study examined the compressive and flexural strength of the modified mortar over time, as well as its microscopic properties and chemical composition after 28 days. The results indicated that bacterial additions of 5% and 10% increased the compressive strength of the mortar after 28 and 56 days compared to the control. A 5% bacteria concentration resulted in significant improvements in strength, showing the best concentration for increasing mortar strength. The addition of 5% bacteria significantly enhanced the early flexure strength, while the 10% showed superior long-term strength after 56 days. Scanning electron microscopy (SEM) revealed high CaCO3 deposits in the bacterial samples, indicating microbial-induced calcite precipitation that filled the small cracks and increased strength. Fourier-transform infrared spectroscopy (FTIR) confirmed the presence of hydroxyl, carbonate, and silicate groups, with bacterial samples having a higher carbonate content, indicating an increase in calcium carbonate formation and microstructure. Conclusions The ideal bacterial concentration was 5% as it improved the compressive and flexural strength while also promoting a more flexible microstructure. This study supports the employment of microorganisms in the production of more durable and environmentally friendly building materials, enhancing the sustainability of building practices.

Abstract Background Anterior open bite is a challenging condition for pediatric dentists and orthodontists as it causes aesthetic, speech, feeding, and psychological problems; this emphasizes the need for early diagnosis and interception of this malocclusion. Aim This study aimed to evaluate the effects of prefabricated metal-bonded tongue tamers and customized bonded spurs in the early treatment of anterior open bite. Materials and methods A sample of seventy-five children aged 7–9 years were assigned into three groups in which anterior open bite was treated using tongue tamers (group-I), customized composite bonded spurs(group-II), and conventional fixed palatal cribs (group-III). Study model and cephalometric x-ray evaluations were done before and after a three-month follow-up. Data was gathered and statistically analyzed using ANOVA and Bonferroni tests. Results Model overbite at central and lateral incisors was increased in all groups. The highest increase was found in group-I(3.16 ± 1.17, 2.81 ± 0.94)and group-III(3.08 ± 1.10,2.99 ± 1.00) though the difference was not statistically significant. Also, cephalometric overbite was non-significantly increased in all groups with a high increase in group-III(3.13 ± 1.10). Overjet decreased in all groups, with the greatest reduction observed in group-I(-2.58 ± 1.02 and -2.47 ± 0.80 in model and cephalometric respectively) and was significantly different from group-II using pairwise analysis. There was a decrease in SNA and ANB in all groups with more significant improvement in groups-I(-1.20 ± 0.88,-1.65 ± 0.74) and -III(-1.31 ± 0.92, -1.62 ± 0.75) than group-II(-0.63 ± 0.46, -0.90 ± 0.43). Moreover, FMA measurements decreased significantly in group-I (-2.6 ± 1.11). Regarding SNB measurement, there was a non-significant increase in all three groups with the highest increase in group-I(0.49 ± 0.48) followed by group-III(0.34 ± 0.63). The U1/FHP and L1/GoGn angles were non-significantly decreased in all tested groups with the highest decrease in group-I(-1.76 ± 1.00 and-2.54 ± 0.87 respectively). Conclusion Early treatment of anterior open bite, along with tongue tamers' simplicity and aesthetics promoted the malocclusion correction and occlusal function restoration. Background Trichodynia or hair pain refers to the painful sensation occurring in the hair and or scalp in the setting of hair shedding. It has been reported mainly in association with telogen effluvium and androgenetic alopecia (AGA). Recently, trichodynia cases have been growing in numbers. Several theories have been proposed to explain the pathogenesis of trichodynia, suggesting a major role for substance P (SP) accompanied by perifollicular inflammation. Objective To assess tissue and serum levels of SP in trichodynia occurring in AGA patients. Patients and methods This is a case–control study that included 35 patients of trichodynia in AGA as well as 35 age-matched and sex-matched healthy participants included as controls. The diagnosis was made by clinical, trichoscopic examination, and scalp biopsy. Tissue and serum levels of SP were measured using the enzyme-linked immunoassay technique. Abstract Background Globally, salinity poses a threat to crop productivity by hindering plant growth and development via osmotic stress and ionic cytotoxicity. Plant extracts have lately been employed as exogenous adjuvants to improve endogenous plant defense mechanisms when grown under various environmental stresses, such as salinity. This study investigated the potential of melatonin (Mt; 0, 50, and 100 mM) as an antioxidant and licorice root extract (LRE; 0.0 and 3%) as an organic biostimulant applied sequentially as a foliar spray on faba bean (Vicia faba L.) grown in cadmium (Cd)-contaminated saline soil conditions [Cd = 4.71 (mg kg− 1 soil) and ECe = 7.84 (dS m− 1)]. Plants not receive any treatment and sprayed with H2O were considered controls. The experimental treatments were laid out in strip plot in a randomized complete block design replicated thrice, where the LRE and Mt were considered as vertical and horizontal strips, respectively. Growth characteristics, photosynthetic pigments, nutrient uptake, physiology and metabolic responses, anatomical features, and yield were assessed. Results Cadmium (Cd) and salinity-induced stress significantly altered leaf