Abstract Paneth cells (PCs), a subset of intestinal epithelial cells (IECs) found at the base of small intestinal crypts, play an essential role in maintaining intestinal homeostasis. Altered PCs function is associated with diverse intestinal pathologies, including ileal Crohn’s disease (CD). CD patients with ileal involvement have been previously demonstrated to display impairment in PCs and decreased levels of anti-microbial peptides. Although the immunosuppressive drug Azathioprine (AZA) is widely used in CD therapy, the impact of AZA on IEC differentiation remains largely elusive. In the present study, we hypothesized that the orally administered drug AZA also exerts its effect through modulation of the intestinal epithelium and specifically via modulation of PC function. AZA-treated CD patients exhibited an ileal upregulation of AMPs on both mRNA and protein levels compared to non-AZA treated patients. Upon in vitro AZA stimulation, intestinal epithelial cell line MODE-K exhibited heightened expression levels of PC marker in concert with diminished cell proliferation but boosted mitochondrial OXPHOS activity. Moreover, differentiation of IECs, including PCs differentiation, was boosted in AZA-treated murine small intestinal organoids and was associated with decreased D-glucose consumption and decreased growth rates. Of note, AZA treatment strongly decreased Lgr5 mRNA expression as well as Ki67 positive cells. Further, AZA restored dysregulated PCs associated with mitochondrial dysfunction. AZA-dependent inhibition of IEC proliferation is accompanied by boosted mitochondria function and IEC differentiation into PC.

Abstract Breast cancer is one of the leading causes of death in females, mainly because of metastasis. Oncometabolites, produced via metabolic reprogramming, can influence metastatic signaling cascades. Accordingly, and based on our previous results, we propose that metabolites from highly metastatic breast cancer cells behave differently from less-metastatic cells and may play a significant role in metastasis. For instance, we aim to identify these metabolites and their role in breast cancer metastasis. Less metastatic cells (MCF-7) were treated with metabolites secreted from highly metastatic cells (MDA-MB-231) and the gene expression of three epithelial-to-mesenchymal transition (EMT) markers including E-cadherin, N-cadherin and vimentin were examined. Some metabolites secreted from MDA-MB-231 cells significantly induced EMT activity. Specifically, hypoxanthine demonstrated a significant EMT effect and increased the migration and invasion effects of MCF-7 cells through a hypoxia-associated mechanism. Hypoxanthine exhibited pro-angiogenic effects via increasing the VEGF and PDGF gene expression and affected lipid metabolism by increasing the gene expression of PCSK-9. Notably, knockdown of purine nucleoside phosphorylase, a gene encoding for an important enzyme in the biosynthesis of hypoxanthine, and inhibition of hypoxanthine uptake caused a significant decrease in hypoxanthine-associated EMT effects. Collectively for the first time, hypoxanthine was identified as a novel metastasis-associated metabolite in breast cancer cells and represents a promising target for diagnosis and therapy.

The use of 12-bit levels of grayscale in medical image representation allows for more precise image analysis and diagnosis, and encrypting such images is essential for protecting patient privacy and security. Existing 8-bit image encryption schemes become inefficient when applied to 12-bit images. In this paper, we design a cryptographically strong encryption scheme to improve the efficiency of encrypting 12-bit medical images. The core of the proposed scheme is a key-dependent $12\times 12$ S-box, which plays a crucial role in enhancing the security and efficiency of the encryption scheme. Most notably, test results show that $12\times 12$ S-boxes offer significantly stronger confusion and key-sensitivity than their $8\times 8$ counterparts. This finding enables the proposed $12\times 12$ S-box-based scheme to process 12-bit images 3.3 times faster than an $8\times 8$ S-box while passing all security tests. Experimental results show that the proposed scheme can encrypt 12-bit images at speeds up to 300MB/s. Moreover, the proposed scheme has also been shown to efficiently handle 8-bit images. Comparative results highlight the security and efficiency advantages of the proposed scheme over existing medical image encryption schemes.