integrity, photosynthetic efficiency, total soluble sugars (TSS), free proline (FPro), total phenolic, DPPH, and total soluble proteins (TSP), non-enzymatic and enzymatic antioxidants, growth characteristics and yield-related traits. However, the application of LRE + Mt considerably improved these negative effects, with higher improvements were observed due to application of LRE + Mt100. Application of LRE + Mt significantly reduced hydrogen peroxide (H2O2) accumulation, lipid peroxidation and Cd content in leaves and seeds, all of which had increased due to Cd stress. Application of LRE + Mt significantly mitigated the Cd-induced oxidative damage by increasing the activity of reactive oxygen species (ROS) scavenging enzymes such as superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase, in parallel with enhanced ascorbate and reducing glutathione content. Exogenous application of LRE + Mt significantly increased osmolyte content, including FPro, TSS, and total phenols and mitigated Cd-induced reduction to considerable levels. Conclusions Our findings showed that LRE + Mt increased V. faba plants’ morphological, physiological, and biochemical properties, reducing Cd stress toxicity, and promoting sustainable agricultural practices. Abstract Environmental heat stress causes significant economic loss in the poultry industry. Therefore, interest has increased in using feed additives to reduce the negative impacts of heat stress on the chickens and improve production performance. This study aimed to assess the effect of supplementing with Nigella sativa nanoparticles (Nano-NS) as an anti-stress and growth promoter in broiler diets under hot climatic conditions. A total of 375 male one-day-old Ross 308 chicks were randomly divided into a control group and four treatment groups (75 chicks/group). The first group fed a basal diet without additives, the second group fed a basal diet supplemented with avilamycin at 50 mg/kg, and the other groups fed a basal diet supplemented with 30, 40, and 50 mg/kg Nano-NS, respectively. Abstract Formulation and adjuvant technologies can facilitate the use of insecticides that have higher biological efficiency application features. Safety, physicochemical properties by increasing consumer demand for safe food and enhancing operator safety. The aim of this current work was to develop a green efficient, and stable pesticide formulation. Therefore, certain nano emulsions with and without Adjuvants Calcium Alkyl Benzene Sulphonate (Atlox 4838B), and non-ionic surfactant based on trisiloxane ethoxylate (ARGAL), were testing against Sitophilus oryzae (Coleoptera: Curculionidae). Certain analytical techniques were used for determining the characterization of the nano emulsions (Sesame, Clove, and Cinnamon). Results showed that all formulations were penal, achieving nanometric size for all compounds. Scanning Electron Microscopy (SEM) micrographs revealed spherical or quasi-spherical morphologies for the tested nanoemulsion formulation nanodroplets. Furthermore, dynamic light scattering (DLS) showed that the particle size of the formulation with the adjuvants showed a slight increase in the droplet size compared to the formulations without adjuvants. In comparison to the tested nanoemulsions with adjuvants, the viscosity of the nanoemulsions without adjuvants was lower. All studied formulations, both with and without adjuvant, showed an acidic to slightly acidic pH, except for sesame (NE) with AtloxTM 4838B, which showed a neutral pH, and they were kinetically stable with no phase separation, creaming, or crystallization. Furthermore, supporting the stability of these nanoemulsion particles was the absence of a separation phase following centrifugation, freeze–thaw cycles, and heating–cooling cycles. Abstract This study was performed to reveal the metabolic effects and molecular mechanisms that govern the dietary incorporation of clenbuterol on growth performance, haemato-biochemical changes, histological alteration, and gene expression regulating glucose and lipid metabolism in normal and high-fat diets fed in Nile tilapia (Oreochromis niloticus). Six experimental diets were formulated, incorporating different concentrations of clenbuterol. The 1st three groups were supplemented with a diet comprising 6% fat, with clenbuterol of 0, 5, and 10 g/kg diet was designated as F6 clenb0, F6clenb5, and F6clenb10, respectively. The other treatment groups were fed a diet of 12% fat, with clenbuterol 0, 5, and 10 g/kg diet, respectively termed F12 clenb0, F12 clenb5, and F12 clenb10. The results revealed that compared to the control group, HFD exhibited a marked reduction in FBW, BWG, PER, and body protein percent but significantly increased the FCR, IPF, liver fat percent, and body ash percent with altered hematological parameters, raised serum biomarkers of hepatic and renal injury. HFD signally raised mRNA expression of pro-inflammatory cytokines, and declined nrf2 and antioxidative function-related genes. Also increased mRNA expression of lipogenic genes as FAS and SREBP-1c and gluconeogenic genes as pepck and g6pc while downregulated, pparα, cpt1, acox1. Nevertheless, clenbuterol supplementation significantly reversed the aforementioned findings induced by HFD. Clenbuterol inclusion significantly improves growth performance and antioxidant defenses by modulating nrf2 signaling and reducing inflammatory response, reduces fatty acid synthesis, and enhances mitochondrial β-oxidation not only functioning as a lipid regulator and effectively alleviating fat accumulation in the liver but playing an essential role in the control of glucose metabolism by reducing hepatic glucose production in high-fat diet-fed Nile tilapias well.

Abstract Avian coccidiosis is one of the many disorders that seriously harm birds’ digestive systems. Nowadays the light is shed on using Phytochemical/herbal medicines as alternative natural anti-coccidial chemical-free standards. Consequently, this study aimed to investigate the impact of lawsonia inermis powder (LIP), and Acacia nilotica aqueous extract (ANAE), on growth performance, serum biochemical, antioxidant status, cytokine biomarkers, total oocyst count and intestinal histopathology of broiler chickens challenged with coccidiosis. Two hundred and forty-one-day-old Ross chicks were randomly distributed into 8 groups, four were challenged with coccidia, while the other four were unchallenged. Each group consisted of 3 replicates of 10 chicks each. The birds were challenged with Eimeria species orally on day 14 of age. Group 1B was unchallenged, and Group 2 A was challenged with coccidiosis and both were fed the basic diet without additives. Groups 3 A, 4 A, and 5 A were challenged and fed on the basic diet supplemented with LIP (40 g/kg of diet), ANAE (5 g/kg of diet), and LIP + ANAE combination, respectively. Groups 3B, 4B, and 5B were unchallenged and fed on the basic diet supplemented with LIP (40 g/kg of diet) and ANAE (5 g/kg of diet) and LIP + ANAE combination, respectively. The best results of growth performance parameters were recorded in G5B, and G5A followed by the group fed on ANAE and then the group fed on LIP compared with the control. All challenged broilers had higher aspartate aminotransferase (AST), alanine transaminase (ALT), urea, creatinine, glucose, MDA, IL-4 &TNF-α levels compared to all unchallenged broilers. Challenged broilers had lower serum cholesterol, triglycerides, total protein, albumin, globulin, SOD, GPX & IL-10 levels compared to non-challenged broilers. Histopathological examination of the small intestine and cecum of challenged treated groups with LIP + ANAE showed good mucosal integrity, few leukocytes infiltration, and low total oocyst count in broilers manure, followed by ANAE then LIP groups. In conclusion, dietary supplementation of lawsonia inermis powder and Acacia nilotica aqueous extract either alone or in combination had positive effects on broiler performance, blood metabolites, antioxidant status, cellular response, and intestinal morphology during the exposure to Eimeria spp. as a potential natural anti-coccidial.

Abstract Background FFPE tissue samples are commonly used in biomedical research and are a valuable source for next-generation sequencing in oncology, however, extracting RNA from these samples can be difficult the quantity and quality achieved can impact the downstream analysis. This study compared the effectiveness of seven different commercially available RNA extraction kits specifically designed for use with FFPE samples in terms of the quantity and quality of RNA recovered. Methods This study used 9 samples of FFPE tissue from three different types of tissue (Tonsil, Appendix and lymph node of B-cell lymphoma) to evaluate RNA extraction methods. Three sections of 20 µm of each sample were combined per sample. The slices were distributed in a systematic manner to prevent any biases. Each of the 7 commercially available RNA extraction kits were used according to manufacturer's instructions, with each sample being tested in triplicate resulting in a total of 189 extractions. The concentration, RNA quality score (RQS) and DV200 of each extraction was analysed using a nucleic acid analyser to determine the quantity and quality of the recovered RNA. Results This study found that despite processing the FFPE samples in the same standardized way, there were disparities in the quantity and quality of RNA recovered across the different tissue types. Additionally, the study found notable differences in the quantity of RNA recovered when using different extraction kits. In terms of quality, three of the kits performed better than the other four in terms of RQS and DV200 values. Conclusion Though many laboratories have developed their own protocols for specific tissue types, using commercially available kits is still a popular option. Although these kits use similar processes and extraction procedures, the amount and quality of RNA obtained can vary greatly between kits. In this study, among the kits tested, while the Roche kit, provided a nearly systematic better-quality recovery than other kits, the ReliaPrep FFPE Total RNA miniprep from Promega yielded the best ratio of both quantity and quality on the tested tissue samples.