Background: Obesity is a complex metabolic disorder that is associated with several diseases. Recently, precision nutrition (PN) has emerged as a tailored approach to provide individualised dietary recommendations. Aim: This review discusses the major intrinsic and extrinsic components considered when applying PN during the management of obesity and common associated chronic conditions. Results: The review identified three main PN components: gene–nutrient interactions, intestinal microbiota, and lifestyle factors. Genetic makeup significantly contributes to inter-individual variations in dietary behaviours, with advanced genome sequencing and population genetics aiding in detecting gene variants associated with obesity. Additionally, PN-based host-microbiota evaluation emerges as an advanced therapeutic tool, impacting disease control and prevention. The gut microbiome’s composition regulates diverse responses to nutritional recommendations. Several studies highlight PN’s effectiveness in improving diet quality and enhancing adherence to physical activity among obese patients. PN is a key strategy for addressing obesity-related risk factors, encompassing dietary patterns, body weight, fat, blood lipids, glucose levels, and insulin resistance. Conclusion: PN stands out as a feasible tool for effectively managing obesity, considering its ability to integrate genetic and lifestyle factors. The application of PN-based approaches not only improves current obesity conditions but also holds promise for preventing obesity and its associated complications in the long term.

This study presents a comprehensive investigation into the evolutionary trajectories of Rhipicephalus ticks (Ixodidae) through the interpretation of molecular phylogenetics, elucidating their chromatographic spectrum. The use of advanced chromatographic tools in this study explored the dynamics chemical profiling, providing valuable insights into the evolutionary history and ecological adaptations. Prevalence of Rhipicephalus ticks was 4.5% in sheep and 3.9% in goats. The ITS2 sequence of the Rhipicephalus sanguineus (OK642408) and Rhipicephalus microplus (OK642409) form a distinct clade with sequences from other countries. The 16S rRNA sequences of R. sanguineus (OK560870) clustered with sequences form three lineages, tropical, temperate, and south‐eastern. The Cox I gene‐identified Rhipicephalus turanicus (OK623472) and R. microplus (OK623463) form separate clades with sequences. The HPLC chromatogram of tick samples reveals a diverse array of identified hydrocarbons, explained the complex chemical composition of their exoskeletons. This analytical approach provides valuable insights into the specific hydrocarbon profiles, allowing for potential applications in species differentiation, ecological studies, and a deeper understanding of the functional roles played by hydrocarbon compounds in tick physiology. The findings revealed the potential of applying molecular phylogenetics tools with chromatography not only to enhance our understanding of tick evolution but also to inform strategies for disease control and management in regions where Rhipicephalus ticks (Ixodidae) are endemic. The brown wheat mite, Petrobia tritici, poses a significant threat to wheat fields. While fertilizers can increase crop productivity, imbalanced application may exacerbate plant susceptibility to pests. This study aimed to evaluate the impact of various NPK fertilization programs on P. tritici infestations over two consecutive cropping seasons. The results revealed significant differences in mite infestation among the treatment groups (p < 0.001). The lowest populations (1.1 and 3.0 mites/leaf) were observed in the treatments sprayed with phosphoric acid (at 0.75 and 1.00 cm/L), where the infestation appeared approximately 120 days after sowing; in contrast, it appeared early at 75 days in the other treatments. Conversely, treatments lacking potassium fertilizer presented the greatest degree of mite injury levels (49.5–57.7 mites/leaf). Although these treatments provided moderate leaf nutrition and crop yield, the highest nutritional content and total yield (10.49 and 9.71 1 t/ha for the two years) were observed in the treatment that received 224:70:100 kg fad−1 commercial fertilizers (=178:25:114 kg ha−1 NPK units) as soil fertilization, which was followed by the treatment with a foliar application of phosphoric acid (1.00 cm/L) with a total yield of 9.34 and 8.53 1 t/ha for the two years. In this treatment, the P. tritici density was moderately high at 9.40 and 6.32 mites/leaf over the two years, respectively. The consistency of P. tritici density and total yield ranking across both years indicated reliable estimates of the impact of fertilization. This study suggests that potassium sulfate application is crucial for reducing P. tritici density and that foliar phosphoric acid application instead of soil application reduces the number of P. tritici and delays its occurrence. Plants are subjected to various biotic and abiotic stresses that significantly impact their growth and productivity. To achieve balanced crop growth and yield, including for leafy vegetables, the continuous application of micronutrient is crucial. This study investigates the effects of different concentrations of copper sulphate (0, 75, 125, and 175 ppm) on the morphological and biochemical features of