IntroductionColorectal cancer (CRC) is a prevalent malignancy with significant morbidity and mortality worldwide. A deeper understanding of the interaction of cancer cells with other cells in the tumor microenvironment is crucial to devise effective therapeutic strategies. MUC2, a major component of the protective mucus layer in the gastrointestinal tract, has been implicated in CRC progression and immune response regulation.MethodIn this study, we sought to elucidate the relationship between MUC2 expression and immune infiltration within CRC using in vitro models involving two well-established cell lines, HT-29 and LS-174T. By employing CRISPR-mediated MUC2 knockout, we investigated the influence of MUC2 on tumor immune infiltration and its interplay with T cells and NK cells enriched peripheral blood mononuclear cells (PBMCs) in 3D spheroid cultures.ResultsWhile MUC2 was more abundant in LS-174T cell line compared to HT-29, its knockout resulted in increased immune infiltration solely in the HT-29 cell line, but not in the LS-174T cell line. We revealed that the removal of MUC2 protein was compensated in LS-174T by the expression of other gel-forming mucin proteins (MUC6, MUC5B) commonly expressed in the gastrointestinal epithelium, while this was not observed in HT-29 cell line.ConclusionOur study is the first to demonstrate that MUC2 functions as a physical barrier to immune infiltration in colorectal cancer (CRC) in vitro. In HT-29 cells, MUC2 knockout increased immune infiltration, while in LS-174T cells, compensatory expression of other mucins (MUC6, MUC5B) maintained the barrier. These findings reveal the complexity of mucin biology in CRC and suggest that targeting mucin pathways could be a novel therapeutic approach.

Background Hepatitis B virus (HBV) is a global health issue, particularly among healthcare personnel, including students because of its occupational exposure pattern. Healthcare Workers and medical students are recommended to have better knowledge, attitudes and good practices and vaccination toward infection control in general and HBV in particular. This study aimed to assess the knowledge, attitudes, and practices of medical students from North Sudan regarding HBV and its vaccination coverage. Methods A cross-sectional study was conducted from January to June 2023 among medical students in North Sudan. Data were collected using a structured scale using online Google Forms. Descriptive and comparative analyses were performed using SPSS version 26. Statistical significance was set at p ≤ 0.05. Results A total of 426 medical students were included in the study. The majority had good knowledge about HBV (86%) and its transmission methods (77%). The majority showed a favorable attitude (77%). The practice score was however very low (15.5%). Older age and advanced academic level were significantly associated with good knowledge (aOR: 3.9; CI 95% [3.69–12.92]; p = 0.016, and aOR: 2.6; CI 95% [1.16–6.15]; p = 0.020, respectively). Only a third of the students were vaccinated (33.0%), with only a few of them received complete doses (18.6%). Conclusion Students from medical colleges in North Sudan had good knowledge and favorable attitudes toward HBV. However, good practice was low. Older age and advanced academic level were significantly associated with knowledge level. Vaccination coverage was also low among the students in this study.

IntroductionLipopolysaccharide (LPS), a constituent of the outer membrane of Gram-negative bacteria, is a powerful inducer of systemic inflammation and has been extensively utilized in experimental models to simulate inflammatory responses and septic disorders. Recent research indicates that oxytocin (OXY), a neuropeptide typically linked to social bonding and reproductive functions, may influence inflammatory processes. This work examines the impact of OXY on LPS-induced testicular damage, aiming to elucidate its therapeutic potential in addressing inflammatory disorders and broadening the comprehension of its functions beyond conventional neuroendocrine roles.MethodsEighteen male albino rats were divided into three groups; the control group received no treatment; the LPS group received 0.5 mL of saline solution containing 5 mg/kg LPS intraperitoneally (orchitis model); and the LPS + OXY group received LPS and OXY (0.1 mg/kg) intraperitoneally every 12 h for 72 h.Results and discussionAnimals subjected to LPS were found to have severe orchitis, as evidenced by increased oxidative stress and surging inflammatory mediators (TNF-α, IL-1β, and IL-6), with declined IL-10 levels. Besides, LPS increased the malondialdehyde (MDA) and decreased the glutathione (GSH) levels, inducing an oxidative stress cascade. In addition, there are dramatic increases in the TLR4, MyD88, NF-κB, and PK2/PKR1 protein expression levels. All these events could alter the sperm count, morphology, and testicular architecture.ConclusionInterestingly, OXY could mitigate LPS-induced oxidative damage and inflammation in testicular tissue alongside restoring the disrupted sperm count, motility, and morphology. This therapeutic potential of OXY might be accounted for by its anti-inflammatory, antioxidant, and antiapoptotic activities.

Dynamic characteristics are of significant interest to researchers in the field of damage detection. Among these, natural frequencies stand out due to their high accuracy and resistance to noise. However, relying solely on natural frequencies is often insufficient for determining the depth and location of damage. To address this limitation, additional masses can be strategically placed at different locations on structural elements, altering the natural frequencies. Each mass placement creates a distinct dynamic scenario with a unique frequency profile, enabling a more comprehensive analysis. In this study, additional masses were introduced at specific elements of the beam structure within the numerical model which were then strategically placed at various locations along the beam. The resulting shifts in natural frequencies served as inputs to the Grey Wolf Optimizer (GWO), which identified elements with stiffness reductions indicative of damage. A custom MATLAB code was developed to perform finite element analysis on the numerical model. The results were validated against previously published experimental data, demonstrating the method’s reliability with a 5% difference. A parametric study involving both simple and continuous span beams was performed. The procedure effectively detected damage severities of 10%, 25%, and 50%, with corresponding errors of 4.3%, 0.44%, and 0.02%, respectively.

Background: One of the best ways to impart important skills to trainees is through simulation-based training, which is more reliable than the conventional clinical examination method. It is used in pediatric nursing training to allow nurses to practice and improve their clinical and conversational skills during an actual child encounter. A heel-prick is a complex psychomotor task that requires skill and knowledge from the pediatric nurse performing the procedure while applying for the National Newborn Screening Program. Aim: This study aimed to assess the impact of simulation-based training on pediatric nurses' knowledge and performance regarding the heel-prick technique used during newborn blood screening tests in select hospitals in Saudi Arabia and Egypt. Methods: This study utilized an experimental pre-test and post-test design. The G*Power Program® Version 3.1.9.4 was employed to determine the sample size to fulfil the study's objectives. The sample consisted of 50 nurses recruited from the Maternity & Children Hospital Bisha, Al-Namas General Hospital in the Kingdom of Saudi Arabia, and Pediatric Assiut University Hospital in Egypt; they were randomly divided into two groups of 25 nurses, intervention and control. The sample size was calculated assuming α level of 0.05, a β level of 0.20, and a desired power of 80%. The study was conducted from June 1st to August 30th, 2023. Two data collection instruments were used: a structured questionnaire and an observational checklist for the heel-prick technique in newborn bloodspot screening sample collection. Before the training intervention, a pre-test assessing knowledge and performance was administered. Intervention and control groups received two hours of instruction every 5 days. The researcher utilized PowerPoint presentations, videos, and interactive learning session intervention to deliver the instructional content to increase nurses' knowledge and also employed mannequins for a newborn blood spot test simulator to train participants on performance. A post-test 1 was administered immediately after the training sessions. Additionally, post-test 2 was conducted 20 days after the study and followed up after 40 days of intervention (post-test 3). Results: The study participants ranged in age from 25 to 30 years, with the majority being female. A statistically significant difference was observed in the knowledge and performance of pediatric nurses following simulation-based training. The majority of nurses demonstrated correct responses after the simulation-based learning intervention. However, the nurses' knowledge varied, as evidenced by the