Spinacia oleracea and Avena sativa. Morphological parameters such as plant height, leaf area, root length, and fresh and dry weights were optimized at a concentration of 75 ppm copper sulfate. At this concentration, chlorophyll a & b levels increased significantly in Spinacia oleracea (462.9 and 249.8 휇푔/푔), and Avena sativa (404.7 and 437.63휇푔/푔). However, carotenoid content and sugar levels in Spinacia oleracea were negatively affected, while sugar content in Avena sativa increased at 125 ppm (941.6 µg/ml). Protein content increased in Spinacia oleracea (75 ppm, 180.3 µg/ml) but decreased in Avena sativa. Phenol content peaked in both plants at 75 ppm (362.2 and 244.5 µg/ml). Higher concentrations (175 ppm) of copper sulfate reduced plant productivity and health. Plants exposed to control and optimal concentrations (75 and 125 ppm) of copper sulpate exhibited the best health and growth compared to those subjected to higher concentrations. Maximum plant height, leaf area, root length, fresh and dry weights were observed at lower concentrations (75 and 125 ppm) of copper sulfate, while higher concentrations caused toxicity. Optimal copper sulfate levels enhanced chlorophyll a, chlorophyll b, total chlorophyll, protein, and phenol contents but inhibited sugar and carotenoid contents in both Spinacia oleracea and Avena sativa. The aim of the present research was to evaluate the efficiency of different vegetation indices (VI) obtained from satellites with varying spatial resolutions in discriminating the phenological stages of soybean crops. The experiment was carried out in a soybean cultivation area irrigated by central pivot, in Balsas, MA, Brazil, where weekly assessments of phenology and leaf area index were carried out. Throughout the crop cycle, spectral data from the study area were collected from sensors, onboard the Sentinel-2 and Amazônia-1 satellites. The images obtained were processed to obtain the VI based on NIR (NDVI, NDWI and SAVI) and RGB (VARI, IV GREEN and GLI), for the different phenological stages of the crop. The efficiency in identifying phenological stages by VI was determined through discriminant analysis and the Algorithm Neural Network–ANN, where the best classifications presented an Apparent Error Rate (APER) equal to zero. The APER for the discriminant analysis varied between 53.4% and 70.4% while, for the ANN, it was between 47.4% and 73.9%, making it not possible to identify which of the two analysis techniques is more appropriate. The study results demonstrated that the difference in sensors spatial resolution is not a determining factor in the correct identification of soybean phenological stages. Although no VI, obtained from the Amazônia-1 and Sentinel-2 sensor systems, was 100% effective in identifying all phenological stages, specific indices can be used to identify some key phenological stages of soybean crops, such as: flowering (R1 and R2); pod development (R4); grain development (R5.1); and plant physiological maturity (R8). Age-related neurodegenerative disorders like Alzheimer's disease (AD) and Parkinson's disease (PD) are characterized by deposits of protein aggregates, or amyloid, in various regions of the brain. Historically, aggregation of a single protein was observed to be correlated with these different pathologies: tau in AD and α-synuclein (αS) in PD. However, there is increasing evidence that the pathologies of these two diseases overlap, and the individual proteins may even promote each other's aggregation. Both tau and αS are intrinsically disordered proteins (IDPs), lacking stable secondary and tertiary structure under physiological conditions. In this study we used a combination of biochemical and biophysical techniques to interrogate the interaction of tau with both soluble and fibrillar αS. Fluorescence correlation spectroscopy (FCS) was used to assess the interactions of specific domains of fluorescently labeled tau with full length and C-terminally truncated αS in both monomer and fibrillar forms. We found that full-length tau as well as individual tau domains interact with monomer αS weakly, but this interaction is much more pronounced with αS aggregates. αS aggregates also mildly slow the rate of tau aggregation, although not the final degree of aggregation. Our findings suggest that co-occurrence of tau and αS in disease are more likely to occur through monomer-fiber binding interactions, rather than monomer-monomer or co-aggregation. For a rooted cluster algebra (Q) over a valued quiver Q , a symmetric cluster variable is any cluster variable belonging to a cluster associated with a quiver σ (Q) , for some permutation σ. The subalgebra of (Q) generated by all symmetric cluster variables, is called the symmetric mutation subalgebra and is denoted by ℬ (Q). In this paper, we identify the class of cluster algebras that satisfy ℬ (Q) = (Q) , which contains almost every quiver of finite mutation type. In the process of proving the main result, we provide a classification of quivers mutations classes that relates their maximum weights to the shapes of the initial quivers. Consequently, there exists a pressing need to enhance our treatment strategies by placing greater emphasis on planning and dosimetric factors, given the availability of more conformal techniques and delineation criteria, achieving optimal goals of radiotherapy treatment. The primary aim of this article is to discuss how the extent of regional nodal coverage influences the choice between IMRT and 3D radiation therapy for patients. Patients and Methods: A total of 15 patients diagnosed with left breast cancer with disease involved lymph nodes were included in this study. Delivering the recommended dose required the use of a linear accelerator (LINAC) with photon beams energy of 6 mega voltage (6MV). Each patient had full breast radiation using two planning procedures: intensity-modulated radiotherapy (IMRT) and three-dimensional radiotherapy (3D conformal). Following the guidelines set forth by the Radiation Therapy Oncology Group (RTOG), the planned treatment coverage was carefully designed to fall between 95% and 107% of the recommended dose. Additionally, Dose Volume Histograms (DVHs) were generated the dose distribution within these anatomical contours. Results and Conclusion: The DVH parameters were subjected to a comparative analysis, focusing on the doses absorbed by both Organs at Risk (OARs) and the Planning Target Volume (PTV). The findings suggest that low doses in IMRT plan might raise the risk of adverse oncological outcomes or potentially result in an increased incidence of subsequent malignancies. Consequently, the adoption of inverse IMRT remains limited, and the decision to opt for this therapy should be reserved for situations where it is genuinely necessary to uphold a satisfactory quality of life. Additionally, this approach helps in reducing the likelihood of developing thyroid problems and mitigates the risk of injuries to the supraclavicular area and the proximal head of the humerus bone. In this work, tungsten oxide (WO3) particles (in micro and nanoscales) have been mixed with silicon epoxy resin to create a novel composite to enhance the radiation attenuation properties of an epoxy resin shield. Six epoxy resin samples were created using various WO3 micro/nanoparticles concentrations. Several radioactive sources with varying energies were employed along with a high-purity germanium detector to evaluate the prepared samples' shielding capabilities. The linear attenuation coefficients (LACs) were measured experimentally using the narrow beam method. Based on the experimentally obtained values of LACs, other radiation shielding parameters, including half value length (HVL), tenth value layer (TVL), radiation shielding efficiency (RSE), and mean free path (MFP), were computed. The EW-20m30n sample (50% epoxy + 20% micro WO3 + 30% nano WO3) had the lowest MFP, HVL, and TVL at any given energy. Thus, in terms of material thickness, the EW-20m30n sample offers the best shielding characteristics. Furthermore, because of its superior density, the almost even mixture of nanoparticles and microparticles in the EW-20m30n sample produces a RSE of ≅ 100% in the low energy zone. Conversely, it was found that the samples' ability to protect decreased as the energy increased. Calculating HVL, TVL, and MFP revealed a high energy dependence that grew as the incident photon energy rose. On the other hand, for 50% micro WO3 and 50% epoxy, the EW-50m sample had the least ideal shielding characteristics. Categorically, our study has demonstrated that adding WO3 micro/nanoparticles to epoxy resin polymer has improved the polymer's radiation shielding properties. [ABSTRACT FROM AUTHOR]

Introduction and Background: Diabetes is a chronic metabolic disease characterized by elevated blood glucose levels and is one of the main global health concerns. Synthetic sugar substrate has many side effects such as leukemia, bladder cancer, hepatotoxicity, breast cancer, headache, and brain toxicity. The WHO and FDA has recently banned some of the synthetic sugar alternatives due to their carcinogenic effects. Objective and Methodology: Therefore, the main objective of the current study was to investigate the safety and binding affinity of Stevioside with Glucose Transpoter-4 (GLUT-4), Akt, Insulin Receptor (IR) and Insulin Receptor Substrate-1 (IRS-1) to confirmed that Stevioside is one the potent natural sweetener/drug for diabetes. This study delves into the molecular interaction between Stevioside and key diabetic proteins: GLUT-4, Akt, IR and IRS-1. A precise molecular docking approach was used to simulate the binding affinity of Stevioside to these proteins. The pharmacokinetic properties of the molecule should be taken into consideration as important variables throughout the virtual screening process. Results: The result of active site analysis of GLUT-4, Akt, IR and IRS-1 showed a zone of 2158.359 Ų, 579.259 Ų, 762.651 Ų, and 152.167 Ų and a volume of 2765.094 ų, 355.567 ų, 686.806 ų, and 116.874 ų, respectively. Docking analysis of the Stevioside compound showed the highest docking energy with scores of -9.9 with GLUT-4, -6.7 with Akt, -8.0 with IR and -8.8 with IRS-1. Studies indicated that it remains undigested by stomach acids and enzymes and is not absorbed in the upper small intestine. Further, tests revealed no hepatotoxicity, AMES toxicity, or skin sensitivity, making it a promising candidate for safe consumption as drug metabolism. Conclusion and Recommendations: Instead of other sugar alternatives, Stevioside will help diabetic patients with a lower chance of infections, lowered blood pressure/blood sugar, and increased glucose uptake in diabetic muscles. In this work, we studied the effect of bismuth oxide