mean scores of their total knowledge regarding the heel-prick for the newborn bloodspot screening test. Immediately after the learning intervention session, the mean scores were 37.86 ± 1.28 and 34.84 ± 1.22 in the experimental and control groups, respectively. At 20 days post-training, the mean scores were 40.16 ± 1.11 and 33.55 ± 1.18 in the experimental and control groups, respectively, with a highly statistically significant difference (P-value = 0.0001). Moreover, at 40 days post-training, the mean scores were 39.54 ± 1.09 and 29.66 ± 1.32 in the experimental and control groups, respectively, and a statistically significant difference was found (P-value = 0.0001). Conversely, the level of nurses' performance before training was below average, with 52% and 56% in the experimental and control groups, respectively. Immediately after the simulation-based learning, the majority of nurses in the experimental group demonstrated a good level of performance in post-test 1, post-test 2, and post-test 3, with a statistically significant difference compared to the nurses in the control group (P-value = 0.001). Conclusion: The use of newborn blood spot test simulators is useful during simulation-based training in raising the overall level of the pediatric nurses' knowledge and performance, especially through 1, 2, & 3 post-training tests. The findings have several practical implications, and one is that pediatric nurses' performance and knowledge concerning newborn blood screening tests should be prioritized to guarantee patient safety and quality of care in pediatric patient scenarios. It is essential for nursing trainers to effectively train pediatric nurses' by using simulators to improve their caring knowledge and performance in educational settings. By improving nurses' knowledge and performance, we can reduce the dangers caused by their training on real neonates; also, it can be an attractive way to train nurses when trained by simulators and ultimately enhance the overall quality of nursing services in the hospital. Liposomal encapsulated phytogenics, such as liposomal hesperetin, are considered novel substitutes for antibiotics in the broiler industry owing to their improved nutritional and therapeutic properties. Therefore, our key goal was to investigate liposomal hesperetin impact on broiler growth performance, health, antioxidant status, tight junction proteins (TJP), and resistance against Listeria monocytogenes. Four broiler groups were fed 0, 150, 250, or 400 mg/kg of liposomal hesperetin-supplemented diets and experimentally infected with L. monocytogenes strain. Herein, liposomal hesperetin, especially at higher concentrations, augmented broilers FCR with upregulation of genes encoding TJP (occludin, JAM-2, MUC-2), and antioxidant attributes (GPX-1, SOD-1, CAT, HO-1, NQO1, COX2), which reflect enhancing health and welfare of broilers. Muscle antioxidant biomarkers were enhanced; meanwhile, muscle MDA, ROS, and H2O2 levels were reduced in response to 400 mg/kg of liposomal hesperetin. Liposomal hesperetin fortification reduced L. monocytogenes loads and expression levels of its virulence-related genes (flaA, hlyA, and ami). Remarkably, histopathological alterations in intestinal and brain tissues of L. monocytogenes-infected broilers were restored post-inclusion at higher levels of liposomal hesperetin, which reflects increase of the birds' resistance to L. monocytogenes infection. Transcription levels of genes encoding cytokines/chemokines (MyD88, AVBD6, CCL20, IL-1β, IL-18), and autophagy (Bcl-2, LC3, AMPK, AKT, CHOP, Bip, p62, XBP1) were ameliorated following dietary liposomal hesperetin fortification, which suggests enhancement of the birds' immunity and health. Collectively, our research recommends liposomal hesperetin application in broiler diets owing to its promoting impact on growth performance, antioxidant status, immunity, health, and welfare besides its antibacterial, and antivirulence characteristics to fight against L. monocytogenes. Background: Self- medication leading to wastage of limited resources in developing countries, prolonged suffering, increase resistance to drugs and may result in significant medical complications such as adverse drug responses and dependence. Self-medication is extensively used by health professionals. Undergraduate medical students as being the future physicians representing a main pillar in health care system thus have special significance. This study aimed to estimate the prevalence of self-medication and identify its pattern among medical students in Tanta university, Egypt. Methods: A four-month cross-sectional study was carried out among Egyptian undergraduate medical students at Tanta University, from first to final year. (November 2023 – February 2024). The students were chosen using a two-stage cluster sampling method, and data on sociodemographic and self-medication patterns were collected using a predesigned semi-structured self-administered questionnaire. Results: The prevalence of self-medication was 71%. The key determinants of self-medication was students' medical knowledge from self- experience and studies (55.9%). Headache was a common complaint for self-medication (80.4%). Majority of them (88.3%) use pharmaceutical products where analgesics lies on top (92.4%). The degree of popularity of the medicine was the primary reason for drug selection (52.5%) and recommendation of the pharmacist was the main determinate of selecting type of drug (43.6%). However, 30.2% experienced side effects, of which 50% went to private physician and 33.5% stopped taking their medications. Half of the students took antibiotics for self-medication. Conclusions: Self-medication is a common practice among medical students. where headache was the common symptom and the most commonly utilized medications for self-medication were analgesics.