particle size and its attenuation capacity as a filler additive in epoxy resins. Six samples were prepared according to the amount of microparticles and nanoparticles in the sample and were coded as ERB-1, ERB-2, ERB-3, ERB-4, ERB-5, and ERB-6. One of the composite epoxies contained Bi2O3 microparticles at a 50:50 ratio (ERB-6) and was chosen as the control composite, and the number of microparticles (MPs) was gradually decreased and replaced by nanoparticles (NPs) to produce epoxy-containing Bi2O3 nanoparticles at a 50:50 ratio (ERB-1). The morphological and thermal characteristics of the studied composites were tested. The attenuation capability of the prepared composites, which is determined by the Bi2O3 particle size, was determined experimentally using a semiconductor detector, an HPGe-detector, and three different gamma-ray point sources (Am-241, Co-60, and Cs-137). The linear attenuation coefficient (LAC) of ERB-3, which contained 30% nanoparticles and 20% microparticles, had the highest value compared to the other composites at all the energies discussed, while the ERB-6 composite had the lowest value at all energies. The radiation-shielding efficiency (RSE) of the prepared samples was determined at all discussed energies; at 662 keV, the radiation-shielding efficiency values were 15.97%, 13.94%, and 12.55% for ERB-3, ERB-1, and ERB-6, respectively. The statistics also proved that the attenuation capacities of the samples containing a combination of nanoparticles and microparticles were much superior to those of the samples containing only microparticles or nanoparticles. Background: Three Amino acid Loop Extension (TALE) belongs to the homeobox group of genes that are important constituents of plant systems. The TALE gene family is instrumental not only in growth and development but also plays an essential role in regulating plant response to environmental adversaries. Results: In the present study, we isolated 21 CsTALE genes from the cucumber (Cucumis sativus L.) genome database. Bioinformatics tools were put in place to understand the structural and functional components of the CsTALE gene family. The evolutionary analysis dissected them into seven subclades (KNOX-I, KNOX-II, and BELL-I to BELL-V). The cis-acting elements in the promoter region of CsTALE genes disclosed that they are key regulators of hormonal and stress-related processes. Additionally, the STRING database advocated the concerting role of CsTALE proteins with other key transcription factors potent in plant developmental biology. The CsmiR319 and CsmiR167a-3p targeting the CsTALE15 and CsTALE16, respectively, further assert the importance of the CsTALE gene family posttranscriptional-related processes. Tissue-specific gene expression unfolded the fundamental involvement of CsTALE genes as they were expressed throughout the developmental stages. Under waterlogging stress, the CsTALE17 expressed significantly higher values in WL, WL-NAA, and WL-ETH but not in WL-MeJA-treated samples. Conclusions: The present study reveals the evolution and functions of the CsTALE gene family in cucumber. A botnet attack is a major cybersecurity threat that involves coordinated control of a network of infected computers, enabling large-scale distributed denial of service (DDoS) attacks, malware spreading, and other cybercrime activities. Proactive security measures and advanced threat intelligence systems are essential to detect and mitigate these assaults. This paper proposes the Harris Hawks Optimization (HHO) algorithm, which employs exploration and exploitation techniques to find optimal solutions for analyzing botnet attack pathways. The proposed approach involves HHO as a feature selector for extracting features from anomalous network traffic. The algorithm’s impact on botnet IP positioning performance is analyzed, considering different optimization modes and control center accuracy. The paper is organized into sections covering attack path establishment and analysis, system testing and verification, and a central leadership entity controls it [1]. Botnets are created based on the use of malicious software packages to infect important and sensitive devices in the network, thus making servers, computers, and Internet of Things devices vulnerable [2]. To detect these attacks and limit their impact requires many proactive security measures such as strong network security settings, regular software upgrades, etc. [3]. HHO is a powerful method that has the potential to solve many functional optimization problems and provides a suitable environment for engineering applications, as it mimics the exploration and exploitation phases during the foraging process of Harris Hawks [4]. A model based on HHO algorithm is proposed in this paper that has the ability to track and analyze bot attack paths by extracting a set of features during abnormal network traffic. The results were analyzed and their impact on the performance of robot networks was discussed, based on the use of different experimental results. After configuring the network topology and determining the attack path based on HHO, the performance of the algorithm and its effectiveness in preventing IP addresses from being spoofed are verified. The results showed convergence in being able to correct attack paths and effective performance in repelling the interference of fake IP addresses. [ABSTRACT FROM AUTHOR]
